    • This paper describes the scientific aims and potentials as well as the preliminary technical design of RUDE, an innovative tool for multi-disciplinary investigations in a wide field of scientific, technological and industrial applications. IRIDE will be a high intensity "particles factory", based on a combination of high duty cycle radio-frequency superconducting electron linacs and of high energy lasers. Conceived to provide unique research possibilities for particle physics, for condensed matter physics, chemistry and material science, for structural biology and industrial applications, IRIDE will open completely new research possibilities and advance our knowledge in many branches of science and technology. [RIDE is also supposed to be realized in subsequent stages of development depending on the assigned priorities.

  • The Gaia -ESO survey: Mixing processes in low-mass stars traced by lithium abundance in cluster and field stars

    • Aims. We aim to constrain the mixing processes in low-mass stars by investigating the behaviour of the Li surface abundance after the main sequence. We take advantage of the data from the sixth internal data release of Gaia-ESO, IDR6, and from the Gaia Early Data Release 3, EDR3s. Methods. We selected a sample of main-sequence, sub-giant, and giant stars in which the Li abundance is measured by the Gaia-ESO survey. These stars belong to 57 open clusters with ages from 130 Myr to about 7 Gyr and to Milky Way fields, covering a range in [Fe/H] between -1.0and +0.5 dex, with few stars between -1.0 and -2.5dex. We studied the behaviour of the Li abundances as a function of stellar parameters. We inferred the masses of giant stars in clusters from the main-sequence turn-off masses, and for field stars through comparison with stellar evolution models using a maximum likelihood technique. We compared the observed Li behaviour in field giant stars and in giant stars belonging to individual clusters with the predictions of a set of classical models and of models with mixing induced by rotation and thermohaline instability. Results. The comparison with stellar evolution models confirms that classical models cannot reproduce the observed lithium abundances in the metallicity and mass regimes covered by the data. The models that include the effects of both rotation-induced mixing and thermohaline instability account for the Li abundance trends observed in our sample in all metallicity and mass ranges. The differences between the results of the classical models and of the rotation models largely differ (up to 2 dex), making lithium the best element with which to constrain stellar mixing processes in low-mass stars. We discuss the nature of a sample of Li-rich stars. Conclusions. We demonstrate that the evolution of the surface abundance of Li in giant stars is a powerful tool for constraining theoretical stellar evolution models, allowing us to distinguish the effect of different mixing processes. For stars with well-determined masses, we find a better agreement of observed surface abundances and models with rotation-induced and thermohaline mixing. Rotation effects dominate during the main sequence and the first phases of the post-main-sequence evolution, and the thermohaline induced mixing after the bump in the luminosity function.

    • HIRES is the high-resolution spectrograph of the European Extremely Large Telescope at optical and near-infrared wavelengths. It consists of three fibre-fed spectrographs providing a wavelength coverage of 0.4-1.8 µm (goal 0.35-2.4 µm) at a spectral resolution of 100,000. The fibre-feeding allows HIRES to have several, interchangeable observing modes including a SCAO module and a small diffraction-limited IFU in the NIR. Therefore, it will be able to operate both in seeing- and diffraction-limited modes. Its modularity will ensure that HIRES can be placed entirely on the Nasmyth platform, if enough mass and volume is available, or part on the Nasmyth and part in the Coud`e room. ELT-HIRES has a wide range of science cases spanning nearly all areas of research in astrophysics and even fundamental physics. Among the top science cases there are the detection of biosignatures from exoplanet atmospheres, finding the fingerprints of the first generation of stars (PopIII), tests on the stability of Nature’s fundamental couplings, and the direct detection of the cosmic acceleration. The HIRES consortium is composed of more than 30 institutes from 14 countries, forming a team of more than 200 scientists and engineers.

  • The Gaia-ESO Survey: The analysis of high-resolution UVES spectra of FGK-type stars

    • Context. The ongoing Gaia-ESO Public Spectroscopic Survey is using FLAMES at the VLT to obtain high-quality medium-resolution Giraffe spectra for about 10(5) stars and high-resolution UVES spectra for about 5000 stars. With UVES, the Survey has already observed 1447 FGK-type stars. Aims. These UVES spectra are analyzed in parallel by several state-of-the-art methodologies. Our aim is to present how these analyses were implemented, to discuss their results, and to describe how a final recommended parameter scale is defined. We also discuss the precision (method-to-method dispersion) and accuracy (biases with respect to the reference values) of the final parameters. These results are part of the Gaia-ESO second internal release and will be part of its first public release of advanced data products. Methods. The final parameter scale is tied to the scale defined by the Gaia benchmark stars, a set of stars with fundamental atmospheric parameters. In addition, a set of open and globular clusters is used to evaluate the physical soundness of the results. Each of the implemented methodologies is judged against the benchmark stars to define weights in three different regions of the parameter space. The final recommended results are the weighted medians of those from the individual methods. Results. The recommended results successfully reproduce the atmospheric parameters of the benchmark stars and the expected T-eff-log g relation of the calibrating clusters. Atmospheric parameters and abundances have been determined for 1301 FGK-type stars observed with UVES. The median of the method-to-method dispersion of the atmospheric parameters is 55K for T-eff, 0.13dex for log g and 0.07 dex for [Fe/H]. Systematic biases are estimated to be between 50-100 K for T-eff, 0.10-0.25 dex for log g and 0.05-0.10 dex for [Fe/H]. Abundances for 24 elements were derived: C, N, O, Na, Mg, Al, Si, Ca, Sc, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Y, Zr, Mo, Ba, Nd, and Eu. The typical method-to-method dispersion of the abundances varies between 0.10 and 0.20 dex. Conclusions. The Gaia-ESO sample of high-resolution spectra of FGK-type stars will be among the largest of its kind analyzed in a homogeneous way. The extensive list of elemental abundances derived in these stars will enable significant advances in the areas of stellar evolution and Milky Way formation and evolution.

