    • Wiskott-Aldrich syndrome (WAS) is caused by loss-of-function mutations in the WASp gene. Decreased cellular responses in WASp-deficient cells have been interpreted to mean that WASp directly regulates these responses in WASp-sufficient cells. Here, we identify an exception to this concept and show that WASp-deficient dendritic cells have increased activation of Rac2 that support cross-presentation to CD8(+) T cells. Using two different skin pathology models, WASp-deficient mice show an accumulation of dendritic cells in the skin and increased expansion of IFN gamma-producing CD8(+) T cells in the draining lymph node and spleen. Specific deletion of WASp in dendritic cells leads to marked expansion of CD8(+) T cells at the expense of CD4(+) T cells. WASp-deficient dendritic cells induce increased cross-presentation to CD8(+) T cells by activating Rac2 that maintains a near neutral pH of phagosomes. Our data reveals an intricate balance between activation of WASp and Rac2 signalling pathways in dendritic cells.

    • This paper provides a comprehensive characterisation of HVOF- and HVAF-sprayed Cr3C2–25 wt.% NiCr hardmetal coatings. One commercial powder composition with two different particle size distributions was processed using five HVOF and HVAF thermal spray systems.All coatings contain less Cr3C2 than the feedstock powder, possibly due to the rebound of some Cr3C2-rich particles during high-velocity impact onto the substrate.Dry sand-rubber wheel abrasive wear testing causes both grooving and pull-out of splat fragments. Mass losses depend on inter- and intra-lamellar cohesion, being higher (≥70 mg after a wear distance of 5904 m) for the coatings deposited with the coarser feedstock powder or with one type of HVAF torch.Sliding wear at room temperature against alumina involves shallower abrasive grooving, small-scale delamination and carbide pull-outs, and it is controlled by intra-lamellar cohesion. The coatings obtained from the fine feedstock powder exhibit the lowest wear rates (≈5x10−6 mm3/(Nm)). At 400 °C, abrasive grooving dominates the sliding wear behaviour; wear rates increase by one order of magnitude but friction coefficients decrease from ≈0.7 to ≈0.5. The thermal expansion coefficient of the coatings (11.08x10−6 °C−1 in the 30–400 °C range) is sufficiently close to that of the steel substrate (14.23x10−6 °C−1) to avoid macro-cracking

  • Tribology of HVOF- and HVAF-sprayed WC-10Co4Cr hardmetal coatings : A comparative assessment

    • his paper provides a comprehensive assessment of the sliding and abrasive wear behaviour of WC–10Co4Cr hardmetal coatings, representative of the existing state-of-the-art. A commercial feedstock powder with two different particle size distributions was sprayed onto carbon steel substrates using two HVOF and two HVAF spray processes.Mild wear rates of < 10-7 mm3/(Nm) and friction coefficients of ≈ 0.5 were obtained for all samples in ball-on-disk sliding wear tests at room temperature against Al2O3 counterparts. WC–10Co4Cr coatings definitely outperform a reference electrolytic hard chromium coating under these test conditions. Their wear mechanisms include extrusion and removal of the binder matrix, with the formation of a wavy surface morphology, and brittle cracking. The balance of such phenomena is closely related to intra-lamellar features, and rather independent of those properties (e.g. indentation fracture toughness, elastic modulus) which mainly reflect large-scale inter-lamellar cohesion, as quantitatively confirmed by a principal component analysis. Intra-lamellar dissolution of WC into the matrix indeed increases the incidence of brittle cracking, resulting in slightly higher wear rates. At 400 °C, some of the hardmetal coatings fail because of the superposition between tensile residual stresses and thermal expansion mismatch stresses (due to the difference between the thermal expansion coefficients of the steel substrate and of the hardmetal coating). Those which do not fail, on account of lower residual stresses, exhibit higher wear rates than at room temperature, due to oxidation of the WC grains.The resistance of the coatings against abrasive wear, assessed by dry sand–rubber wheel testing, is related to inter-lamellar cohesion, as proven by a principal component analysis of the collected dataset. Therefore, coatings deposited from coarse feedstock powders suffer higher wear loss than those obtained from fine powders, as brittle inter-lamellar detachment is caused by their weaker interparticle cohesion, witnessed by their systematically lower fracture toughness as well.

