    • The purpose of this study was to characterize different ethnic groups in Kyrgyzstan regarding cardiovascular disease (CVD) and mental distress, and to investigate the association between CVD and mental distress. The mental distress was measured in terms of sleep disturbance, burnout, and stress.Materials and Methods: A cross-sectional study was carried out among six ethnic groups in Kyrgyzstan, aged 18 years and above. The sample was stratified for age, education, family status, and income. We used the Karolinska Sleep Questionnaire to assess sleep disturbance, the physical and emotional subscale of the Shirom Melamed Burnout Questionnaire to assess burnout, and the 10-item Perceived Stress Scale to assess perceived stress.Results: The distribution of CVD differed significantly between the six ethnic groups, with higher prevalence among East Europeans, and Western Asians and lower among Other minorities and Central Asians. In all ethnic groups in Kyrgyzstan, individuals with CVD had increased odds of sleep disturbance and burnout. There was a significant difference in burnout and stress between persons with and without CVD in Kyrgyz and East European ethnic groups.Conclusion: There was a significant difference in burnout and stress between persons with and without CVD in Kyrgyz and East European ethnic groups. In addition to CVD prevention, mitigating sleep disturbance and preventing burnout in the general population should be aimed at in public health measures.

  • Considerations and consequences of allowing DNA sequence data as types of fungal taxa

    • Nomenclatural type definitions are one of the most important concepts in biological nomenclature. Being physical objects that can be re-studied by other researchers, types permanently link taxonomy (an artificial agreement to classify biological diversity) with nomenclature (an artificial agreement to name biological diversity). Two proposals to amend the International Code of Nomenclature for algae, fungi, and plants (ICN), allowing DNA sequences alone (of any region and extent) to serve as types of taxon names for voucherless fungi (mainly putative taxa from environmental DNA sequences), have been submitted to be voted on at the 11th International Mycological Congress (Puerto Rico, July 2018). We consider various genetic processes affecting the distribution of alleles among taxa and find that alleles may not consistently and uniquely represent the species within which they are contained. Should the proposals be accepted, the meaning of nomenclatural types would change in a fundamental way from physical objects as sources of data to the data themselves. Such changes are conducive to irreproducible science, the potential typification on artefactual data, and massive creation of names with low information content, ultimately causing nomenclatural instability and unnecessary work for future researchers that would stall future explorations of fungal diversity. We conclude that the acceptance of DNA sequences alone as types of names of taxa, under the terms used in the current proposals, is unnecessary and would not solve the problem of naming putative taxa known only from DNA sequences in a scientifically defensible way. As an alternative, we highlight the use of formulas for naming putative taxa (candidate taxa) that do not require any modification of the ICN.

  • Inventering av stora rovdjur i Gävleborgs län 2012-2013

    • I Gävleborgs län finns fasta stammar av Sveriges samtliga stora rovdjur: björn, järv, kungsörn, lodjur och varg. Länsstyrelsen har det regionala ansvaret för rovdjursinventeringar, det vill säga att kartlägga rovdjursförekomsten i länet. I den här rapporten redovisas resultaten från rovdjurs-inventeringar samt länsstyrelsens bedömning av järvförekomst i Gävleborg inventeringssäsongen 2012/2013. Syftet med inventeringarna är att övervaka rovdjursstammarnas storlek, status och utveckling. Inventeringsresultaten utgör ett viktigt underlag i länsstyrelsens arbete med att förvalta rovdjuren utifrån uppsatta nationella och regionala mål. Kunskap om rovdjursstammarnas utveckling kan också vara av betydelse inom övrig viltförvaltning.  Jägares och allmänhetens rapportering av rovdjursobservationer till länsstyrelsen är till stor hjälp i arbetet med att göra så bra inventeringar som möjligt utifrån de resurser som finns. Rapporteringen gör enklast via hemsidorna www.rovobs.se och www.skandobs.se. Länsstyrelsens fältpersonal följer löpande upp inrapporterade observationer. Inventeringsmetoderna i fält efter lo och varg är huvudsakligen snoking och mindre områdesinventeringar utförda av länsstyrelsens personal. Vid lodjursinventeringen 2012/2013 kvalitetssäkrades 21 lodjursföryngringar, det vill säga 21 honor som fött ungar 2012. Antalet lodjur i Gävleborg kan därmed uppskattas till cirka 137 vintern 2012/2013. Två av föryngringarna delades med Uppsala län och en med Dalarnas län.  Gävleborg berördes av fem vargrevir vintern 2012/2013: Haverö, Prästskogen, Kukumäki, Sjösveden och Korså. I två av reviren, Prästskogen och Korså, bekräftades föryngringar 2012. Länsstyrelsen bedömer att föryngring också kan ha skett i Sjösvedenreviret, men det har inte kunnat bekräftas. Utifrån inventeringsresultatet kan länets vargpopulation vintern 2012/2013 uppskattas till 20-30 vargar. Björn inventerades hösten 2012 genom en spillningsinventering genomförd av länsstyrelsen och Jägareförbundet. Det preliminära resultatet visar att björnstammen i Gävleborg och Dalarna uppgår till 700-800 björnar, jämt fördelade över de två länen. Inventeringen visar också att björnarna finns i hela Gävleborg med kärnområden i Ljusdals, Ovanåkers, Bollnäs och Ockelbo kommuner, och att stammen är glesare vid kusten och i de södra länsdelarna. Ingen riktad järvinventering har gjorts i länet de senaste åren, men järvspår dokumenteras av länsstyrelsen under lodjurs- och varginventeringen. Vintern 2012/2013 dokumenterades järv och järvspår framför allt i länets nordliga och nordvästra delar. Länsstyrelsens bedömning är att järven ökar något i dessa områden och det finns indikationer på att stammen sprider sig söderut i länet. Kungsörn inventeras av Kungsörnsgruppen, på uppdrag av länsstyrelsen, genom besök vid kända boplatser under häckningstiden. Tre lyckade häckningar konstaterades i Gävleborg 2012.

