    • A passive satellite radiometer operating at submillimetre wavelengths can measure cloud ice water path (IWP), ice particle size, and cloud altitude. The paper first discusses the scientific background for such measurements. Formal scientific mission requirements are derived, based on this background and earlier assessments. The paper then presents a comprehensive prototype instrument and mission concept, and demonstrates that it meets the requirements. The instrument is a conically scanning 12-channel radiometer with channels between 183 and 664 GHz, proposed to fly in tandem with one of the Metop satellites. It can measure IWP with a relative accuracy of approximately 20% and a detection threshold of approximately 2 g m−2. The median mass equivalent sphere diameter of the ice particles can be measured with an accuracy of approximately 30 µm, and the median IWP cloud altitude can be measured with an accuracy of approximately 300 m. All the above accuracies are median absolute error values; root mean square error values are approximately twice as high, due to rare outliers.

    • Hemoglobin (Hb) adducts are used to measure reactive compounds/metabolites in vivo. Schiff base adducts from aldehydes to N-termini in Hb have been measured by GC-MS/MS after stabilisation through reduction, and detachment by a modified Edman procedure. This paper describes a further development using 5-hydroxymethylfurfural (HMF) and its probable metabolite, 2,5-furandialdehyde (FDA), as model compounds. Reference compounds were synthesized and characterized. The conditions for the reduction of the Schiff bases were optimized using NaBH(3)CN as a mild reducing agent, and steps used in the earlier method could be deleted. The adduct from FDA could not be specifically analysed, as selective reduction of the imine could not be achieved. In a few samples of human blood, background levels of 10-35 pmol/g globin of the HMF adduct were observed. Half-lifes of the reversible Schiff base adduct from HMF were determined to 3.4h at 37 degrees C and 10.9h at 25 degrees C. The developed method showed good sensitivity and reproducibility for the analysis of the Schiff base from HMF, with improvements regarding simplicity of work-up procedures due to mild conditions. The developed method could be explored for application to adducts from other aldehydes bound as Schiff bases to N-termini in Hb.

    • Observational results of diurnal variations of humidity from Odin-SMR and AURA-MLS, and cloud ice mass from Odin-SMR and CloudSat are presented for the first time. Comparisons show that the retrievals of humidity and cloud ice from these two satellite combinations are in good agreement. The retrieved data are combined from four almost evenly distributed times of the day allowing mean values, amplitudes and phases of the diurnal variations around 200 hpa to be estimated. This analysis is applied to six climatologically distinct regions, five located in the tropics and one over the subtropical northern Pacific Ocean. The strongest diurnal cycles are found over tropical land regions, where the amplitude is ∼7 RHi for humidity and ∼50% for ice mass. The greatest ice mass for these regions is found during the afternoon, and the humidity maximum is observed to lag this peak by ∼6 h. Over tropical ocean regions the variations are smaller and the maxima in both ice mass and humidity are found during the early morning. Observed results are compared with output from three climate models (ECHAM, EC-EARTH and CAM3). Direct measurement-model comparisons were not possible because the measured and modelled cloud ice masses represent different quantities. To make a meaningful comparison, the amount of snow had to be estimated from diagnostic parameters of the models. There is a high probability that the models underestimate the average ice mass (outside the 1-σ uncertainty). The models also show clear deficiencies when it comes to amplitude and phase of the regional variations, but to varying degrees. © 2010 Author(s).

  • Contribution of antibody-based protein profiling to the human chromosome-centric proteome project (C-HPP)

    • A gene-centric Human Proteome Project has been proposed to characterize the human protein-coding genes in a chromosome-centered manner to understand human biology and disease. Here, we report on the protein evidence for all genes predicted from the genome sequence based on manual annotation from literature (UniProt), antibody-based profiling in cells, tissues and organs and analysis of the transcript profiles using next generation sequencing in human cell lines of different origins. We estimate that there is good evidence for protein existence for 69% (n = 13985) of the human protein-coding genes, while 23% have only evidence on the RNA level and 7% still lack experimental evidence. Analysis of the expression patterns shows few tissue-specific proteins and approximately half of the genes expressed in all the analyzed cells. The status for each gene with regards to protein evidence is visualized in a chromosome-centric manner as part of a new version of the Human Protein Atlas (www.proteinatlas.org).

