  • An operational decision support tool for stump harvest

    • A multi-criteria decision support tool was developed to optimise stump harvesting for energy in Sweden. The decision tool takes account of multiple, sometimes conflicting, criteria relating to stump harvest; energy and climate, economics, biodiversity, and soil and water. Data on harvested stems are used as primary input data in the tool. Such data are routinely collected in harvester computers. The tool effectively deals with mixed sets of data; quantitative harvest data are re-calculated to metric (e.g. stump biomass), and qualitative data (e.g. biodiversity implications) are incorporated. A digital terrain map derived from air-borne laser scanning provides basic data for estimating soil wetness, while digital maps of water courses, key habitats and protected areas, or other sensitive habitats, are used to identify potentially and practically harvestable stumps.In four sub-models, an index from 0 to 10 is calculated for each stump, with 0 representing ‘Not at all suitable’ and 10 ‘Highly suitable for extraction’. Through this, a stump of high value for wood-living species is assigned a low index in the biodiversity sub-model and a large, easily accessible stump is assigned a high index in the economic sub-model. When calculating the net index, the sub-indices can be weighted according to the preferences of the end-user.An energy and climate sub-model incorporates greenhouse gas (GHG) emissions from forest operations and the effect of advancing GHG emissions when stump biomass is incinerated instead of being left to decompose. In the economic sub-model the potential monetary return from each stump is calculated based on estimated revenue from harvested stump biomass and the costs of stump harvesting and forwarding operations (based on cost functions and GIS calculations of transport distances).The biodiversity sub-model considers four types of wood-dependent organisms (lichens, mosses, insects and fungi) in terms of their habitat requirements, vulnerability, sun exposure preferences, locality, etc. A panel of external experts has drawn up a grading scale of stump values for the different taxonomic groups. The proximity to key habitats and exposure to sunlight are derived from a spatial model.Soil and water issues are handled within a sub-model estimating the consequences for long-term soil fertility (nutrient cycling and soil compaction) and water (leaching of plant nutrients and mercury, and particle transport due to soil damage by heavy machinery).The tool offers the end-user possibilities to prioritise and plan for cost-effective stump harvesting, while minimising negative environmental impacts.

  • A multi-criteria decision support model for optimal stump harvesting

    • A multi-criteria decision support model for optimal stump harvesting Sweden was developed. The model quantifies the effect of harvesting each individual stump over a harvesting object in four criteria's: - Biodiversity (Biodiversity value index) - Economy (SEK) - Greenhouse gas emissions (CO2) - Soil and water (Soil and water preservation index) The four criteria's are sometimes in conflict to each other, and uses values that are not directly comparable. The intended use for this model is to contribute with the objective evaluation of all four criteria's in the decision in what stumps to harvest and what stumps to leave in the harvesting object. The model uses individual stump data (e.g. position, tree species and stump biomass) and harvesting object GIS data (roads, elevation map, soil map, terrain map). Primary data on individual stumps comes from the logging system in the stem harvesters: GPS and operator classification. Such data are routinely collected in harvesters. Official map data for the harvesting object are available from the Swedish mapping, cadastral and land registration authority (Lantmäteriet). This includes the topographic map and elevation maps data in 2 m resolution. Also, GIS data are collected in the inspections before harvesting the stems. Through this, a stump of high biodiversity value is assigned a low harvesting index in the biodiversity sub-model and a large, easily accessible stump is assigned a high harvesting index in the economic sub-model. When calculating the total net index, the harvesting index from each criteria has to be weighed together using one coefficient for each criteria. The weighing coefficient for each criteria is chosen according to the preferences of the decision maker. The tool offers the end-user possibilities to prioritise and plan for cost-effective stump harvesting, while minimising negative environmental impacts.

    • To identify genetic loci influencing central obesity and fat distribution, we performed a meta-analysis of 16 genome-wide association studies (GWAS, N = 38,580) informative for adult waist circumference (WC) and waist-hip ratio (WHR). We selected 26 SNPs for follow-up, for which the evidence of association with measures of central adiposity (WC and/or WHR) was strong and disproportionate to that for overall adiposity or height. Follow-up studies in a maximum of 70,689 individuals identified two loci strongly associated with measures of central adiposity; these map near TFAP2B (WC, P = 1.9x10(-11)) and MSRA (WC, P = 8.9x10(-9)). A third locus, near LYPLAL1, was associated with WHR in women only (P = 2.6x10(-8)). The variants near TFAP2B appear to influence central adiposity through an effect on overall obesity/fat-mass, whereas LYPLAL1 displays a strong female-only association with fat distribution. By focusing on anthropometric measures of central obesity and fat distribution, we have identified three loci implicated in the regulation of human adiposity.

