    • Pancreatic islets implanted heterotopically (i.e., into the kidney, spleen, or liver) become poorly revascularized following transplantation. We hypothesized that islets implanted into the pancreas Would become better revascularized. Islets isolated from transgenic mice expressing enhanced yellow fluorescent protein (EYFP) in all somatic cells were Cultured before they were implanted into the pancreas or beneath the renal capsule of athymic mice. Vascular density was evaluated in histological sections 1 month posttransplantation. EYFP was used as reporter for the transgene to identify the transplanted islets. Islet endothelial cells were visualized by staining with the lectin Bandeiraea simplicifolia (BS-1). Capillary   numbers in intrapancreatically implanted islets were only slightly lower than those counted in endogenous islets, whereas islets implanted   beneath the renal capsule had a markedly lower vascular density. In order to determine if this high graft vascular density at the   intrapancreatic site reflected expansion of remnant donor endothelial  cells or increased ingrowth of blood vessels from the host. also islets  from Tie2-green fluorescent protein (GFP) mice (i.e., islets with fluorescent endothelial cells) were transplanted into the pancreas or beneath the renal capsule of athymic mice. These islet grafts revealed that the new vascular structures formed in the islet grafts contained very few GFP-positive cells, and thus mainly were of recipient origin. The reason(s) for the much better ingrowth of blood vessels at the intrapancreatic site merits further studies, because this may help us form strategies to overcome the barrier for ingrowth of host vessels also into islets in heterotopic implantation sites.

  • Implantation site-dependent dysfunction of transplanted pancreatic islets

    • OBJECTIVE—Clinical islet transplantations are performed through infusion of islets via the portal vein into the liver. This study aimed at characterizing the influence of the implantation microenvironment on islet graft metabolism and function. RESEARCH DESIGN AND METHODS—Islets were transplanted into their normal environment, i.e., the pancreas, or intraportally into the liver of mice. One month posttransplantation, the transplanted islets were retrieved and investigated for changes in function and gene expression. RESULTS—Insulin content, glucose-stimulated insulin release, (pro)insulin biosynthesis, and glucose oxidation rate were markedly decreased in islets retrieved from the liver, both when compared with islets transplanted into the pancreas and endogenous islets. Islets transplanted into the pancreas showed normal insulin content, (pro)insulin biosynthesis, and glucose oxidation rate but increased basal insulin secretion and impaired glucose stimulation index. Gene expression data for retrieved islets showed downregulation of pancreatic and duodenal homeobox gene-1, GLUT-2, glucokinase, mitochondrial glycerol-phosphate dehydrogenase, and pyruvate carboxylase, preferentially in intraportally transplanted islets. CONCLUSIONS—Islets transplanted into their normal microenvironment, i.e., the pancreas, display gene expression changes when compared with endogenous islets but only moderate changes in metabolic functions. In contrast, site-specific properties of the liver markedly impaired the metabolic functions of intraportally transplanted islets.

    • Background and purpose: Health related quality of life (HRQoL) was assessed in the randomised, prospective ARTSCAN study comparing conventional radiotherapy (CF) with accelerated radiotherapy (AF) for head and neck cancer. Material and methods: 750 patients with squamous cell carcinoma (of any grade and stage) in the oral cavity, oro-, or hypopharynx or larynx (except T1-2, NO glottic carcinoma) without distant metastases were randomised to either conventional fractionation (2 Gy/day, 5 days/week in 49 days, total dose 68 Gy) or accelerated fractionation (1.1 + 2.0 Gy/day, 5 days/week in 35 days, total dose 68 Gy). HRQoL was assessed with EORTC QLQ-C30, QLQ-H&N35 and HADS at baseline, at end of radiotherapy (eRT) and at 3 and 6 months and 1, 2 and 5 years after start of treatment. Results: The AF group reported HRQoL was significantly lower at eRT and at 3 months for most symptoms, scales and functions. Few significant differences were noted between the groups at 6 months and 5 years. Scores related to functional oral intake never reached baseline. Conclusion: In comparison to CF, AF has a stronger adverse effect on HRQoL in the acute phase.

