Common variants at the MHC locus and at chromosome 16q24.1 predispose to Barrett's esophagus.

- Barrett's esophagus is an increasingly common disease that is strongly associated with reflux of stomach acid and usually a hiatus hernia, and it strongly predisposes to esophageal adenocarcinoma (EAC), a tumor with a very poor prognosis. We report the first genome-wide association study on Barrett's esophagus, comprising 1,852 UK cases and 5,172 UK controls in the discovery stage and 5,986 cases and 12,825 controls in the replication stage. Variants at two loci were associated with disease risk: chromosome 6p21, rs9257809 (Pcombined=4.09×10(-9); odds ratio (OR)=1.21, 95% confidence interval (CI)=1.13-1.28), within the major histocompatibility complex locus, and chromosome 16q24, rs9936833 (Pcombined=2.74×10(-10); OR=1.14, 95% CI=1.10-1.19), for which the closest protein-coding gene is FOXF1, which is implicated in esophageal development and structure. We found evidence that many common variants of small effect contribute to genetic susceptibility to Barrett's esophagus and that SNP alleles predisposing to obesity also increase risk for Barrett's esophagus.

Robust estimation of bacterial cell count from optical density

- Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

- That leadership development is a contested terrain, like any organizational terrain, can scarcely be considered a new idea, yet research into the intricacies of resistance in this context is very much in its infancy. This article takes recent critical scholarship on resistance as its starting point to explore the interdependencies of power, resistance and struggle in a leadership development environment. Drawing on extensive online interactions collected from an 18-month, cross-sector programme with emergent leaders, this article asks whether the different stakeholders in leadership development could benefit from a more open exploration of power and resistance. Such dynamics offer new insights into the relationship between participants and facilitators and raise a series of alternative questions, challenges and strategies for leadership development.

AusTraits, a curated plant trait database for the Australian flora

- We introduce the AusTraits database - a compilation of values of plant traits for taxa in the Australian flora (hereafter AusTraits). AusTraits synthesises data on 448 traits across 28,640 taxa from field campaigns, published literature, taxonomic monographs, and individual taxon descriptions. Traits vary in scope from physiological measures of performance (e.g. photosynthetic gas exchange, water-use efficiency) to morphological attributes (e.g. leaf area, seed mass, plant height) which link to aspects of ecological variation. AusTraits contains curated and harmonised individual- and species-level measurements coupled to, where available, contextual information on site properties and experimental conditions. This article provides information on version 3.0.2 of AusTraits which contains data for 997,808 trait-by-taxon combinations. We envision AusTraits as an ongoing collaborative initiative for easily archiving and sharing trait data, which also provides a template for other national or regional initiatives globally to fill persistent gaps in trait knowledge.

Current Transition Practice for Primary Immunodeficiencies and Autoinflammatory Diseases in Europe: a RITA-ERN Survey.

- Due to the absence of curative treatments for inborn errors of immunity (IEI), children born with IEI require long-term follow-up for disease manifestations and related complications that occur over the lifespan. Effective transition from pediatric to adult services is known to significantly improve adherence to treatment and long-term outcomes. It is currently not known what transition services are available for young people with IEI in Europe.To understand the prevalence and practice of transition services in Europe for young people with IEI, encompassing both primary immunodeficiencies (PID) and systemic autoinflammatory disorders (AID).A survey was generated by the European Reference Network on immunodeficiency, autoinflammatory, and autoimmune diseases Transition Working Group and electronically circulated, through professional networks, to pediatric centers across Europe looking after children with IEI.Seventy-six responses were received from 52 centers, in 45 cities across 17 different countries. All services transitioned patients to adult services, mainly to specialist PID or AID centers, typically transferring up to ten patients to adult care each year. The transition process started at a median age of 16-18years with transfer to the adult center occurring at a median age of 18-20years. 75% of PID and 68% of AID centers held at least one joint appointment with pediatric and adult services prior to the transfer of care. Approximately 75% of PID and AID services reported having a defined transition process, but few centers reported national disease-specific transition guidelines to refer to.Transition services for children with IEI in Europe are available in many countries but lack standardized guidelines to promote best practice.

Genetic studies of body mass index yield new insights for obesity biology.

- Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

- Leadership development theory and practice is increasingly turning its gaze on identity as a primary focus for development efforts. Most of this literature focuses on how the identities of participants are strengthened, repaired and evolved. This article focuses on identity work practices that are underdeveloped in the literature: the deconstruction, unravelling and letting go that can be experienced when working upon one's self. We group these experiences, among others, under the conceptual term identity undoing' and, based on findings from an 18-month ethnographic study of a leadership development program, we offer five manifestations of how it can be experienced. Through foregrounding the undoing of identity, we are able to look more closely at how power relations shape the leadership development experience. In order to raise questions and propositions for leadership and its development we use a micro-sociological and interactionist approach to explore the interplay between identity and power.

- The subject of this study is a community theatre for, with and by girls aged 13–17. The theatre practice is situated within the field of applied theatre and as a communitarian practice engages with artistic, social and political ambitions by staging the everyday of marginalised people. When describing and researching these kinds of theatre practices, emphasis is often placed on their democratic potential and positive notions of empowerment. However, the challenges which arise when participants with little or no previous theatre experience are to perform their lives in front of an audience are not often discussed in research. Accordingly, the aim of this ethnographic study is to explore how the participants’ lives are turned into creative and artistic expressions in the drama room and staged in front of an audience. By working with post-constructionist and spatial theories, humans as well as space, place and affectivity are acknowledged as co-producing actors. These theoretical positions, articulated by philosophers such as Rosi Braidotti, Gillian Deleuze and Michael Foucault, enable an analysis of how place, playfulness and vulnerability influence the theatre practice and how it simultaneously produces both possibilities and constraints for the girls. In four articles, the study addresses how tensions are produced in the process of staging the everyday. Article I engages with Foucault’s concept of heterotopia, addressing how the drama room is co-created with local context while also being an isolated room. Through an analytical mapping, the article discusses how theatre practice enables the girls to act creatively in relation to limiting norms in the drama room and other places such as school. Article II focuses on how vulnerability is both a generative and restrictive force when creating artistic expressions. The study points towards the importance of professional leadership that can navigate through multilayered spaces that are explorative as well as artistic. Article III breaks with the somewhat taken-for-granted role of playfulness in community theatre and applied theatre. The analysis implies that although playfulness creates agency in relation to restricting norms, playfulness can at the same time reinforce these norms. The article also challenges ideas of how playfulness bridges the gap between creative explorations and professional art. Article IV explores how the content of the theatre practice is created through negotiations between bodies, affects and feeling as well as space and place. Through the concept of an extended relational didactics, the article also discusses how the leader can relate to a content that is highly unpredictable and co-created by several actors. Altogether, the four articles show how community theatre is both a relational and spatial practice that is consistently transforming. Moreover, they show how frustration, confusion and exclusion as well as pride, joy and agency are part of the practice by exploring tensions such as those that exist between ethics and aesthetics. The study suggests that the participants’ experience is more likely to be positive if the leaders are able to navigate through the multiple spaces that are produced in the theatre practice while also being firmly established in the local context.
