  • Decline in ribosomal fidelity contributes to the accumulation and stabilization of the master stress response regulator sigma S upon carbon starvation

    • The {sigma}S subunit of RNA polymerase is a master regulator of Escherichia coli that retards cellular senescence and bestows cells with general stress protective functions during growth arrest. We show that mutations and drugs triggering translational errors elevate {sigma}S levels and stability. Furthermore, mutations enhancing translational fidelity attenuate induction of the rpoS regulon and prevent stabilization of {sigma}S upon carbon starvation. Destabilization of {sigma}S by increased proofreading requires the presence of the {sigma}S recognition factor SprE (RssB) and the ClpXP protease. The data further suggest that {sigma}S becomes stabilized upon starvation as a result of ClpP sequestration and this sequestration is enhanced by oxidative modifications of aberrant proteins produced by erroneous translation. ClpP overproduction counteracted starvation-induced stabilization of {sigma}S, whereas overproduction of a ClpXP substrate (ssrA-tagged GFP) stabilized {sigma}S in exponentially growing cells. We present a model for the sequence of events leading to the accumulation and activation of {sigma}S upon carbon starvation, which are linked to alterations in both ribosomal fidelity and efficiency.

  • The Political Dynamics of Portfolio Design in European Democracies

    • The design of government portfolios – that is, the distribution of competencies among government ministries and office holders – has been largely ignored in the study of executive and coalition politics. This article argues that portfolio design is a substantively and theoretically relevant phenomenon that has major implications for the study of institutional design and coalition politics. The authors use comparative data on portfolio design reforms in nine Western European countries since the 1970s to demonstrate how the design of government portfolios changes over time. Specifically, they show that portfolios are changed frequently (on average about once a year) and that such shifts are more likely after changes in the prime ministership or the party composition of the government. These findings suggest a political logic behind these reforms based on the preferences and power of political parties and politicians. They have major implications for the study of institutional design and coalition politics.

    • This article examines patterns of oppositional behaviour in the German Bundestag’s Committee on European Union Affairs (EAC) for two separate legislative periods (2005–2009 and 2009–2013). The study makes two contributions to previous research. It, first, shed some much-needed empirical light on political opposition in the Bundestag by examining how much and what kind of opposition exists in the German EAC. Secondly, the article examines the differences in oppositional behaviour of the Members of Parliament between the two legislative periods following an institutional reform in 2009 that afforded the Bundestag with increased opportunity structures in Europen Union affairs.

Contributions from ten leading scholars in the fields of economics, law, and political science provide in-depth analyses of three key dimensions of EU foreign policy, namely: the internal challenges facing the EU, as its 28 member countries struggle to coordinate their actions; the external challenges facing the EU on the global arena, in areas where global imbalances are particularly pervasive, and where measures taken by the Union can have an important impact; and the EU´s performance on the global arena, in the eyes of other key actors.     • Voluntarily organized sport is the cornerstone of Swedish sports and has an important societal role. In this chapter, the Regional Sports Confederation and Sports Education Skåne, which is the gathering body of the non-profit sport clubs in the region, represents the voluntary, non-profit sector. The Leisure Department, City of Malmö, represents the public sector. The football club Malmö FF represents the for-profit sector, despite not being a completely commercial club. Data was collected through semi-structured interviews and analysis of documents.The three sectors have overlapping interests and a lot to gain from extended cooperation and knowledge transfer to bring about change. All our informants state a clear awareness of the importance of sustainability and highlight sustainable development and the Sustainable Development Goals (SDG) at the leading level of their organizations. However, their capacity and incentives to act differ, partly due to each sectors’ specific logics. Malmö FF, strongly affected by commercial logics, stands out as the organization most active in realizing the SDGs. The Leisure Department, limited by political requirements, works hard to handle basic political requirements with an ambition to do good and even be a role model among municipalities. For the sports movement, the possibility of collective actions is challenged by voluntarism and the strong autonomy of sports clubs. Perhaps it would be logical to argue for a stricter control of the sports clubs’ operations to ensure proper handling of sustainability issues, but according to the Swedish sport ideology, the incentives should rather be created from the bottom-up.

