  • Bystander cell death and stress response is inhibited by the radical scavenger α(1)-microglobulin in irradiated cell cultures.

    • Alpha-particle irradiation of cells damages not only the irradiated cells but also nontargeted bystander cells. It has been proposed that the bystander effect is caused by oxidants and free radicals generated by the radiation. Recent studies have shown that α(1)-microglobulin protects against cell damage caused by oxidants and free radicals. Using a novel experimental system that allows irradiation of 0.02% of a human hepatoma monolayer, leaving 99.98% as bystander cells, we investigated the influence of oxidative stress and the cell-protective effects of α(1)-microglobulin during α-particle irradiation. The results showed an increase in cell death in both irradiated cells and bystander cells. A significant increase in apoptosis, oxidation markers and expression of the stress response genes heme oxygenase 1, superoxide dismutase, catalase, glutathione peroxidase 1, p21 and p53 were observed. Addition of α(1)-microglobulin reduced the amount of dead cells and inhibited apoptosis, formation of oxidation markers, and up-regulation of stress response genes. The results emphasize the role of oxidative stress in promoting bystander effects. Furthermore, the results suggest that α(1)-microglobulin protects nonirradiated cells by eliminating oxidants and free radicals generated by radiation and imply that α(1)-microglobulin can be used in radiation therapy of tumors to minimize damage to surrounding tissues.

  • Fabrication and Characterization of Ultra-Thin PIN Silicon Detectors for Counting the Passage of MeV Ions

    • This paper describes the fabrication and initial characterization of an ultra-thin silicon PIN detector using a new technique in silicon nanotechnology. In collaboration with the Nuclear Physics Division and the Lund Nano Lab at Lund University, we have developed and manufactured ultra thin Delta E-detectors for spectroscopic applications. The fabrication process has been carried out using a double-polished silicon substrate n-type wafer and locally thinning by means of a 10:1 solution of 25% tetramethyl ammonium hydroxide (TMAH) with Isopropyl alcohol. More than 100 detectors of different thicknesses, down to 5 mu m with active areas ranging from 0.71 to 0.172 mm(2), have been fabricated. The main design considerations of our thin detectors were a very low leakage current below 12 nA and a low full depletion voltage at a reverse bias less than 1.5 V. Finally, most of our thin detectors offer an energy resolution (FWHM) as low as 31 keV for 5.487 MeV alpha particles from a Am-241 source.

    • This paper describes the performance evaluation of new Delta E detectors to be used as pre-cell hit detectors in living cell irradiation experiments at the Lund Ion Beam Analysis facility (LIBAF). Using these detectors with a thickness down to 4.15 mu m fabricated at Lund University, an experiment was setup in which Delta E-detectors were used together with a stop E-detector in a telescope system under coincidence measurements. The characteristics of Delta E detectors were based on the optimal detection of the passage of 2.55 MeV protons. The results of these tests demonstrate that the detector telescope clearly separates the protons, this due to high signal-to-noise ratio and good energy resolution of the Delta E-detectors. The best performing detector was shown to have a detection efficiency of 95% at thickness of 9.7 mu m. This type of high-performing detector is suitable for the planned role of the Delta E-detector in the future cell irradiation experiments. (C) 2013 Elsevier B.V. All rights reserved.

  • Efficient ultra-thin transmission silicon detectors for a single-ion irradiation system at the Lund Ion Beam Analysis Facility

    • This paper describes the fabrication of efficient ultra-thin silicon transmission detectors for use as pre-cell detectors in single-ion experiments on living cells at the Lund Ion Beam Analysis Facility. More than 40 detectors of different thicknesses down to 5 mu m have been fabricated and packaged. The main design considerations were very low leakage current (below 9 nA) and low full depletion voltage at biases less than 0.5 V at room temperature. In addition, we have shown that cooling the device can reduce the leakage current to 3 nA. The experimental testing of the pre-cell detection system is based on counting the passage of ions through the transmission (Delta E) detector before hitting the stopping (E) detector placed behind it, to ensure the accurate delivery of specific doses of radiation to the sample. Optimal detection of the fabricated detectors for the passage of an external beam of 2.2 MeV protons was obtained by cooling the device to below 2 degrees C. Cooling the Delta E detectors provides up to 20% better energy resolution and up to 98% detection efficiency for 2.2 MeV protons. The development of this kind of efficient pre-cell detector enables a range of new experiments to be conducted on thick biological samples.

  • Modification of spray-deposited CdO thin films by 1.5 MeV proton irradiation

    • Thin films of cadmium oxide (CdO) with different thicknesses in the range of 78–250 nm were prepared and deposited on glass substrates by using a spray pyrolysis technique. A microbeam facility (Lund Ion Beam Analysis facility) was used to irradiate the prepared CdO thin films with a 1.5 MeV proton beam for fluence in the range of 2 × 1012–2 × 1014 ions/cm2. The surface morphology of the CdO films was measured by atomic force microscopy, and it was observed that the grain size and surface roughness were increased as a function of ion fluence. A UV spectrophotometer was used to study the optical properties of the samples, and the results revealed that the absorbance and optical band gaps increased with an increase in the proton fluence and CdO film's thickness. This work explores the possibility of CdO material modification using low-energy proton irradiation.

