    • The PREDICTS project-Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)-has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

  • Global urban environmental change drives adaptation in white clover

    • Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural dines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale.

  • The PREDICTS database : a global database of how local terrestrial biodiversity responds to human impacts

    • Biodiversity continues to decline in the face of increasing anthropogenic pressures such as habitat destruction, exploitation, pollution and introduction of alien species. Existing global databases of species' threat status or population time series are dominated by charismatic species. The collation of datasets with broad taxonomic and biogeographic extents, and that support computation of a range of biodiversity indicators, is necessary to enable better understanding of historical declines and to project - and avert - future declines. We describe and assess a new database of more than 1.6 million samples from 78 countries representing over 28,000 species, collated from existing spatial comparisons of local-scale biodiversity exposed to different intensities and types of anthropogenic pressures, from terrestrial sites around the world. The database contains measurements taken in 208 (of 814) ecoregions, 13 (of 14) biomes, 25 (of 35) biodiversity hotspots and 16 (of 17) megadiverse countries. The database contains more than 1% of the total number of all species described, and more than 1% of the described species within many taxonomic groups - including flowering plants, gymnosperms, birds, mammals, reptiles, amphibians, beetles, lepidopterans and hymenopterans. The dataset, which is still being added to, is therefore already considerably larger and more representative than those used by previous quantitative models of biodiversity trends and responses. The database is being assembled as part of the PREDICTS project (Projecting Responses of Ecological Diversity In Changing Terrestrial Systems - ). We make site-level summary data available alongside this article. The full database will be publicly available in 2015.

  • Global Impact of the COVID-19 Pandemic on Stroke Volumes and Cerebrovascular Events: A 1-Year Follow-up.

    • Declines in stroke admission, IV thrombolysis (IVT), and mechanical thrombectomy volumes were reported during the first wave of the COVID-19 pandemic. There is a paucity of data on the longer-term effect of the pandemic on stroke volumes over the course of a year and through the second wave of the pandemic. We sought to measure the effect of the COVID-19 pandemic on the volumes of stroke admissions, intracranial hemorrhage (ICH), IVT, and mechanical thrombectomy over a 1-year period at the onset of the pandemic (March 1, 2020, to February 28, 2021) compared with the immediately preceding year (March 1, 2019, to February 29, 2020).We conducted a longitudinal retrospective study across 6 continents, 56 countries, and 275 stroke centers. We collected volume data for COVID-19 admissions and 4 stroke metrics: ischemic stroke admissions, ICH admissions, IVT treatments, and mechanical thrombectomy procedures. Diagnoses were identified by their ICD-10 codes or classifications in stroke databases.There were 148,895 stroke admissions in the 1 year immediately before compared with 138,453 admissions during the 1-year pandemic, representing a 7% decline (95% CI [95% CI 7.1-6.9]; p < 0.0001). ICH volumes declined from 29,585 to 28,156 (4.8% [5.1-4.6]; p < 0.0001) and IVT volume from 24,584 to 23,077 (6.1% [6.4-5.8]; p < 0.0001). Larger declines were observed at high-volume compared with low-volume centers (all p < 0.0001). There was no significant change in mechanical thrombectomy volumes (0.7% [0.6-0.9]; p = 0.49). Stroke was diagnosed in 1.3% [1.31-1.38] of 406,792 COVID-19 hospitalizations. SARS-CoV-2 infection was present in 2.9% ([2.82-2.97], 5,656/195,539) of all stroke hospitalizations.There was a global decline and shift to lower-volume centers of stroke admission volumes, ICH volumes, and IVT volumes during the 1st year of the COVID-19 pandemic compared with the prior year. Mechanical thrombectomy volumes were preserved. These results suggest preservation in the stroke care of higher severity of disease through the first pandemic year.This study is registered under NCT04934020.