  • Towards better lighting recommendations for open surgery

    • This study assesses the lighting requirements for operating theatres and the lighting conditions in a specific test case and proposes alternate ways of achieving a favourable visual environment. The hypothesis was that an increased illuminance level from general lighting and lower luminance contrasts between the essential task areas affect perceived lighting quality, visual ability and performance, as well as tiredness of medical staff performing surgical work. The investigation consisted of three parts: a laboratory study, followed by a field study and concluding with a retrospective study. In the laboratory study, each medical staff subject was exposed to four lighting scenarios with three illuminances from the operating light (12 lighting conditions), with no patients present. In the field and the retrospective study, the existing lighting scenario was compared to the highest-rated test lighting scenario (with illuminances more than double). The field study and the retrospective study took place with medical staff working under either the test general lighting condition or the existing general lighting condition. Increased illuminance from the general lighting and a higher correlated colour temperature did not improve the operating staff’s visual ability. However, medical staff rated the change in lighting quality resulting from the increased general illuminance as an improvement in the work environment, both in the laboratory study and in the field study. Medical staff subjects using the operating theatre rated both perceived visual ability and lighting quality higher under the test lighting scenario. Tiredness was also rated lower under that test scenario. Based on the results from this study, general lighting levels of 2000 lx in the periphery of the operating room, 4000 lx surrounding the operating table and 5500 lx on the operating table are recommended by the authors. With reference to the selections made by the surgeons for the illuminance in the operating cavity, it is suggested that the central beam illuminance from the surgical luminaires be between 50,000 lx and 100,000 lx.

    • A new concept for treatment of infections is induction of our own antimicrobial peptides and the presented novel class of inducer, aroylated phenylenediamines (APDs), gives up to 20 to 30-fold induction of the human antimicrobial peptide LL-37, in vitro. In addition, oral administration of an APD in a rabbit model of Shigellosis resulted in recovery from the infection in a few days implying that APD’s are promising candidates for treatment of infections.

  • Synergonomi och ögonbesvär hos personal på ögonsjukhus.

    • Ögonläkare, ögonsjuksköterskor, optiker och annan personal på ögonsjukhus arbetar ofta med synkrävande arbetsuppgifter i dämpad belysning eller helt utan allmänbelysning. Inför den planerade byggnationen av ett nytt ögonsjukhus i Stockholm ca år 2018 gjordes en enkätstudie för att kartlägga ögonbesvär och trötthet hos personalen på S:t Eriks Ögonsjukhus. Studien som är ett multidisciplinärt samarbetsprojekt mellan S:t Erik, Karolinska Institutet och Kungliga Tekniska Högskolan, syftar till att optimera belysning, dagsljusinsläpp och ljusmiljö vid det nya ögonsjukhuset.Totalt 265 anställda på S:t Eriks Ögonsjukhus samt 60 röntgenläkare och 45 barnläkare, varav de senare två  utgjorde jämförelsegrupper, inviterades till  studien.  Enkäten som distribuerades baserades på synergonomienkäter av Knave och Hemphälä och bestod av 31 validerade frågor om subjektiva ögonbesvär, nuvarande belysning, tillgång till dagsljus och välbefinnande. Ögonbesvären räknades om till ett gruppmedelvärde (ögonbesvärsindex) med avseende på svårighetsgrad och frekvens. Studien är godkänd av Etikprövningsnämnden. Nittiosex av 265 (33%) anställda på S:t Eriks Ögonsjukhus hade t o m juni 2012 besvarat enkäten tillsammans med 23 röntgenläkare (38%) och 14 barnläkare (31%). Ögonbesvär som torrhets- och gruskänsla var vanligt förekommande hos alla yrkesgrupper på ögonsjukhuset och generellt vanligare hos kvinnor. Då samtliga grupper jämfördes med avseende på ögonbesvärsindex var skillnaden mellan män och kvinnor statistiskt säkerställd (p<0,05). Röntgenläkarna rapporterade högst ögonbesvärsindex och barnläkarna lägst, skillnaden mellan dessa yrkesgrupper var signifikant (p<0,05). Ögonpersonal och röntgenläkare som rapporterade att de ofta arbetade i mörker, associerade detta med  ökad trötthet i högre grad än  med barnläkarna (p<0,05).Ögonbesvär var vanliga hos ögonpersonalen inkluderade i studien. Den grupp som arbetade mest i mörker hade mer ögonbesvär än den grupp som arbetade minst i mörker. Kvinnor hade mer ögonbesvär än män. Arbete i mörker ökade den subjektiva känslan av trötthet . Optimala ljusförhållanden och bra synergonomi bör ges hög prioritet vid planering av ett nytt ögonsjukhus.