    •  Länsstyrelsen ansvarar för att årliga rovdjursinventeringar genomförs i Gävleborgs län. Syftet är attövervaka rovdjursstammarnas storlek, utbredning, genetiska status och hur stammarna utvecklasöver tiden. I den här rapporten redovisas resultaten från inventeringar av lodjur, varg, järv och kungsörn, samtlänsstyrelsens bedömning av förekomsten av björn i Gävleborg under inventeringssäsongen2014/2015. Vid lodjursinventeringen 2014/2015 kvalitetssäkrades 15 familjegrupper, det vill säga 15 honor somfött ungar 2014. Lodjursstammen i Gävleborg kan därmed uppskattas till cirka 83 individer vintern2014/2015. Fyra av familjegrupperna delades med angränsande län. Gävleborg berördes av tio vargrevir vintern 2014/2015: Prästskogen/Haverö, Naggen, Kukumäki,Sjösveden, Tandsjön, Lingbo, Åmot/Ockelbo, Bondskog, Björnås och Glamsen. I sju av revirenbekräftades att föryngring skett våren 2014. Sex av reviren delades med andra län. Det innebär attGävleborg anses ha haft 4,7 familjegrupper vintern 2014/2015, enligt Viltskadecenters fördelning.Vargarna i Korsåreviret, som tidigare delades mellan Gävleborg och Dalarna, har denna säsong vistatsväster om länsgränsen och har inte spårats i Gävleborg. Tandsjö-reviret försvann underinventeringssäsongen genom skyddsjakt i februari 2015. Ingen inventering av björn har genomförts denna säsong. Länsstyrelsen bedömer att björnstammensstorlek och spridning i länet sannolikt överensstämmer med resultatet från 2012 års spillningsinventering. Björnstammen uppskattades då till cirka 380 individer i Gävleborgs län. En riktad järvinventering har för första gången på många år genomförts i Gävleborgs län vintern2014/2015. Med hjälp av fotostationer har bilder på järv erhållits på flera ställen i hela norra delen avGävleborg och en föryngring 2014 har kvalitetssäkrats. Vid inventeringen av kungsörn i Gävleborg 2014 konstaterades totalt 25 revir, varav 8 hade lyckadehäckningar och 3 av dessa var dubbelkullar, det vill säga kullar med två ungar. Storleken på länetsfasta kungsörnspopulation 2014 uppskattas till 50-65 individer. Inventeringarna av lo, järv och varg utfördes av länsstyrelsens personal, medan kungsörninventerades av Kungsörnsgruppen Gävleborg på uppdrag av länsstyrelsen. Allmänhetensrapportering av rovdjursobservationer har varit till stor hjälp i arbetet med att göra så brainventeringar som möjligt utifrån de resurser som finns.

    • Circulating tumor cells (CTC) are shed in peripheral blood at advanced metastatic stages of solid cancers. Surface-marker-based detection of CTC predicts recurrence and survival in colorectal, breast, and prostate cancer. However, scarcity and variation in size, morphology, expression profile, and antigen exposure impairs reliable detection and characterization of CTC. We have developed a noncontact, label-free microfluidic acoustophoresis method to separate prostate cancer cells from white blood cells (WBC) through forces generated by ultrasonic resonances in microfluidic channels. Implementation of cell prealignment in a temperature-stabilized (±0.5 °C) acoustophoresis microchannel dramatically enhanced the discriminatory capacity and enabled the separation of 5 μm microspheres from 7 μm microspheres with 99% purity. Next, we determined the feasibility of employing label-free microfluidic acoustophoresis to discriminate and divert tumor cells from WBCs using erythrocyte-lysed blood from healthy volunteers spiked with tumor cells from three prostate cancer cell-lines (DU145, PC3, LNCaP). For cells fixed with paraformaldehyde, cancer cell recovery ranged from 93.6% to 97.9% with purity ranging from 97.4% to 98.4%. There was no detectable loss of cell viability or cell proliferation subsequent to the exposure of viable tumor cells to acoustophoresis. For nonfixed, viable cells, tumor cell recovery ranged from 72.5% to 93.9% with purity ranging from 79.6% to 99.7%. These data contribute proof-in-principle that label-free microfluidic acoustophoresis can be used to enrich both viable and fixed cancer cells from WBCs with very high recovery and purity.

    • The use of acoustic forces to manipulate particles or cells at the microfluidic scale (i.e. acoustophoresis), enables non-contact, label-free separation based on intrinsic cell properties such as size, density and compressibility. Acoustophoresis holds great promise as a cell separation technique in several research and clinical areas. However, it has been suggested that the force acting upon cells undergoing acoustophoresis may impact cell viability, proliferation or cell function via subtle phenotypic changes. If this were the case, it would suggest that the acoustophoresis method would be a less useful tool for many cell analysis applications as well as for cell therapy.