  • Nasal administration of a probiotic assemblage in allergic rhinitis: A randomised placebo-controlled crossover trial

    • Background: Topical probiotics have been suggested as a treatment option for allergic rhinitis, as they may skew the immune response towards a beneficial type-1 non-allergic profile. So far observations in man have exclusively involved oral intake. The aim of this study was to examine whether a topical/nasal administration of a probiotic assemblage (PA) affects quality of life, symptoms and signs of allergic rhinitis in a nasal allergen challenge (NAC) model.Methods: In a placebo-controlled and crossover design, 24 patients with seasonal allergic rhinitis were randomised to topical/nasal administration with a PA of Lactobacillus rhamnosus SP1, Lactobacillus paracasei 101/37 and Lactococcus lactis L1A or placebo for 3 weeks. Participants and investigators were blind to treatment allocation. The last week of each treatment period was combined with a NAC series. Efficacy variables were "Mini-Rhinoconjunctivitis Quality of Life Questionnaire" (Mini-RQLQ), "Total Nasal Symptom Score" (TNSS), "Peak Nasal Inspiratory Flow" (PNIF) and "Fractional Exhaled Nitric Oxide" (FeNO). In addition, to assess whether or not the PA produced any pro-inflammatory effect per se, soluble analytes were monitored in nasal lavage fluids. Finally, bacterial cultures, sampled using swabs from the middle nasal meatus, were assessed for the presence of the PA by MALDI-TOF analysis.Results: Administration of the PA did not produce any nasal symptoms (cf. placebo). An innate immune response was discerned within the PA run (cf. baseline), but no change in nasal lavage fluid levels of cytokines/mediators was observed cf. placebo except for IL-17/IL-17A (a minor increase in the PA run). Administration of the PA did neither affect Mini-RQLQ, TNSS, PNIF nor FeNO. No evidence of persistent colonization was observed.Conclusions: Topical/nasal administration of a PA comprising Lactobacillus rhamnosus SP1, Lactobacillus paracasei 101/37 and Lactococcus lactis L1A, while likely evoking a minor innate immune response yet being safe, does not affect quality of life, symptoms or signs of allergic rhinitis.

    • OBJECTIVE: Osteoprotegerin (OPG) is a tumor necrosis factor receptor-related cytokine, initially found to inhibit osteoclastogenesis. In the present study we investigated the effect of OPG treatment on atherosclerosis. METHODS AND RESULTS: Hypercholesterolemic apoe(-/-) mice were treated with recombinant 15 mg/kg OPG or vehicle injections twice a week for 10 consecutive weeks. Mice treated with OPG showed increased amounts of smooth muscle cells and collagen within the atherosclerotic lesions. OPG treatment did not affect atherosclerotic lesion size (8.2% versus 7.6%) or total vessel area but led to a 250% increase in lesion collagen, formation of mature collagen fibers in subendothelial fibrous caps, and upregulated mRNA for lysyl oxidase that promotes collagen crosslinking. In cell culture studies, OPG promoted cell proliferation in rat aortic smooth muscle cells. In contrast, OPG treatment did not affect markers of vascular or systemic inflammation. CONCLUSIONS: OPG treatment promotes smooth muscle accumulation, collagen fiber formation, and development of fibrous caps but does not affect inflammatory properties of atherosclerotic lesions. Its effects may contribute to plaque stabilization.

  • Nonlinear Behaviour of Concrete Buttress Dams under High-Frequency Excitations Taking into Account Topographical Amplifications

    • Concrete buttress dams could potentially be susceptible to high-frequency vibrations, especially in the cross-stream direction, due to their slender design. Previous studies have mainly focused on low-frequency vibrations in stream direction using a simplified foundation model with the massless method, which does not consider topographic amplifications. This paper therefore investigates the nonlinear behaviour of concrete buttress dams subjected to high-frequency excitations, considering cross-stream vibrations. For comparison, the effect of low-frequency excitations is also investigated. The influence of the irregular topography of the foundation surface on the amplification of seismic waves at the foundation surface and thus in the dam is considered by a rigorous method based on the domain-reduction method using the direct finite element method. The sensitivity of the calculated response of the dam to the free-field modelling approach is investigated by comparing the result with analyses using an analytical method based on one-dimensional wave propagation theory and a massless approach. Available deconvolution software is based on the one-dimensional shear wave propagation to transform the earthquake motion from the foundation surface to the corresponding input motion at depth. Here, a new deconvolution method for both shear and pressure wave propagation is developed based on an iterative time-domain procedure using a one-dimensional finite element column. The examples presented showed that topographic amplifications of high-frequency excitations have a significant impact on the response of this type of dam. Cross-stream vibrations reduced the safety of the dam due to the opening of the joints and the increasing stresses. The foundation modelling approach had a significant impact on the calculated response of the dam. The massless method produced unreliable results, especially for high-frequency excitations. The free-field modelling with the analytical method led to unreliable joint openings. It is therefore recommended to use an accurate approach for foundation modelling, especially in cases where nonlinearity is considered.

  • Numerical modelling considerations for analysis of concrete hydraulic structures subjected to high-frequency seismic loads

