    • PURPOSE: Whether coffee modulates response to endocrine therapy in breast cancer patients is currently unknown. The CYP1A2 and CYP2C8 enzymes contribute to tamoxifen and caffeine metabolism. The purpose was to investigate the impact of coffee consumption on tumor characteristics and risk for early events in relation to breast cancer treatment and CYP1A2 and CYP2C8 genotypes. METHODS: Questionnaires regarding lifestyle were completed preoperatively by 634 patients in southern Sweden. CYP1A2*1F and CYP2C8*3 were genotyped. Clinical data and tumor characteristics were obtained from patients' charts, population registries, and pathology reports. Coffee consumption was categorized as low (0-1 cups/day), moderate (2-4 cups/day), or high (5+ cups/day). RESULTS: The proportion of estrogen receptor negative (ER-) tumors increased with increasing coffee consumption (p (trend) = 0.042). Moderate to high consumption was associated with lower frequency of discordant receptor status (ER + PgR-) OR 0.38 (0.23-0.63) compared to low consumption. Median follow-up time was 4.92 (IQR 3.01-6.42) years. Tamoxifen-treated patients with ER+ tumors (n = 310) who consumed two or more cups/day had significantly decreased risk for early events compared to patients with low consumption, adjusted HR 0.40 (0.19-0.83). Low consumption combined with at least one CYP1A2*1F C-allele (n = 35) or CYP2C8*3 (n = 13) was associated with a high risk for early events in tamoxifen-treated patients compared to other tamoxifen-treated patients, adjusted HRs 3.49 (1.54-7.91) and 6.15 (2.46-15.36), respectively. CONCLUSION: Moderate to high coffee consumption was associated with significantly decreased risk for early events in tamoxifen-treated patients and modified hormone receptor status. If confirmed, new recommendations regarding coffee consumption during tamoxifen treatment may be warranted.

    • De mångfacetterade krav som idag ställs på yrkesverksamma högskole- och civilingenjörer handlar inte enbart om rena ämnesmässiga kunskaper och förmågor, utan även om t.ex. erfarenhet och förmåga att delta i och leda projekt samt (kopplat till detta) kommunikativ skicklighet (såväl muntlig som skriftlig). En annan uppgift som yrkesverksamma ingenjörer måste kunna bemästra i sin profession är att (individuellt, eller som medlemmar i olika typer av styrgrupper) bedöma och utvärdera andras ingenjörsmässiga arbete, en oerhört central uppgift i ett projekt- och kvalitetssäkringsperspektiv.Det övergripande syftet med arbetet har varit att studera hur kamratbedömning kan implementeras i redan existerande kurser, för att på så sätt ge de studerande möjlighet att träna på att ge och ta emot kritisk bedömning, att förbättra de studerandes lärande via ett större aktivt engagemang under kursen samt att se om potential finns att effektivisera undervisningen m.a.p. lärarinsatsen, genom att lyfta över en del av enklare rutinkontroller till de studerande själva.Kamratbedömning har implementerats i två sinsemellan likartade kurser inom beräkningsmekanik vid högskole- resp. civilingenjörsprogrammen i Maskinteknik, LiTH (600 studenter ingick i studien). Mer specifikt har till två enklare inledande laborationsuppgifter inkluderats ett moment av kamratbedömning. Utvärderingen av det genomförda arbetet har skett via en kvalitativ studie, där de studerandes perspektiv/synpunkter inhämtats i samband med skriftlig redovisning av inlämningsuppgifter. Utöver detta har den kursansvarige gjort observationer av hur kamratutvärderingsarbetet fortskridit under laborationstid.Resultatet visar att de studerande har uppskattat granskningen av såväl det egna som kamraternas arbeten, och att de sett det som ett led i lärandeprocessen. Även professionsperspektivet har lyfts fram där de studerande betonar vikten av moment och uppgifter i utbildningen som har en direkt relevans i det kommande yrkeslivet. Den kursansvariges bedömning är vidare att andelen ”viktiga” och ”relevanta” frågor ökat, vilket ger indikationer på att kamratbedömning kan vara en såväl pedagogiskt som effektivitetsmässigt gynnsam metodik. De studerande har dock upplevt logistiken/administrationen kring kamratvärderingen som i vissa avseenden besvärlig.Sammanfattningsvis är det vår slutsats att kamratbedömning har en stor potential att berika utbildningar m.a.p. förmåga till kritisk granskning, djupinlärning och effektivisering, men att verksamheten inte bara skall implementeras kursvis (med risk för suboptimering och ineffektivitet), utan att den bör inlemmas i ett större perspektiv, med progression genom utbildningen som riktmärke.

    • The purpose of the dissertation is to study, describe and analyses which comparative measurement activities preschool children construct and use, and how they solve problems and communicate when they use the comparative measurement activities whilst playing with Lego (“the Lego play”). The measurement activities chosen are length/height, quantity and time.The empirical material is based on data from two preschool classes with children aged 2 – 5 year, It was collected through participant observation (video captures) of the children’s Lego play. The theoretical starting points in this study are grounded in childhood sociology and the sociocultural perspective. The study assumes the childhood sociology perspective by viewing the children as competent and active in creating meaning as well as controlling and influencing their own and others’ social environment. The sociocultural perspective gives prominence to development and learning, and its related tools and concepts are used to analyses the results of the study. That is, the Lego play is studied in a social context from the child’s perspective, and the sociocultural perspective describes and analyses the child’s use of mathematics and the acquisition of knowledge in the Lego play in a sociocultural context.The results show that children measuring length/height and quantity explored a store of measurement tools in order to make comparisons, and adapted these to the context in question. These were own body, other body, artefacts, numbers and counting. The measurements were used individually and with others, and the solving of the own or shared problems constituted a large share of the time spent constructing models during Lego play. By contrast, the time concept was used mainly as a tool when the children played with their finished Lego models. Thus, a time perspective was added to the child’s finished model, which inspired thoughts and reflections about time used in the Lego play. The children used the time concepts of the present, the past and the future, and also considered the concept of velocity in the context of the timescale. The children’s communication had a large impact on the Lego play, and they expressed their ideas verbally, physically and through action. The children’s use of mathematics was prominent and meaningful during the Lego play.

