  • Breaking Free and Settling Down : Contradictory Cultural Meanings of Rural Retirement Migration among Swedish Seniors
  • 2013
  • Ingår i: Nordic Seniors on the Move. Mobility and Migration in Later Life. - 2001-7510 .- 2001-7529. - 9789198145809 ; 4:4, s. 27-50
  • Bokkapitel (refereegranskat)abstract
    • In this chapter, I argue that moving as a way to counter the negative aspects of ageing (actual as well as symbolic), is both dependent on mobility and contradictory to it. The overall purpose is to investigate the relation between moving and mobility in the accounts told by people who have chosen to migrate as seniors in order to enable a certain lifestyle. In what way does the discursive ideal of freedom and mobility, apparent in media and in popular science today, influence the way these seniors imagine, experience and talk about their move? To what extent do their accounts of retirement migration include traces of continuity and change, freedom and security, as well as mobility and rootedness? Could the move function as a way of breaking free and settling down simultaneously?

  • Narrating the moral geography of rape in Swedish newspapers
  • 2019
  • Ingår i: Rape Narratives in Motion. - Cham : Springer International Publishing. - 9783030138516 - 9783030138523 ; , s. 119-146
  • Bokkapitel (refereegranskat)abstract
    • In the master narrative of rape some women’s testimonies of sexual violence become particularly tainted with doubt and disbelief. Conversely, only some men’s sexually violent acts are unconditionally marked by guilt and blame. The chapter stresses the spatial dimension of this master narrative of rape. With an analysis of two hyper-medialised Swedish gang rape cases, it is highlighted how a moral geography is evoked in news narratives of rape. More specifically, it is argued that location, and narratives of women and men crossing boundaries, walking in line, being in place or invading space are used as proxy for the socio-spatial dimensions of power and morality.

  • Expression profiling of GIST: CD133 is associated with KIT exon 11 mutations, gastric location, and poor prognosis.
  • 2011
  • Ingår i: International journal of cancer. Journal international du cancer. - : Wiley. - 1097-0215 .- 0020-7136. ; 129:5, s. 1149-1161
  • Tidskriftsartikel (refereegranskat)abstract
    • In gastrointestinal stromal tumors (GISTs), KIT exon 11 deletions are associated with poor prognosis. The aim of this study was to determine the gene expression profiles of GISTs carrying KIT exon 11 deletions and to identify genes associated with poor prognosis. Expression profiling was performed on 9 tumors with KIT exon 11 deletions and 7 without KIT exon 11 mutations using oligonucleotide microarrays. In addition, gene expression profiles for 35 GISTs were analyzed by meta-analysis. Expression of CD133 (prominin-1) protein was examined by tissue microarray (TMA) analysis of 204 GISTs from a population-based study in western Sweden. Survival analysis was performed on patients subjected to R0 resection (n=180) using the Cox proportional hazards model. Gene expression profiling, meta-analysis, and qPCR showed up regulation of CD133 in GISTs carrying KIT exon 11 deletions. Immunohistochemical analysis on TMA confirmed CD133 expression in 28% of all tumors. CD133 positivity was more frequent in gastric GISTs (48%) than in small intestinal GISTs (4%). CD133 positivity was also more frequent in GISTs with KIT exon 11 mutations (41%) than in tumors with mutations in KIT exon 9, PDGFRA, or wild-type tumors (0-17%). Univariate survival analysis showed a significant correlation between the presence of CD133 protein and shorter overall survival (hazard ratio=2.23, P=0.027). Multivariate analysis showed that CD133 provided additional information on patient survival compared to age, sex, NIH risk group and mutational status. CD133 is expressed in a subset of predominantly gastric GISTs with KIT exon 11 mutations and poor prognosis.

