  • PROVE-Pre-Eclampsia Obstetric Adverse Events: Establishment of a Biobank and Database for Pre-Eclampsia

    • Pre-eclampsia is a leading cause of maternal and perinatal morbidity and mortality. The burden of disease lies mainly in low-middle income countries. The aim of this project is to establish a pre-eclampsia biobank in South Africa to facilitate research in the field of pre-eclampsia with a focus on phenotyping severe disease.The approach of our biobank is to collect biological specimens, detailed clinical data, tests, and biophysical examinations, including magnetic resonance imaging (MRI) of the brain, MRI of the heart, transcranial Doppler, echocardiography, and cognitive function tests.Women diagnosed with pre-eclampsia and normotensive controls are enrolled in the biobank at admission to Tygerberg University Hospital (Cape Town, South Africa). Biological samples and clinical data are collected at inclusion/delivery and during the hospital stay. Special investigations as per above are performed in a subset of women. After two months, women are followed up by telephonic interviews. This project aims to establish a biobank and database for severe organ complications of pre-eclampsia in a low-middle income country where the incidence of pre-eclampsia with organ complications is high. The study integrates different methods to investigate pre-eclampsia, focusing on improved understanding of pathophysiology, prediction of organ complications, and potentially future drug evaluation and discovery.

    • The 1993 Ground-based Cloud Experiment on Great Dun Fell used a wide range of measurements of trace gases, aerosol particles and cloud droplets at five sites to study their sources and sinks especially those in cloud. These measurements have been interpreted using a variety of models. The conclusions add to our knowledge of air pollution, acidification of the atmosphere and the ground, eutrophication and climate change. The experiment is designed to use the hill cap cloud as a flow-through reactor, and was conducted in varying levels of pollution typical of much of the rural temperate continental northern hemisphere in spring-time.

  • Associations Between Soluble fms-Like Tyrosine Kinase-1 and Placental Growth Factor and Disease Severity Among Women With Preterm Eclampsia and Preeclampsia

    • Background The angiogenic factors soluble fms-like tyrosine kinase-1 (sFlt-1) and placental growth factor (PlGF) are postulated to be pathogenic disease drivers of preeclampsia. If true, then circulating levels should become more deranged with increasing disease severity. Methods and Results We investigated the association between circulating sFlt-1 and PlGF levels and severe adverse maternal outcomes among 348 women with preeclampsia. Compared with 125 women with preeclampsia without severe features, 25 women with preeclampsia and any of hemolysis, elevated liver enzymes, low platelet count syndrome, disseminated intravascular coagulation, or severe renal involvement had sFlt-1 levels that were 2.63-fold higher (95% CI, 1.81-3.82), sFlt-1/PlGF levels that were 10.07-fold higher (95% CI, 5.36-18.91) and PlGF levels that were 74% lower (adjusted fold change, 0.26 [95% CI, 0.18-0.39]). Compared with 125 women with preeclampsia without severe features, 37 with eclampsia had sFlt-1 levels that were 2-fold higher (2.02 [95% CI, 1.32-3.09]), sFlt-1/PIGF levels that were 4.71-fold higher (95% CI, 2.30-9.66) and PIGF levels that were 63% lower (0.43-fold change [95% CI, 0.27-0.68]). Compared with those without severe features, preeclampsia with severe hypertension (n=146) was also associated with altered angiogenic levels (sFlt-1, 1.71-fold change [95% CI, 1.39-2.11]; sFlt/PlGF, 2.91 [95% CI, 2.04-4.15]; PlGF, 0.59 [95%CI 0.47-0.74]). We also found that sFlt-1 and PlGF levels were altered by the number of maternal complications experienced. Conclusions Further angiogenic imbalance among women with preeclampsia is likely a pathogenic disease driver responsible for the life-threatening maternal complications.

  • Field validation of the droplet aerosol analyser

    • A new instrument for the study of cloud droplets and its relation to aerosol particles, the droplet aerosol analyser (DAA), was for the first time used in a field campaign. The DAA has the unique feature of measuring the ambient size of cloud droplets or cloud interstitial aerosol particles together with the size of its dry residue. This is obtained with a two-parameter data acquisition technique which results in a three-dimensional data set (ambient size, dry residue size, number concentration). The principle and design of the DAA is briefly described. The DAA was intercompared with differential mobility particle sizers, particulate volume monitors and a forward scattering spectrometer probe with respect to interstitial and cloud droplet dry residue size distribution as well as particle-size-dependent scavenging due to cloud droplet nucleation and for cloud droplet number concentration and size distribution and cloud liquid water concentration. Overall, the DAA showed good agreement with respect to all these six aerosol/cloud properties.