  • The Gaia-ESO survey : Mapping the shape and evolution of the radial abundance gradients with open clusters

    • Context: The spatial distribution of elemental abundances and their time evolution are among the major constraints to disentangling the scenarios of formation and evolution of the Galaxy.Aims: In this paper we used the sample of open clusters available in the final release of the Gaia-ESO survey to trace the Galactic radial abundance and abundance-to-iron ratio gradients, and their time evolution.Methods: We selected member stars in 62 open clusters, with ages from 0.1 to about 7 Gyr, located in the Galactic thin disc at galactocentric radii (R-GC) from about 6 to 21 kpc. We analysed the shape of the resulting [Fe/H] gradient, the average gradients [El/H] and [El/Fe] combining elements belonging to four different nucleosynthesis channels, and their individual abundance and abundance ratio gradients. We also investigated the time evolution of the gradients dividing open clusters in three age bins.Results: The [Fe/H] gradient has a slope of -0.054 dex kpc(-1). It can be better approximated with a two-slope shape, steeper for R-GC <= 11.2 kpc and flatter in the outer regions. We saw different behaviours for elements belonging to different channels. For the time evolution of the gradient, we found that the youngest clusters (age < 1 Gyr) in the inner disc have lower metallicity than their older counterparts and that they outline a flatter gradient. We considered some possible explanations, including the effects of gas inflow and migration. We suggest that the most likely one may be related to a bias introduced by the standard spectroscopic analysis producing lower metallicities in the analysis of low-gravity stars.Conclusions: To delineate the shape of the 'true' gradient, we should most likely limit our analysis to stars with low surface gravity log g> 2.5 and microturbulent parameter xi< 1.8 km s(-1). Based on this reduced sample, we can conclude that the gradient has minimally evolved over the time-frame outlined by the open clusters, indicating a slow and stationary formation of the thin disc over the last 3 Gyr. We found a secondary role of cluster migration in shaping the gradient, with a more prominent role of migration for the oldest clusters.

  • The Gaia-ESO Survey : properties of newly discovered Li-rich giants

    • Aims. We report 20 new lithium-rich giants discovered within the Gaia-ESO Survey, including the first Li-rich giant with an evolutionary stage confirmed by CoRoT (Convection, Rotation and planetary Transits) data. We present a detailed overview of the properties of these 20 stars. Methods. Atmospheric parameters and abundances were derived in model atmosphere analyses using medium-resolution GIRAFFE or high-resolution UVES (Ultraviolet and Visual Echelle Spectrograph) spectra. These results are part of the fifth internal data release of the Gaia-ESO Survey. The Li abundances were corrected for non-local thermodynamical equilibrium effects. Other stellar properties were investigated for additional peculiarities (the core of strong lines for signs of magnetic activity, infrared magnitudes, rotational velocities, chemical abundances, and Galactic velocities). We used Gaia DR2 parallaxes to estimate distances and luminosities. Results. The giants have A(Li) > 2.2 dex. The majority of them (14 of 20 stars) are in the CoRoT fields. Four giants are located in the field of three open clusters, but are not members. Two giants were observed in fields towards the Galactic bulge, but likely lie in the inner disc. One of the bulge field giants is super Li-rich with A(Li) = 4.0 dex. Conclusions. We identified one giant with infrared excess at 22 mu m. Two other giants, with large v sin i, might be Li-rich because of planet engulfment. Another giant is found to be barium enhanced and thus could have accreted material from a former asymptotic giant branch companion. Otherwise, in addition to the Li enrichment, the evolutionary stages are the only other connection between these new Li-rich giants. The CoRoT data confirm that one Li-rich giant is at the core-He burning stage. The other giants are concentrated in close proximity to the red giant branch luminosity bump, the core-He burning stages, or the early-asymptotic giant branch. This is very clear from the Gaia-based luminosities of the Li-rich giants. This is also seen when the CoRoT Li-rich giants are compared to a larger sample of 2252 giants observed in the CoRoT fields by the Gaia-ESO Survey, which are distributed throughout the red giant branch in the T-eff-log g diagram. These observations show that the evolutionary stage is a major factor for the Li enrichment in giants. Other processes, such as planet accretion, contribute at a smaller scale.

  • Outcomes after Haploidentical Stem Cell Transplantation with Post-Transplantation Cyclophosphamide in Patients with Primary Immunodeficiency Diseases