    • The distribution of sulphorhodamine B (SRB), a fluorescent hydrophilic model drug, was investigated in human skin after passive diffusion using four different topical delivery systems. The delivery vehicles applied were two bicontinuous lipid cubic systems, a commercial ointment and water. The lipid cubic systems consisted of either monoolein (MO) or phytantriol (PT) and water. The formulations were applied on full-thickness human skin during 24 h. Thereafter the samples were investigated using two-photon microscopy (TPM). The TPM system consisted of an inverted microscope with a 40× water-immersion objective, laser scan-box, and a pulsed femtosecond titanium:sapphire laser operating at 780 nm. The fluorescence was detected using a 560 nm long-pass filter. Sequential optical sectioning was performed, resulting in images obtained at different tissue depths. TPM revealed that SRB mainly penetrates the skin via the intercellular lipid matrix. Samples exposed to the cubic phases showed a higher accumulation of SRB in micro-fissures, from which a fluorescent network of threadlike structures spread laterally in the tissue. These structures were also detected in some of the ointment samples, but not as frequent. The penetration of SRB into the stratum granulosum was deduced from the fluorescence of SRB present inside polygonal keratinocytes with cell nuclei. Higher SRB fluorescence was obtained in the outermost layer of the epidermis using the bicontinuous cubic phases, compared to when using the reference formulations. Thus, our results suggest that the dominating delivery route using the cubic phases is via micro-fissures caused by microscopic clustering of the keratinocytes in the skin. From these micro--fissures hydrophilic compounds, here modeled by SRB, can diffuse into the surrounding intercellular lipid matrix acting like a source for sustained release.

    • Amyloid precursor protein (APP) and its cleavage product amyloid beta (A beta) have been thoroughly studied in Alzheimer's disease. However, APP also appears to be important for neuronal development. Differentiation of induced pluripotent stem cells (iPSCs) towards cortical neurons enables in vitro mechanistic studies on human neuronal development. Here, we investigated expression and proteolytic processing of APP during differentiation of human iPSCs towards cortical neurons over a 100-day period. APP expression remained stable during neuronal differentiation, whereas APP processing changed. alpha-Cleaved soluble APP (sAPP alpha) was secreted early during differentiation, from neuronal progenitors, while beta-cleaved soluble APP (sAPP beta) was first secreted after deep-layer neurons had formed. Short A beta peptides, including A beta 1-15/16, peaked during the progenitor stage, while processing shifted towards longer peptides, such as A beta 1-40/42, when post-mitotic neurons appeared. This indicates that APP processing is regulated throughout differentiation of cortical neurons and that amyloidogenic APP processing, as reflected by A beta 1-40/42, is associated with mature neuronal phenotypes.

    • The notion of teaching is theoretically conceptualized in a manner relevant to the context of preschool. This concept is grounded in evolutionary and a pedagogical-psychological theory, with reference to Barnett, Rommetveit, Tomasello, and Vygotsky. The basis of teaching is found in people’s tendencies to make known to others what they themselves have seen/realized. Institutions such as preschool and school are understood as societal means for maintaining accumulated experience with the growing generation, something that puts teaching to the foreground. Empirical examples of language teaching in contemporary preschool are given and analyzed. Some important features of the concept of teaching as here theorized and exemplified are responsivity, language practices, the tension between intersub- jectivity and alterity, the variation of children’s experience, play and playfulness. The importance of basing theoretical accounts of teaching as a preschool activity on empirical research is emphasized.

    • I artikeln teoretiseras ett undervisningsbegrepp relevant för förskolans verksamhet. Teoretiskt grundas detta begreppsliggörande i såväl ett evolutionärt som i ett pedagogiskt-psykologiskt perspektiv. Några centrala teoretiker vi bygger på är Barnett, Rommetveit, Tomasello och Vygotskij. Vi lokaliserar grunden till undervisning i människors tendens att göra sådant de själva sett/insett synligt också för andra. Institutioner såsom förskolan och skolan förstås här som sätt för samhället att främja bevarandet av ackumulerande erfarenheter hos den växande generationen, något som gör undervisning centralt. Vi ger empiriska exempel på hur språkundervisning kan gestalta sig i dagens förskola; aktiviteter som vi analyserar utifrån de teoretiska begrepp vi introducerar. Några av de begrepp vi skriver fram för att teoretisera undervisning mer allmänt och språkundervisning mer specifikt i förskolan är responsivitet, kommunikativa praktiker, spänningsfältet mellan intersubjektivitet och alteritet, polyfoni och barns skilda erfarenheter, samt lek och lekfullhet. Vikten av att teoretisera undervisning i förskolan utifrån empirisk forskning betonas.

