  • The effectiveness of an integrated BIM/UAV model in managing safety on construction sites

    • Introduction. A variety of approaches exist to achieve better construction safety performance, but only a few consider a combination of building information modeling (BIM) and unmanned aerial vehicles (UAVs).Method. This article presents a four-dimensional (4D) BIM/UAV-enabled safety management model based on IDEF0 language. In the first step, potential hazards are identified with the help of safety specialists’ experiences and BIM software used in the design of the structure. Then, a UAV monitors the location of the potential hazards. The third step involves the integration of the 4D BIM-based model and the information obtained from the UAV. Finally, the combined data are analyzed and interpreted, and site safety staff are notified about measures to be put in place to prevent accidents.Results. This model shows a strong relationship between the design and construction phases by using BIM in the design phase and UAVs in the construction phase. The proposed safety model was evaluated by construction safety specialists in a two-pronged approach.Conclusion. The number of fatal, non-fatal and property damage-causing accidents may be significantly lower when the proposed system was used.Practical application. This model allows safety specialists to identify hazards and develop suitable mitigation strategies.

  • Tribological properties of powder metallurgical tool steels used in powder compaction pressing dies

    • The tribological properties of two powder metallurgical (PM) tool steels, high and low nitrogen containing, are investigated by means of three different wear tests: ball-on-disc, rubber wheel and scratch test. The ball-on-disc tests showed two distinct friction curves corresponding to each material. In order to simulate the tribosystem existing in metal powder compaction dies, the rubber wheel and the scratch test were modified. The rubber wheel test was performed using ferrous powder instead of sand, and scratch testing was carried out by sliding a powder compact over the tool steels. The scratch tests indicated a higher steady-state coefficient of friction for the low nitrogen containing PM steel as compared with the high nitrogen containing alloy. Additionally, the results from the rubber wheel tests were in agreement with industrial experiences, showing the low nitrogen containing tool steel to suffer from severe galling.     • This study evaluates a simultaneous assessment of organic matter (OM) and trace elements (TE) bio-accessibility in substrate and digestate from a full-scale anaerobic digester by a sequential OM extraction method. Simultaneous release of TE was determined along with the extraction of different OM fractions and the effects of extracting reagents on characteristics of OM were evaluated by nuclear magnetic resonance (NMR) spectroscopy. The reagents used for sequential extraction of OM were not enough selective. However, proteins were particularly removed by 0.1 M NaOH, while 72% H2SO4 mainly extracted hemicellulose and cellulose. The OM fractionation allowed for simultaneous extraction of greater than60% of total As, Cd, Co, Fe, Mn, Ni and Zn, while the extraction was limited for Al, Cr, Cu, Mo, and Pb. In substrate, greater than50% of total As, Co, Mn and Ni and less than40% of total Fe, Zn and Mo were identified in bio-accessible fractions.   • Distinct HLA associations with autoantibody-defined subgroups in idiopathic inflammatory myopathies

    • Background In patients with idiopathic inflammatory myopathies (IIM), autoantibodies are associated with specific clinical phenotypes suggesting a pathogenic role of adaptive immunity. We explored if autoantibody profiles are associated with specific HLA genetic variants and clinical manifestations in IIM. Methods We included 1348 IIM patients and determined the occurrence of 14 myositis-specific or-associated autoantibodies. We used unsupervised cluster analysis to identify autoantibody-defined subgroups and logistic regression to estimate associations with clinical manifestations, HLA-DRB1, HLA-DQA1, HLA-DQB1 alleles, and amino acids imputed from genetic information of HLA class II and I molecules. Findings We identified eight subgroups with the following dominant autoantibodies: anti-Ro52, -U1RNP, -PM/Scl,-Mi2,-Jo1,-Jo1/Ro52,-TIF1 gamma or negative for all analysed autoantibodies. Associations with HLA-DRB1*11, HLA-DRB1*15, HLA-DQA1*03, and HLA-DQB1*03 were present in the anti-U1RNP-dominated subgroup. HLA-DRB1*03, HLA-DQA1*05, and HLA-DQB1*02 alleles were overrepresented in the anti-PM/Scl and anti-Jo1/ Ro52-dominated subgroups. HLA-DRB1*16, HLA-DRB1*07 alleles were most frequent in anti-Mi2 and HLA- DRB1*01 and HLA-DRB1*07 alleles in the anti-TIF1 gamma subgroup. The HLA-DRB1*13, HLA-DQA1*01 and HLA-DQB1*06 alleles were overrepresented in the negative subgroup. Significant signals from variations in class I molecules were detected in the subgroups dominated by anti-Mi2, anti-Jo1/Ro52, anti-TIF1 gamma, and the negative subgroup. Interpretation Distinct HLA class II and I associations were observed for almost all autoantibody-defined subgroups. The associations support autoantibody profiles use for classifying IIM which would likely reflect underlying pathogenic mechanisms better than classifications based on clinical symptoms and/or histopathological features.   • Population-level effects of human papillomavirus vaccination programs on infections with nonvaccine genotypes