    • Allogeneic hematopoietic stem cell transplantation (HCT) can cure primary immunodeficiency diseases (PID). When a HLA-matched donor is not available, a haploidentical family donor may be considered. The use of T cell-replete haploidentical HCT with post-transplantation cyclophosphamide (haplo-PTCy) in children with PID has been reported in few case series. A donor is usually readily available, and haplo-PTCy can be used in urgent cases. We studied the outcomes of 73 patients with PID who underwent haplo-PTCy, including 55 patients who did so as a first transplantation and 18 who did so as a salvage transplantation after graft failure of previous HCT. The median patient age was 1.6 years. Most of the children were male (n = 54) and had active infection at the time of transplantation (n = 50); 10 children had severe organ damage. The diagnosis was severe combined immunodeficiency (SCID) in 34 patients and non-SCID in 39 (Wiskott-Aldrich syndrome; n = 14; chronic granulomatous disease, n = 10; other PID, n = 15). The median duration of follow-up of survivors was 2 years. The cumulative incidence of neutrophil recovery was 88% in the SCID group and 84% in non-SCID group and was 81% for first transplantations and 83% after a salvage graft. At 100 days, the cumulative incidence of acute GVHD grade II-IV and III-IV was 33% and 14%, respectively. The majority of patients reached 200/mu L CD4(+) and 1000/mu L CD3(+) cell counts between 3 and 6 months. The estimated 2-year overall survival was 66%; it was 64% for SCID patients and 65% for non-SCID patients and 63% for first HCT and 77% for salvage transplantations. Twenty-five patients died, most of them due to infection early after transplantation (before 100 days). In conclusion, haplo-PTCy is a feasible procedure, can cure two-thirds of children with PID, and can be used as rescue treatment for previous graft failure.     • The CRyogenic InfraRed Echelle Spectrograph (CRIRES) Upgrade project CRIRES+ extended the capabilities of CRIRES. It transformed this VLT instrument into a cross-dispersed spectrograph to increase the wavelength range that is covered simultaneously by up to a factor of ten. In addition, a new detector focal plane array of three Hawaii 2RG detectors with a 5.3 mu m cutoff wavelength replaced the existing detectors. Amongst many other improvements, a new spectropolarimetric unit was added and the calibration system has been enhanced. The instrument was installed at the VLT on Unit Telescope 3 at the beginning of 2020 and successfully commissioned and verified for science operations during 2021, partly remotely from Europe due to the COVID-19 pandemic. The instrument was subsequently offered to the community from October 2021 onwards. This article describes the performance and capabilities of the upgraded instrument and presents on sky results.

  • Gaia -ESO survey : Lithium abundances in open cluster Red Clump stars

    • Context. It has recently been suggested that all giant stars with masses below 2 M? suffer an episode of surface lithium enrichment between the tip of the red giant branch (RGB) and the red clump (RC). Aims. We test if the above result can be confirmed in a sample of RC and RGB stars that are members of open clusters. Methods. We discuss Li abundances in six open clusters with ages between 1.5 and 4.9 Gyr (turn-off masses between 1.1 and 1.7 M? ). We compare these observations with the predictions of different models that include rotation-induced mixing, thermohaline instability, mixing induced by the first He flash, and energy losses by neutrino magnetic moment. Results. In six clusters, we find close to 35% of RC stars have Li abundances that are similar or higher than those of upper RGB stars. This can be a sign of fresh Li production. Because of the extra-mixing episode connected to the luminosity bump, the expectation has been for RC stars to have systematically lower surface Li abundances. However, we cannot confirm that this possible Li production is ubiquitous. For about 65% of RC giants, we can only determine upper limits in abundances that could be hiding very low Li content. Conclusions. Our results indicate the possibility that Li is being produced in the RC, at levels that would not typically permit the classification of these the stars as Li rich. The determination of their carbon isotopic ratio would help to confirm that the RC giants have suffered extra mixing followed by subsequent Li enrichment. The Li abundances of the RC stars can be qualitatively explained by the models including an additional mixing episode close to the He flash.

    • We report the discovery of two Li-rich giants, with A(Li) ~ 1.50, in an analysis of a sample of 40 giants of the open cluster Trumpler 20 (with turnoff mass ~1.8 M⊙). The cluster was observed in the context of the Gaia-ESO Survey. Methods. The atmospheric parameters and Li abundances were derived using high-resolution UVES spectra. The Li abundances were corrected for nonlocal thermodynamical equilibrium (non-LTE) effects. Results. Only upper limits of the Li abundance could be determined for the majority of the sample. Two giants with detected Li turned out to be Li rich: star MG 340 has A(Li)non-LTE = 1.54 ± 0.21 dex and star MG 591 has A(Li)non-LTE = 1.60 ± 0.21 dex. Star MG 340 is on average ~0.30 dex more rich in Li than stars of similar temperature, while for star MG 591 this difference is on average ~0.80 dex. Carbon and nitrogen abundances indicate that all stars in the sample have completed the first dredge-up. Conclusions. The Li abundances in this unique sample of 40 giants in one open cluster clearly show that extra mixing is the norm in this mass range. Giants with Li abundances in agreement with the predictions of standard models are the exception. To explain the two Li-rich giants, we suggest that all events of extra mixing have been inhibited. This includes rotation-induced mixing during the main sequence and the extra mixing at the red giant branch luminosity bump. Such inhibition has been suggested in the literature to occur because of fossil magnetic fields in red giants that are descendants of main-sequence Ap-type stars.

    • High-resolution infrared spectroscopy plays an important role in astrophysics from the search for exoplanets to cosmology. Yet, many existing infrared spectrographs are limited by a rather small simultaneous wavelength coverage. The AO assisted CRIRES instrument, installed at the ESO VLT on Paranal, is one of the few IR (0.92-5.2 mu m) high-resolution spectrographs in operation since 2006. However it has a limitation that hampers its efficient use: the wavelength range covered in a single exposure is limited to similar to 15 nanometers. The CRIRES Upgrade project (CRIRES+) will transform CRIRES into a cross-dispersed spectrograph and will also add new capabilities. By introducing cross-dispersion elements the simultaneously covered wavelength range will be increased by at least a factor of 10 with respect to the present configuration, while the operational wavelength range will be preserved. For advanced wavelength calibration, new custom made absorption gas cells and etalons will be added. A spectro-polarimetric unit will allow one for the first time to record circularly polarized spectra at the highest spectral resolution. This will be all supported by a new data reduction software which will allow the community to take full advantage of the new capabilities of CRIRES+.