    • The prognostic importance of tumor-specific nuclear insulin receptor (InsR) expression in breast cancer is unclear, while membrane and cytoplasmic localization of InsR is better characterized. The insulin signaling network is influenced by obesity and may interact with the estrogen receptor a (ERα) signaling. The purpose was to investigate the interplay between nuclear InsR, ER, body mass index (BMI), and prognosis. Tumor-specific expression of nuclear InsR was evaluated by immunohistochemistry in tissue microarrays from 900 patients with primary invasive breast cancer without preoperative treatment, included in a population-based cohort in Sweden (2002-2012) in relation to prognosis. Patients were followed for up to 11 years during which 107 recurrences were observed. Nuclear InsR+ expression was present in 214 patients (23.8%) and increased with longer time between surgery and staining (P < 0.001). There were significant effect modifications by ER status and BMI in relation to clinical outcomes. Nuclear InsR+ conferred higher recurrence-risk in patients with ER+ tumors, but lower risk in patients with ER- tumors (Pinteraction = 0.003). Normal-weight patients with nuclear InsR+ tumors had higher recurrence-risk, while overweight or obese patients had half the recurrence-risk compared to patients with nuclear InsR- tumors (Pinteraction = 0.007). Normal-weight patients with a nuclear InsR-/ER+ tumor had the lowest risk for recurrence compared to all other nuclear InsR/ER combinations [HRadj 0.50, 95% confidence interval (CI): 0.25-0.97], while overweight or obese patients with nuclear InsR-/ER- tumors had the worst prognosis (HRadj 7.75, 95% CI: 2.04-29.48). Nuclear InsR was more prognostic than ER among chemotherapy-treated patients. In summary, nuclear InsR may have prognostic impact among normal-weight patients with ER+ tumors and in overweight or obese patients with ER- tumors. Normal-weight patients with nuclear InsR-/ER+ tumors may benefit from less treatment than normal-weight patients with other nuclear InsR/ER combinations. Overweight or obese patients with nuclear InsR-/ER- tumors may benefit from more tailored treatment or weight management.

    • Coffee is associated with decreased breast cancer risk, but the impact of body mass index (BMI) in combination with coffee consumption on prognosis is unclear. The suppressive effect of coffee constituents on the insulin-like growth factor receptor 1 (IGF1R) levels in breast cancer cells may play a role. The aim was to investigate the prognostic impact of coffee consumption and possible associations with tumor-specific IGF1R protein expression and BMI in a population-based cohort in Sweden, comprising 1,014 primary breast cancer patients without pretreatment enrolled 2002-2012 and followed for up to 13 years. Patients with higher coffee consumption had lower tumor IGF1R levels (P = 0.025), but only among the normal-weight patients (P = 0.005). Coffee did not impact the recurrence-risk overall. However, tamoxifen-treated patients with ER+ tumors drinking ≥ 2 cups of coffee/day had lower recurrence-risk [adjusted HR (HRadj) 0.57, 95% CI, 0.34-0.97] compared with patients with lower intake, although only among normal-weight patients (HRadj 0.37, 95% CI: 0.17-0.78; Pinteraction = 0.039). Similarly, coffee consumption ≥ 2 cups/day was associated with significantly lower recurrence-risk among the 640 radiotherapy-treated patients irrespective of BMI (HRadj 0.59, 95% CI 0.36-0.98) and in the 296 normal-weight patients (HRadj 0.36, 95% CI 0.17-0.76) but not in the 329 overweight or obese patients (HRadj 0.88, 95% CI 0.42-1.82) although the interaction was not significant (Pinteraction = 0.093). In conclusion, coffee consumption was negatively associated with tumor-specific IGF1R levels only among normal-weight patients. Though, IGF1R did not explain the association between coffee intake and improved prognosis among normal-weight tamoxifen- or radiotherapy-treated patients. Studies of IGF1R-targeting therapies may benefit from taking BMI and coffee consumption into account.

    • Clinical trials examining insulin-like growth factor-I receptor (IGF1R)-targeting strategies have emphasized that better predictive biomarkers are required to improve patient selection. Immunohistochemical tumor-specific protein expression of IGF1R, insulin receptor (InsR), and phosphorylated IGF1R/InsR (pIGF1R/InsR) individually and combined in relation to breast cancer prognosis was evaluated in a populationbased cohort of 1,026 primary invasive breast cancer patients without preoperative treatment diagnosed in Sweden. IGF1R (n = 923), InsR (n = 900), and pIGF1R/InsR (n = 904) combined cytoplasmic and membrane staining was dichotomized. IGF1Rstrong/InsRmod/strong/pIGF1R/InsRpos tumors were borderline associated with 2-fold risk for events, HRadj (2.00; 95%CI 0.96-4.18). Combined IGF1R and pIGF1R/InsR status only impacted prognosis in patients with InsRmod/strong expressing tumors (Pinteraction = 0.041). IGF1Rstrong expression impacted endocrine treatment response differently depending on patients' age and type of endocrine therapy. Phospho-IGF1R/InsRpos was associated with lower risk for events among non-endocrine-treated patients irrespective of ER status, HRadj (0.32; 95%CI 0.16-0.63), but not among endocrinetreated patients (Pinteraction = 0.024). In non-endocrine-treated patients, pIGF1R/InsRpos was associated with lower risk for events after radiotherapy, HRadj (0.31; 95%CI 0.12-0.80), and chemotherapy, HRadj (0.29; 95%CI 0.09-0.99). This study highlights the complexity of IGF hetero-and homodimer signaling network and its interplay with endocrine treatment, suggesting that combinations of involved factors may improve patient selection for IGF1R-targeted therapy.

