    • Surveys have shown that super-Earth and Neptune-mass exoplanets are more frequent than gas giants around low-mass stars, as predicted by the core accretion theory of planet formation. We report the discovery of a giant planet around the very-low-mass star GJ 3512, as determined by optical and near-infrared radial-velocity observations. The planet has a minimum mass of 0.46 Jupiter masses, very high for such a small host star, and an eccentric 204-day orbit. Dynamical models show that the high eccentricity is most likely due to planet-planet interactions. We use simulations to demonstrate that the GJ 3512 planetary system challenges generally accepted formation theories, and that it puts constraints on the planet accretion and migration rates. Disk instabilities may be more efficient in forming planets than previously thought.

  • TOI-1468: A system of two transiting planets, a super-Earth and a mini-Neptune, on opposite sides of the radius valley

    • We report the discovery and characterization of two small transiting planets orbiting the bright M3.0V star TOI-1468 (LSPM J0106+1913), whose transit signals were detected in the photometric time series in three sectors of the TESS mission. We confirm the planetary nature of both of them using precise radial velocity measurements from the CARMENES and MAROON-X spectrographs, and supplement them with ground-based transit photometry. A joint analysis of all these data reveals that the shorter-period planet, TOI-1468 b (P-b = 1.88 d), has a planetary mass of M-b = 3.21 +/- 0.24M(circle plus) and a radius of R-b = 1.280(-0.039)(+0.038) R-circle plus, resulting in a density of rho(b) = 8.39(-0.92)(+1.05) g cm(-3), which is consistent with a mostly rocky composition. For the outer planet, TOI-1468 c (P-c = 15.53 d), we derive a mass of M-c = 6.64(-0.68)(+0.67) M-circle plus,aradius of R-c = 2.06 +/- 0.04 R-circle plus, and a bulk density of rho(c) = 2.00(-0.19)(+0.21) g cm(-3), which corresponds to a rocky core composition with a H/He gas envelope. These planets are located on opposite sides of the radius valley, making our system an interesting discovery as there are only a handful of other systems with the same properties. This discovery can further help determine a more precise location of the radius valley for small planets around M dwarfs and, therefore, shed more light on planet formation and evolution scenarios.

  • Precise mass and radius of a transiting super-Earth planet orbiting the M dwarf TOI-1235: a planet in the radius gap?

    • We report the confirmation of a transiting planet around the bright weakly active M0.5 V star TOI-1235 (TYC 4384-1735-1, V ≈ 11.5 mag), whose transit signal was detected in the photometric time series of sectors 14, 20, and 21 of the TESS space mission. We confirm the planetary nature of the transit signal, which has a period of 3.44 d, by using precise RV measurements with the CARMENES, HARPS-N, and iSHELL spectrographs, supplemented by high-resolution imaging and ground-based photometry. A comparison of the properties derived for TOI-1235 b with theoretical models reveals that the planet has a rocky composition, with a bulk density slightly higher than that of Earth. In particular, we measure a mass of Mp = 5.9 ± 0.6 M⊕ and a radius of Rp = 1.69 ± 0.08 R⊕, which together result in a density of ρp = 6.7- 1.1+ 1.3 g cm-3. When compared with other well-characterized exoplanetary systems, the particular combination of planetary radius and mass places our discovery in the radius gap, which is a transition region between rocky planets and planets with significant atmospheric envelopes. A few examples of planets occupying the radius gap are known to date. While the exact location of the radius gap for M dwarfs is still a matter of debate, our results constrain it to be located at around 1.7 R⊕ or larger at the insolation levels received by TOI-1235 b (~60 S⊕). This makes it an extremely interesting object for further studies of planet formation and atmospheric evolution.

  • TOI-1801 b: A temperate mini-Neptune around a young M0.5 dwarf

    • We report the discovery, mass, and radius determination of TOI-1801 b, a temperate mini-Neptune around a young M dwarf. TOI-1801 b was observed in TESS sectors 22 and 49, and the alert that this was a TESS planet candidate with a period of 21.3 days went out in April 2020. However, ground-based follow-up observations, including seeing-limited photometry in and outside transit together with precise radial velocity (RV) measurements with CARMENES and HIRES revealed that the true period of the planet is 10.6 days. These observations also allowed us to retrieve a mass of 5.74 +/- 1.46 M-circle plus, which together with a radius of 2.08 +/- 0.12 R-circle plus, means that TOI-1801 b is most probably composed of water and rock, with an upper limit of 2% by mass of H-2 in its atmosphere. The stellar rotation period of 16 days is readily detectable in our RV time series and in the ground-based photometry. We derived a likely age of 600-800 Myr for the parent star TOI-1801, which means that TOI-1801 b is the least massive young mini-Neptune with precise mass and radius determinations. Our results suggest that if TOI-1801 b had a larger atmosphere in the past, it must have been removed by some evolutionary mechanism on timescales shorter than 1 Gyr.