  • Visual ergonomics and eye strain in eye careprofessionals

    • Eye care professionals spend many hours a day in darkness performing visually demanding tasks. A new eye hospital will be built in Stockholm 2018. The current lighting, logistics, and working conditions are analysed in a multidisciplinary project aiming to optimise settings in the new hospital. The main purpose of the present project was to study visual ergonomics and current eye strain in employees at an eye hospital. Ninety-six employees answered a validated questionnaire regarding their experiences of light, visual ergonomics and eye strain problems. Twenty-three radiologists and 14 paediatricians at a university hospital were used as comparison groups. Eye strain was common in all departments at the hospital but was significantly more common only among radiologists compared to paediatricians. Overall, women experienced significantly more eye strain than men.

  • BCG Skin Infection Triggers IL-1R-MyD88-Dependent Migration of EpCAMlow CD11bhigh Skin Dendritic cells to Draining Lymph Node During CD4+ T-Cell Priming

    • The transport of antigen from the periphery to the draining lymph node (DLN) is critical for T-cell priming but remains poorly studied during infection with Mycobacterium bovis Bacille Calmette-Guérin (BCG). To address this we employed a mouse model to track the traffic of Dendritic cells (DCs) and mycobacteria from the BCG inoculation site in the skin to the DLN. Detection of BCG in the DLN was concomitant with the priming of antigen-specific CD4+ T cells at that site. We found EpCAMlow CD11bhigh migratory skin DCs to be mobilized during the transport of BCG to the DLN. Migratory skin DCs distributed to the T-cell area of the LN, co-localized with BCG and were found in close apposition to antigen-specific CD4+ T cells. Consequently, blockade of skin DC traffic into DLN dramatically reduced mycobacterial entry into DLN and muted T-cell priming. Interestingly, DC and mycobacterial entry into the DLN was dependent on IL-1R-I, MyD88, TNFR-I and IL-12p40. In addition, we found using DC adoptive transfers that the requirement for MyD88 in BCG-triggered migration was not restricted to the migrating DC itself and that hematopoietic expression of MyD88 was needed in part for full-fledged migration. Our observations thus identify a population of DCs that contribute towards the priming of CD4+ T cells to BCG infection by transporting bacilli into the DLN in an IL-1R-MyD88-dependent manner and reveal both DC-intrinsic and -extrinsic requirements for MyD88 in DC migration.

    • Antimicrobial peptides are important for a healthy host-microbe homeostasis. In infections characterized by low levels of the human cathelicidin, LL-37, induction of its expression increases clearance of pathogens. Our aim was to discover signaling pathways and compounds capable of affecting the expression of LL-37. We recently observed a synergistic induction of LL-37 expression by stimulating the colonic epithelial cell-line HT-29 with lactose and phenylbutyrate (PBA). Here, we studied regulatory circuits mediating this synergism in HT-29 cells stimulated with lactose (60 g/l) and PBA (2 m<smlcap>M</smlcap>) for 24 h by using mass spectrometry and pathway analyses. Selected pathways were evaluated for their involvement in LL-37 regulation in a <i>CAMP</i> gene-luciferase reporter system. Three pathways were examined in detail: thyroid hormone receptor and retinoid X receptor (TR/RXR) activation, eicosanoid signaling and steroid biosynthesis. Induced expression of LL-37 was observed upon stimulation with triiodothyronine (T3, 2.5 n<smlcap>M</smlcap>-1 µ<smlcap>M</smlcap> for 3-30 h) and thyroxine (T4, 2.5-10 n<smlcap>M</smlcap> for 24 h). Furthermore, the synergism of lactose and PBA was reduced in cells coincubated with inhibitors of phospholipase A2, cyclooxygenase 2 or HMG-CoA reductase. Based on these results, we conclude that proteomics and pathway analyses are valuable tools for dissecting the regulatory networks involved in LL-37 expression.

