    • Summary: The relationship between self-reported falls and fracture risk was estimated in an international meta-analysis of individual-level data from 46 prospective cohorts. Previous falls were associated with an increased fracture risk in women and men and should be considered as an additional risk factor in the FRAX® algorithm. Introduction: Previous falls are a well-documented risk factor for subsequent fracture but have not yet been incorporated into the FRAX algorithm. The aim of this study was to evaluate, in an international meta-analysis, the association between previous falls and subsequent fracture risk and its relation to sex, age, duration of follow-up, and bone mineral density (BMD). Methods: The resource comprised 906,359 women and men (66.9% female) from 46 prospective cohorts. Previous falls were uniformly defined as any fall occurring during the previous year in 43 cohorts; the remaining three cohorts had a different question construct. The association between previous falls and fracture risk (any clinical fracture, osteoporotic fracture, major osteoporotic fracture, and hip fracture) was examined using an extension of the Poisson regression model in each cohort and each sex, followed by random-effects meta-analyses of the weighted beta coefficients. Results: Falls in the past year were reported in 21.4% of individuals. During a follow-up of 9,102,207 person-years, 87,352 fractures occurred of which 19,509 were hip fractures. A previous fall was associated with a significantly increased risk of any clinical fracture both in women (hazard ratio (HR) 1.42, 95% confidence interval (CI) 1.33–1.51) and men (HR 1.53, 95% CI 1.41–1.67). The HRs were of similar magnitude for osteoporotic, major osteoporotic fracture, and hip fracture. Sex significantly modified the association between previous fall and fracture risk, with predictive values being higher in men than in women (e.g., for major osteoporotic fracture, HR 1.53 (95% CI 1.27–1.84) in men vs. HR 1.32 (95% CI 1.20–1.45) in women, P for interaction = 0.013). The HRs associated with previous falls decreased with age in women and with duration of follow-up in men and women for most fracture outcomes. There was no evidence of an interaction between falls and BMD for fracture risk. Subsequent risk for a major osteoporotic fracture increased with each additional previous fall in women and men. Conclusions: A previous self-reported fall confers an increased risk of fracture that is largely independent of BMD. Previous falls should be considered as an additional risk factor in future iterations of FRAX to improve fracture risk prediction.

    • A large international meta-analysis using primary data from 64 cohorts has quantified the increased risk of fracture associated with a previous history of fracture for future use in FRAX.The aim of this study was to quantify the fracture risk associated with a prior fracture on an international basis and to explore the relationship of this risk with age, sex, time since baseline and bone mineral density (BMD).We studied 665,971 men and 1,438,535 women from 64 cohorts in 32 countries followed for a total of 19.5 million person-years. The effect of a prior history of fracture on the risk of any clinical fracture, any osteoporotic fracture, major osteoporotic fracture, and hip fracture alone was examined using an extended Poisson model in each cohort. Covariates examined were age, sex, BMD, and duration of follow-up. The results of the different studies were merged by using the weighted β-coefficients.A previous fracture history, compared with individuals without a prior fracture, was associated with a significantly increased risk of any clinical fracture (hazard ratio, HR = 1.88; 95% CI = 1.72-2.07). The risk ratio was similar for the outcome of osteoporotic fracture (HR = 1.87; 95% CI = 1.69-2.07), major osteoporotic fracture (HR = 1.83; 95% CI = 1.63-2.06), or for hip fracture (HR = 1.82; 95% CI = 1.62-2.06). There was no significant difference in risk ratio between men and women. Subsequent fracture risk was marginally downward adjusted when account was taken of BMD. Low BMD explained a minority of the risk for any clinical fracture (14%), osteoporotic fracture (17%), and for hip fracture (33%). The risk ratio for all fracture outcomes related to prior fracture decreased significantly with adjustment for age and time since baseline examination.A previous history of fracture confers an increased risk of fracture of substantial importance beyond that explained by BMD. The effect is similar in men and women. Its quantitation on an international basis permits the more accurate use of this risk factor in case finding strategies.

  • Update of the fracture risk prediction tool FRAX : a systematic review of potential cohorts and analysis plan

    • Summary: We describe the collection of cohorts together with the analysis plan for an update of the fracture risk prediction tool FRAX with respect to current and novel risk factors. The resource comprises 2,138,428 participants with a follow-up of approximately 20 million person-years and 116,117 documented incident major osteoporotic fractures.Introduction: The availability of the fracture risk assessment tool FRAX® has substantially enhanced the targeting of treatment to those at high risk of fracture with FRAX now incorporated into more than 100 clinical osteoporosis guidelines worldwide. The aim of this study is to determine whether the current algorithms can be further optimised with respect to current and novel risk factors.Methods: A computerised literature search was performed in PubMed from inception until May 17, 2019, to identify eligible cohorts for updating the FRAX coefficients. Additionally, we searched the abstracts of conference proceedings of the American Society for Bone and Mineral Research, European Calcified Tissue Society and World Congress of Osteoporosis. Prospective cohort studies with data on baseline clinical risk factors and incident fractures were eligible.Results: Of the 836 records retrieved, 53 were selected for full-text assessment after screening on title and abstract. Twelve cohorts were deemed eligible and of these, 4 novel cohorts were identified. These cohorts, together with 60 previously identified cohorts, will provide the resource for constructing an updated version of FRAX comprising 2,138,428 participants with a follow-up of approximately 20 million person-years and 116,117 documented incident major osteoporotic fractures. For each known and candidate risk factor, multivariate hazard functions for hip fracture, major osteoporotic fracture and death will be tested using extended Poisson regression. Sex- and/or ethnicity-specific differences in the weights of the risk factors will be investigated. After meta-analyses of the cohort-specific beta coefficients for each risk factor, models comprising 10-year probability of hip and major osteoporotic fracture, with or without femoral neck bone mineral density, will be computed.Conclusions: These assembled cohorts and described models will provide the framework for an updated FRAX tool enabling enhanced assessment of fracture risk (PROSPERO (CRD42021227266)).