  • Comparison of Skimmed Milk and Lanthanum Flocculation for Concentration of Pathogenic Viruses in Water

    • Concentration of viruses in water is necessary for detection and quantification of the viruses present, in order to evaluate microbiological barriers in water treatment plants and detect pathogenic viruses during waterborne outbreaks, but there is currently no standardised procedure. In this study, we implemented a previously described fast and simple lanthanum-based protocol for concentration of norovirus genogroup I (GI), genogroup II (GII) and hepatitis A virus (HAV) in drinking and surface water. We compared the results with those of a widely used skimmed milk flocculation method, followed by nucleic acid extraction and RT-qPCR detection. Three seeding levels, with intended concentrations 5 × 103, 5 × 104 and 5 × 105 genome copies/10 L, were added to drinking water or surface water. All seed levels were detected with both flocculation methods. Samples extracted with skimmed milk flocculation had on average 1.82, 1.86 and 1.38 times higher measured concentration of norovirus GI, GII and HAV, respectively, than those extracted with lanthanum flocculation, across all seeding levels and water types tested. Mengovirus was used as a positive process control. Mengovirus recovery was higher for skimmed milk (40.7% in drinking water, 26.0% in surface water) than for lanthanum flocculation (24.4% in drinking water, 9.7% in surface water). Together, this indicates that skimmed milk flocculation provides higher viral recovery than lanthanum flocculation. However, lanthanum-based flocculation can be performed much faster than skimmed milk flocculation (1.5 h versus 16 h flocculation time) and thus could be a good alternative for rapid monitoring of viruses in water.

    • In recent decades, refugee immigration has had significant impact on educational contexts in Sweden, with preschools the primary arenas for young childrens language learning experiences. The present study examines second language and literacy training practices for immigrant children (aged 1-5) in preschools in Sweden. The empirical data consist of video recordings of teacher-guided play activities. These were designed to create rich linguistic and cultural environments facilitating active and democratic participation by the children. Guided play activities were developed in close collaboration between teachers and researchers during action-based interventions that were aimed at constructing child-oriented participatory language learning practices in ethnically and linguistically diverse ECEC settings. The childrens first languages were Tigrinya, Arabic dialects, Somalian, Kurdish dialects, Russian, Spanish and Swedish. The participatory learning potential of guided play was collaboratively analyzed and assessed, and new and revised activities were implemented, based on the goals of inclusivity and childrens active engagement in play and language learning. The analysis shows that teachers use of multimodal semiotic means, such as questions, texts and cultural artifacts (stories, material story-related objects, play spaces) were important strategies for the preparation of appealing play environments and activities that contributed to childrens curiosity and participation. These resources served as affordances for guided play, allowing the simultaneous scaffolding of childrens play competencies and their language learning. Au cours des dernieres decennies, limmigration de refugies a eu un impact significatif dans le domaine de leducation en Suede, en commencant par les ecoles maternelles qui sont les premiers lieux dapprentissage de la langue pour les jeunes enfants. Cette etude examine les pratiques dalphabetisation et dapprentissage du suedois deuxieme langue, qui sont utilisees dans les ecoles maternelles en Suede pour les enfants immigrants ages de 1 a 5 ans. Les donnees empiriques rassemblees consistent en des enregistrements video dactivites ludiques guidees par un enseignant. Ces activites ont ete concues pour creer des environnements linguistiques et culturels riches facilitant la participation active et democratique des enfants. Elles ont ete developpees dans le cadre dune etroite collaboration entre enseignants et chercheurs au cours dinterventions concretes visant a elaborer des pratiques participatives dapprentissage de la langue prenant en compte la perspective de lenfant dans le contexte dune petite enfance ethniquement et linguistiquement diversifiee. Les langues maternelles des enfants etaient le tigrinya, differents dialectes arabes, le somali, differents dialectes kurdes, le russe, lespagnol et le suedois. Le potentiel dapprentissage participatif de chaque jeu guide a ete analyse et evalue collectivement et de nouvelles activites revisees ont ete mises en place, sur la base des objectifs dinclusivite et dengagement actif des enfants dans le jeu et dans lapprentissage de la langue. Lanalyse montre que lutilisation par les enseignants de moyens semiotiques multimodaux, tels que des questions, des textes et des artefacts culturels (histoires, objets concrets lies a lhistoire, espaces de jeu) constituaient des strategies importantes pour preparer un environnement de jeu et des activites susceptibles dattirer les enfants et de stimuler leur participation et leur curiosite. Ces ressources ont servi daffordances a la realisation de jeux guides, permettant aux enfants de combiner competences ludiques et apprentissage de la langue. En las ultimas decadas, la inmigracion de refugiados ha tenido un impacto significativo en los contextos educativos de Suecia, y los centros de ensenanza preescolar han sido los principales escenarios para las experiencias de aprendizaje de idiomas de los ninos de corta edad. El presente estudio investiga las practicas de aprendizaje de una segunda lengua y de alfabetizacion en ninos inmigrantes (de 1 a 5 anos) en centros de ensenanza preescolar de Suecia. Los datos empiricos consisten en grabaciones de video de actividades ludicas guiadas por el maestro. Estas se disenaron para crear entornos linguisticos y culturales ricos que facilitaran la participacion activa y democratica de los ninos. Las actividades ludicas guiadas se elaboraron en estrecha colaboracion entre maestros e investigadores durante intervenciones basadas en la accion que tenian como objetivo construir practicas participativas de aprendizaje de idiomas orientadas a los ninos en entornos de educacion infantil etnica y linguisticamente diversos. Las lenguas maternas de los ninos eran el tigrina, dialectos arabes, el somali, dialectos kurdos, el ruso, el espanol y el sueco. El potencial de aprendizaje participativo del juego guiado se analizo y evaluo de manera colaborativa, y se llevaron a cabo actividades nuevas y revisadas, basadas en los objetivos de inclusividad y participacion activa de los ninos en el juego y el aprendizaje de idiomas. El analisis muestra que el uso por parte de los maestros de medios semioticos multimodales, tales como preguntas, textos y artefactos culturales (historias, objetos materiales relacionados con las historias, espacios ludicos) fueron estrategias importantes para la preparacion de entornos ludicos atractivos y actividades que contribuyeron a la curiosidad y la participacion de los ninos. Estos recursos sirvieron como formas de abordaje o << affordances >> (nota del traductor: << affordance >> es un neologismo utilizado para definir la << capacidad de un objeto de sugerir su propia utilizacion >>) para el juego guiado, lo que permite el andamiaje simultaneo de las competencias ludicas de los ninos y su aprendizaje del lenguaje.