    • The complexity of many components being coated in the aircraft industry today makes the traditional trial and error approach to obtain uniform coatings inadequate. To reduce programming time and further increase process accuracy a more systematic approach to develop robot trajectories is needed. In earlier work, a mathematical model was developed to predict coating thickness for thermal spray deposition on rotating objects with rotationally invariant surfaces. The model allows for varying spray distance and spray direction but is simple enough to give very short simulation times. An iterative method for robot feed optimization to obtain uniform coatings was also proposed. Currently, the use of the model in engineering practice is being evaluated. A MATLAB implementation of the model has been integrated with a commercial off-line programming system, giving a powerful and efficient tool to predict and optimize coating thickness. Simulations and experimental verifications are presented for two zirconia plasma sprayed parts.

    • The average residual stress distribution as a function of depth in an air plasma-sprayed yttria stabilized zirconia top coat used in thermal barrier coating (TBC) systems was measured using synchrotron radiation X-ray diffraction in reflection geometry on station I15 at Diamond Light Source, UK, employing a series of incidence angles. The stress values were calculated from data deconvoluted from diffraction patterns collected at increasing depths. The stress was found to be compressive through the thickness of the TBC and a fluctuation in the trend of the stress profile was indicated in some samples. Typically this fluctuation was observed to increase from the surface to the middle of the coating, decrease a little and then increase again towards the interface. The stress at the interface region was observed to be around 300 MPa, which agrees well with the reported values. The trend of the observed residual stress was found to be related to the crack distribution in the samples, in particular a large crack propagating from the middle of the coating. The method shows promise for the development of a nondestructive test for as-manufactured samples.

    • Optical and electronic properties of metal (Pb, Bi, Sn, and In) doped Zn4Sb3 are reported in the temperature range 80-300 K, which covers the beta, alpha, and alpha(') structural phases of this thermoelectric material. Metal doping alters the subtle balance between Zn disorder and Zn deficiency present in beta-Zn4Sb3 and changes its low temperature structural behavior. Analysis of infrared reflection data shows that the formation of ordered alpha'-Zn4Sb3 is accompanied by a substantial increase in the free charge-carrier concentration. In contrast, for samples where doping suppresses the occurrence of the low temperature alpha'-phase, the free charge-carrier concentration is only weakly temperature dependent. Different degrees of structural disorder in doped beta-Zn4Sb3 and the ordering processes at low temperatures leading to alpha- and alpha'-Zn4Sb3 are also recognized in the charge-carrier dynamics.

    • Bacterial resistance against classical antibiotics is a growing problem and the development of new antibiotics is limited. Thus, novel alternatives to antibiotics are warranted. Antimicrobial peptides (AMPs) are effector molecules of innate immunity that can be induced by several compounds, including vitamin D and phenyl-butyrate (PBA). Utilizing a luciferase based assay, we recently discovered that the histone deacetylase inhibitor Entinostat is a potent inducer of the CAMP gene encoding the human cathelicidin LL-37. Here we investigate a mechanism for the induction and also find that Entinostat up-regulates human β-defensin 1. Analysis of the CAMP promoter sequence revealed binding sites for the transcription factors STAT3 and HIF-1α. By using short hairpin RNA and selective inhibitors, we found that both transcription factors are involved in Entinostat-induced expression of LL-37. However, only HIF-1α was found to be recruited to the CAMP promoter, suggesting that Entinostat activates STAT3, which promotes transcription of CAMP by increasing the expression of HIF-1α. Finally, we provide in vivo relevance to our findings by showing that Entinostat-elicited LL-37 expression was impaired in macrophages from a patient with a STAT3-mutation. Combined, our findings support a role for STAT3 and HIF-1α in the regulation of LL-37 expression.

