  • Effects of shading and simulated grazing on carbon sequestration in a tropical seagrass meadow

    • 1. There is an ongoing world-wide decline of seagrass ecosystems, one of the world's most efficient carbon sink habitats. In spite of this, there is a clear lack of studies experimentally testing the effects of anthropogenic disturbances on carbon sequestration of seagrass systems. 2. We assessed the effects of two disturbances of global concern on the carbon sink function in a five-month in situ experiment within a tropical seagrass (Thalassia hemprichii) meadow by testing the impacts of shading and simulated grazing at two levels of intensity using shading cloths and clipping of shoot tissue. We measured the effects of these disturbances on the carbon sequestration process by assessing the net community production (NCP), carbon and nitrogen content in tissue biomass, and organic matter and THAA (total hydrolysable amino acids) in the sediment down to 40 cm depth. 3. Treatments of high-intensity shading and high-intensity clipping were similarly impacted and showed a significantly lower NCP and carbon content in the below-ground biomass compared to the seagrass control. No significant effects were seen in organic carbon, total nitrogen, C:N ratio and THAA in the sediment for the seagrass treatments. However, both clipping treatments showed different depth profiles of carbon and THAA compared to the seagrass control, with lower organic carbon and THAA content in the surface sediment. This can be explained by the clipping of shoot tissue causing a less efficient trapping of allochthonous carbon and reduced input of shredded seagrass leaves to the detritus sediment layer. In the clipping plots, erosion of the surface sediment occurred, which was also most likely caused by the removal of above-ground plant biomass. 4. Synthesis. Our findings show that during the course of this experiment, there were no impacts on the sedimentary carbon while the high-intensity disturbances caused a clear depletion of carbon biomass and reduced the seagrass meadow's capacity to sequester carbon. From a long-term perspective, the observed effect on the carbon biomass pool in the high-intensity treatments and the sediment erosion in the clipping plots may lead to loss in sedimentary carbon.

    • The efficiency of thin-layer capping in reducing sediment-to-water fluxes and bioaccumulation of polychlorinated dibenzo-p-dioxins and dibenzofurans, hexachlorobenzene, and octachlorostyrene was investigated in a boxcosm experiment. The influence of cap thickness (0.5-5 cm) and different cap materials was tested using a three-factor experimental design. The cap materials consisted of a passive material (coarse or fine limestone or a marine clay) and an active material (activated carbon (AC) or kraft lignin) to sequester the contaminants. The cap thickness and the type of active material were significant factors, whereas no statistically significant effects of the type of passive material were observed. Sediment-to-water fluxes and bioaccumulation by the two test species, the surface-dwelling Nassarius nitidus and the deep-burrowing Nereis spp., decreased with increased cap thickness and with addition of active material. Activated carbon was more efficient than lignin, and a ∼90% reduction of fluxes and bioaccumulation was achieved with 3 cm caps with 3.3% AC. Small increases in fluxes with increased survival of Nereis spp. indicated that bioturbation by Nereis spp. affected the fluxes.

  • Ecosystem effects of materials proposed for thin-layer capping of contaminated sediments

    • Ecotoxicological effects of 2 carbonaceous and 7 mineral capping materials suggested for in situ remediation of contaminated sediments in the Grenland fjords, Norway, were investigated in a mesocosm experiment. The primary objective was to compare the various materials with regard to potentially harmful effects on the benthic ecosystem. The materials assessed were activated carbon, Kraft-lignin, sand and clay materials, and 3 industrial by-products. Using sediment box-core samples with intact benthic communities, effects on structural (bacterial, macro-and meiofauna diversity) and functional (sediment-to-water nutrient fluxes, oxygen fluxes and bacterial production) endpoints were assessed. Significant deviations from the control (no capping) were detected for all of the tested materials for at least one endpoint. Generally, materials similar to the indigenous sediment (clay, sand) had relatively low deviations from the control, whereas industrial products (plaster, 2 types of crushed marble) resulted in deviations for most endpoints and large reductions in community richness and abundance. For example, at the end of the experimental period, the number of macrofauna taxa was <10 in these treatments, compared to >27 in uncapped mesocosm and field control sediments. The results from the study show that reducing harmful ecosystem effects from thin-layer capping by selecting capping materials based on robust, multi-endpoint mesocosm bench-tests is both possible and recommendable. Potential ecosystem impacts are particularly important to consider when large areas and areas with adequate ecological status are considered for thin-layer capping.