    • CRIRES, the ESO high resolution infrared spectrometer, is a unique instrument which allows astronomers to access a parameter space which up to now was largely uncharted. In its current setup, it consists of a single-order spectrograph providing long-slit, single-order spectroscopy with resolving power up to R=100,000 over a quite narrow spectral range. This has resulted in sub-optimal efficiency and use of telescope time for all the scientific programs requiring broad spectral coverage of compact objects (e.g. chemical abundances of stars and intergalactic medium, search and characterization of extra-solar planets). To overcome these limitations, a consortium was set-up for upgrading CRIRES to a cross-dispersed spectrometer, called CRIRES+. This paper presents the updated optical design of the cross-dispersion module for CRIRES+. This new module can be mounted in place of the current pre-disperser unit. The new system yields a factor of >10 increase in simultaneous spectral coverage and maintains a quite long slit (10"), ideal for observations of extended sources and for precise sky-background subtraction.

  • The Gaia -ESO Survey: Galactic evolution of lithium at high metallicity

    • Context. Reconstructing the Galactic evolution of lithium (Li) is the main tool used to constrain the source(s) of Li enrichment in the Galaxy. Recent results have suggested a decline in Li at supersolar metallicities, which may indicate reduced production. Aims. We exploit the unique characteristics of the Gaia-ESO Survey open star cluster sample to further investigate this issue and to better constrain the evolution of Li at high metallicity. Methods. We trace the upper envelope of Li abundance versus metallicity evolution using 18 clusters and considering members that should not have suffered any Li depletion. Results. At variance with previous claims, we do not find any evidence of a Li decrease at high metallicity. The most metal-rich clusters in the sample ([Fe/H] = ∼0.3) actually show the highest Li abundances, with A(Li) > 3.4. Our results clearly show that previous findings, which were based on field stars, were affected by selection effects. The metal-rich population in the solar neighbourhood is composed of relatively old and cool stars that have already undergone some Li depletion; hence, their measured Li does not represent the initial interstellar medium abundance, but a lower limit to it.

    • The results of hematopoietic stem cell transplant (HSCT) for primary immunodeficiency diseases (PID) have been improving over time. Unfortunately, developing countries do not experience the same results. This first report of Brazilian experience of HSCT for PID describes the development and results in the field. We included data from transplants in 221 patients, performed at 11 centers which participated in the Brazilian collaborative group, from July 1990 to December 2015. The majority of transplants were concentrated in one center (n=123). The median age at HSCT was 22months, and the most common diseases were severe combined immunodeficiency (SCID) (n=67) and Wiskott-Aldrich syndrome (WAS) (n=67). Only 15 patients received unconditioned transplants. Cumulative incidence of GVHD grades II to IV was 23%, and GVHD grades III to IV was 10%. The 5-year overall survival was 71.6%. WAS patients had better survival compared to other diseases. Most deaths (n=53) occurred in the first year after transplantation mainly due to infection (55%) and GVHD (13%). Although transplant for PID patients in Brazil has evolved since its beginning, we still face some challenges like delayed diagnosis and referral, severe infections before transplant, a limited number of transplant centers with expertise, and resources for more advanced techniques. Measures like newborn screening for SCID may hasten the diagnosis and ameliorate patients' conditions at the moment of transplant.

    • Limited clinical data are available to assess whether the sequencing of cyclophosphamide (Cy) and total body irradiation (TBI) changes outcomes. We evaluated the sequence in 1769 (CyTBI, n = 948; TBICy, n = 821) recipients of related or unrelated hematopoietic cell transplantation who received TBI (1200 to 1500 cGY) for acute leukemia from 2003 to 2010. The 2 cohorts were comparable for median age, performance score, type of leukemia, first complete remission, Philadelphia chromosome positive acute lymphoblastic leukemia, HLA-matched siblings, stem cell source, antithymocyte globulin use, TBI dose, and type of graft-versus-host disease (GVHD) prophylaxis. The sequence of TBI did not significantly affect transplantation-related mortality (24% versus 23% at 3 years, P = .67; relative risk, 1.01; P = .91), leukemia relapse (27% versus 29% at 3 years, P = .34; relative risk,.89, P = .18), leukemia-free survival (49% versus 48% at 3 years, P = .27; relative risk,.93; P = .29), chronic GVHD (45% versus 47% at 1 year, P = .39; relative risk,.9; P = .11), or overall survival (53% versus 52% at 3 years, P = .62; relative risk,.96; P = .57) for CyTBI and TBICy, respectively. Corresponding cumulative incidences of sinusoidal obstruction syndrome were 4% and 6% at 100 days (P = .08), respectively. This study demonstrates that the sequence of Cy and TBI does not impact transplantation outcomes and complications in patients with acute leukemia undergoing hematopoietic cell transplantation with myeloablative conditioning.

    • The brain is a complex, multiscale dynamical system composed of many interacting regions. Knowledge of the spatiotemporal organization of these interactions is critical for establishing a solid understanding of the brain’s functional architecture and the relationship between neural dynamics and cognition in health and disease. The possibility of studying these dynamics through careful analysis of neuroimaging data has catalyzed substantial interest in methods that estimate time-resolved fluctuations in functional connectivity (often referred to as “dynamic” or time-varying functional connectivity; TVFC). At the same time, debates have emerged regarding the application of TVFC analyses to resting fMRI data, and about the statistical validity, physiological origins, and cognitive and behavioral relevance of resting TVFC. These and other unresolved issues complicate interpretation of resting TVFC findings and limit the insights that can be gained from this promising new research area. This article brings together scientists with a variety of perspectives on resting TVFC to review the current literature in light of these issues. We introduce core concepts, define key terms, summarize controversies and open questions, and present a forward-looking perspective on how resting TVFC analyses can be rigorously and productively applied to investigate a wide range of questions in cognitive and systems neuroscience.