  • Trans-ancestry genome-wide association meta-analysis of prostate cancer identifies new susceptibility loci and informs genetic risk prediction

    • Prostate cancer is a highly heritable disease with large disparities in incidence rates across ancestry populations. We conducted a multiancestry meta-analysis of prostate cancer genome-wide association studies (107,247 cases and 127,006 controls) and identified 86 new genetic risk variants independently associated with prostate cancer risk, bringing the total to 269 known risk variants. The top genetic risk score (GRS) decile was associated with odds ratios that ranged from 5.06 (95% confidence interval (CI), 4.84-5.29) for men of European ancestry to 3.74 (95% CI, 3.36-4.17) for men of African ancestry. Men of African ancestry were estimated to have a mean GRS that was 2.18-times higher (95% CI, 2.14-2.22), and men of East Asian ancestry 0.73-times lower (95% CI, 0.71-0.76), than men of European ancestry. These findings support the role of germline variation contributing to population differences in prostate cancer risk, with the GRS offering an approach for personalized risk prediction. A meta-analysis of genome-wide association studies across different populations highlights new risk loci and provides a genetic risk score that can stratify prostate cancer risk across ancestries.

  • Is there a potential international market for Danish welfare pork? : A consumer survey from Denmark, Sweden, and Germany

    • This cross-country study investigates the potential to improve pig welfare by exploiting consumer demand, domestically and in export markets, for welfare pork produced in indoor production systems. The analysis is based on questionnaire data collected in 2019 focusing on demand for Danish welfare pork both in Denmark and in two nearby export markets, Sweden and Germany. To reduce hypothetical bias, a willingness-to-pay indicator is combined with an indicator of positive interest in buying a fictive Danish welfare labelled pork. We find that the market potential is relatively weak. Our findings indicate that there is some, albeit limited, potential in Denmark and Germany while demand is practically non-existing in Sweden, probably because the pig welfare guaranteed by Swedish legislation is similar to what is provided by the fictive welfare label employed in the study. Hence, consumer demand alone cannot secure enhanced pig welfare. Moreover, we found national differences in the characteristics of consumers who are interested in Danish welfare pork. © 2021 Elsevier Ltd

    • Motion vision provides important cues for many tasks. Flying insects, for example, may pursue small, fast moving targets for mating or feeding purposes, even when these are detected against self-generated optic flow. Since insects are small, with size-constrained eyes and brains, they have evolved to optimize their optical, neural and behavioral target visualization solutions. Indeed, even if evolutionarily distant insects display different pursuit strategies, target neuron physiology is strikingly similar. Furthermore, the coarse spatial resolution of the insect compound eye might actually be beneficial when it comes to detection of moving targets. In conclusion, tiny insects show higher than expected performance in target visualization tasks.

    • Microdialysis enables the chemistry of the extracellular interstitial space to be monitored. Use of this technique in patients with acute brain injury has increased our understanding of the pathophysiology of several acute neurological disorders. In 2004, a consensus document on the clinical application of cerebral microdialysis was published. Since then, there have been significant advances in the clinical use of microdialysis in neurocritical care. The objective of this review is to report on the International Microdialysis Forum held in Cambridge, UK, in April 2014 and to produce a revised and updated consensus statement about its clinical use including technique, data interpretation, relationship with outcome, role in guiding therapy in neurocritical care and research applications.