    • PURPOSE: Isoform-specific tumor estrogen receptor β (ERβ) expression may hold prognostic information in breast cancer, especially among endocrine-treated breast cancer patients. The study's purpose was to evaluate ERβ isoform 1 (ERβ1) expression in relation to tumor characteristics, ESR2 genotypes, and prognosis in different treatment groups.EXPERIMENTAL DESIGN: A population-based prospective cohort of 1,026 patients diagnosed with primary invasive breast cancer in Lund, Sweden, between October 2002 and June 2012 was followed until June 2014 (median 5 years). Associations between immunohistochemical ERβ1 expression, patient and tumor characteristics, as well as outcome within treatment groups were analyzed.RESULTS: Tumor ERβ1 expression was available for 911 patients (89%) and was not associated with ESR2 genotypes. ERβ1 positivity, defined as >75% (ERβ175(+), 72.7%), was positively associated with established favorable tumor characteristics. Overall, ERβ175(+) was associated with lower risk of breast cancer events [HRadj = 0.60; 95% confidence interval (CI), 0.41-0.89]. The magnitude of the association was larger in patients with ERα(-) tumors (HRadj = 0.30; 95% CI, 0.12-0.76), compared with ERα(+) tumors (HRadj = 0.66; 95% CI, 0.42-1.03). Among the 232 chemotherapy-treated patients, ERβ175(+) tumors were associated with lower risk of breast cancer events compared with ERβ175(-) tumors (HRadj = 0.31; 95% CI, 0.15-0.64). Among the 671 chemonaïve patients, ERβ175 status was not associated with the outcome.CONCLUSION: High ERβ1 expression was a favorable prognostic marker in this breast cancer cohort, especially in chemotherapy-treated patients, but not in endocrine therapy-treated patients.     • The objective of the present study is to produce knowledge about how children, things, documents, computers, teachers, templates and other entities take part in and produce documentation in preschool systematic quality development work. The thesis asks questions about how and what is produced as quality in preschool documentation. Through a posthumanist agential realist approach, the study engages empirically in video recordings of preschool documentation practices from one Swedish preschool, and in documentation from four other Swedish preschools.The results, presented in four articles, show that documentation practice is complex and entangled with a variety of entities, human and non-human, and spaces and time. The study shows how children’s participation in production of documentation does not depend on which documentation method is used. In the study, objects such as computers, images and coloured labels, together with teachers’ attitudes and agendas, have an impact on the way that children are involved in producing documentation. The results also show that the products of documentation, intra-acting with a local template, with past and present preschool traditions and policies, produce rather than represent certain elements of quality such as teachers’ actions, children’s interests and thematic work. The thesis produces knowledge about how the focus of preschool documentation shifts away from individual children and towards teachers’ activities, and creates possibilities for what preschool practices and quality may become.

    • Two-photon laser scanning fluorescence microscopy (TPM) has been shown to be advantageous for imaging optically turbid media such as human skin. The ability of performing three-dimensional imaging without presectioning of the samples makes the technique not only suitable for noninvasive diagnostics but also for studies of topical delivery of xenobiotics. Here, TPM is used as a method to visualize both autofluorescent and exogenous fluorophores in skin. Samples exposed to sulforhodamine B have been scanned from two directions to investigate attenuation effects. It is shown that optical effects play a major role. Thus, TPM is excellent for visualizing the localization and distribution of fluorophores in human skin, although quantification might be difficult. Furthermore, an image-analysis algorithm has been implemented to facilitate interpretation of TPM images of autofluorescent features of nonmelanoma skin cancer obtained ex vivo. The algorithm was designed to detect cell nuclei and currently has a sensitivity and specificity of 82% and 78% to single cell nuclei.     • Members of the SREBP family of transcription factors control cholesterol and lipid homeostasis and play important roles during adipocyte differentiation. The transcriptional activity of SREBPs is dependent on the coactivators p300 and CBP. We now present evidence that SREBPs are acetylated by the intrinsic acetyltransferase activity of p300 and CBP. In SREBP1a, the acetylated lysine residue resides in the DNA-binding domain of the protein. Coexpression with p300 dramatically increases the expression of both SREBP1a and SREBP2, and this effect is dependent on the acetyltransferase activity of p300, indicating that acetylation of SREBPs regulates their stability. Indeed, acetylation or mutation of the acetylated lysine residue in SREBP1a stabilizes the protein. We demonstrate that the acetylated residue in SREBP1a is also targeted by ubiquitination and that acetylation inhibits this process. Thus, our studies define acetylation-dependent stabilization of transcription factors as a novel mechanism for coactivators to regulate gene expression.

    • Two-photon excitation fluorescence correlation spectroscopy (TPFCS) has been applied in connection to measurements of the point spread function (PSF) for quantitative analysis of sulphorhodamine B (SRB) in excised human skin. The PSF was measured using subresolution fluorescent beads embedded in the skin specimen. The PSF, measured as full width at half maximum (FWHM) was found to be 0.41 ± 0.05 μm in the lateral direction, and 1.2 ± 0.4 μm in the axial direction. The molecular diffusion of SRB inside the skin ranged between 0.5 and 15.0 × 10 −8 cm2/s. The diffusion coefficient is not dependent on depths down to 40 μm. The fluorophores were found to accumulate on the upper layers of the skin. This work is the first TPFCS study in human skin. The results show that TPFCS can be used for quantitative analyses of fluorescent compounds in human skin.