    • An outline is presented here of the Po Valley Fog Experiment 1989, carried out within the EUROTRAC‐GCE project. This experiment is a joint effort by several European research groups from 5 countries. The physical and chemical behaviour of the fog multiphase system was studied experimentally following the temporal evolution of the relevant chemical species in the different phases (gas, droplet, interstitial aerosol) and the evolution of micrometeorological and microphysical conditions, from the pre‐fog situation through the whole fog evolution, to the post‐fog period. Some general results, useful for describing the general features of the fog system, are presented here, while specific scientific questions on the different processes taking place within the system itself will be addressed in other companion papers of this same issue.

  • Nucleation and growth of Ti2 AlN thin films deposited by reactive magnetron sputtering onto MgO(111)

    • The nucleation and growth of Ti2 AlN thin films on MgO(111) substrates during dual direct current reactive magnetron cosputtering from Ti and Al targets in an Ar N2 atmosphere at a substrate temperature of 690 °C have been investigated. Time and thickness dependent in situ specular x-ray reflectivity and x-ray diffraction in combination with cross-sectional transmission electron microscopy and Rutherford backscattering spectroscopy reveal the formation of competing phases for slight N superstoichiometry with respect to Ti2 AlN. The stoichiometry deviations initiate the layer-by-layer growth of a ∼380 Å thick epitaxial N-substoichiometric cubic (Ti1-x Alx) Ny layer. N-vacancy driven diffusion of Ti and Al leads to decomposition of this metastable solid solution into nanosized cubic TiN y′ and AlN y″ domains as well as to a solid-state reaction with the MgO(111) by formation of a Mg2 (Al:Ti) O4 spinel, reducing the transformed (Ti1-x Alx) Ny layer thickness down to ∼60 Å. Local AlN y″ domains serve as templates for Ti2 AlN nucleation at higher thicknesses. At the same time TiN y′ and AlN y″ serve as a sink for excess gas phase N during the subsequent polycrystalline Ti2 AlN growth with random (Ti1-x Alx) Ny renucleation as a tissue phase along Ti2 AlN grain boundaries. The individual Ti2 AlN grains with vertical sizes up to the total thickness retain local epitaxy to the substrate, with basal planes nonparallel to the substrate interface. Concurrently the (Ti1-x Alx) Ny layer is further reduced by inward Ti2 AlN grain growth along the basal planes. © 2007 American Institute of Physics.

    • Introduction We aimed to assess cognitive function in women with preeclampsia stratified by severity, before and after onset of disease. Material and methods Prospective study performed at a referral hospital in Cape Town, South Africa. Pregnant women between 20 and 42 weeks of gestation with eclampsia, pulmonary edema and preeclampsia without severe features, and a normotensive pregnancy were approached. Women were included at diagnosis of preeclampsia or at admission for delivery (women with normotensive pregnancies). Two cognitive assessments, the Cognitive Failure Questionnaire to assess the cognitive function subjectively before inclusion in the study, and Montreal Cognitive Assessment to assess the current cognitive function objectively before discharge from the hospital after delivery, were performed. Results We included 61 women with eclampsia, 28 with preeclampsia complicated by pulmonary edema, 38 with preeclampsia without severe features, and 26 with normotensive pregnancies. There was no difference in cognitive function from early pregnancy between groups. Women with eclampsia and preeclampsia complicated by pulmonary edema scored lower on the Montreal Cognitive Assessment at time of discharge compared with women with normotensive pregnancies. The results were attenuated in pulmonary edema after adjustment for confounders. Conclusions Women with preeclampsia complicated by pulmonary edema and in particular eclampsia had impaired cognitive function after onset of disease compared with women with normotensive pregnancies. The impairment did not seem to be present before onset of disease. Women with preeclampsia without severe features did not have impaired cognitive function.

    • The aim of this study was to evaluate the Collaborative and Proactive Solutions (CPS) intervention in an alternative educational setting for students with behavioural problems. The effect of the CPS intervention on students’ off-task behaviour was studied using a single-subject experimental design that included two students with behavioural problems via systematic direct observations and direct behaviour ratings. The usability of the CPS intervention was investigated through questionnaires and interviews with the participating students and teacher assistant. The results revealed no significant effects of the CPS intervention on students’ off-task behaviour. The students and teacher assistant viewed the CPS intervention as acceptable but the teacher assistant’s ratings of the feasibility of the intervention were lower, together with the ratings of the extent to which the intervention matched the socio-political climate of the educational setting.