  • Gastrointestinal stromal tumors (GISTs) express somatostatin receptors and bind radiolabeled somatostatin analogs.
  • 2013
  • Ingår i: Acta oncologica (Stockholm, Sweden). - 1651-226X .- 0284-186X. ; 52:4, s. 783-792
  • Tidskriftsartikel (refereegranskat)abstract
    • Background. Gastrointestinal stromal tumors (GISTs) can be effectively treated with tyrosine kinase inhibitors (TKIs). However, some patients with GIST develop drug resistance, and alternative treatment strategies are therefore needed. The aim of this study was to analyze the expression of somatostatin receptors (SSTR) in GIST as a target for peptide receptor-mediated radiotherapy (PRRT). Material and methods. Expression profiling of SSTR1-5 was performed on biopsies from 34 GISTs (16 gastric tumors, 15 small intestinal tumors, and three rectal tumors). SSTR scintigraphy ((111)In-octreotide) and measurement of (111)In activity in tumor specimens was performed in seven patients. Uptake and internalization of (177)Lu- octreotate was studied in primary cell cultures from two patients. Results. Quantitative PCR analysis showed expression of SSTR1 and SSTR2 in the majority of tumors, while SSTR3-5 were expressed at low levels. Immunohistochemical analysis confirmed the presence of SSTR1 and SSTR2 proteins in all GISTs, and SSTR3-5 in a subset of tumors. Diagnostic imaging by SSTR scintigraphy, using (111)In-octreotide, demonstrated tumor uptake of (111)In in three of six GIST patients. Measurement of (111)In activity in excised tumor specimens from five patients gave tumor-to-blood (T/B) activity ratios of between eight and 96. Tumor cells in primary culture (gastric and small intestinal GIST) specifically bound and internalized (177)Lu when incubated with the therapeutic compound (177)Lu-octreotate for 4-48 hours (p < 0.05). Conclusion. Peptide receptor-mediated radiotherapy via SSTR may provide a novel treatment strategy in carefully selected GIST patients with TKI-resistant tumors.

  • Combining CAR T cells and the Bcl-2 family apoptosis inhibitor ABT-737 for treating B-cell malignancy
  • 2013
  • Ingår i: Cancer Gene Therapy. - : Springer Science and Business Media LLC. - 0929-1903 .- 1476-5500. ; 20:7, s. 386-393
  • Tidskriftsartikel (refereegranskat)abstract
    • B-cell malignancies upregulate the B-cell lymphoma 2 (Bcl-2) family inhibitors of the intrinsic apoptosis pathway, making them therapy resistant. However, small-molecule inhibitors of Bcl-2 family members such as ABT-737 restore a functional apoptosis pathway in cancer cells, and its oral analog ABT-263 (Navitoclax) has entered clinical trials. Gene engineered chimeric antigen receptor (CAR) T cells also show promise in B-cell malignancy, and as they induce apoptosis via the extrinsic pathway, we hypothesized that small-molecule inhibitors of the Bcl-2 family may potentiate the efficacy of CAR T cells by engaging both apoptosis pathways. CAR T cells targeting CD19 were generated from healthy donors as well as from pre-B-ALL (precursor-B acute lymphoblastic leukemia) patients and tested together with ABT-737 to evaluate apoptosis induction in five B-cell tumor cell lines. The CAR T cells were effective even if the cell lines exhibited different apoptosis resistance profiles, as shown by analyzing the expression of apoptosis inhibitors by PCR and western blot. When combining T-cell and ABT-737 therapy simultaneously, or with ABT-737 as a presensitizer, tumor cell apoptosis was significantly increased. In conclusion, the apoptosis inducer ABT-737 enhanced the efficacy of CAR T cells and could be an interesting drug candidate to potentiate T-cell therapy.

