  • Key features for more successful place-based sustainability research on social-ecological systems : a Programme on Ecosystem Change and Society (PECS) perspective

    • The emerging discipline of sustainability science is focused explicitly on the dynamic interactions between nature and society and is committed to research that spans multiple scales and can support transitions toward greater sustainability. Because a growing body of place-based social-ecological sustainability research (PBSESR) has emerged in recent decades, there is a growing need to understand better how to maximize the effectiveness of this work. The Programme on Ecosystem Change and Society (PECS) provides a unique opportunity for synthesizing insights gained from this research community on key features that may contribute to the relative success of PBSESR. We surveyed the leaders of PECS-affiliated projects using a combination of open, closed, and semistructured questions to identify which features of a research project are perceived to contribute to successful research design and implementation. We assessed six types of research features: problem orientation, research team, and contextual, conceptual, methodological, and evaluative features. We examined the desirable and undesirable aspects of each feature, the enabling factors and obstacles associated with project implementation, and asked respondents to assess the performance of their own projects in relation to these features. Responses were obtained from 25 projects working in 42 social-ecological study cases within 25 countries. Factors that contribute to the overall success of PBSESR included: explicitly addressing integrated social-ecological systems; a focus on solutionand transformation-oriented research; adaptation of studies to their local context; trusted, long-term, and frequent engagement with stakeholders and partners; and an early definition of the purpose and scope of research. Factors that hindered the success of PBSESR included: the complexities inherent to social-ecological systems, the imposition of particular epistemologies and methods on the wider research group, the need for long periods of time to initiate and conduct this kind of research, and power asymmetries both within the research team and among stakeholders. In the self-assessment exercise, performance relating to team and context-related features was ranked higher than performance relating to methodological, evaluation, and problem orientation features. We discuss how these insights are relevant for balancing place-based and global perspectives in sustainability science, fostering more rapid progress toward inter-and transdisciplinary integration, redefining and measuring the success of PBSESR, and facing the challenges of academic and research funding institutions. These results highlight the valuable opportunity that the PECS community provides in helping build a community of practice for PBSESR.

  • Germline heterozygous variants in genes associated with familial hemophagocytic lymphohistiocytosis as a cause of increased bleeding

    • Familial hemophagocytic lymphohistiocytosis (FHL) is caused by biallelic variants in genes regulating granule secretion in cytotoxic lymphocytes. In FHL3–5, the affected genes UNC13D, STX11 and STXBP2 have further been shown to regulate the secretion of platelet granules, giving rise to compromised platelet function. Therefore, we aimed to investigate platelet degranulation in patients heterozygous for variants in UNC13D, STX11 and STXBP2. During the work-up of patients referred to the Coagulation Unit, Skåne University Hospital, Malmö, Sweden and the Department of Hematology, Rigshospitalet, Copenhagen, Denmark due to bleeding tendencies, 12 patients harboring heterozygous variants in UNC13D, STX11 or STXBP2 were identified using targeted whole exome sequencing. Transmission electron microscopy (TEM) was used to assess the secretion of platelet dense granules following thrombin stimulation. Platelet degranulation, activation and aggregation were further assessed by flow cytometry (FC) and light transmission aggregometry (LTA) with lumi-aggregometry. In total, eight out of twelve (67%) patients showed impaired degranulation by at least one of the assays (TEM, FC and LTA). In the 12 patients, eight different heterozygous variants were identified. One variant was strongly associated with impaired degranulation, while four of the variants were associated with impaired granule secretion to a slightly lesser extent. One additional variant was found in six out of the twelve patients, and was associated with varying degrees of degranulation impairment. Accordingly, six out of the eight (75%) identified variants were associated with impaired platelet degranulation. Our results suggest that heterozygous variants in UNC13D, STX11 and STXBP2 are sufficient to cause platelet secretion defects resulting in increased bleeding.