    • Objective To identify and characterize genetic loci associated with the risk of developing ANCA-associated vasculitides (AAV). Methods Genetic association analyses were performed after Illumina sequencing of 1853 genes and subsequent replication with genotyping of selected single nucleotide polymorphisms in a total cohort of 1110 Scandinavian cases with granulomatosis with polyangiitis or microscopic polyangiitis, and 1589 controls. A novel AAV-associated single nucleotide polymorphism was analysed for allele-specific effects on gene expression using luciferase reporter assay. Results PR3-ANCA(+) AAV was significantly associated with two independent loci in the HLA-DPB1/HLA-DPA1 region [rs1042335, P = 6.3 x 10(-61), odds ratio (OR) 0.10; rs9277341, P = 1.5 x 10(-44), OR 0.22] and with rs28929474 in the SERPINA1 gene (P = 2.7 x 10(-10), OR 2.9). MPO-ANCA(+) AAV was significantly associated with the HLA-DQB1/HLA-DQA2 locus (rs9274619, P = 5.4 x 10(-25), OR 3.7) and with a rare variant in the BACH2 gene (rs78275221, P = 7.9 x 10(-7), OR 3.0), the latter a novel susceptibility locus for MPO-ANCA(+) granulomatosis with polyangiitis/microscopic polyangiitis. The rs78275221-A risk allele reduced luciferase gene expression in endothelial cells, specifically, as compared with the non-risk allele. Conclusion We identified a novel susceptibility locus for MPO-ANCA(+) AAV and propose that the associated variant is of mechanistic importance, exerting a regulatory function on gene expression in specific cell types.

  • CKD : A Call for an Age-Adapted Definition

    • Current criteria for the diagnosis of CKD in adults include persistent signs of kidney damage, such as increased urine albumin-to-creatinine ratio or a GFR below the threshold of 60 ml/min per 1.73 m2 This threshold has important caveats because it does not separate kidney disease from kidney aging, and therefore does not hold for all ages. In an extensive review of the literature, we found that GFR declines with healthy aging without any overt signs of compensation (such as elevated single-nephron GFR) or kidney damage. Older living kidney donors, who are carefully selected based on good health, have a lower predonation GFR compared with younger donors. Furthermore, the results from the large meta-analyses conducted by the CKD Prognosis Consortium and from numerous other studies indicate that the GFR threshold above which the risk of mortality is increased is not consistent across all ages. Among younger persons, mortality is increased at GFR <75 ml/min per 1.73 m2, whereas in elderly people it is increased at levels <45 ml/min per 1.73 m2 Therefore, we suggest that amending the CKD definition to include age-specific thresholds for GFR. The implications of an updated definition are far reaching. Having fewer healthy elderly individuals diagnosed with CKD could help reduce inappropriate care and its associated adverse effects. Global prevalence estimates for CKD would be substantially reduced. Also, using an age-specific threshold for younger persons might lead to earlier identification of CKD onset for such individuals, at a point when progressive kidney damage may still be preventable.

    • BACKGROUND: Connections between mood changes and weather have been described throughout the ages, and in more recent years, there have been reports on a relationship between atmospheric pressure and neurotransmitter levels in cerebrospinal fluid.METHODS: To further investigate this issue under strictly standardized conditions, we have lumbar-punctured 8 healthy males under low (963 hPa) and high (1064 hPa) barometric pressure, using a pressure chamber.RESULTS: Under high pressure, the tyrosine concentrations in the cerebrospinal fluid (CSF) were lower, while the cholecystokinin tetrapeptide (CCK-4) levels were higher. No differences between low and high pressure were found for tryptophan, 5-hydroxyindolacetic acid (5-HIAA), dopamine (DA), and sulphated cholecystokinin octapeptide (CCK-8S). The serum level of CCK-8S was higher under high pressure. On comparing concentration ratios between the second and the first CSF fraction, we found significantly increased ratios for homovanillic acid (HVA) and 4-hydroxy-3-methoxyphenylglycol (HMPG), but a decreased ratio for tyrosine under high pressure. The difference in the concentration ratios of HVA between low and high pressure correlated negatively with age. Intraspinal pressure correlated negatively with tapping time at low pressure.CONCLUSION: Our results are in line with the hypothesis that atmospheric pressure influences CSF levels of monoamine compounds and cholecystokinin peptides.

    • The See Change survey was designed to make z > 1 cosmological measurements by efficiently discovering high-redshift Type Ia supernovae (SNe Ia) and improving cluster mass measurements through weak lensing. This survey observed twelve galaxy clusters with the Hubble Space Telescope (HST) spanning the redshift range z = 1.13-1.75, discovering 57 likely transients and 27 likely SNe Ia at z similar to 0.8-2.3. As in similar previous surveys, this proved to be a highly efficient use of HST for supernova observations; the See Change survey additionally tested the feasibility of maintaining, or further increasing, the efficiency at yet higher redshifts, where we have less detailed information on the expected cluster masses and star formation rates. We find that the resulting number of SNe Ia per orbit is a factor of similar to 8 higher than for a field search, and 45% of our orbits contained an active SN Ia within 22 rest-frame days of peak, with one of the clusters by itself yielding 6 of the SNe Ia. We present the survey design, pipeline, and supernova discoveries. Novel features include fully blinded supernova searches, the first random forest candidate classifier for undersampled IR data (with a 50% detection threshold within 0.05 mag of human searchers), real-time forward-modeling photometry of candidates, and semi-automated photometric classifications and follow-up forecasts. We also describe the spectroscopic follow-up, instrumental in measuring host galaxy redshifts. The cosmology analysis of our sample will be presented in a companion paper.