    • Concrete hydraulic structures are of great importance in today's society. When situated in areas with hard bedrock, these structures may become extra vulnerable to seismic excitations as these here are dominated by high-frequency vibrations which can have disastrous consequences for slender structures. The aim of this thesis was to investigate special considerations that must be made when conducting analyses of such hydraulic structures during high-frequency excitations. Underground and on the ground structures were investigated separately. Underground concrete pipelines and concrete buttress dams were selected for the study because their behaviour when exposed to seismic excitations is dominated by their stiffness. The most effective models and modelling methods for the seismic analyses of such structures were implemented and evaluated. Two-dimensional finite element (FE) models were developed for the dynamic analysis of underground concrete pipelines loaded by seismic waves propagating from bedrock through soil. The interaction between the bedrock and the surrounding soil was investigated with respect to rock geometry and soil properties. The surface of dam foundations is commonly irregular, resulting in nonuniform motions at the dam-foundation interface. The free-field modelling methods for concrete dam foundations were adapted in order to accurately describe the propagation of earthquake vibrations from the source to the ground surface. The implementation of a threedimensional FE model for concrete buttress dams was investigated. Two different methods for free-field modelling are presented, which can be implemented independently of the software used. The seismic loads are applied as effective earthquake forces at non-reflecting boundaries. In the first method, the free-field motions at the non-reflecting boundaries are determined by the so-called domain reduction method using the direct FE calculation. In the second method, the free-field motions are analytically determined based on the onedimensional wave propagation theory. The results are also compared with the massless foundation modelling approach, in which the topographical amplifications are neglected. It was demonstrated that a two-dimensional model can effectively account for pipeline behaviour. The most important aspect of the models is the ability to capture bending deformations, as segmented structures such as pipelines are vulnerable in this respect. Nonuniform bedrock reduces the safety of concrete pipeline, especially because of bending deformations in the pipe and joints. The massless method gave unreliable results for analyses of dams, especially for high-frequency excitations. The analytical method was also unreliable in estimating the non-linear behaviour of the dams. But, a new time domain deconvolution method was developed to transform the earthquake motion from the foundation surface to the corresponding input motion at depth. It wasfound that free-field modelling of foundations using the direct FE method can accurately capture the topographic amplifications of the seismic excitations. It was shown that a three-dimensional model is required for seismic evaluation of concrete buttress dams. The topographic amplification of high-frequency waves at the surface of canyons had a significant effect on the response of this type of dam.

    • Consumers often have a positive attitude to the option of receiving personalized nutrition advice based upon genetic testing, since the prospect of enhancing or maintaining one’s health can be perceived as empowering. Current direct-to-consumer services over the Internet, however, suffer from a questionable level of truthfulness and consumer protection, in addition to an imbalance between far-reaching promises and contrasting disclaimers. Psychological and behavioral studies indicate that consumer acceptance of a new technology is primarily explained by the end user’s rational and emotional interpretation as well as moral beliefs. Results from such studies indicate that personalized nutrition must create true value for the consumer. Also, the freedom to choose is crucial for consumer acceptance. From an ethical point of view, consumer protection is crucial, and caution must be exercised when putting nutrigenomic-based tests and advice services on the market. Current Internet offerings appear to reveal a need to further guaranty legal certainty by ensuring privacy, consumer protection and safety. Personalized nutrition services are on the borderline between nutrition and medicine. Current regulation of this area is incomplete and undergoing development. This situation entails the necessity for carefully assessing and developing existing rules that safeguard fundamental rights and data protection while taking into account the sensitivity of data, the risks posed by each step in their processing, and sufficient guarantees for consumers against potential misuse.

  • Models for analysis of shotcrete on rock exposed to blasting

    • In underground construction and tunnelling, the strive for a more time-efficient construction process naturally focuses on the possibilities of reducing the times of waiting between stages of construction. The ability to project shotcrete (sprayed concrete) on a rock surface at an early stage after blasting is vital to the safety during construction and function of e.g. a tunnel. A complication arises when the need for further blasting affects the hardening of newly applied shotcrete. If concrete, cast or sprayed, is exposed to vibrations at an early age while still in the process of hardening, damage that threatens the function of the hard concrete may occur. There is little, or no, established knowledge on the subject and there are no guidelines for practical use. It is concluded from previous investigations that shotcrete can withstand high particle velocity vibrations without being seriously damaged. Shotcrete without reinforcement can survive vibration levels as high as 0.5−1 m/s while sections with loss of bond and ejected rock will occur for vibration velocities higher than 1 m/s. The performance of young and hardened shotcrete exposed to high magnitudes of vibration is here investigated to identify safe distances and shotcrete ages for underground and tunnelling construction, using numerical analyses and comparison with measurements and observations. The work focuses on finding correlations between numerical results, measurement results and observations obtained during tunnelling. The outcome will be guidelines for practical use. The project involves development of sophisticated dynamic finite element models for which the collected information and data will be used as input, accomplished by using the finite ele­ment program Abaqus. The models were evaluated and refined through comparisons between calculated and measured data. First, existing simple engineering models were compared and evaluated through calculations and comparisons with existing data. The first model tested is a structural dynamic model that consists of masses and spring elements. The second is a model built up with finite beam elements interconnected with springs. The third is a one-dimensional elastic stress wave model. The stress response in the shotcrete closest to the rock when exposed to P-waves striking perpendicularly to the shotcrete-rock interface was simulated. Results from a non-destructive laboratory experiment were also used to provide test data for the models. The experiment studied P-wave propagation along a concrete bar, with proper­ties similar to rock. Cement based mortar with properties that resembles shotcrete was applied on one end of the bar with a hammer impacting the other. The shape of the stress waves travelling towards the shotcrete was registered using accelerometers positioned along the bar. Due to the inhomogeneous nature of the rock, the stress waves from the blasting attenuate on the way from the point of explosion towards the shotcrete on the rock surface. Material damping for the rock mass is therefore accounted for, estimated from previous in-situ measurements. The vibration resistance of the shotcrete-rock support system depends on the material properties of the shotcrete and here were age-dependent properties varied to investigate the behaviour of young shotcrete subjected to blast loading. The numerical simulations require insertion of realistic material data for shotcrete and rock, such as density and modulus of elasticity. The calculated results were in good correspondence with observations and measurements in-situ, and with the previous numerical modelling results. Compared to the engineering models, using a sophisticated finite element program facilitate modelling of more complex geometries and also provide more detailed results. It was demonstrated that wave propagation through rock towards shotcrete can be modelled using two dimensional elastic finite elements in a dynamic analysis. The models must include the properties of the rock and the accuracy of the material parameters used will greatly affect the results. It will be possible to describe the propagation of the waves through the rock mass, from the centre of the explosion to the reflection at the shotcrete-rock interface. It is acceptable to use elastic material formulations until the material strengths are exceeded, i.e. until the strains are outside the elastic range, which thus indicates material failure. The higher complexity of this type of model, compared to the engineering models, will make it possible to model more sophisticated geometries. Examples of preliminary recommendations for practical use are given and it is demonstrated how the developed models and suggested analytical technique can be used to obtain further detailed limit values.