    • Background: Rheumatoid arthritis is a chronic inflammatory disease with a well-recognised female preponderance. In this post-hoc analysis of the NORD-STAR trial, we aimed to examine sex differences in remission rates with three different biological treatments combined with methotrexate versus active conventional treatment over 24 weeks, in patients with early rheumatoid arthritis. Methods: NORD-STAR was a multicentre, investigator-initiated, assessor-blinded, phase 4, randomised, controlled trial of early rheumatoid arthritis, done in Denmark, Finland, Iceland, Norway, Sweden, and the Netherlands. Newly diagnosed patients, naive to disease-modifying antirheumatic drugs, aged 18 years or older with early rheumatoid arthritis and with a symptom duration less than 24 months were randomly assigned (1:1:1:1) to receive active conventional treatment, certolizumab-pegol, abatacept, or tocilizumab. Sex was reported in case report forms by study physicians or by study nurses. Data on gender were not collected. Remission outcomes were analysed with logistic generalised estimating equations (GEE), using a logit link and exchangeable correlation matrix. The model included treatment, time, sex, and the relevant interactions. For this post-hoc analysis, the co-primary outcomes were differences in Clinical Disease Activity Index (CDAI) remission (CDAI score ≤2·8) between sexes over time and at week 24, assessed with interaction terms (men vs women within each treatment comparison) and using active conventional treatment as the reference. We present adjusted average marginal differences in remission rates (risk differences) with 95% CIs. Findings: Between Dec 14, 2012, and Dec 11, 2018, 812 patients were enrolled and randomly assigned; 217 received active conventional treatment, 203 received certolizumab-pegol, 204 received abatacept, and 188 received tocilizumab. All 812 patients were included in this analysis; 561 (69%) were women and 251 (31%) were men. Observed CDAI remission rates at 24 weeks were numerically higher among men than among women despite comparable disease activity at baseline (55% vs 50% with active conventional treatment, 57% vs 52% with certolizumab-pegol, 65% vs 51% with abatacept, and 61% vs 40% with tocilizumab). In the adjusted analysis, with active conventional treatment as the reference, the only significant difference between men and women was in the tocilizumab group (pinteraction=0·015); men in the tocilizumab group had a higher probability of CDAI remission, on average over time, than did men in the active conventional treatment group (0·12; 95% CI 0·00 to 0·23), whereas women in the tocilizumab group had a lower probability of remission than did women in the active conventional treatment group (–0·05, 95% CI –0·13 to 0·02). Interpretation: Numerically higher remission rates were observed in men than in women in all four treatment groups at week 24, suggesting that this generalised sex difference is not related to the treatment. The difference between men and women was significantly greater with tocilizumab, an interleukin (IL)-6 inhibitor, than with active conventional treatment, suggesting a possible additional sex-based effect specific for IL-6 blockade.     • To identify genetic loci influencing central obesity and fat distribution, we performed a meta-analysis of 16 genome-wide association studies (GWAS, N = 38,580) informative for adult waist circumference (WC) and waist-hip ratio (WHR). We selected 26 SNPs for follow-up, for which the evidence of association with measures of central adiposity (WC and/or WHR) was strong and disproportionate to that for overall adiposity or height. Follow-up studies in a maximum of 70,689 individuals identified two loci strongly associated with measures of central adiposity; these map near TFAP2B (WC, P = 1.9x10(-11)) and MSRA (WC, P = 8.9x10(-9)). A third locus, near LYPLAL1, was associated with WHR in women only (P = 2.6x10(-8)). The variants near TFAP2B appear to influence central adiposity through an effect on overall obesity/fat-mass, whereas LYPLAL1 displays a strong female-only association with fat distribution. By focusing on anthropometric measures of central obesity and fat distribution, we have identified three loci implicated in the regulation of human adiposity.

    • Narrowband radio transmitters like radio amateurs and broadcast radio stations are considered to be a serious problem for high-bitrate data transmission over twisted pairs. Due to its high power level, radio frequency interference (RFI) has the potential of overloading the receiver's analog-to-digital converter (ADC). Once the ADC is in saturation, any countermeasure taken in the digital domain will fail, so the problem has to be faced at least partly in the analog domain. This paper proposes an adaptive, mixed-signal, narrowband interference canceller employing a modified recursive least-squares (RLS) algorithm which is split into an analog and a digital part. The mixed-signal approach enables the circuit to generate an interference-cancelling signal of several MHz while operating the adaptive algorithm at some kHz. The structure is fast enough to prevent the ADC from overloading due to radio amateur interference, thus protecting the data transmission from interruption. Simulation results as well as measurements indicate a practical disturbance rejection potential of about 40-50 dB

    • Recent estimates of ion escape rates from Venus, based on ASPERA-4 data, differ by more than a factor of 4. Whereas the ASPERA-4 instrument provides state-of-the art observations, the limited field of view of the instrument and the strongly limited geographical coverage of the spacecraft orbit means that significant assumptions must be used in the interpretation of the data. We complement previous studies by using a method of average distribution functions to obtain as good statistics as possible while taking the limited field of view into account. We use more than 3 years of data, more than any of the previous studies, and investigate how the choice of a geographical reference frame or a solar wind electric field oriented reference frame affects the results. We find that the choice of reference frame cannot explain the difference between the previously published reports. Our results, based on a larger data set, fall in between the previous studies. Our conclusion is that the difference between previous studies is caused by the large variability of ion outflow at Venus. It matters significantly for the end result which data are selected and which time period is used. The average escape rates were found to be 5.2±1.0×1024 s−1for heavy ions (m/q ≥16) and 14±2.6×1024 s−1for protons. We also discuss the spatial distribution of the planetary ion outflow in the solar wind electric field reference frame.

