  • Complex evolution of Holocene hydroclimate, fire and vegetation revealed by molecular, minerogenic and biogenic proxies, Marais Geluk wetland, eastern Free State, South Africa

    • The Holocene climate history of Southern Africa remains inconclusive despite the increasing number of proxy records from the region. This might be related to the diversity of proxy records, how the proxies are interpreted, or that proxies may respond to more than one forcing (e.g. hydroclimate, fire, temperature.). Here, a 175-cm peat sequence from Free State, South Africa (28°17′53″S, 29°25′10.9″E), was analyzed using a comprehensive set of novel and conventional proxies, including isotopic (δ13C), elemental (CS-XRF), mineral (pXRD), molecular (FTIR-ATR and pyrolysis-GC-MS), grain size (Malvern 3000) and GSSC phytolith composition. The chronology was constructed through AMS radiocarbon dating (n = 7). The early Holocene (10,380–7000 cal yr BP) was characterized by an initial wet phase, followed by relative dryness, at least seasonally, evidenced by slow accumulation rates, low organic content and dominance of terrestrial vegetation in the organic matter matrix. From 7000 cal yr BP, decreasing temperatures, as evidenced in regional climate reconstructions, were associated at Marias Geluk with higher biogenic silica and organic matter content and an increase of moisture-adapted grasses, indicating increasingly mesic conditions. This trend was amplified after 6000 cal yr BP, co-occurring with a southward displacement of the ITCZ. Complex proxy dynamics were observed between 4300 and 2180 cal yr BP, with bulk organic proxies indicating a drier environment (lower carbon content, slow accumulation rates, enriched δ13C values) but the phytolith record pointing towards relatively mesic conditions. The period was also associated with increased fire frequency, that also reached the local wetland. We suggest that the period was associated with seasonally mesic conditions together with increased fire incidence, which affected some of the organic proxies. Increased fire activity was also recorded in the region, while the hydroclimatic indications differed. The last 2000 years, during which human activity is known to have increased in the region, was characterized by lower fire incidence and variable, but relatively moist, conditions. The hydroclimatic inferences for the last 2000 years are in line with previous studies from the region, but additional studies are needed to decipher if the decline in fire incidence was associated to climate forcings, human activities, or a combination of both. The multiproxy approach applied here - in particular the inclusion of FTIR-ATR and pyrolysis GC-MS - revealed a complex interplay between vegetation dynamics, hydrology and paleofire variability. This study confirms that relatively small Holocene temperature variations (compared to northern higher latitudes) were associated with major hydrological variability at Marais Geluk, and reinforces concerns from earlier studies that the hydroclimate of the region is vulnerable to climate change. The result s also show that the southward displacement of the ITCZ, and associated tropical air masses, likely had significant effects on regional hydrology and fire incidence.

  • Inter- and intra-laboratory comparison of a multibiodosimetric approach to triage in a simulated, large scale radiation emergency

    • Purpose: The European Union's Seventh Framework Programme-funded project 'Multi-disciplinary biodosimetric tools to manage high scale radiological casualties' (MULTIBIODOSE) has developed a multiparametric approach to radiation biodosimetry, with a particular emphasis on triage of large numbers of potentially exposed individuals following accidental exposures. In November 2012, an emergency exercise took place which tested the capabilities of the MULTIBIODOSE project partners. The exercise described here had a dual purpose: Intercomparison of (i) three biodosimetric assays, and (ii) the capabilities of the seven laboratories, with regards to provision of triage status for suspected radiation exposed individuals. Materials and methods: Three biological dosimetry tools - the dicentric, micronucleus and gamma-H2AX (the phosphorylated form of member X of histone H2A, in response to DNA double-strand breaks) foci assays - were tested, in addition to provision of the triage status results (low exposure: <1 Gy; medium exposure: 1-2 Gy; high exposure: >2 Gy) by the MULTIBIODOSE software. The exercise was run in two modes: An initial triage categorisation of samples (based on the first dose estimates for each assay received from each laboratory) followed by collation of the full set of estimated doses (all the results from all modes of each assay carried out by the participating laboratories) calculated using as many modes of operation as possible of the different assays developed during the project. Simulated acute whole body and partial body exposures were included. Results: The results of the initial triage categorisation and the full comparison of assays and methods within and between laboratories are presented here. Conclusions: The data demonstrate that the MULTIBIODOSE approach of applying multiparametric tools to radiation emergencies is valid and effective.