  • Distribution of intraportally implanted microspheres and fluorescent islets in mice

    • The aim of the study was to evaluate the distribution of intraportally transplanted islets in mice. We initially administered 2000 polystyrene microspheres with a diameter of 50 microm intraportally into normoglycemic C57BL/6 mice. In separate experiments other mice were injected similarly with 300 microspheres each with a diameter of 100 or 200 microm. One week later the animals were killed, and the lungs and livers were removed and divided into lobes. The number of microspheres in each individual liver lobe and in the lungs was counted using a stereomicroscope. In other experiments, athymic C57BL/6 mice were similarly implanted with 250 islets isolated from transgenic mice expressing the enhanced yellow fluorescent protein in the islet cells. The distribution of microspheres and islets was independent of size, and fairly homogenous within the liver, with the exception of the caudate lobe, which contained fewer microspheres and islets, respectively. Approximately one third of all microspheres and islets were present as aggregates. Eighty-five to 90% of the implanted microspheres were identified in the liver sections, whereas 60-65% of the implanted islets were recovered. Aggregates or single fluorescent cells were observed in the liver of islet-implanted mice. We conclude that islets and microspheres implanted into the liver distribute fairly homogenously and quite a few of them exist as aggregates or, with respect to islets, as fragments.

    • All manufacturing processes have variation which may violate the fulfillment of assembly, functional, geometrical or esthetical requirements and difficulties to reach desired form in all areas. The cost for geometry defects rises downstream in the process chain. Therefore, it is vital to discover these defects as soon as they appear. Then adjustments can be done in the process without losing products or time. In order to find a solution for this, a project with the overall scope “development of an intelligent process control system” has been initiated. This project consists of five different work packages: Inline measurement, Process Evaluation, Corrective actions, Flexible tooling and demonstrator cell. These work packages address different areas which are necessary to fulfill the overall scope of the project. The system shall both be able to detect geometrical defects, propose adjustments and adjust simple process parameters. The results are demonstrated in a demo cell located at Chalmers University of Technology. In the demonstrator all the different areas have been verified in an industrial case study – assembly of GOR Volvo S80. Efficient offline programming for robot based measurement, efficient process evaluation based on case base reasoning (CBR) methodology, flexible fixtures and process adjustments based on corrective actions regarding in going part positioning.

  • Play with Science in Inquiry Based Science Education

    • In science education students sometimes engage in imaginary science-oriented play where ideas about science and scientists are put to use. Through play, children interpret their experiences, dramatize, give life to and transform what they know into a lived narrative. In this paper we build on the work of Vygotsky on imagination and creativity. Previous research on play in primary and secondary school has focused on play as a method for formal instruction rather than students’ spontaneous informal play. Our aim is to explore students’ informal play as part of activity in lower secondary school science. The empirical study was conducted as part of a larger study on learning, narrative knowing and remembering in inquiry based science education in two Swedish compulsory schools. Data were collected during 10 weeks using video- and audiotape recordings. The results show that the studied classroom practices offer students a variety of resources that may be used to engage in spontaneous play with science. We discern two categories of play: role-playing and game-playing involving transformations of classroom tasks and opening up for different social positioning in relation to science. Implications for teaching science is that teachers, in order to promote students learning about science as a cultural endeavor and about themselves in relation to science and scientist communities may encourage students’ informal play.

  • Students' Ways of Using Prior Experience in Inquiry Based Science Education – the case of NTA

    • Inquiry has been identified as a 'key-approach' and recommended as an important pedagogy to improve primary science education in Europe. Inquiry is also a key component in the US national science education standards. In Sweden, inquiry pedagogy in primary science has been introduced through a large curriculum program called NTA (Science and Technology for all). The aim of this paper is to shed light on what inquiry based science education (IBSE) practices are emerging in Swedish primary schools through the introduction of the NTA-program and what activities students are engaging in, in these practices. In particular we focus on ways of working with 'what is already known' which is held to be a central aspect to IBSE in e.g. the definition of inquiry in the US national science education standards. The research questions investigated are: In what actions do students engage in when working with ‘what is already known’ in IBSE practice? How is 'what is already known' acted with in practice? Data was collected, during a 10-week-period using video- and audiotape recordings of classroom work, in two 6th grade classes where teachers and students worked with an NTA-unit called 'the Chemistry of food'. Our analysis is conceptualized within a cultural-historical activity-theoretical perspective. Results show that students participate in three different actions: A) separation of tasks, B) playing the lottery and C) challenging test results with ‘what is already known’. These different actions involve different types of use of 'what is already known' , and the actions correspond to, and acquire meaning in relation to, different goals. Common to the different ways of working is that we can discern two different types of ‘what is already known’: (1) referring to facts, and (2) making use of ideas about science culture.

