    • This dissertation contains an exploratory study of how four Swedish media companies has decided to use social media in their daily work, and also how they in fact use them. What are the goals with implementing social media, how do they differ between different companies and do the goals transfer into the newsrooms?The four companies studied are Sveriges Radio, Aftonbladet, Dalarnas Tidningar and Gotlands Media. Media companies that differ from one another both in size, geographical coverage and economy. They range from small local newspapers to national public service radio and the biggest tabloid on the Swedish market. The methods used are qualitative research interviews and field studies as well as quantitative data analysis. The theoretical chapter helps define traditional journalism and journalistic practice. The concepts of boundaries, participatory cultures and normalizing of work methods are also discussed and helps build a context for the analysis of the empirical data.Some key findings from the study are that there are more similarities than differences between the various companies in both how they aim to use social media and how they in fact use them. That the digital development in the surrounding society are much faster than the implementation of new technologies in the newsrooms. Also, that there are differences between how the companies aim to use social media and the actual usage. For instance, all companies strive for a more active dialog with the audience in their visionary work, but has a hard time turning this into reality.There is also notable that social media in this study do not change the journalistic fundamental routines and traditions. The studied journalists are still in control of their own content and they aim to meet the same standard in journalistic quality when working in social media. It is also evident that Facebook is invaluable for all the studied companies, which gives Facebook an interesting indirect power over the journalistic work process and product. Finally, the study finds that it is hard to engage the audience in an active collaboration even when working with social media.

  • Campaigning Against Europe? : The Role of Euroskeptic Fringe and Mainstream Parties in the 2009 European Parliament Election

    • In this article, we analyze political parties' campaign communication during the 2009 European Parliamentary election in 11 countries (Austria, Bulgaria, Czech Republic, Germany, Hungary, The Netherlands, Poland, Portugal, Spain, Sweden, and the UK). We study which types of issues Euroskeptic fringe and Euroskeptic mainstream parties put on their campaign agendas and the kind and extent of EU opposition they voice. Further, we seek to understand whether Euroskeptic and non-Euroskeptic parties co-orient themselves toward each other within their national party systems with regard to their campaigns. To understand the role of Euroskeptic parties in the 2009 European Parliamentary elections, we draw on a systematic content analysis of parties' posters and televised campaign spots. Our results show that it is Euroskeptic parties at the edges of the political spectrum who discuss polity questions of EU integration and who most openly criticize the union. Principled opposition against the project of EU integration, however, can only be observed in the UK. Finally, we find indicators for co-orientation effects regarding the tone of EU mobilization: In national political environments where Euroskeptic parties strongly criticize the EU, pro-European parties at the same time publicly advance pro-EU positions.

It could be described as one of the worst catastrophes experienced during the post-war period in these countries. This book examines how this dramatic and unexpected event affected public communication patterns and practices in countries like Finland and Sweden. The communicative relations between government actors, the media and citizens always significantly affect the development of crucial democratic values such as trust, accountability and legitimacy. The book covers different topics related to this issue, such as strategic political communication, media coverage, newsroom practices, public opinion and the use of new media in Finland and Sweden after the tsunami disaster.

  • Cancer therapy and risk of congenital malformations in children fathered by men treated for testicular germ-cell cancer : A nationwide register study