    • Three types of thin-layer caps with activated carbon (AC) were tested in situ in experimental plots (10 x 10 m) in Trondheim harbor, Norway, using AC + clay, AC-only or AC + sand. One year after capping, intact sediment cores were collected from the amended plots for ex situ surveys of the capping efficiency in reducing the PAH and PCB aqueous concentrations and bioaccumulation by the polychaete Hediste diversicolor and the clam Abra nitida. Reduced pore water concentrations were observed in all AC treatments. The capping efficiency was in general AC + clay > AC-only > AC + sand. AC + clay reduced bioaccumulation of PAH and PCB congeners between 40% and 87% in the worms and between 67% and 97% in the clams. Sediment capped with AC-only also led to reduced bioaccumulation of PCBs, while AC + sand showed no reduction in bioaccumulation. Thus the best thin-layer capping method in this study was AC mixed with clay.

  • In situ remediation of contaminated sediments using thin-layer capping : efficiency in contaminant retention and ecological implications

    • Hydrophobic organic contaminants (HOCs) often reside in sediments sorbed to particles, most tightly to particles with high content of organic carbon. If persistent, such pollutants can accumulate in the sediment for many years and constitute a contamination risk for sediment-living organisms and organisms at higher trophic levels, including humans.Since traditional remediation techniques are associated with complications (e.g. release of contaminants during dredging operations, disturbance of benthic faunal communities), or constraints (handling of large amounts of contaminated sediment and water, limitations due to depth and size of the area, high costs), there is a need for new alternative methods.In situ remediation through thin-layer capping (a few centimeter cover) with a sorbing material such as activated carbon (AC) has been proposed as an alternative remediation method. Compared to traditional remediation techniques, AC amendment in a thin layer means less material handling and lower costs and is assumed to be less disruptive to benthic communities. The objectives of this thesis were to investigate the ecological effects from thin layer capping as well as the efficiency in contaminant retention.Thin layer capping amended with AC proved to reduce availability of HOCs to the tested organisms, the gastropod Nassarius nitidus (Paper II), the clam Abra nitida (Paper III) and to polychaete worms  (Paper II and III). The remediation technique also decreased the sediment-to-water fluxes of the contaminants (Paper II and III).However, AC amended thin-layer capping was also found to cause negative biological effects. In laboratory studies with only a few species the negative effects were minor, or difficult to discern with the endpoints used (Paper II and III). In a larger multi-species mesocosm (boxcore) study, on the other hand, the negative effects were more prominent (Paper I) and in a large scale field study the benthic community was found to be profoundly disturbed by the AC amendment, with the effects persisting or even worsening ca one year (14 months) post amendment (Paper IV).

  • Large-scale field study on thin-layer capping of dioxin contaminated sediments in the Grenland fjords, Norway : Effects on marine benthic fauna

    • The Grenland fjords on the South East Coast of Norway are heavily contaminated with dioxins and furans after emissions from past industrial activities. Thin layer capping with a 1.1 to 3.7 cm cap was tested as a remediation option in a large-scale in situ study in two different parts of the fjord system, in the Ormefjord at 25-30 meters depth and in the Eidangerfjord at 80-100 meters depth. Three different capping materials: Limestone gravel, Clay, and powdered activated carbon (AC) mixed into clay (AC+clay) was compared to untreated reference fields in order to evaluate their effects on contaminant sequestering and possible effects on the benthic communities. Sediment to water fluxes of contaminants were significantly reduced by the capping materials, especially with AC-clay and is reported in a companion study.This study discusses the ecological effects of the remediation 1 and 14 months post treatment. Capping with Clay and Lime had minor and short-lasting effects on benthic fauna. Capping with AC+clay, however, had led to profound and more long-lasting perturbations of the macrofauna. An initial massive decline in filter feeders and suspension feeders was observed after 1 month in the shallower Ormefjord. The negative effects got worse after 14 months and resulted in dramatic reductions of all feeding guilds. The number of species, organism abundances and biomass in the AC+clay field were ca 80-90 % lower compared to the reference fields after 14 months.The negative effects were less pronounced at the deeper (80-100 meters) location in the Eidangerfjord and were also stable with time, suggesting that the benthic community the deeper habitat was more resilient to the capping compared to the shallower community in Ormefjord.The differences in response of the two communities are hypothesized to be due to the higher macrofaunal diversity in the deeper location, as well as to differences in abiotic factors such as available food and temperature. Results from this study show that amendment with powdered AC can lead to serious perturbations of the benthic community, at least initially, i.e. one year post capping in this study. These results stresses that further long-term monitoring of these benthic communities is necessary before capping with AC+clay could be advocated as a potential remediation option.