    • The use of on-axis geometry in scanning transmission ion microscopy (STIM) has been widely used for thin biological sample structure identification. In this configuration, the lateral resolution is optimised so that micron or submicron beam spots are easily achieved even for classic microbeam lines. Off-axis STIM was more particularly employed for rapid imaging, and also (when associated to a scattering set-up) for normalising elemental contents obtained by other ion beam analysis techniques in organic thin samples. Due to the very small beam current required, on-axis STIM is a stand-alone technique. Off-axis STIM can be advantageous as it enables the simultaneous utilisation of PIXE and RBS techniques. In this paper, the STIM images obtained with an on-axis geometry, a standard off-axis geometry and a recently developed on-off geometry are presented and discussed. Data from skin samples are used for comparison purposes aiming at studying skin permeability to sunscreens. Skin is a stratified tissue and the precise identification of skin layers is needed to ascertain the penetration depth of the physical filter from the formulation. In addition to the intrinsic difference in image quality due to the beam resolution, the influence of the detector type, implanted silicon detector versus Si pin diode, is discussed on the basis of their energy resolution.. their resistance to beam damage as well as the effect

    • The objective of the CELLION project is radiation research at low doses. The main cell responses to low dose irradiation are bystander effects, genomic instability and adaptive responses. In order to study these effects it is convenient to make the cells addressable in space and time through locking the cell position. A new alternative dish has been developed for irradiation procedures at the Lund Nuclear Probe. The versatile dish can be used both to cultivate and to hold the cells during the irradiation procedure. The irradiation dish is made of an epoxy-based photopolymer named SU-8 chosen by its flexibility, non-toxicity and biological compatibility to cell attachment. It has been fabricated using a UV lithographic technique. The irradiation dish forms a 2 × 2 mm2 grid which contains 400 squares. Each square has 80 μm side and is separated from neighbouring ones by 20 μm wide walls. The location of each square is marked by a row letter and column number patterned outside the grid. The Cell Irradiation Facility at the Lund Nuclear Probe utilizes protons to irradiate living cells. A post-cell detection set up is used to control the applied dose, detecting the number of protons after passing through the targeted cell. The transmission requirement is fulfilled by our new irradiation dish. So far, the dish has been used to perform non-targeted irradiation of Hepatoma cells. The cells attach and grow easily on the SU-8 surface. In addition, the irradiation procedure can be performed routinely and faster since the cells are incubated and irradiated in the same surface.

    • The Lund Nuclear Microprobe (LNM) has been adapted to be used as a Single Ion Hit Facility (SIHF) for proton cell irradiation experiments at low dose. In order to test the capabilities of the system, Ni dot arrays and artificial cells have been fabricated with the photopolymer SU-8 and common lithographic techniques. The primary purpose of the Ni dot arrays was to determine the targeting accuracy of the beam in vacuum and in air. Additionally, this sample was employed to evaluate the system performance during cell target irradiation experiments. The Ni dot arrays were also used for beam characterization. The artificial cells were originally fabricated to test the software for cell recognition and localization, developed and implemented at the LNM. However, this sample became very functional to make small adjustments at the irradiation chamber for cell irradiation experiments.     • The CELLION project is directed towards the studies on cellular response to targeted single ions. This paper gives an account of the modifications made at the Lund sub-micron beam line in order to create a Single Ion Hit Facility for biological applications within the CELLION project. The most relevant new feature is the specially developed software used to locate the cells. A program for cell recognition and localization that gives the coordinates of the centre of the cells has been designed. The recognition is made online. A picture taken by a microscope is used as input parameter for the recognition program. Using V79 Chinese hamster cells, the recognition procedure can be done in less than 0.5 s for a picture size of 800 x 600 pixels with approximately 96% efficiency. This paper also reports target accuracy test results and the first non-targeted irradiation procedure performed at the Lund sub-micron beam line. (c) 2007 Elsevier B.V. All rights reserved.

  • Exposure parameters for MeV proton beam writing on SU-8

    • Proton beam writing was performed on a lithographic resist to determine the main parameters required to achieve the minimum feature size, maximum pattern lateral density and maximum aspect ratio. A 2.5 MeV proton beam focused to sizes between 1.5 and 2.5 mu m was used to expose SU-8 negative resist. The number of protons per pixel was varied in the exposure of SU-8 with thicknesses between 5 and 95 pm. Patterns consisting of single pixels, single-pixel lines and multi-pixel areas with different densities were fabricated. The smallest structures achieved were posts 1.5 pin in diameter with 4:1 structure-space ratio in 15 pm thick resist and the highest aspect ratio structures of 20:1 in 40 pm resist were produced. It was found that the minimum feature size depended only on the beam size, and +/- 10% post size accuracy could be achieved within 40-70% variation of the number of protons. MeV proton beam allows a direct fabrication of complex shapes without a mask in single-step irradiation and. in addition, no proximity correction is needed. We present examples of MeV proton beam written single and multi-pixel microstructures with easily reproducible high aspect ratios and densities. (c) 2006 Elsevier B.V. All rights reserved.

    • A new nuclear microscopy technique called ion photon emission microscopy or IPEM was recently invented. IPEM allows analysis involving single ions, such as ion beam induced charge (IBIC) or single event upset (SEU) imaging using a slightly modified optical microscope. The spatial resolution of IPEM is currently limited to more than 10 mu m by the scattering and reflection of ion-induced photons, i.e. light blooming or spreading, in the ionoluminescent phosphor layer. We are developing a '' Microscopic Gridded Phosphor '' (also called Black Matrix) where the phosphor nanocrystals are confined within the gaps of a micrometer scale opaque grid, which limits the amount of detrimental light blooming. MeV-energy proton beam lithography is ideally suited to lithographically form masks for the grid because of high aspect ratio, pattern density and sub-micron resolution of this technique. In brief, the fabrication of the grids was made in the following manner: (1) a MeV proton beam focused to 1.5-2 mu m directly fabricated a matrix of pillars in a 15 mu m thick SU-8 lithographic resist; (2) 7:1 aspect ratio pillars were then formed by developing the proton exposed area; (3) Ni (Au) was electrochemically deposited onto Cu-coated Si from a sulfamate bath (or buffered CN bath); (4) the SU-8 pillars were removed by chemical etching; finally (5) the metal micro-grid was freed from its substrate by etching the underlying Cu layer. Our proposed metal micro-grids promise an order-of-magnitude improvement in the resolution of IPEM.

