    • We compute and investigate four types of imprint of a stochastic background of primordial magnetic fields (PMFs) on the cosmic microwave background (CMB) anisotropies: the impact of PMFs on the CMB temperature and polarization spectra, which is related to their contribution to cosmological perturbations; the effect on CMB polarization induced by Faraday rotation; the impact of PMFs on the ionization history; magnetically-induced non-Gaussianities and related non-zero bispectra; and the magnetically-induced breaking of statistical isotropy. We present constraints on the amplitude of PMFs that are derived from different Planck data products, depending on the specific effect that is being analysed. Overall, Planck data constrain the amplitude of PMFs to less than a few nanoGauss, with different bounds that depend on the considered model. In particular, individual limits coming from the analysis of the CMB angular power spectra, using the Planck likelihood, are B-1 (Mpc) < 4.4 nG (where B1 Mpc is the comoving field amplitude at a scale of 1 Mpc) at 95% confidence level, assuming zero helicity. By considering the Planck likelihood, based only on parity-even angular power spectra, we obtain B-1 (Mpc) < 5.6 nG for a maximally helical field. For nearly scale-invariant PMFs we obtain B-1 (Mpc) < 2.0 nG and B-1 (Mpc) < 0.9 nG if the impact of PMFs on the ionization history of the Universe is included in the analysis. From the analysis of magnetically-induced non-Gaussianity, we obtain three different values, corresponding to three applied methods, all below 5 nG. The constraint from the magnetically-induced passive-tensor bispectrum is B-1 (Mpc) < 2.8 nG. A search for preferred directions in the magnetically-induced passive bispectrum yields B-1 (Mpc) < 4.5 nG, whereas the compensated-scalar bispectrum gives B-1 (Mpc) < 3 nG. The analysis of the Faraday rotation of CMB polarization by PMFs uses the Planck power spectra in EE and BB at 70 GHz and gives B-1 (Mpc) < 1380 nG. In our final analysis, we consider the harmonic-space correlations produced by Alfven waves, finding no significant evidence for the presence of these waves. Together, these results comprise a comprehensive set of constraints on possible PMFs with Planck data.

  • Planck intermediate results XL. The Sunyaev-Zeldovich signal from the Virgo cluster

    • The Virgo cluster is the largest Sunyaev-Zeldovich (SZ) source in the sky, both in terms of angular size and total integrated flux. Planck's wide angular scale and frequency coverage, together with its high sensitivity, enable a detailed study of this big object through the SZ effect. Virgo is well resolved by Planck, showing an elongated structure that correlates well with the morphology observed from X-rays, but extends beyond the observed X-ray signal. We find good agreement between the SZ signal (or Compton parameter, y(c)) observed by Planck and the expected signal inferred from X-ray observations and simple analytical models. Owing to its proximity to us, the gas beyond the virial radius in Virgo can be studied with unprecedented sensitivity by integrating the SZ signal over tens of square degrees. We study the signal in the outskirts of Virgo and compare it with analytical models and a constrained simulation of the environment of Virgo. Planck data suggest that significant amounts of low-density plasma surround Virgo, out to twice the virial radius. We find the SZ signal in the outskirts of Virgo to be consistent with a simple model that extrapolates the inferred pressure at lower radii, while assuming that the temperature stays in the keV range beyond the virial radius. The observed signal is also consistent with simulations and points to a shallow pressure profile in the outskirts of the cluster. This reservoir of gas at large radii can be linked with the hottest phase of the elusive warm/hot intergalactic medium. Taking the lack of symmetry of Virgo into account, we find that a prolate model is favoured by the combination of SZ and X-ray data, in agreement with predictions. Finally, based on the combination of the same SZ and X-ray data, we constrain the total amount of gas in Virgo. Under the hypothesis that the abundance of baryons in Virgo is representative of the cosmic average, we also infer a distance for Virgo of approximately 18 Mpc, in good agreement with previous estimates.