  • Applying the World Café Method to Involve Users in an Interactive Analysis of Research Results
  • 2017
  • Ingår i: Innovation in Aging. - Oxford : Oxford University Press. - 2399-5300. ; 1:S1, s. 749-750
  • Tidskriftsartikel (refereegranskat)abstract
    • This presentation will discuss how the World Café method can be used to analyze data interactively together with users, in this case, organizers of senior camps. As part of the Senior Camp Study we were particularly interested in ways of “doing age” and how discourses on age influenced the organization of senior camps. The World Café was arranged with two main purposes: 1) involve users to discuss preliminary findings to validate and problematize the results; 2) use the discussions recorded during the World Café session as data to deepen the analysis. We experience that combining those two purposes was a challenge in the sense that the users were more keen on validating the results and explaining their standpoint than problematize around conceptions of age. We will elaborate and discuss this further in our presentation and also share our experiences of involving users in this stage of the research process. © The Authors 2017. • 2016
  • Ingår i: 23 NGK 2016. 23rd Nordic Congress of Gerontology. Tampere, Finland. - Tampere.
  • Konferensbidrag (refereegranskat)abstract
    • Senior camps for as a summerstay or resort have been established on several places in Sweden. These camps build on notions of what it means to age and to ”be elderly” in relation to activity, health, functional abilities and participation in society. In a cross-scientific and holistic study at CASE, Lund university these senior camps have been studied from three perspectives 1) History of ideas. What is the cultural meaning of senior camps? 2) Ethnology: how is age made visible and important at senior camps? 3) Occupational Science: What are the individual social and health aspects of the senior camp? In this presentation we focus on the results from part study 1. The data consists of texts concerning senior camps as well as interviews with directors for the senior camps. The camps are pictured as places where “elderly people almost become children again”. Simultaneously these camps are inprinted with nostalgic ideas of “Swedishnes” and “The good Old Age”. What structural and ideological values are made visible in these utterances and activities? We will discuss the contextual and ideological conditions for senior camps in Sweden and their relations to the historical heritage to other forms of summer camps in Sweden.

  • Pensioneer camps - about doing age and making difference
  • 2015
  • Ingår i: IAGG-ER 8th Congress Dublin 2015 - Unlocking the Demographic Dividend. - : The Irish Gerontological Society. ; , s. 337-337
  • Konferensbidrag (refereegranskat)abstract
    • "Pensioneer camps", summer camps aimed at older people, has for some years been established in several locations in Sweden. What unites the camps is the claimed positive effect they have for older people's health and quality of life. The stay is said to break isolation and lead to new habits and friendships that endure. This multidisciplinary study focuses on the phenomenon "Pensioneer camp" with the overall aim to study its health and life enhancing potential. Pensioneer camps are examined from a structural cultural-historical perspective focusing on the discourses in action, from an ethnographic perspective, focusing on identity construction and performance of age and from an activity perspective, focusing on quality of life and health in everyday life before and after the camp stay. Key issues are: What impact have older or more general discourses of "summer camp" on the activities? What age coded practices are made possible and how is age performed? How is the camp experienced by the elderly and it can be said to have any long-lasting impact in their lives after their stay? By studying both the individual, cultural and organizational aspects in relation to Pensioneer camps it enables both in- depth scientific knowledge of older people's quality of life and health in general, and, recommendations for future local investment in Pensioneer camps in terms of possibilities and limitations. © Irish Gerontological Society, 2015

  • Ingår i: Clinical Linguistics & Phonetics. - : Informa UK Limited. - 0269-9206 .- 1464-5076. ; 30:12, s. 944-958
  • Tidskriftsartikel (refereegranskat)abstract
    • The aims of the study were to investigate healthy subjects' performance on a clinical test of high-level language (HLL) and how it is related to demographic characteristics and verbal working memory (VWM). One hundred healthy subjects (20-79 years old) were assessed with the Swedish BeSS test (Laakso, Brunnegard, Hartelius, & Ahlsen, 2000) and two digit span tasks. Relationships between the demographic variables, VWM and BeSS were investigated both with bivariate correlations and multiple regression analysis. The results present the norms for BeSS. The correlations and multiple regression analysis show that demographic variables had limited influence on test performance. Measures of VWM were moderately related to total BeSS score and weakly to moderately correlated with five of the seven subtests. To conclude, education has an influence on the test as a whole but measures of VWM stood out as the most robust predictor of HLL.