    • Biocatalytical methods are presented which provide useful building blocks for pheromone synthesis. Examples of the utility of this approach are the preparation of building blocks for the synthesis of stereochemically pure isomers of pine sawfly pheromones and some other pheromones. Enantiom- erically pure ( 98% ee) 2-methyl-1-alkanols 2 were obtained via baker's yeast reduction of suitable α,β-unsaturated aldehydes, and by using lipases from Pseudomonas to effect resolution by transesterification of suitable racemic precursors to 2-methyl-1-alkanols 2 which gave high enantiomeric ratios E > 100. The resolution by esterification mediated by lipase from Candida rugosa of racemic 2-methylalkanoic acids also gave high enantiomeric ratios E> 100 after having improved the reaction conditions by regulating water activity, by choice of the appropriate complimentary substrate alcohol and by adjusting the initial equivalents of the latter present at the start. Also discussed is the separation of diastereomers of diprionol 1, which is naturally occurring in the pine sawfly Neodiprion sertifer, where it is the direct precursor of its pheromone.

    • Estimation of kidney function is often part of daily clinical practice, mostly done by using the endogenous GFR-markers creatinine or cystatin C. A recommendation to use both markers in parallel in 2010 has resulted in new knowledge concerning the pathophysiology of kidney disorders by identification of a new set of kidney disorders, selective glomerular hypofiltration syndromes. These syndromes, connected to strong increases in mortality and morbidity, are characterised by a selective reduction in the glomerular filtration of 5-30 kDa molecules, such as cystatin C, compared to the filtration of small molecules < 1kDa dominating the glomerular filtrate e.g., water, urea, creatinine. At least two types of such disorders, shrunken or elongated pore syndrome, are possible according to the pore model for glomerular filtration. Selective glomerular hypofiltration syndromes are prevalent in investigated populations, and patients with these syndromes often display normal measured GFR or creatinine-based GFR-estimates. The syndromes are characterised by proteomic changes promoting the development of atherosclerosis, indicating antibodies and specific receptor-blocking substances as possible new treatment modalities. Presently, the KDIGO guidelines for diagnosing kidney disorders do not recommend cystatin C as a general marker of kidney function and will therefore not allow the identification of a considerable number of patients with selective glomerular hypofiltration syndromes. Furthermore, as cystatin C is uninfluenced by muscle mass, diet or variations in tubular secretion and cystatin C-based GFR-estimation equations do not require controversial race or sex terms, it is obvious that cystatin C should be a part of future KDIGO guidelines.

    • Background: The hepatitis C virus (HCV) prevalence in Sweden is estimated to be <0.5%, but unclear in pregnant women. The dominating route of transmission is drug use (DU), blood transfusions constituted a risk before 1992. The aim was to examine the anti-HCV prevalence and risk factors for HCV among pregnant women and their partners to evaluate screening strategies.Methods: Pregnant women and partners in Örebro County and in southern Stockholm were offered HCV-screening when visiting an antenatal clinic in 2013-2016, and completed a questionnaire concerning the country of birth, knowledge of HCV-status and HCV risk factors.Results: In Örebro 2,827 pregnant women and 707 partners, and in Stockholm 1,281 pregnant women and 320 partners participated. Anti-HCV was positive in 34 (0.7%) (25 pregnant women) and the associated risk factors were DU (n = 27), partner with HCV (n = 24) and not born in Sweden (n = 8). HCV RNA was positive in 23 (0.4%), 4 previously unknown and 10 who had been lost to follow-up. The most effective risk factor-based screening model for pregnant women included DU, blood transfusions, born in high prevalence country, partner with HCV, resulting in 538 (13%) pregnant women tested with 96% sensitivity, 87% specificity.Conclusions: In this study of expecting parents in two Swedish regions, the anti-HCV prevalence was 0.7% and 0.4% were viraemic, of which about 60% were previously unknown or lost to follow-up. Awaiting more studies, including cost-benefit analysis evaluating universal screening, we recommend this improved risk factor-based screening model to identify HCV-infected individuals who need follow-up and therapy.

  • Mortality and morbidity after groin hernia surgery : the role of nationwide registers in finding and analysing rare outcomes

    • Introduction: Groin hernia surgery is one of the most common surgical procedures world-wide. Although mainly uncomplicated, the large volume of these operations makes it important to consider severe postoperative complications. The Swedish Hernia Register (SHR) started in 1992 and has grown to include more than 95% of all groin hernia operations performed in Sweden empowering it to be merged with other registers in population-based studies. The aim of this thesis is to merge SHR with other nation-wide registers to analyse postoperative mortality, cardiovascular morbidity, surgical hazards, as well as to study the influence of prostatectomy upon the risk for subsequent groin hernia surgery.Methods: SHR was interlinked with the Cause of Death Register to find standardised mortality ratio, the National Prostate Cancer Register to find incidence of groin hernia surgery after prostatectomy compared to a control group and with the National Patient Register to find morbidity within 30 days of groin hernia surgery. In paper II, medical records of deceased patients were retrieved and scrutinised.Results: Elective groin hernia surgery was found to be a low risk procedure even for elderly patients. The mortality risk within 30 days of emergency surgery was raised sevenfold compared to that of the background population. Women had a threefold increased risk of postoperative mortality compared to men. Patients with bowel obstruction, not examined for groin hernia in the emergency room, were subject to more radiological examinations and were operated significantly later than patents with a clinical diagnosis of groin hernia. Compared to men, significantly fewer women were examined for groin hernia in the emergency ward, 61% vs. 78%, (P=0.04). High age, co-morbidity, emergency operation, and regional anaesthesia were risk factors for cardiovascular events. Compared to open anterior mesh repair, all other methods were associated with increased risk of surgical complication, intra-operatively or postoperatively. A threefold increase in groin hernia surgery was seen after radical prostatectomy, conventional as well as minimally invasive.Discussion: Women are significantly overrepresented concerning mortality after groin hernia surgery. This thesis shows the importance of nation-based registers in the analysis of infrequent phenomena in surgical care.