Intact sediment cores were collected from the in situ remediated plots to study the capping efficiency of the various AC treatments in reducing the aqueous concentrations and the bioaccumulation of polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) in the polychaete Hediste diversicolor and the clam Abra nitida. Reduced aqueous concentrations were observed in all AC-treatments, but generally AC+clay appeared to be superior to the other tested treatments. Capping efficiency by AC+clay, in terms of reduced bioaccumulation of PAHs and PCBs, ranged between 40 % and 87% in the worms and between 67% and 97% in the clams. Sediment capped with AC-only also led to reduced bioaccumulation of PCBs, while AC+sand showed no reduction in bioaccumulation. The worms had lower relative lipid content in the AC-only treatment after exposure.

    • The effects of suppression of the carbonic anhydrase (CA) activity by a CA-inhibitor, acetazolamide (AA), on the photosynthetic activities of photosystem II (PS II) particles from higher plants were investigated. AA along with CA-activity inhibits the PS II photosynthetic electron transfer and the AA-induced suppression is totally reversed by the addition of bicarbonate (3-5 mM). Similar effect of recovery in the PS II photosynthetic activity was also revealed upon the addition of known artificial electron donors (potassium ferrocyanide and TMPD). Significance and possible functions of CA for the PS II donor side are discussed.

    • In situ amendment of contaminated sediments using activated carbon (AC) is a recent remediation technique, where the strong sorption of contaminants to added AC reduces their release from sediments and uptake into organisms. The current study describes a marine underwater field pilot study in Trondheim harbor, Norway, in which powdered AC alone or in combination with sand or clay was tested as a thin-layer capping material for polycyclic aromatic hydrocarbon (PAH)-contaminated sediment. Several novel elements were included, such as measuring PAH fluxes, no active mixing of AC into the sediment, and the testing of new manners of placing a thin AC cap on sediment, such as AC+clay and AC+sand combinations. Innovative chemical and biological monitoring methods were deployed to test capping effectiveness. In situ sediment-to-water PAH fluxes were measured using recently developed benthic flux chambers. Compared to the reference field, AC capping reduced fluxes by a factor of 2-10. Pore water PAH concentration profiles were measured in situ using anew passive sampler technique, and yielded a reduction factor of 2-3 compared to the reference field. The benthic macrofauna composition and biodiversity were affected by the AC amendments, AC + clay having a lower impact on the benthic taxa than AC-only or AC + sand. In addition, AC + clay gave the highest AC recoveries (60% vs 30% for AC-only and AC + sand) and strongest reductions in sediment-to-water PAH fluxes and porewater concentrations. Thus, application of an AC-clay mixture is recommended as the optimal choice of the currently tested thin-layer capping methods for PAHs, and more research on optimizing its implementation is needed.

    • The effects of a 60 min exposure to photosynthetic photon flux densities ranging from 300 to 2200-mu-mol m-2 s-1 on the photosynthetic light response curve and on PS II heterogeneity as reflected in chlorophyll a fluorescence were investigated using the unicellular green alga Chlamydomonas reinhardtii. It was established that exposure to high light acts at three different regulatory or inhibitory levels; 1) regulation occurs from 300 to 780-mu-mol m-2 s-1 where total amount of PS II centers and the shape of the light response curve is not significantly changed, 2) a first photoinhibitory range above 780 up to 1600-mu-mol m-2 s-1 where a progressive inhibition of the quantum yield and the rate of bending (convexity) of the light response curve can be related to the loss of Q(B)-reducing centers and 3) a second photoinhibitory range above 1600-mu-mol m-2 s-1 where the rate of light saturated photosynthesis also decreases and convexity reaches zero. This was related to a particularly large decrease in PS II(alpha) centers and a large increase in spill-over in energy to PS I.