  • Bead Arrays for Antibody and Complement Profiling Reveal Joint Contribution of Antibody Isotypes to C3 Deposition
  • 2014
  • Ingår i: PLOS ONE. - : Public Library of Science (PLoS). - 1932-6203. ; 9:5, s. e96403-
  • Tidskriftsartikel (refereegranskat)abstract
    • The development of antigen arrays has provided researchers with great tools to identify reactivities against self or foreign antigens from body fluids. Yet, these approaches mostly do not address antibody isotypes and their effector functions even though these are key points for a more detailed understanding of disease processes. Here, we present a bead array-based assay for a multiplexed determination of antigen-specific antibody levels in parallel with their properties for complement activation. We measured the deposition of C3 fragments from serum samples to reflect the degree of complement activation via all three complement activation pathways. We utilized the assay on a bead array containing native and citrullinated peptide antigens to investigate the levels of IgG, IgM and IgA autoantibodies along with their complement activating properties in serum samples of 41 rheumatoid arthritis patients and 40 controls. Our analysis revealed significantly higher IgG reactivity against the citrullinated fibrinogen beta and filaggrin peptides as well as an IgA reactivity that was exclusive for citrullinated fibrinogen b peptide and C3 deposition in rheumatoid arthritis patients. In addition, we characterized the humoral immune response against the viral EBNA-1 antigen to demonstrate the applicability of this assay beyond autoimmune conditions. We observed that particular buffer compositions were demanded for separate measurement of antibody reactivity and complement activation, as detection of antigen-antibody complexes appeared to be masked due to C3 deposition. We also found that rheumatoid factors of IgM isotype altered C3 deposition and introduced false-positive reactivities against EBNA-1 antigen. In conclusion, the presented bead-based assay setup can be utilized to profile antibody reactivities and immune-complex induced complement activation in a high-throughput manner and could facilitate the understanding and diagnosis of several diseases where complement activation plays role in the pathomechanism.

    • We study the dynamics of the interaction between the solar wind ions and a partially ionized atmosphere around a comet, at a distance of 2.88 AU from the Sun during a period of low nucleus activity. Comparing particle data and magnetic field data for a case study, we highlight the prime role of the solar wind electric field in the cometary ion dynamics. Cometary ion and solar wind proton flow directions evolve in a correlated manner, as expected from the theory of mass loading. We find that the main component of the accelerated cometary ion flow direction is along the antisunward direction and not along the convective electric field direction. This is interpreted as the effect of an antisunward polarization electric field adding up to the solar wind convective electric field.

    • Context. The first long-term in-situ observation of the plasma environment in the vicinity of a comet, as provided by the European Rosetta spacecraft.Aims. Here we offer characterisation of the solar wind flow near 67P/Churyumov-Gerasimenko (67P) and its long term evolution during low nucleus activity. We also aim to quantify and interpret the deflection and deceleration of the flow expected from ionization of neutral cometary particles within the undisturbed solar wind.Methods. We have analysed in situ ion and magnetic field data and combined this with hybrid modeling of the interaction between the solar wind and the comet atmosphere.Results. The solar wind deflection is increasing with decreasing heliocentric distances, and exhibits very little deceleration. This is seen both in observations and in modeled solar wind protons. According to our model, energy and momentum are transferred from the solar wind to the coma in a single region, centered on the nucleus, with a size in the order of 1000 km. This interaction affects, over larger scales, the downstream modeled solar wind flow. The energy gained by the cometary ions is a small fraction of the energy available in the solar wind.Conclusions. The deflection of the solar wind is the strongest and clearest signature of the mass-loading for a small, low-activity comet, whereas there is little deceleration of the solar wind.

  • Tidskriftsartikel (refereegranskat)abstract
    • The flow direction of low-energy ions around comet 67P/Churyumov–Gerasimenko has previously been difficult to constrain due to the influence of the spacecraft potential. The Ion Composition Analyzer of the Rosetta Plasma Consortium (RPC-ICA) on Rosetta measured the distribution function of positive ions with energies down to just a few eV/q throughout the escort phase ofthe mission. Unfortunately, the substantial negative spacecraft potential distorted the directional information of the low-energy data. In this work, we present the flow directions of low-energy ions around comet 67P, corrected for the spacecraft potential using Particle-In-Cell simulation results. We focus on the region in and around the diamagnetic cavity, where low-energy ions are especially important for the dynamics. We separate between slightly accelerated ‘burst’ features and a more constant ‘band’ of low-energy ions visible in the data. The ‘bursts’ are flowing radially outwards from the nucleus with an antisunward component while the ‘band’ is predominantly streaming back towards the comet. This provides evidence of counter-streaming ions, which has implications for the overall expansion velocity of the ions. The backstreaming ions are present also at times when the diamagnetic cavity was not detected, indicating that the process accelerating the ions back towards the comet is not connected to the cavity boundary.