    • Background Because of the potential mutagenic effects of chemo- and radiotherapy, there is concern regarding increased risk of congenital malformations (CMs) among children of fathers with cancer. Previous register studies indicate increased CM risk among children conceived after paternal cancer but lack data on oncological treatment. Increased CM risk was recently reported in children born before paternal cancer. This study aims to investigate whether anti-neoplastic treatment for testicular germ-cell cancer (TGCC) implies additional CM risk. Methods and findings In this nationwide register study, all singletons born in Sweden 1994-2014 (n = 2,027,997) were included. Paternal TGCC diagnoses (n = 2,380), anti-neoplastic treatment, and offspring CMs were gathered from the Swedish Norwegian Testicular Cancer Group (SWENOTECA) and the Swedish Medical Birth Register. Children were grouped based on +/- paternal TGCC; treatment regimen: surveillance (n = 1,340), chemotherapy (n = 2,533), or radiotherapy (n = 360); and according to time of conception: pre- (n = 2,770) or post-treatment (n = 1,437). Odds ratios (ORs) for CMs were calculated using logistic regression with adjustment for parental ages, maternal body mass index (BMI), and maternal smoking. Children conceived before a specific treatment acted as reference for children conceived after the same treatment. Among children fathered by men with TGCC (n = 4,207), 184 had a CM. The risk of malformations was higher among children of fathers with TGCC compared with children fathered by men without TGCC (OR 1.28, 95% confidence interval [CI] 1.19-1.38, p = 0.001, 4.4% versus 3.5%). However, no additional risk increase was associated with oncological treatment when comparing post-treatment-to pretreatment-conceived children (chemotherapy, OR = 0.82, 95% CI 0.54-1.25, p = 0.37, 4.1% versus 4.6%; radiotherapy, OR = 1.01, 95% CI 0.25-4.12, p = 0.98, 3.2% versus 3.0%). Study limitations include lack of data on use of cryopreserved or donor sperm and on seminoma patients for the period 1995-2000-both tending to decrease the difference between the groups with TGCC and without TGCC. Furthermore, the power of analyses on chemotherapy intensity and radiotherapy was limited. Conclusions No additional increased risk of CMs was observed in children of men with TGCC treated with radio- or chemotherapy. However, paternal TGCC per se was associated with modestly increased risk for offspring malformations. Clinically, this information can reassure concerned patients.

  • Part-load thermal efficiency enhancement in gas turbine combined cycles by exhaust gas recirculation

    • Gas turbine power plants are popular for offshore power generation due to high power density and their reliability. However, growing usage of renewable energies put gas turbines in a load following backup operation. These power plants suffer part-load efficiency losses when operating at less than full capacity, resulting in higher carbon dioxide (CO2) emission from natural gas combined cycles or higher consumption of carbon-free fuels in decarbonized gas turbines. In this article, a solution is proposed for enhancement of power plant part-load thermal efficiency based on exhaust gas recirculation in the gas turbine cycle. Recirculating exhaust gas into the gas turbine have been studied by several researchers and engineers due to its benefit for carbon-free combustion and carbon capture mechanisms. The proposed operation strategy is evaluated for single-spool and two-spool gas turbines operating jointly with a steam bottoming cycle harvesting the waste heat for further power production. In the suggested strategy, eliminating the necessity to cool down the recirculated gas resulted in less equipment footprint for the power plant which makes it more favorable for offshore applications. An in-house design and simulation tool is developed for evaluating gas turbines with modern gas recirculating systems and a flexibility in operation with carbon-free fuel mixtures. The enhancement in efficiency boost, emission reduction, and fuel consumption is quantified demonstrating the improvements with the proposed solution.

    • Sclerosing epithelioid fibrosarcoma (SEF) and low-grade fibromyxoid sarcoma (LGFMS) are 2 distinct types of sarcoma, with a subset of cases showing overlapping morphologic and immunohistochemical features. LGFMS is characterized by expression of the MUC4 protein, and about 90% of cases display a distinctive FUS-CREB3L2 gene fusion. In addition, SEF is often MUC4 positive, but is genetically less well studied. Fluorescence in situ hybridization (FISH) studies have shown involvement of the FUS gene in the majority of so-called hybrid LGFMS/SEF and in 10% to 25% of sarcomas with pure SEF morphology. In this study, we investigated a series of 10 primary tumors showing pure SEF morphology, 4 cases of LGFMS that at local or distant relapse showed predominant SEF morphology, and 1 primary hybrid LGFMS/SEF. All but 1 case showed diffuse expression for MUC4. Using FISH, reverse transcription polymerase chain reaction, and/or mRNA sequencing in selected cases, we found recurrent EWSR1-CREB3L1 fusion transcripts by reverse transcription polymerase chain reaction in 3/10 pure SEF cases and splits and deletions of the EWSR1 and/or CREB3L1 genes by FISH in 6 additional cases. All 5 cases of LGFMS with progression to SEF morphology or hybrid features had FUS-CREB3L2 fusion transcripts. Our results indicate that EWSR1 and CREB3L1 rearrangements are predominant over FUS and CREB3L2 rearrangements in pure SEF, highlighting that SEF and LGFMS are different tumor types, with different impacts on patient outcome.