    • We analyzed human papillomavirus (HPV) prevalences during prevaccination and postvaccination periods to consider possible changes in nonvaccine HPV genotypes after introduction of vaccines that confer protection against 2 high-risk types, HPV16 and HPV18. Our meta-analysis included 9 studies with data for 13,886 girls and women ≤19 years of age and 23,340 women 20–24 years of age. We found evidence of cross-protection for HPV31 among the younger age group after vaccine introduction but little evidence for reductions of HPV33 and HPV45. For the group this same age group, we also found slight increases in 2 nonvaccine high-risk HPV types (HPV39 and HPV52) and in 2 possible high-risk types (HPV53 and HPV73). However, results between age groups and vaccines used were inconsistent, and the increases had possible alternative explanations; consequently, these data provided no clear evidence for type replacement. Continued monitoring of these HPV genotypes is important.

  • Turbulent Structure of a Simplified Urban Fluid Flow Studied Through Stereoscopic Particle Image Velocimetry

    • Stereoscopic particle image velocimetry was used to provide a three-dimensional characterization of the flow around a simplified urban model defined by a 5 by 7 array of blocks, forming four parallel streets, perpendicular to the incoming wind direction corresponding to a zero angle of incidence. Channeling of the flow through the array under consideration was observed, and its effect increased as the incoming wind direction, or angle of incidence (AOI), was changed from to , , and . The flow between blocks can be divided into two regions: a region of low turbulence kinetic energy (TKE) levels close to the leeward side of the upstream block, and a high TKE area close to the downstream block. The centre of the arch vortex is located in the low TKE area, and two regions of large streamwise velocity fluctuation bound the vortex in the spanwise direction. Moreover, a region of large spanwise velocity fluctuation on the downstream block is found between the vortex legs. Our results indicate that the reorientation of the arch vortex at increasing AOI is produced by the displacement of the different TKE regions and their interaction with the shear layers on the sides and top of the upstream and downstream blocks, respectively. There is also a close connection between the turbulent structure between the blocks and the wind gusts. The correlations among gust components were also studied, and it was found that in the near-wall region of the street the correlations between the streamwise and spanwise gusts were dominant for all four AOI cases. At higher wall-normal positions in the array, the correlation decreased with increasing AOI, whereas the coefficient increased as AOI increased, and at all three correlations exhibited relatively high values of around 0.4.