The second purpose is to take a critical look at these opinions. The third is to present a constructive proposal as to how the Bible should be used in Christian ethics.This analysis is carried out using analytical questions. The main questions are as follows: What is biblical ethics? What is the conception of revelation and tradition and their relation to the Bible? How is the interpretation performed? What is the relation between the Bible and other sources of knowledge? What is Christian ethics? This work results in five different models. The critical and constructive purposes are worked out with the assistance of four different criteria.The theologians and ethicists whose ideas are analysed propose five different models. G_ranBexell provides a law- and gospel model with a clear center in biblical and Christian ethics. This center consists of the gospel and the commandment of love. Lisa Cahill provides a feminist model. As one starting point before bible interpretation begins, she sides with women's equal rights. She also contributes to a teleological virtue ethics. Bruce Birch and Larry Rasmussen focus on the role of the Christian community in their multifaceted community model and recommend a multifaceted use of the Bible. In their liberation model Clodovis Boff and George Pixley advocate a preferential option for the poor. Stephen Fowl and Gregory Jones' narrative model propounds the importance of character and develops a virtue ethics.The author develops his own contribution as an agape model. It is called agape model bothbecause he pronounces God's love in Christ as a central theme in the Bible and in Christian ethics, and because he understands the dimensions of the moral life, centered in the commandment of love, as reflexes of this central theme.

Syftet är attövervaka rovdjursstammarnas storlek, utbredning, genetiska status och hur stammarna utvecklasöver tiden. I den här rapporten redovisas resultaten från inventeringar av lodjur, varg och kungsörn, samtlänsstyrelsens bedömning av förekomsten av björn och järv i Gävleborg under inventeringssäsongen2013/2014. Vid lodjursinventeringen 2013/2014 kvalitetssäkrades 15 familjegrupper, det vill säga 15 honor somfött ungar 2013. Lodjursstammen i Gävleborg kan därmed uppskattas till cirka 83 individer vintern2013/2014. Tre av familjegrupperna delades med angränsande län. Gävleborg berördes av åtta vargrevir vintern 2013/2014: Haverö/Prästskogen, Naggen, Kukumäki,Sjösveden, Korså, Lingbo, Åmot/Ockelbo och Glamsen. I tre av reviren – Kukumäki, Sjösveden ochKorså – bekräftades att föryngring skett våren 2013. Utifrån inventeringsresultatet kan länetsvargpopulation vintern 2013/2014 uppskattas till cirka 30 vargar. Ingen inventering av björn har genomförts denna säsong. Länsstyrelsen bedömer att björnstammensstorlek och spridning i länet sannolikt överensstämmer med resultatet från 2012 års spillningsinventering. Björnstammen uppskattades då till cirka 380 individer i Gävleborgs län.Ingen riktad järvinventering har gjorts i länet de senaste åren, men järvspår dokumenteras avlänsstyrelsen under lodjurs- och varginventeringen. Vintern 2013/2014 observerades järv ochjärvspår framför allt i länets nordliga och nordvästra delar. Vid inventeringen av kungsörn i Gävleborg 2013 konstaterades totalt 21 revir, varav 5 hade lyckadehäckningar. Storleken på länets fasta kungsörnspopulation 2013 uppskattas till 50-60 individer. Inventeringarna av lo och varg utfördes av länsstyrelsens personal, medan kungsörn inventerades avKungsörnsgruppen Gävleborg, på uppdrag av länsstyrelsen. Allmänhetens rapportering avrovdjursobservationer har varit till stor hjälp i arbetet med att göra så bra inventeringar som möjligtutifrån de resurser som finns.

