  • Differential vital staining of normal fibroblasts and melanoma cells by an anionic conjugated polyelectrolyte

    • Molecular probes for imaging of live cells are of great interest for studying biological and pathological processes. The anionic luminescent conjugated polythiophene (LCP) polythiophene acetic acid (PTAA), has previously been used for vital staining of cultured fibroblasts as well as transformed cells with results indicating differential staining due to cell phenotype. Herein, we investigated the behavior of PTAA in two normal and five transformed cells lines. PTAA fluorescence in normal cells appeared in a peripheral punctated pattern whereas the probe was more concentrated in a one-sided perinuclear localization in the five transformed cell lines. In fibroblasts, PTAA fluorescence was initially associated with fibronectin and after 24 h partially localized to lysosomes. The uptake and intracellular target in malignant melanoma cells was more ambiguous and the intracellular target of PTAA in melanoma cells is still elusive. PTAA was well tolerated by both fibroblasts and melanoma cells, and microscopic analysis as well as viability assays showed no signs of negative influence on growth. Stained cells maintained their proliferation rate for at least 12 generations. Although the probe itself was nontoxic, photoinduced cellular toxicity was observed in both cell lines upon irradiation directly after staining. However, no cytotoxicity was detected when the cells were irradiated 24 h after staining, indicating that the photoinduced toxicity is dependent on the cellular location of the probe. Overall, these studies certified PTAA as a useful agent for vital staining of cells, and that PTAA can potentially be used to study cancer-related biological and pathological processes.

  • Pregnancy Associated Plasma Protein-A as a Cardiovascular Risk Marker in Patients with Stable Coronary Heart Disease During 10 Years Follow-Up-A CLARICOR Trial Sub-Study

    • Elevated pregnancy-associated plasma protein A (PAPP-A) is associated with mortality in acute coronary syndromes. Few studies have assessed PAPP-A in stable coronary artery disease (CAD) and results are conflicting. We assessed the 10-year prognostic relevance of PAPP-A levels in stable CAD. The CLARICOR trial was a randomized controlled clinical trial including outpatients with stable CAD, randomized to clarithromycin versus placebo. The placebo group constituted our discovery cohort (n = 1.996) and the clarithromycin group the replication cohort (n = 1.975). The composite primary outcome was first occurrence of cardiovascular event or death. In the discovery cohort, incidence rates (IR) for the composite outcome were higher in those with elevated PAPP-A (IR 12.72, 95% Confidence Interval (CI) 11.0-14.7 events/100 years) compared to lower PAPP-A (IR 8.78, 8.25-9.34), with comparable results in the replication cohort. Elevated PAPP-A was associated with increased risk of the composite outcome in both cohorts (discovery Hazard Ratio (HR) 1.45, 95% CI 1.24-1.70; replication HR 1.29, 95% CI 1.10-1.52). In models adjusted for established risk factors, these trends were attenuated. Elevated PAPP-A was associated with higher all-cause mortality in both cohorts. We conclude that elevated PAPP-A levels are associated with increased long-term mortality in stable CAD, but do not improve long-term prediction of death or cardiovascular events when added to established predictors.

    • The Swedish Civil Air Traffic Control (SCAT) dataset consists of 13 weeks of data collected from the area control in Sweden flight information region. The dataset consists of detailed data from almost 170,000 flights as well as airspace data and weather forecasts. The flight data includes system updated flight plans, clearances from air traffic control, surveillance data and trajectory prediction data. Each week of data is continuous but the 13 weeks are spread over one year to provide variations in weather and seasonal traffic patterns. The dataset does only include scheduled flights not involved in any incident reports. Sensitive data such as military and private flight has been removed.The SCAT dataset can be useful for any research related to air traffic control, e.g. analysis of transportation patterns, environmental impact, optimization and automation/AI.

  • Performance using low-cost gaze-control for simulated flight tasks

    • In the current study, interaction using gaze control was compared to computer mouse using the MATB-II (Multi-Attribute Task Battery) environment. The study had two aims; the first was to explore the utility of low-cost technologies in a rapid prototyping and testing environment for aviation. The second aim was to use such an environment to compare a novel interaction device (a low-cost gaze control device) to a familiar interaction device (computer mouse). Method: Thirty participants performed two scenarios with each interaction device. The software MATB-II provided simulated flight tasks and recorded performance. Mental workload was assessed by the NASA Task Load Index (TLX) questionnaire after each scenario. Results: The results showed that gaze control resulted in significantly higher overall mental workload than computer mouse. Performance was better with mouse in two of the four tasks. Conclusions: Concerning the first aim, the study demonstrated the value of low-cost technology for initial user testing before using more expensive high-fidelity environments. Concerning the second aim, the computer mouse resulted in better performance and lower mental workload. This may either be due to higher user familiarity with computer mouse interaction or to limitations of the gaze control equipment and insufficient adjustments of the interface design to optimize for gaze control.