    • There is a need for tools enabling quantitative imaging of biological tissue for pharmaceutical applications. In this study, two-photon fluorescence microscopy (TPM) has been combined with fluorescence correlation spectroscopy (FCS), demonstrating proof-of-principle providing quantitative data of fluorophore concentration and diffusion in human skin. Measurements were performed on excised skin exposed to either rhodamine B (RB) or rhodamine B isothiocyanate (RBITC), chosen based on their similarity in fluorescence yield and molecular weight, but difference in chemical reactivity. The measurements were performed at tissue depths in the range 0 and 20 pm, and the diffusion coefficients at skin depths 5 and 10 mu m were found to be significantly different (P < 0.05). Overall median values for the diffusion coefficients were found to be 4.0 x 10(-13) m(2)/s and 2.0 x 10(-13) m(2)/s for RB and RBITC, respectively. These values correspond to the diffusion of a hard sphere with a volume eight times larger for RBITC compared to RB. This indicates that the RBITC have bound to biomolecules in the skin, and the measured signal is obtained from the RBITC-biomolecule complexes, demonstrating the potential of the TPM-FCS method to track molecular interactions in an intricate biological matrix such as human skin.

    • The herpes simplex virus type 1 (HSV-1) origin binding protein, OBP, is a DNA helicase specifically stimulated by the viral single strand DNA-binding protein, ICP-8. The stimulation is dependent on direct protein-protein interactions between the C-terminal domain of OBP, delta OBP, and ICP 8 (Boehmer, P.E., Craigie, M.C., Stow, N.D., and Lehman, I.R. (1994) J. Biol. Chem. 269, 29329-29334). We have now observed that this interaction is dramatically influenced by the nature of the DNA ligand. Stable complexes between delta OBP, ICP 8, and double-stranded DNA, presented either as a specific duplex oligonucleotide or a restriction fragment containing the HSV-1 origin of replication, oriS, can be detected by gel chromatography and gel electrophoresis. In contrast, a single-stranded oligonucleotide, oligo(dT)65, will completely disrupt the complex between delta OBP and ICP 8. We therefore suggest that the interaction between delta OBP and ICP 8 serves to position the single strand DNA-binding protein with high precision onto single-stranded DNA at a replication fork or at an origin of DNA replication.

    • Breast-feeding is a known protective factor against breast cancer. Breast-feeding duration is influenced by hormone levels, milk production, and lifestyle factors. The aims were to investigate how breast-feeding duration and milk production affected tumor characteristics and risk for early breast cancer events in primary breast cancer patients. Between 2002 and 2008, 634 breast cancer patients in Lund, Sweden, took part in an ongoing prospective cohort study. Data were extracted from questionnaires, pathology reports, and patients' charts from 592 patients without preoperative treatment. Breast-feeding duration ≤12 months of the first child was associated with higher frequency of ER+/PgR+ tumors (P=0.02). Median follow-up time was 4.9 years. Higher risk for early events was observed for breast-feeding duration of first child >12 months (LogRank P=0.001), total breast-feeding duration >12 months (LogRank P=0.008), as well as 'excessive milk production' during breast-feeding of the first child (LogRank P=0.001). Patients with 'almost no milk production' had no events. In a multivariable model including both 'excessive milk production' and breast-feeding duration of the first child >12 months, both were associated with a two-fold risk for early events, adjusted HRs 2.33 (95% CI: 1.25-4.36) and 2.39 (0.97-5.85), respectively, while total breast-feeding duration was not. 'Excessive milk production' was associated with a two-fold risk of early distant metastases, adjusted HR 2.59 (1.13-5.94), but not duration. In conclusion, 'excessive milk production' during breast-feeding was associated with higher risk for early events independent of tumor characteristics, stressing the need to consider host factors in the evaluation of prognostic markers.

Syftet med konferensen var att lyfta upp begreppet omsorg, diskutera dess innebörd, rädda det från retorisk förflackning och samtidigt via dess koppling till verksamheter belysa ett vidare sammanhang. När bamforskning diskuteras har jag på senare tid funnit det väsentligt att peka på att den har en kapacitet att kasta ljus över centrala samhällsfrågor. Forskning om barn bör inte bara vara en forskning om en viss grupp i samhället. Den nyare bamdomsforskningen söker tillämpa ett bamperspektiv. Detta innebär att fenomen studeras såsom de framstår från barnens perspektiv och att inverkan av olika sociala reformer, politiska förändringar och produktion av varor på barns liv belyses. Det handlar alltså både om att söka efter barnens perspektiv och att söka efter konsekvenserna för barnen av olika samhälleliga förändringar. Nya fålt som t.ex. barndomshistoria, barndomssociologi och barndomsantropologi har uppstått och begreppet generation har införts för att markera vikten av att anlägga ett generationsperspektiv på frågor som vanligtvis grupperats på annat sätt.