    • We present an abundance analysis of the very metal-poor, carbon-enhancedstar CS 29497-030. Our results indicate that this unusually hot turnoffstar (Teff = 6650 K, log g = 3.5) has a metallicity [Fe/H] =-2.8, and exhibits large overabundances of carbon ([C/Fe] = +2.38),nitrogen ([N/Fe] = +1.88), and oxygen ([O/Fe] = +1.67). This star alsoexhibits a large enhancement in its neutron-capture elements; thepattern follows that expected to arise from the s-process. Inparticular, the Pb abundance is found to be very high with respect toiron ([Pb/Fe] = +3.5), and also with respect to the second peaks-process elements (e.g., Ba, La, Ce, Nd), which fits into the newlyintroduced classification of lead (Pb) stars. The known spectroscopicbinary status of this star, along with the observed s-process abundancepattern, suggest that it has accreted matter from a companion, whichformerly was an Asymptotic Giant-Branch (AGB) star. In a preliminaryanalysis, we have also identified broad absorption lines of metallicspecies that suggest a large axial rotational velocity for this star,which may be the result of spin-up associated with the accretion ofmaterial from its previous AGB companion. In addition, this star isclearly depleted in the light element Li.   • First stars X. The nature of three unevolved carbon-enhanced metal-poor stars

    • Context. On the order of 20% of the very metal-poor stars in the Galaxy exhibit large carbon enhancements. It is important to establish which astrophysical sites and processes are responsible for the elemental abundance patterns of this early Galactic population. Aims. We seek to understand the nature of the progenitors of three main-sequence turnoff Carbon-Enhanced Metal-Poor (CEMP) stars, CS 31080-095, CS 22958-042, and CS 29528-041, based on a detailed abundance analysis. Methods. From high-resolution VLT/UVES spectra (R similar to 43 000), we determine abundances or upper limits for Li, C, N, O, and other important elements, as well as C-12/C-13 isotopic ratios. Results. All three stars have -3.30 <= [Fe/H]<= -2.85 and moderate to high CNO abundances. CS 22958-042 is one of the most carbon-rich CEMP stars known ([C/Fe] = +3.2), while CS 29528-041 (one of the few N-enhanced metal-poor stars known) is one of the most nitrogen rich ([N/Fe] = +3.0). Oxygen is very high in CS 31080-095 ([O/Fe] = +2.35) and in CS 22958-042 ([O/Fe] = +1.35). All three stars exhibit [Sr/Fe] < 0; Ba is not detected in CS 22958-042 ([Ba/Fe] < -0.53),but it is moderately enhanced ([Ba/Fe] similar to 1) in the other two stars. CS 22958-042 displays one of the largest sodium overabundances yet found in CEMP stars ([Na/Fe] = +2.8). CS 22958-042 has C-12/C-13 = 9, similar to most other CEMP stars without enhanced neutron-capture elements, while C-12/C-13 = 40 in CS 31080-095. CS 31080-095 and CS 29528-041 have A(Li) similar to 1.7, below the Spite Plateau, while Li is not detected in CS 22958-042. Conclusions. CS 22958-042 is a CEMP-no star, but the other two stars are in no known class of CEMP star and thus either constitute a new class or are a link between the CEMP-no and CEMP-s classes, adding complexity to the abundance patterns for CEMP stars. We interpret the abundance patterns in our stars to imply that current models for the presumed AGB binary progenitors lack an extra-mixing process, similar to those apparently operating in RGB stars.

  • A Novel Interception Strategy in a Miniature Robber Fly with Extreme Visual Acuity

    • Our visual system allows us to rapidly identify and intercept a moving object. When this object is far away, we base the trajectory on the target's location relative to an external frame of reference [1]. This process forms the basis for the constant bearing angle (CBA) model, a reactive strategy that ensures interception since the bearing angle, formed between the line joining pursuer and target (called the range vector) and an external reference line, is held constant [2-4]. The CBA model may be a fundamental and widespread strategy, as it is also known to explain the interception trajectories of bats and fish [5, 6]. Here, we show that the aerial attack of the tiny robber fly Holcocephala fusca is consistent with the CBA model. In addition, Holcocephala fusca displays a novel proactive strategy, termed "lock-on" phase, embedded with the later part of the flight. We found the object detection threshold for this species to be 0.13°, enabled by an extremely specialized, forward pointing fovea (∼5 ommatidia wide, interommatidial angle Δφ = 0.28°, photoreceptor acceptance angle Δρ = 0.27°). This study furthers our understanding of the accurate performance that a miniature brain can achieve in highly demanding sensorimotor tasks and suggests the presence of equivalent mechanisms for target interception across a wide range of taxa.

    • Predatory animals have evolved to optimally detect their prey using exquisite sensory systems such as vision, olfaction and hearing. It may not be so surprising that vertebrates, with large central nervous systems, excel at predatory behaviors. More striking is the fact that many tiny insects, with their miniscule brains and scaled down nerve cords, are also ferocious, highly successful predators. For predation, it is important to determine whether a prey is suitable before initiating pursuit. This is paramount since pursuing a prey that is too large to capture, subdue or dispatch will generate a substantial metabolic cost (in the form of muscle output) without any chance of metabolic gain (in the form of food). In addition, during all pursuits, the predator breaks its potential camouflage and thus runs the risk of becoming prey itself. Many insects use their eyes to initially detect and subsequently pursue prey. Dragonflies, which are extremely efficient predators, therefore have huge eyes with relatively high spatial resolution that allow efficient prey size estimation before initiating pursuit. However, much smaller insects, such as killer flies, also visualize and successfully pursue prey. This is an impressive behavior since the small size of the killer fly naturally limits the neural capacity and also the spatial resolution provided by the compound eye. Despite this, we here show that killer flies efficiently pursue natural (Drosophila melanogaster) and artificial (beads) prey. The natural pursuits are initiated at a distance of 7.9 ± 2.9 cm, which we show is too far away to allow for distance estimation using binocular disparities. Moreover, we show that rather than estimating absolute prey size prior to launching the attack, as dragonflies do, killer flies attack with high probability when the ratio of the prey's subtended retinal velocity and retinal size is 0.37. We also show that killer flies will respond to a stimulus of an angular size that is smaller than that of the photoreceptor acceptance angle, and that the predatory response is strongly modulated by the metabolic state. Our data thus provide an exciting example of a loosely designed matched filter to Drosophila, but one which will still generate successful pursuits of other suitable prey.