    • Preclinical trials of cancer drugs in animal models are important for drug development. The Rip1Tag2 (RT2) transgenic mouse, a model of pancreatic neuroendocrine tumours (PNET), has provided immense knowledge about PNET biology, although tumour progression occurs in a location inaccessible for real-time monitoring. To overcome this hurdle we have developed a novel platform for intravital 3D imaging of RT2 tumours to facilitate real-time studies of cancer progression. Pre-oncogenic islets retrieved from RT2 mice were implanted into the anterior chamber of the eye (ACE) of host mice, where they engrafted on the iris, recruited blood vessels and showed continuous growth. Noninvasive confocal and two-photon laser-scanning microscopy through the transparent cornea facilitated high-resolution imaging of tumour growth and angiogenesis. RT2 tumours in the ACE expanded up to 8-fold in size and shared hallmarks with tumours developing in situ in the pancreas. Genetically encoded fluorescent reporters enabled high-resolution imaging of stromal cells and tumour cell migration. Sunitinib treatment impaired RT2 tumour angiogenesis and growth, while overexpression of the vascular endothelial growth factor (VEGF)-B increased tumour angiogenesis though tumour growth was impaired. In conclusion, we present a novel platform for intravital high-resolution and 3D imaging of PNET biology and cancer drug assessment.

    • BackgroundCurrent prognostic markers cannot adequately predict the clinical outcome of breast cancer patients. Therefore, additional biomarkers need to be included in routine immune panels. FOXA1 was a significant predictor of favorable outcome in primary breast cancer, while Nestin expression is preferentially found in triple-negative tumors with increased rate of nodal metastases, and reduced survival. No studies have investigated the prognostic value of FOXA1 and Nestin expression in breast cancer metastases.MethodsBreast cancer metastases (n=164) from various anatomical sites were retrospectively analyzed by immunohistochemistry for FOXA1, Nestin and GATA3 expression. Cox regression analysis assessed the prognostic value of FOXA1 and Nestin expression.ResultsIn breast cancer metastases, FOXA1 expression was associated with Nestin-negativity, GATA3-positivity, ER-positivity, HER2-positivity and non-triple-negative status (P<0.05). In contrast, Nestin expression was associated with FOXA1-negative, GATA3-negative, ER-negative, and triple-negative metastases (P<0.05). Univariate Cox regression analysis showed FOXA1 expression was predictive of overall survival (OS, P=0.00048) and metastasis-free survival (DMFS, P=0.0011), as well as, distant metastasis-free survival in ER-positive patients (P=0.036) and overall survival in ER-negative patients (P=0.024). Multivariate analysis confirmed the significance of FOXA1 for both survival endpoints in metastatic breast cancer patients (OS, P=0.0033; DMFS, P=0.015).ConclusionsIn our study, FOXA1 was expressed mostly in ER-positive breast cancer metastases. Expression of Nestin was related to triple-negative metastases, where brain was the most frequent metastatic site. These findings highlight the clinical utility of FOXA1 and Nestin expression and warrant their inclusion in routine immunohistochemical panels for breast carcinoma.

    • Collaborative robots becomes more and more common in lab environment and soon also in industry. In order to create resource- and volume flexibility, dynamic and smart automation could be seen as an answer. This paper has investigated the collaborative robots UR3 and UR5 for O-ring assembly and final assembly, compared to the current state which is performed manually. The methodology Dynamo++ was used for measurement and analysis in terms of LoA (cognitive and physical), cycle-time and quality. Furthermore, automation strategy, safety and easiness of programming was investigated. Results show that collaborative robots have great potential in the middle product volume area. A lot of time, layout space and money could be saved with these solutions. However, standards and safety has to be investigated further in order to reach its fully potential

    • Predictive maintenance (PdM) using Machine learning (ML) is a top-rated business case with respect to the availability of data and potential business value for future sustainability and competitiveness in the manufacturing industry. However, applying ML within actual industrial practice of PdM is a complex and challenging task due to high dimensionality and lack of labeled data. To cope with this challenge, this paper presents a systematic framework based on an unsupervised ML approach by aiming to construct health indicators, which has a crucial impact on making the data meaningful and usable for monitoring machine performance (health) in PdM applications. The results are presented by using real-world industrial data coming from a manufacturing company. In conclusion, the designed health indicators can be used to monitor machine performance over time and further be used in a supervised setting for the purpose of prognostic like remaining useful life estimation in implementing PdM in the industry.

