    • From 1985 to 2016, the prevalence of underweight decreased, and that of obesity and severe obesity increased, in most regions, with significant variation in the magnitude of these changes across regions. We investigated how much change in mean body mass index (BMI) explains changes in the prevalence of underweight, obesity, and severe obesity in different regions using data from 2896 population-based studies with 187 million participants. Changes in the prevalence of underweight and total obesity, and to a lesser extent severe obesity, are largely driven by shifts in the distribution of BMI, with smaller contributions from changes in the shape of the distribution. In East and Southeast Asia and sub-Saharan Africa, the underweight tail of the BMI distribution was left behind as the distribution shifted. There is a need for policies that address all forms of malnutrition by making healthy foods accessible and affordable, while restricting unhealthy foods through fiscal and regulatory restrictions.

  • Diminishing benefits of urban living for children and adolescents' growth and development

    • Optimal growth and development in childhood and adolescence is crucial for lifelong health and well-being1–6. Here we used data from 2,325 population-based studies, with measurements of height and weight from 71 million participants, to report the height and body-mass index (BMI) of children and adolescents aged 5–19 years on the basis of rural and urban place of residence in 200 countries and territories from 1990 to 2020. In 1990, children and adolescents residing in cities were taller than their rural counterparts in all but a few high-income countries. By 2020, the urban height advantage became smaller in most countries, and in many high-income western countries it reversed into a small urban-based disadvantage. The exception was for boys in most countries in sub-Saharan Africa and in some countries in Oceania, south Asia and the region of central Asia, Middle East and north Africa. In these countries, successive cohorts of boys from rural places either did not gain height or possibly became shorter, and hence fell further behind their urban peers. The difference between the age-standardized mean BMI of children in urban and rural areas was <1.1 kg m–2 in the vast majority of countries. Within this small range, BMI increased slightly more in cities than in rural areas, except in south Asia, sub-Saharan Africa and some countries in central and eastern Europe. Our results show that in much of the world, the growth and developmental advantages of living in cities have diminished in the twenty-first century, whereas in much of sub-Saharan Africa they have amplified.

    • Ti2AlC coatings have been fabricated by cold-spray deposition. The microstructure evolution as a function of basic spray parameters temperature and pressure onto AA6060 aluminium alloy and 1.0037 steel substrates has been studied. Adherent and dense 50–80 μm thick Ti2AlC coatings were deposited on soft AA6060 substrates under gas temperature and pressure of 600 °C and 3.4 MPa, respectively, whilst comparable results were obtained on harder 1.0037 steel by using higher temperature (800 °C) and pressure (3.9 MPa).

  • Identification of circulating microRNA patterns in patients in psoriasis and psoriatic arthritis

    • ObjectivemiRNAs are small non-coding RNAs that control gene expression. Specific intra- and extracellular miRNA signatures have been identified in various diseases. Whether certain miRNA signatures are associated with psoriasis (PsO) and PsA is currently unknown. We aimed to search for circulating miRNA signatures associated with PsO and PsA patients.MethodsExpression of miRNAs was analysed by reverse transcription quantitative real-time PCR (RT-qPCR) in the serum of PsA, PsO patients and healthy controls. Demographic and disease-specific characteristics and imaging data from hand MRI were recorded. In the discovery phase, 192 miRNA assays were analysed in 48 samples (PsA, PsO, controls: each N = 16). For validation, 17 selected miRNAs were measured in the total population.ResultsA total of 141 patients and controls were analysed (51 PsA, 40 PsO, 50 controls). In the discovery phase 51 miRNAs in PsO and 64 miRNAs in PsA were down- or upregulated compared with controls, with 33 miRNAs being changed in both (adj. P &lt; 0.05). The 17 top candidates from discovery were assessed in the validation phase, 9 of them discriminated PsA and PsO from controls [area under the curve (AUC) ≥0.70, all P &lt; 0.05]. Four miRNAs (miR-19b-3p, miR-21-5p, miR-92a-3p and let-7b-5p) were significantly differently regulated between PsO and PsA. A combination of these miRNAs increased the AUC to 0.92 in multivariate regression model to discriminate PsO and PsA.ConclusionmiRNA signatures in PsA and PsO patients differ from controls. Nine miRNAs were differentially regulated in PsA and PsO patients, five of them previously reported to be involved in bone and cartilage metabolism, indicating an intimate association of psoriatic inflammation and bone/cartilage changes.

    • Topographies are one of the challenges for the development of the metal additive manufacturing promising technique. The article investigates multi scale topographies (from form to roughness) of as built surfaces generated by selective laser melting (SLM). Different building inclinations of samples were observed both for upskin and downskin surfaces with a wide range of measuring techniques. The two main aims are: (i) to make a critical review of measuring techniques at different scales, (ii) to enlighten the different surface generation phenomena (and the corresponding scale) occurring during additive manufacturing. The effect of heat treatment on each scale of the topography is also discussed. It is found that the focus variation technique is well suited for AM surfaces. Concerning the observation of the inclined surfaces, some parameters are emphasized as good indicators of typical signatures of AM surfaces: isotropy for the weld track component, the skewness and Rsm for the staircase effect, fractal dimension for the presence of partly melted particles. The different parameters studied helps to model and understand the different surface generation phenomena aforementioned. © 2018 Elsevier Inc.