  • Determinants of morbidity and mortality following emergency abdominal surgery in children in low-income and middle-income countries

    • Background: Child health is a key priority on the global health agenda, yet the provision of essential and emergency surgery in children is patchy in resource-poor regions. This study was aimed to determine the mortality risk for emergency abdominal paediatric surgery in low-income countries globally.Methods: Multicentre, international, prospective, cohort study. Self-selected surgical units performing emergency abdominal surgery submitted prespecified data for consecutive children aged <16 years during a 2-week period between July and December 2014. The United Nation's Human Development Index (HDI) was used to stratify countries. The main outcome measure was 30-day postoperative mortality, analysed by multilevel logistic regression.Results: This study included 1409 patients from 253 centres in 43 countries; 282 children were under 2 years of age. Among them, 265 (18.8%) were from low-HDI, 450 (31.9%) from middle-HDI and 694 (49.3%) from high-HDI countries. The most common operations performed were appendectomy, small bowel resection, pyloromyotomy and correction of intussusception. After adjustment for patient and hospital risk factors, child mortality at 30 days was significantly higher in low-HDI (adjusted OR 7.14 (95% CI 2.52 to 20.23), p<0.001) and middle-HDI (4.42 (1.44 to 13.56), p=0.009) countries compared with high-HDI countries, translating to 40 excess deaths per 1000 procedures performed.Conclusions: Adjusted mortality in children following emergency abdominal surgery may be as high as 7 times greater in low-HDI and middle-HDI countries compared with high-HDI countries. Effective provision of emergency essential surgery should be a key priority for global child health agendas.

  • Predicting bee community responses to land-use changes : effects of geographic and taxonomic biases

    • Land-use change and intensification threaten bee populations worldwide, imperilling pollination services. Global models are needed to better characterise, project, and mitigate bees' responses to these human impacts. The available data are, however, geographically and taxonomically unrepresentative; most data are from North America and Western Europe, overrepresenting bumblebees and raising concerns that model results may not be generalizable to other regions and taxa. To assess whether the geographic and taxonomic biases of data could undermine effectiveness of models for conservation policy, we have collated from the published literature a global dataset of bee diversity at sites facing land-use change and intensification, and assess whether bee responses to these pressures vary across 11 regions (Western, Northern, Eastern and Southern Europe; North, Central and South America; Australia and New Zealand; South East Asia; Middle and Southern Africa) and between bumblebees and other bees. Our analyses highlight strong regionally-based responses of total abundance, species richness and Simpson's diversity to land use, caused by variation in the sensitivity of species and potentially in the nature of threats. These results suggest that global extrapolation of models based on geographically and taxonomically restricted data may underestimate the true uncertainty, increasing the risk of ecological surprises.

    • NASA’s Interface Region Imaging Spectrograph (IRIS) provides high-resolution observations of the solar atmosphere through ultraviolet spectroscopy and imaging. Since the launch of IRIS in June 2013, we have conducted systematic observation campaigns in coordination with the Swedish 1 m Solar Telescope (SST) on La Palma. The SST provides complementary high-resolution observations of the photosphere and chromosphere. The SST observations include spectropolarimetric imaging in photospheric Fe I lines and spectrally resolved imaging in the chromospheric Ca II 8542 Å, Hα, and Ca II K lines. We present a database of co-aligned IRIS and SST datasets that is open for analysis to the scientific community. The database covers a variety of targets including active regions, sunspots, plages, the quiet Sun, and coronal holes.

    • We have set out to develop a novel, implantable microelectrode array that has the capabilities to detect neurotransmitters with enhanced sensitivity, selectivity, and temporal sampling capabilities compared to other current technologies. We have shown that this device maintains recording performance during chronic measurements of extracellular neurotransmitter levels for at least 7 days postimplantation, single-unit neuronal activity for as long as 6 months, and provides enhanced biocompatibility compared to current technologies. As we continue to refine and improve our recording capability, we are able to incorporate the chronic microelectrode array technology into multimodal experimental paradigms, such as behavioral testing, pharmacological intervention (local and systemic), or combined measurements of neurotransmitter levels and neuronal activity (local field potential). Furthermore, the improvements made with the microelectrode technology discussed in this chapter have the potential to conduct longitudinal analyses that can benefit a wide range of translational efforts, including studies on learning and memory, aging, neurodegenerative disease progression, and traumatic brain injury neuropathology.