    • Studies of de novo cytokinin biosynthesis in isopentenyltransferase (ipt)-transformed Arabidopsis thaliana, involving in vivo deuterium labeling and mass spectrometry, showed that the biosynthetic rate of zeatinriboside-5'-monophosphate was around 66-fold higher than that of isopentenyladenosine-5'-monophosphate (iPMP), the proposed primary product of the Agrobacterium ipt. Double tracer analysis, using [(2)H(6)] isopentenyladenosine and deuterium oxide, provided evidence for an alternative, iPMP-independent, biosynthetic pathway for zeatin-type cytokinins, present in both ipt-expressing and wild-type Arabidopsis thaliana. Reduction of the biosynthetic flux in the alternative pathway by use of mevastatin, an inhibitor for 3-hydroxy-3-methylglutaryl CoA reductase, indicated a terpenoid origin for the side-chain precursor of the iPMP independent pathway.

    • Synchronized phasor measurement units (PMUs) provide GPS-time tagged high-sampling rate positive-sequence voltage and current phasors. When placed in high-voltage substations in power networks, PMUs can provide real-time information that is necessary for the development of Smart Transmission Grid software applications for improving power system monitoring, control and protection. The development of these applications, particularly for use within control centers for on-line purposes, is limited by the availability of and access to real-time PMU data and other information. One attractive approach for application development is the use of real-time simulators to which PMUs can be interfaced as hardware-in-the-loop (HIL) devices to harvest PMU data. However, this approach has technical and economical limitations, which can be tackled by the virtualization of PMU devices. This article describes the development of an entirely software-based synchronized phasor measurement unit for use within real-time simulators that will allow the emulation of a large number of real-life PMUs, which in turn can be used for creating new phasor-based applications.

    • We report broadband UBV and/or BVRCIC CCD photometry for a total of 1857 stars in the thick-disk and halo populations of the Galaxy. The majority of our targets were selected as candidate field horizontal-branch or other A-type stars (FHB/ A, N = 576), or candidate low-metallicity stars (N = 1221), from the HK and Hamburg/ESO objective-prism surveys. Similar data for a small number of additional stars from other samples are also reported. These data are being used for several purposes. In the case of the FHB/ A candidates they are used to accurately separate the lower gravity FHB stars from various higher gravity A-type stars, a subsample that includes the so-called blue metal poor stars, halo and thick-disk blue stragglers, main-sequence A-type dwarfs, and Am and Ap stars. These data are also being used to derive photometric distance estimates to high-velocity hydrogen clouds in the Galaxy and for improved measurements of the mass of the Galaxy. Photometric data for the metal-poor candidates are being used to refine estimates of stellar metallicity for objects with available medium-resolution spectroscopy, to obtain distance estimates for kinematic analyses, and to establish initial estimates of effective temperature for analysis of high-resolution spectroscopy of the stars for which this information now exists.

    • Context. The detailed chemical abundances of extremely metal-poor (EMP) stars are key guides to understanding the early chemical evolution of the Galaxy. Most existing data, however, treat giant stars that may have experienced internal mixing later. Aims. We aim to compare the results for giants with new, accurate abundances for all observable elements in 18 EMP turno. stars. Methods. VLT/UVES spectra at R similar to 45 000 and S/N similar to 130 per pixel (lambda lambda 330-1000 nm) are analysed with OSMARCS model atmospheres and the TURBOSPECTRUM code to derive abundances for C, Mg, Si, Ca, Sc, Ti, Cr, Mn, Co, Ni, Zn, Sr, and Ba. Results. For Ca, Ni, Sr, and Ba, we find excellent consistency with our earlier sample of EMP giants, at all metallicities. However, our abundances of C, Sc, Ti, Cr, Mn and Co are similar to 0.2 dex larger than in giants of similar metallicity. Mg and Si abundances are similar to 0.2 dex lower (the giant [Mg/Fe] values are slightly revised), while Zn is again similar to 0.4 dex higher than in giants of similar [Fe/H] (6 stars only). Conclusions. For C, the dwarf/giant discrepancy could possibly have an astrophysical cause, but for the other elements it must arise from shortcomings in the analysis. Approximate computations of granulation (3D) effects yield smaller corrections for giants than for dwarfs, but suggest that this is an unlikely explanation, except perhaps for C, Cr, and Mn. NLTE computations for Na and Al provide consistent abundances between dwarfs and giants, unlike the LTE results, and would be highly desirable for the other discrepant elements as well. Meanwhile, we recommend using the giant abundances as reference data for Galactic chemical evolution models.