    • Y Comets are constantly interacting with the solar wind. When the comet activity is high enough, this leads to the creation of a magnetic field free region around the nucleus known as the diamagnetic cavity. It has been suggested that the ion-neutral drag force is balancing the magnetic pressure at the cavity boundary, but after the visit of Rosetta to comet 67P/Churyumov-Gerasimenko the coupling between ions and neutrals inside the cavity has been debated, at least for moderately active comets. In this study, we use data from the ion composition analyser to determine the bulk speeds and temperatures of the low-energy ions in the diamagnetic cavity of comet 67P. The low-energy ions are affected by the negative spacecraft potential, and we use the Spacecraft Plasma Interaction Software to model the resulting influence on the detected energy spectra. We find bulk speeds of 5-10 km s(-1) with a most probable speed of 7 km s(-1), significantly above the velocity of the neutral particles. This indicates that the collisional coupling between ions and neutrals is not strong enough to keep the ions at the same speed as the neutrals inside the cavity. The temperatures are in the range 0.7-1.6 eV, with a peak probability at 1.0 eV. We attribute the major part of the temperature to the fact that ions are born at different locations in the coma, and hence are accelerated over different distances before reaching the spacecraft.

  • Ingår i: Environmental Research Letters. - : IOP Publishing. - 1748-9326. ; 6:3, s. 035103-
  • Tidskriftsartikel (refereegranskat)abstract
    • Wind turbines emit low frequency noise (LFN) and large turbines generally generate more LFN than small turbines. The dominant source of LFN is the interaction between incoming turbulence and the blades. Measurements suggest that indoor levels of LFN in dwellings typically are within recommended guideline values, provided that the outdoor level does not exceed corresponding guidelines for facade exposure. Three cross-sectional questionnaire studies show that annoyance from wind turbine noise is related to the immission level, but several explanations other than low frequency noise are probable. A statistically significant association between noise levels and self-reported sleep disturbance was found in two of the three studies. It has been suggested that LFN from wind turbines causes other, and more serious, health problems, but empirical support for these claims is lacking.