  • Geographic variation in fitness-related traits of the bladderwrack Fucus vesiculosus along the Baltic Sea-North Sea salinity gradient

    • In the course of the ongoing global intensification and diversification of human pressures, the study of variation patterns of biological traits along environmental gradients can provide relevant information on the performance of species under shifting conditions. The pronounced salinity gradient, co-occurrence of multiple stressors, and accelerated rates of change make the Baltic Sea and its transition to North Sea a suitable region for this type of study. Focusing on the bladderwrack Fucus vesiculosus, one of the main foundation species on hard-bottoms of the Baltic Sea, we analyzed the phenotypic variation among populations occurring along 2,000km of coasts subjected to salinities from 4 to >30 and a variety of other stressors. Morphological and biochemical traits, including palatability for grazers, were recorded at 20 stations along the Baltic Sea and four stations in the North Sea. We evaluated in a common modeling framework the relative contribution of multiple environmental drivers to the observed trait patterns. Salinity was the main and, in some cases, the only environmental driver of the geographic trait variation in F.vesiculosus. The decrease in salinity from North Sea to Baltic Sea stations was accompanied by a decline in thallus size, photosynthetic pigments, and energy storage compounds, and affected the interaction of the alga with herbivores and epibiota. For some traits, drivers that vary locally such as wave exposure, light availability or nutrient enrichment were also important. The strong genetic population structure in this macroalgae might play a role in the generation and maintenance of phenotypic patterns across geographic scales. In light of our results, the desalination process projected for the Baltic Sea could have detrimental impacts on F.vesiculosus in areas close to its tolerance limit, affecting ecosystem functions such as habitat formation, primary production, and food supply.

    • Cuticular hydrocarbons (CHCs) form the boundary between insects and their environments and often act as essential cues for species, mate, and kin recognition. This complex polygenic trait can be highly variable both among and within species, but the causes of this variation, especially the genetic basis, are largely unknown. In this study, we investigated phenotypic and genetic variation of CHCs in the seaweed fly, Coelopa frigida, and found that composition was affected by both genetic (sex and population) and environmental (larval diet) factors. We subsequently conducted behavioral trials that show CHCs are likely used as a sexual signal. We identified general shifts in CHC chemistry as well as individual compounds and found that the methylated compounds, mean chain length, proportion of alkenes, and normalized total CHCs differed between sexes and populations. We combined these data with whole genome resequencing data to examine the genetic underpinnings of these differences. We identified 11 genes related to CHC synthesis and found population-level outlier SNPs in 5 that are concordant with phenotypic differences. Together these results reveal that the CHC composition of C. frigida is dynamic, strongly affected by the larval environment, and likely under natural and sexual selection.

    • We used immunochemistry to demonstrate expression of acetylcholine's nicotinic 7-receptor subtype inhuman colon cancer cell line HT-29. Moreover, RT-PCR and immunochemistry showed that cholineacetyltransferase and acetylcholine esterase, the enzymes responsible for acetylcholine synthesis anddegradation, respectively, localise in HT-29 cells. Bromoacetylcholine bromide, an inhibitor of cholineacetyltransferase, significantly attenuated basal cell growth. Our findings suggest that acetylcholine mightserve as an autocrine/paracrineor speculatively, even intracrinesignalling molecule in cell line HT-29, thuscontributing to carcinogenesis/cancer progression

    • This is the first report describing the cloning and characterization of sterol carrier protein-2 (SCP-2) from plants. Arabidopsis thaliana SCP-2 (AtSCP-2) consists of 123 amino acids with a molecular mass of 13.6 kDa. AtSCP-2 shows 35% identity and 56% similarity to the human SCP-2-like domain present in the human D-bifunctional protein (DBP) and 30% identity and 54% similarity to the human SCP-2 encoded by SCP-X. The presented structural models of apo-AtSCP-2 and the ligand-bound conformation of AtSCP-2 reveal remarkable similarity with two of the structurally known SCP-2s, the SCP-2-like domain of human DBP and the rabbit SCP-2, correspondingly. The AtSCP-2 models in both forms have a similar hydrophobic ligand-binding tunnel, which is extremely suitable for lipid binding. AtSCP-2 showed in vitro transfer activity of BODIPY-phosphatidylcholine (BODIPY-PC) from donor membranes to acceptor membranes. The transfer of BODIPY-PC was almost completely inhibited after addition of 1-palmitoyl 2-oleoyl phosphatidylcholine or ergosterol. Dimyristoyl phosphatidic acid, stigmasterol, steryl glucoside, and cholesterol showed a moderate to marginal ability to lower the BODIPY-PC transfer rate, and the single chain palmitic acid and stearoyl-coenzyme A did not affect transfer at all. Expression analysis showed that AtSCP-2 mRNA is accumulating in most plant tissues. Plasmids carrying fusion genes between green fluorescent protein and AtSCP-2 were transformed with particle bombardment to onion epidermal cells. The results from analyzing the transformants indicate that AtSCP-2 is localized to peroxisomes.

