  • Genotypic and phenotypic spectrum of infantile liver failure due to pathogenic TRMU variants

    • Purpose: This study aimed to define the genotypic and phenotypic spectrum of reversible acute liver failure (ALF) of infancy resulting from biallelic pathogenic TRMU variants and determine the role of cysteine supplementation in its treatment. Methods: Individuals with biallelic (likely) pathogenic variants in TRMU were studied within an international retrospective collection of de-identified patient data. Results: In 62 individuals, including 30 previously unreported cases, we described 47 (likely) pathogenic TRMU variants, of which 17 were novel, and 1 intragenic deletion. Of these 62 individuals, 42 were alive at a median age of 6.8 (0.6-22) years after a median follow-up of 3.6 (0.1-22) years. The most frequent finding, occurring in all but 2 individuals, was liver involvement. ALF occurred only in the first year of life and was reported in 43 of 62 individuals; 11 of whom received liver transplantation. Loss-of-function TRMU variants were associated with poor survival. Supplementation with at least 1 cysteine source, typically N-acetylcysteine, improved survival significantly. Neurodevelopmental delay was observed in 11 individuals and persisted in 4 of the survivors, but we were unable to determine whether this was a primary or a secondary consequence of TRMU deficiency. Conclusion: In most patients, TRMU-associated ALF was a transient, reversible disease and cysteine supplementation improved survival.

  • Limbic-predominant age-related TDP-43 encephalopathy (LATE) : consensus working group report

    • We describe a recently recognized disease entity, limbic-predominant age-related TDP-43 encephalopathy (LATE). LATE neuropathological change (LATE-NC) is defined by a stereotypical TDP-43 proteinopathy in older adults, with or without coexisting hippocampal sclerosis pathology. LATE-NC is a common TDP-43 proteinopathy, associated with an amnestic dementia syndrome that mimicked Alzheimer's-type dementia in retrospective autopsy studies. LATE is distinguished from frontotemporal lobar degeneration with TDP-43 pathology based on its epidemiology (LATE generally affects older subjects), and relatively restricted neuroanatomical distribution of TDP-43 proteinopathy. In community-based autopsy cohorts, similar to 25% of brains had sufficient burden of LATE-NC to be associated with discernible cognitive impairment. Many subjects with LATE-NC have comorbid brain pathologies, often including amyloid-beta plaques and tauopathy. Given that the oldest-old' are at greatest risk for LATE-NC, and subjects of advanced age constitute a rapidly growing demographic group in many countries, LATE has an expanding but under-recognized impact on public health. For these reasons, a working group was convened to develop diagnostic criteria for LATE, aiming both to stimulate research and to promote awareness of this pathway to dementia. We report consensus-based recommendations including guidelines for diagnosis and staging of LATE-NC. For routine autopsy workup of LATE-NC, an anatomically-based preliminary staging scheme is proposed with TDP-43 immunohistochemistry on tissue from three brain areas, reflecting a hierarchical pattern of brain involvement: amygdala, hippocampus, and middle frontal gyrus. LATE-NC appears to affect the medial temporal lobe structures preferentially, but other areas also are impacted. Neuroimaging studies demonstrated that subjects with LATE-NC also had atrophy in the medial temporal lobes, frontal cortex, and other brain regions. Genetic studies have thus far indicated five genes with risk alleles for LATE-NC: GRN, TMEM106B, ABCC9, KCNMB2, and APOE. The discovery of these genetic risk variants indicate that LATE shares pathogenetic mechanisms with both frontotemporal lobar degeneration and Alzheimer's disease, but also suggests disease-specific underlying mechanisms. Large gaps remain in our understanding of LATE. For advances in prevention, diagnosis, and treatment, there is an urgent need for research focused on LATE, including in vitro and animal models. An obstacle to clinical progress is lack of diagnostic tools, such as biofluid or neuroimaging biomarkers, for ante-mortem detection of LATE. Development of a disease biomarker would augment observational studies seeking to further define the risk factors, natural history, and clinical features of LATE, as well as eventual subject recruitment for targeted therapies in clinical trials.

