  • Macromolecular Viral Entry Inhibitors as Broad-Spectrum First-Line Antivirals with Activity against SARS-CoV-2

    • Inhibitors of viral cell entry based on poly(styrene sulfonate) and its core–shell nanoformulations based on gold nanoparticles are investigated against a panel of viruses, including clinical isolates of SARS-CoV-2. Macromolecular inhibitors are shown to exhibit the highly sought-after broad-spectrum antiviral activity, which covers most analyzed enveloped viruses and all of the variants of concern for SARS-CoV-2 tested. The inhibitory activity is quantified in vitro in appropriate cell culture models and for respiratory viral pathogens (respiratory syncytial virus and SARS-CoV-2) in mice. Results of this study comprise a significant step along the translational path of macromolecular inhibitors of virus cell entry, specifically against enveloped respiratory viruses.

  • Advanced Molecular Tweezers with Lipid Anchors against SARS-CoV-2 and Other Respiratory Viruses

    • The COVID-19 pandemic caused by SARS-CoV-2 presents a global health emergency. Therapeutic options against SARS-CoV-2 are still very limited but urgently required. Molecular tweezers are supramolecular agents that destabilize the envelope of viruses resulting in a loss of viral infectivity. Here, we show that first-generation tweezers, CLR01 and CLR05, disrupt the SARS-CoV-2 envelope and abrogate viral infectivity. To increase the antiviral activity, a series of 34 advanced molecular tweezers were synthesized by insertion of aliphatic or aromatic ester groups on the phosphate moieties of the parent molecule CLR01. A structure-activity relationship study enabled the identification of tweezers with a markedly enhanced ability to destroy lipid bilayers and to suppress SARS-CoV-2 infection. Selected tweezer derivatives retain activity in airway mucus and inactivate the SARS-CoV-2 wildtype and variants of concern as well as respiratory syncytial, influenza, and measles viruses. Moreover, inhibitory activity of advanced tweezers against respiratory syncytial virus and SARS-CoV-2 was confirmed in mice. Thus, potentiated tweezers are broad-spectrum antiviral agents with great prospects for clinical development to combat highly pathogenic viruses.

  • Performance of an in situ passive sampling system for metals in stormwater

    • A passive sampler has been developed and is demonstrated in situ for urban runoff. The passive sampler is compared to conventional composite (time-dependent and flow-weighted) bottle sampling during and between storm events. The sampling was carried out at established stormwater stations; before and after a stormwater detention pond. In situ deployment of the passive sampler provides the metal concentrations, corresponding to the electrochemically available fraction of total metal, for time-dependent samples collected in parallel. The sampler provides improved accuracy compared to bottle sampling because contamination during sample transport and handling is minimised. Laboratory handling is reduced by direct analysis of the accumulated metals on the receiving membrane by laser ablation inductively coupled plasma mass spectrometry. Passive sampling also solves the problem of metal speciation change during transport to the laboratory, which is a potential problem for bottle samples. The low cost and convenience of the passive sampler and subsequent analysis should allow significantly more extensive spatial and temporal monitoring of metals in the aquatic environment than has previously been possible.

  • Mesomelia-Synostoses Syndrome Results from Deletion of SULF1 and SLCO5A1 Genes at 8q13