    • Depot medroxyprogesterone acetate (DMPA) is an injectable hormonal contraceptive used by millions of women worldwide. However, experimental studies have associated DMPA use with genital epithelial barrier disruption and mucosal influx of human immunodeficiency virus (HIV) target cells. We explored the underlying molecular mechanisms of these findings. Ectocervical biopsies and cervicovaginal lavage (CVL) specimens were collected from HIV-seronegative Kenyan sex workers using DMPA (n = 32) or regularly cycling controls (n = 64). Tissue samples were assessed by RNA-sequencing and quantitative imaging analysis, whereas protein levels were measured in CVL samples. The results suggested a DMPA-associated upregulation of genes involved in immune regulation, including genes associated with cytokine-mediated signaling and neutrophil-mediated immunity. A transcription factor analysis further revealed DMPA-associated upregulation of RELA and NFKB1 which are involved in several immune activation pathways. Several genes significantly downregulated in the DMPA versus the control group were involved in epithelial structure and function, including genes encoding keratins, small proline-rich proteins, and cell-cell adhesion proteins. Pathway analyses indicated DMPA use was associated with immune activation and suppression of epithelium development, including keratinization and cornification processes. The cervicovaginal microbiome composition (Lactobacillus dominant and non-Lactobacillus dominant) had no overall interactional impact on the DMPA associated tissue gene expression. Imaging analysis verified that DMPA use was associated with an impaired epithelial layer as illustrated by staining for the selected epithelial junction proteins E-cadherin, desmoglein-1 and claudin-1. Additional staining for CD4(+) cells revealed a more superficial location of these cells in the ectocervical epithelium of DMPA users versus controls. Altered protein levels of SERPINB1 and ITIH2 were further observed in the DMPA group. Identification of specific impaired epithelial barrier structures at the gene expression level, which were verified at the functional level by tissue imaging analysis, illustrates mechanisms by which DMPA adversely may affect the integrity of the genital mucosa. Author summarySexual transmission accounts for the majority of all new HIV infections in women, and alterations to the mucosal environment of the female genital tract have been associated with an increase in the risk of acquiring HIV. Observational epidemiological studies have implied that the use of the injectable hormonal contraceptive depot medroxyprogesterone acetate (DMPA) may be associated with increased HIV-acquisition. However, a prospective clinical study has not confirmed this association and the controversial findings are currently evaluated in the context of international reproductive health policies. Several studies using various model systems indicate that DMPA affects the integrity of the genital epithelial barrier as well as the mucosal immune system, but the exact mechanisms remain largely unknown. To characterize the effect of DMPA on the genital mucosal environment, we used a multi-omics approach to assess paired genital secretions and cervical tissue samples from long-term regular DMPA users living in Kenya. This unique cohort represents a population at risk of HIV infection in which DMPA is one of the most commonly used hormonal contraceptives. We identified impaired cervical epithelial barrier structures, including DMPA-associated reduction in the expression of cell-cell adhesion molecules, keratins, small proline-rich proteins and a thinner upper epithelial layer with more superficially located CD4(+) cells. Gene set enrichment pathway analyses indicated DMPA use was associated with immune activation and suppression of epithelium development including keratinization and cornification pathways. Protein analysis identified altered levels of selected anti-proteases. Our findings illustrate mechanisms by which DMPA adversely may affect the integrity of the genital mucosa.

  • Secondary cytoreductive surgery and hyperthermic intraperitoneal chemotherapy for recurrent colorectal peritoneal metastases
  • 2024
  • Ingår i: Surgery Open Science. - : Elsevier. - 2589-8450. ; 20, s. 45-50
  • Tidskriftsartikel (refereegranskat)abstract
    • Background:Secondary treatment of recurrent colorectal peritoneal metastases after previous cytoreductive surgery (CRS) and hyperthermic intraperitoneal chemotherapy (HIPEC) is poorly investigated. Objectives:To evaluate the overall survival outcome of secondary (repeat) CRS + HIPEC compared to palliative treatment in recurrent peritoneal disease. Methods:Patients with colorectal peritoneal metastases treated with an index CRS + HIPEC and subsequently having recurrent peritoneal disease were identified from the prospective Swedish national HIPEC registry. Patients were divided into interventional group (secondary CRS + HIPEC) or palliative group. Multivariable logistic regression, propensity-score matching, and survival outcomes were calculated. Results:Among 575 patients who underwent complete CRS between 2010 and 2021, 208 (36 %) were diagnosed with a subsequent recurrent peritoneal disease. Forty-two patients (20 %) were offered secondary CRS + HIPEC. Propensity-score matching of secondary interventional cases with palliative cases succeeded in 88 % (n = 37) in which female sex, lower peritoneal cancer index at index surgery, longer disease-free interval, and absence of extra-peritoneal metastases were identified as the most relevant matching covariates. Median OS from date of recurrence was 38 months (95%CI 30-58) in the interventional group and 19 months (95%CI: 15-24) in the palliative group (HR 0.35 95%CI: 0.20-0.63, p = 0.0004). Sensitivity analyses confirmed the results. As reference, the median OS from index CRS + HIPEC in the whole colorectal registry (n = 575) was 41 months (95%CI: 38-45).Conclusion:After matching for relevant factors, the hazard ratio for death was significantly reduced in patients who were offered a secondary CRS + HIPEC procedure for recurrent peritoneal disease. Selection bias is inherent, but survival outcomes were comparable to those achieved after the initial procedure.