    • Objective. The purpose of this experimental study was to elucidate alterations in fetal energy metabolism in relation to ECG changes during extreme fetal asphyxia, postnatal resuscitation and the immediate post-resuscitatory phase. Study design. Five near-term fetal sheep were subjected to umbilical cord occlusion until cardiac arrest followed by delivery, resuscitation and postnatal pressure-controlled ventilation. Four sheep served as sham controls and were delivered immediately after ligation of the umbilical cord. Fetal ECG was analysed online for changes of the ST segment. Fetal metabolism was monitored by intracerebral and subcutaneous microdialysis catheters. Results. Fetal ECG reacted on cord occlusion with an increase in the T-wave height followed by changes in intracerebral levels of oxidative parameters. Cerebral lactate/pyruvate ratio and glutamate increased to median (range) of 240 (200-744) and 34.0 (22.6-60.5) mmol/l, respectively; both parameters returned to baseline after resuscitation. Cerebral glucose decreased to 0.1 (0.08-0.12) mmol/l after occlusion and increased above baseline upon resuscitation. In subcutaneous tissue as well as blood the increase in lactate occurred with a delay compared to cerebral levels. Conclusion. The fetal ECG changes related to asphyxia preceded the increase in excitotoxicity as determined by increase in cerebral glutamate during asphyxia. Cerebral lactate increase was superior to subcutaneous lactate increase.

  • Durability of shotcrete tunnel linings due to steel fibre corrosion in cracks

    • Steel fibre reinforcement in homogenous, intact, wet-mix shotcrete show an excellent durability against corrosion. The alkaline concrete environment gives good protection but the relatively thin shotcrete layers may crack due to shrinkage and other deformations. Durability requirements today often demands service-life of more than 100 years, which is not realistic with a maintained load-bearing capacity. Special measures must therefore be taken in the design of shotcrete rock support, such as addition of extra amount of fibres if the shotcrete is cracked or increased structural thickness, which is here demonstrated with examples. Due to the complex situation with requirements on service-life, climate impact and cost-efficiency, the design of future shotcrete supports will be optimized based on life cycle and life cycle cost analyses.

    • A laboratory method for investigation of cored samples of steel fibre reinforced sprayed concrete (shotcrete) is described. A pilot study on computed tomography (CT) has been conducted, with focus on how the technique can be used for non-destructive testing where the cores remain intact after scanning and evaluation. The CT method require advanced integrated equipment for X-ray scanning and image detection, together with a computerized visualization system that can reproduce a threedimensional virtual, transparent model of the studied object. The method is well suited to describe orientation and distribution of steel fibres within the concrete. Interfaces between rock-concrete and concrete-concrete, between layers of differentsequences ofspraying, can also be identified. The results from the CT investigations can be presented as qualitative data that in 3D shows locations of steel fibres, aggregates, etc., and also as quantitative data showing relative distributions of cement paste, aggregates, steel fibres and voids, which is here demonstrated by a selection of examples. The method is well suited for practical analysis of sprayed concrete in situ specimens and it is recommended that it is established as a standard method for special inspections and performance evaluation of rock support in tunnels and subspace structures.