    • A MeV proton beam with a current in the range of fA-nA is usually measured using a secondary beam signal that is dependent on a sample or requires noise-free accurate charge integration in a Faraday cup. We propose a simple on-line beam current measurement setup that can be used to directly measure beam current during ion beam analysis. A fast beam blanker is used to modulate the beam position after the last set of collimator slits in and out of a mini Faraday cup connected to fA-sensitive electrometer. Less than 100 fA current can be measured during an undisturbed irradiation with kHz sampling frequency. The fast, simple and easily controlled current measurement set-up is going to be routinely used for ion beam analysis and modification at the new Lund sub-micron beam line. (c) 2006 Elsevier B.V.     • A new beam blanking system was installed at the Lund Nuclear Microprobe and employed in proton beam lithography (PBL) for polymer microstructures fabrication. The blanker consists of two parallel plates connected to a high voltage generator. Measurement of the beam blanking time on a sample was performed by means of the standard PIXE system. The beam is blanked and returns to a sample within 200 ns. The blanking system is designed for the new sub-micrometer beamline under installation in the accelerator laboratory. A number of pilot MeV ion beam lithography experiments were performed to illustrate the possibility to use the blanking system in combination with the existing data acquisition and scanning system. A 2.5 MeV proton beam was used to irradiate 50 mum SU-8 negative resist. The blanker was shown to be a necessary part of the lithography system. It enables blanking between each pixel and hence fabrication of various patterns down to a single pixel. The blanker has significantly simplified beam control and enhanced process time and spatial resolution. Three-dimensional microstructures with 20:1 aspect ratio were fabricated.

    • A transition from gabbro to eclogite has been investigated at Vinddoldalen in south-central Norway, with the aim to link reaction textures to metamorphic zircon growth and to obtain a direct U-Pb zircon age of the metamorphic process. In the different rocks of the transition zone zircon occurs as (I) igneous prismatic grains, (II) metamorphic polycrystalline rims and pseudomorphs after baddeleyite, and (III) as tiny (< 10 mu m) bead-like zircon grains. Textural relations suggest that type II zircon formed by breakdown of baddeleyite in the presence of silica, whereas Fe-Ti oxides were the main Zr source for the type III zircon. Subsolidus liberation of Zr and formation of bead zircon took place by oxyexsolution of titanomagnetite during fluid-assisted metamorphism, and by resorption of Fe-Ti oxide in rock domains that were completely recrystallized to eclogite. SIMS (secondary ion mass spectrometry) and TIMS (thermal ionization mass spectrometry) dating provides comparable U-Pb ages of magmatic zircon and baddeleyite. Baddeleyite (TIMS) yielded an age of 1457 +/- 11 Ma for the gabbro emplacement. Bead-type metamorphic zircon from eclogite gave 425 +/- 10 Ma (TIMS) dating the metamorphic transition from gabbro to eclogite in the upper basement of the Lower Allochthon in the south-central Scandinavian Caledonides.

    • One of multiple functions of metalloproteins is to provide detoxification to excess metal levels in organisms. Here we address the induction and persistence of a range of low to high molecular weight copper- and zinc binding proteins in the collembolan species Tetrodontophora bielanensis exposed to copper- and zinc-enriched food, followed by a period of recovery from metal exposure, in absence and presence of food. After 10 days of feeding copper and zinc contaminated yeast, specimens were either moved to ample of leaf litter material from their woodland stand of origin or starved (no food offered). The molecular weight distribution of metal binding proteins was determined by native polyacryl gel electrophoresis. One gel was stained with Comassie brilliant blue and a duplicate gel dried and scanned for the amount of copper and zinc by particle-induced X-ray emission. Specimens exposed to copper and recovered from it with ample of food had copper bound to two groups of rather low molecular weight proteins (40-50 kDa) and two of intermediate size (70-80 kDa). Most zinc in specimens from the woodland stand was bound to two large proteins of about 104 and 106 kDa. The same proteins were holding some zinc in metal-exposed specimens, but most zinc was found in proteins <40 kDa in size. Specimens recovered from metal exposure in presence of ample of food had the same distribution pattern of zinc binding proteins, whereas starved specimens had zinc as well as copper mainly bound to two proteins of 8 and 10 kDa in size. Thus, the induction and distribution of copper- and zinc-binding proteins depend on exposure conditions, and the presence of low molecular weight binding proteins, characteristic of metallothioneins, was mainly limited to starving conditions.

    • A new setup for photon tagged nuclear reaction analysis pNRA is being developed at Lund's ion beam analysis facility LIBAF. Particle induced gamma ray emission PIGE and nuclear reaction analysis NRA are two methods that have been extensively used for light isotope measurement in ion beam analysis IBA. There is an abundance of nuclear reactions between light elements and MeV protons, deuterons and alpha particles. This means that in principle all elements from lithium all the way up to chlorine can be analyzed using those techniques. Detection limits can be improved for some elements, if those two methods are fused together into pNRA. The new setup for pNRA will benefit from advances in detector technology that occurred during the last 20 years. A LaBr3 scintillator detector and an annular double sided silicon strip detector DSSSD are used in coincidence to detect a gamma and a charged particle respectively. Both detectors are connected to a VME based data acquisition system. Of primary interest in this work is the analysis of isotopic ratios of light elements in geological samples, which are usually thick with a complex matrix. This setup can be for instance used to measure isotopic fractionation of oxygen and boron. We will present the setup and discuss its capabilities. (C) 2014 The Authors. Published by Elsevier B.V.

    • The double sided silicon strip detector (DSSSD) is a segmented silicon detector commonly used in the fields of high energy physics and nuclear physics. This type of detector is used for analysis of reactions produced by charged particles. This makes it well suited for a number of analytical methods commonly used in ion beam analysis (IBA), such as Rutherford Backscattering (RBS) and elastic recoil detection (ERDA). One such detector was installed and tested at Lund Ion Beam Analysis Facility (LIBAF) recently. This is a modification to the existing setup used to measure hydrogen concentrations and depth profiles. When completed it will be used primarily for geological applications. Exact knowledge of the hydrogen content is important in a number of fields, but high enough accuracy can be difficult to achieve with most methods. In IBA normally some variant of ERDA, such as the proton-proton (p-p) coincidence method is used. We describe how the p-p coincidence technique was optimized to get the most out of our experimental setup. Previously this type of spectroscopy has been performed with two detector channels. In the present setup we expand that number from 2 to 96 channels, 64 on the front and 32 on the back of the detector. The intersecting strips give 2048 distinct detector elements or 1024 possible coincidences as dictated by the reaction kinematics. This increase in complexity requires a more detailed data analysis but it rewards us with higher sensitivity and a better background suppression. (C) 2013 Elsevier B.V. All rights reserved.