  • Early salvage radiation therapy combined with short-term hormonal therapy in recurrent prostate cancer after radical prostatectomy: Single-institution 4-year data on outcome, toxicity, health-related quality of life and co-morbidities from 184 consecutive patients treated with 70 Gy.
  • 2013
  • Ingår i: International journal of oncology. - : Spandidos Publications. - 1791-2423 .- 1019-6439. ; 42:1, s. 109-17
  • Tidskriftsartikel (refereegranskat)abstract
    • The aim of this study was to investigate the role of 70Gy salvage radiotherapy (SRT) combined with short-term neoadjuvant hormonal therapy (NHT) in the treatment of recurrent disease after radical prostatectomy (RP), and to consider quality of life (QoL), survival outcomes and impact of co-morbidities on treatment-related rectal-genitourinary toxicity. Electronic records of 184 SRT patients treated consecutively between October 2001 and February 2007 were analyzed. Median age was 64 years (median follow-up 48months). NHT was given to 165 patients (median 3 months). Pre-RP and pre-SRT PSA, PSA doubling time, Gleason score (GS), seminal vesicle invasion (SVI) and detectable post-SRT PSA were recorded. Any detectable PSA or PSA >0.1 ng/ml + nadir was considered biochemical failure (BcF). The Charlson co-morbidity index was used to correlate co-morbidities and rectal-genitourinary toxicity. Scores from the health-related QoL EORTC QLQ-C30 and PR-25 questionnaires were also evaluated. In 116 (63%) patients, a long-lasting curative effect was indicated by undetectable PSA levels. In univariate analysis, using BcF as an outcome variable, p<0.001 was found for GS, pre-SRT PSA, SVI and detectable post-SRT PSA. Multivariate analysis showed p=0.01 for SVI, p=0.09 for GS, and detectable post-SRT PSA (p=0.01); with metastases as an outcome variable, only SVI was significant (p=0.007). Cancer-specific and overall survival were 99 and 95%, respectively. Although microscopy showed SVI or GS 8-10 in the prostatectomy specimens 17/40(43%) and 13/29 (45%), respectively, of patients still showed undetectable PSA at long-term follow-up (median 55 months) after SRT. Likewise, 11/31 (36%) patients with pre-SRT PSA >1.0 ng/ml and 80/134 (60%) patients with PSA doubling time (PSADT) <10 still showed undetectable PSA after 50 months. Slightly elevated acute and late rectal-genitourinary grade 3-4 toxicity was observed. No association with co-morbidity/toxicity was found. EORTC QLQ-C30 scores were similar to or slightly better than reference values. SRT with 70 Gy combined with 3-month NHT results in long-term undetectable PSA in >50% of patients with recurrence after RP with acceptable rectal-genitourinary toxicity and without negatively affecting long-term QoL. Non-metastatic patients should not be disqualified from receiving SRT although presenting with poor prognostic factors at surgery.

  • Distinct cervical tissue-adherent and luminal microbiome communities correlate with mucosal host gene expression and protein levels in Kenyan sex workers
  • 2023
  • Ingår i: Microbiome. - : Springer Nature. - 2049-2618. ; 11:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Background The majority of studies characterizing female genital tract microbiota have focused on luminal organisms, while the presence and impact of tissue-adherent ectocervical microbiota remain incompletely understood. Studies of luminal and tissue-associated bacteria in the gastrointestinal tract suggest that these communities may have distinct roles in health and disease. Here, we performed a multi-omics characterization of paired luminal and tissue samples collected from a cohort of Kenyan female sex workers. Results We identified a tissue-adherent bacterial microbiome, with a higher alpha diversity than the luminal microbiome, in which dominant genera overall included Gardnerella and Lactobacillus, followed by Prevotella, Atopobium, and Sneathia. About half of the L. iners-dominated luminal samples had a corresponding Gardnerella-dominated tissue microbiome. Broadly, the tissue-adherent microbiome was associated with fewer differentially expressed host genes than the luminal microbiome. Gene set enrichment analysis revealed that L. crispatus-dominated tissue-adherent communities were associated with protein translation and antimicrobial activity, whereas a highly diverse microbial community was associated with epithelial remodeling and pro-inflammatory pathways. Tissue-adherent communities dominated by L. iners and Gardnerella were associated with lower host transcriptional activity. Tissue-adherent microbiomes dominated by Lactobacillus and Gardnerella correlated with host protein profiles associated with epithelial barrier stability, although with a more pro-inflammatory profile for the Gardnerella-dominated microbiome group. Tissue samples with a highly diverse composition had a protein profile representing cell proliferation and pro-inflammatory activity. Conclusion We identified ectocervical tissue-adherent bacterial communities in all study participants of a female sex worker cohort. These communities were distinct from cervicovaginal luminal microbiota in a significant proportion of individuals. We further revealed that bacterial communities at both sites correlated with distinct host gene expression and protein levels. The tissue-adherent bacterial community could possibly act as a reservoir that seed the lumen with less optimal, non-Lactobacillus, bacteria.