  • Advancing sustainability through mainstreaming a social–ecological systems perspective

    • The concept of social-ecological systems is useful for understanding the interlinked dynamics of environmental and societal change. The concept has helped facilitate: (1) increased recognition of the dependence of humanity on ecosystems; (2) improved collaboration across disciplines, and between science and society; (3) increased methodological pluralism leading to improved systems understanding; and (4) major policy frameworks considering social-ecological interactions. Despite these advances, the potential of a social-ecological systems perspective to improve sustainability outcomes has not been fully realized. Key priorities are to: (1) better understand and govern social-ecological interactions between regions; (2) pay greater attention to long-term drivers; (3) better understand the interactions among power relations, justice, and ecosystem stewardship; and (4) develop a stronger science-society interface.

  • Measuring ecosystem multifunctionality across scales

    • Multifunctionality refers to the capacity of an area to supply multiple ecosystem functions or services. While many conceptual and methodological advances have focused on defining and quantifying multifunctionality, the challenge of dealing with cross-scale dynamics of multifunctionality remains open. This study proposes a new way of measuring multifunctionality across spatial scales, illustrated with a European-wide dataset of 18 ecosystem services. Our assessment captures not only the diversity of ecosystem services supplied within each municipality (alpha-multifunctionality), but also the unique contribution of each municipality to the regional ecosystem service diversity (beta-multifunctionality). This cross-scale analysis helps better understanding the spatial distribution of ecosystem services, which is required to design management and policies at the right scale. Our analysis shows that alpha-multifunctionality follows a latitudinal gradient across Europe and strongly decreases towards the city centers of metropolitan areas. By relating alpha- and beta-multifunctionality to land use intensity, we show that low-intensity management systems support higher ecosystem multifunctionality across Europe. Municipalities of low alpha-multifunctionality often contribute significantly to regional multifunctionality, by providing ecosystem services of a specific value to the region. Our method to measure both alpha- and beta-multifunctionality thus provides a new way to inform reconciliation of competing land uses when maximizing alpha-multifunctionality is not reasonable.

    • A new animal model for the purpose of studying superficial infections is presented. In this model, an infection is established by disruption of the skin barrier by partial removal of the epidermal layer by tape stripping and subsequent application of the pathogens Staphylococcus aureus and Streptococcus pyogenes. The infection and the infection route are purely topical, in contrast to those used in previously described animal models in mice, such as the skin suture-wound model, where the infection is introduced into the deeper layers of the skin. Thus, the present model is considered more biologically relevant for the study of superficial skin infections in mice and humans. Established topical antibiotic treatments are shown to be effective. The procedures involved in the model are simple, a feature that increases throughput and reproducibility. This new model should be applicable to the evaluation of novel antimicrobial treatments of superficial infections caused by S. aureus and S. pyogenes.

    • FusE mutants are fusidic acid-resistant small colony variants (SCVs) of Staphylococcus aureus that can be selected with aminoglycosides. All FusE SCVs have mutations in rplF, encoding ribosomal protein L6. However, individual FusE mutants including some with the same mutation in rplF display auxotrophy for either hemin or menadione, suggesting that additional mutations are involved. Here we show that FusE SCVs can be divided into three genetic sub-groups and that some carry an additional mutation, in one of the genes required for hemin biosynthesis, or in one of the genes required for menadione biosynthesis. Reversion analysis and genome sequencing support the hypothesis that these combinations of mutations in the rplF, hem, and/or men genes can account for the SCV and auxotrophic phenotypes of FusE mutants.