    • Mixtures of ethyl (hydroxyethyl)cellulose (EHEC) and sodium dodecanoate (SDoD) were investigated using surface tension, conductivity and transmittance measurements. The parameters of the surfactant to polymer association processes such as the critical aggregation concentration (cac) and saturation of the polymer by SDoD (psp) were determined from the plots of surface tension and specific conductivity versus surfactant concentration. The transmittance versus [SDoD] profiles exhibited bands that, in conjunction with conductivity and surface tension data, gave support to the explanation of the behavior of EHEC-SDoD-water solutions. Transmittance bands determined in the presence of EHEC and surfactant are sensitive to the process of surfactant to polymer binding. In this process, a discontinuity point represented by a second broader peak in the transmittance plots was identified, implying a slight inhibition of the breakdown of the network process. This is explained as a kind of intermediate re-structural network of the complexes produced by the formation of regular SDoD micelles. It was demonstrated that the second conductivity breakpoint marks the polymer saturation by surfactant clusters, and the effect of the EHEC concentration on the process of binding can be described by a linear correlation when the psp breakpoint is plotted against the polymer concentration.     • Increased apoptotic shedding has been linked to intestinal barrier dysfunction and development of inflammatory bowel diseases (IBD). In contrast, physiological cell shedding allows the renewal of the epithelial monolayer without compromising the barrier function. Here, we investigated the role of live cell extrusion in epithelial barrier alterations in IBD.DesignTaking advantage of conditional GGTase and RAC1 knockout mice in intestinal epithelial cells (Pggt1biΔIECandRac1iΔIECmice), intravital microscopy, immunostaining, mechanobiology, organoid techniques and RNA sequencing, we analysed cell shedding alterations within the intestinal epithelium. Moreover, we examined human gut tissue and intestinal organoids from patients with IBD for cell shedding alterations and RAC1 function.ResultsEpithelialPggt1bdeletion led to cytoskeleton rearrangement and tight junction redistribution, causing cell overcrowding due to arresting of cell shedding that finally resulted in epithelial leakage and spontaneous mucosal inflammation in the small and to a lesser extent in the large intestine. Both in vivo and in vitro studies (knockout mice, organoids) identified RAC1 as a GGTase target critically involved in prenylation-dependent cytoskeleton dynamics, cell mechanics and epithelial cell shedding. Moreover, inflamed areas of gut tissue from patients with IBD exhibited funnel-like structures, signs of arrested cell shedding and impaired RAC1 function. RAC1 inhibition in human intestinal organoids caused actin alterations compatible with arresting of cell shedding.ConclusionImpaired epithelial RAC1 function causes cell overcrowding and epithelial leakage thus inducing chronic intestinal inflammation. Epithelial RAC1 emerges as key regulator of cytoskeletal dynamics, cell mechanics and intestinal cell shedding. Modulation of RAC1 might be exploited for restoration of epithelial integrity in the gut of patients with IBD.

    • Semi-arid areas, defined as those areas of the world where water is an important limitation for plant growth, have become the subject of increased interest due to the impacts of current global changes and sustainability of human lifestyles. While many ground-based reports of declining vegetation productivity have been published over the last decades, a number of recent publications have shown a nuanced and, for some regions, positive picture. With this background, the paper provides an analysis of trends in vegetation greenness of semi-arid areas using AVHRR GIMMS from 1981 to 2007. The vegetation index dataset is used as a proxy for vegetation productivity and trends are analyzed for characterization of changes in semi-arid vegetation greenness. Calculated vegetation trends are analyzed with gridded data on potential climatic constraints to plant growth to explore possible causes of the observed changes. An analysis of changes in the seasonal variation of vegetation greenness and climatic drivers is conducted for selected regions to further understand the causes of observed inter-annual vegetation changes in semi-arid areas across the globe. It is concluded that semi-arid areas, across the globe, on average experience an increase in greenness (0.015 NDVI units over the period of analysis). Further it is observed that increases in greenness are found both in semi-arid areas where precipitation is the dominating limiting factor for plant production (0.019 NDVI units) and in semi-arid areas where air temperature is the primarily growth constraint (0.013 NDVI units). Finally, in the analysis of changes in the intra-annual variation of greenness it is found that seemingly similar increases in greenness over the study period may have widely different explanations. This implies that current generalizations, claiming that land degradation is ongoing in semi-arid areas worldwide, are not supported by the satellite based analysis of vegetation greenness.   • TOI-4438 b: a transiting mini-Neptune amenable to atmospheric characterization