    • Using observations, experiments and model simulations we investigate the possibility and range of long-distance dispersal by seed rafting in eelgrass, Zostera marina, at the Swedish west coast. A field experiment showed that the breaking strength of post-flowering rhipidia decreased significantly over a 30-day period, indicating that ontogenetic changes of the morphology facilitate dislodgement of the rhipidia at the optimal time for dispersal. We recorded positive buoyancy of detached post-flowering rhipidia for at least 26 days. Finally, we found evidence for sexual reproduction via seeds in the study area. These results, together with field measurements of wind-driven surface-transport velocity of rhipidia and wind data from 7 years, allowed us to model a dispersal potential of up to 150 km in one season along the Swedish west coast, which corresponds well with published estimates of dispersal potential from both observational and genetic studies. The results add to evidence that rafting of seeds on floating reproductive shoots is an important dispersal strategy for eelgrass, allowing for colonisation of new areas and for recolonisation after die-back events.   • Stepwise function of natural growth for Scylla serrata in East Africa : a valuable tool for assessing growth of mud crabs in aquaculture

    • Predicting growth is critical in aquaculture, but models of growth are largely missing for mud crab species. Here, we present the first model of natural growth in juvenile and adult mud crabs Scylla serrata from East Africa using a stepwise growth function based on data on intermoult periods and growth at moult from field mark-recapture, pond and laboratory studies. The results showed a sigmoid growth pattern in carapace width and suggest that S.serrata in East Africa will reach 300g and sexual maturity similar to 9.9months after settlement, and a commercial size of 500g after 12.4months. Analyses of the literature identified several issues with the common praxis to compare standard growth measures between aquaculture studies with different initial size or growing periods. Using the new growth function to estimate the proportional difference between modelled and obtained growth as an alternative method, we show that growth rates of S.serrata cultured in cage systems, which are dominant in East Africa, was <40% of the estimated natural growth and growth obtained in pond systems. The analysis also indicated that growth rates of S.serrata in Southeast Asia was over 50% higher compared with similar culture systems in East Africa, and that different species of mud crabs had large differences in growth rates. This study shows that growth in the present mud crab aquaculture systems in East Africa is below their expected potential. Further work is needed to identify the factors behind this observation.

    • Seagrasses have declined in many places around the world, and the Swedish Skagerrak coast is no exception. Between the 1980s and 2000, the cover of eelgrass (Zostera marina L.) on the Swedish Skagerrak coast decreased about 60%. In the present study, the sites that were investigated in the 1980s and 2000 were revisited (1655 ha) in 2003 and 2004 to investigate long and short term temporal and spatial differences in eelgrass coverage. The distribution of eelgrass was mapped from a small boat using an aquascope and a GPS receiver. No variation in total (about 150 km) or regional (about 20 km) eelgrass cover was found between 2000 and 2004, but there were substantial changes (both losses and gains) in meadow size on a local (less than about 2 km) scale. The results have implications for management of shallow soft bottoms, particularly because it was shown that even though no eelgrass is found in a single survey in a specific area, there could be eelgrass growing there in the following years.

Resultatet av dessa fältundersökningar har sedan använts i en modell som kan användas för att studera konsekvenser av mänskliga störningar och av olika strategier för att skydda dessa områden.

  • Genetic alterations associated with multiple primary malignancies.

    • Breast cancer (BC) patients are frequently at risk of developing other malignancies following treatment. Although studies have been conducted to elucidate the etiology of multiple primary malignancies (MPM) after a BC diagnosis, few studies have investigated other previously diagnosed primary malignancies (OPPM) before BC. Here, genome-wide profiling was used to identify potential driver DNA copy number alterations and somatic mutations that promote the development of MPMs. To compare the genomic profiles for two primary tumors (BC and OPPM) from the same patient, tumor pairs from 26 young women (≤50years) diagnosed with one or more primary malignancies before breast cancer were analyzed. Malignant melanoma was the most frequent OPPM, followed by gynecologic- and hematologic malignancies. However, significantly more genetic alterations were detected in BC compared to the OPPM. BC also showed more genetic similarity as a group than the tumor pairs. Clonality testing showed that genetic alterations on chromosomes 1, 3, 16, and 19 were concordant in both tumors in 13 patients. TP53 mutations were also found to be prevalent in BC, MM, and HM. Although all samples were classified as genetically unstable, chromothripsis-like patterns were primarily observed in BC. Taken together, few recurrent genetic alterations were identified in both tumor pairs that can explain the development of MPMs in the same patient. However, larger studies are warranted to further investigate key driver mutations associated with MPMs.