  • Hur kuratorn balanserar känslor och byråkrati : blanketters betydelse i vårdmötet
  • 2022
  • Ingår i: Socialmedicinsk Tidskrift. - : Stiftelsen Socialmedicinsk Tidskrift. - 0037-833X. ; 99:1, s. 99-107
  • Tidskriftsartikel (refereegranskat)abstract

  • Tidskriftsartikel (refereegranskat)abstract
    • The article focuses on the work of clinical social workers who, on the one hand, respond to the emotions that children and families experience after the child is diagnosed with the chronic disease diabetes, on the other hand, deal with various bureaucratic processes aimed at ensuring that the child’s medical needs are met in the family’s new life. The work is directed towards two different worlds that are interdependent, yet bringing them together fre-quently necessitates a balancing act. The purpose is to examine how the social worker deal with these two worlds in the encounter with the child and the family. A result is the document’s role when the social worker translates the family’s new everyday life into a language that can generate assistance or allowance from the state.

  • The Use of Occupations to Challenge Perceptions of Age at a Senior Camp
  • 2016
  • Konferensbidrag (refereegranskat)abstract
    • Background: Senior camps have been established with the purpose to decrease social exclusion and improve health amongst elderly people. These senior camps build on notions of what it means to age and to ”be old” in relation to functional abilities, activity, participation and health.Method:Eight persons who participated in a senior camp were interviewed in their homes before and after the stay. The data was analysed using a narrative case-centred approach. Results: Seniors going to the camp were looking forward to meet friends and do things they did not commonly do. The pace at the camp, just being and not doing much, was appreciated by some while others whished for more organized activities. Being together with others, including eating together, talking and even sharing rooms was appreciated. Being close to nature was described to contribute both to peace of mind and to become spirited and strong, feelings that persisted after returning home. Being at a camp close to nature and the serving of traditional food was linked to memories from the childhood. Conclusion: Occupations were used both to consolidate and to challenge perceptions of age at the camp. The stay contributed to a feeling of belonging and experiences of doing during the camp and being in nature influenced the participants’ everyday life.Application to Practice: The experience of pace and place at the camp was important for a successful stay that could contribute to wellbeing. This presentation will elaborate on how occupations were used to challenge perceptions of age.

  • Learning-Environment Uncertainty and Students' Approaches to Learning : A Self-Determination Theory Perspective
  • 2023
  • Ingår i: Scandinavian Journal of Educational Research. - : Routledge. - 0031-3831 .- 1470-1170. ; 67:4, s. 559-573
  • Tidskriftsartikel (refereegranskat)abstract
    • In the literature on the relationship between learning environments and students' approaches to learning, much attention has been directed to aspects that foster a deep approach. Considerably less attention has been directed to aspects that result in the opposite, namely a surface approach. Indeed, there is a small literature focusing on how learning environments can frustrate the basic psychological needs of individuals and, as a result, foster a surface approach. However, hitherto, this stream has focused on controlling elements in learning environments. We add to this latter literature, through focusing on learning-environment uncertainty. This notion emerged from a qualitative study of 19 students at a Swedish university, where we identified three types of learning-environment uncertainty, related to the basic psychological needs for autonomy, competence, and relatedness. The findings suggest that a surface approach to learning can be understood as a coping strategy that students adopt to reduce such uncertainties.