  • Effect of aspirin on the diagnostic accuracy of the faecal immunochemical test for colorectal advanced neoplasia

    • Aspirin (ASA) is a drug that can cause gastrointestinal lesions and symptoms. Colorectal cancer (CRC) is the most prevalent type of cancer in Western countries. We assessed the effect of aspirin on the diagnostic accuracy of the faecal immunochemical test (FIT) for CRC and/or advanced neoplasia (AN) in patients undergoing colonoscopy for gastrointestinal symptoms. Methods We conducted a prospective multicentre observational study of diagnostic tests that included patients with gastrointestinal symptoms undergoing colonoscopy between March 2012 and 2014 (the COLONPREDICT study). Symptoms were assessed and a FIT and blood tests assessing haemoglobin and carcinoembryonic antigen (CEA) levels were performed. Results The study included 3052 patients: A total of 2567 did not take aspirin (non-user group) and 485 (16%) took aspirin (user group). Continuous treatment with ASA did not change the AUC (0.88, 0.82; p = 0.06), sensitivity (92%, 88%; p = 0.5) or specificity (71%, 67%; p = 0.2) of the FIT for CRC detection. Similarly, we found no differences in the AUC (0.81, 0.79; p = 0.6), sensitivity (74%, 75.5%; p = 0.3) or specificity (76%, 73.6%; p = 0.3) for AN detection. Patients with an aspirin use of ≥ 300 mg/day had a lower prevalence of AN and the sensitivity, specificity and AUC for AN for these patients were 54%, 68% and 0.66, significantly lower than for the non-user group ( p = 0.03). Conclusions Aspirin does not modify the diagnostic accuracy of FIT for CRC and/or AN in patients with gastrointestinal symptoms. Aspirin use of ≥ 300 mg/day decreases the accuracy of the test.

    • The topic of animal ethics has advanced in tourism studies since its inception in 2000, based on a diverse range of studies on species involvement, types of uses and contexts, level of engagement, states of animals, and theoretical perspectives. While there is still considerable scope to amplify research on animal-based tourism, a gap exists in tourism pedagogy amidst the field’s emphasis on a new expanding consciousness platform. We review the depth of existing scholarship on animal ethics in tourism and develop an agenda for advancing animal ethics pedagogy for the future. Our intent is to issue a call to action for curriculum committees, programme administrators, and educators to recognise and act on this critical moral domain in tourism education.

    • Temporomandibular disorders (TMD) include craniocervical pain conditions with unclear etiologies. Central changes are suspected; however, few neuroimaging studies of TMD exist. Single-voxel proton magnetic resonance spectroscopy (H-1-MRS) was used before and after pressure-pain testing to assess glutamate (Glu), glutamine (Gln), N-acetylaspartate (NAA), and choline (Cho) levels in the right and left posterior insulae of 11 individuals with myofascial TMD and 11 matched control individuals. Glu levels were significantly lower in all individuals after pain testing. Among those with TMD, left-insular Gln levels were related to reported pain, left posterior insular NAA and Cho levels were significantly higher at baseline than in control individuals, and NAA levels were significantly correlated with pain-symptom duration, suggesting adaptive changes. The results suggest that significant central cellular and molecular changes can occur in individuals with TMD.

    • It has been shown that the noradrenergic (NE) locus coeruleus (LC)-hippocampal pathway plays an important role in learning and memory processing, and that the development of this transmitter pathway is influenced by neurotrophic factors. Although some of these factors have been discovered, the regulatory mechanisms for this developmental event have not been fully elucidated. Glial cell line-derived neurotrophic factor (GDNF) is a potent neurotrophic factor influencing LC-NE neurons. We have utilized a GDNF knockout animal model to explore its function on the LC-NE transmitter system during development, particularly with respect to target innervation. By transplanting various combinations of brainstem (including LC) and hippocampal tissues from wildtype or GDNF knockout fetuses into the brains of adult wildtype mice, we demonstrate that normal postnatal development of brainstem LC-NE neurons is disrupted as a result of the GDNF null mutation. Tyrosine hydroxylase immunohistochemistry revealed that brainstem grafts had markedly reduced number and size of LC neurons in transplants from knockout fetuses. NE fiber innervation into the hippocampal co-transplant from an adjacent brainstem graft was also influenced by the presence of GDNF, with a significantly more robust innervation observed in transplants from wildtype fetuses. The most successful LC/hippocampal co-grafts were generated from fetuses expressing the wildtype GDNF background, whereas the most severely affected transplants were derived from double transplants from null-mutated fetuses. Our data suggest that development of the NE LC-hippocampal pathway is dependent on the presence of GDNF, most likely through a target-derived neurotrophic function.