    • In this work, we discuss the possibility of a new approach to measuring oxygen isotope ratios in fossil meteorite samples, specifically one based on nuclear reaction analysis (NRA). Variations of oxygen ratios within meteoritic chromite grains can help to determine the type of meteorite to which the grains originally belonged. In this work, we have evaluated the possibility to use the reaction O-18(p, alpha)N-15 just above the 846 keV resonance to estimate the relative oxygen-18 content in a number of test samples. Another technique has to be employed for oxygen-16 measurements. A large area segmented silicon detector is used to detect the produced alpha particles. Results of the experimental O-18 measurements for a number of samples including four extraterrestrial chromite grains are presented and compared with SIMNRA simulations. The advantage of a segmented silicon detector in the form of inherent pile-up suppression can be clearly seen in the current work. (C) 2011 Elsevier B.V. All rights reserved.

    • The possibility to study variations in the oxygen isotopic ratio with photon tagged nuclear reaction analysis (pNRA) is evaluated in the current work. The experiment described in the article was performed at Lund Ion Beam Analysis Facility (LIBAF) with a 2 MeV deuteron beam. Isotopic fractionation of light elements such as carbon, oxygen and nitrogen is the basis of many analytical tools in hydrology, geology, paleobiology and paleogeology. IBA methods provide one possible tool for measurement of isotopic content. During this experimental run we focused on measurement of the oxygen isotopic ratio. The measurement of stable isotopes of oxygen has a number of applications; the particular one driving the current investigation belongs to the field of astrogeology and specifically evaluation of fossil extraterrestrial material. There are three stable isotopes of oxygen; O-16, O-17 and O-18. We procured samples highly enriched with all three isotopes. Isotopes O-16 and O-18 were easily detected in the enriched samples, but no significant signal from O-17 was detected in the same samples. The measured yield was too low to detect O-18 in a sample with natural abundances of oxygen isotopes, at least in the current experimental setup, but the spectral line from the reaction with O-16 was clearly visible. Metoden kallas PIXE (Particle Induced X-ray Emisson) och medger att mycket små förekomster (109 – 1012 g) av grundämnen kanbestämmas i små prov. Metoden är snabb. Ett par minuters analys ger en samtidig bestämning av ett stort antal grundämnen. Analysmetoden används nu i ett stort antal laboratorier runt om i världen och har funnit tillämpning inom en rad olika områden såsom medicin, biologi, yrkes- och omgivningshygien, meteorologi, geologi samt arkeologi. I Lund har metoden huvudsakligen använts för omfattande studier av föroreningar i arbetsmiljö och yttre miljö, varför vi först redogör för vårt arbete inom dessa tillämpningsområden, varefter vi beskriver de fysikaliska principerna för analysmetoden och utvecklingsarbetet med den.

    • α1-microglobulin (A1M) is a 26 kDa plasma and tissue protein with reductase activity and radical- and heme-binding anti-oxidative functions. In addition, exposure of A1M to hemoglobin has been shown to induce proteolytic elimination of a C-terminal tetrapeptide yielding a heme-degrading form, truncated A1M (t-A1M). Myeloperoxidase (MPO), a heme-containing enzyme that catalyzes the production of free radicals and hypochlorite, is released by neutrophils during the inflammatory response to bacterial infections. MPO-induced low density lipoprotein (LDL)-oxidation in blood has been suggested as a causative factor in atherosclerosis. In this study we have hypothesized that A1M interacts with MPO in a similar mode as with hemoglobin, and is a regulator of its activity. The results show that A1M is proteolytically cleaved, with formation of t-A1M, after exposure to MPO, and that t-A1M contains iron and heme-degradation products. The reaction is dependent of pH, time and concentration of substrates and a pH-value around 7 is shown to be optimal for cleavage. Furthermore, A1M inhibits MPO- and hydrogen peroxide-induced oxidation of LDL. The results suggest that A1M may have a role as an inhibitor of the damaging effects of the neutrophil respiratory burst on bystander tissue components.

    • In this paper we investigate the applicability of small scintillators with glass rod lightguides to measure both neutrons and gamma rays from the sample. Experimental test ofthese detectors and their suitability for the task can be performed at Lund University usingnatural uranium rods, therefore the MCNPX simulation set-up corresponds to the Lund con-figuration. The long-term goal of this research is to develop a method of joint-multiplicitycounting for fuel evaluation into a technology capable of quantifying plutonium in the fuelpool, where a lot of factors, such as presence of neutron absorbers (boron acid), can affect al-most all parameters, such as multiplication etc. Therefore, part of the present investigationswas devoted to the development of a direct method for determination of the concentrationof boron acid in the fuel pool using scintillation detectors with further correction of mea-surement results. For this purpose we suggest a method which utilizes a relation betweengamma lines with energy of 480 keV and 2.23 MeV for the direct evaluation of concentrationof boron acid in water. Test results of the new method, and an answer to questions regardingthe ability to measure both neutron and gamma rays using small scintillation detectors withglass rod light guides are provided in this paper.

    • Quantification and mapping possibilities of lithium in geological material, by Nuclear Reaction Analysis (NRA), was evaluated at the Lund Ion Beam Analysis Facility (LIBAF). LiF and two Standard Reference Materials, (SRM 610 and SRM 612) were used in the investigation. The main part of the data was obtained at the beam energy 635keV studying the high Q-value reaction 7Li(p, α)4He, but reaction yield and detection limits were also briefly investigated as a function of the energy. A double-sided silicon strip detector (DSSSD) was used to detect the α-particles emitted in the reaction in the backward direction. The combination of the high Q-value, a reasonably good cross-section and the possibility to use a high beam current have been demonstrated to allow for measurement of concentrations down below 50ppm. Proton energies below 800keV were demonstrated to be appropriate energies for extracting lithium in combination with boron analysis.