  • Planck intermediate results XLIV. Structure of the Galactic magnetic field from dust polarization maps of the southern Galactic cap

    • Using data from the Planck satellite, we study the statistical properties of interstellar dust polarization at high Galactic latitudes around the south pole (b < -60 degrees). Our aim is to advance the understanding of the magnetized interstellar medium (ISM), and to provide a modelling framework of the polarized dust foreground for use in cosmic microwave background (CMB) component-separation procedures. We examine the Stokes I, Q, and U maps at 353 GHz, and particularly the statistical distribution of the polarization fraction (p) and angle (Psi), in order to characterize the ordered and turbulent components of the Galactic magnetic field (GMF) in the solar neighbourhood. The Q and U maps show patterns at large angular scales, which we relate to the mean orientation of the GMF towards Galactic coordinates (l(0); b(0)) = (70 degrees +/- 5 degrees, 24 degrees +/- 5 degrees). The histogram of the observed p values shows a wide dispersion up to 25%. The histogram Psi of has a standard deviation of 12 degrees about the regular pattern expected from the ordered GMF. We build a phenomenological model that connects the distributions of p and Psi to a statistical description of the turbulent component of the GMF, assuming a uniform effective polarization fraction (p(0)) of dust emission. To compute the Stokes parameters, we approximate the integration along the line of sight (LOS) as a sum over a set of N independent polarization layers, in each of which the turbulent component of the GMF is obtained from Gaussian realizations of a power-law power spectrum. We are able to reproduce the observed p and distributions using a p0 value of 26%, a ratio of 0.9 between the strengths of the turbulent and mean components of the GMF, and a small value of N. The mean value of p (inferred from the fit of the large-scale patterns in the Stokes maps) is 12 +/- 1%. We relate the polarization layers to the density structure and to the correlation length of the GMF along the LOS. We emphasize the simplicity of our model (involving only a few parameters), which can be easily computed on the celestial sphere to produce simulated maps of dust polarization. Our work is an important step towards a model that can be used to assess the accuracy of component-separation methods in present and future CMB experiments designed to search the B mode CMB polarization from primordial gravity waves.

    • Recent models for the large-scale Galactic magnetic fields in the literature have been largely constrained by synchrotron emission and Faraday rotation measures. We use three different but representative models to compare their predicted polarized synchrotron and dust emission with that measured by the Planck satellite. We first update these models to match the Planck synchrotron products using a common model for the cosmic-ray leptons. We discuss the impact on this analysis of the ongoing problems of component separation in the Planck microwave bands and of the uncertain cosmic-ray spectrum. In particular, the inferred degree of ordering in the magnetic fields is sensitive to these systematic uncertainties, and we further show the importance of considering the expected variations in the observables in addition to their mean morphology. We then compare the resulting simulated emission to the observed dust polarization and find that the dust predictions do not match the morphology in the Planck data but underpredict the dust polarization away from the plane. We modify one of the models to roughly match both observables at high latitudes by increasing the field ordering in the thin disc near the observer. Though this specific analysis is dependent on the component separation issues, we present the improved model as a proof of concept for how these studies can be advanced in future using complementary information from ongoing and planned observational projects.

    • The international precision oncology program INFORM enrolls relapsed/refractory pediatric cancer patients for comprehensive molecular analysis. We report a two-year pilot study implementing ex vivo drug sensitivity profiling (DSP) using a library of 75–78 clinically relevant drugs. We included 132 viable tumor samples from 35 pediatric oncology centers in seven countries. DSP was conducted on multicellular fresh tumor tissue spheroid cultures in 384-well plates with an overall mean processing time of three weeks. In 89 cases (67%), sufficient viable tissue was received; 69 (78%) passed internal quality controls. The DSP results matched the identified molecular targets, including BRAF, ALK, MET, and TP53 status. Drug vulnerabilities were identified in 80% of cases lacking actionable (very) high-evidence molecular events, adding value to the molecular data. Striking parallels between clinical courses and the DSP results were observed in selected patients. Overall, DSP in clinical real-time is feasible in international multicenter precision oncology programs.