  • Immediate effects of the COVID-19 pandemic on the use of wildlife as food among indigenous people and local communities in South America

    • The COVID-19 pandemic has had a range of effects on the environment and particularly on wildlife, through diverse and sometimes contradictory impact pathways. In this study, based on data collected among indigenous people and local communities from South America (Colombia, Ecuador, Guyana, and Peru), we investigated changes in the use of wildlife resources for food during the first months of the COVID-19 pandemic. Our study generated unique data collected from 756 households in 60 communities and nine sites. We confirm the hypothesis that wildlife use increased as a short-term response to food insecurity during the pandemic, and fish played a more significant role than wild meat in that endeavor. The increase in wild-meat consumption as a response to food insecurity was conditioned by prices and availability (unsuccessful hunts). Wildlife use did not increase as an alternative means to generate income, because communities were cut off from the market economy for several months. Also, whereas the reliance on wildlife emerged as an immediate solution during the first months of the crisis, longer-term strategies prioritized at household level involved diversifying food sources through domestic meat and crop production. Among all available animal-based proteins, local chicken came just after fish as the animal-based source of protein whose consumption increased the most during the first months of the crisis, as a response to food insecurity. We caution that relying on wildlife as a safety net may constitute a poverty trap in cases where the resource is depleted. Although not specifically studied here, access to land and the transmission of traditional knowledge/skills are possible additional determinants of the role that wildlife may play in times of crisis, and this is proposed as an area for future research. Results also attest to local communities expecting more support from their respective national governments, and confirm results from Walters et al. (2021) that governments were generally absent or unable to react quickly during the pandemic, leaving households (or their local leaders) with the responsibility to innovate with local solutions and pro-actively adapt to the rapid impacts of the crisis.

    • This work analyzes the effect of the anhydride excess on the nonisothermal curing kinetics and on the final properties of synthetic and biobased epoxy resins. Diglycidyl ether of bisphenol A (DGEBA) and epoxidized soybean oil (ESO) were crosslinked using methyltetrahydrophthalic anhydride (MTHPA) as a curing agent and 1-methylimidazole (1MI) as an initiator. It was shown that the ESO/MTHPA/1MI system reacts slower than the DGEBA/MTHPA/1MI system, giving place to a more significant evaporation of the curing agent during the reaction. As a result, an excess of anhydride improves the final thermal properties of the ESO/MTHPA/1MI network, contrary to the behavior observed for DGEBA/MTHPA/1MI. The knowledge of the kinetics of the curing process and the optimal amount of the curing agent for each system is of critical importance for a more efficient processing of these materials.

    • Direct electrochemical measurements of glutamate release in vivo were combined with optogenetics in order to examine light-induced control of glutamate neurotransmission in the rodent brain. Self-referenced recordings of glutamate using ceramic-based microelectrode arrays (MEAs) in hippocampus and frontal cortex demonstrated precise optical control of light-induced glutamate release through channelrhodopsin (ChR2) expression in both rat hippocampus and frontal cortex. Although the virus was only injected unilaterally, bilateral and rostro-caudal expression was observed in slice imaging, indicating diffusion and active transport of the viral particles. Methodology for the optogenetic control of glutamate signaling in the rat brain is thoroughly explained with special attention paid to MEA enzyme coating and cleaning for the benefit of other investigators. These data support that optogenetic control of glutamate signaling is robust with certain advantages as compared to other methods to modulate the in vivo control of glutamate signaling.