Dessa är inte DNA-profilerade men kvalitetsäkrade som stationära vargar enligtinventeringsföreskrifterna. Vid inventeringen av kungsörn i Gävleborg 2015 konstaterades totalt 23 intakta revir med 14häckande par, varav 8 hade lyckade häckningar. Storleken på länets fasta kungsörnspopulation 2015uppskattas till drygt 50 individer. Ingen inventering av björn har genomförts denna säsong. Länsstyrelsen bedömer att björnstammensstorlek i länet minskat något sedan den senaste inventeringen som genomfördes 2012.Björnstammen upp-skattades då till cirka 380 individer i Gävleborgs län. Järvinventeringen blev bristfällig vintern 2015/2016. Huvudsyftet med inventeringen är att fastställaantalet föryngringar som skett inom länet. Järvarna föder sina ungar i februari och honan lämnar sinlya efter någon vecka med ungarna. Snön försvann redan i början på mars och förutsättningarna attspåra en hona tillbaka till lyan försvann med snön. Länsstyrelsen bedömer att det sker mellan 4-8föryngringar inom Gävleborgs län. Spår av järv blir allt vanligare söderut i länet. Länets järvpopulationär som tätast i de norra och nordvästra delarna. Inventeringarna av lo, järv och varg utfördes av länsstyrelsens personal, medan kungsörninventerades av Kungsörnsgruppen Gävleborg på uppdrag av länsstyrelsen.     • I denna rapport redovisas resultaten från länsstyrelsens inventeringar av lodjur ochvarg i Gävleborg under inventeringssäsongen 2011/2012, samt information om förekomstenav björn, järv och kungsörn i länet.Länsstyrelserna har det regionala ansvaret för rovdjursinventeringar i länen. Inventeringarnai Gävleborg genomförs i enlighet med Naturvårdsverkets föreskrifter ochallmänna råd (NFS 2007:10) om inventering av björn, varg, järv, lodjur och kungsörn.Resultaten från lodjurs- och varginventeringen är huvudsakligen baserade påsnöspårning genom snoking och mindre områdesinventeringar. I varginventeringenanvänds också DNA-analys av insamlad spillning. Jägares och allmänhetens rapporteringav rovdjursobservation till länsstyrelsens hemsida rovobs.se har varit till storhjälp i arbetet med att inventera länets rovdjur.Vid inventeringen av lodjur i Gävleborg 2011/2012 konstaterades totalt 19 familjegrupper/föryngringar (det vill säga hona med ungar), varav 1 delas med Uppsla län.Resultat från varginventeringen visar att Gävleborg 2011/2012 berördes av sex vargrevir,varav tre delas med andra län. I tre av länets revir bekräftades föryngringar.Vid slutet av spårsäsongen återfanns den så kallade Galvenhannen i länets norradelar. DNA-analys av spillning visade att han hade bildat par med en tik från norskaKynnareviret. Denna parbildning anses vara genetiskt mycket viktig för den Skandinaviskavargstammen. Parets nya revir har namnet Prästskogen. I vargreviret Korså2 har ett skifte av alfahane skett, vilket upptäcktes genom DNA-analys av spillning.Den nya hannen härstammar från reviret Kynna 2.Länets björnstam har inte inventerats på flera år. Länsstyrelsen bedömer dock attlänet har en fast björnstam med kärnområden i Ljusdals, Ovanåkers och Bollnäskommuner. Under hösten 2012 sker en länstäckande spillningsinventering.I länet har ingen riktad järvinventering utförts på flera år.     • Density functional theory calculations are employed to study the asymmetric transfer hydrogenation of ketones catalyzed by rhodiumarene complexes containing hydroxamic acid-functionalized amino acid ligands. Firstly, the ligandmetal binding is investigated and it is shown that both the N,N and O,O binding modes Are viable. For each of these, the full free energy profile for the transfer hydrogenation is calculated according to the outer-sphere reaction mechanism. Three factors are demonstrated to influence the stereoselectivity of the process, namely the energy difference between the metalligand binding modes, the energy difference between the intermediate hydrogenated catalyst, and the existence of a stabilizing CHp interaction between the Cp* ligand of the catalyst and the phenyl moiety of the substrate. Theoretical reproduction of the selectivity of a slightly modified ligand that is shown experimentally to yield the opposite enantioselectivity corroborates these results. Finally, a technical observation made is that inclusion of dispersion interactions (using the B3LYP-D2 correction or the M06 functional) proved to be very important for reproducing the enantioselectivity.

    • The enantioselectivities of lipases from Pseudomonas cepacia (PFL, Amano PS, etc.) towards a series of primary 2-methyl-substituted alcohols using vinyl acetate as the acyl donor in transesterifications in organic solvents were studied. In terms of enantioselectivity, the best results were found for 3-aryl-2-methylpropan-1-ols with enantiomeric ratios (E-values) over 100 in most cases, whereas other 3-substituted primary 2-methylpropan-1-ols generally displayed lower enantioselectivities: 3-cycloalkyl-2-methylpropan-1-ols (E ≈ 20) and 2-methylalkan-1-ols (E ≈ 10). Moving the aryl group closer or further away from the chiral centre resulted in low enantioselectivities: 2-arylpropan-1-ols (E < 10), 2-methyl-4-(2-thienyl)butan-l-ol (E = 12), 2-methyl-5-(2-thienyl)pentan-l-ol (E=3.2) and 2-methyl-6-(2-thienyl)hexan-l-ol (E=3.8).

  • Social capital and the life cycle model : The transformation of the destination of Åre

    • This article analyzes the development of the Swedish tourism destination of Åre and its transformation from one of many mountain villages into Scandinavia’s leading alpine ski resort. In reference to Butler’s life cycle model, this evolution is analyzed and some historical turning points in Åre’s development identified. The role of social capital is added to the historical analysis, based on the assumption that there is a link between a destination’s development and its ability to reproduce its social capital. The analysis shows that the social capital at the early stage was, to a large extent, built by and around local actors which predominated the village. The transformation to an international ski resort was possible only by the entrance of new actors, increasingly from the national and international arenas. These new actors have largely come to play leading roles in Åre’s development. At the same time, the new actors’ different values and networks have impacted Åre’s social capital.