  • RECOVERY OF PHOTOSYNTHESIS AND PHOTOSYSTEM-II FLUORESCENCE IN CHLAMYDOMONAS-REINHARDTII AFTER EXPOSURE TO 3 LEVELS OF HIGH LIGHT

    • Recovery from 60 min of photoinhibitory treatment at photosynthetic photon flux densities of 500, 1400 and 2200-mu-mol m-2 s-1 was followed in cells of the green alga Chlamydomonas reinhardtii grown at 125-mu-mol m-2 s-1. These light treatments re resent photoregulation, moderate photoinhibition and strong photoinhibition, respectively. Treatment in photoregulatory light resulted in an increased maximal rate of oxygen evolution (P(max)) and an increased quantum yield (PHI), but a 15% decrease in F(V)/F(M). Treatment at moderately photoinhibitory light resulted in a 30% decrease in F(V)/F(M) and an approximately equal decrease in PHI. Recovery in dim light restored F(V)/F(M) within 15 and 45 min after high light treatment at 500 and 1 400-mu-mol m-2 s-1 respectively. Convexity (THETA), a measure of the extent of co-limitation between PS II turnover and whole-chain electron transport, and PHI approached, but did not reach the control level during recovery after exposure to 1 400-mu-mol m 2 s-1, whereas P(max) increased above the control. Treatment at 2200-mu-mol m-2 s-1 resulted in a strong reduction of the modeled parameters PHI, THETA and P(max). Subsequent recovery was initially rapid but the rate decreased, and a complete recovery was not reached within 120 min. Based on the results, it is hypothesized that exposure to high light results in two phenomena. The first, expressed at all three light intensities, involves redistribution within the different aspects of PS II heterogeneity rather than a photoinhibitory destruction of PS II reaction centers. The second, most strongly expressed at 2200-mu-mol m-2 s-1, is a physical damage to PS II shown as an almost total loss of PS II(alpha) and PS II Q(B)-reducing centers. Thus recovery displayed two phases, the first was rapid and the only visible phase in algae exposed to 500 and 1 400-mu-mol m-2 s-1. The second phase was slow and visible only in the later part of recovery in cells exposed to 2 200-mu-mol m-2 s-1.

    • OBJECTIVE: Definition of the molecular basis of the Reunion and the Bombay red cell and salivary H-deficient phenotypes. METHODS: Sequence and expression of FUT1 and FUT2 genes from H-deficient individuals. Family segregation analysis of the mutations responsible for the fucosyltransferase defects of H, secretor and Lewis systems. RESULTS: The Indian red cell H null Bombay phenotype depends on a new mutation of the FUT1 gene. T725-->G changing Leu242-->Arg. Their salivary nonsecretor phenotype is secondary to a complete deletion of the FUT2 gene. The red cell H weak Reunion phenotype depends on another new mutation of FUT1, C349-->T which induces a change of His117-->Tyr. Their salivary nonsecretor phenotype is due to the known Caucasian inactivating mutation G428-->A. CONCLUSION: Single prevalent FUT1 and FUT2 point mutations and a deletion are responsible for the Indian Bombay H null and the Reunion H weak phenotypes found on Reunion island. This is in contrast with other H-deficient phenotypes where sporadic nonprevalent inactivating mutations are the rule.

  • Blue Carbon Storage in Tropical Seagrass Meadows Relates to Carbonate Stock Dynamics, Plant–Sediment Processes, and Landscape Context : Insights from the Western Indian Ocean