    • The Medellin Metaharzburgitic Unit (MMU), emplaced onto the western continental margin of Pangea during Triassic time, is located in the Central Cordillera of Colombia and consists of metaharzburgites, minor metadunites and chromitite bodies (Patio Bonito and San Pedro ore deposits). The ultramafic rocks contain relicts of mantle-derived olivine, chromian spinel and minor orthopyroxene, and a later metamorphic mineral assemblage composed by tremolite, chlorite, talc, fine-grained recrystallized olivine, serpentine-group minerals, magnetite, and secondary chromian spinel, formed during the thermal evolution of the unit. The Cr# [Cr/(Cr+Al) atomic ratio] of the accessory primary chromian spinel in the metaperidotites ranges from 0.58 to 0.62 and overlaps those of supra-subduction peridotites from ophiolites. According to textural and compositional variations, the accessory chromian spinel in the metaperidotites can be classified into three groups: i) partially altered chromian spinel with an Al-rich core, ii) porous, Cr-Fe2+-enriched and Al-Mg-depleted chromian spinel, and iii) homogeneous Fe3+-rich chromian spinel. These variations can be related to superimposed medium-T metamorphism that reached amphibolite facies (ca. 600 ºC). Chromitite bodies associated with the metaperidotites have massive and semi-massive textures, and mainly consist of chromian spinel crystals, which show large unaltered cores surrounded by thin alteration rims of ferrian chromian spinel and chlorite. Chromitites are Al-rich (#Cr <0.6) and strongly depleted in platinum group elements (ΣPGE <41 ppb). The primary petrological and geochemical characteristics preserved in the metaperidotites and chromitites indicate that the MMU formed at shallow levels of a suboceanic lithospheric mantle related to a supra-subduction zone (back-arc basin/incipient arc scenario), and that the chromitites crystallized from a tholeiitic magma (back-arc basin basalt type).

    • Cell science has made significant progress by focusing on understanding individual cellular processes through reductionist approaches. However, the sheer volume of knowledge collected presents challenges in integrating this information across different scales of space and time to comprehend cellular behaviors, as well as making the data and methods more accessible for the community to tackle complex biological questions. This perspective proposes the creation of next-generation virtual cells, which are dynamic 3D models that integrate information from diverse sources, including simulations, biophysical models, image-based models, and evidence-based knowledge graphs. These virtual cells would provide statistically accurate and holistic views of real cells, bridging the gap between theoretical concepts and experimental data, and facilitating productive new collaborations among researchers across related fields.

    • Biotic responses to global change include directional shifts in organismal traits. Body size, an integrative trait that determines demographic rates and ecosystem functions, is thought to be shrinking in the Anthropocene. Here, we assessed the prevalence of body size change in six taxon groups across 5025 assemblage time series spanning 1960 to 2020. Using the Price equation to partition this change into within-species body size versus compositional changes, we detected prevailing decreases in body size through time driven primarily by fish, with more variable patterns in other taxa. We found that change in assemblage composition contributes more to body size changes than within-species trends, but both components show substantial variation in magnitude and direction. The biomass of assemblages remains quite stable as decreases in body size trade off with increases in abundance.

    • Wide-Area monitoring systems are a requirement in the current power systems in order to provide an appropriate monitoring, supervision and protection from undesired events. Electromechanical oscillations can provoke critical situations and affect the power transmission capability. This paper applies the Multi-Prony Analysis (MPA) to estimate the critical modes by using the tie-lines power measurements from Phasor Measuerement Unit (PMU)-data. The modes observation including an online sliding window give the frequency and damping variation allowing to detect and classify the range of the oscillation.