  • Single-cell analyses of X Chromosome inactivation dynamics and pluripotency during differentiation

    • Pluripotency, differentiation, and X Chromosome inactivation (XCI) are key aspects of embryonic development. However, the underlying relationship and mechanisms among these processes remain unclear. Here, we systematically dissected these features along developmental progression using mouse embryonic stem cells (mESCs) and single-cell RNA sequencing with allelic resolution. We found that mESCs grown in a ground state 2i condition displayed transcriptomic profiles diffused from preimplantation mouse embryonic cells, whereas EpiStem cells closely resembled the post-implantation epiblast. Sex-related gene expression varied greatly across distinct developmental states. We also identified novel markers that were highly enriched in each developmental state. Moreover, we revealed that several novel pathways, including PluriNetWork and Focal Adhesion, were responsible for the delayed progression of female EpiStem cells. Importantly, we “digitalized” XCI progression using allelic expression of active and inactive X Chromosomes and surprisingly found that XCI states exhibited profound variability in each developmental state, including the 2i condition. XCI progression was not tightly synchronized with loss of pluripotency and increase of differentiation at the single-cell level, although these processes were globally correlated. In addition, highly expressed genes, including core pluripotency factors, were in general biallelically expressed. Taken together, our study sheds light on the dynamics of XCI progression and the asynchronicity between pluripotency, differentiation, and XCI.

    • The paper presents an outline of the CHEMDROP field experiment, carried out in November 1994 at the field station of S. Pietro Capofiume in the Po Valley, Italy. The main objective of the project was to address the issue of the size-dependent chemical composition of fog droplets, by experimentally investigating the following processes, which are expected to affect (or be affected by) the chemical composition of fog droplets as a function of size: a) the connection of the size-dependent chemical composition of CCN to the size-dependent composition of fog droplets; b) the gas/liquid partitioning of the gaseous species NH3, SO2, HCHO, HNO3 in fog; c) the Fe(II)/Fe(III) redox cycle in fog water. Some general results and overall conclusions of the experiment are reported in this paper, while more specific scientific questions are discussed in other companion papers in this issue. CHEMDROP results show that several processes concur in determining the size-dependence of fog droplets chemical composition: nucleation scavenging of pre-existing CCN, fog dynamical evolution and gas/liquid exchange between interstitial air and fog droplets. Chemical transformations in the liquid phase can cause further changes in the chemical composition of the droplets. Only by taking into account the combination of all these processes, is it possible to explain the inhomogeneities in fog droplet chemical composition.

  • Polycomb repressive complex 2 shields naïve human pluripotent cells from trophectoderm differentiation

    • The first lineage choice in human embryo development separates trophectoderm from the inner cell mass. Naïve human embryonic stem cells are derived from the inner cell mass and offer possibilities to explore how lineage integrity is maintained. Here, we discover that polycomb repressive complex 2 (PRC2) maintains naïve pluripotency and restricts differentiation to trophectoderm and mesoderm lineages. Through quantitative epigenome profiling, we found that a broad gain of histone H3 lysine 27 trimethylation (H3K27me3) is a distinct feature of naïve pluripotency. We define shared and naïve-specific bivalent promoters featuring PRC2-mediated H3K27me3 concomitant with H3K4me3. Naïve bivalency maintains key trophectoderm and mesoderm transcription factors in a transcriptionally poised state. Inhibition of PRC2 forces naïve human embryonic stem cells into an ‘activated’ state, characterized by co-expression of pluripotency and lineage-specific transcription factors, followed by differentiation into either trophectoderm or mesoderm lineages. In summary, PRC2-mediated repression provides a highly adaptive mechanism to restrict lineage potential during early human development.

  • Microstructure and nonbasal-plane growth of epitaxial Ti2AlN thin films

    • Thin films of the Mn+1AXn (MAX) phase (M: early transition metal, A:A-group element, X: C and/or N, n=1-3) Ti2AlN were epitaxially grown onto single-crystal MgO(111) and MgO(100) substrates by dc reactive magnetron cosputtering from Ti and Al targets in an Ar/N2 gas mixture at a temperature of 690°C. To promote the nucleation of the MAX phase, a fcc (Ti0.63Al0.37)N seed layer was deposited before changing to Ti2AlN growth parameters. The nucleation processes have been studied by real-time in situ specular x-ray reflectivity. Independent of substrate orientation, the seed layer shows no roughening until its final thickness of approximately 100 Å, indicating pseudomorphic layer-by-layer growth. The MAX phase shows heteroepitaxial layer-by-layer growth on MgO(111), with increased surface roughening up to approximately 200 Å, whereas on MgO(100) the growth mode changes to Volmer-Weber-type already after three monolayers. X-ray scattering in Bragg-Brentano geometry of the final, approximately 1000 Å thick, Ti2AlN film reveals lattice parameters of c=13.463 Å and a=2.976 Å on the MgO(111) substrate and c=13.740 Å and a=2.224 Å on the MgO(100) substrate. From pole figure measurements the orientational relationship between film and substrate lattice was determined to be MgO{111}〈110〉//Ti2AlN{1012} 〈1210〉, regardless of the substrate orientation. This tilted, nonbasal-plane growth leads to a threefold grain orientation of Ti 2AlN along the MgO〈110〉 directions and a polycrystalline morphology confirmed by cross-sectional transmission electron microscopy. The growth can be assumed to take place in a lateral step-flow mode, i.e., emerging low surface free-energy (0001) planes, on which arriving atoms can diffuse until finding a step where they are bound to A facets. This growth process is irrespective of orientational relationship between substrate and film. However, in the present low-temperature case the partitioning of arriving Al and Ti atoms during nucleation is suppressed, which as a result of interfacial adaptation between substrate and film induces standing a-type planes during growth. © 2006 American Institute of Physics.