    • The current economic crisis is global in scope, thus aff ecting both countries in the North and in the South. As a conse-quence, national European governments are modifying their policies in order to cut down on public spending. This article compares how public service media policies are shaped in this new situation in two countries that represent very con-trasting models of public service media: Spain and Sweden. The story of public service media survival in times of rapid development of media technology and liberal political hegemony is a common theme in contemporary media and communication research. This study adds to this theme, by exploring the conditions for public service media further by a more explicit focus on how newly elected governments approach public service media policies in times of economic crises. The basic aim of the study is to compare how public service media conditions may change in countries with very diff erent public service media models.

    • Fifteen identified C-18 fatty acyl-containing saponin structures from Quillaja saponaria Molina have been investigated by electrospray ionization ion-trap multiple-stage mass spectrometry (ESI-IT-MS(n)) in positive ion mode. Their MS(1)-MS(3) spectra were analyzed and ions corresponding to useful fragments, important for the structural identification of Quillaja saponins, were recognized. A few key fragments could describe the structural variations in the C-3 and the C-28 oligosaccharides of the Quillaja saponins. A flowchart involving a stepwise procedure based on key fragments from the MS(1)-MS(3) spectra of these saponins, together with key fragments from these saponins and 13 previously investigated saponins, was constructed for the identification of structural elements in Quillaja saponins. Peak intensity ratios in MS(3) spectra were found to be correlated to structural features of the investigated saponins and is therefore of value for the identification of regioisomers.     • A method for separation and detection of major and minor components in complex Mixtures has been developed, utilising two-dimensional high-performance liquid chromatography (2D-HPLC) combined with electrospray ionisation ion-trap multiple-stage mass spectrometry (ESI-ITMS(n)). Chromatographic conditions were matched with mass spectrometric detection to maximise the number of components that could be separated. The described procedure has proven useful to discern several hundreds of saponin components when applied to Quillaja saponaria Molina bark extracts. The discrimination of each saponin component relies oil the fact that three coordinates (x,y,z) for each component can be derived from the retention time of the two chromatographic steps (x,y) and the M/Z-values from the multiple-stage mass spectrometry (z(n), n = 1, 2....). Thus an improved graphical representation was obtained by combining retention times from the two-stage separation with +MS(1) (z(1)) and the additional structural information from the second mass stage +MS(2) (z(2), z(3)) corresponding to the main fragment ions. By this approach three-dimensional plots can be made that reveal both the chromatographic and structural properties of a specific mixture which can be useful in fingerprinting of complex mixtures.   • Life in the dark : far-red absorbing cyanobacteria extend photic zones deep into terrestrial caves

    • Chlorophyll (Chl) f and d are the most recently discovered chlorophylls, enabling cyanobacteria to harvest near-infrared radiation (NIR) at 700-780 nm for oxygenic photosynthesis. Little is known about the occurrence of these pigments in terrestrial habitats. Here, we provide first details on spectral photon irradiance within the photic zones of four terrestrial cave systems in concert with a detailed investigation of photopigmentation, light reflectance and microbial community composition. We frequently found Chl f and d along the photic zones of caves characterized by low light enriched in NIR and inhabited by cyanobacteria producing NIR-absorbing pigments. Surprisingly, deeper parts of caves still contained NIR, an effect likely attributable to the reflectance of specific wavelengths by the surface materials of cave walls. We argue that the stratification of microbial communities across the photic zones of cave entrances resembles the light-driven species distributions in forests and aquatic environments.