  • Effect of momentum relaxation on exciton spin dynamics in diluted magnetic semiconductor ZnMnSe CdSe superlattices

    • cw hot photoluminescence (PL) complemented by transient PL measurements is employed to evaluate momentum and spin relaxation of heavy hole (HH) excitons in ZnMnSe CdSe superlattices. The rate of acoustic-phonon assisted momentum relaxation is concluded to be comparable to the total rate of exciton decay processes, about (2-3) × 1010 s-1, independent of applied magnetic fields. In magnetic fields when the Zeeman splitting ? of the exciton states is below the energy of the longitudinal optical (LO) phonon (?LO), a surprisingly strong suppression of spin relaxation rate from the bottom of the upper spin band is observed, which becomes comparable to that of momentum scattering via acoustic phonons. On the other hand, dramatic acceleration of the spin relaxation process by more than one order of magnitude is found for the excitons with a high momentum K. The findings are interpreted as being due to electron and hole spin flip processes via exchange interaction with isolated Mn2+ ions. Experimental evidence for the efficient interaction between the hot excitons and Mn impurities is also provided by the observation of spin flip transitions within Mn2+ - Mn2+ pairs that accompany the momentum relaxation of the hot HH excitons. In higher magnetic fields ?= ?LO, abrupt shortening of the spin flip time is observed. It indicates involvement of a new and more efficient spin relaxation process and is attributed to direct LO-assisted exciton spin relaxation with a subpicosecond spin relaxation time. © 2005 The American Physical Society.

    • We show, by time-resolved magneto-photoluminescence (PL) spectroscopy in combination with selective laser excitation, that optical polarization of the ZnCdSe spin detector induced by spin injection from the ZnMnSe spin injector persists over a much longer time scale than the lifetime of the ZnMnSe excitons. This finding provides compelling experimental evidence that the dominant mechanism for the observed spin injection in the ZnMnSe/ZnCdSe structures should not be due to injection of the excitonic spins of the diluted magnetic semiconductor (DMS). It is rather due to e.g. a delayed spin injection arising from tunneling of individual carriers or/and trapped spins in ZnMnSe.

    •  Spin depolarization of a ZnCdSe quantum-well spin detector (SD) in ZnMnSe/ZnCdSe light-emitting quantum structures is investigated by cw and time-resolved optical orientation spectroscopy. It is shown that spin depolarization is governed by three distinct spin relaxation processes with the corresponding polarization decay times of 850, 30, and <10 ps. The dominant and the fastest process is attributed to spin relaxation accompanying energy relaxation of hot excitons (and hot carriers) within the SD, providing evidence that it can be an important source of spin loss, leading to the observed limited efficiency of optical spin injection in the structures.

    • We report on time-resolved photoluminescence studies of exciton dynamics in a ZnMnSe/ZnSSe type-II quantum well. Strong separation of the electrons, confined to the nonmagnetic semiconductor ZnSSe, and holes, confined to the diluted magnetic semiconductor ZnMnSe, results in an exciton lifetime exceeding 3 ns. Two excitonic emission lines are observed in this structure at low temperatures. The lower-energy line was associated with the formation of an exciton magnetic polaron (EMP), whereas the higher-energy line was attributed to the emission of nonmagnetically localized excitons. The intensity of the EMP line was found to saturate at a relatively low density of photoexcited carriers well below the exciton Mott density. This behavior evidences a limited density of sites where the hole localization radius is small enough to ensure the fast formation of EMP's with a noticeable binding energy. © 2006 The American Physical Society.

    • In this work, we address the issue of spin relaxation and its relevance to spin detection in ZnO-based materials, by spin-polarized, time-resolved magneto-optical spectroscopy. We have found that spin relaxation is very fast, i.e. about 100 ps for donor bound excitons in wurtzite ZnO, despite of a weak spin–orbit interaction. We also reveal that alloying of ZnO with Cd enhances spin relaxation, prohibiting ZnCdO/ZnO structures for efficient optical spin detection. On the other hand, a variation in strain field induced by lattice mismatch with substrates does not seem to lead to a noticeable change in spin relaxation. The observed fast spin relaxation, together with the limitation imposed by the band structure, are thus identified as the two most important factors that limit the efficiency of optical spin detection in the studied ZnO-based materials.

    • Two dominant factors limiting efficiency of optical spin detection in ZnO-based materials system are identified from time-resolved optical orientation and magneto-optical studies. The first is related to the fundamental band structure of the materials characterized by a weak spin-orbit interaction. It leads to cancellation of circular polarization from the optical transitions between the conduction band and the A and B valence band states, which would otherwise carry the desired information on spin polarization of carriers. The second limiting factor is shown to be efficient carrier/exciton spin relaxation, i.e., about 45-80 ps.

    • Optical carrier/exction and spin injection processes from a ZnMnSe dilute magnetic semiconductor (DMS) to CdSe quantum dots (QD’s) are studied in detail by means of spinpolarized magneto- photoluminescence (PL) and PL excitation spectroscopies. Efficiency of carrier/exciton transfer is found to be practically independent of width (Lb) of a ZnSe barrier layer inserted between the DMS and QD’s. This is tentatively explained in terms of photonexchange energy transfer. In sharp contrast, spin injection efficiency is found to be largely suppressed in the structures with large Lb, pointing towards increasing spin loss.