    • We determine the detailed differences in geometry and band structure between wurtzite (Wz) and zinc blende (Zb) InAs nanowire (NW) surfaces using scanning tunneling microscopy/spectroscopy and photoemission electron microscopy. By establishing unreconstructed and defect-free surface facets for both Wz and Zb, we can reliably measure differences between valence and conduction band edges, the local vacuum levels, and geometric relaxations to the few-millielectronvolt and few-picometer levels, respectively. Surface and bulk density functional theory calculations agree well with the experimental findings and are used to interpret the results, allowing us to obtain information on both surface and bulk electronic structure. We can thus exclude several previously proposed explanations for the observed differences in conductivity of Wz-Zb NW devices. Instead, fundamental structural differences at the atomic scale and nanoscale that we observed between NW surface facets can explain the device behavior.

  • Spatial Control of Multiphoton Electron Excitations in InAs Nanowires by Varying Crystal Phase and Light Polarization

    • We demonstrate the control of multiphoton electron excitations in InAs nanowires (NWs) by altering the crystal structure and the light polarization. Using few-cycle, near-infrared laser pulses from an optical parametric chirped-pulse amplification system, we induce multiphoton electron excitations in InAs nanowires with controlled wurtzite (WZ) and zincblende (ZB) segments. With a photoemission electron microscope, we show that we can selectively induce multiphoton electron emission from WZ or ZB segments of the same wire by varying the light polarization. Developing ab initio GW calculations of first to third order multiphoton excitations and using finite-difference time-domain simulations, we explain the experimental findings: While the electric-field enhancement due to the semiconductor/vacuum interface has a similar effect for all NW segments, the second and third order multiphoton transitions in the band structure of WZ InAs are highly anisotropic in contrast to ZB InAs. As the crystal phase of NWs can be precisely and reliably tailored, our findings open up for new semiconductor optoelectronics with controllable nanoscale emission of electrons through vacuum or dielectric barriers.

  • Learning at Work and Productivity in Swedish Business Firms : Based on the Swedish MEADOW Survey 2009/2010

    • The background of the thesis is the ongoing struggle for better understanding of driving forces for economic growth. Some important and rewarding new steps have been taken the last decades in the research of economic growth theory. For example, the theory includes not only the use of technology but also the creation and implementation of new technology entwined with management and learning practices at work. However, the development of knowledge on the matter can be described as slow. The explanation is argued to be the lack of data, which complicates and prevents the development of theory. The lack of data and the intertwined dependence with the development of theory in the matter constitutes the starting point for the research in the thesis.The purpose of this thesis is to contribute to the development of measures of management of work and learning practices at work that can be used in productivity models. The analyses are based on the micro-level data from the Swedish MEADOW Survey 2009/2010, at Statistics Sweden. Factor and regression analysis are performed, other analytical techniques are also used.Some key findings of how management and learning practices at work can be described and measured show that the business firms use the different practices in bundles. The findings indicate synergy effects on performance when more than one management and learning practice is applied. Two different management approaches are tested, both models predict productivity. However, there is a difference in the productivity model a such when it includes the two management approaches; the measure of the contemporary use has a better fitness then the theoretical model. Focus and delimitations of research questions in the forthcoming doctoral dissertation are discussed, see Future Research – the Logic Next Step, in Chapter 6. Final Reflections.

    • In recent years it has become increasingly clear that chemical interactions play a fundamental role in aquatic habitats and have far-reaching evolutionary and ecological consequences. A plethora of studies have shown that aquatic organisms from most taxa and functional groups respond to minute concentrations of chemical substances released by other organisms. However, our knowledge of this 'chemical network' is still negligible. Chemical interactions can be divided into two larger sub-areas based on the function of the chemical substance. First, there are interactions where chemical substances are toxic to other organisms and are used as a defense against consumers (including both herbivores and predators) or a weapon against competitors (allelopathy). Second, chemical substances mey be used as a source for information of the environment; for example: how can I find the optimal habitat, the best food, the nicest partner, and avoid being eaten? Aquatic organisms are able to detect and respond to extremely low concentrations of chemical cues to answer all these questions. The book aims at connecting these intriguing chemical interactions with traditional knowledge of organism interactions. Chemical ecology in aquatic systems covers a wide range of studies, both plant and animal, from different geographic regions and habitats-pelagic as well as benthic. Most of the chemical interactions are similar in freshwater and marine habitats and this book therefor strives at integrating work on both systems. This accessible, research-level text is aimed at graduate students and professional researchers in the fields of limnology, marine ecology, evolutionary biology, behavioral ecology, and chemical ecology.