    • Proton-induced reaction (p,α) is one type of nuclear reaction analysis (NRA) suitable especially for light element quantification. In the case of lithium quantification presented in this work, accelerated protons with an energy about of 850 keV were used to induce the 7Li(p,α)4He reaction in standard reference and geological samples such as tourmaline and other Li-minerals. It is shown that this technique for lithium quantification allowed for measurement of concentrations down below one ppm. The possibility to relate the lithium content with the boron content in a single analysis was also demonstrated using tourmaline samples, both in absolute concentration and in lateral distribution. In addition, Particle induced X-ray emission (PIXE) was utilized as a complementary IBA technique for simultaneous mapping of elements heavier than sodium.

    • With the recent advances towards modern Ion Beam Analysis (IBA), going from one- or few-parameter detector systems to multi-parameter systems, it has been necessary to expand and replace the more than twenty years old CAMAC based system. A new VME multi-parameter (presently up to 200 channels) data acquisition and control system has been developed and implemented at the Lund Ion Beam Analysis Facility (LIBAF). The system is based on the VX-511 Single Board Computer (SBC), acting as master with arbiter functionality and consists of standard VME modules like Analog to Digital Converters (ADC's), Charge to Digital Converters (QDC's), Time to Digital Converters (TDC's), scaler's, IO-cards, high voltage and waveform units. The modules have been specially selected to support all of the present detector systems in the laboratory, with the option of future expansion. Typically, the detector systems consist of silicon strip detectors, silicon drift detectors and scintillator detectors, for detection of charged particles, X-rays and γ-rays. The data flow of the raw data buffers out from the VME bus to the final storage place on a 16 terabyte network attached storage disc (NAS-disc) is described. The acquisition process, remotely controlled over one of the SBCs ethernet channels, is also discussed. The user interface is written in the Kmax software package, and is used to control the acquisition process as well as for advanced online and offline data analysis through a user-friendly graphical user interface (GUI). In this work the system implementation, layout and performance are presented. The user interface and possibilities for advanced offline analysis are also discussed and illustrated.

    • The cellular part of the skin, the epidermis, is a very thin structure, approximately 120 microns thick, a fact which has hindered the exploration of the physiology of the epidermis in normal and pathological conditions. An additional complication is the fact that the epidermis contains layers of cells at different stages of differentiation. Therefore, conventional physiological capillary probes cannot, with any satisfactory precision, be located within a specified cell of a specified layer of the skin in vivo. Hence, alternative ways for the exploration of skin physiology have been sought for. In the past, analysis of the elemental content of skin was done was done as bulk measurements, and surprisingly wide ranges of elemental content were recorded. The width of these ranges was most certainly due to the sampling methods used rather than the sensitivity of the chosen method of analysis. Also, these older measurements did not discriminate between the different strata, and therefore the information provided little if any substance for a functional analysis of processes involved in normal and pathological differentiation of the epidermis. Particle probes, however, have been able to overcome such methodological problems. Over a period of 15 years we have studied normal human skin, normal-looking, paralesional skin from psoriatics, and skin from persons suffering from atopic dermatitis using PIXE analysis. In recent years, trace elements have been shown to work as secondary messengers or regulatory substances. As an example, calcium (Ca2+) has proven to be a very important signalling substance in a great variety of cellular systems. Studies with the transmission electron microscope (TEM) as well as histochemical methods have allowed an understanding of the role of Ca2+ in the differentiation process of the epidermis. Ca2+ has also been shown to play an important role in apoptosis (programmed cell death), which is currently a hot subject for the obvious reason that the final differentiation step between the stratum granulosum level and the stratum corneum represents a particular aspect of programmed cell death. The importance of the balance between calcium and zinc in apoptosis has been clearly demonstrated in a number of cellular systems, but we have still to clarify the validity of topical treatment with Zn ointments in different skin conditions. Substantial iron (Fe) losses via psoriatic lesions were demonstrated more than two decades ago, and these data were given new meaning when we found that a more discrete loss occurs in clinically normal-looking psoriatic skin. Obviously, such findings stress the importance of understanding the relation between the elemental content and normal and abnormal physiology. The ultimate goal of particle probe studies is to provide an understanding of the formation of a mature stratum corneum with a functional barrier reflected in physiological/biochemical mechanisms behind the properties of changed skin in patients afflicted with skin disorders of genetic or constitutional origin. This paper aims to give an overview of the state of the art in skin physiology made possible through the use of particle probes.

    • Using a scanning nuclear microprobe, the distribution of elements and trace elements of skin cross sections of normal skin, non-lesional psoriatic skin and in dry atopic skin have been mapped. In non-lesional psoriatic skin and in dry atopic skin the epidermal Ca-gradient is higher than that of normal skin. In addition, abnormally high Fe and Zn levels were recorded in the stratum granulosum and corneum regions in the pathological skin. It is suggested that these findings correlate to an increased cell turnover in the basal cell layer of the psoriatic and atopic skins. The ratio of Ca/Zn in stratum corneum of paralesional psoriatic skin is approximately 8:1 compared to 12: 1 in normal skin and 15: 1 in atopic skin. This suggests that the differentiation process in paralesional psoriatic skin may actually be an example of disturbed programmed cell death.

    • Particle probe methods (electron probe and proton probe X-ray microanalysis) have been applied to investigate the distribution of elements and water over the different layers of the epidermis. For major elements, electron probe X-ray microanalysis (XRMA) provides the advantage of superior spatial resolution, but for trace element analysis the more sensitive proton probe (particle induced X-ray emission, PIXE) analysis has to be used. On a dry weight basis, the concentration of S is rather constant across the epidermis, whereas the concentrations of P, K, Cl and Na show gradients with high levels in stratum germinativum (basale) and stratum spinosum but low levels in the stratum granulosum and stratum corneum. Essentially, Fe and Zn are confined to the basal region in normal skin. The concentration of Ca, however, increased steadily from the basal region to the stratum corneum. The probe technique allows quantitative analysis of stratum-specific changes in elemental content in a variety of pathological conditions, e.g., changes induced by nickel, detergents and other chemicals, or in psoriatic skin. Of particular interest are findings of increased Fe and Zn in non-involved psoriatic skin. Since the different layers of the skin have different elemental concentrations and react differently under pathological conditions, the probe techniques are far superior to bulk chemical analysis in elucidating physiological and pathological processes in the skin.