    • En stor andel svenska barn mellan sex och nio år gamla går efter skolan till ”fritids”. Fritidshem kännetecknas av både utbildning och omsorg, och därmed av både formella och informella aktiviteter. På senare år har verksamheten blivit mer och mer influerad av skolan och numera finns ett eget kapitel för fritidshem i den nationella läroplanen för grundskolan. Fritidshemmen genomgår därför en förändring som kommer att medföra att mer formella aktiviteter införs, exempelvis inom teknik, samtidigt som praktiskt taget ingen forskning har gjorts på detta. Syftet med det här kapitlet är att presentera några resultat från en pågående forskningsstudie om teknikens roll i aktiviteter på fritidshem. Observationerna fokuserade på de dagliga aktiviteterna på fritidshemmen där teknik hade en central roll. Vi använder begreppet gränsobjekt för att analysera teknik i aktiviteterna. Resultaten visar att det är en speciell sorts teknikundervisning som uppstår i fritidshem, på gränsen mellan den informella och formella verksamheten: från det informella lekrelaterade byggandet med Lego och träklotsar till den mer formella datorundervisningen. I dessa aktiviteter finns ett tydligt fritidsinslag, framför allt i form av ett fritt val av teknik och vad man vill lära sig. Det faktum att barn kan välja fritt pekar inte bara på att teknik i fritidshem är ett gränsobjekt med en stor tolkningsflexibilitet, utan också att teknikundervisning i fritidshem skulle kunna vara en lustfylld och effektiv väg till teknisk allmänbildning.

    • It has long been speculated that psychedelic use could provoke the onset of psychosis, but there is little evidence to support this conjecture. Using a longitudinal research design with samples representative of the US and UK adult populations with regard to sex, age, and ethnicity (n = 9732), we investigated associations between psychedelic use and change in the number of psychotic symptoms during the two-month study period. In covariate-adjusted regression models, psychedelic use during the study period was not associated with a change in the number of psychotic symptoms unless it interacted with a personal or family history of bipolar disorder, in which case the number of symptoms increased, or with a personal (but not family) history of psychotic disorders, in which case the number of symptoms decreased. Taken together, these findings indicate that psychedelic use may affect psychotic symptoms in individuals with a personal or family history of certain disorders characterized by psychotic symptomatology.

Between 2002 and 2011, 994 primary breast cancer patients without preoperative treatment were enrolled in Lund, Sweden and followed until December 2012. History of OC use was obtained from preoperative questionnaires. Tumor characteristics, clinical data, and date of death were obtained from pathology reports, patient charts, and population registries. Among the 948 patients with invasive cancer and no metastasis detected on the post-operative screen, 74 % had ever used OCs. Patients were followed for up to nine years (median follow-up 3 years), and 100 breast cancer events were recorded. Ever OC use was not associated with prognosis, irrespective of duration. However, any OC use before age 20 was associated with a threefold increased risk for breast cancer events in patients <50 years but not in patients ≥50 years (P interaction = 0.009). In patients ≥50 years with estrogen receptor positive tumors, previous OC use at any age was associated with a significantly decreased risk of breast cancer events among patients who received aromatase inhibitors compared to patients who never used OCs (adjusted HR 0.37: 95 % CI 0.15-0.87). OC use was not associated with tamoxifen-response. If confirmed, history of OC use may yield valuable prognostic and treatment predictive information in addition to currently used criteria.

    • A majority of Swedish children between six and nine years old attend afterschool activities in a leisure-time centre, fritidshem, until their parents finish work for the day. Leisure-time centres are characterised by “educare” activities, that is, both educational and carerelated activities and thus both formal and informal learning settings. The centres are part of the Swedish educational system and activities should be related to the national curriculum for the compulsory school. The centres are right now in an interesting transition which will lead to the introduction of more formal learning activities, for instance, in technology, at the same time as virtually no research has been carried out on technology activities in the centres. The aim of this paper is therefore to study the role of technology in activities in leisure-time centres. The results are drawn from field work conducted in three Swedish leisure-time centres. The observations focused upon the daily activities in the leisure centres, specifically activities where technology play a central part. The data collection was completed with interviews with teachers. We use the concept of boundary object to analyse the technological activities. The findings indicate that there is a special technology education that emerges in the context of the leisuretime center – leisure-time technology education – at the intersection between informal and formal technology learning, which ranges from the informal play-related construction of Lego and wooden blocks to the more formal computer instruction. The amorphousness of leisure-time technology – the fact that children can choose what they want it to be and what to learn –points to it being a boundary object with a very open interpretative flexibility.

    • Det övergripande syftet med denna licentiatstudie är att undersöka hur teknik som undervisningsområde kommer till uttryck inom fritidsverksamheter. Syftet inrymmer ett fokus på vad fritidslärarna gör för att organisera undervisning. Studien utgår ifrån följande frågeställningar: hur ser fritidslärarna på undervisning i teknik i fritidshem; och hur organiseras teknik som en del av undervisnings-praktiken i fritidshem? Studien är kvalitativ samt inspirerad av etnografi och Grounded theory. Studiens teoretiska perspektiv är symbolisk interaktionism, samt Mitchams modell för manifestation av teknik från 1994. Studien är en fält-studie på tre olika fritidshem. I studiens bakgrundsbeskrivning påvisas att fritidshem har haft en historia där hantverk och olika estetiska aktiviteter varit vanligt förekommande aktiviteter. Fritidshemmen har också, i och med en tydligare styrning kring lärandet, fått del i läroplanen Lgr11 (reviderad 2016), där tekniken är en framskriven del av undervisningen. Studien handlar därför om hur fritidshemspersonalen reflekterar och förhandlar och definierar för att göra olika teknikaktiviteter. Fritidshemspersonalen gör, enligt studien, här en nyorientering för att tolka innehållet i den egna läroplansdelen. I denna nyorientering använder fritidshemspersonalen sig av tre olika strategier för att hantera detta med teknik som undervisning, detta i ett gränsland mellan tidigare idétraditioner och en utökad styrning.