  • Cross-border dynamic in comparative studies of media systems : Towards a new model

    • In globalized and digitalized economies, how could we compare the role of media in politics in different countries? The growth of transnational social media platforms; the new borderless patterns of media distribution and consumption, and the consequential need for transnational and harmonized digital media policies have caused challenges for scholars in the field of comparative studies (Siebert & Schram 1956; Hallin & Mancini 2004). A problem is that traditional conceptualizations of media systems still dominate (cf. Humprecht et al. 2022; Marques & Vos 2023). Therefore, the purpose is to theorize emerging media systems constituted by combinations of domestic/cross-border institutionalized/de-institutionalized relations. Through a literature review and results from the EU-funded project Mediadelcom, we argue as follows:(1) The power of de-institutionalizing digital communication (Mancini 2020), and its challenging of legacy media organizations, requires that the polarized pluralist/corporatist/liberal classifications of Hallin & Mancini (2004) become more changeable and dynamically combined dimensions within and across nation-states, and updated with theory about global politics and communication (Hellmueller & Berglez 2022). For example, this enables understandings of the, often transnationally interconnected, expansion of polarized politics across countries’ digital public spheres.(2) Nation-states should be grouped along categories of domestic-cross-border conditions and relations (prel. the multipolar media system, global pluralist media system, and de-globalizing media system). Here, different types and levels of institutionalized/de-institutionalized movement across borders thus becomes an important unit of comparison. (3) Thereby we propose a framework for comparative studies of cross-border dynamic (CBD) media systems; their role within political systems, and vice versa.ReferencesHallin, D., Mancini, P.     • The purpose of this paper is to discuss Swedish media developments between 2000 and 2020 in terms of critical junctures. This includes examination of media developments in relation to four defined domains (Legal and Ethical Regulation: Journalism; Media Usage Patterns, and Media User-Related Competencies). In this paper we ask how the Swedish developments within the four domains can be understood in terms of opportunities and risks connected to deliberate communication. In the Swedish case, what seems to be significative is the relative absence of clearly defined country-specific junctures. Mostly, we observe many small, incremental changes and gradual developments of risks.

    • In this report, we present available data about Swedish media development during the period 2000-2020, but also relevant context to understand the production and availability of these data, and the main monitoring actors. To be precise, in accordance with the theoretical framework of the Mediadelcom project, we focus on data about legal and ethical legislation, journalistic production, media usage, and media competencies. The overall conclusion is that, in the Swedish case, there is in most instances no lack of reliable data about media development, which also means that there are good prospects for mapping such development. Instead, the challenge in this context is often how to select data, and a combination of different data, to give an objective overview of the media development in relation to topics such as deliberative communication and deliberative democracy.

    • This article offers a longitudinal perspective on communications during election campaigns from a political-party perspective, where strategic considerations about digital media are compared across time. Our analysis is grounded on the concepts of hybridisation and data-driven campaigning, where digital technology tends to play a central role without replacing all traditional campaign features. Empirically, the study is based on a longitudinal analysis of four election campaigns in Sweden during 2010–2022. The analysis shows that Swedish political parties have gradually integrated digital campaign features in their structure and strategy. The process is not linear, but rather back and forth, as party perceptions of the importance of communication channels vary across time. The results imply a development where all parties, regardless of size and ideology, are increasingly making rational judgments of which combinations of old and new campaign methods and communication channels are most effective.

    • The study explores the perspectives of political parties regarding news media coverage in election campaigns. By analyzing official post-election analyses produced by Swedish political parties from 2010 to 2022, the study offers a novel approach to the study of political party views of news media. The findings largely support the arguments proposed by mediatization literature, highlighting the significance of media in party communication. Parties display eagerness to attract positive media attention while expressing regret over inadequate or negative publicity. More surprisingly, there is a lack of references to media bias in the reports, suggesting that the hostile media effect is not a major concern among Swedish parties. Despite criticisms related to irrelevant reporting and perceived negative coverage, party perceptions towards the media remains predominantly neutral. The study contributes to the understanding of the complex relationship between political parties and news media in the context of election campaigns.