  • Infection biology of Chlamydia pneumoniae

    • There are two main human pathogens in the family of Chlamydiaceae. Different serovars of Chlamydia trachomatis cause sexually-transmitted disease and eye infections whereas C. pneumoniae (TWAR) is a common cause of community-acquired respiratory infection. Chlamydia species are obligate, intracellular bacteria sharing a unique developmental cycle that occurs within a protected vacuole termed an inclusion. These microorganisms can be distinguished by two different forms: the infectious, metabolically inert elementary body (EB) and the reproducing non-infectious form, termed the reticulate body (RB). The cycle is terminated when re-differentiation of RBs back to infectious EBs occurs. Chlamydia possesses a type III secretion system (T3SS) essential for delivery of effector proteins into the host for host-cell interactions. This virulence system has been systematically characterized in several mammalian pathogens. Due to lack of a tractable genetic system for Chlamydia species, we have employed chemical genetics as a strategy to investigate molecular aspects of the T3SS. We have identified that the T3S-inhibitors INP0010 and INP0400 block the developmental cycle and interfere with secretion of T3S effector proteins in C. pneumoniae and C. trachomatis, without any cytotoxic effect. We have further shown that INP0010 decreases initiation of transcription in C. pneumoniae during the early mid-developmental cycle as demonstrated by a novel calculation, useful for measurement of transcription initiation in any intracellular pathogen. The mechanism regulating the signal(s) for primary as well as terminal differentiation of RBs has not been defined in Chlamydia. We show using T3S-inhibitors that INP0010 targets the T3SS and thereby arrests RB proliferation as well as RB to EB re-differentiation of C. pneumoniae as where INP0400 targets the T3SS and provokes a bacterial dissociation from the inclusion membrane presumed to mimic the natural occurrence of terminal differentiation. The effect of INP0010 on iron-responsive genes indicates a role for T3S in iron acquisition. Accordingly, our results suggest the possibility that C. pneumoniae acquires iron via the intracellular trafficking pathway of endocytosed transferrin. Moreover, we have for the first time presented data showing generalized bone loss from C. pneumoniae infection in mice. The infection was associated with increased levels of the bone resorptive cytokines IL-6 and IL-1beta. In addition, an increased sub-population of T-cells expressed RANKL during infection. Additionally, C. pneumoniae established an infection in a human osteoblast cell line in vitro with a similar cytokine profile as seen in vivo, supporting a causal linkage. Collectively, these data may indicate a previously unknown pathological role of C. pneumoniae in generalized bone loss.

    • Intracellular parasitism by Chlamydiales is a complex process involving transmission of metabolically inactive particles that differentiate, replicate, and re-differentiate within the host cell. A type three secretion system (T3SS) has been implicated in this process. We have here identified small molecules of a chemical class of acylated hydrazones of salicylaldehydes that specifically blocks the T3SS of Chlamydia. These compounds also affect the developmental cycle showing that the T3SS has a pivotal role in the pathogenesis of Chlamydia. Our results suggest a previously unexplored avenue for development of novel anti-chlamydial drugs.

    • We report broadband UBV and/or BVRCIC CCD photometry for a total of 1857 stars in the thick-disk and halo populations of the Galaxy. The majority of our targets were selected as candidate field horizontal-branch or other A-type stars (FHB/ A, N = 576), or candidate low-metallicity stars (N = 1221), from the HK and Hamburg/ESO objective-prism surveys. Similar data for a small number of additional stars from other samples are also reported. These data are being used for several purposes. In the case of the FHB/ A candidates they are used to accurately separate the lower gravity FHB stars from various higher gravity A-type stars, a subsample that includes the so-called blue metal poor stars, halo and thick-disk blue stragglers, main-sequence A-type dwarfs, and Am and Ap stars. These data are also being used to derive photometric distance estimates to high-velocity hydrogen clouds in the Galaxy and for improved measurements of the mass of the Galaxy. Photometric data for the metal-poor candidates are being used to refine estimates of stellar metallicity for objects with available medium-resolution spectroscopy, to obtain distance estimates for kinematic analyses, and to establish initial estimates of effective temperature for analysis of high-resolution spectroscopy of the stars for which this information now exists.

    • We have studied clusters of Cr on a Au(111) surface by means of a first-principles method that allows for non-collinear magnetic ordering. We find that the Cr atoms possess a large magnetic moment, of order 4 μB/atom, and have essentially antiferromagnetic interatomic exchange interactions. This leads to collinear magnetic arrangements in linear clusters (dimers and trimers) and non-collinear ordering in clusters where antiferromagnetism becomes frustrated, such as in triangular geometries.

  • A Theoretical Study of Magnetism in Nanostructured Materials

    • A first-principles linear scaling real-space method for investigating non-collinear magnetic behaviour of nanostructured materials has been developed. With this method, the magnetic structures of small supported transition metal clusters have been examined. The geometric constraints imposed on the clusters by the underlying surface is found to cause non-collinear behaviour for V, Cr, and Mn clusters on Cu(111). Fe clusters supported on Cu and Ni have been studied and both spin and orbital moments are found to be enhanced for the Fe atoms, which is attributed to the recuced symmetry present at the surface. Atoms in Co clusters have been found to order antiferromagnetically, and some times in a non-collinear fasion, when deposited on a W surface. Small clusters of fcc Fe embedded in Cu have been examined and a new type of ordering, not present in larger fcc Fe systems was found. Several theoretical studies of Fe and Co based nanostructures consisting of multilayers or embedded clusters have been conducted, with the aim of predicting high moment materials for use in data storage applications. In agreement with previous experiments an enhancement of the magnetic moment is found compared to the magnetic moment of bcc Fe. The enhancement has been shown to be caused by increased spin moments for Fe atoms in close proximity with Co atoms, and this enhancement depends on the number of Co neighbours. As a result of these studies, a possible method of increasing the magnetic moment of cluster based materials has been proposed. Fermi surface analysis have been performed both on bulk materials, in order to investigate mechanisms for stabilizing non-collinear magnetic states, and in layered structures where the effect of the Fermi surface on the interlayer exchange coupling has been investigated. In addition to the development of a real-space electronic structure method for non-collinear magnetism, a density matrix purification method has been implemented in the framework of linear muffin-tin orbitals.