    • A co-existence of chemical dependence and other psychiatric syndromes is commonly referred to as “dual-diagnosis.” This categorization is commonly made by social workers in several European countries assigned the primary responsibility for the care of drug and alcohol dependence. Here, we examined the validity of this categorization through systematic, structured patient evaluation following a minimum of 3 weeks of abstinence from drugs and alcohol. Less than one-third of patients originally labelled as suffering from “dual-diagnosis” by the social services did in fact obtain any Axis I DSM IIIR diagnosis, and less than half of the patients had any psychiatric diagnosis other than dependence. Syndromes commonly discussed in the context of self-medication, i.e., unipolar depression and anxiety syndromes, were not over-represented compared to a population sample, while chronic psychoses and bipolar syndromes were highly significantly more common. We conclude that the dual-diagnosis concept, unless substantiated through stringent diagnostic procedures by psychiatrically trained personnel, may be of questionable utility in caring for patients presenting with psychiatric symptoms and substance dependence. A systematic individual evaluation in an alcohol- and drug-free state of sufficient duration is necessary to obtain a basis for an adequate individual treatment plan.

    • The fluoroquinolone sensitivity of Pseudomonas aeruginosa and Klebsiella pneumoniae was studied by determinations of Minimum Inhibitory Concentrations (MICs) of norfloxacin and ciprofloxacin in consecutive clinical isolates. In P. aeruginosa, resistance or reduced susceptibility (MIC > or = 8 mg/l for norfloxacin and > or = 2 mg/l for ciprofloxacin) was found in 17% (9/54) of the isolates. Reduced susceptibility (MIC > or = 2 mg/l for norfloxacin) was found in 12% (10/84) of K. pneumoniae isolates. P. aeruginosa strains isolated in 1991 had reduced susceptibility to both norfloxacin and ciprofloxacin significantly more often than isolates from 1984-85 (p = 0.02). 10/84 K. pneumoniae strains from 1991 had reduced susceptibility to norfloxacin, compared with 1/34 from 1984-85. This difference was not statistically significant (p = 0.11).

  • A Population-based Investigation of the Autoantibody Profile in Mothers of Children with Atrioventricular Block

    • The objective of the study was to investigate the antigen specificity and occurrence of individual autoantibodies in mothers of children diagnosed with atrioventricular (AV) block in a nation-wide setting. Patients with AV block detected before 15 years of age were identified using national quality registries as well as a network of pediatric and adult cardiologists and rheumatologists at the six university hospitals in Sweden. Patients with gross heart malformations, surgically or infectiously induced blocks were excluded. Blood samples were obtained from the mothers and maternal autoantibody profile, including the occurrence of antibodies against Ro52, Ro60, La, SmB, SmD, RNP-70k, RNP-A, RNP-C, CENP-C, Scl-70, Jo-1, ribosomal RNP and histones was investigated in 193 mothers of children with AV block by immunoblotting and ELISA. Autoantibody reactivity was detected in 48% (93/193) of the mothers of children with AV block. In autoantibody-positive mothers, the vast majority, 95% (88/93), had antibodies against Ro52, while 63% (59/93) had autoantibodies to Ro60 and 58% (54/93) had autoantibodies to La. In addition, 13% (12/93) of the autoantibody-positive mothers had antibodies to other investigated antigens besides Ro52, Ro60 and La, and of these anti-histone antibodies were most commonly represented, detected in 8% (7/93) of the mothers. In conclusion, this Swedish population-based study confirms that maternal autoantibodies may associate with heart block in the child. Further, our data demonstrate a dominant role of Ro52 antibodies in association with AV block.

  • Efficient protein binders for the C-reactive protein from a designed chemically modified peptide library

    • A polypeptide conjugate synthesized by coupling a small organic molecule to the side chain of an amino acid residue in a designed 42-residue polypeptide binds the C-reactive protein (CRP) essentially irreversibly. The specificity in human serum is equal to that of an avian antibody although the size is only 1/30 and the structure unordered. The polypeptide conjugate binds CRP several orders of magnitude more tightly than the small molecule due to the fact that one amino acid has been modified to include a more strongly interacting side chain. The polypeptide was selected from a 16-membered set of sequences with no prior relationship to the target protein and designed to fold into a helix-loop-helix motif. The results suggest that synthetic amino acid alphabets with more strongly interacting side chains can be used to form polypeptides with improved binding properties in comparison to those engineered by biological methods.

    • High-affinity binders for the C-reactive protein (CRP), with dissociation constants in the pM to nM range and selectivities in human serum comparable to those of antibodies, were obtained by conjugation of 16 designed polypeptides to phosphocholine, a small molecule that binds CRP with a KDvalue of 5I . The polypeptides were not designed specifically to recognize CRP and bind by an adapted fit mechanism.