    • Magneto-optical spectroscopy in combination with tunable laser spectroscopy is employed to study the effect of sample design on optical spin-injection efficiency from a Zn(Cd)MnSe diluted magnetic semiconductor (DMS) into non-magnetic CdSe quantum dots (QDs). Two samples with inverted growth sequence of the DMS and QD layers were grown for this purpose. Despite observation of clear evidence for exciton/carrier-injection from the DMS to the QDs in both structures, their spin-injection efficiency is markedly different. The structure grown with the QD layer topmost exhibits rather efficient spin-injection, while the structure grown with the DMS layer topmost does not show any sign of spin-injection. Even in the former case, the spin-polarization of the injected excitons/carriers is deduced to be about 32%, decreasing from 100% before the injection. The observed spin loss is shown to occur during the spin-injection across the heterointerfaces. © 2007 Elsevier B.V. All rights reserved.

    • We investigate ensemble electron spin dephasing in self-assembled InGaAs/GaAs quantum dots (QDs) of different lateral sizes by employing optical Hanle measurements. Using low excitation power, we are able to obtain a spin dephasing time T-2* (in the order of ns) of the resident electron after recombination of negative trions in the QDs. We show that T-2* is determined by the hyperfine field arising from the frozen fluctuation of nuclear spins, which scales with the size of QDs following the Merkulov-Efros-Rosen model. This scaling no longer holds in large QDs, most likely due to a breakdown in the lateral electron confinement. (C) 2015 AIP Publishing LLC.

    • To determine the regulatory mechanism of the expression of the mouse platelet-derived growth factor (PDGF) beta-receptor gene, a 1.9-kb 5' flanking genomic fragment was cloned and analyzed. Site-directed mutagenesis of a CCAAT motif, located 60 bp upstream of the transcriptional-start site, completely abolished the promoter activity [Ballagi, A. E., Ishisaki, A., Nelin, J.-O. & Funa, K. (1995) Biochem. Biophys. Res. Commun. 210, 165-1751. The sequence around the intact CCAAT motif was protected by in vitro DNase-I-footprinting analysis. Electrophoresis-mobility-shift assays with anti-[nuclear factor Y(NF-Y)]Ig revealed binding of the NF-Y complex to the CCAAT box. Furthermore, the double-stranded oligonucleotides corresponding to the sequence around the CCAAT motif were conjugated with DNA-affinity magnetic beads. The binding proteins were affinity purified and identified as the NF-Y transcription factor by western blotting. Our results indicate that NF-Y controls the basal transcription activity of the mouse PDGF beta-receptor gene.

    • Dynamics of spin injection has been studied in double quantum wells (DQWs) composed of diluted magnetic and non-magnetic semiconductors. Picosecond-transient photoluminescence (PL) of excitons in the DQWs has been measured in magnetic field. In the Cd1-xMnxTe-based DQWs, the PL intensity of the magnetic well (MW) excitons decays faster with decreasing barrier width from 12 to 2 nm. This provides the evidence for carrier tunneling from the MW to the non-magnetic well (NW) through the barrier layer. The degree of circular polarization in the transient PL of the NW exciton in magnetic field shows marked evidence for spin injection and rapid spin relaxation in the DQW system. In the Zn1-yMnySe-based DQWs, the degree of circle polarization in the NW exciton PL at 3T shows a rise with a time constant of 400 ps, while the PL of the MW exciton decays within 50 ps. The observed result is interpreted by the individual spin injection for electrons and holes from the MW to the NW. The results of the transient PL of excitons in the DQWs are compared with the transient absorptions of excitons studied by the pump-and-probe spectroscopy. (c) 2006 Elsevier B.V. All rights reserved.

    • Pathological accumulation of abnormally phosphorylated tau protein in astrocytes is a frequent, but poorly characterized feature of the aging brain. Its etiology is uncertain, but its presence is sufficiently ubiquitous to merit further characterization and classification, which may stimulate clinicopathological studies and research into its pathobiology. This paper aims to harmonize evaluation and nomenclature of aging-related tau astrogliopathy (ARTAG), a term that refers to a morphological spectrum of astroglial pathology detected by tau immunohistochemistry, especially with phosphorylation-dependent and 4R isoform-specific antibodies. ARTAG occurs mainly, but not exclusively, in individuals over 60 years of age. Tau-immunoreactive astrocytes in ARTAG include thorn-shaped astrocytes at the glia limitans and in white matter, as well as solitary or clustered astrocytes with perinuclear cytoplasmic tau immunoreactivity that extends into the astroglial processes as fine fibrillar or granular immunopositivity, typically in gray matter. Various forms of ARTAG may coexist in the same brain and might reflect different pathogenic processes. Based on morphology and anatomical distribution, ARTAG can be distinguished from primary tauopathies, but may be concurrent with primary tauopathies or other disorders. We recommend four steps for evaluation of ARTAG: (1) identification of five types based on the location of either morphologies of tau astrogliopathy: subpial, subependymal, perivascular, white matter, gray matter; (2) documentation of the regional involvement: medial temporal lobe, lobar (frontal, parietal, occipital, lateral temporal), subcortical, brainstem; (3) documentation of the severity of tau astrogliopathy; and (4) description of subregional involvement. Some types of ARTAG may underlie neurological symptoms; however, the clinical significance of ARTAG is currently uncertain and awaits further studies. The goal of this proposal is to raise awareness of astroglial tau pathology in the aged brain, facilitating communication among neuropathologists and researchers, and informing interpretation of clinical biomarkers and imaging studies that focus on tau-related indicators.