    • This chapter addresses the topic of whether news media in different countries are still able to reach the general public and generate a shared public sphere as a prerequisite of democratic countries. The empirical part of the chapter focuses on the extent to which the different segments of the society use news media like newspapers, radio, television, and social media, comparing the results from 18 countries participating in the 2021 Media for Democracy Monitor (MDM) research project. We conclude that most people in most countries still use the news media regularly, although country-specific gaps exist related to sociodemographic factors like age, gender, and especially education and income. Most conspicuous is an intergenerational gap insofar.

    • The dissertation analyses changes in the role of editors-in-chief in ten leading Norwegian and Swedish media houses – today owned by either Bonnier or Schibsted – in light of the potential tensions between journalistic ideals and market demands. This duality is studied over a period of 30 years, from 1985 to 2015. The most defining changes in the structural framework under which editors-in-chief work are the ongoing technological revolution, the transformation from an analogue into a digital society, and structural, economic changes related to this development. Methodologically, the study builds on data from qualitative in-depth interviews, mainly with 33 past and present editors-in-chief. It also contains a study of how the role of editors-in-chief has been reported and discussed in the magazines of two media branch organisations. The changing role of editors-in-chief is analysed within an institutional perspective. The main empirical results are as follows: (1) Owners and company management have considered the recruitment of editors-in-chief to be highly important throughout the period, and they have used their influence actively. Internal recruitment processes are a standard procedure. Very few of those chosen are women; men recruit other men. The last decade shows a recruitment process becoming more centralized and professionalized. (2) Most editors-in-chief represented in the study have a background in the newsroom. This has traditionally been the main qualification. (3) Regular meetings have structured most of the working hours for editors-in-chief. From an institutional perspective, meetings have played a norm-setting and ritualised role. During the last decade, some of those meetings have included not only journalists but also employees from other departments. (4) Those respondents who were active during the last period investigated perceive the increased speed of work on a daily basis and the more complex editorial role as the main changes and challenges. (5) Many of the respondents are so-called ‘silent’ editors. Due to a lack of time, they do not write much in their own papers. Lately, this has changed to some extent, especially among Swedish editors. This finding is one of the major differences between Norwegian and Swedish editors-in-chief. (6) Editors are still responsible for journalistic content, but demands on the part of commercial management have gradually become more important, and strategic decisions have become more centralized. The metaphor about the need to balance the demands of the ‘Marketplace and Cathedral’ has been replaced by the metaphor ‘We are all in the same boat’. The journalistic institution is under pressure. (7) Despite the immense technological and economic changes in the business and in the structural framework, there is also stability in the role due to the robust nature of journalism as an institution. The role of editor-in-chief is complex, and during the last 30 years, it has become even more so. The structural conditions have affected the role in various ways. While the basic tasks of editors-in-chief remain rooted in editorial work, downsizing and market demands have simultaneously undermined the autonomy and power of editors-in-chief, especially in relation to central media group management.

    • The business model construct has been widely used during the last decade, partly because of its potential to provide a holistic view of how companies do business. A test of how prefabrication could form the basis of a construction firm’s business model can lead to an understanding of the potential for the competitiveness and profitability of industrialized building. The aim is to adapt a general business model construct and use it to empirically identify the most frequently used and the most viable business model. The theoretical perspective is employed to examine how a company does business and which activities and resources are mobilized through the distinction between strategic and operational effectiveness. The multiple case studies include five major Swedish companies that produce prefabricated timber building systems and the analysis is grounded in pattern-finding. The business model construct includes: market position, offering, and operational platform. The result indicates five business model elements: prefabrication mode, role in the building process, end-user segments, system augmentation and complementary resources. Applying this construct to the five case companies revealed that one out of seven models was found to be viable in terms of both ‘market share’ and decision-makers’ opinions. One important conclusion is to take the prefabrication mode as the starting point for business model design and then adapt the other elements to a good fit.