    • Globally, seagrass ecosystems are considered major blue carbon sinks and thus indirect contributors to climate change mitigation. Quantitative estimates and multi-scale appraisals of sources that underlie long-term storage of sedimentary carbon are vital for understanding coastal carbon dynamics. Across a tropical–subtropical coastal continuum in the Western Indian Ocean, we estimated organic (Corg) and inorganic (Ccarb) carbon stocks in seagrass sediment. Quantified levels and variability of the two carbon stocks were evaluated with regard to the relative importance of environmental attributes in terms of plant–sediment properties and landscape configuration. The explored seagrass habitats encompassed low to moderate levels of sedimentary Corg (ranging from 0.20 to 1.44% on average depending on species- and site-specific variability) but higher than unvegetated areas (ranging from 0.09 to 0.33% depending on site-specific variability), suggesting that some of the seagrass areas (at tropical Zanzibar in particular) are potentially important as carbon sinks. The amount of sedimentary inorganic carbon as carbonate (Ccarb) clearly corresponded to Corg levels, and as carbonates may represent a carbon source, this could diminish the strength of seagrass sediments as carbon sinks in the region. Partial least squares modelling indicated that variations in sedimentary Corg and Ccarb stocks in seagrass habitats were primarily predicted by sediment density (indicating a negative relationship with the content of carbon stocks) and landscape configuration (indicating a positive effect of seagrass meadow area, relative to the area of other major coastal habitats, on carbon stocks), while seagrass structural complexity also contributed, though to a lesser extent, to model performance. The findings suggest that accurate carbon sink assessments require an understanding of plant–sediment processes as well as better knowledge of how sedimentary carbon dynamics are driven by cross-habitat links and sink–source relationships in a scale-dependent landscape context, which should be a priority for carbon sink conservation.

    • This experimental study quantified and compared particle-mixing and solute transport by the polychaetes Marenzelleria neglecta (2 g ww, 3200 ind.m(-2)) and Hediste diversicolor (2 g ww, 800 ind.m(-2)) in Baltic Sea sediments. Particle tracers (luminophores) were added to the sediment surface and their vertical distribution in the sediment was measured after 10 d. The rate of particle mixing was quantified using a gallery-diffusion model calculating the biodiffusion coefficient D(b) and the non-local transport parameter r. Bioirrigation was measured by adding an inert solute tracer (bromide) to the overlying water 1, 1.5 and 2 d before the end of the experiment, and quantified by calculating the net bromide flux and fitting the bromide profiles to a 1D diffusion model providing an apparent biodiffusion coefficient D(a). The two polychaete worms displayed similar particle-mixing and solute transport efficiencies (based on total biomass) despite different modes of bioturbation. However, H. diversicolor was a more efficient particle-reworker and M. neglecta a more efficient bioirrigator, on an individual level. H. diversicolor buried a higher percentage (13%) of luminophores below the top 0.5 cm surface layer than M. neglecta (6%). D(b) did not differ between the two species (2.4 x 10(-3) cm(2) d(-1)) indicating a similar rate of diffusive mixing of the top sediment, however, the non-local transport parameter r was 2.5 y(-1) for H. diversicolor and zero for M. neglecta, suggesting no significant particle-transport below the biodiffusive layer by M. neglecta. The average individual net bromide fluxes obtained were ca. 0.01 mL min(-1) for H. diversicolor and 0.003 mL min(-1) for M. neglecta, corresponding to an area-specific rate of ca. 12 L m(-2) d(-1) at the used densities. D(a) did not differ between the two polychaetes, suggesting a higher individual solute exchange efficiency of M. neglecta considering the much higher ventilation rates reported for H. diversicolor than for Marenzelleria sp. The ongoing colonization of Baltic Sea sediments by M. neglecta at high densities may thus lead to an enhanced soluble release of both nutrients and contaminants. These results add information to the understanding of the potential effects of the invasion of M. neglecta on sediment biogeochemistry when competing with and/or replacing native species.

    • A potentiometric titration method was used to study the adverse effect of arsenate (As(V) and arsenite (As(III) on inorganic carbon uptake in suspensions of the green alga Scenedesmus obliquus. The measurements were performed in a closed CO2-system with diluted synthetic seawater (1 parts per thousand salinity) as ionic medium. Usually, the algal chlorophyll concentration was 0.4 mg dm-3, while the arsenate- and arsenite-concentrations were varied within the limits 0.1 to 200 mumol dm-3. In some experiments arsenate toxicity was studied in the presence of 1 to 100 mumol dm-3 of phosphate (P(V)). With concentrations of arsenate or arsenite less than 0.1 mumol dm-3 no toxic effects were observed. However, at As-concentrations of 200 mumol dm-3, the algal carbon uptake was reduced by 41% with arsenate and 29% with arsenite, i.e., arsenate is more toxic to Scenedesmus obliquus than arsenite. The toxicity of arsenate was negligible in the presence of a ten fold excess of phosphate. This is probably due to chemical similarities between arsenate and phosphate causing competition between the ions for the binding sites. The importance of taking the speciation as well as the buffer capacity of the algal system into account, when calculating the carbon uptake, is also discussed.