    • The presence of monosomal karyotype (MK+) in acute myeloid leukemia (AML) is associated with dismal outcomes. We evaluated the impact of MK+ in AML (MK+AML, n = 240) and in myelodysplastic syndrome (MDS) (MK+MDS, n = 221) on hematopoietic cell transplantation outcomes compared with other cytogenetically defined groups (AML, n = 3360; MDS, n = 1373) as reported to the Center for International Blood and Marrow Transplant Research from 1998 to 2011. MK+AML was associated with higher disease relapse (hazard ratio, 1.98; P < .01), similar transplantation-related mortality (TRM) (hazard ratio, 1.01; P = .90), and worse survival (hazard ratio, 1.67; P < .01) compared with those outcomes for other cytogenetically defined AML. Among patients with MDS, MK+ MDS was associated with higher disease relapse (hazard ratio, 2.39; P < .01), higher TRM (hazard ratio, 1.80; P < .01), and worse survival (HR, 2.02; P < .01). Subset analyses comparing chromosome 7 abnormalities (del7/7q) with or without MK+ demonstrated higher mortality for MK+ disease in for both AML (hazard ratio, 1.72; P < .01) and MDS (hazard ratio, 1.79; P < .01). The strong negative impact of MK+ in myeloid malignancies was observed in all age groups and using either myeloablative or reduced-intensity conditioning regimens. Alternative approaches to mitigate disease relapse in this population are needed.

    • The rising incidence of pediatric obesity may significantly affect bone marrow transplantation (BMT) outcomes. We analyzed outcomes in 3687 children worldwide who received cyclophosphamide-based BMT regimens for leukemias between 1990 and 2007. Recipients were classified according to age-adjusted body mass index (BMI) percentiles as underweight (UW), at risk of UW (RUW), normal, overweight (OW), or obese (OB). Median age and race were similar in all groups. Sixty-one percent of OB children were from the United States/Canada. Three-year relapse-free and overall survival ranged from 48% to 52% (P = .54) and 55% to 58% (P = .81) across BMI groups. Three-year leukemia relapses were 33%, 33%, 29%, 25%, and 21% in the UW, RUW, normal, OW, and OB groups, respectively (P < .001). Corresponding cumulative incidences for transplant-related mortality (TRM) were 18%, 19%, 21%, 22%, and 28% (P < .01). Multivariate analysis demonstrated a decreased risk of relapse compared with normal BMI (relative risk [RR] = 0.73; P < .01) and a trend toward higher TRM (RR = 1.28; P = .014). BMI in children is not significantly associated with different survival after BMT for hematologic malignancies. Obese children experience less relapse posttransplant compared with children with normal BMI; however, this benefit is offset by excess in TRM.

    • PURPOSE Allogeneic hematopoietic cell transplantation (HCT) can cure bone marrow failure in patients with Fanconi anemia (FA). Data on outcomes in patients with pretransplantation cytogenetic abnormalities, myelodysplastic syndrome (MDS), or acute leukemia have not been separately analyzed. PATIENTS AND METHODS We analyzed data on 113 patients with FA with cytogenetic abnormalities (n = 54), MDS (n = 45), or acute leukemia (n = 14) who were reported to the Center for International Blood and Marrow Transplant Research from 1985 to 2007. Results Neutrophil recovery occurred in 78% and 85% of patients at days 28 and 100, respectively. Day 100 cumulative incidences of acute graft-versus-host disease grades B to D and C to D were 26% (95% CI, 19% to 35%) and 12% (95% CI, 7% to 19%), respectively. Survival probabilities at 1, 3, and 5 years were 64% (95% CI, 55% to 73%), 58% (95% CI, 48% to 67%), and 55% (95% CI, 45% to 64%), respectively. In univariate analysis, younger age was associated with superior 5-year survival (≤ v > 14 years: 69% [95% CI, 57% to 80%] v 39% [95% CI, 26% to 53%], respectively; P = .001). In transplantations from HLA-matched related donors (n = 82), younger patients (≤ v > 14 years: 78% [95% CI, 64% to 90%] v 34% [95% CI, 20% to 50%], respectively; P < .001) and patients with cytogenetic abnormalities only versus MDS/acute leukemia (67% [95% CI, 52% to 81%] v 43% [95% CI, 27% to 59%], respectively; P = .03) had superior 5-year survival. CONCLUSION Our analysis indicates that long-term survival for patients with FA with cytogenetic abnormalities, MDS, or acute leukemia is achievable. Younger patients and recipients of HLA-matched related donor transplantations who have cytogenetic abnormalities only have the best survival.

    • We report on long-term survival in 157 patients with Fanconi anemia (FA) who survived 2 years or longer after their first transplantation with a median follow-up of 9 years. Marrow failure (80%) was the most common indication for transplantation. There were 20 deaths beyond 2 years after transplantation, with 12 of the deaths occurring beyond 5 years after transplantation. Donor chimerism was available for 149 patients: 112 (76%) reported > 95% chimerism, 27 (18%) reported 90% to 95% chimerism, and 8 (5%) reported 20% to 89% donor chimerism. Two patients have < 20% donor chimerism. The 10- and 15-year probabilities of survival were 90% and 79%, respectively. Results of multivariate analysis showed higher mortality risks for transplantations before 2003 (hazard ratio [HR], 7.87; P =.001), chronic graft-versus-host disease (GVHD) (HR, 3.80; P =.004) and squamous cell carcinoma after transplantation (HR, 38.17; P <.0001). The predominant cause of late mortality was squamous cell carcinoma, with an incidence of 8% and 14% at 10 and 15 years after transplantation, respectively, and was more likely to occur in those with chronic GVHD. Other causes of late mortality included chronic GVHD, infection, graft failure, other cancers, and hemorrhage. Although most patients are disease free and functional long term, our data support aggressive surveillance for long periods to identify those at risk for late mortality.