    • IT has for a long time been viewed as a success factor for police investigation, but now IT has also become an obstacle. Many modern crimes are of a nature where the evidence needed for police investigations is found in both the analogue world and the digital world. We argue that modern criminals act as hybrids, i.e. they are using IT as natural components in their criminal activities. In this paper we analyze the modern criminal as a hybrid and analyze the effect this perspective has on modern police work. The result is that the crime arena should be seen as a hybrid arena, where the criminals and the police both should take the roles as hybrids. Today, there is a division among the police officers, some are acting in the digital world; others in the analogue world. This gives the criminal a bit of an advantage.

  • MAKING SENSE OF DIGITAL FOOTPRINTS IN POLICE PRACTICE

    • Purpose Digital footprints are yet another weapon in law enforcement but have so far gained limited attention in research on police practice. Here we conduct an analyse of how a small police task force has managed to make sense of a large set of digital footprints. The aim is to increase the knowledge about the role digital footprints play and how these footprints are captured and managed during a 21st century criminal investigation. Methodology A case study approach have been applied and the theory of sensemaking as foremost an analytical lens helping the data analysis with a goal to better understand how the police officers worked in the chosen case study. Findings The use of large sets of digital footprints in the police investigations is a sensemaking process, and the use of digital footprints exists in two temporal structures. In the pre-arrest(1) the use of digital footprints is characterized by sensemaking, i.e. the police officers use the footprints as triggers to traditional police work to understand what is going on. In the post-arrest(2) the use of digital footprints is characterized as evidential aggregation, and analyzing and validating of digital footprints with those collected in previous temporal structure. Value Knowledge generated from this research gives design implcations for information system that not only fulfil the needs in police practice on a general level but in the actual work of capturing and managing digital footprints. The article also provides knowledge on how digital footprints are forming police practice in 21st century.

    • Over 30 years ago, Browne and Franklin (1973) demonstrated that parties in a coalition tend to receive portfolio payoffs in almost perfect proportionality to their seat share. Even though this result has been confirmed in several studies, few researchers have asked what the underlying mechanism is that explains why parties receive a proportional payoff. The aim of this paper is to investigate the causal mechanism linking party size and portfolio payoffs. To fulfil this aim, a small-n analysis is performed. By analysing the predictions from a statistical analysis of all post-war coalition governments in 14 Western European countries, two predicted cases are selected; the coalitions that formed after the 1976 Swedish election and the 1994 German election. In these case studies two hypotheses are evaluated: that the proportional distribution of ministerial posts is the result of a social norm, or that parties obtain payoffs according to their bargaining strength. The results give no support to the social norm hypothesis. Instead, we suggest that proportionality serves as a bargaining convention for the actors involved, thus rendering proportional payoffs more likely.

    • Nutritionally induced changes in RNA polymerase availability have been hypothesized to be an evolutionary primeval mechanism for regulation of gene expression and several contrasting models have been proposed to explain how such 'passive' regulation might occur. We demonstrate here that ectopically elevating Escherichia coli RNA polymerase (Esigma(70)) levels causes an increased expression and promoter occupancy of ribosomal genes at the expense of stress-defense genes and amino acid biosynthetic operons. Phenotypically, cells overproducing Esigma(70) favours growth and reproduction at the expense of motility and damage protection; a response reminiscent of cells with no or diminished levels of the alarmone guanosine tetraphosphate (ppGpp). Consistently, we show that cells lacking ppGpp displayed markedly elevated levels of free Esigma(70) compared with wild-type cells and that the repression of ribosomal RNA expression and reduced growth rate of mutants with constitutively elevated levels of ppGpp can be suppressed by overproducing Esigma(70). We conclude that ppGpp modulates the levels of free Esigma(70) and that this is an integral part of the alarmone's means of regulating a trade-off between growth and maintenance.

    • In a recent paper Wagenaar (1988) suggested that overconfidence can be used as an indicator of reconstructive processes which allow responses based on inference to be distinguished from responses based on retrieval. The ecological models (Björkman, in press; Gigerenzer, Hoffrage, & Kleinbölting, 1991; Juslin, 1993a, 1993b, 1994) provide a more positive view of the calibration of reconstructive responses. In this paper we compare these two views and argue that overconfidence cannot be considered a reliable indicator of reconstructive processes since people may be well calibrated for tasks that require inference, provided that tasks are selected in an unbiased manner. Instead, we discuss two different models: the response-independence model which is appropriate to retrieval, and the response-dependence model which applies to inference. These two models predict different distributions of solution probabilities and they therefore provide a criterion by which we can distinguish between direct retrieval and reconstruction. In two empirical studies modelled after Experiment 1 in Wagenaar's (1988) paper it is shown that calibration can be very similar and quite reasonable both for tasks that are dominated by inference and tasks that are dominated by retrieval processes. In Experiment 2 we show that the two conditions nevertheless differ in regard to the distributions of solution probabilities in the manner predicted by the two response models presented in the paper. It is proposed that the issue of which is the most appropriate interpretation of solution probabilities is neglected, and that the criterion should be of interest also to applications outside the domain of calibration research.