  • Capping efficiency of various carbonaceous and mineral materials for in situ remediation of marine sediments contaminated with PCDD/Fs, OCS and HCB

    • The efficiency of various thin-layer capping materials in reducing the sediment-to-water flux and benthic organism bioaccumulation of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), hexachlorobenzene (HCB) and octachlorostyrene (OCS) was investigated in a boxcosm experiment. The influence of cap layer thickness (0.5-5 cm) and different cap materials were tested using a three-factor experimental design. The capping materials consisted of a passive carrier (coarse or fine limestone material, or a marine clay sediment), and an active material (activated carbon (AC) or kraft lignin) to sequester the contaminants and decrease their bioavailability. Macrofauna was added to the boxes to get a semi-natural bioturbation. The sediment-to-water flux was measured using passive (SPMD) samplers, and the bioaccumulation by the surface-dwelling gastropod Hinia reticulata and the deep-burrowing polychaetes Nereis spp. was determined. Results showed substantial decreases in both flux and bioaccumulation as a result of thin-layer capping. The thickness of the capping layer and the choice of active material were important factors, while the use of different types of passive materials was not statistically significant for any of the observed endpoints. Flux and bioaccumulation decreased with increased cap thickness, and could be further decreased with addition of active material. Activated carbon was more efficient than lignin, and a ~90% reduction of the flux and bioaccumulation, compared to uncapped control sediment, could be achieved with 3 cm caps with 3.3% AC (g C/g ww clay). The reduction was generally larger in the surface-dwelling H. reticulata than in Nereis spp., and the magnitude of the reduction was frequently similar between Nereis spp. and sediment-to-water fluxes. The latter was interpreted to indicate a link between Nereis spp. bioirrigation and sediment-to-water fluxes. Furthermore, the reduction in sediment-to-water flux was dependent on the hydrophobicity of the congeners, with less hydrophobic congeners achieving a larger reduction than more hydrophobic congeners.

    • The regulation of the supply of oxaloacetate (OAA) for mitochondrial metabolism via phosphoenolpyruvate carboxylase (PEPC) by covalent modification is studied in barley (Hordeum vulgare L.) leaf protoplasts in light or darkness as well as under photorespiratory or non-photorespiratory conditions. Extracts for studies on in vivo PEPC phosphorylation were prepared from barley leaf protoplasts by rapid filtration, fractionating the cell within less than 1 s. Measurements of in vitro PEPC activity were performed on samples quickly frozen in liquid nitrogen to break the cell and stop metabolism and thus preserve the in vivo activation state. The relative PEPC phosphorylation state increased upon illumination and decreased upon redarkening under photorespiratory and non-photorespiratory conditions. PEPC activity measured in the presence of malate (3 mM) under photorespiratory conditions showed the same response indicating that a light-induced increase in PEPC activity and decrease in malate sensitivity is caused by an increased phosphorylation level of the PEPC protein. PEPC activity was pH dependent. At the physiological cytosolic pH, activity was suboptimal, but most sensitive towards malate inhibition and glucose 6-phosphate stimulation. The presence of malate increased the sensitivity of PEPC activity towards pH changes. The response of PEPC activity to changing pH was not affected by changes in the activation state of the enzyme. The K-m (phosphoenolpyruvate, PEP) is about 1 mM. Upon illumination the K-m (PEP) decrease significantly. V-max was unaffected by the light treatment. The presence of physiological concentrations of glucose 6-phosphate decreased K-m (PEP) 5- to 10-fold and increased V-max by about 35%. The effect of glucose 6-phosphate was strongest (up to 7-fold) at subsaturating PEP concentrations stimulating PEPC activity to nearly maximal rates. The results show that an increase in PEPC phosphorylation state causes an increase in PEPC activity as well as in substrate affinity leading to an increased production of OAA in the light.