    • We study propagation dynamics of exciton spins in a system composed of high-density self-assembled quantum dots (QDs) of CdSe and a diluted magnetic semiconductor (DMS) layer of ZnMnSe, where spin-polarized excitons are generated in the DMS layer and are subsequently injected into the QDs. The degree of circular polarization, P, of excitonic photoluminescence at 5 T in the coupled QDs shows a rapid increase with increasing delay time after a linearly polarized pulsed excitation, indicating the exciton-spin injection from the ZnMnSe to CdSe-QDs. The P value tends to decay gradually because of the exciton-spin relaxation in the QDs after the injection. The spin-polarized excitons in the QD ensemble migrate simultaneously from the QDs with higher exciton energies to those with lower exciton energies, where the exciton-spins relax inside each dot during the migration. Therefore, the observed P values in the QD emission band are affected by this feeding of the relaxed spins.

    • We show that circularly polarized emission light from InGaAs/GaAs quantum dot (QD) ensembles under optical spin injection from an adjacent GaAs layer can switch its helicity depending on emission wavelengths and optical excitation density. We attribute this anomalous behavior to simultaneous contributions from both positive and negative trions and a lower number of photo-excited holes than electrons being injected into the QDs due to trapping of holes at ionized acceptors and a lower hole mobility. Our results call for caution in reading out electron spin polarization by optical polarization of the QD ensembles and also provide a guideline in improving efficiency of spin light emitting devices that utilize QDs.

    • Power-dependent time-resolved optical spin orientation measurements were performed on In0.1Ga0.9As quantum well(QW) and In0.5Ga0.5As quantum dot (QD) tunnel-coupled structures with an 8-nm-thick GaAs barrier. A fast transient increase of electron spin polarization was observed at the QW ground state after circular-polarized pulse excitation. The temporal maximum of polarization increased with increasing pumping fluence owing to enhanced spin blocking in the QDs, yielding a highest amplification of 174% with respect to the initial spin polarization. Further elevation of the laser power gradually quenched the polarizationdynamics, which was induced by saturated spin filling of both the QDs and the QW phase spaces.

    • Time-resolved optical spin orientation spectroscopy was employed to investigate the temperature-dependent electron spin injection in In0.1Ga0.9As quantum well (QW) and In0.5Ga0.5As quantum dots (QDs) tunnel-coupled nanostructures with 4, 6, and 8 nm-thick GaAs barriers. The fast picosecond-ranged spin injection from QW to QDexcited states (ES) was observed to speed up with temperature, as induced by pronounced longitudinal-optical (LO)-phonon-involved multiple scattering process, which contributes to a thermally stable and almost fully spin-conserving injection within 5–180 K. The LO-phonon coupling was also found to cause accelerated electron spin relaxation of QD ES at elevated temperature, mainly via hyperfine interaction with random nuclear field.

    • Transient magneto-optical spectroscopy of InGaNGaN and InGaNGaMnN quantum wells reveals a spin relaxation process with a characteristic time of 50 ps. We show that the observed spin relaxation is mediated by spin flips of individual carriers rather than by direct exciton spin flips, and is proposed to occur near the bottom of the exciton band (K=0). Nearly complete thermalization between spin sublevels of the excitons, observed immediately after the pulsed photoexcitation, is attributed to even faster spin relaxation of photogenerated hot carriers/excitons accompanying momentum and energy relaxation at high K vectors. © 2005 American Institute of Physics.

    • Spin polarization of nonmagnetic CdSe quantum dots (QDs) coupled to adjacent ZnMnSe diluted magnetic semiconductor (DMS) is investigated by CW and time-resolved magneto-optical spectroscopy under tunable laser excitation. Efficient enhancement in the degree of σ− circular polarization of photoluminescence from the CdSe QDs is observed under optical excitation at the σ+-active exciton state of the DMS. The fact that the enhancement persists much longer than the exciton lifetime of the DMS rules out a role of the DMS excitons. A possible explanation is discussed in terms of antiferromagnetic coupling between the excitons in QDs and aligned Mn ions in DMS.