    • Malignant cells are known to have increased glucose uptake and accelerated glucose metabolism. Using liquid chromatography and mass spectrometry, we found that treatment of acute lymphoblastic leukemia (ALL) cells with the glucocorticoid (GC) dexamethasone (Dex) resulted in profound inhibition of glycolysis. We thus demonstrate that Dex reduced glucose consumption, glucose utilization and glucose uptake by leukemic cells. Furthermore, Dex treatment decreased the levels of the plasma membrane-associated glucose transporter GLUT1, thus revealing the mechanism for the inhibition of glucose uptake. Inhibition of glucose uptake correlated with induction of cell death in ALL cell lines and in leukemic blasts from ALL patients cultured ex vivo. Addition of di-methyl succinate could partially overcome cell death induced by Dex in RS4;11 cells, thereby further supporting the notion that inhibition of glycolysis contributes to the induction of apoptosis. Finally, Dex killed RS4;11 cells significantly more efficiently when cultured in lower glucose concentrations suggesting that modulation of glucose levels might influence the effectiveness of GC treatment in ALL. In summary, our data show that GC treatment blocks glucose uptake by leukemic cells leading to inhibition of glycolysis and that these effects play an important role in the induction of cell death by these drugs.

    • We report a new determination of [O/Fe, the relative logarithmicabundance of O/Fe with respect to the Sun, for the very metal-poor starBD+23 3130 ([Fe/H=-2.6). The value was derived from the forbidden line[O I at 630 nm and from six weak Fe II lines, with a S/N ratiosubstantially larger than those obtained before, thanks to theefficiency of the VLT-UVES instrument at Paranal. We obtain[O/Fe=0.71+/-0.25, a value 0.36 dex higher than the value obtained fromthe same lines by Fulbright and Kraft [AJ 118 (1999) 527, but 0.46lower than the one derived by Israelian et al. [ApJ 507 (1998) 805 fromthe UV OH bands.

    • In the framework of the ESO Large Programme ``First Stars'', veryhigh-quality spectra of some 70 very metal-poor dwarfs and giants wereobtained with the ESO VLT and UVES spectrograph. These stars are likelyto have descended from the first generation(s) of stars formed after theBig Bang, and their detailed composition provides constraints on issuessuch as the nature of the first supernovae, the efficiency of mixingprocesses in the early Galaxy, the formation and evolution of the haloof the Galaxy, and the possible sources of reionization of the Universe.This paper presents the abundance analysis of an homogeneous sample of35 giants selected from the HK survey of Beers et al. (cite{BPS92},cite{Be99}), emphasizing stars of extremely low metallicity: 30 of our35 stars are in the range -4.1 <[Fe/H]< -2.7, and 22 stars have[Fe/H] < -3.0. Our new VLT/UVES spectra, at a resolving power ofR∼45 000 and with signal-to-noise ratios of 100-200 per pixel overthe wavelength range 330-1000 nm, are greatly superior to those of theclassic studies of McWilliam et al. (cite{MPS95}) and Ryan et al.(cite{RNB96}).The immediate objective of the work is to determine precise,comprehensive, and homogeneous element abundances for this large sampleof the most metal-poor giants presently known. In the analysis wecombine the spectral line modeling code ``Turbospectrum'' with OSMARCSmodel atmospheres, which treat continuum scattering correctly and thusallow proper interpretation of the blue regions of the spectra, wherescattering becomes important relative to continuous absorption (λ< 400 nm). We obtain detailed information on the trends of elementalabundance ratios and the star-to-star scatter around those trends,enabling us to separate the relative contributions of cosmic scatter andobservational/analysis errors.Abundances of 17 elements from C to Zn have been measured in all stars,including K and Zn, which have not previously been detected in starswith [Fe/H] < -3.0. Among the key results, we discuss the oxygenabundance (from the forbidden [OI] line), the different and sometimescomplex trends of the abundance ratios with metallicity, the very tightrelationship between the abundances of certain elements (e.g., Fe andCr), and the high [Zn/Fe] ratio in the most metal-poor stars. Within theerror bars, the trends of the abundance ratios with metallicity areconsistent with those found in earlier literature, but in many cases thescatter around the average trends is much smaller than found in earlierstudies, which were limited to lower-quality spectra. We find that thecosmic scatter in several element ratios may be as low as 0.05 dex.The evolution of the abundance trends and scatter with decliningmetallicity provides strong constraints on the yields of the firstsupernovae and their mixing into the early ISM.     • The ages of the oldest stars in the Galaxy indicate when star formationbegan, and provide a minimum age for the Universe. Radioactive dating ofmeteoritic material and stars relies on comparing the present abundanceratios of radioactive and stable nuclear species to the theoreticallypredicted ratios of their production. The radioisotope 232Th(half-life 14Gyr) has been used to date Galactic stars, but it decays byonly a factor of two over the lifetime of the Universe. 238U(half-life 4.5Gyr) is in principle a more precise age indicator, buteven its strongest spectral line, from singly ionized uranium at awavelength of 385.957nm, has previously not been detected in stars. Herewe report a measurement of this line in the very metal-poor starCS31082-0018, a star which is strongly overabundant in itsheavy elements. The derived uranium abundance, log(U/H) = -13.7 +/- 0.14+/- 0.12 yields an age of 12.5 +/- 3Gyr, though this is still modeldependent. The observation of this cosmochronometer gives the mostdirect age determination of the Galaxy. Also, with improved theoreticaland laboratory data, it will provide a highly precise lower limit to theage of the Universe.