    • It is demonstrated that the magnetic interactions can be drastically different for nanosized systems compared to those of bulk or surfaces. Using a real-space formalism we have developed a method to calculate noncollinear magnetization structures and hence exchange interactions. Our results for magnetic Mn clusters supported on a Cu(111) surface show that the magnetic ordering as a rule is noncollinear and cannot always be described by using a simple Heisenberg Hamiltonian. We argue that the use of ab initio calculations allowing for noncollinear coupling between atomic spins constitutes an efficient and reliable way of analyzing nanosized magnets.

    • The magnetic properties of Fe/Co(001) superlattices have been studied using fully-relativistic first-principles theories. The average magnetic moment shows a behavior similar to bulk Fe-Co alloys, i.e., an enhanced magnetic moment for low Co concentrations, as described by the Slater-Pauling curve. The maximum of the magnetization curve, however, is lowered and shifted towards the Fe-rich compositions. The increased average magnetic moment for the Fe-rich superlattices, compared to bulk Fe, is due to an enhancement of the Fe spin moment close to the Fe-Co interface. The orbital moments were found to be of the same size as in bulk. The effect of interface roughness on the magnetic properties was investigated, and it was found that-despite local fluctuations due to the varying coordination-the average magnetic moment is only slightly affected. From a mapping of first-principles interactions onto the screened generalized perturbation method, we calculate the temperatures for when Fe/Co superlattices break up into an alloy configuration. Furthermore, the tetragonal distortion of the superlattice structure was found to only have a minor effect on the magnetic moments. Also, the calculated easy axis of magnetization is in the film plane for all compositions studied. It lies along the [100] direction for Fe-rich superlattices and along the [110] direction for Co-rich compositions. The transition of the easy axis occurs around a Co concentration of 50%.

Different geometries such as triangles, pyramids, and wires are considered and the cluster sizes have been varied between two and ten atoms. The calculations have been performed using a real-space linear muffin-tin orbital method. The Fe clusters are found to order ferromagnetically regardless of the cluster geometry. For Mn and Cr clusters, antiferromagnetic exchange interactions between nearest neighbors are found to cause collinear antiferromagnetic ordering when the geometry allows it. If the antiferromagnetism is frustrated by the cluster geometry, noncollinear ordering is found. A comparison between the calculated structures and ground states obtained from simplified Heisenberg Hamiltonians show that the exchange interaction varies for different atoms in the clusters as a result of the different local structure.

    • The magnetic properties of Co nanostructures and a Co monolayer on W(001) have been studied in the framework of density functional theory. Different geometries such as planar and three-dimensional clusters have been considered, with cluster sizes varying between 2 and 13 atoms. The calculations were performed using the real-space linear muffin-tin orbital method (RS-LMTO-ASA). With respect to the stability of the magnetic state, we predict an antiferromagnetic (AFM) structure for the ground state of the planar Co clusters and a ferromagnetic (FM) state for the three-dimensional clusters. For the three-dimensional clusters, one of the AFM arrangements leads to frustration due to the competing FM and AFM exchange interactions between different atoms in the cluster, and gives rise to a non-collinear state with energy close to that of the FM ground state. The relative role of the Co-Co and Co-W exchange interactions is also investigated.

    • We have calculated spin and orbital moments for Fe clusters of sizes up to 700 atoms embedded as impurities in a bcc Co matrix. The calculations have been carried out using relativistic first-principles real-space density functional theory, and we have made a comparison with earlier experimental studies. For Fe atoms close to theFeCo interface, the spin moments are found to increase while atoms far from the interface exhibit bulklike moments. The Co moments remain essentially unchanged and close to the moment of bulk bcc Co. With increasing cluster size, the average moments of the cluster atoms decrease due to the decreased surface to volume ratio. The orbital moments of both Fe and Co are calculated to be small and they stay almost constant regardless of cluster size. Our results for spin moments agree with experimental data but the calculated orbital moments are slightly underestimated. A simplified model indicates that a compound of close-packed Fe clusters surrounded by Co show higher average total moments compared to bulk and multilayer systems with a similar concentration. This increase seems to disappear when cluster-cluster interactions are taken into account. The general trend is that for a given alloy concentration of FexCo1−x, clustering tends to lower the average magnetic moment compared to that of ordered structures and random alloys.

    • The dynamics of a synthetic antiferromagnet (a metallic trilayer) have been explored and are shown to exhibit ultrafast switching on a time scale of tens of ps. This conclusion is based on first-principles, atomistic spin dynamics simulations. The simulations are performed at finite temperature, as well as at T = 0 K (the macrospin limit), and we observe a marked temperature dependence of the switching phenomenon. It is shown that, to reach very high switching speeds, it is important that the two ferromagnetic components of the synthetic antiferromagnet have oppositely directed external fields to one another. Then a complex collaboration between precession switching of an internal exchange field and the damping switching of the external field occurs, which considerably accelerates the magnetization dynamics. We discuss a possible application of this fast switching as a magnetic random access memory device, which has as a key component intrinsic antiferromagnetic couplings and an applied Oersted field.