Glycine, serine, pyruvate, acetyl-CoA, glycolate, fructose 1,6-bisphosphate, fructose 2,6-bisphosphate and ADP had no significant effect on PEPC activity. Malate, aspartate and glutamate were strong inhibitors of PEPC activity decreasing the activity more in light versus darkness. However, at the physiological cytosolic concentration of these metabolites under the respective conditions, inhibition of PEPC activity was about the same with the exception of aspartate which inhibits more under non-photorespiratory than under photorespiratory conditions. 2-Oxoglutarate and glyoxylate decreased PEPC activity by 20 to 40% in the range of its physiological cytosolic concentration. Inhibition by physiological cytosolic concentrations of glutamine was limited. Glucose B-phosphate, fructose B-phosphate, 3-phosphoglycerate, dihydroxyacetonphosphate and P-i stimulated PEPC activity significantly in their physiological cytosolic concentration range. Physiological cytosolic concentrations of glucose 6-phosphate and fructose 6-phosphate activated PEPC activity to about the same extent under all conditions applied, while 3-phosphoglycerate and dihydroxyacetonphosphate stimulating Stronger under non-photorespiratory versus photorespiratory conditions. Moreover, dihydroxyacetonphosphate stimulated PEPC activity more in light versus darkness under non-photorespiratory conditions. P-i activation of PEPC activity decreases in light versus darkness under non-photorespiratory conditions. Stimulation of PEPC activity by citrate in its physiological concentration range is limited. Glucose 1-phosphate and AMP activated PEPC activity only at concentrations higher than their physiological levels in the cytosol. Determinations of PEPC activity in the presence of different malate/glucose 6-phosphate ratios revealed that glucose 6-phosphate totally relieved the inhibitory effect of malate. The regulatory properties of PEPC activity will be discussed in relation to its functions in C-3 plants.

  • Molecular cloning and biochemical characterization of carbonic anhydrase from Populus tremula x tremuloides

    • A leaf cDNA library from hybrid aspen, Populus tremula x tremuloides, was constructed. From this two different cDNA clones, denoted CAla and CAlb, encoding a chloroplastic carbonic anhydrase (CA) were isolated and DNA sequenced. Analysis of the deduced amino acid sequences showed that the isolated CAs belong to the beta-CA family, and have identities around 70% to other dicotyledonous plant CAs. The two hybrid aspen cDNA clones display a high nucleotide sequence identity, only 12 nucleotides differ. Since only one gene copy of this soluble chloroplastic CA is present in the nuclear genome, we postulate that the two isolated cDNA clones are alleles. Northern blot hybridization revealed a CA transcript of ca. 1300 bases, 140 bases shorter than in pea. Western and northern blot hybridizations on crude protein extracts and on total RNA, respectively, isolated from stem and leaves, showed that hybrid aspen CA is expressed specifically in the leaf under the growth conditions used. Based on the deduced amino acid sequence, the mature hybrid aspen CA enzyme subunit has a molecular mass of 24.8 kDa. The enzyme was over-expressed in Escherichia coli, and purified by affinity chromatography. Biochemical characterization showed that the protein structure and the CO2-hydration activity are similar to the pea enzyme. Molecular characterization of a CA from a perennial plant has not previously been performed, and it demonstrates that both the structure and activity of hybrid aspen CA resembles CAs from annual plants.

    • The pollution state in the Skagerrak and Kattegat was investigated by determination of pollutant concentrations and toxicity of sediment samples from 11 stations in the area. A comparison was made with the sediment from a reference site near the Faroe Islands. Polycyclic aromatic hydrocarbons (PAH) and organochlorines were determined in whole sediment and heavy metals and ammonia were analysed in filtered pore water. Toxicity was bioassayed in whole sediment with Nitocra spinipes and Daphnia magna, in pore water with Mytilus edulis larvae and in solvent extracts from sediment with tests measuring etoxyresorufin-O-deethylase (EROD) activity in Oncorhyncus mykiss and rate of denitrification. Sites close to Goteborg and in an area from the Oslo fjord to the Norwegian Trench were most polluted. Sediment from the Faroe Islands was least polluted and also least toxic. Multivariate statistical analysis indicates that the different tests were sensitive to different kinds of pollutants. Effects on mussel larvae correlated strongest with the occurrence of ammonia, manganese, cadmium and PAHs, Nitocra with alpha-hexachlorocyclohexane (HCH) and p,p'-DDD, Daphnia with arsenic and gamma-HCH, fish EROD activity with benzo[ghi]perylene and unknown compounds associated with organic carbon, and denitrification with chlordanes, dieldrin and a few PAHs. The results indicate that sampling sites close to Goteborg are so polluted that harmful effects on the ecosystem probably occur.