    • We report the confirmation and mass determination of a mini-Neptune transiting the M3.5 V star TOI-4438 (G 182-34) every 7.44 days. A transit signal was detected with NASA's TESS space mission in the sectors 40, 52, and 53. In order to validate the planet TOI-4438 b and to determine the system properties, we combined TESS data with high-precision radial velocity measurements from the CARMENES spectrograph, spanning almost one year, and ground-based transit photometry. We found that TOI-4438 b has a radius of Rb = 2.52 ± 0.13 R⊕ (5% precision), which together with a mass of Mb = 5.4 ± 1.1 M⊕ (20% precision), results in a bulk density of ρb = 1.85-0.44+0.51 g cm-3 (~28% precision), aligning the discovery with a volatile-rich planet. Our interior structure retrieval with a pure water envelope yields aminimum water mass fraction of 46% (1σ). TOI-4438 b is a volatile-rich mini-Neptune with likely H/He mixed with molecules, such as water, CO2, and CH4. The primary star has a J-band magnitude of 9.7, and the planet has a high transmission spectroscopy metric (TSM) of 136 ± 13. Taking into account the relatively warm equilibrium temperature of Teq = 435 ± 15 K, and the low activity level of its host star, TOI-4438 b is one of the most promising mini-Neptunes around an M dwarf for transmission spectroscopy studies.

  • Transformations through proximity flying : a phenomenological investigation

    • Participation in extreme sports has been linked to personal transformations in everyday life. Descriptions of lived experience resulting from transformative experiences are limited. Proximity flying, a relatively new discipline involving BASE jumping with a wingsuit where participants fly close to solid structures, is arguably one of the most extreme of extreme sports. The aim of this paper, part of a larger phenomenological study on the lived experience of proximity flying, is to explicate the ways in which participating in proximity flying influences the everyday lives of participants. Interpretative phenomenological analysis was used to explicate the lived experience of six proximity pilots. An analysis of interview transcripts revealed three significant themes describing the lived experience of participants. First, experiences of change were described as positive and skills developed through proximity flying were transferable into everyday life. Second, transformative experiences were considered fundamental to participants' perspectives on life. Third, experience of transformation influenced their sense of personal identity and facilitated flourishing in other aspects of everyday life. Participants were clear that their experiences in proximity flying facilitated a profound process of transformation which manifest as changes in everyday capabilities and behaviors, values and sense of identity.

  • Gliese 12 b: A Temperate Earth-sized Planet at 12 pc Ideal for Atmospheric Transmission Spectroscopy

    • Recent discoveries of Earth-sized planets transiting nearby M dwarfs have made it possible to characterize the atmospheres of terrestrial planets via follow-up spectroscopic observations. However, the number of such planets receiving low insolation is still small, limiting our ability to understand the diversity of the atmospheric composition and climates of temperate terrestrial planets. We report the discovery of an Earth-sized planet transiting the nearby (12 pc) inactive M3.0 dwarf Gliese 12 (TOI-6251) with an orbital period (P(or)b) of 12.76 days. The planet, Gliese 12 b, was initially identified as a candidate with an ambiguous P-orb from TESS data. We confirmed the transit signal and P-orb using ground-based photometry with MuSCAT2 and MuSCAT3, and validated the planetary nature of the signal using high-resolution images from Gemini/NIRI and Keck/NIRC2 as well as radial velocity (RV) measurements from the InfraRed Doppler instrument on the Subaru 8.2 m telescope and from CARMENES on the CAHA 3.5 m telescope. X-ray observations with XMM-Newton showed the host star is inactive, with an X-ray-to-bolometric luminosity ratio of log L-X/L-bol approximate to - 5.7. Joint analysis of the light curves and RV measurements revealed that Gliese 12 b has a radius of 0.96 +/- 0.05 R-circle plus, a 3 sigma mass upper limit of 3.9M(circle plus), and an equilibrium temperature of 315 +/- 6 K assuming zero albedo. The transmission spectroscopy metric (TSM) value of Gliese 12 b is close to the TSM values of the TRAPPIST-1 planets, adding Gliese 12 b to the small list of potentially terrestrial, temperate planets amenable to atmospheric characterization with JWST.