  • Global effect of modifiable risk factors on cardiovascular disease and mortality

    • Background: Five modifiable risk factors are associated with cardiovascular disease and death from any cause. Studies using individual-level data to evaluate the regional and sex-specific prevalence of the risk factors and their effect on these outcomes are lacking.Methods: We pooled and harmonized individual-level data from 112 cohort studies conducted in 34 countries and 8 geographic regions participating in the Global Cardiovascular Risk Consortium. We examined associations between the risk factors (body-mass index, systolic blood pressure, non-high-density lipoprotein cholesterol, current smoking, and diabetes) and incident cardiovascular disease and death from any cause using Cox regression analyses, stratified according to geographic region, age, and sex. Population-attributable fractions were estimated for the 10-year incidence of cardiovascular disease and 10-year all-cause mortality.Results: Among 1,518,028 participants (54.1% of whom were women) with a median age of 54.4 years, regional variations in the prevalence of the five modifiable risk factors were noted. Incident cardiovascular disease occurred in 80,596 participants during a median follow-up of 7.3 years (maximum, 47.3), and 177,369 participants died during a median follow-up of 8.7 years (maximum, 47.6). For all five risk factors combined, the aggregate global population-attributable fraction of the 10-year incidence of cardiovascular disease was 57.2% (95% confidence interval [CI], 52.4 to 62.1) among women and 52.6% (95% CI, 49.0 to 56.1) among men, and the corresponding values for 10-year all-cause mortality were 22.2% (95% CI, 16.8 to 27.5) and 19.1% (95% CI, 14.6 to 23.6).Conclusions: Harmonized individual-level data from a global cohort showed that 57.2% and 52.6% of cases of incident cardiovascular disease among women and men, respectively, and 22.2% and 19.1% of deaths from any cause among women and men, respectively, may be attributable to five modifiable risk factors.     • A wide gap exists between the state of the art in developing Wireless Sensor Network (WSN) software and current practices concerning the design, execution, and maintenance of business processes. WSN software is most often developed based on low-level OS abstractions, whereas business process development leverages high-level languages and tools. This state of affairs places WSNs at the fringe of industry. The makeSense system addresses this problem by simplifying the integration of WSNs into business processes. Developers use BPMN models extended with WSN-specific constructs to specify the application behavior across both traditional business process execution environments and the WSN itself, which is to be equipped with application-specific software. We compile these models into a high-level intermediate language-Also directly usable by WSN developers-And then into OS-specific deployment-ready binaries. Key to this process is the notion of meta-Abstraction, which we define to capture fundamental patterns of interaction with and within the WSN. The concrete realization of meta-Abstractions is application-specific; developers tailor the system configuration by selecting concrete abstractions out of the existing codebase or by providing their own. Our evaluation of makeSense shows that i) users perceive our approach as a significant advance over the state of the art, providing evidence of the increased developer productivity when using makeSense; ii) in large-scale simulations, our prototype exhibits an acceptable system overhead and good scaling properties, demonstrating the general applicability of makeSense; and, iii) our prototype-including the complete tool-chain and underlying system support-sustains a real-world deployment where estimates by domain specialists indicate the potential for drastic reductions in the total cost of ownership compared to wired and conventional WSN-based solutions.

    • The reversible thermal unfolding of the archaeal histone-like protein Ssh10b from the extremophile Sulfolobus shibatae was studied using differential scanning calorimetry and circular dichroism spectroscopy. Analytical ultracentrifugation and gel filtration showed that Ssh10b is a stable dimer in the pH range 2.5-7.0. Thermal denaturation data fit into a two-state unfolding model, suggesting that the Ssh10 dimer unfolds as a single cooperative unit with a maximal melting temperature of 99.9 degrees C and an enthalpy change of 134 kcal/mol at pH 7.0. The heat capacity change upon unfolding determined from linear fits of the temperature dependence of DeltaH(cal) is 2.55 kcal/(mol K). The low specific heat capacity change of 13 cal/(mol K residue) leads to a considerable flattening of the protein stability curve (DeltaG (T)) and results in a maximal DeltaG of only 9.5 kcal/mol at 320 K and a DeltaG of only 6.0 kcal/mol at the optimal growth temperature of Sulfolobus.