    • Mot bakgrund av samhälleliga utmaningar kopplade till demografi, klimat, sysselsättning och krav på resurseffektivitet kan industriellt träbyggande bidra med följande i ett tidsperspektiv fram till 2025:Bygga upp en kapacitet för industriell träbyggande som kan leverera 50 % av flerbostadshusen som byggs på den svenska marknaden (17 500 lägenheter varav 15 000 producerade inom landet) och samtidigt nå en 30-35%-ig materialandel inom segmentet lokaler.Därigenom skapa 8 000 nya jobb inom prefabricering i fabriksmiljö och bidra till att flytta 6 000 jobb från storstad till landsbygd. Detta kan vara en dellösning på bristen på inhemsk byggarbetskraft – att flytta ut jobben på landet och att ”göra om” snickare till maskinoperatörer. Detta kan även vara en möjlighet för integrering av nyanländ arbetskraft.Utnyttja träets potential för minskad klimatbelastning jämfört med andra byggmaterial. Ett industriellt flerbostadshus har 40 % lägre CO2 utsläpp än ett jämförbart betonghus. Motsvarande siffra för lokaler är minus 35-40% (baserat på sammanfattande och nya analyser av Tyréns).Minska klimatbelastningen med 0,7-0,8 miljoner ton CO2 ekvivalenter genom att substituera från betong till trä – en besparing som utgör knappt 3 % av den totala mängd utsläpp som Sveriges handlande och icke handlande sektorer ska anpassa sig till för år 2030. Om träets kollagring adderas till substitutionseffekten så blir den kalkylmässiga besparingen 2-4 miljoner ton CO2 ekvivalenter.Bygga resurseffektivare med industriellt byggande och framförallt utnyttja den potential till fortsatta effektivitetsförbättringar som ligger i att öka volymerna. Detta sker genom högre nivåer på byggproduktionen i kombination med kraftigt ökade marknadsandelar. Industriellt byggande har bättre kontroll över processer som även inkluderar underentreprenörer och konsulter och kan genom bättre integration och partnerskap verka dämpande på en marknad som kännetecknas av stora svängningar i lägen präglade av överhettning respektive nedgång och stiltje.En avslutande kommentar gäller behovet av kapacitet på den svenska marknaden för bostadsbyggande. Svensk skogs- och träindustri verkar i dagsläget stiga in med stora kapacitetsutbyggnader i ett läge där det inte är särskilt troligt att svensk betongindustri fortsätter att expandera. Vi bedömer att osäkerheten vad gäller klimatfrågan är för stor för att man från betongsidan ska ge sig in på stora investeringar i nuläget. De stora byggbolagen verkar ägarmässigt frikoppla sig från betongbranschen och är därmed ännu mera obundna att gå över till en trästrategi när incitamenten blir tillräckligt stora och tydliga. Detta är en stor möjlighet för industriell träbyggande.

    • The changing market conditions for the Swedish sawmill industry place a focus on a better understanding of the pros and cons of value-added and forward integration strategies. The purpose of this article is to describe and explain recent value-added strategies in the Swedish sawmill industry. The study includes strategies from 13 of the 15 largest sawmill companies for the period between 2002 and 2005, describing a differentiation between value added in primary sawmill production and forward integration into secondary production. It also aims to relate some basic conditions, such as company size, company growth and corporate strategy, to value added and forward integration to profitability. The results show strong positive and significant correlations between forward integration, value added in primary production (somewhat weaker) and unit revenue, and profitability measured as return on investment. There were no strong or significant correlations between size and profitability, playing down the importance of economies of scale (among these already large companies). An interesting result is the much higher profitability of the buying sawmill companies compared with the forest corporations, stressing the importance of both a long-term strategy when investing in value-added activities and ultimately the priorities of ownership.