    • Ecosystem exchanges of CO2 and CH4 were studied by chamber techniques in five different vegetation types in a high arctic valley at Zackenberg, NE Greenland. The vegetation types were categorized as Cassiope heath, hummocky fen, continuous fen, grass land and Salix arctica snowbed. Integrated daytime fluxes for the different vegetation types of the valley showed that the fen areas and the grassland, were significant sources of CH4 with a mean efflux of 6.3 mg CH4 m(-2) h(-1) and sinks for CO2, with almost -170 mg CO2 m(-2) hr(-1). The heath and snowbed areas had much lower carbon sequestration rates of about -25 mg CO2 m(-2) hr(-1) and were also sinks for CH4. Methane emissions from the valley dominated in the hummocky fens. Computation of area integrated mean daytime flux values across all vegetation types of the entire valley bottom revealed that it was a sink of CO2 in the order of -96+/-33 mg CO2 m-2 hr-1 and a source of 1.9+/-0.7 m(-2) CH4 m(-2) hr(-1). These values were in accordance with eddy correlation measurements reported elsewhere in this issue and reflect a high-carbon exchange despite the high arctic location. In the fens, where the water table was at or above the soil surface, methane emissions increased with net ecosystem CO2 flux. In places with the water table below the soil surface, such as particularly in the hummocky parts of the fen, oxidation tended to become the dominant controlling factor on methane efflux.

    • Objectives: In patients with axial spondyloarthritis (axSpA) or psoriatic arthritis (PsA) initiating secukinumab, we aimed to assess and compare the proportion of patients achieving 6-, 12- and 24-month patient-reported outcomes (PRO) remission and the 24-month retention rates. Patients and methods: Patients with axSpA or PsA from 16 European registries, who initiated secukinumab in routine care were included. PRO remission rates were defined as pain, fatigue, Patient Global Assessment (PGA) ≤2 (Numeric Rating Scale (NRS) 0–10) and Health Assessment Questionnaire (HAQ) ≤0.5, for both axSpA and PsA, and were calculated as crude values and adjusted for drug adherence (LUNDEX). Comparisons of axSpA and PsA remission rates were performed using logistic regression analyses (unadjusted and adjusted for multiple confounders). Kaplan-Meier plots with log-rank test and Cox regression analyses were conducted to assess and compare secukinumab retention rates. Results: We included 3087 axSpA and 3246 PsA patients initiating secukinumab. Crude pain, fatigue, PGA and HAQ remission rates were higher in axSpA than in PsA patients, whereas LUNDEX-adjusted remission rates were similar. No differences were found between the patient groups after adjustment for confounders. The 24-month retention rates were similar in axSpA vs. PsA in fully adjusted analyses (HR [95 %CI] = 0.92 [0.84–1.02]). Conclusion: In this large European real-world study of axSpA and PsA patients treated with secukinumab, we demonstrate for the first time a comparable effectiveness in PRO remission and treatment retention rates between these two conditions when adjusted for confounders.

    • We report on a dedicated effort to identify and study metal-poor stars strongly enhanced in r-process elements ([r/Fe]>1 dex; hereafter r-IIstars), the Hamburg/ESO R-process Enhanced Star survey (HERES).Moderate-resolution (∼2 Å) follow-up spectroscopy has been obtained for metal-poor giant candidates selected from the Hamburg/ESO objective-prism survey (HES) as well as the HK survey to identify sharp-lined stars with [Fe/H]<-2.5 dex. For several hundred confirmed metal-poor giants brighter than B∼ 16.5 mag (most of them from theHES), ``snapshot'' spectra (R∼ 20 000; S/N ∼ 30 per pixel) are being obtained with VLT/UVES, with the main aim of finding the 2-3% r-II stars expected to be among them. These are studied in detail by means of higher resolution and higher S/N spectra. In this paper we describe a pilot study based on a set of 35 stars, including 23 from the HK survey,eight from the HES, and four comparison stars. We discovered two new r-II stars, CS 29497-004 ([Eu/Fe]=1.64± 0.22) and CS 29491-069([Eu/Fe]=1.08± 0.23). A first abundance analysis of CS 29497-004 yields that its abundances of Ba to Dy are on average enhanced by 1.5 dex with respect to iron and the Sun and match a scaled solar r-process pattern well, while Th is underabundant relative to that pattern by 0.3dex, which we attribute to radioactive decay. That is, CS 29497-004 seems not to belong to the class of r-process enhanced stars displaying an ``actinide boost'', like CS 31082-001 (Hill et al. 2002), or CS30306-132 (Honda et al. 2004b).     • Context: Low-grade inflammation in adipose tissue may contribute to insulin resistance in obesity. However, the roles of individual inflammatory mediators in adipose tissue are poorly understood. Objectives: The objective of this study was to determine which inflammation markers are most overexpressed at the gene level in adipose tissue in human obesity and how this relates to corresponding protein secretion. Design: We examined gene expression profiles in 17 lean and 20 obese subjects. The secretory pattern of relevant corresponding proteins was examined in human sc adipose tissue or isolated fat cells in vitro and in vivo in several obese or lean cohorts. Results: In ranking gene expression, defined pathways associated with obesity and immune and defense responses scored high. Among seven markedly overexpressed chemokines, only monocyte chemoattractant protein 1 (MCP1) was released from adipose tissue and isolated fat cells in vitro. In obesity, the secretion and expression of MCP1 in adipose tissue pieces were more than 6- and 2-fold increased, respectively, but there was no change in circulating MCP1 levels. There was no net release of MCP1, but there was a net release of leptin, in vivo from adipose tissue into the circulation. Conclusions: Obesity is associated with the increased expression of several chemokine genes in adipose tissue. However, only MCP1 is secreted into the extracellular space, where it primarily acts as a local factor, because little or no spillover into the circulation occurs. MCP1 influences the function of adipocytes, is a recruitment factor for macrophages, and may be a crucial link among chemokines between adipose tissue inflammation and insulin resistance.