  • Från utsläppsscenarier till lokal nederbörd och översvämningsrisker

    • Inom det av MSB finansierade projektet ”Nederbörd och översvämningar i framtidens Sverige − ett system till stöd för klimatanpassning” har SMHI ansvarat för hydrologisk och hydraulisk modellering samt framtagande av tidsserier med lokalt klimat för framtida förhållanden. Två metoder att bearbeta klimatdata har använts; SMHI:s Distributionsbaserad skalering (DBS) och en statistisk metod utarbetad vid Göteborgs universitet. Hydrologiska modeller har satts upp för Ätran och Torneälven och har sedan använts för beräkningar med de framtagna tidsserierna, som omfattar både SRES- och RCP-scenarier. Flöden med 100 års och 200 års återkomsttid har därefter beräknats liksom Beräknat högsta flöde (Bhf). Hydrauliska modeller för Falkenberg vid Ätran och Haparanda vid Torneälven har satts upp och beräkningar av vattennivåer har gjorts utifrån flödesberäkningarna och antaganden om havsnivåer. Därefter har översvämningsskikt applicerats på kartor. Samtliga beräkningar pekar mot ökande årsmedeltemperatur och årsmedelnederbörd i framtiden. För Ätrans avrinningsområde beräknas att antalet dygnstemperaturer kring noll grader och kring 15°C minskar till förmån för fler dagar med 5-10°C och från 15°C uppemot 25°C. Dagar med mindre än 10 mm nederbörd blir färre och dagar med 10-40 mm ökar. För Torneälvens avrinningsområde ser de riktigt låga dygnstemperaturerna ut att minska och istället blir det fler dagar med temperaturer strax under noll grader. En ökning av antalet dagar med medeltemperaturer kring 15°C ökar. Dagar med några mm nederbörd minskar och dagar med 5-25 mm ökar. Samtliga beräkningar pekar på ökad intensiv nederbörd och antalet torra dagar minskar enligt nästan alla beräkningar. Generellt sett ökar samtliga beräknade flöden för Ätran och för Torneälven ser de ut att minska mot slutet av seklet. Det dimensionerande snötäcket minskar för Ätran med 30-80%. Även för Torneälven ses minskande snötäcke i de flesta scenarier på ca 10-60%. Medeltillrinningen ser ut att öka både för Torneälven och för Ätran. Den maximala tillrinningen minskar troligen för Torneälven, vilket till stor del kan kopplas till mindre snömängder, och ökar för Ätran, vilket beror på ökad nederbörd. För Torneälven ses en förskjutning mot tidigare flödestoppar på våren, kopplat till en tidigare snösmältning. Havsvattenståndet har stor betydelse för vattennivåerna i Ätran ända upp till Tullbron. Uppströms Tullbron är det storleksordningen på flödet som styr hur hög vattennivån blir i Ätran. Havet påverkar vattennivåerna i mynningen vid Torneälven och längs kusten. Uppströms i Torneälven är det storleksordningen på flödet som styr vattennivåerna, och därmed risken för översvämning. Summary

    • Stishovite (SiO2 with the rutile structure and octahedrally coordinated silicon) is an important high-pressure mineral. It has previously been considered to be essentially anhydrous. In this study, hydrothermal treatment of silica glass and coesite at 350–550 °C near 10 GPa produces stishovite with significant amounts of H2O in its structure. A combination of methodologies (X-ray diffraction, thermal analysis, oxide melt solution calorimetry, secondary ion mass spectrometry, infrared and nuclear magnetic resonance spectroscopy) indicate the presence of 1.3 ± 0.2 wt % H2O and NMR suggests that the primary mechanism for the H2O uptake is a direct hydrogarnet-like substitution of 4H+ for Si4+, with the protons clustered as hydroxyls around a silicon vacancy. This substitution is accompanied by a substantial volume decrease for the system (SiO2 + H2O), although the stishovite expands slightly, and it is only slightly unfavorable in energy. Stishovite could thus be a host for H2O at convergent plate boundaries, and in other relatively cool high-pressure environments.