    • The MM500 study is an initiative to map the protein levels in malignant melanoma tumor samples, focused on in-depth histopathology coupled to proteome characterization. The protein levels and localization were determined for a broad spectrum of diverse, surgically isolated melanoma tumors originating from multiple body locations. More than 15,500 proteoforms were identified by mass spectrometry, from which chromosomal and subcellular localization was annotated within both primary and metastatic melanoma. The data generated by global proteomic experiments covered 72% of the proteins identified in the recently reported high stringency blueprint of the human proteome. This study contributes to the NIH Cancer Moonshot initiative combining detailed histopathological presentation with the molecular characterization for 505 melanoma tumor samples, localized in 26 organs from 232 patients.

    • Short-chain dehydrogenases/reductases form a large, evolutionarily old family of NAD(P)(H)-dependent enzymes with over 60 genes found in the human genome. Despite low levels of sequence identity (often 10-30%), the three-dimensional structures display a highly similar alpha/beta folding pattern. We have analyzed the role of several conserved residues regarding folding, stability, steady-state kinetics, and coenzyme binding using bacterial 3beta/17beta-hydroxysteroid dehydrogenase and selected mutants. Structure determination of the wildtype enzyme at 1.2-Angstrom resolution by x-ray crystallography and docking analysis was used to interpret the biochemical data. Enzyme kinetic data from mutagenetic replacements emphasize the critical role of residues Thr-12, Asp-60, Asn-86, Asn-87, and Ala-88 in coenzyme binding and catalysis. The data also demonstrate essential interactions of Asn-111 with active site residues. A general role of its side chain interactions for maintenance of the active site configuration to build up a proton relay system is proposed. This extends the previously recognized catalytic triad of Ser-Tyr-Lys residues to form a tetrad of Asn-Ser-Tyr-Lys in the majority of characterized short-chain dehydrogenases/reductase enzymes.

    • Magnetic fields play an important role in shaping the structure and evolution of the interstellarmedium (ISM) of galaxies, but the details of this relationship remain unclear. With SKA1, the 3Dstructure of galactic magnetic fields and its connection to star formation will be revealed. A highlysensitive probe of the internal structure of the magnetoionized ISM is the partial depolarizationof synchrotron radiation from inside the volume. Different configurations of magnetic field andionized gas within the resolution element of the telescope lead to frequency-dependent changesin the observed degree of polarization. The results of spectro-polarimetric observations are tiedto physical structure in the ISM through comparison with detailed modeling, supplemented withthe use of new analysis techniques that are being actively developed and studied within the communitysuch as Rotation Measure Synthesis. The SKA will enable this field to come into its ownand begin the study of the detailed structure of the magnetized ISM in a sample of nearby galaxies,thanks to its extraordinary wideband capabilities coupled with the combination of excellentsurface brightness sensitivity and angular resolution.

    • Determining magnetic field properties in different environments of the cosmic large-scale structure as well as their evolution over redshift is a fundamental step toward uncovering the origin of cosmic magnetic fields. Radio observations permit the study of extragalactic magnetic fields via measurements of the Faraday depth of extragalactic radio sources. Our aim is to investigate how much different extragalactic environments contribute to the Faraday depth variance of these sources. We develop a Bayesian algorithm to distinguish statistically Faraday depth variance contributions intrinsic to the source from those due to the medium between the source and the observer. In our algorithm the Galactic foreground and the measurement noise are taken into account as the uncertainty correlations of the galactic model. Additionally, our algorithm allows for the investigation of possible redshift evolution of the extragalactic contribution. This work presents the derivation of the algorithm and tests performed on mock observations. With cosmic magnetism being one of the key science projects of the new generation of radio interferometers we have made predictions for the algorithm's performance on data from the next generation of radio interferometers. Applications to real data are left for future work.