    • Incompressible Navier-Stokes solvers based on the projection method often require an expensive numerical solution of a Poisson equation for a pressure-like variable. This often involves linear system solvers based on iterative and multigrid methods which may limit the ability to scale to large numbers of processors. The artificial compressibility method (ACM) [6] introduces a time derivative of the pressure into the incompressible form of the continuity equation creating a coupled closed hyperbolic system that does not require a Poisson equation solution and allows for explicit time-marching and localized stencil numerical methods. Such a scheme should theoretically scale well on large numbers of CPUs, GPU'€™s, or hybrid CPU-GPU architectures. The original ACM was only valid for steady flows and dual-time stepping was often used for time-accurate simulations. Recently, Clausen [7] has proposed the entropically damped artificial compressibility (EDAC) method which is applicable to both steady and unsteady flows without the need for dual-time stepping. The EDAC scheme was successfully tested with both a finite-difference MacCormack'€™s method for the two-dimensional lid driven cavity and periodic double shear layer problem and a finite-element method for flow over a square cylinder, with scaling studies on the latter to large numbers of processors. In this study, we discretize the EDAC formulation with a new optimized high-order centered finite-difference scheme and an explicit fourth-order Runge-€“Kutta method. This is combined with an immersed boundary method to efficiently treat complex geometries and a new robust outflow boundary condition to enable higher Reynolds number simulations on truncated domains. Validation studies for the Taylor-€“Green Vortex problem and the lid driven cavity problem in both 2D and 3D are presented. An eddy viscosity subgrid-scale model is used to enable large eddy simulations for the 3D cases. Finally, an application to flow over a sphere is presented to highlight the boundary condition and performance comparisons to a traditional incompressible Navier-€“Stokes solver is shown for the 3D lid driven cavity. Overall, the combined EDAC formulation and discretization is shown to be both effective and affordable.

    • It has repeatedly been shown that consumers consider the higher price of organic produce to be a barrier to buying organic food products. The present paper therefore asks whether organic consumption affects consumers’ actual total food expenditures. Food purchase data for a panel of Danish households during 2006–2014 were used to compare the total food expenditures of different organic household segments. A fixed effects model and a cross-section model both indicated that although food expenditures rose slightly with organic consumption, the increase was considerably less than had been suggested by organic price premiums at product level. © 2020 Taylor & Francis Group, LLC.

    • CS 22949-037 is one of the most metal-poor giants known ([Fe/H]~-4.0),and it exhibits large overabundances of carbon and nitrogen (Norris etal.). Using VLT-UVES spectra of unprecedented quality, regardingresolution and S/N ratio, covering a wide wavelength range (from lambda= 350 to 900 nm), we have determined abundances for 21 elements in thisstar over a wide range of atomic mass. The major new discovery is anexceptionally large oxygen enhancement, [O/Fe] = 1.97+/-0.1, as measuredfrom the [O I] line at 630.0 nm. We find an enhancement of [N/Fe] of2.56+/- 0.2, and a milder one of [C/Fe] = 1.17+/-0.1, similar to thosealready reported in the literature. This implies Zstar =0.01Zsun. We also find carbon isotopic ratios12C/13C =4+/-2.0 and 13C/14N=0.03 +0.035-0.015, close to the equilibrium valueof the CN cycle. Lithium is not detected. Na is strongly enhanced([Na/Fe] = +2.1 +/- 0.2), while S and K are not detected. Thesilicon-burning elements Cr and Mn are underabundant, while Co and Znare overabundant ([Zn/Fe]=+0.7). Zn is measured for the first time insuch an extremely metal-poor star. The abundances of the neutron-captureelements Sr, Y, and Ba are strongly decreasing with the atomic number ofthe element: [Sr/Fe] ~ +0.3, [Y/Fe] ~ -0.1, and [Ba/Fe] ~ -0.6. Amongpossible progenitors of CS 22949-037, we discuss the pair-instabilitysupernovae. Such very massive objects indeed produce large amounts ofoxygen, and have been found to be possible sources of primary nitrogen.However, the predicted odd/even effect is too large, and the predictedZn abundance much too low. Other scenarios are also discussed. Inparticular, the yields of a recent model (Z35Z) from Heger and Woosleyare shown to be in fair agreement with the observations. The onlydiscrepant prediction is the very low abundance of nitrogen, possiblycurable by taking into account other effects such as rotationallyinduced mixing. Alternatively, the absence of lithium in our star, andthe values of the isotopic ratios 12C/13C and13C/14N close to the equilibrium value of the CNcycle, suggest that the CNO abundances now observed might have beenaltered by nuclear processing in the star itself. A 30-40Msun supernova, with fallback, seems the most likelyprogenitor for CS 22949-037.
