    • We present energy-resolved cross-correlation measurements of extreme ultraviolet (XUV) pulses generated as high harmonics of femtosecond pulses from a 1 kHz titanium-sapphire laser. The harmonic pulses are probed by a fraction of the fundamental laser pulse at 800 nm, in a noncollinear geometry, allowing us to vary independently the parameters of the harmonic pump and near-infrared probe pulses. We measure the so-called "sidebands" in the photoelectron spectrum of argon corresponding to the absorption of a harmonic photon plus or minus one probe photon. Spectrally resolving the cross-correlation signal allows us to characterize the time-dependent frequency of the XUV pulse.

  • Time-frequency characterization of femtosecond extreme ultraviolet pulses.

    • We present energy-resolved cross-correlation measurements of an extreme ultraviolet (XUV) pulse, generated as the fifth harmonic (15.5 eV) of an intense 80 fs laser pulse centered at 400 nm. Spectrally resolving the cross-correlation signal allows us to characterize the time-dependent frequency of the XUV pulse. We find that the fifth harmonic has a small negative chirp in excess of that predicted by perturbation theory. In addition, by manipulating the chirp of the driving laser we can induce and measure a positive or a negative chirp on the XUV pulse.

  • The predissociation of highly excited states in acetylene by time-resolved photoelectron spectroscopy

    • We study the dynamics of highly excited states in acetylene initiated by an ultrashort vacuum ultraviolet laser pulse. Electronic states lying in the 4s-3d Rydberg region are excited with one femtosecond pulse, and the dynamic development of the states is monitored by a second short pulse which ionizes the system. We show that even for femtosecond pulses where the bandwidth of the exciting pulse covers several electronic states, it is possible to extract short decay lifetimes through time-resolved photoelectron spectroscopy by using a frequency-modulated (chirped) excitation pulse. We report decay lifetimes for the F 4(0)(2) and E 4-5(0)(2) states in acetylene, and for the E 4(0)(2) and E 5(0)(2) states in d-acetylene. The time evolution measured in the electron spectra is compared to decay spectra measured using ion yield and the differences in these results are discussed. (C) 2003 American Institute of Physics.

  • Effects of water activity and low molecular weight humectants on skin permeability and hydration dynamics : a double-blind, randomized and controlled study

    • OBJECTIVES: The mammalian skin is a barrier that effectively separates the water-rich interior of the body from the normally dryer exterior. Changes in the external conditions, for example ambient humidity, have been shown to affect the skin barrier properties. The prime objective of this study was to evaluate the effect of water activity of a topical formulation on skin hydration and permeability. A second objective was to gain more understanding on how two commonly used humectants, urea and glycerol, affect skin barrier function in vivo. METHODS: Simple aqueous formulations were applied under occlusion to the volar forearm of healthy volunteers. Following 4-h exposure, skin water loss (by transepidermal water loss measurements), skin hydration (by Corneometry) and skin permeability (by time to vasodilation due to benzyl nicotinate exposure) were monitored. RESULTS: The results demonstrate that a relatively small change in the water activity of a topical formulation is sufficient to induce considerable effects on stratum corneum hydration and permeability to exogenous substances. Exposing the skin to high water activity leads to increased skin hydration and also increased permeability. Furthermore, urea and glycerol promote skin hydration and permeability even at reduced water activity of the applied formulation. CONCLUSION: These results highlight the importance of considering the water activity in topically applied formulations and the potential benefit of using humectants. The results may impact formulation optimization in how to facilitate skin hydration and to modify skin permeability by temporarily open and close the skin barrier.