    • Genome-wide association studies (GWAS) have identified a large number of disease-associated SNPs, but in few cases the functional variant and the gene it controls have been identified. To systematically identify candidate regulatory variants, we sequenced ENCODE cell lines and used public ChIP-seq data to look for transcription factors binding preferentially to one allele. We found 9962 candidate regulatory SNPs, of which 16 % were rare and showed evidence of larger functional effect than common ones. Functionally rare variants may explain divergent GWAS results between populations and are candidates for a partial explanation of the missing heritability. The majority of allele-specific variants (96 %) were specific to a cell type. Furthermore, by examining GWAS loci we found >400 allele-specific candidate SNPs, 141 of which were highly relevant in our cell types. Functionally validated SNPs support identification of an SNP in SYNGR1 which may expose to the risk of rheumatoid arthritis and primary biliary cirrhosis, as well as an SNP in the last intron of COG6 exposing to the risk of psoriasis. We propose that by repeating the ChIP-seq experiments of 20 selected transcription factors in three to ten people, the most common polymorphisms can be interrogated for allele-specific binding. Our strategy may help to remove the current bottleneck in functional annotation of the genome.

  • Public Trust and Journalistic Transparency : An experimental study of disclosure and participatory effects in online news

    • Media matter. Most citizens’ in contemporary democracies get their information about current affairs and politics through the media. Political communication studies have for long time analysed the interplay between media content and journalistic style and political attitudes and public trust in political institutions. This paper adds to this discussion by addressing another dimension: the possible impact of journalistic transparency – offered in online-journalism – on political trust.Methodologically, the study was based on a web-based experiment including 1,320 respondents. The treatment groups comprised the same version of an online news article with additional indicators for disclosure transparency and participatory transparency. The article covered a local political issue and politicians form both ruling and opposition political parties appeared in the text.The results indicate that transparency effects on political trust may be overestimated. This experimental study did not confirm any significant positive correlation between transparency and the public trust towards local politicians appearing in the news.

    • Common variants in 94 loci have been associated with breast cancer including 15 loci with genome-wide significant associations (P<5 x 10(-8)) with oestrogen receptor (ER)-negative breast cancer and BRCA1-associated breast cancer risk. In this study, to identify new ER-negative susceptibility loci, we performed a meta-analysis of 11 genome-wide association studies (GWAS) consisting of 4,939 ER-negative cases and 14,352 controls, combined with 7,333 ER-negative cases and 42,468 controls and 15,252 BRCA1 mutation carriers genotyped on the iCOGS array. We identify four previously unidentified loci including two loci at 13q22 near KLF5, a 2p23.2 locus near WDR43 and a 2q33 locus near PPIL3 that display genome-wide significant associations with ER-negative breast cancer. In addition, 19 known breast cancer risk loci have genome-wide significant associations and 40 had moderate associations (P<0.05) with ER-negative disease. Using functional and eQTL studies we implicate TRMT61B and WDR43 at 2p23.2 and PPIL3 at 2q33 in ER-negative breast cancer aetiology. All ER-negative loci combined account for similar to 11% of familial relative risk for ER-negative disease and may contribute to improved ER-negative and BRCA1 breast cancer risk prediction.

    • In metabolic profiling, multivariate data analysis techniques are used to interpret one-dimensional (1D) 1H NMR data. Multivariate data analysis techniques require that peaks are characterised by the same variables in every spectrum. This location constraint is essential for correct comparison of the intensities of several NMR spectra. However, variations in physicochemical factors can cause the locations of the peaks to shift. The location prerequisite may thus not be met, and so, to solve this problem, alignment methods have been developed. However, current state-of-the-art algorithms for data alignment cannot resolve the inherent problems encountered when analysing NMR data of biological origin, because they are unable to align peaks when the spatial order of the peaks changes—a commonly occurring phenomenon. In this paper a new algorithm is proposed, based on the Hough transform operating on an image representation of the NMR dataset that is capable of correctly aligning peaks when existing methods fail. The proposed algorithm was compared with current state-of-the-art algorithms operating on a selected plasma dataset to demonstrate its potential. A urine dataset was also processed using the algorithm as a further demonstration. The method is capable of successfully aligning the plasma data but further development is needed to address more challenging applications, for example urine data.

