    • Efforts to extract a Greenland ice core with a complete record of the Eemian interglacial (130,000 to 115,000 years ago) have until now been unsuccessful. The response of the Greenland ice sheet to the warmer-than-present climate of the Eemian has thus remained unclear. Here we present the new North Greenland Eemian Ice Drilling ('NEEM') ice core and show only a modest ice-sheet response to the strong warming in the early Eemian. We reconstructed the Eemian record from folded ice using globally homogeneous parameters known from dated Greenland and Antarctic ice-core records. On the basis of water stable isotopes, NEEM surface temperatures after the onset of the Eemian (126,000 years ago) peaked at 8 +/- 4 degrees Celsius above the mean of the past millennium, followed by a gradual cooling that was probably driven by the decreasing summer insolation. Between 128,000 and 122,000 years ago, the thickness of the northwest Greenland ice sheet decreased by 400 +/- 250 metres, reaching surface elevations 122,000 years ago of 130 +/- 300 metres lower than the present. Extensive surface melt occurred at the NEEM site during the Eemian, a phenomenon witnessed when melt layers formed again at NEEM during the exceptional heat of July 2012. With additional warming, surface melt might become more common in the future.

    • An important share of paleoclimatic information is buried within the lowermost layers of deep ice cores. Because improving our records further back in time is one of the main challenges in the near future, it is essential to judge how deep these records remain unaltered, since the proximity of the bedrock is likely to interfere both with the recorded temporal sequence and the ice properties. In this paper, we present a multiparametric study (delta D-delta O-18(ice), delta O-18(atm), total air content, CO2, CH4, N2O, dust, high-resolution chemistry, ice texture) of the bottom 60 m of the EPICA (European Project for Ice Coring in Antarctica) Dome C ice core from central Antarctica. These bottom layers were subdivided into two distinct facies: the lower 12 m showing visible solid inclusions (basal dispersed ice facies) and the upper 48 m, which we will refer to as the basal clean ice facies. Some of the data are consistent with a pristine paleoclimatic signal, others show clear anomalies It is demonstrated that neither large-scale bottom refreezing of subglacial water, nor mixing (be it internal or with a local basal end term from a previous/initial ice sheet configuration) can explain the observed bottom-ice properties. We focus on the high-resolution chemical profiles and on the available remote sensing data on the subglacial topography of the site to propose a mechanism by which relative stretching of the bottom-ice sheet layers is made possible, due to the progressively confining effect of subglacial valley sides. This stress field change, combined with bottom-ice temperature close to the pressure melting point, induces accelerated migration recrystallization, which results in spatial chemical sorting of the impurities, depending on their state (dissolved vs. solid) and if they are involved or not in salt formation. This chemical sorting effect is responsible for the progressive build-up of the visible solid aggregates that therefore mainly originate from within, and not from incorporation processes of debris from the ice sheet's substrate. We further discuss how the proposed mechanism is compatible with the other ice properties described. We conclude that the paleoclimatic signal is only marginally affected in terms of global ice properties at the bottom of EPICA Dome C, but that the timescale was considerably distorted by mechanical stretching of MIS20 due to the increasing influence of the subglacial topography, a process that might have started well above the bottom ice. A clear paleoclimatic signal can therefore not be inferred from the deeper part of the EPICA Dome C ice core. Our work suggests that the existence of a flat monotonic ice bedrock interface, extending for several times the ice thickness, would be a crucial factor in choosing a future oldest ice drilling location in Antarctica.

It consists of 18 amino acids, is located in the C-terminal region of the protein and corresponds functionally to a cellulose binding domain (CBD), the first one to be reported in a fungal xylanase. The amino acid sequence of this peptide shows no homology with any known CBD

    • In Saccharomyces cerevisiae, phosphate uptake is mainly dependent on the proton-coupled Pho84 permease under phosphate-limited growth conditions. Phosphate addition causes Pho84-mediated activation of the protein kinase A (PKA) pathway as well as rapid internalization and vacuolar breakdown of Pho84. We show that Pho84 undergoes phosphate-induced phosphorylation and subsequent ubiquitination on amino acids located in the large middle intracellular loop prior to endocytosis. The attachment of ubiquitin is dependent on the ubiquitin conjugating enzymes Ubc2 and Ubc4. In addition, we show that the Pho84 endocytotic process is delayed in strains with reduced PKA activity. Our results suggest that Pho84-mediated activation of the PKA pathway is responsible for its own downregulation by phosphorylation, ubiquination, internalization, and vacuolar breakdown.

  • Melting and refreezing beneath Roi Baudouin Ice Shelf (East Antarctica) inferred from radar, GPS, and ice core data

    • Ice-penetrating radar profiles across the grounding line of a small ice-rise promontory located within the Roi Baudouin Ice Shelf in the Dronning Maud Land sector of East Antarctica show downward dipping englacial radar-detected reflectors. Model results indicate that this reflector pattern is best fit by including basal melting of at least 15 cm a(-1). This rate of melting is low compared with rates observed on larger ice shelves in both West and East Antarctica. Ice cores extracted from a rift system close to the ice-rise promontory show several meters of marine ice accreted beneath the shelf. These observations of low rates of basal melting, and limited distribution of accreted marine ice suggest that either Antarctic surface water may reach the ice shelf base or that circulation beneath the shelf is likely dominated by the production of high salinity shelf water rather than the incursion of circumpolar deep water, implying a weak sub-shelf circulation system here. Many of the ice shelves located along the coast of Dronning Maud Land are, like Roi Baudouin Ice Shelf, characterized by frequent ice rises and promontories. Therefore, it is highly likely that these are also of shallow bathymetry and are subject to similarly weak side-shelf basal melting and refreezing.