  • PDRs4All: IV. An embarrassment of riches: Aromatic infrared bands in the Orion Bar

    • Context. Mid-infrared observations of photodissociation regions (PDRs) are dominated by strong emission features called aromatic infrared bands (AIBs). The most prominent AIBs are found at 3.3, 6.2, 7.7, 8.6, and 11.2 µm. The most sensitive, highest-resolution infrared spectral imaging data ever taken of the prototypical PDR, the Orion Bar, have been captured by JWST. These high-quality data allow for an unprecedentedly detailed view of AIBs. Aims. We provide an inventory of the AIBs found in the Orion Bar, along with mid-IR template spectra from five distinct regions in the Bar: the molecular PDR (i.e. the three H2 dissociation fronts), the atomic PDR, and the H II region. Methods. We used JWST NIRSpec IFU and MIRI MRS observations of the Orion Bar from the JWST Early Release Science Program, PDRs4All (ID: 1288). We extracted five template spectra to represent the morphology and environment of the Orion Bar PDR. We investigated and characterised the AIBs in these template spectra. We describe the variations among them here. Results. The superb sensitivity and the spectral and spatial resolution of these JWST observations reveal many details of the AIB emission and enable an improved characterization of their detailed profile shapes and sub-components. The Orion Bar spectra are dominated by the well-known AIBs at 3.3, 6.2, 7.7, 8.6, 11.2, and 12.7 µm with well-defined profiles. In addition, the spectra display a wealth of weaker features and sub-components. The widths of many AIBs show clear and systematic variations, being narrowest in the atomic PDR template, but showing a clear broadening in the H II region template while the broadest bands are found in the three dissociation front templates. In addition, the relative strengths of AIB (sub-)components vary among the template spectra as well. All AIB profiles are characteristic of class A sources as designated by Peeters (2022, A&A, 390, 1089), except for the 11.2 µm AIB profile deep in the molecular zone, which belongs to class B11.2. Furthermore, the observations show that the sub-components that contribute to the 5.75, 7.7, and 11.2 µm AIBs become much weaker in the PDR surface layers. We attribute this to the presence of small, more labile carriers in the deeper PDR layers that are photolysed away in the harsh radiation field near the surface. The 3.3/11.2 AIB intensity ratio decreases by about 40% between the dissociation fronts and the H II region, indicating a shift in the polycyclic aromatic hydrocarbon (PAH) size distribution to larger PAHs in the PDR surface layers, also likely due to the effects of photochemistry. The observed broadening of the bands in the molecular PDR is consistent with an enhanced importance of smaller PAHs since smaller PAHs attain a higher internal excitation energy at a fixed photon energy. Conclusions. Spectral-imaging observations of the Orion Bar using JWST yield key insights into the photochemical evolution of PAHs, such as the evolution responsible for the shift of 11.2 µm AIB emission from class B11.2 in the molecular PDR to class A11.2 in the PDR surface layers. This photochemical evolution is driven by the increased importance of FUV processing in the PDR surface layers, resulting in a “weeding out” of the weakest links of the PAH family in these layers. For now, these JWST observations are consistent with a model in which the underlying PAH family is composed of a few species: the so-called ‘grandPAHs’.

    • Renal dysfunction is a recognized risk factor for mortality after allogeneic hematopoietic cell transplantation (alloHCT), yet our understanding of the effect of different levels of renal dysfunction at time of transplantation on outcomes remains limited. This study explores the impact of different degrees of renal dysfunction on HCT outcomes and examines whether the utilization of incremental degrees of renal dysfunction based on estimated glomerular filtration rate (eGFR) improve the predictability of the hematopoietic cell transplantation comorbidity index (HCT-CI). The study population included 2 cohorts: cohort 1, comprising patients age >= 40 years who under went alloHCT for treatment of hematologic malignancies between 2008 and 2016 (n = 13,505; cohort selected given a very low incidence of renal dysfunction in individuals age <40 years), and cohort 2, comprising patients on dialysis at the time of HCT (n = 46). eGFR was measured using the Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) method. The patients in cohort 1 were assigned into 4 categories-eGFR >= 90 mL/min (n = 7062), eGFR 60 to 89 mL/min (n = 5264), eGFR 45 to 59 mL/min (n = 897), and eGFR <45 mL/min (n=282)-to assess the impact of degree of renal dysfunction on transplantation outcomes. Transplantation outcomes in patients on dialysis at the time of alloHCT were analyzed separately. eGFR <60 mL/min was associated with an increased risk for nonrelapse mortality (NRM) and requirement for dialysis post-HCT. Compared with the eGFR >= 90 group, the hazard ratio (HR) for NRM was 1.46 (P = .0001) for the eGFR 45 to 59 mL/min group and 1.74 (P = .004) for the eGFR <45 mL/min group. Compared with the eGFR >= 90 mL/min group, the eGFR 45 to 59 mL/min group (HR, 2.45; P < .0001) and the eGFR <45 mL/min group (HR, 3.09; P < .0001) had a higher risk of renal failure necessitating dialysis after alloHCT. In addition, eGFR <45 mL/min was associated with an increased overall mortality (HR, 1.63; P < .0001). An eGFR-based revised HCT-CI was also developed and shown to be predictive of overall survival (OS) and NRM, with predictive performance similar to the original HCT-CI. Among 46 patients on dialysis at alloHCT, the 1-year probability of OS was 20%, and that of NRM was 67%. The degree of pretransplantation renal dysfunction is an independent predictor of OS, NRM, and probability of needing dialysis after alloHCT. An eGFR-based HCT-CI is a validated index for predicting outcomes in adults with hematologic malignancies undergoing alloHCT. The outcomes of alloHCT recipients on dialysis are dismal; therefore, one should strongly weigh the significant risks of being on hemodialysis as a factor in determining alloHCT candidacy.