    • We present calculations of the exchange interactions and critical temperatures for several diluted magnetic semiconductor systems. It is shown that the exchange interactions are dominated by short-ranged interactions that have a strong directional dependence. Using a combination of first-principles calculations of the exchange interactions together with Monte Carlo simulations of the classical Heisenberg model, in which the positional disorder and spin fluctuations are properly included, the calculated critical temperatures are in good agreement with experimantal observations. It is shown that agreement between theory and experiment, as regards ordering temperatures, is obtained only when the magnetic atoms are randomly positioned in a simulation cell which proves that disorder effects play a very important role. The effect of strong electron-electron interaction has been studied by means of the LSDA+U scheme. We investigate in detail the nature of the anisotropic exchange interactions by means of a Fermi surface analysis.

    • The ICES/HELCOM Working Group on Integrated Assessments of the Baltic Sea (WGIAB) meeting was held in Helsinki (Finland), 18-22 April 2016. The meeting was attended by 26 participants from five countries and chaired by Laura Uusitalo, Fin-land, Saskia Otto, Germany, Martin Lindegren, Denmark, and Lena Bergström, Swe-den. This was the first year of the new three-year Terms of Reference (ToR) for WGIAB. The main working activities in 2016 were to A) develop the trait-based ap-proach of understanding the ecosystem function, and B) explore the social-ecological system, including indicator development, revising the conceptual model, and devel-oping case studies. As a primary outcome of the ToR A, we built on our previous work on integrated ecosystem assessments (IEAs) in the Baltic Sea, but extended it beyond considering changes in abundances of a few dominant species, to accounting for community-wide changes in a number of key traits across multiple trophic levels. These traits represent various ecosystem functions upon which we derive important ecosystem services. By investigating temporal changes in the community weighted mean traits of phyto-plankton, zooplankton, zoobenthos, and fish, we demonstrated whether trait reor-ganizations at the level of entire communities occurred in the Central Baltic Sea as a result of the 1980s regime shift. Using in total 29 traits combined for all groups we found indications of two breakpoints across all four taxonomic groups over the last decades, i.e. one around 1990 and one around 2000. Further work will focus on ex-ploring the nature of the changes in trait composition and on standardizing the num-ber of traits and data types (i.e. binary, continuous or categorical) across taxonomic group.In addition, we collected data on key functional groups and abiotic variables in all main sub-basins of the Baltic Sea, setting the stage for a cross-regional comparison of temporal patterns and trends in lower trophic level in the face of recent develop-ments in climate-related drivers.With reference to Tor B, to explore how social indicators could be used in parallel with biological indicators in an integrated assessment framework, we developed a conceptual model of interrelationships between ecosystem and society. We used the model as a basis for mapping factors to be accounted for in the ecosystem-based management using the Baltic salmon and clupeid species as case studies. The models depict 1) the structure of the foodweb relevant to the target species, 2) the key com-munity level and population traits that contribute to the state of the species, 3) main pressures affecting the foodweb and their effects on the species, 4) key management measures, and 5) benefits that the species can produce for society.To support the development of Ecosystem Overview the group members evaluated the probability of occurrence and the magnitude of the effect of 15 pressures occur-ring in the Baltic Sea. The top five pressures identified were input of nutrients, in-creased temperature, decreased salinity, input of hazardous substances, and input or spread of non-indigenous species.The work will continue intersessionally and the next meeting of WGIAB is planned to be held in Lisbon, Portugal, back-to-back with WGCOMEDA and WGEAWESS.

    • Intresset for systemförvaltning har ökat under senare ar, inte minst mot bakgrund av att stora resurser av de totala ADB-kostnaderna anses åtgå till förvaltning av befintliga system. Syftet med den har studien är att undersöka vad systemförvaltning egentligen innebär. Den här studien kan sagas utgöra en grundläggande verksamhetsanalys av systemförvaltningsarbetet med inriktning på andringshantering och dess problembild. Sju fallstudier har genomförts i svenska organisationer som bedriver systemförvaltning. Utifrån dessa fallstudier har begrepp och kategorier genererats med hjälp av ”grounded theory” - ansats. Rapporten bestar av en inledande de] där frågeställningar, forskningsmetod och teoretiska förutsättningar redovisas. Därefter redovisas det empiriska materialet från fallstudien. Resultatet av studien visar att systemförvaltning idag bedrivs främst ADB-inriktat De verksamhetsinriktade aspekterna tillgodoses inte tillräckligt. I rapporten presenteras därför en är- och en bör-definition av systemförvaltning. Ar-definitionen uttrycker hur systemförvaltning bedrivs idag (på ADB-inriktat sätt). Bor-definitionen uttrycker vad systemförvaltning borde vara utifrån en verksamhetsinriktad syn. Systemförvaltningens aktiviteter har identifierats och presenteras i rapporten. Avgränsning mot närliggande aktiviteter görs genom att en ny livscykelmodell for informationssystemarbete presenteras. Ett antal ansvarsroller som är nödvändiga i samband med systemförvaltning har identifierats och presenteras i rapporten.

    • We have investigated the electronic structure and the incommensurate magnetic configuration of the pressure-induced superconductor CeRhIn5. Noncollinear first-principles calculations were performed in the local-density approximation plus U scheme. The observed magnetic configuration is described accurately in our calculations, especially considering the minute energy scale which is relevant (microelectron volt). The band structure and Fermi surfaces were investigated and nesting was found to be responsible for the complex noncollinear magnetic state of CeRhIn5.