  • Metachronous and synchronous occurrence of 5 primary malignancies in a female patient between 1997 and 2013 : A case report with germline and somatic genetic analysis

    • The number of patients with multiple primary malignancies has been increasing steadily in recent years. In the present study, we describe a unique case of an 81-year-old woman with 5 metachronous and synchronous primary malignant neoplasms. The patient was first diagnosed with an endometrium adenocarcinoma in 1997 and a colon adenocarcinoma in 2002. Eleven years after her colon surgery, in 2013, the patient presented with 3 other primary malignancies within a 4-month time span: An invasive malignant melanoma on the lower leg, an invasive mucinous breast carcinoma in the right breast, and a pleomorphic spindle cell sarcoma on the left upper arm. Subsequent routine medical checkups in 2013-2017 revealed no metastases of the primary malignancies. The patient mentioned a familial aggregation of malignant tumors, including 2 sisters with breast cancer and a brother with lung cancer. Interestingly, next-generation sequencing analysis of the patient's blood sample detected no mutations in the BRCA1, BRCA2, TP53, PTEN, CDH1, PALB2, RAD51C, RAD51D, MLH1, MSH2, MSH6, PMS2, EPCAM, APC, MUTYH, STK11, BMPR1A, SMAD4, PTEN, POLE, POLD1, GREM1, and GALNT12 genes. Therefore, whole genome sequencing is warranted to identify cancer-related genetic alterations in this patient with quintuple primary malignancies.

    • Multiple primary malignancies (MPMs) caused by breast cancer treatment are well described, but only few studies to date describe which other previous primary malignancies (OPPMs) occur before breast cancer. The purpose of the present study was to evaluate the prevalence of OPPMs in patients with breast cancer between 2007 and 2018 in Western Sweden.Patient selection was performed using both pathology reports at Sahlgrenska University Hospital (Sweden) and the Swedish Cancer Registry. All newly diagnosed breast cancer patients were screened for presence of OPPM.In total, 8031 breast cancer patients were diagnosed at Sahlgrenska University Hospital between 2007 and 2018. The prevalence of breast cancer patients with OPPMs (n=414) increased from on average 2.6% to 8.2% during this 12-year period and ranged from 17 to 59 patients annually.The most striking increase in prevalence was found among the gynecological tumors (endometrium and ovarian adenocarcinomas), malignant melanomas and gastrointestinal malignancies.These findings were validated using data of the Swedish Cancer Registry.The overall survival rates for cancer patients have improved tremendously during the past 40years, in part due to individually tailored therapies and screening programs. Our study revealed an increasing trend of OPPMs in breast cancer patients.

    • Research on the development of preterm infant feeding behavior has focused mainly on bottlefeeding, using invasive methods or observations by professionals. In this study, a clinical method for observing breastfeeding was developed in collaboration between observers and mothers for the purpose of enabling neonatal personnel and mothers to describe developmental stages in preterm infant breastfeeding behavior. Tests of interobserver reliability resulted in acceptable agreement between observers, but a somewhat lower level of agreement between observers and mothers. The scale showed a good capacity to discriminate between infant gestational ages and can be used for helping mothers to identify their infants' emerging competence.

The dynamical systems of this kind, having attracting fixed points, can in a natural way be divided into equivalence classes, and we investigate whether something can be said about the number of those equivalence classes, for a certain degree of the polynomial g(x).

    • Recent development in the predictive maintenance field has focused on incorporating artificial intelligence techniques in the monitoring and prognostics of machine health. The current predictive maintenance applications in manufacturing are now more dependent on data-driven Machine Learning algorithms requiring an intelligent and effective analysis of a large amount of historical and real-time data coming from multiple streams (sensors and computer systems) across multiple machines. Therefore, this article addresses issues of data pre-processing that have a significant impact on generalization performance of a Machine Learning algorithm. We present an intelligent approach using unsupervised Machine Learning techniques for data pre-processing and analysis in predictive maintenance to achieve qualified and structured data. We also demonstrate the applicability of the formulated approach by using an industrial case study in manufacturing. Data sets from the manufacturing industry are analyzed to identify data quality problems and detect interesting subsets for hidden information. With the approach formulated, it is possible to get the useful and diagnostic information in a systematic way about component/machine behavior as the basis for decision support and prognostic model development in predictive maintenance.

    • Habitat structural complexity affects the behaviour and physiology of individuals, and responses to the environment can be immediate or influence performance later in life through delayed effects. Here, we investigated how structural enrichment, both pre-release in the hatchery rearing environment and post-release in the wild, influenced winter growth and site fidelity of brown trout stocked into side channels of a regulated river. Experiencing structural enrichment in the rearing environment during 3 months in autumn had no pre-release effect on growth, but a delayed positive effect after release during the subsequent winter. Moreover, trout recaptured in wood-treated sections of the side channels had grown more than trout recaptured in control sections. Wood enrichment in the side channels also increased overwinter site fidelity. These results show that adding structure during a relatively short period may alter growth trajectories, and adding wood to side channels is a cost-effective method to enhance winter habitat carrying capacity for juvenile salmonids in regulated rivers.