  • A simple and updated pneumatic method for uniaxial ice compression in the laboratory : experimental settings and creep test results on glacier ice

    • Creep tests provide invaluable data to better understand the physical properties of ice under various conditions. We describe here a simple, updated pneumatic apparatus for experimental studies of ice rheological properties. The apparatus is designed to perform two simultaneous compression creep tests either in a cold room or in atmospheric conditions when coupled to an external cooling circulator. We present results from calibration tests of the apparatus and provide new data from creep tests performed on temperate glacier ice samples. These calibration and creep results show that the apparatus is able to provide fast and reliable mechanical ice characterization. The secondary creep rates measured in this study range between 1.59 x 10(-8) s(-1) (at 0.21 MPa) and 4.38 x 10(-7) s(-1) (at 0.71 MPa) at -10 degrees C for quasi-isotropic ice, which is consistent with former standard published data. The corresponding mean parameter, A, is 5.20 x 10(-16) s(-1) kPa(-3), which also compares well with the range of reported other studies.

  • Plasma deposition of polymer composite films incorporating nanocellulose whiskers

    • In a trend for sustainable engineering and functionalization of surfaces, we explore the possibilities of gas phase processes to deposit nanocomposite films. From an analysis of pulsed plasma polymerization of maleic anhydride in the presence of nanocellulose whiskers, it seems that thin nanocomposite films can be deposited with various patterns. By specifically modifying plasma parameters such as total power, duty cycle, and monomer gas pressure, the nanocellulose whiskers are either incorporated into a buckled polymer film or single nanocellulose whiskers are deposited on top of a polymeric film. The density of the latter can be controlled by modifying the exact positioning of the substrate in the reactor. The resulting morphologies are evaluated by optical microscopy, AFM, contact angle measurements and ellipsometry.

    • Samples from two ice cores drilled at Lomonosovfonna, Svalbard, covering the period 1957-2009, and 1650-1995, respectively, were analyzed for NO(3)(-)concentrations, and NO3- stable isotopes (N-15 and O-18). Post-1950 N-15 has an average of (-6.91.9), which is lower than the isotopic signal known for Summit, Greenland but agrees with values observed in recent Svalbard snow and aerosol. Pre-1900 N-15 has an average of (4.21.6)parts per thousand suggesting that natural sources, enriched in the N-15 isotope, dominated before industrialization. The post-1950 O-18 average of (75.1 +/- 4.1)parts per thousand agrees with data from low and polar latitudes, suggesting similar atmospheric NOy (NOy=NO+NO2+HNO3) processing pathways. The combination of anthropogenic source N-15 and transport isotope effect was estimated as -29.1 parts per thousand for the last 60years. This value is below the usual range of NOx (NOx=NO+NO2) anthropogenic sources which is likely the result of a transport isotope effect of -32 parts per thousand. We suggest that the N-15 recorded at Lomonosovfonna is influenced mainly by fossil fuel combustion, soil emissions, and forest fires; the first and second being responsible for the marked decrease in N-15 observed in the post-1950s record with soil emissions being associated to the decreasing trend in N-15 observed up to present time, and the third being responsible for the sharp increase of N-15 around 2000.

    • The Na(+)-coupled, high-affinity Pho89 plasma membrane phosphate transporter in Saccharomyces cerevisiae has so far been difficult to study because of its low activity and special properties. In this study, we have used a pho84Deltapho87Deltapho90Deltapho91Delta quadruple deletion strain of S. cerevisiae devoid of all transporter genes specific for inorganic phosphate, except for PHO89, to functionally characterize Pho89 under conditions where its expression is hyperstimulated. Under these conditions, the Pho89 protein is strongly upregulated and is the sole high-capacity phosphate transporter sustaining cellular acquisition of inorganic phosphate. Even if Pho89 is synthesized in cells grown at pH 4.5-8.0, the transporter is functionally active under alkaline conditions only, with a K(m) value reflecting high-affinity properties of the transporter and with a transport rate about 100-fold higher than that of the protein in a wild-type strain. Even under these hyperexpressive conditions, Pho89 is unable to sense and signal extracellular phosphate levels. In cells grown at pH 8.0, Pho89-mediated phosphate uptake at alkaline pH is cation-dependent with a strong activation by Na(+) ions and sensitivity to carbonyl cyanide m-chlorophenylhydrazone. The contribution of H(+)- and Na(+)-coupled phosphate transport systems in wild-type cells grown at different pH values was quantified. The contribution of the Na(+)-coupled transport system to the total cellular phosphate uptake activity increases progressively with increasing pH.