  • Genetic Determinants of Resistance to Fusidic Acid among Clinical Bacteremia Isolates of Staphylococcus aureus

    • Resistance to fusidic acid in Staphylococcus aureus is caused by mutation of the elongation factor G (EF-G) drug target (FusA class) or by expression of a protein that protects the drug target (FusB and FusC classes). Recently, two novel genetic classes of small-colony variants (SCVs) were identified among fusidic acid-resistant mutants selected in vitro (FusA-SCV and FusE classes). We analyzed a phylogenetically diverse collection of fusidic acid-resistant bacteremia isolates to determine which resistance classes were prevalent and whether these were associated with particular phylogenetic lineages. Each isolate was shown by DNA sequencing and plasmid curing to carry only one determinant of fusidic acid resistance, with approximately equal frequencies of the FusA, FusB, and FusC genetic classes. The FusA class (mutations in fusA) were distributed among different phylogenetic types. Two distinct variants of the FusC class (chromosomal fusC gene) were identified, and FusC was also distributed among different phylogenetic types. In contrast, the FusB class (carrying fusB on a plasmid) was found in closely related types. No FusE-class mutants (carrying mutations in rplF) were found. However, one FusA-class isolate had multiple mutations in the fusA gene, including one altering a codon associated with the FusA-SCV class. SCVs are frequently unstable and may undergo compensatory evolution to a normal growth phenotype after their initial occurrence. Accordingly, this normal-growth isolate might have evolved from a fusidic acid-resistant SCV. We conclude that at least three different resistance classes are prevalent among fusidic acid-resistant bacteremia isolates of S. aureus.

  • A novel telecoupling framework to assess social relations across spatial scales for ecosystem services research

    • Access to ecosystem services and influence on their management are structured by social relations among actors, which often occur across spatial scales. Such cross-scale social relations can be analysed through a telecoupling framework as decisions taken at local scales are often shaped by actors at larger scales. Analyzing these cross-scale relations is critical to create effective and equitable strategies to manage ecosystem services. Here, we develop an analytical framework -i.e. the 'cross-scale influence-dependence framework'- to facilitate the analysis of power asymmetries and the distribution of ecosystem services among the beneficiaries. We illustrate the suitability of this framework through its retrospective application across four case studies, in which we characterize the level of dependence of multiple actors on a particular set of ecosystem services, and their influence on decision-making regarding these services across three spatial scales. The 'cross-scale influence-dependence framework' can improve our understanding of distributional and procedural equity and thus support the development of policies for sustainable management of ecosystem services.

    • Social-ecological interactions have been shown to generate interrelated and reoccurring sets of ecosystem services, also known as ecosystem service bundles. Given the potential utility of the bundles concept, along with the recent surge in interest it is timely to reflect on the concept, its current use and potential for the future. Based on our ecosystem service bundle experience, expertise, and ecosystem service bundle analyses, we have found critical elements for advancing the utility of ecosystem service bundle concept and deepening its impact in the future. In this paper we 1) examine the different conceptualizations of the ecosystem service bundle concept; 2) show the range of benefits of using a bundles approach; 3) explore key issues for improving research on ecosystem service bundles, including indicators, scale, and drivers and relationships between ecosystem services; and 4) outline priorities for the future by facilitating comparisons of ecosystem service bundle research.

    • Small-colony variants (SCVs) of Staphylococcus aureus are a slow-growing subpopulation whose phenotypes can include resistance to aminoglycosides, defects in electron transport, and enhanced persistence in mammalian cells. Here we show that a subset of mutants selected as SCVs by reduced susceptibility to aminoglycosides are resistant to the antibiotic fusidic acid (FA) and conversely that a subset of mutants selected for resistance to FA are SCVs. Mutation analysis reveals different genetic classes of FA-resistant SCVs. One class, FusA-SCVs, have amino acid substitution mutations in the ribosomal translocase EF-G different from those found in classic FusA mutants. Most of these mutations are located in structural domain V of EF-G, but some are in domain I or III. FusA-SCVs are auxotrophic for hemin. A second class of FA-resistant SCVs carry mutations in rplF, coding for ribosomal protein L6, and are designated as FusE mutants. FusE mutants fall into two phenotypic groups: one auxotrophic for hemin and the other auxotrophic for menadione. Accordingly, we have identified new genetic and phenotypic classes of FA-resistant mutants and clarified the genetic basis of a subset of S. aureus SCV mutants. A clinical implication of these data is that FA resistance could be selected by antimicrobial agents other than FA.