    • BACKGROUND: Cognitive impairment is common following out-of-hospital cardiac arrest (OHCA), but the nature of the impairment is poorly understood. Our objective was to describe cognitive impairment in OHCA survivors, with the hypothesis that OHCA survivors would perform significantly worse on neuropsychological tests of cognition than controls with acute myocardial infarction (MI). Another aim was to investigate the relationship between cognitive performance and the associated factors of emotional problems, fatigue, insomnia, and cardiovascular risk factors following OHCA.METHODS: This was a prospective case-control sub-study of The Targeted Hypothermia versus Targeted Normothermia after Out-of-Hospital Cardiac Arrest (TTM2) trial. Eight of 61 TTM2-sites in Sweden, Denmark, and the United Kingdom included adults with OHCA of presumed cardiac or unknown cause. A matched non-arrest control group with acute MI was recruited. At approximately 7 months post-event, we administered an extensive neuropsychological test battery and questionnaires on anxiety, depression, fatigue, and insomnia, and collected information on the cardiovascular risk factors hypertension and diabetes.RESULTS: Of 184 eligible OHCA survivors, 108 were included, with 92 MI controls enrolled. Amongst OHCA survivors, 29% performed z-score ≤ - 1 (at least borderline-mild impairment) in ≥ 2 cognitive domains, 14% performed z-score ≤ - 2 (major impairment) in ≥ 1 cognitive domain while 54% performed without impairment in any domain. Impairment was most pronounced in episodic memory, executive functions, and processing speed. OHCA survivors performed significantly worse than MI controls in episodic memory (mean difference, MD = - 0.37, 95% confidence intervals [- 0.61, - 0.12]), verbal (MD = - 0.34 [- 0.62, - 0.07]), and visual/constructive functions (MD = - 0.26 [- 0.47, - 0.04]) on linear regressions adjusted for educational attainment and sex. When additionally adjusting for anxiety, depression, fatigue, insomnia, hypertension, and diabetes, executive functions (MD = - 0.44 [- 0.82, - 0.06]) were also worse following OHCA. Diabetes, symptoms of anxiety, depression, and fatigue were significantly associated with worse cognitive performance.CONCLUSIONS: In our study population, cognitive impairment was generally mild following OHCA. OHCA survivors performed worse than MI controls in 3 of 6 domains. These results support current guidelines that a post-OHCA follow-up service should screen for cognitive impairment, emotional problems, and fatigue.TRIAL REGISTRATION: ClinicalTrials.gov, NCT03543371.     • Background: This study is designed to provide detailed knowledge on cognitive impairment after out-of-hospital cardiac arrest (OHCA) and its relation to associated factors, and to validate the neurocognitive screening of the Targeted Hypothermia versus Targeted Normothermia after Out-of-Hospital Cardiac Arrest trial (TTM2-trial), assessing effectiveness of targeted temperature management after OHCA. Methods: This longitudinal multi-center clinical study is a sub-study of the TTM2-trial, in which a comprehensive neuropsychological examination is performed in addition to the main TTM2-trial neurocognitive screening. Approximately 7 and 24 months after OHCA, survivors at selected study sites are invited to a standardized assessment, including performance-based tests of cognition and questionnaires of emotional problems, fatigue, executive function and insomnia. At 1:1 ratio, a matched control group from a cohort of acute myocardial infarction (MI) patients is recruited to perform the same assessment. We aim to include 100 patients per group. Potential differences between the OHCA patients and the MI controls at 7 and 24 months will be analyzed with a linear regression, using composite z-scores per cognitive domain (verbal, visual/constructive, working memory, episodic memory, processing speed, executive functions) as primary outcome measures. Results from OHCA survivors on the main TTM2-trial neurocognitive screening battery will be compared with neuropsychological test results at 7 months, using sensitivity and specificity analyses. Discussion: In this study we collect detailed information on cognitive impairment after OHCA and compare this to a control group of patients with acute MI. The validation of the TTM2 neurocognitive screening battery could justify its inclusion in routine follow-up. Our results may have a potential to impact on the design of future follow-up strategies and interventions after OHCA. Trial registration: ClinicalTrials.gov, NCT03543371. Registered 1 June 2018

    • The folate metabolism enzyme MTHFD2 (methylenetetrahydrofolate dehydrogenase/cyclohydrolase) is consistently overexpressed in cancer but its roles are not fully characterized, and current candidate inhibitors have limited potency for clinical development. In the present study, we demonstrate a role for MTHFD2 in DNA replication and genomic stability in cancer cells, and perform a drug screen to identify potent and selective nanomolar MTHFD2 inhibitors; protein cocrystal structures demonstrated binding to the active site of MTHFD2 and target engagement. MTHFD2 inhibitors reduced replication fork speed and induced replication stress followed by S-phase arrest and apoptosis of acute myeloid leukemia cells in vitro and in vivo, with a therapeutic window spanning four orders of magnitude compared with nontumorigenic cells. Mechanistically, MTHFD2 inhibitors prevented thymidine production leading to misincorporation of uracil into DNA and replication stress. Overall, these results demonstrate a functional link between MTHFD2-dependent cancer metabolism and replication stress that can be exploited therapeutically with this new class of inhibitors. Helleday and colleagues describe a nanomolar MTHFD2 inhibitor that causes replication stress and DNA damage accumulation in cancer cells via thymidine depletion, demonstrating a potential therapeutic strategy in AML tumors in vivo.