    •  This paper addresses the in Sweden quite new phenomena "gender sensitive pedagogy" which can mainly be found in preschool. The study leans on feminist theory, specifically theory of the construction of gender in educational settings, power relations often maintained in school settings and sociological theory of childhood. Pedagogues in preschool and advisory "gender pedagogues" have been interviewed of their opinions on gender sensitive pedagogy, what it might be, what problems it would solve and how to learn to create this "gender sensitive pedagogy". This study is a part of a three-step research project of gender sensitive pedagogy in Sweden. The first study was directed towards in Sweden very popular and very much used books concerning how to work with this kind of pedagogy, the second was aiming to find preschool workgroups views on the matter and the last one studied "advisory gender pedagogues" that are teachers or preschool teachers that have got an additional training to help preschool and schools to work for equality and to apply gender sensitive pedagogy in their work. We have conducted focus group interviews. Four were carried out with preschool work groups and two with advisory teams. Three cities in mid Sweden have been the locations for the studies. The findings show that the pedagogues regard gender sensitive pedagogy as something that affect all levels of the preschool institutions for example organisation, interaction, activities and offers to the children. A lot of reflection and training for the preschool themes are required, aiming to construct "know how" in relation to -gender sensitive pedagogy-. Parents should also be involved in the upbringing of equal and gender sensitive new citizens according to the pedagogues-.

    • The dominant approach to mainstreaming climate adaptation into sectoral policies relies on an ‘upscaling’ model in which it is envisaged to extract lessons from local change processes to inspire generic sub-national and national policies. One of the central methodological questions, which remain unanswered in climate change adaptation research, is exactly how public policy can learn from highly contextual experiences of community-based adaptation and what role should be played by case study research. In this paper we undertake a comparison between three large research projects in Sweden, Canada and Indonesia, which aim to study and/or foster local adaptation in selected case studies through a process of social learning. We present a novel framework based on mapping of ‘sense-making perspectives’, which enables analysis of the multiple ways case study research can support local climate adaptation and link such efforts to higher level public policy. The analysis demonstrates how methodological choices shape how case study research works at the interface between planned (steered/regulatory policy) and self-organised adaptation of stakeholders (non-coercive policy). In this regard, there is a need for a high degree of transparency from the research community to enable local professionals to decide on their stakes and interests when inviting researchers into their grounded activities. We conclude that case study research can achieve new significance if viewed as a platform to leverage stakeholder competencies when informing existing social structures and enable the implementation of political objectives, but equally driving the very reinvention and improvement of these institutions.

    • I den svenska läroplanen för förskolan (Skolverket, 2018) har krav på ett språkutvecklande arbete genom högläsning specificerats samtidigt som förskolans undervisningsuppdrag har förtydligats. Utifrån ett sociokulturellt perspektiv utgår denna artikel från en teoretisk premiss att förskolans professionskunskap befinner sig i en remedieringsprocess, där högläsning som professionellt yrkesutövande omförhandlas i ljuset av ett delvis nytt uppdrag. Syftet är att bidra med kunskap om hur denna remediering går till och vad den resulterar i för förståelse för högläsning i samtida förskola. Det empiriska materialet utgörs av fyra fokusgruppssamtal. Resultatet visar hur förskollärare remedierar professionell högläsning genom att kontrastera och omförhandla innebörden av, villkoren för och deltagandet i förskolans högläsning. I sina resonemang ger förskollärarna uttryck för en vidgad repertoar vad gäller högläsning där traditionella praktiker lever kvar samtidigt som nya praktiker har tillkommit och andra aspekter har blivit till figur för det professionella arbetet. Implikationerna av dessa resultat diskuteras i relation till vad som förväntas av den läsande förskolläraren och det läsande/lyssnande barnet i dessa remedierade högläsningspraktiker.

    • In breast cancer, high levels of the inflammatory cytokine interleukin-6 (IL-6) have been associated with disease-free survival and treatment resistance. Increased serum levels of IL-6 have been correlated with increased levels of NF-κβ and aromatase expression in adipose tissue. Several IL6 single nucleotide polymorphisms have been associated with breast cancer prognosis, but the impact may differ depending on tumour oestrogen receptor (ER) status. This translational study investigated the association between IL6 genotypes, ER-status, and treatment on the risk of early events among breast cancer patients.

    • The COX2 rs5277 (306G>C) polymorphism has been associated with inflammation-associated cancers. In breast cancer, tumor COX-2 expression has been associated with increased estrogen levels in estrogen receptor (ER)-positive and activated Akt-pathway in ER-negative tumors. Our study investigated the impact of COX2 genotypes on early breast cancer events and treatment response in relation to tumor ER status and body constitution. In Sweden, between 2002 and 2008, 634 primary breast cancer patients, aged 25-99 years, were included. Disease-free survival was assessed for 570 rs5277-genotyped patients. Body measurements and questionnaires were obtained preoperatively. Clinical data, patient- and tumor-characteristics were obtained from questionnaires, patients' charts, population registries and pathology reports. Minor allele(C) frequency was 16.1%. Genotype was not linked to COX-2 tumor expression. Median follow-up was 5.1 years. G/G genotype was not associated with early events in patients with ER-positive tumors, adjusted HR 0.77 (0.46-1.29), but conferred an over 4-fold increased risk in patients with ER-negative tumors, adjusted HR 4.41 (1.21-16.02)(pinteraction = 0.015). Chemotherapy-treated G/G-carriers with a breast volume ≥850 ml had an increased risk of early events irrespective of ER status, adjusted HR 8.99 (1.14-70.89). Endocrine-treated C-allele carriers with ER-positive tumors and a breast volume ≥850 ml had increased risk of early events, adjusted HR 2.30 (1.12-4.75). COX2 genotype, body constitution and ER status had a combined effect on the risk of early events and treatment response. The high risk for early events in certain subgroups of patients suggests that COX2 genotype in combination with body measurements may identify patients in need of more personalized treatment.