  • Prevalence and recognition of chronic kidney disease in Stockholm healthcare

    • Background. Chronic kidney disease (CKD) is common, but the frequency of albuminuria testing and referral to nephrology care has been difficult to measure. We here characterize CKD prevalence and recognition in a complete healthcare utilization cohort of the Stockholm region, in Sweden. Methods. We included all adult individuals (n = 1 128 058) with at least one outpatient measurement of IDMS-calibrated serum creatinine during 2006-11. Estimated glomerular filtration rate (eGFR) was calculated via the CKD-EPI equation and CKD was solely defined as eGFR <60 mL/min/1.73 m(2). We also assessed the performance of diagnostic testing (albuminuria), nephrology consultations, and utilization of ICD-10 diagnoses. Results. A total of 68 894 individuals had CKD, with a crude CKD prevalence of 6.11% [95% confidence interval (CI): 6.07-6.16%] and a prevalence standardized to the European population of 5.38% (5.33-5.42%). CKD was more prevalent among the elderly (28% prevalence >75 years old), women (6.85 versus 5.24% in men), and individuals with diabetes (17%), hypertension (17%) or cardiovascular disease (31%). The frequency of albuminuria monitoring was low, with 38% of diabetics and 27% of CKD individuals undergoing albuminuria testing over 2 years. Twenty-three per cent of the 16 383 individuals satisfying selected KDIGO criteria for nephrology referral visited a nephrologist. Twelve per cent of CKD patients carried an ICD-10 diagnostic code of CKD. Conclusions. An estimated 6% of the adult Stockholm population accessing healthcare has CKD, but the frequency of albuminuria testing, nephrology consultations and registration of CKD diagnoses was suboptimal despite universal care. Improving provider awareness and treatment of CKD could have a significant public health impact.

  • Targeting PFKFB3 radiosensitizes cancer cells and suppresses homologous recombination

    • The glycolytic PFKFB3 enzyme is widely overexpressed in cancer cells and an emerging anticancer target. Here, we identify PFKFB3 as a critical factor in homologous recombination (HR) repair of DNA double-strand breaks. PFKFB3 rapidly relocates into ionizing radiation (IR)-induced nuclear foci in an MRN-ATM-gamma H2AX-MDC1-dependent manner and co-localizes with DNA damage and HR repair proteins. PFKFB3 relocalization is critical for recruitment of HR proteins, HR activity, and cell survival upon IR. We develop KAN0438757, a small molecule inhibitor that potently targets PFKFB3. Pharmacological PFKFB3 inhibition impairs recruitment of ribonucleotide reductase M2 and deoxynucleotide incorporation upon DNA repair, and reduces dNTP levels. Importantly, KAN0438757 induces radiosensitization in transformed cells while leaving non-transformed cells unaffected. In summary, we identify a key role for PFKFB3 enzymatic activity in HR repair and present KAN0438757, a selective PFKFB3 inhibitor that could potentially be used as a strategy for the treatment of cancer.

    • In this paper, we present and compare the two different XUV interferometric techniques using high-order harmonics that have been developed so far. The first scheme is based on the interference between two spatially separated phase-locked harmonic sources while the second uses two temporally separated harmonic sources. These techniques have been applied to plasma diagnostics in feasibility experiments where electron densities up to a few 1020 e[minus sign/cm3 have been measured with a temporal resolution of 200 fs. We present the main characteristics of each technique and discuss their respective potentials and limitations.

    • We demonstrate a new scheme for extreme ultraviolet (xuv) Fourier-transform spectroscopy based on the generation of two phase-locked high-harmonic beams. It allows us to measure for the first time interferograms at wavelengths as short as 90 nm, and open the perspective of performing high-resolution Fourier-transform absorption spectroscopy in the xuv. Our measurements also demonstrate that a precise control of the relative phase of harmonic pulses can be obtained with an accuracy on an attosecond time scale, of importance for future xuv pump-xuv probe attosecond spectroscopy.

    • OBJECTIVE: To investigate-by molecular, classical and functional methods-the microbiota in biopsies and faeces from patients with active Crohn's disease (CD) and controls.DESIGN: The microbiota in biopsies was investigated utilizing a novel molecular method and classical cultivation technology. Faecal samples were investigated by classical technology and four functional methods, reflecting alterations in short chain fatty acids pattern, conversion of cholesterol and bilirubin and inactivation of trypsin.RESULTS: By molecular methods we found more than 92% similarity in the microbiota on the biopsies from the two groups. However, 4.6% of microbes found in controls were lacking in CD patients. Furthermore, NotI representation libraries demonstrate two different clusters representing CD patients and controls, respectively. Utilizing conventional technology, Bacteroides (alt. Parabacteroides) was less frequently detected in the biopsies from CD patients than from controls. A similar reduction in the number of Bacteroides was found in faecal samples. Bacteroides is the only group of bacteria known to be able to inactivate pancreatic trypsin. Faecal tryptic activity was high in CD patients, and inversely correlated to the levels of Bacteroides.CONCLUSIONS: CD patients have compositional and functional alterations in their intestinal microbiota, in line with the global description hypothesis rather than the candidate microorganism theory. The most striking functional difference was high amount of faecal tryptic activity in CD patients, inversely correlated to the levels of Bacteroides in faeces.