  • On Fusidic Acid Resistance in Staphylococcus Aureus

    • Controlling bacterial infections with antibiotics is central to modern health care. However, increasing bacterial resistance to antibiotics threatens effective therapy. This thesis concerns the use of the antibiotic fusidic acid, and novel analogues of fusidic acid, to treat topical infections caused by the bacterial pathogen Staphylococcu aureus. It also addresses genetic mechanisms by which S. aureus develops resistance to fusidic acid.Pre-clinical microbiological tests were made on two structurally different groups of fusidic acid analogues developed by Leo Pharma. These drugs were tested against S. aureus and Streptococcus pyogenes strains, measuring MIC, in vitro concentration-dependent bacteriocidal or bacteriostatic effects, and in vivo efficacy in clearing topical infections. We developed a new superficial skin infection animal model (the ‘tape-stripping model’) designed for testing topical antibiotics, including the novel fusidic acid analogues, against S. aureus and S. pyogenes. Some new compounds giving promising results will be further tested and developed by Leo Pharma. Fusidic acid inhibits protein synthesis by binding to elongation factor EF-G on the ribosome. Previously described resistance mechanisms are mutations in the gene coding for EF-G (fusA), or, in some strains, the presence of a gene (fusB, fusC or fusD) coding for a protein that protects EF-G from fusidic acid.We discovered two novel classes of spontaneous FusR mutants in S. aureus with the small colony variant (SCV) phenotype which is associated with persistent infections. The FusR SCV’s are very frequent, slow growing, cross-resistant to aminoglycosides, and auxotrophic for hemin or menadione. Some of the FusR SCV mutations are in structural domain V of EF-G (classic fusA mutations map overwhelmingly in domain III). The remaining FusR SCV’s are unmapped but their additive effect on MIC together with the fusB plasmid suggests the possibility that their mechanism of resistance is also associated with the translation machinery.

    • An increasing number of voices highlight the need for science itself to transform and to engage in the co-production of knowledge and action, in order to enable the fundamental transformations needed to advance towards sustainable futures. But how can global sustainability-oriented research networks engage in co-production of knowledge and action? The present article introduces a strategic tool called the ‘network compass’ which highlights four generic, interrelated fields of action through which networks can strive to foster co-production. It is based on the networks’ particular functions and how these can be engaged for co-production processes. This tool aims to foster self-reflection and learning within and between networks in the process of (re)developing strategies and activity plans and effectively contributing to sustainability transformations.

  • Automatic classification of the Sub-Techniques (Gears) used in cross-country ski skating employing a mobile phone

    • The purpose of the current study was to develop and validate an automatic algorithm for classification of cross-country (XC) ski-skating gears (G) using Smartphone accelerometer data. Eleven XC skiers (seven men, four women) with regional-to-international levels of performance carried out roller skiing trials on a treadmill using fixed gears (G2left, G2right, G3, G4left, G4right) and a 950-m trial using different speeds and inclines, applying gears and sides as they normally would. Gear classification by the Smartphone (on the chest) and based on video recordings were compared. Formachine-learning, a collective database was compared to individual data. The Smartphone application identified the trials with fixed gears correctly in all cases. In the 950-m trial, participants executed 140 ± 22 cycles as assessed by video analysis, with the automatic Smartphone application giving a similar value. Based on collective data, gears were identified correctly 86.0% ± 8.9% of the time, a value that rose to 90.3% ± 4.1% (P < 0.01) with machine learning from individual data. Classification was most often incorrect during transition between gears, especially to or from G3. Identification was most often correct for skiers who made relatively few transitions between gears. The accuracy of the automatic procedure for identifying G2left, G2right, G3, G4left and G4right was 96%, 90%, 81%, 88% and 94%, respectively. The algorithm identified gears correctly 100% of the time when a single gear was used and 90% of the time when different gears were employed during a variable protocol. This algorithm could be improved with respect to identification of transitions between gears or the side employed within a given gear.