    • This article offers a comprehensive comparison of oppositional behaviour in European Union (EU) affairs in six countries: Austria, Denmark, Germany, Ireland, Sweden and the United Kingdom. By drawing on a unique data set of hand-coded statements made by Members of Parliament during European Affairs Committee (EAC) meetings it provides new knowledge on political opposition. The data uncover that there is more opposition expressed in EU affairs than has been assumed by previous research. The results also reveal that there exists considerable cross-national variation in oppositional behaviour during EAC deliberations in the six countries. The study finally shows that Eurosceptic parties are key drivers of opposition in EU affairs, especially when it comes to opposition directed at the EU political system.

    • This study provides new insights regarding how the EU dimension relates to the left-right dimension in national politics. It does so by examining to what extent parties’ oppositional behaviour in EU affairs in parliament is a function of their distance to the government, other parties, the voters and party supporters on the EU dimension and the left-right dimension, respectively. The results suggest that oppositional behaviour is determined by two parallel dynamics: When expressing opposition, political parties are receptive both to the positions of the government and other parties on the EU dimension, and to the positions of voters and supporters on the left-right dimension. Our results thus indicate that the politicization of EU affairs in national parliaments primarily is an elite phenomenon. In their relation to the voters and supporters, parties still primarily navigate along the traditional left-right dimension.

    • This study attempts to break new ground by systematically comparing oppositional behaviour of Members of Parliament (MPs) in the two key arenas found in parliament: the plenary and the committee. Do MPs express more opposition frontstage in the plenary than backstage in the committee when discussing European affairs? The article sheds new light on political opposition in parliament by using a dataset of hand-coded statements made by MPs during plenary debates and European Affairs committee deliberations in five national parliaments. The results lend strong support to the main hypothesis that MPs express more opposition during plenary debates than committee deliberations.

    • Is oppositional behaviour in legislatures gendered? Despite a growing literature on gender and legislative behaviour, there is a conspicuous absence of research on differences between female and male members of parliament (MPs) when it comes to one of the key aspects of parliamentary debates: the voicing of opposition. This article connects the literature on gender and legislative behaviour with opposition scholarship. We examine the role of gender for oppositional behaviour in four legislatures with a focus on debates concerning European Union affairs. Our results show that female MPs generally are equally likely to express opposition as their male colleagues. This result, however, is conditioned by government status. Whilst there is no significant difference between the proportion of opposition expressed by female and male MPs from opposition parties, we find that male MPs representing government parties express more than twice as much opposition as their female colleagues.

    • According to the ‘opposition deficit thesis’ European Union politics is characterized by a conspicuous absence of opposition to the policies promoted by the EU. However, once we start looking for systematic evidence supporting the claim of an opposition deficit in EU politics, we come up surprisingly empty handed. This article seeks to rectify the lacuna in previous research by examining how much and what type of opposition actually exist in EU politics. The study formulates four hypotheses on the state of political opposition in EU politics, and breaks new ground by testing their validity by use of unique data collected from the Swedish European Affairs Committee for the period 1995–2016. The results show there is in fact considerably more opposition present in EU politics than has previously been assumed. The opposition deficit thesis thus seems to be more myth than reality.

    • The α-1,3-glucosyltransferase WaaG is involved in the synthesis of the core region of lipopolysaccharides in E. coli. A fragment-based screening for inhibitors of the WaaG glycosyltrasferase donor site has been performed using NMR spectroscopy. Docking simulations were performed for three of the compounds of the fragment library that had shown binding activity towards WaaG and yielded 3D models for the respective complexes. The three ligands share a hetero-bicyclic ring system as a common structural motif and they compete with UDP-Glc for binding. Interestingly, one of the compounds promoted binding of uridine to WaaG, as seen from STD NMR titrations, suggesting a different binding mode for this ligand. We propose these compounds as scaffolds for the design of selective high-affinity inhibitors of WaaG. Binding of natural substrates, enzymatic activity and donor substrate selectivity were also investigated by NMR spectroscopy. Molecular dynamics simulations of WaaG were carried out with and without bound UDP and revealed structural changes compared to the crystal structure and also variations in flexibility for some amino acid residues between the two WaaG systems studied.