    • The Applied Nuclear Physics Group at Lund University is constructing a prototype CANS (Compact Accelerator-driven Neutron Source). The CANS is based around a 3 MV, single-ended, Pelletron accelerator, which is used to impinge a 2.8 MeV deuterium beam into a beryllium target. The anticipated neutron production will be on the order of 1010 n/s in 4π sr. A further upgrade to the ion source of the Pelletron is expected to increase neutron production to 1011 n/s. Neutron energies will be up to 9 MeV with peak emission at ∼5 MeV. Shielding and moderation will be provided by a large water tank surrounding the target, with three exit ports to allow neutrons of different energies to be directed to experiments. The design is supported by simulation results which predict fast-neutron fluxes of 9×104 to 5×106 n/cm2/s, and thermal-neutron fluxes of 1×104 to 5×104 n/cm2/s to be readily obtainable with a 10 µA deuteron beam.

    • In situ growth rates of the toxin-producing dinoflagellate Dinophysis norvegica collected in the central Baltic Sea were estimated during the summers of 1998 and 1999. Flow cytometric measurements of the DNA cell cycle of D. norvegica yielded specific growth rates (μ) ranging between 0.1 and 0.4 per day, with the highest growth rates in stratified populations situated at 15–20 m depth. Carbon uptake rates, measured using 14C incubations followed by single-cell isolation, at irradiances corresponding to depths of maximum cell abundance were sufficient to sustain growth rates of 0.1–0.2 per day. The reason for D. norvegica accumulation in the thermocline, commonly at 15–20 m depth, is thus enigmatic. Comparison of depth distributions of cells with nutrient profiles suggests that one reason could be to sequester nutrients. Measurements of single-cell nutrient status of D. norvegica, using nuclear microanalysis, revealed severe deficiency of both nitrogen and phosphorus as compared to the Redfield ratio. It is also possible that suitable prey or substrate for mixotrophic feeding is accumulating in the thermocline. The fraction of cells containing digestive vacuoles ranged from 2 to 22% in the studied populations. Infection by the parasitic dinoflagellate Amoebophrya sp. was observed in D. norvegica in all samples analysed. The frequency of infected cells ranged from 1 to 3% of the population as diel averages, ranging from 0.2 to 6% between individual samples. No temporal trends in infection frequency were detected. Estimated loss rates based on observed infection frequencies were 0.5–2% of the D. norvegica population daily, suggesting that these parasites were not a major loss factor for D. norvegica during the periods of study.

    • For the generation of sound, some woodwind musical instruments, e.g. oboe, bassoon, clarinet and saxophone, are provided with mouthpieces made from reeds. These reeds are the culms of Arundo donax, a tall, cane-like perennial grass. A general problem is that the material is of varying quality, yet externally differences cannot be observed. Hence, large proportions of the prepared reeds are unusable. One hypothesis is that the changes in quality are correlated with differences in the chemical and anatomical structure of the tissue. Therefore, a comparison of superior and inferior mouthpieces, used by professional musicians, was undertaken to determinate potential indicators of quality. Nuclear microprobe analysis of reeds was carried out and complemented by scanning electron and light microscopy. The elemental levels of Si, P, S, Cl, K and Ca were compared between good and poor mouthpieces using appropriate statistical tests. No statistically significant differences could be identified. Microscopical observations showed that partial occlusion of vessels by tylose formation was associated with material deemed unusable.

    • The design and first results from a Double Sided Silicon Strip Detector (DSSSD) recently installed at the Lund Nuclear Microprobe facility (NMP) are presented. The detector has 64 sector strips and 32 ring strips, which in combination give more than 2000 detector cells, each with characteristics comparable with a standard surface barrier detector (SBD). The detector has been tested both with radioactive sources and with different ion beams and energies.     • Piloderma sp., a wood ash-colonizing ectomycorrhizal (EM) fungus, was grown symbiotically with Norway spruce in microcosms which contained granules of hardened wood ash. Mycelium close to the granules was sampled 3 times over a period of 11 weeks and the elemental content was investigated with particle induced X-ray emission. Mycelium from microcosms without wood ash was used as controls. The contents of P and K were similar in mycelium growing close to wood ash granules to those in control mycelium, while the Ca content increased from 23 +/- 21 mg g(-1) in controls to 63 +/- 8 mg g(-1) in mycelium growing close to wood ash granules. The Ca content was also increased in other parts of the mycelium more distant from the wood ash. Piloderma sp. may have a role in the short-term storage of Ca released from wood ash, rather than in releasing and storing P.

We compared the elemental composition of the field AMF to those of the soil and the associated plants. The spores and hyphae from the saline soils showed strongly elevated levels of Ca, Cl, Mg, Fe, Si, and K compared to their growth environment. In contrast, the spores of both the field-derived AMF and the in vitro grown G. intraradices contained lower or not elevated Na levels compared to their growth environment. This resulted in higher K:Na and Ca:Na ratios in spores than in soil, but lower than in the associated plants for the field AMF. The K:Na and Ca:Na ratios of G. intraradices grown in monoxenic cultures were also in the same range as those of the field AMF and did not change even when those ratios in the growth medium were lowered several orders of magnitude by adding NaCl. These results indicate that AMF can selectively take up elements such as K and Ca, which act as osmotic equivalents while they avoid uptake of toxic Na. This could make them important in the alleviation of salinity stress in their plant hosts.