    • Thermal spraying is used widely in many industrial sectors in the North European Countries: Finland, Sweden, Norway and Denmark. Important areas where thermal spraying is used petroleum, paper, metals, transport, defence and high-tech machinery industries. In Finland thermal spray technology has wideest use in the pulp and paper industries with several companies spraying various types of rolls and cylinders. In Sweden, thermal spray technology is of great importance in the manufacture of aero engines and in industrial gas turbine applications. In Norway thermal spraying is widely used in various offshore applications, including subsea oil drilling enterprises. Development and innovation in thermal spray technology in the Nordic countries is dominated by research conducted in Finland and Sweden, followed by Denmark and Norway. In particular, research groups from Sweden and Finland, as well as from Norway, have routinely presented their research results at international conferences and in international journals.

    • To investigate associations of morbidity with subsequent sickness absence (SA) and disability pension (DP) among initially nulliparous women with no, one or several childbirths during follow-up.DesignLongitudinal register-based cohort study.SettingSweden.ParticipantsNulliparous women, aged 18 to 39 years and living in Sweden on 31 December 2004 and the three preceding years (n=492 504).Outcome measuresAnnual mean DP and SA days (in SA spells >14 days) in the 3 years before and after inclusion date in 2005.MethodsWomen were categorised into three groups: no childbirth in 2005 nor during the follow-up, first childbirth in 2005 but not during follow-up, and having first childbirth in 2005 and at least one more during follow-up. Microdata were obtained for 3 years before and 3 years after inclusion regarding SA, DP, mortality and morbidity (ie, hospitalisation and specialised outpatient healthcare, also excluding healthcare for pregnancy, childbirth and puerperium). HRs and 95% CIs for SA and DP in year 2 and 3 after childbirth were estimated by Cox regression; excluding those on DP at inclusion.ResultsAfter controlling for study participants’ prior morbidity and sociodemographic characteristics, women with one childbirth had a lower risk of SA and DP than those who remained nulliparous, while women with more than one childbirth had the lowest DP risk. Morbidity after inclusion that was not related to pregnancy, childbirth or the puerperium was associated with a higher risk of future SA and DP, regardless of childbirth group. Furthermore, morbidity both before and after childbirth showed a strong association with SA and DP (HR range: 2.54 to 13.12).ConclusionWe found a strong positive association between morbidity and both SA and DP among women, regardless of childbirth status. Those who gave birth had lower future SA and DP risk than those who did not.

  • Change in Practice of Radioactive Iodine Administration in Differentiated Thyroid Cancer: A Single-Centre Experience

    • <b><i>Objective:</i></b> Our study aimed to analyse temporal trends in radioactive iodine (RAI) treatment for thyroid cancer over the past decade; to analyse key factors associated with clinical decisions in RAI dosing; and to confirm lower activities of RAI for low-risk patients were not associated with an increased risk of recurrence. <b><i>Methods:</i></b> Retrospective analysis of 1,323 patients who received RAI at a quaternary centre in Australia between 2008 and 2018 was performed. Prospectively collected data included age, gender, histology, and American Joint Committee on Cancer stage (7th ed). American Thyroid Association risk was calculated retrospectively. <b><i>Results:</i></b> The median activities of RAI administered to low-risk patients decreased from 3.85 GBq (104 mCi) in 2008–2016 to 2.0 GBq (54 mCi) in 2017–2018. The principal driver of this change was an increased use of 1 GBq (27 mCi) from 1.3% of prescriptions in 2008–2011 to 18.5% in 2017–2018. In patients assigned as low risk per ATA stratification, lower activities of 1 GBq or 2 GBq (27 mCi or 54 mCi) were not associated with an increased risk of recurrence. In patients assigned to intermediate- or high-risk categories who received RAI as adjuvant therapy, there was no difference in risk of recurrence between 4 GBq (108 mCi) and 6 GBq (162 mCi). <b><i>Conclusions:</i></b> Our data demonstrate an evolution of RAI activities consistent with translation of ATA guidelines into clinical practice. Use of lower RAI activities was not associated with an increase in recurrence in low-risk thyroid cancer patients. Our data also suggest lower RAI activities may be as efficacious for adjuvant therapy in intermediate- and high-risk patients.