High substrate temperature deposition results in epitaxial Ti2AlN growth with basal planes parallel to the substrate surface. In contrast to reported high thermal stability for bulk Ti-Al-N M n+1AXn phases in air, Ti2AlN thin films in vacuum decompose already at ∼800°C. The decomposition proceeds by outward Al diffusion and evaporation, followed by detwinning of the as-formed Ti2N atomic layers into cubic TiNx and intermediate phases. © 2006 American Institute of Physics.

    • Phase, microstructure, and strain evolution during annealing of arc deposited TiZrAlN coatings are studied using in situ x-ray scattering and ex situ transmission electron microscopy. We find that the decomposition route changes from nucleation and growth of wurtzite AlN to spinodal decomposition when the Zr-content is decreased and the Al-content increases. Decomposition of Ti0.31Zr0.24Al0.45N results in homogeneously distributed wurtzite AlN grains in a cubic, dislocation-dense matrix of TiZrN consisting of domains of different chemical composition. The combination of high dislocation density, variation of chemical composition within the cubic grains, and evenly distributed wurtzite AlN grains results in high compressive strains, -1.1%, which are retained after 3 h at 1100 degrees C. In coatings with higher Zr-content, the strains relax during annealing above 900 degrees C due to grain growth and defect annihilation. (C) 2018 Elsevier B.V. All rights reserved.

  • Investigation of the spectroscopic properties of single defects in hexagonal boron nitride

    • Among the quantum systems capable of emitting single photons, the class of recently discovered defects in hexagonal boron nitride (hBN) is especially interesting, as these defects offer much desired characteristics such as narrow emission lines and photostability [1]. Like for any new class of quantum emitters, the first challenges to solve are the understanding of their photophysics as well as to find ways to facilitate integration in photonics structures. Here, we will show our investigation of the optical transition in hBN with different methods: Employing excitation with a short laser pulse the emission properties in case of linear and non-linear excitation can be compared [2]. We find clear antibunching signals that prove the single emitter character in both excitation cases. To gain further knowledge, we also obtain saturation curves. From a comparison of one- and two-photon case insights about the level structure of the defects can be obtained. The possibility to perform two-photon excitation makes this single photon emitter an interesting candidate as a biosensor.

  • Socioeconomic determinants of infant mortality: a worldwide study of 152 low-, middle-, and high-income countries

    • Background: To reach the Millennium Development Goals for health, influential international bodies advocate for more resources to be directed to the health sector, in particular medical treatment. Yet, health has many determinants beyond the health sector that are less evident than proximate predictors. Aim: To assess the relative importance of major socioeconomic determinants of population health, measured as infant mortality rate (IMR), at country level. Methods: National-level data from 152 countries based on World Development Indicators 2003 were used for multivariate linear regression analyses of five socioeconomic predictors of IMR: public spending on health, GNI/capita, poverty rate, income equality (Gini index), and young female illiteracy rate. Analyses were performed on a global level and stratified for low-, middle-, and high-income countries. Results: In order of importance, GNI/capita, young female illiteracy, and income equality predicted 92% of the variation in national IMR whereas public spending on health and poverty rate were non-significant determinants when adjusted for confounding. In low-income countries, female illiteracy was more important than GNI/capita. Income equality (Gini index) was an independent predictor of IMR in middle-income countries only. In high-income countries none of these predictors was significant. Conclusions: The relative importance of major health determinants varies between income levels, thus extrapolating health policies from high- to low-income countries is problematic. Since the size, per se, of public health spending does not independently predict health outcomes, functioning health systems are necessary to make health investments efficient. Potential health gains from improved female education and economic growth should be considered in low- and middle-income countries