    • Submicroscopic nanoparticles (NPs) in air have received much attention due to their possible effects on health and wellbeing. Adverse health impacts of air pollution may not only be associated with level of exposure, but also mediated by the perception of the pollution and by beliefs of the exposure being hazardous. The aim of this study was to test a model that describes interrelations between NP pollution, perceived air quality, health risk perception, stress, and sick building syndrome. In the NanoOffice study, the level of NPs was measured and a survey on health risk perception was conducted among 260 employees in twelve office buildings in northern Sweden. Path analyses were performed to test the validity of the model. The data refute the model proposing that the NP exposure level significantly influences stress, chronic diseases, or SBS symptoms. Instead, the perceived exposure influences the perceived risk of NP, and the effect of perceived exposure on SBS and chronic disease is mediated by stress. There was little concern about nanoparticles, despite relatively high levels in some facilities. Perceived pollution and health risk perception may explain a large part of the environmentally induced symptoms and diseases, particularly in relatively low levels of pollution. The research results raise important questions on the physiologically or psychologically mediated health effects of air pollution.

The aim of this study was to test a model that describes interrelations between air pollution (particles < 10 μ m, PM10), perceived pollution, health risk perception, health symptoms and diseases.Methods: A population-based questionnaire study was conducted among 1000 Estonian residents (sample was stratified by age, sex, and geographical location) about health risk perception and coping. The PM10 levels were modelled in 1 × 1 km grids using a Eulerian air quality dispersion model. Respondents were ascribed their annual mean PM10 exposure according to their home address. Path analysis was performed to test the validity of the model.Results: The data refute the model proposing that exposure level significantly influences symptoms and disease. Instead, the perceived exposure influences symptoms and the effect of perceived exposure on disease is mediated by health risk perception. This relationship is more pronounced in large cities compared to smaller towns or rural areas.Conclusions: Perceived pollution and health risk perception, in particular in large cities, play important roles in understanding and predicting environmentally induced symptoms and diseases at relatively low levels of air pollution.

  • Cholecystectomy in Sweden 2000 - 2003 : a nationwide study on procedures, patient characteristics, and mortality

    • Background: Epidemiological data on characteristics of patients undergoing open or laparoscopic cholecystectomy are limited. In this register study we examined characteristics and mortality of patients who underwent cholecystectomy during hospital stay in Sweden 2000 – 2003.Methods: Hospital discharge and death certificate data were linked for all patients undergoing cholecystectomy in Sweden from January 1st 2000 through December 31st 2003. Mortality risk was calculated as standardised mortality ratio (SMR) i.e. observed over expected deaths considering age and gender of the background population.Results: During the four years of the study 43072 patients underwent cholecystectomy for benign biliary disease, 31144 (72%) using a laparoscopic technique and 11928 patients (28%) an open procedure (including conversion from laparoscopy). Patients with open cholecystectomy were older than patients with laparoscopic cholecystectomy (59 vs 49 years, p < 0.001), they were more likely to have been admitted to hospital during the year preceding cholecystectomy, and they had more frequently been admitted acutely for cholecystectomy (57% Vs 21%, p < 0.001). The proportion of women was lower in the open cholecystectomy group compared to the laparoscopic group (57% vs 73%, p < 0.001). Hospital stay was 7.9 (8.9) days, mean (SD), for patients with open cholecystectomy and 2.6 (3.3) days for patients with laparoscopic cholecystectomy, p < 0.001. SMR within 90 days of index admission was 3.89 (3.41–4.41) (mean and 95% CI), for patients with open cholecystectomy and 0.73 (0.52–1.01) for patients with laparoscopic cholecystectomy. During this period biliary disease accounted for one third of all deaths in both groups. From 91 to 365 days after index admission, SMR for patients in the open group was 1.01 (0.87–1.16) and for patients in the laparoscopic group 0.56 (0.44–0.69).Conclusion: Laparoscopic cholecystectomy is performed on patients having a lower mortality risk than the general Swedish population. Patients with open cholecystectomy are more sick than patients with laparoscopic cholecystectomy, and they have a mortality risk within 90 days of admission for cholecystectomy, which is four times that of the general population. Further efforts to reduce surgical trauma in open biliary surgery are motivated.

    • gamma-Butyrobetaine hydroxylase is a 2-oxoglutarate-dependent dioxygenase that catalyzes the hydroxylation of gamma-butyrobetaine to carnitine, the last step in the biosynthesis of carnitine from lysine. The primary structure of the enzyme from Pseudomonas sp. AK1 has been determined. Sequence analysis of the intact protein and of peptides from essentially three different digests established the presence of a peptide chain containing 383 residues, and an N-terminal truncated form of 382 residues. The two chains have molecular masses of 43,321 Da and 43,207 Da, respectively, and are identical except for the presence or absence of an N-terminal asparagine residue; the shorter form starts with an alanine residue. In preparations of the dimeric protein, the two chains occur in an approximate ratio of 1:1. There are nine cysteine residues and 13 histidine residues, i.e. amino acids which have been postulated as ligands for iron binding. In spite of functional similarities, there appears to be no clear sequence similarities with any of the other mammalian 2-oxoglutarate-dependent dioxygenases so far characterized.