The James Webb Space Telescope (JWST) has captured the most detailed and sharpest infrared (IR) images ever taken of the inner region of the Orion Nebula, the nearest massive star formation region, and a prototypical highly irradiated dense photo-dissociation region (PDR). Aims. We investigate the fundamental interaction of far-ultraviolet (FUV) photons with molecular clouds. The transitions across the ionization front (IF), dissociation front (DF), and the molecular cloud are studied at high-angular resolution. These transitions are relevant to understanding the effects of radiative feedback from massive stars and the dominant physical and chemical processes that lead to the IR emission that JWST will detect in many Galactic and extragalactic environments. Methods. We utilized NIRCam and MIRI to obtain sub-arcsecond images over ∼150′′ and 42′′ in key gas phase lines (e.g., Pa α, Br α, [FeII] 1.64 µm, H2 1–0 S(1) 2.12 µm, 0–0 S(9) 4.69 µm), aromatic and aliphatic infrared bands (aromatic infrared bands at 3.3–3.4 µm, 7.7, and 11.3 µm), dust emission, and scattered light. Their emission are powerful tracers of the IF and DF, FUV radiation field and density distribution. Using NIRSpec observations the fractional contributions of lines, AIBs, and continuum emission to our NIRCam images were estimated. A very good agreement is found for the distribution and intensity of lines and AIBs between the NIRCam and NIRSpec observations. Results. Due to the proximity of the Orion Nebula and the unprecedented angular resolution of JWST, these data reveal that the molecular cloud borders are hyper structured at small angular scales of ∼0.1–1′′ (∼0.0002–0.002 pc or ∼40–400 au at 414 pc). A diverse set of features are observed such as ridges, waves, globules and photoevaporated protoplanetary disks. At the PDR atomic to molecular transition, several bright features are detected that are associated with the highly irradiated surroundings of the dense molecular condensations and embedded young star. Toward the Orion Bar PDR, a highly sculpted interface is detected with sharp edges and density increases near the IF and DF. This was predicted by previous modeling studies, but the fronts were unresolved in most tracers. The spatial distribution of the AIBs reveals that the PDR edge is steep and is followed by an extensive warm atomic layer up to the DF with multiple ridges. A complex, structured, and folded H0/H2 DF surface was traced by the H2 lines. This dataset was used to revisit the commonly adopted 2D PDR structure of the Orion Bar as our observations show that a 3D “terraced” geometry is required to explain the JWST observations. JWST provides us with a complete view of the PDR, all the way from the PDR edge to the substructured dense region, and this allowed us to determine, in detail, where the emission of the atomic and molecular lines, aromatic bands, and dust originate. Conclusions. This study offers an unprecedented dataset to benchmark and transform PDR physico-chemical and dynamical models for the JWST era. A fundamental step forward in our understanding of the interaction of FUV photons with molecular clouds and the role of FUV irradiation along the star formation sequence is provided.

    • Bone marrow (BM) is the preferred graft source for hematopoietic stem cell transplantation (HSCT) in severe aplastic anemia (SAA) compared with mobilized peripheral blood stem cells (PBSCs). We hypothesized that this recommendation may not apply to those regions where patients present later in their disease course, with heavier transfusion load and with higher graft failure rates. Patients with SAA who received HSCT from an HLA-matched sibling donor from 1995 to 2009 and reported to the Center for International Blood and Marrow Transplant Research or the Japan Society for Hematopoietic Cell Transplantation were analyzed. The study population was categorized by gross national income per capita and region/countries into 4 groups. Groups analyzed were high-income countries (HIC), which were further divided into United States-Canada (n=486) and other HIC (n=1264); upper middle income (UMIC) (n=482); and combined lower-middle, low-income countries (LM-LIC) (n=142). In multivariate analysis, overall survival (OS) was highest with BM as graft source in HIC compared with PBSCs in all countries or BM in UMIC or LM-LIC (P<.001). There was no significant difference in OS between BM and PBSCs in UMIC (P=.32) or LM-LIC (P=.23). In LM-LIC the 28-day neutrophil engraftment was higher with PBSCs compared with BM (97% versus 77%, P<.001). Chronic graft-versus-host disease was significantly higher with PBSCs in all groups. Whereas BM should definitely be the preferred graft source for HLA-matched sibling HSCT in SAA, PBSCs may be an acceptable alternative in countries with limited resources when treating patients at high risk of graft failure and infective complications.

    • Transition metal oxides are promising electrocatalysts for water oxidation, i.e., the oxygen evolution reaction (OER), which is critical in electrochemical production of non-fossil fuels. The involvement of oxidation state changes of the metal in OER electrocatalysis is increasingly recognized in the literature. Tracing these oxidation states under operation conditions could provide relevant information for performance optimization and development of durable catalysts, but further methodical developments are needed. Here, we propose a strategy to use single-energy X-ray absorption spectroscopy for monitoring metal oxidation-state changes during OER operation with millisecond time resolution. The procedure to obtain time-resolved oxidation state values, using two calibration curves, is explained in detail. We demonstrate the significance of this approach as well as possible sources of data misinterpretation. We conclude that the combination of X-ray absorption spectroscopy with electrochemical techniques allows us to investigate the kinetics of redox transitions and to distinguish the catalytic current from the redox current. Tracking of the oxidation state changes of Co ions in electrodeposited oxide films during cyclic voltammetry in neutral pH electrolyte serves as a proof of principle.