  • Neuron-enriched RNA-binding Proteins Regulate Pancreatic Beta Cell Function and Survival

    • Pancreatic beta cell failure is the central event leading to diabetes. Beta cells share many phenotypic traits with neurons, and proper beta cell function relies on the activation of several neuronal-like transcription programs. Regulation of gene expression by alternative splicing plays a pivotal role in brain, where it affects neuronal development, function and disease. The role of alternative splicing in beta cells remains unclear, but recent data indicate that splicing alterations modulated by both inflammation and susceptibility genes for diabetes contribute to beta cell dysfunction and death. Here we used RNA sequencing to compare the expression of splicing-regulatory RNA-binding proteins in human islets, brain and other human tissues, and identified a cluster of splicing regulators that are expressed in both beta cells and brain. Four of them, namely Elavl4, Nova2, Rbox1 and Rbfox2 were selected for subsequent functional studies in insulin-producing rat INS-1E, human EndoC-βH1 cells, and in primary rat beta cells. Silencing of Elavl4 and Nova2 increased beta cell apoptosis, while silencing of Rbfox1 and Rbfox2 increased insulin content and secretion. Interestingly, Rbfox1 silencing modulates the splicing of the actin-remodeling protein gelsolin, increasing gelsolin expression and leading to faster glucose-induced actin depolymerization and increased insulin release. Taken together, these findings indicate that beta cells share common splicing regulators and programs with neurons. These splicing regulators play key roles in insulin release and beta cell survival and their dysfunction may contribute to the loss of functional beta cell mass in diabetes.

  • Oxygen isotopes of land snail shells in high latitude regions

    • The present study investigates the environmental significance of the oxygen isotopic composition of several modern land snail species collected along two north-to-south transects in Alaska and Scandinavia at latitudes between 60 and 70 °N. We tested the hypothesis that land snail shell δ18O values primarily track precipitation δ18O. The results show that shell δ18O values from Scandinavia were ∼5.1‰ enriched in 18O with respect to snails from Alaska, equivalent to differences in precipitation δ18O values between the two regions. Within the Alaskan transect, shell δ18O values increased with observed increasing air temperature and precipitation δ18O, whereas shell δ18O values from Scandinavia did not correlate to instrumental climate data because of a reduced climatic gradient across the locations sampled. In addition, shell δ18O values differed significantly among sympatric species, with larger species consistently exhibiting higher δ18O values, which implies that species-level isotopic variations should be considered at the local and microhabitat scale. However, when snail shell δ18O values from this study are combined with previously published data from North America and Europe, we see evidence that shell δ18O values track precipitation δ18O across latitudes, even when different species are combined because climate gradients are greater than variations among taxa. © 2022 Elsevier Ltd

    • Methodological developments and new paleoanthropological data remain jointly central to clarifying the timing and systemic interrelationships between the Middle-Upper Paleolithic (MP-UP) archaeological transition and the broadly contemporaneous anatomically modern human-archaic biological turnover. In the recently discovered cave site of Mughr el-Hamamah, Jordan, in situ flint artifacts comprise a diagnostic early Upper Paleolithic (EUP) assemblage. Unusually well-preserved charcoal from hearths and other anthropogenic features associated with the lithic material were subjected to acid-base-wet oxidation-stepped combustion (ABOx-SC) pretreatment. This article presents the ABOx-SC accelerator mass spectrometry (AMS) radiocarbon dates on nine charcoal specimens from a single palimpsest occupation layer. Date calibration was carried out using the INTCAL13 radiocarbon calibration dataset. With the bulk of the material dating to 45-39 ka cal BP (thousands of years calibrated before present), the Mughr el-Hamamah lithic artifacts reveal important differences from penecontemporaneous sites in the region, documenting greater technological variability than previously known for this time frame in the Levant. The radiocarbon data from this EUP archaeological context highlight remaining challenges for increasing chronological precision in documenting the MP-UP transition.

    • The fundamental challenge in the synthesis/design optimization of energy systems is the definition of system configuration and design parameters. The traditional way to operate is to follow the previous experience, starting from the existing design solutions. A more advanced strategy consists in the preliminary identification of a superstructure that should include all the possible solutions to the synthesis/design optimization problem and in the selection of the system configuration starting from this superstructure through a design parameter optimization. This top–down approach cannot guarantee that all possible configurations could be predicted in advance and that all the configurations derived from the superstructure are feasible. To solve the general problem of the synthesis/design of complex energy systems, a new bottom–up methodology has been recently proposed by the authors, based on the original idea that the fundamental nucleus in the construction of any energy system configuration is the elementary thermodynamic cycle, composed only by the compression, heat transfer with hot and cold sources and expansion processes. So, any configuration can be built by generating, according to a rigorous set of rules, all the combinations of the elementary thermodynamic cycles operated by different working fluids that can be identified within the system, and selecting the best resulting configuration through an optimization procedure. In this paper, the main concepts and features of the methodology are deeply investigated to show, through different applications, how an artificial intelligence can generate system configurations of various complexity using preset logical rules without any “ad hoc” expertise.