  • Effects of solriamfetol treatment on body weight in participants with obstructive sleep apnea or narcolepsy

    • Objectives: This analysis characterized changes in weight in participants with obstructive sleep apnea (OSA) or narcolepsy treated with solriamfetol (SunosiTM) 37.5 (OSA only), 75, 150, or 300 mg/d. Methods: In two 12-week, randomized, placebo-controlled trials and one 1-year open-label extension study, changes in weight were evaluated from baseline to end of study (week 12 or week 40 of the open -label extension [after up to 52 weeks of solriamfetol treatment]) in participants with OSA or narcolepsy. Results: After 12 weeks of solriamfetol treatment, median percent change in weight from baseline across all solriamfetol doses was-0.84%, compared with 0.54% for placebo, in participants with OSA; and-0.07%, compared with 3.08% for placebo, in participants with narcolepsy. After up to 52 weeks of solriamfetol treatment, overall median percent change in weight from baseline was-1.76%, which showed a dose-dependent pattern (75 mg, 0.57%; 150 mg,-1.2%; 300 mg,-2.5%).Results were similar in subgroups of participants with OSA or narcolepsy, with overall median percent changes in weight of-2.2% and-1.1%, respectively. After up to 52 weeks of solriamfetol treatment, the percentage of participants with weight loss >= 5% relative to baseline was 25.7% overall and increased in a dose -dependent manner (75 mg, 4.5%; 150 mg, 17.3%; 300 mg, 32.4%). Results were similar among sub-groups of participants with OSA or narcolepsy, with 26.4% and 24.2% of participants experiencing weight loss >= 5%, respectively. No weight-related treatment-emergent adverse events were serious.Conclusions: Solriamfetol treatment was associated with decreases in body weight in a dose-related manner.(c) 2022 The Authors.   • Two super-Earths at the edge of the habitable zone of the nearby M dwarf TOI-2095

    • The main scientific goal of TESS is to find planets smaller than Neptune around stars that are bright enough to allow for further characterization studies. Given our current instrumentation and detection biases, M dwarfs are prime targets in the search for small planets that are in (or near) the habitable zone of their host star. In this work, we use photometric observations and CARMENES radial velocity (RV) measurements to validate a pair of transiting planet candidates found by TESS. The data were fitted simultaneously, using a Bayesian Markov chain Monte Carlo (MCMC) procedure and taking into account the stellar variability present in the photometric and spectroscopic time series. We confirm the planetary origin of the two transiting candidates orbiting around TOI-2095 (LSPM J1902+7525). The star is a nearby M dwarf (d = 41.90 ± 0.03 pc, Teff = 3759 ± 87 K, V = 12.6 mag), with a stellar mass and radius of M∗ = 0.44 ± 0.02 M· and R∗ = 0.44 ± 0.02 R·, respectively. The planetary system is composed of two transiting planets: TOI-2095b, with an orbital period of Pb = 17.66484 ± (7 A - 10- 5) days, and TOI-2095c, with Pc = 28.17232 ± (14 A - 10- 5) days. Both planets have similar sizes with Rb = 1.25 ± 0.07 R· and Rc = 1.33 ± 0.08 R· for planet b and planet c, respectively. Although we did not detect the induced RV variations of any planet with significance, our CARMENES data allow us to set stringent upper limits on the masses of these objects. We find Mb < 4.1 M· for the inner and Mc < 7.4 M· for the outer planet (95% confidence level). These two planets present equilibrium temperatures in the range of 300 350 K and are close to the inner edge of the habitable zone of their star.