  • Characterisation and Applications of Ultrashort Extreme Ultraviolet Pulses

    • When a strong laser field interacts with free atoms, harmonics up to very high orders are created. This extreme ultraviolet (XUV) radiation has interesting properties, such as spatial and temporal coherence, short pulse duration and high brightness. This thesis concentrates on the temporal aspects of this radiation. A method to characterise the pulse duration and time-dependent frequency (chirp) of such harmonic pulses has been developed. To perform the characterisation, the energy and amplitude of a photoelectron peak, generated through ionization of a rare gas after absorption of one XUV and one infrared laser photon, is measured as a function of the delay between the two pulses. In addition, by manipulating the chirp of the driving laser, positive or negative chirp could be induced on the harmonic radiation. By creating two sources of mutually coherent harmonics and analysing the interference in the far field, the first-order autocorrelation function has been measured. By overlapping the two sources, a high order autocorrelation trace of the laser was recorded. An XUV monochromator capable of extracting single harmonic frequencies without stretching them in time has been designed. Experiments have been performed on a prototype working in the infrared wavelength region. High-order harmonics have been used to perform XUV interferometric measurements. The beams were divided in the infrared before the generation of harmonics. The thickness of aluminium filters and the electron density of a laser-produced plasma were determined. In another application, harmonics have been used to perform lifetime measurements of Rydberg states in acetylene.

  • Design of an extreme-ultraviolet monochromator free from temporal stretching

    • High-order harmonic generation in gases by use of femtosecond lasers is a source of ultrashort pulses in the extreme-ultraviolet (XUV). For many applications it is necessary to select radiation of only one specific harmonic order without affecting the duration of the ultrashort pulse. A three-grating monochromator that meets this demand has been designed and modeled by ray tracing as well as by wave-optical simulations. The only remaining temporal stretching of an XUV pulse is due to distortion of the pulse front on the gratings and is predicted to be similar to1 fs. The design has been successfully tested in the near infrared. Finally, the monochromator is also capable of eliminating any existing linear chirp in the harmonic pulses, thus compressing them to shorter durations. (C) 2004 Optical Society of America.

    • There have been concerns about serious infusion-related adverse drug reactions (ADR) with rituximabin chronic lymphocytic leukemia (CLL). We therefore conducted an observational trial in which CLL patients planned for rituximab-containing therapy were eligible. Ninety-six patients from 19 centers were enrolled. The most common regimen was rituximab, fludarabine and cyclophosphamide. Fifty-six patients experienced ADR during rituximab infusion. Reactions greater than= grade 3 occurred in five patients and no cases of tumor lysis syndrome were recorded. Despite a high number of circulating tumor cells few severe ADR were noted. Thus, rituximab containing regimens can be considered safe for CLL patients in general practice.

  • Endophilin A2 deficiency protects rodents from autoimmune arthritis by modulating T cell activation

    • The introduction of the CTLA-4 recombinant fusion protein has demonstrated therapeutic effects by selectively modulating T-cell activation in rheumatoid arthritis. Here we show, using a forward genetic approach, that a mutation in the SH3gl1 gene encoding the endocytic protein Endophilin A2 is associated with the development of arthritis in rodents. Defective expression of SH3gl1 affects T cell effector functions and alters the activation threshold of autoreactive T cells, thereby leading to complete protection from chronic autoimmune inflammatory disease in both mice and rats. We further show that SH3GL1 regulates human T cell signaling and T cell receptor internalization, and its expression is upregulated in rheumatoid arthritis patients. Collectively our data identify SH3GL1 as a key regulator of T cell activation, and as a potential target for treatment of autoimmune diseases. The autoimmune disorder, rheumatoid arthritis (RA), has been associated with multiple pathophysiological factors. Here the authors show that deficiency in endophilin A2 in rodents protects them from experimental arthritis by altering T cell activation threshold and effector functions, thereby hinting a potential target for RA therapy.