    • The exchange of carbohydrates and mineral nutrients in the arbuscular mycorrhizal symbiosis must be controlled by both partners in order to sustain an evolutionarily stable mutualism. Plants down-regulate their carbon flow to the fungus when nutrient levels are sufficient, while the mechanism controlling fungal nutrient transfer is unknown. Here we show that the fungus accumulates nutrients when connected to a host that is of less benefit to the fungus, indicating a potential of the fungus to control the transfer of nutrients. We used a monoxenic in vitro model of root organ cultures associated with G. intraradices, in which we manipulated the carbon availability to the plant. We found that G. intraradices accumulated up to 7 times more nutrients in its spores, and up to 9 times more in its hyphae, when the carbon pool available to the associated roots was halved. The strongest effect was found for phosphorus, considered to be the most important nutrient in the arbuscular mycorrhizal symbiosis. Other elements such as potassium and chorine were also accumulated, but to a lesser extent, while no accumulation of iron or manganese was found. Our results suggest a functional linkage between carbon and phosphorus exchange.

    • Sturgeon populations in the Danube River have experienced severe decline during the last several decades, mostly due to the poorly regulated fishery, river fragmentation and water pollution. This study focuses on gaining better understanding of sturgeon life history primarily by addressing the assessment of microelement accumulation in sturgeon pectoral fin rays, especially of strontium and calcium, as a method that can reveal migration patterns of anadromous sturgeons. Analysis was performed on pectoral fin samples of three anadromous Danube sturgeon species (beluga, Russian sturgeon and stellate sturgeon) by the use of a Nuclear Microprobe technique. The most frequent pattern in analyzed samples was represented by a low Sr:Ca ratio in the innermost annuli, followed by an increased ratio in the middle annuli segment, and often with a decreased ratio in the outermost annuli. Probability density estimate has revealed three distinguished maxima of the Sr: Ca ratio, 7.08x10(-3), 8.98x10(-3) and 9.90x10(-3), which might correspond, respectively, to fresh, brackish and saltwater. Although the analysis of the Sr: Ca ratio in sturgeon pectoral fin rays has revealed changes that might indicate probable migration between habitats with different water salinity, further studies are needed for improvement of this method. This study represents the first analysis of this kind that was conducted on sturgeon species from the Black Sea basin.

    • A general mass calibration procedure of a PIXE set-up is described. As an example, the results of the calibration of the PIXE set-up in Lund are given. For the calibration commercially available standards were used. The parameters of a physical model were adjusted to fit a calibration curve. To decrease errors due to inhomogeneities of the standard foils, the average from four different pieces of each foil was taken. The accuracy of the calibrated system in Lund is estimated to be better than 5% for individual elements and still less for ratios of adjacent elements. For long-term quality control of the results, a special sample has been designed. This consists of a thick silver plate with a small copper spot in the centre. It is very sensitive to small changes in a PIXE set-up. By analysing this sample periodically, mistakes in the arrangement are promptly detected-a feature which is much appreciated in a mixed research, development and routine analysis laboratory. Also a measure of the long-term stability is obtained. For the Lund system, the long-term stability is 2.5% for a homogeneous sample and 4% for a small (inhomogeneous) sample.

  • Metaller i luftburna partiklar i Landskrona 2017

    • Mellan 16 februari och 22 mars 2017 genomfördes 24 mätningar av finfraktionen (PM2.5) och grovfraktionen (PM10 – PM2.5) av grundämnen i luftburna partiklar på mätstationerna i Stadshuset och Lundåkrahamnen i Landskrona. Detta gjordes som ett led i att kontrollera luftkvaliten i Landskrona stad, särskilt med tanke på industrierna i Landskrona, som tidigare har givit höga halter av framförallt metaller i luften för åren 1977, 1988, 2003, och 2008. Bägge mätstationerna representerar urbana bakgrundsstationer, där Stadshusets mätningar är i trafikmiljö, medan Lundåkrahamnens mätningar är i industriområde.Flertalet ämnen kunde detekteras med PIXE-analysmetoden; Al, Si, P, S, Cl, K, Ca, Ti, Cr, Mn, Fe, Ni, Cu, Zn, Br, Sr, Pb. V och As låg mestadels under detektionsgränsen. Ga, Ge, Se, Rb, Y, Zr, Pd, Cd, och Sn var aldrig detekterbara.För grovfraktionen både vid Stadshuset och Lundåkrahamnen har mätningar under år 2008 generellt gett de lägsta partikelhlaterna. Förutom detta år har det rent generellt varit en nedåtgående trend för halterna för följande ämnen sedan 1977; K, Ca, Ti, Mn, Fe, Ni, Zn, och Pb. Nästan oförändrade halter har observerats för Al, Si, S, Cu, och Br. Halterna av Cl och Cr har rent av gått upp. Det bör dock nämnas att Cl kommer från den naturliga källan havssprej, varför halterna av Cl och andra naturliga ämnen mer beror på meteorologiska förutsättningar än minskningar i mänskliga utsläpp. För finfraktionen har vi nedåtgående trend sedan 1977 för; S, K, Ca, Ti, Mn, Fe, Ni, Cu, Zn, Br, och Pb. Oförändrade halter för Cr, och uppåtgående trend för Cl.P mättes för första gången både i grov- och finfraktion. Detta ämne kan till exempel komma från jordstoftpartiklar, samt från havet (övergödning). Men, ingen vidare korrelation kunde skönjas med jordstoftpartiklar och havssprejpartiklar. Ämnet har betydelse för bördiga jordar och övergödning, varför det är viktigt att fortsätta mäta detta ämne.Käll/receptor-modellering har utförts för att beräkna källtilldelningen. PMF-modellen kördes med en lätt polarisering av källorna (FPEAK-paramter = 0.5) och för grovfraktion och finfraktion för sig, och de två stationerna var och en för sig. Det gjordes alltså 4 olika PMF-körningar.Resultaten visade att vi i princip har 7 olika källor för partiklar och grundämneselement i Landskrona;1. Havssprej2. Jordstoftpartiklar3. Pb-källa4. Fe/Zn-källa5. Cr/Ni-källa6. Cu/Zn-källa7. Åldrad källaHavssprej både i Stadshuset och Lundåkrahamnen utgörs av grova partiklar med Cl, Br som främsta ämnen. Även finfraktionen innehåller en hög halt Cl och Br, vilket förmodligen beror på att även finfraktionen innehåller en relativt hög andel grova partiklar. Högsta halterna är förknippade med havsluft västerifrån.Jorstoftpartiklar karakteriseras av elementen Si, K, Ca, Ti, och Fe, vilka är typiska för jordskorpan. Framförallt grova partiklar ger höga halter av dessa ämnen, men även finfraktionen kan innehålla en andel grova partiklar liksom havssprejkällan.Pb-källan är förknippad med vindar från Boliden-Bergsoe-fabriken, vilket den även har varit för föregående mätningar, både vid Stadshuset och Lundåkrahamnen, såväl i fin- som i grovfraktionen.Fe/Zn-källan är vid några tillfällen associerad med vindar från hamnområdet eller Scandust, för både grov- och finfraktionen i Stadshuset och Lundåkrahamnen. Det går dock inte att härleda denna källa till specifik aktivitet i detta område i Landskrona. En del av Fe/Zn-källan samvarierar med jordsoftkällan, vilket bevisar att en del av Fe och Zn kommer från jordstoft. Detta är väntat eftersom jordskorpan även innehåller dessa element.Liknande resonemang gäller för Cr/Ni-källan. Vi kan dessutom inte utesluta att Cr/Ni i finfraktionen dessutom kommer från långdistanstransport av fossila förbränningspartiklar, eftersom fossil olja ofta innehåller Cr och Ni.Återigen gäller liknande resonemang för Cu/Zn-källan. Dessutom är det möjligt att en del Cu/Zn-damm och fina partiklar möjligen härstammar från färgborttagning från småbåtar och målning.Den sista källan är en långdistanstransporterad källa innehållandes S, framförallt i finfraktionen. Denna kommer från kondensation av svaveldioxid eller svavelsyra under långdistanstransport under flera timmar eller dygn (sekundär källa), eller direkt under förbränning av fossila bränslen (primär källa). Man kan förvänta sig att denna källa har minskat i och med regleringen av sulfatinnehållet i sjöfartsbränslet sedan 2015 i Östersjön och Nordsjön, eller generellt bättre rening av S från kolkraftverk eller andra industrier med förbränningsprocesser sedan konventionen om gränsöverskridande luftföroreningar (CLRTAP) kom igång ordentligt på 80-talet. En svag trend är synlig. Å andra sidan kan variationer i S bero på variationer i meteorologiska förhållanden, och inte enbart på grund av utsläppsminskningar.