    • HD 235088 (TOI-1430) is a young star known to host a sub-Neptune-sized planet candidate. We validated the planetary nature of HD 235088 b with multiband photometry, refined its planetary parameters, and obtained a new age estimate of the host star, placing it at 600- 800 Myr. Previous spectroscopic observations of a single transit detected an excess absorption of He I coincident in time with the planet candidate transit. Here, we confirm the presence of He I in the atmosphere of HD 235088 b with one transit observed with CARMENES. We also detected hints of variability in the strength of the helium signal, with an absorption of -0.91 ± 0.11%, which is slightly deeper (2γ) than the previous measurement. Furthermore, we simulated the He I signal with a spherically symmetric 1D hydrodynamic model, finding that the upper atmosphere of HD 235088 b escapes hydrodynamically with a significant mass loss rate of (1.5-5) × 1010 g s-1 in a relatively cold outflow, with T = 3125 ±375 K, in the photon-limited escape regime. HD 235088 b (Rp = 2.045 ± 0.075 R⊕) is the smallest planet found to date with a solid atmospheric detection - not just of He I but any other atom or molecule. This positions it a benchmark planet for further analyses of evolving young sub-Neptune atmospheres.

    • Visible surface films, termed slicks, can extensively cover freshwater and marine ecosystems, with coastal regions being particularly susceptible to their presence. The sea-surface microlayer (SML), the upper 1-mm at the air-water interface in slicks (herein slick SML) harbors a distinctive bacterial community, but generally little is known about SML viruses. Using flow cytometry, metagenomics, and cultivation, we characterized viruses and bacteria in a brackish slick SML in comparison to non-slick SML as well as seawater below slick and non-slick areas (subsurface water = SSW). Size-fractionated filtration of all samples distinguished viral attachment to hosts and particles. The slick SML contained higher abundances of virus-like particles, prokaryotic cells, and dissolved organic carbon compared to non-slick SML and SSW. The community of 428 viral operational taxonomic units (vOTUs), 426 predicted as lytic, distinctly differed across all size fractions in the slick SML compared to non-slick SML and SSW. Specific metabolic profiles of bacterial metagenome-assembled genomes and isolates in the slick SML included a prevalence of genes encoding motility and carbohydrate-active enzymes (CAZymes). Several vOTUs were enriched in slick SML, and many virus variants were associated with particles. Nine vOTUs were only found in slick SML, six of them being targeted by slick SML-specific clustered-regularly interspaced short palindromic repeats (CRISPR) spacers likely originating from Gammaproteobacteria. Moreover, isolation of three previously unknown lytic phages for Alishewanella sp. and Pseudoalteromonas tunicata, abundant and actively replicating slick SML bacteria, suggests that viral activity in slicks contributes to biogeochemical cycling in coastal ecosystems.

In this perspective, we delineate next-generation biotechnologies for CCSU supported by engineering design principles derived from ecological processes inspired by three major biomes (plant-soil, deep biosphere, and marine). These are to interface with existing industrial infrastructure and, in some cases, tap into the carbon sink potential of nature. To develop ecosystem-inspired biotechnology, it is important to identify accessible control points of CO2 and CH4 within a given system as well as value-chain opportunities that drive innovation. In essence, we must supplement natural biogeochemical carbon sinks with new bioengineering solutions. © 2020 The Authors Atmospheric carbon emissions are driving global tipping points that must be addressed by reducing emissions in combination with carbon capture for sequestration and utilization. We outline that there is much to be gained by translating ecological processes that underpin global carbon cycles into engineering principles that harness plant-soil, deep biosphere and marine microbiomes for creation of new value chains and access to Earth's major carbon storage pools. Rapid innovation is required, and biotechnology has a certain role to play. © 2020 The Authors

    • The rare earth element (REE) composition of a fossil bone reflects its chemical alteration during diagenesis. Consequently, fossils presenting low REE concentrations and/or REE profiles indicative of simple diffusion, signifying minimal alteration, have been proposed as ideal candidates for paleomolecular investigation. We directly tested this prediction by conducting multiple biomolecular assays on a well-preserved fibula of the dinosaur Edmontosaurus from the Cretaceous Hell Creek Formation previously found to exhibit low REE concentrations and steeply-declining REE profiles. Gel electrophoresis identified the presence of organic material in this specimen, and subsequent immunofluorescence and enzyme-linked immunosorbant assays identified preservation of epitopes of the structural protein collagen I. Our results thereby support the utility of REE profiles as proxies for soft tissue and biomolecular preservation in fossil bones. Based on considerations of trace element taphonomy, we also draw predictions as to the biomolecular recovery potential of additional REE profile types exhibited by fossil bones.