    • Carbonic anhydrases catalyse the reversible hydration of CO2, increasing the interconversion between CO2 and HCO3- + H+ in living organisms. The three evolutionarily unrelated families of carbonic anhydrases are designated alpha-, beta -and gamma -CA. Animals have only the alpha -carbonic anhydrase type of carbonic anhydrase, but they contain multiple isoforms of this carbonic anhydrase. In contrast, higher plants, algae and cyanobacteria may contain members of all three CA families. Analysis of the Arabidopsis database reveals at least 14 genes potentially encoding carbonic anhydrases. The database also contains expressed sequence tags (ESTs) with homology to most of these genes. Clearly the number of carbonic anhydrases in plants is much greater than previously thought. Chlamydomonas, a unicellular green alga, is not far behind with five carbonic anhydrases already identified and another in the EST database. In algae, carbonic anhydrases have been found in the mitochondria, the chloroplast thylakoid, the cytoplasm and the periplasmic space. In C-3 dicots, only two carbonic anhydrases have been localized, one to the chloroplast stroma and one to the cytoplasm. A challenge for plant scientists is to identify the number, location and physiological roles of the carbonic anhydrases.

  • Carbonic anhydrase activities in pea thylakoids. : A photosystem II core complex-associated carbonic anhydrase

    • Pea thylakoids with high carbonic anhydrase (CA) activity (average rates of 5000 µmol H+ (mg Chl)–1 h–1 at pH 7.0) were prepared. Western blot analysis using antibodies raised against the soluble stromal beta-CA from spinach clearly showed that this activity is not a result of contamination of the thylakoids with the stromal CA but is derived from a thylakoid membrane-associated CA. Increase of the CA activity after partial membrane disintegration by detergent treatment, freezing or sonication implies the location of the CA in the thylakoid interior. Salt treatment of thylakoids demonstrated that while one part of the initial enzyme activity is easily soluble, the rest of it appears to be tightly associated with the membrane. CA activity being measured as HCO3 – dehydration (dehydrase activity) in Photosystem II particles (BBY) was variable and usually low. The highest and most reproducible activities (approximately 2000 µmol H+ (mg Chl)–1 h–1) were observed in the presence of detergents (Triton X-100 or n-octyl-beta-D-glucopyranoside) in low concentrations. The dehydrase CA activity of BBY particles was more sensitive to the lipophilic CA inhibitor, ethoxyzolamide, than to the hydrophilic CA inhibitor, acetazolamide. CA activity was detected in PS II core complexes with average rate of 13,000 µmol H+ (mg Chl)–1 h–1 which was comparable to CA activity in BBY particles normalized on a PS II reaction center basis.

    • Aims Quality of life is an important outcome measure in studies of urinary incontinence. Electronic collection of data has several advantages. We examined the reliability of the Swedish version of the highly recommended condition-specific quality of life questionnaire International Consultation on Incontinence Modular Questionnaire-Lower Urinary Tract Symptoms Quality of Life (ICIQ-LUTSqol), in paper and web-based formats in women with stress urinary incontinence.Methods Women aged 1870 years, with stress urinary incontinence at least once weekly, were recruited via the project's website and answered the ICIQ-LUTSqol questionnaire. Respondents completed either the paper version twice (n?=?78), or paper and web-based versions once each (n?=?54). The ICIQ validation protocol was followed.Results The mean interval between answers was 18.1 (SD?=?3.1) days in the paper versus paper setting and 15.0 (SD?=?7.8) days in the paper versus web-based setting. Internal consistency was excellent, with Cronbach's alpha coefficients of 0.87 for the paper version and 0.86 for the web-based version. There was a high degree of agreement of overall scores with intraclass correlations in the paper versus paper and paper versus web-based settings: 0.95 (P?Conclusions The questionnaire is reliable in women with stress urinary incontinence, and it can be used in either a paper or a web-based version.Neurourol.