    • The central Arctic Ocean (CAO) plays an important role in the global carbon cycle, but the current and future exchange of the climate-forcing trace gases methane (CH4) and carbon dioxide (CO2) between the CAO and the atmosphere is highly uncertain. In particular, there are very few observations of near-surface gas concentrations or direct air-sea CO2 flux estimates and no previously reported direct air-sea CH4 flux estimates from the CAO. Furthermore, the effect of sea ice on the exchange is not well understood. We present direct measurements of the air-sea flux of CH4 and CO2, as well as air-snow fluxes of CO2 in the summertime CAO north of 82.5 N from the Synoptic Arctic Survey (SAS) expedition carried out on the Swedish icebreaker Oden in 2021. Measurements of air-sea CH4 and CO2 flux were made using floating chambers deployed in leads accessed from sea ice and from the side of Oden, and air-snow fluxes were determined from chambers deployed on sea ice. Gas transfer velocities determined from fluxes and surface-water-dissolved gas concentrations exhibited a weaker wind speed dependence than existing parameterisations, with a median sea-ice lead gas transfer rate of 2.5cmh-1 applicable over the observed 10m wind speed range (1-11ms-1). The average observed air-sea CO2 flux was -7.6mmolm-2d-1, and the average air-snow CO2 flux was -1.1mmolm-2d-1. Extrapolating these fluxes and the corresponding sea-ice concentrations gives an August and September flux for the CAO of -1.75mmolm-2d-1, within the range of previous indirect estimates. The average observed air-sea CH4 flux of 3.5μmolm-2d-1, accounting for sea-ice concentration, equates to an August and September CAO flux of 0.35μmolm-2d-1, lower than previous estimates and implying that the CAO is a very small (‰ 1%) contributor to the Arctic flux of CH4 to the atmosphere.

  • Solid phase extraction and metabolic profiling of exudates from living copepods

    • Copepods are ubiquitous in aquatic habitats. They exude bioactive compounds that mediate mate finding or induce defensive traits in prey organisms. However, little is known about the chemical nature of the copepod exometabolome that contributes to the chemical landscape in pelagic habitats. Here we describe the development of a closed loop solid phase extraction setup that allows for extraction of exuded metabolites from live copepods. We captured exudates from male and female Temora longicornis and analyzed the content with high resolution LC-MS. Chemometric methods revealed 87 compounds that constitute a specific chemical pattern either qualitatively or quantitatively indicating copepod presence. The majority of the compounds were present in both female and male exudates, but nine compounds were mainly or exclusively present in female exudates and hence potential pheromone candidates. Copepodamide G, known to induce defensive responses in phytoplankton, was among the ten compounds of highest relative abundance in both male and female extracts. The presence of copepodamide G shows that the method can be used to capture and analyze chemical signals from living source organisms. We conclude that solid phase extraction in combination with metabolic profiling of exudates is a useful tool to develop our understanding of the chemical interplay between pelagic organisms.

    • Sterol carrier protein-2 (SCP-2) is a small intracellular basic protein domain implicated in peroxisomal beta-oxidation. We extend our knowledge of plant SCP-2 by characterizing SCP-2 from Euphorbia lagascae. This protein consists of 122 amino acids including a PTS1 peroxisomal targeting signal. It has a molecular mass of 13.6 kDa and a pI of 9.5. It shares 67% identity and 84% similarity with SCP-2 from Arabidopsis thaliana. Proteomic analysis revealed that E. lagascae SCP-2 accumulates in the endosperm during seed germination. It showed in vitro transfer activity of BODIPY-phosphatidylcholine (BODIPY-PC). The transfer of BODIPY-PC was almost completely inhibited after addition of phosphatidylinositol, palmitic acid, stearoyl-CoA and vernolic acid, whereas sterols only had a very marginal inhibitory effect. We used protein modelling and site-directed mutagenesis to investigate why the BODIPY-PC transfer mediated by E. lagascae SCP-2 is not sensitive to sterols, whereas the transfer mediated by A. thaliana SCP-2 shows sterol sensitivity. Protein modelling suggested that the ligand-binding cavity of A. thaliana SCP-2 has four methionines (Met12, 14, 15 and 100), which are replaced by leucines (Leu11, 13, 14 and 99) in E. lagascae SCP-2. Changing Leu99 to Met99 was sufficient to convert E. lagascae SCP-2 into a sterol-sensitive BODIPY-PC-transfer protein, and correspondingly, changing Met100 to Leu100 abolished the sterol sensitivity of A. thaliana SCP-2.