    • Since the breakdown of the 'permissive consensus' in the early 1990s we have witnessed an input turn in the debate on EU legitimacy. Many scholars have been arguing for enhancing the input legitimacy of the EU through promoting stakeholder participation and deliberation on important European issues. Yet, others warn that this strategy might not help in increasing overall legitimacy of the EU, since increased input legitimacy could be thought to undermine its output legitimacy by making decision-making less efficient. This article assesses, empirically, the relationship between input and output legitimacy within the context of the EU chemicals policy overhaul. Contrary to what some scholars suggest, we find evidence from a survey that measures aimed at increasing the input legitimacy of the EU also hold the promise of increasing its output legitimacy.

    • This book is an empirical assessment of whether participatory governance reforms within the European Union enhance or endanger democracy. The alleged democratic deficit of the EU and its waning legitimacy have inspired scholars and practitioners to look for new ways of improving democracy and efficiency within it. Proponents of participatory governance, including the European Commission, argue that introducing new forums for interest-group participation and deliberation at the EU level will help create a more democratic and effective Union. This book examines the veracity of this claim. It looks at whether it is possible to enhance decision-making efficiency and democratic legitimacy by involving interest group representatives in the formulation and implementation of policy. In doing so, Participatory Governance in the EU employs data from a unique expert survey of stakeholders and EU officials.

    • There is a lively academic debate over whether political cleavages in the European Union ( EU) follow mainly territorial ( national) or non-territorial ( ideological) patterns. This article analyses the cleavages that structure the conflict over European chemicals policy, the so-called REACH system. Taking positions on this major policy as an empirical example, we test these competing theories on the nature of cleavages on environmental policy issues in the EU. We use data from an expert survey of more than 600 individuals to fulfil this aim. The results show that neither of the hypotheses is unequivocally supported. But the data indicate that cleavages based on non-territorial interests are much more important than territorial interests in explaining positions on REACH.

    • Traumatic brain injury (TBI) is the leading cause of death and disability among young persons. A key to improve outcome for patients with TBI is to reduce the time from injury to definitive care by achieving high triage accuracy. Microwave technology (MWT) allows for a portable device to be used in the pre-hospital setting for detection of intracranial hematomas at the scene of injury, thereby enhancing early triage and allowing for more adequate early care. MWT has previously been evaluated for medical applications including the ability to differentiate between hemorrhagic and ischemic stroke. The purpose of this study was to test whether MWT in conjunction with a diagnostic mathematical algorithm could be used as a medical screening tool to differentiate patients with traumatic intracranial hematomas, chronic subdural hematomas (cSDH), from a healthy control (HC) group. Twenty patients with cSDH and 20 HC were measured with a MWT device. The accuracy of the diagnostic algorithm was assessed using a leave-one-out analysis. At 100% sensitivity, the specificity was 75%-i.e., all hematomas were detected at the cost of 25% false positives (patients who would be overtriaged). Considering the need for methods to identify patients with intracranial hematomas in the pre-hospital setting, MWT shows promise as a tool to improve triage accuracy. Further studies are under way to evaluate MWT in patients with other intracranial hemorrhages.

  • Is public procurement efficiency conditioned by market types? A critical test in park and road sectors in Sweden