    • A methodology for early design-stage evaluation of biorefinery processes that uses multicriteria decision-making (MCDM) panels was developed with the goal of screening out less promising options. Panel members were asked to rate the relative importance of a set of evaluation criteria for biorefinery process implementation at a pulp and paper mill. Three different panels were conducted, each with this same objective. This report compares and assesses the results from the panels. Two of the MCDM panels were comprised of biorefinery specialists of similar backgrounds from government and academia, and the third panel consisted of pulp and paper industry decision makers. In general, consensus was high between all three panels, and especially between the two biorefinery specialist panels; however, certain differences were noted with the industry panel. For three out of eight criteria, these differences were attributable to a difference in the perspective of the forest industry decision makers and biorefinery specialists. For example, for the return on investment criteria, the specialist panels gave a significantly higher importance to current information from technology suppliers because it provides an important short-term perspective. In contrast, the forest industry decision makers thought that information about a particular technology should be critiqued and adjusted based on a systematic assessment of likely future process performance. Differences between the other criteria were attributed to differing interpretations of the criteria.

    • HD 165052 is a short-period massive eccentric binary system that undergoes apsidal motion. As the rate of apsidal motion is directly related to the internal structure constants of the binary components, its study allows getting insight into the internal structure of the stars. We use medium- and high-resolution spectroscopic observations of HD 165052 to provide constraints on the fundamental properties of the binary system and the evolutionary state of its components. We apply a spectral disentangling code to reconstruct artefact-free spectra of the individual stars and derive the radial velocities (RVs) at the times of the observations. We perform the first analysis of the disentangled spectra with the non-local thermodynamic equilibrium model atmosphere code CMFGEN to determine the stellar properties. We derive the first self-consistent orbital solution of all existing RV data, including those reported in the literature, accounting for apsidal motion. We build, for the very first time, dedicated stellar evolution tracks with the Cles code requesting the theoretical effective temperatures and luminosities to match those obtained from our spectroscopic analysis. The binary system HD 165052, consisting of an O6.5 V((f)) primary (T-eff,T-P = 37 500 +/- 1000 K) and an O7 V((f)) secondary (T-eff,T-S = 36 000 +/- 1000 K), displays apsidal motion at a rate of (11.30(-0.49)(+0.64))degrees yr(-1). Evolutionary masses are compared to minimum dynamical masses to constrain the orbital inclination. Evolutionary masses M-ev,M-P = 24.8 +/- 1.0 M-circle dot and M-ev,M-S = 20.9 +/- 1.0 M-circle dot and radii R-ev,R-P = 7.0(-0.4)(+0.5) R-circle dot and R-ev,R-S = 6.2(-0.3)(+0.4) R-circle dot are derived, and the inclination is constrained to 22.1 degrees <= i <= 23.3 degrees. Theoretical apsidal motion rates, derived assuming an age of 2.0 +/- 0.5 Myr for the binary, are in agreement with the observational determination. The agreement with theoretical apsidal motion rates enforces the inferred values of the evolutionary stellar masses and radii.

  • Cobalt containing glass fibres and their synergistic effect on the HIF-1 pathway for wound healing applications

    • Introduction and Methods: Chronic wounds are a major healthcare problem, but their healing may be improved by developing biomaterials which can stimulate angiogenesis, e.g. by activating the Hypoxia Inducible Factor (HIF) pathway. Here, novel glass fibres were produced by laser spinning. The hypothesis was that silicate glass fibres that deliver cobalt ions will activate the HIF pathway and promote the expression of angiogenic genes. The glass composition was designed to biodegrade and release ions, but not form a hydroxyapatite layer in body fluid.Results and Discussion: Dissolution studies demonstrated that hydroxyapatite did not form. When keratinocyte cells were exposed to conditioned media from the cobalt-containing glass fibres, significantly higher amounts of HIF-1α and Vascular Endothelial Growth Factor (VEGF) were measured compared to when the cells were exposed to media with equivalent amounts of cobalt chloride. This was attributed to a synergistic effect of the combination of cobalt and other therapeutic ions released from the glass. The effect was also much greater than the sum of HIF-1α and VEGF expression when the cells were cultured with cobalt ions and with dissolution products from the Co-free glass, and was proven to not be due to a rise in pH. The ability of the glass fibres to activate the HIF-1 pathway and promote VEGF expression shows the potential for their use in chronic wound dressings.