  • HLA and KIR Associations of Cervical Neoplasia

    • Background: Cervical cancer is the fourth most common cancer in women, and we recently reported human leukocyte antigen (HLA) alleles showing strong associations with cervical neoplasia risk and protection. HLA ligands are recognised by killer immunoglobulin-like receptors (KIRs) expressed on a range of immune cell subsets, governing their proinflammatory activity. We hypothesized that the inheritance of particular HLA-KIR combinations would increase cervical neoplasia risk.Methods: Here, we used HLA and KIR dosages imputed from SNP genotype data from 2,143 cervical neoplasia cases and 13,858 healthy controls of European decent.Results: Four novel HLA alleles were identified in association with cervical neoplasia: HLA-DRB3*9901 (OR=1.24, P=2.49×10-9), HLA-DRB5*0101 (OR=1.29, P=2.26×10-8), HLA-DRB5*9901 (OR=0.77, P=1.90×10-9) and HLA-DRB3*0301 (OR=0.63, P=4.06×10-5), due to their linkage disequilibrium with known cervical neoplasia-associated HLA-DRB1 alleles. We also found homozygosity of HLA-C1 group alleles is a protective factor for HPV16-related cervical neoplasia (C1/C1, OR=0.79, P=0.005). This protective association was restricted to carriers of either KIR2DL2 (OR=0.67, P=0.00045) or KIR2DS2 (OR=0.69, P=0.0006).Conclusions: Our findings suggest that HLA-C1 group alleles play a role in protecting against HPV16-related cervical neoplasia, mainly through a KIR-mediated mechanism.

    • The microbial community of 21 full-scale biogas reactors was examined using 454 pyrosequencing of 16S rRNA gene sequences. These reactors included seven (six mesophilic and one thermophilic) digesting sewage sludge (SS) and 14 (ten mesophilic and four thermophilic) codigesting (CD) various combinations of wastes from slaughterhouses, restaurants, households, etc. The pyrosequencing generated more than 160 000 sequences representing 11 phyla, 23 classes, and 95 genera of Bacteria and Archaea. The bacterial community was always both more abundant and more diverse than the archaeal community. At the phylum level, the foremost populations in the SS reactors included Actinobacteria, Proteobacteria, Chloroflexi, Spirochetes, and Euryarchaeota, while Firmicutes was the most prevalent in the CD reactors. The main bacterial class in all reactors was Clostridia. Acetoclastic methanogens were detected in the SS, but not in the CD reactors. Their absence suggests that methane formation from acetate takes place mainly via syntrophic acetate oxidation in the CD reactors. A principal component analysis of the communities at genus level revealed three clusters: SS reactors, mesophilic CD reactors (including one thermophilic CD and one SS), and thermophilic CD reactors. Thus, the microbial composition was mainly governed by the substrate differences and the process temperature.

    • Codigesting fats, oils, and greases with municipal wastewater sludge can greatly improve biomethanerecovery at wastewater treatment facilities. Process loading rates of fats, oils, and greases have beenpreviously tested with little knowledge of the digester microbial community structure, and high transientfat loadings have led to long chain fatty acid (LCFA) accumulation and digester upsets. This studyutilized recently-developed quantitative PCR assays for syntrophic LCFA-degrading bacteria along with16S amplicon sequencing to relate changes in microbial community structure to LCFA accumulationduring transient loading increases to an anaerobic codigester receiving waste restaurant oil andmunicipal wastewater sludge. The 16S rRNA gene concentration of the syntrophic b-oxidizing genusSyntrophomonas increased to ~15% of the Bacteria community in the codigester, but stayed below 3% inthe control digester that was fed only wastewater sludge. Methanosaeta and Methanospirillum were thedominant methanogenic genera enriched in the codigester, and together comprised over 80% of theArchaea community by the end of the experimental period. Constrained ordination showed that changesin the codigester Bacteria and Archaea community structures were related to measures of digester performance.Notably, the effluent LCFA concentration in the codigester was positively correlated to thespecific loading rate of waste oil normalized to the Syntrophomonas 16S rRNA concentration. Specificloading rates of 0e1.5 1012 g VS oil/16S gene copies-day resulted in LCFA concentrations below 30 mg/g TS, whereas LCFA accumulated up to 104 mg/g TS at higher transient loading rates. Based on thecommunity-dependent loading limitations found, enhanced biomethane production from high loadingsof fats, oils and greases can be achieved by promoting a higher biomass of slow-growing syntrophicconsortia, such as with longer digester solids retention times. This work also demonstrates the potentialfor controlling the loading rate of fats, oils, and greases based on the analysis of the codigester communitystructure, such as with quantitative PCR measurements of syntrophic LCFA-degrading bacteriaabundance.