    • The increasing disturbances in monocultures around the world are testimony to their instability under global change. Many studies have claimed that temporal stability of productivity increases with species richness, although the ecological fundamentals have mainly been investigated through diversity experiments. To adequately manage forest ecosystems, it is necessary to have a comprehensive understanding of the effect of mixing species on the temporal stability of productivity and the way in which it is influenced by climate conditions across large geographical areas. Here, we used a unique dataset of 261 stands combining pure and two-species mixtures of four relevant tree species over a wide range of climate conditions in Europe to examine the effect of species mixing on the level and temporal stability of productivity. Structural equation modelling was employed to further explore the direct and indirect influence of climate, overyielding, species asynchrony and additive effect (i.e. temporal stability expected from the species growth in monospecific stands) on temporal stability in mixed forests. We showed that by adding only one tree species to monocultures, the level (overyielding: +6%) and stability (temporal stability: +12%) of stand growth increased significantly. We identified the key effect of temperature on destabilizing stand growth, which may be mitigated by mixing species. We further confirmed asynchrony as the main driver of temporal stability in mixed stands, through both the additive effect and species interactions, which modify between-species asynchrony in mixtures in comparison to monocultures. Synthesis and applications. This study highlights the emergent properties associated with mixing two species, which result in resource efficient and temporally stable production systems. We reveal the negative impact of mean temperature on temporal stability of forest productivity and how the stabilizing effect of mixing two species can counterbalance this impact. The overyielding and temporal stability of growth addressed in this paper are essential for ecosystem services closely linked with the level and rhythm of forest growth. Our results underline that mixing two species can be a realistic and effective nature-based climate solution, which could contribute towards meeting EU climate target policies.

    • While the majority of previous research suggests there are positive relationships between digital media use and political participation and knowledge, most studies have relied on cross-sectional surveys and have thus not been able to firmly establish the chain of causality. Also, there is little research investigating use of different forms of digital media and their relative effects on political participation and knowledge. This study examines (a) the effects of digital media use on political participation and knowledge and (b) whether different forms of digital media use affect people differently. Drawing on two representative panel surveys, the study demonstrates that there are only weak effects of digital media use on political learning, but that the use of some digital media forms has appreciable effects on political participation.

  • Building a precision medicine infrastructure at a national level : The Swedish experience

    • Precision medicine has the potential to transform healthcare by moving from one-size-fits-all to personalised treatment and care. This transition has been greatly facilitated through new high-throughput sequencing technologies that can provide the unique molecular profile of each individual patient, along with the rapid development of targeted therapies directed to the Achilles heels of each disease. To implement precision medicine approaches in healthcare, many countries have adopted national strategies and initiated genomic/precision medicine initiatives to provide equal access to all citizens. In other countries, such as Sweden, this has proven more difficult due to regionally organised healthcare. Using a bottom-up approach, key stakeholders from academia, healthcare, industry and patient organisations joined forces and formed Genomic Medicine Sweden (GMS), a national infrastructure for the implementation of precision medicine across the country. To achieve this, Genomic Medicine Centres have been established to provide regionally distributed genomic services, and a national informatics infrastructure has been built to allow secure data handling and sharing. GMS has a broad scope focusing on rare diseases, cancer, pharmacogenomics, infectious diseases and complex diseases, while also providing expertise in informatics, ethical and legal issues, health economy, industry collaboration and education. In this review, we summarise our experience in building a national infrastructure for precision medicine. We also provide key examples how precision medicine already has been successfully implemented within our focus areas. Finally, we bring up challenges and opportunities associated with precision medicine implementation, the importance of international collaboration, as well as the future perspective in the field of precision medicine.