  • In Vivo Effects of Mesenchymal Stromal Cells in Two Patients With Severe Acute Respiratory Distress Syndrome

    • Mesenchymal stromal cells (MSCs) have been investigated as a treatment for various inflammatory diseases because of their immunomodulatory and reparative properties. However, many basic questions concerning their mechanisms of action after systemic infusion remain unanswered. We performed a detailed analysis of the immunomodulatory properties and proteomic profile of MSCs systemically administered to two patients with severe refractory acute respiratory distress syndrome (ARDS) on a compassionate use basis and attempted to correlate these with in vivo anti-inflammatory actions. Both patients received 2 x 10(6) cells per kilogram, and each subsequently improved with resolution of respiratory, hemodynamic, and multiorgan failure. In parallel, a decrease was seen in multiple pulmonary and systemic markers of inflammation, including epithelial apoptosis, alveolar-capillary fluid leakage, and proinflammatory cytokines, microRNAs, and chemokines. In vitro studies of the MSCs demonstrated a broad anti-inflammatory capacity, including suppression of T-cell responses and induction of regulatory phenotypes in T cells, monocytes, and neutrophils. Some of these in vitro potency assessments correlated with, and were relevant to, the observed in vivo actions. These experiences highlight both the mechanistic information that can be gained from clinical experience and the value of correlating in vitro potency assessments with clinical effects. The findings also suggest, but do not prove, a beneficial effect of lung protective strategies using adoptively transferred MSCs in ARDS. Appropriate randomized clinical trials are required to further assess any potential clinical efficacy and investigate the effects on in vivo inflammation.     • A scoring system for a standardized composite movement of the shoulder--the Hand in Neck (HIN) manoeuvre--is presented. The EMG activity of the supraspinatus muscle was studied in 5 healthy subjects at different performance levels of this test. It is shown that the supraspinatus muscle is about four times more active during normal performance than at the subnormal levels, which among themselves do not differ. EMG activity was also studied during normal performance of another standardized manoeuvre--the Pour out of a Pot (POP) test. Based on a comparison of the EMG data with clinical data from patients with the subacromial impingement syndrome it is suggested that an abnormal HIN test indicates the presence of a traction responsive pain generator in the supraspinatus tendon. In the same patient group, the combination of a normal HIN test and an abnormal POP test indicates pain generated by compression of subacromial structures.

    • This paper describes the implementation and evaluation of a collision model for CFD spray simulations. The collision model is intended to be used for prediction of fuel sprays from specially designed impinging spray nozzles. The new model is enhanced in terms of collision frequency, post-collisional characteristics and collision induced break-up. The model is evaluated in terms of macro scale properties, such as spray penetration and spray cone angle.

    • Atom probe tomography has been used to the study the distribution of hydrogen and deuterium in the oxide scale of two common zirconium alloys after autoclave testing in H2O and D2O, respectively. Comparison between hydrogen and deuterium in the mass spectra allows for separation of hydrogen as a corrosion product from adsorbed H2 gas from the vacuum chamber. Enrichment of hydrogen and deuterium, as OH+ and OD+, was observed in grain boundaries. The grain boundaries were identified through segregation of iron. This lends experimental support to existing theories for the mechanism of hydrogen pick-up in zirconium alloys.

  • Inventering av stora rovdjur i Gävleborgs län 2017 - 2018

    • Länsstyrelsen ansvarar för att årliga rovdjursinventeringar genomförs i Gävleborgs län. Syftet är att övervaka rovdjursstammarnas storlek, utbredning, genetiska status och hur stammarna utvecklas över tiden. I den här rapporten redovisas resultaten från inventeringar av lodjur, varg, björn och kungsörn samt länsstyrelsens bedömning av förekomsten av järv i Gävleborg under inventeringssäsongen 2017/2018.Vid lodjursinventeringen 2017/2018 kvalitetssäkrades 12,5 länsegna familjegrupper. Lodjursstammen i Gävleborg vintern 2017/2018 uppskattades till cirka 70 individer.Gävleborg berördes vintern 2017/2018 av 15 vargrevir, varav sju delades med andra län. Länet hade fyra länsegna familjegrupper och tre revirmarkerande par.Resultatet från 2017 års kungsörnsinventering i länet är 12 lyckade häckningar och tre misslyckade. Kungsörnsbeståndet uppskattas utgöras av 24 besatta revir.En björnspillningsinventering genomfördes under hösten 2017. För Gävleborgs län analyserades 1325 spillningsprov som beräknas motsvara 508 st individer. Ett antal av dessa individer delas dock med intilliggande län, vilket innebär att Gävleborg totalt sett förvaltar något färre än 508 individer inom länet.Länsstyrelsen bedömer att det årligen sker mellan fyra och sju järvföryngringar inom Gävleborgs län. Populationen är som tätast i länets norra och nordvästra delar.

  • How to Handle Genetic Information : A Comparison of Attitudes among Patients and the General Population

    • Background: So far there are no studies comparing the attitudes of patients with hereditary conditions to the attitudes of the general public on how to handle genetic risk information which mutation carriers refuse to disclose to relevant family members. The purpose of the present study was to investigate whether such patients and members of the general public want to be informed about the existence of hereditary conditions within their family, and under which conditions they want healthcare providers to breach confidentiality. Methods: It was hypothesized that the desire to be informed would be influenced by characteristics of both the disease and the individual. Systematically varying 3 disease characteristics (fatality, penetrance and treatment availability) yielded 8 versions of a questionnaire, which was administered to general population samples in Norway and Sweden (N = 3,207) and to patient samples in both countries (N = 822). Individual differences in uncertainty avoidance, coping style and consideration for future consequences were also assessed. Results and Conclusion: A majority of both patients and the general public want to be informed about the existence of hereditary conditions within their family. However, patients are more positive towards being informed, both with and without the relative's consent, than the general public. The main predictor of the desire to be informed was uncertainty avoidance in both samples.