    • We present a detailed chemical analysis of 22 stars along thecolour-magnitude sequence of the intermediate-age (1.7 Gyr) open clusterIC 4651, based on high-resolution, high S/N ratio spectra from UVES/VLT.IC 4651 thus becomes one of the few open clusters for which a detailedcomposition analysis exists for stars spanning 3.5 mag, from solar-typemain-sequence stars to giants above the RGB clump. In a strictcomparison with the Sun, we find for the cluster a well-defined Feabundance of [Fe/H] = 0.10 ± 0.03 (internal errors), with areddening E(b-y) = 0.091. We also derive abundances for the αelements Mg, Si, Ti, and Ca and find a moderate enhancement of the threeformer elements, in excellent agreement with the results for field starsof similar Fe abundance. Among the Fe group elements, Cr and Ni areslightly overabundant ([X/Fe] ∼ 0.05). As also observed in M 67, theNa abundance among the giants is more than 0.2 dex higher than in thedwarfs. We interpret this effect as real, and due to dredge-up of23Na in the giants. Four turnoff stars, all fairly rapidrotators (v sin i ≥ 25 km s-1), appear to have higher[Fe/H] than the other stars; our tests show that, while a spuriousenhancement of 0.1 dex can be produced by the effect of high rotation onour spectral analysis, this cannot explain the whole difference. Thesestars appear overabundant because we tend to overestimate theireffective temperatures by forcing excitation equilibrium. Li abundanceshave been computed for all the stars and show a well-defined pattern:the Solar-type stars have an almost constant Li abundance, just belowthat of the Hyades, and the Li-dip is pronounced and well determined.Turnoff stars just above the dip have a ``cosmic'' Li abundance, butwithin a very small range of magnitudes (0.25 mag) higher on theturnoff, the Li level drops by more than a factor 10. This cannot be duejust to dilution; rather some extra mixing is required. Among thegiants, two probable clump stars show detectable Li, while all the other(likely) RGB stars do not - as is also observed in the similar clusterNGC 3680. None of these patterns can be explained by classical stellarevolution models. Again, some extra mixing is required. We show thatrotating stellar models including the most recent developments formeridional circulation and turbulence by shear instabilities explainvery well the behaviour of the lithium abundance along thecolour-magnitude diagram of IC 4651, including subgiant and giantphases. The possibility remains open that the giants exhibiting thehighest lithium abundances are actual RGB bump stars which have justbeen through the so-called ``lithium flash".Based on observations collected at the ESO VLT, Paranal Observatory,Chile.

  • Gravitational redshifts in main-sequence and giant stars

    • Context. Precise analyses of stellar radial velocities is able to reveal intrinsic causes of the wavelength shifts of spectral lines (other than Doppler shifts due to radial motion), such as gravitational redshifts and convective blueshifts. Aims. Gravitational redshifts in solar-type main-sequence stars are expected to be some 500 m s(-1) greater than those in giants. We search for this difference in redshifts among groups of open-cluster stars that share the same average space motion and thus have the same average Doppler shift. Methods. We observed 144 main-sequence stars and cool giants in the M67 open cluster using the ESO FEROS spectrograph and obtained radial velocities by means of cross-correlation with a spectral template. Binaries and doubtful members were not analyzed, and average spectra were created for different classes of stars. Results. The M67 dwarf and giant radial-velocity distributions are each well represented by Gaussian functions, which share the same apparent average radial velocity to within similar or equal to 100 m s(-1). In addition, dwarfs in M67 appear to be dynamically hotter (sigma = 0.90 km s(-1)) than giants (sigma = 0.68 km s(-1)). Conclusions. We fail to detect any difference in the gravitational redshifts of giants and MS stars. This is probably because of the differential wavelength shifts produced by the different hydrodynamics of dwarf and giant atmospheres. Radial-velocity differences measured between unblended lines in averaged spectra vary with line-strength: stronger lines are more blueshifted in dwarfs than in giants, apparently removing any effect of the gravitational redshift. Synthetic high-resolution spectra are computed from three dimensional (3D) hydrodynamic model atmospheres for both giants and dwarfs, and synthetic wavelength shifts obtained. In agreement with observations, 3D models predict substantially smaller wavelength-shift differences than expected from gravitational redshifts only. The procedures developed could be used to test 3D models for different classes of stars, but will ultimately require high-fidelity spectra for measurements of wavelength shifts in individual spectral lines.

    • OBJECTIVE: Prenatal diagnosis of isolated coarctation of the aorta suffers from high false positive and false negative rates. The aim of our study was to develop Z-scores for the aortic isthmus in normal fetuses as a reference for fetuses with suspected coarctation. METHODS: The aortic isthmal diameter, immediately proximal to the insertion of the arterial duct, was measured prospectively in the transverse (three vessel and trachea) and sagittal views in 221 normal fetuses at 18 to 37 weeks' gestation. The ductal diameter was measured immediately before it entered the descending aorta in the same view. All measurements were repeated three times by a single investigator and averaged. A second investigator re-measured the images of 50 cases to assess interobserver variability. Z-scores were created relating isthmal and ductal diameters to femur length and gestational age. The ratio between the isthmal and ductal diameters was calculated. RESULTS: The formula used to calculate Z-scores for the three diameters was: [ln(measured isthmal diameter) - (m ln(femur length or gestational age) + c)]/root MSE, where c is the intercept, m is a multiplier and MSE is the mean squared error. The ratio between isthmal and ductal diameters was close to a constant value of 1 (95% CI 0.97-1.01), regardless of the value of femur length or gestational age. CONCLUSION: We have defined Z-scores for the fetal aortic isthmus and arterial duct measured in the three vessels and trachea view and for the isthmus in the sagittal plane. In suspected coarctation, these Z-scores and the isthmal to ductal ratio may help in longitudinal assessment of the aortic arch and aid in the prenatal diagnosis of coarctation.