    • In many IBA applications the main aim is to obtain quantitative figures characterizing the sample. Normally charge, i.e. number of probe particles, is used for normalization and is measured either by collecting the charge deposited in the sample or by collecting the particle in a post-sample Faraday cup or in combination. Both these techniques have drawbacks and results can be difficult to compare for samples with different matrix composition. In this work, we present an upgraded design and test results from the Lund NMP pre-sample charge measurement system. The system presented is based on a pre-sample beam deflection controlled by the beam scanning system for the nuclear microprobe. It can be operated in different modes, but during normal operation the beam is blanked once per pixel and the corresponding charge is collected during the beam-off period. The system does not only measure an average of the beam current during data collection, but actually a pixel-by-pixel normalization is possible. Data of the system performance are presented and in addition illustrations of how quantitative measurements both for PIXE and elastic scattering can be made more reliable. (C) 2010 Elsevier B.V.     • Stable carbon isotopes play an important role in the interpretation of biological activity, particularly when the fossil record is studied. In combination with morphological and chemical data, isotopic information can be useful in paleontology and astrobiology. In this paper the development of a microanalytical ion beam system for the measurement of the carbon isotope ratio with high spatial resolution and good statistics is presented. The technique used is elastic scattering of alpha particles with an energy of 2.75 MeV. At this energy the C-13 cross-section is enhanced relative to the Rutherford cross-section and while the C-12 cross-section is reduced. The optimisation of the system is described, including sample preparation and the design of the particle detection system for high efficiency combined with good energy resolution.     • Hydrogen normally occurs as hydroxyl ions related to defects at specific crystallographic sites in the structures, and is normally characterized by infrared spectroscopy (FTIR). For quantification purposes the FTIR technique has proven to be less precise since calibrations against independent methods are needed. Hydrogen analysis by the NMP technique can solve many of the problems, due to the low detection limit, high lateral resolution, insignificant matrix effects and possibility to discriminate surface-adsorbed water. The technique has been shown to work both on thin samples and on thicker geological samples. To avoid disturbance from surface contamination the hydrogen is analyzed inside semi-thick geological samples. The technique used is an elastic recoil technique where both the incident projectile (proton) and the recoiled hydrogen are detected in coincidence in a segmented detector. Both the traditional annular system with the detector divided in two halves and the new double-sided silicon strip detector (DSSSD) has been used. In this work we present an upgraded version of the technique, studying two sets of mineral standards combined with pre-sample charge normalization. To improve the processing time of data we suggest a very simple semi-empirical approach to be used for data evaluation. The advantages and drawbacks with the approach are discussed and a possible extension of the model is suggested. (C) 2013 Elsevier B.V. All rights reserved.

    • Interest in high spatial resolution boron analyses from a geochemical perspective is related to the recognition of boron as an important tracer of chemical recycling in the Earth, due to the high solubility of boron in aqueous fluids and silicate melts. Although boron is not a nominal component in common silicates they may still contain significant B-concentrations and hence constitute important boron reservoirs in the deeper parts of the Earth. Boron analyses have been performed at the Lund Ion Beam Analytical Facility for almost 20 years. For the analysis the nuclear reaction p+B-11 is used with beam energy just below 700 keV where the reaction has a broad resonance. In this paper we describe an upgrade of the system with a double sided silicon strip detector, which allows for much higher count rates compared to the old annular surface detector based system. A gain close to 20 in the data rate allows for high resolution mapping of boron distributions in crystals. This is illustrated by a number of examples. In addition, the detection limits for boron in geological samples are improved, now around 5 ppmw. In this work we address issues with data quality, especially charge normalization, lifetime correction and subtraction of different background components. (C) 2014 The Authors.