    • Purpose – The purpose of this paper is to investigate to what extent contracting out practices has led to perceived resource cuts in the Swedish park and road sectors. Design/methodology/approach – E-survey sent to managers in park and technical departments in the municipalities. Response rate: 39.7 percent. Findings – Decreased costs were found in 25 percent of the responding road departments, in 20 percent of park departments. 49 percent of the road departments responded “ no change ” or “ increase ” (48 percent parks). Findings indicate that tendering practices tend to be most commonly centered in Swedish municipalities situated in the three metropolitan regions (Stockholm, Gothenburg and Malmö with suburbs), while such practices are not as common in mid-sized and rural municipalities. In addition, contracting out practices correspond with higher per capita budgets, not with smaller. Research limitations/implications – The research implications are that contracting out tend to occur “ where the resources are, ” while poorer municipalities either have to contract even if it is more expensive, or, contracting out is combined with other provision models. The limitation of the data are that it based on managers’ perceptions, while hard data have been impossible to map. Practical implications – The uniform assumption that contracting out saves public resources need to be conditioned with context, and initial resources at hand. Contracting out is one option among several possible strategies, especially for mid-sized and smaller municipalities with small budgets. Social implications – The rationalities of local government procurements are bounded by the local market situations and local the budget capacities. Originality/value – This is the first study of manager ’ s perceptions in the Swedish park and road contexts.     • Relations between the public sector and the market have a long history in Sweden. The opening section discusses relations between public and private governance at the national level across the past four decades in Sweden. Next, the discussion is narrowed to the municipal park and road sectors of the local municipalities, as the early marketization reforms were initiated there. In the closing section several lessons are discussed. First, the actual selling-off of public companies on the open market primarily relates to the national level, while corporatization and market imitation have been increasingly common features of the local administrative level. Second, in local government discussions of management form characterized the early period, while outcome and quality control dominated in later years. Third, contracting out has taken place due to procurement regulations. For the local road sector, the level external contracting has been roughly the same throughout the period, while for the park sector we found an increase from 3 percent in 1987 to 28 percent in 2015. Materials are primary data (INOPS 2015) combined with secondary data from previous scientific reports and governmental studies. The results are multifaceted. First, the market imitation has increasingly become the features of the local administrative level – but not to the expense of local government legitimacy. Second, the alternatives of management forms characterized the early period, while outcome and quality control dominated the more recent years. Third, for the local road sector, the level of external contracting has been stable throughout the period; while for the park sector we found an increase from 3 percent in 1987 to 28 percent in 2015. The general mimicking of the private sector seems to have kept local governance and legitimacy in line with the expectations of the Swedes.

    • A number of high profile crises and disasters have driven the EU to increase cooperation among its member states in the area of civil protection and to enhance its capacity to conduct civil protection operations in Europe and around the world. However, in the light of recent transboundary crises in the EU, manifested by the refugee crisis, terrorist attacks, and natural disasters, it is far from clear how effective such cooperative EU arrangements can be due to differences in the way national civil protection has been organized and due to the question of whether sufficient trust exists within and between the involved organizations. In this article, drawing from a unique study of civil protection agencies in 17 EU member states, and utilizing theories on crisis management, public administration and trust, we shed light on the factors that promote national and EU-level effectiveness in civil protection and crisis management.

    • Despite long tradition of stable parliamentary democracy, the Nordic countries are not unaffected by the profound changes taking place in the parliaments of Europe. This book seeks to address these challenges to the Nordic parliamentary systems. Despite their long tradition of stable parliamentary democracy, the Nordic countries have not gone unaffected by the profound changes taking place in Europe. In recent years, some scholars have claimed that executive-legislative relations are undergoing fundamental changes in the Nordic countries and elsewhere. More powers are suggested to be transferred to political executives at the expense of parliaments, while powers inside government cabinets are increasingly concentrated in the hands of prime ministers. This volume seeks to address these challenges to the Nordic parliamentary systems. Due to the alleged strong reliance on power-sharing in these countries between the executive and the legislature on the one hand, and between the prime minister and his/her cabinet ministers on the other, the Nordic countries should be particularly resistant to such trends. Yet, the authors find that parliamentary government in the Nordic countries appears to be undergoing significant changes. It seems therefore that the Nordic parliamentary systems are now at an important crossroads. The evidence presented in this volume, however, yields a rather mixed picture of where the development is going.

    • This study investigates whether the European Union's funding of civil society organizations (CSOs) can foster equal access to existing channels of participation. This is analyzed by using data from 7,889 CSOs that are registered in the EU Transparency Register. Two criteria should be met for funding to combat inequalities among CSOs: the funding must support underrepresented interests, and it must increase their participation in the policy process. The results of a logistic regression analysis show that the Commission's funding scheme does support underrepresented interests but makes participation more unequal by also supporting already overrepresented groups. Furthermore, the analysis shows that the probability of a CSO having access to formal channels of participation significantly increases if it has received funding. The conclusion is that funding could be used more efficiently if the Commission were to consistently support underrepresented interests.