    • Mesomelia-synostoses syndrome (MSS) or mesomelic dysplasia with acral synostoses Verloes-David-Pfeiffer type is a rare autosomal-dominant disorder characterized by mesomelic limb shortening, acral synostoses, and multiple congenital malformations. So far, five patients in four unrelated families have been reported worldwide with MMS. By using whole-genome oligonucleotide array CGH, we have identified an interstitial deletion at 8q13 in all patients. The deletions vary from 582 Kb to 738 Kb in size, but invariably encompass only two genes: SULF1, encoding the heparan sulfate 6-O-endosulfatase 1, and SLCO5A1, encoding the solute carrier organic anion transporter family member 5A1. SULF1 acts as a regulator of numerous growth factors in skeletal embryonic development whereas the function of SLCO5A1 is yet unknown. Breakpoint sequence analyses performed in two families showed nonrecurrent deletions. Real-time quantitative RT-PCR analysis showed the highest levels of SULF1 transcripts in human osteoblasts and cartilage whereas SLCO5A1 was highly expressed in human fetal and adult brain and heart. Our results strongly suggest that haploinsufficiency of SULF1 contributes to this mesomelic chondrodysplasia, highlighting the critical role of endosulfatase in human skeletal development. Codeletion of SULF1 and SLCO5A1-which does not result from a low-copy repeats (LCRs)-mediated recombination event in at least two families-was found in all patients, so we suggest that haploinsufficiency of SULF1 combined with haploinsufficiency of SLCO5A1 (or the altered expression of a neighboring gene through position effect) could be necessary in the pathogenesis of MSS.

    • IL-22 is a recently discovered cytokine that belongs to the family of IL-10 related cytokines. It is produced by activated T-cells and innate lymphoid cells and has been suggested to be involved in tissue repair. As both inflammation and repair play important roles in atherosclerosis we investigated if IL-22 deficiency influences the disease process in Apoe(-/-) mice.

    • Increased precipitation and risk of flooding are major effects due to climate change that Swedish municipalities need to consider, while facing an ongoing growth in population and densification of urban areas. In this context, urban stormwater management represents a growing challenge. The vulnerability of the society towards climate change depends on the capability of the city to responds to environmental issues. This report presents the challenges and the needs for the implementation of sustainable stormwater solutions encountered in the urban planning process for the city of Gothenburg. The decision making process can be facilitated by the adoption of a stormwater toolbox, which functionalities are designed to support the stakeholders at each step of the planning process. The modules of the toolbox should be designed around a collaboration platform that assists with transparent information flows and allocation of responsibilities. The specific modules (e.g. hydrology, cost-benefit analysis, experience database) should support the needs along the different phases in the process. This study was financially supported by Climate KIC. The Knowledge and Innovation Communities (KICs) are partnerships set up by the European Institute of Innovation and Technology, EIT, that bring together businesses, research centers and universities with the purpose of developing innovative products and services, starting new companies and training a new generation of entrepreneurs.   • Utvärdering av biofilter vid HSB Living Lab : samlade erfarenheter och råd

    • Föreliggande rapport är en del av forskningsprojektet ”Testbädd för fördröjning och rening av dagvatten”. Projektet är ett tvärvetenskapligt samarbetsprojekt mellan Göteborg stad, RISE Research Institutes of Sweden, Chalmers Tekniska Högskola, Luleå Tekniska Universitet, Gryaab, HSB och ett antal teknikföretag. Projektet är ett fyrårigt projekt som i huvudsak har finansierats av Vinnova, HSB, Göteborgs Stad och Gryaab.Centralt för projektet är de två testbäddsansläggningarna som uppförts på HSB Living Lab i Göteborg. Syftet med bäddarna var att fungera som en testarena för innovativa dagvattenlösningar. Under projektets löptid har två olika typer av biofilter utvärderats på platsen, eller som de också kallas raingardens, regnbädd eller växtbäddar. Denna rapport är en sammanställning av den information som samlats in kopplat till dessa två biofilter.HSB Living Lab är en forsknings och demonstrationsarena där HSB tillsammans med 12 olika samarbetspartners utvecklar nya sätt att bygga och forma framtidens boende. Stor vikt läggs på ny kunskap beträffande social, ekonomisk och ekologisk hållbarhet och nya smarta tekniska lösningar som kan användas i framtidens produktion. Huset är en flyttbar byggnad med fyra våningar där allt går att byta ut och förändra. En boendedel med studentbostäder och en utställningsdel där det finns kontor, möteslokal, show room för forskningsresultat, tvättstudio och mycket mer. Närmare 2000 sensorer mäter i realtid allt som pågår. Huset stod klart för inflyttning 1a juni 2016 och redan då fanns de två testbäddarna för utvärdering av lokal fördröjning med biofilter på plats.
