    • Malignant salivary gland tumors make up a small proportion of malignancies worldwide, yet vary widely in terms of histology, patterns of spread, and recurrence. A better understanding of this variability will guide appropriate treatment recommendations and lead to improved outcomes. Recent molecular genetic studies have uncovered a translocation-generated gene fusion network in salivary gland carcinomas that can be used for diagnosis, treatment decisions, and development of specific targeted therapies. The gene fusions encode novel fusion oncoproteins that function as transcriptional coactivators, tyrosine kinase receptors, and transcription factors involved in growth-factor signaling and cell-cycle regulation. While surgery currently is the primary therapy for operable tumors, radiation plays an important role in the postoperative setting, as well as in the definitive setting for inoperable lesions. An awareness of the risk factors for tumor recurrence and spread is important for both adjuvant therapy referrals and for radiation treatment planning purposes. Additionally, chemotherapy is being used increasingly in both the concurrent setting as a radiosensitizer, as well as in the palliative setting for metastatic tumors. Future trials investigating concurrent chemotherapy and radiation, as well as the use of targeted agents based on evolving molecular discoveries, will elucidate optimal personalized approaches for this challenging disease.

  • A comparative study on the degradation of cotton linters induced by carbonate and hydroxyl radicals generated from peroxynitrite

    • Carbonate (CO3.(-)) and hydroxyl (HO.) radicals were chemically produced in cotton linter suspensions using peroxynitrite as a radical precursor. Both radicals could degrade cotton linters, as shown by viscosity and GPC-SEC measurements. As evidenced by the viscosity measurements, the presence of oxygen during the cotton linter treatments slightly increased cellulose degradation by both radicals. For the carbonate radical, more than 90% of the viscosity losses could be recovered by reductive NaBH4 treatment before measuring the viscosity, whereas only approximately 40% of the viscosity was recovered after hydroxyl radical degradation and subsequent NaBH4 treatment. This indicates that carbonate radicals mainly abstract H-atoms adjacent to hydroxyl groups, i.e., at C-2, C-3 and C-6. This intramolecular selectivity may reflect a polar effect, whereby hydrogen atom abstractions from these positions are favoured. In addition, abstraction at C-6 would be sterically and statistically favoured.

    • The mechanism by which the carbonate radical anion reacts with D-glucose in alkaline aqueous solutions has been studied by means of gamma-radiolysis. From the product analysis it is concluded that the reaction sequence is initiated by a one-electron transfer between the carbonate radical anion and deprotonated D-glucose. In the presence of molecular oxygen, the major, if not only products of this reaction sequence are formic acid, arabinose and gluconic acid and reaction schemes are proposed to account for the observed formation of these products.

  • Use of amended tailings as mine waste cover

    • The present study addresses the effectiveness of green liquor dregs (GLD, a residue from paper pulp-making) as a paste additive with different proportions in tailings for stabilizing mine waste by testing of uniaxial compressive strength and hydraulic conductivity. Selected samples were also investigated for freeze–thaw effect on hydraulic conductivity, and the water retention capacity was discussed based on previous results. Fly ash was also added to the paste to study the auxiliary functions such as solidification in the sealing material. GLD have the potential for use as a barrier layer material for stabilizing mine tailings by decreasing water percolation and improving water retention properties. However, the solidified paste of GLD-amended tailings possesses low uniaxial compressive strength, but the addition of fly ash to the paste increased the uniaxial compressive strength by up to 2–3 times 1 month after it was solidified, with a further two- to threefold strength increase after 3-months curing time. Although the hydraulic conductivity of the tailings paste decreased as a consequence of mixing with both GLD and FA, the difference was within one order of magnitude. The hydraulic conductivity was also reduced as the water/solid ratio of pure GLD decreased. The climatic freeze–thaw cyclic process led to 20 % increase in hydraulic conductivity. A proportion of 7:2:1 for tailings:GLD:FA was found to be a geotechnical satisfactory recipe to seal the mine waste. If porosity is reduced, improved water retention capacity and no cracking in the tailings can be assured, oxygen diffusion is limited, and oxidation of the mine waste is decelerated, thus mitigating acid rock drainage.

    • This study focuses on the effect of green liquor dregs (GLD) and fly ash (FA) on geotechnical stabilization of tailings, using shear strength test.The objectives were to evaluate thickening of tailings using GLD to paste to improve its water retention and to reduce waste percolation, to identify optimal proportion of additives regarding geotechnical stabilization of tailings, and to study the role of FA in the decrease of porosity and thus to reduce oxygen diffusion via hardening process.The results showed that GLD is a potential material for tailings stabilization by decreasing water percolation and improving water retention property. However, the paste of GLD amended tailings withstands low shear strength. The addition of FA to the paste greatly improves shear strength which is up to 2-3 times higher. Moreover, the fly ash hardening process effectively reinforces the strength of GLD amended tailings paste. Longer curing period (3 months) for specimens leads to up to 2-3 folds higher strength compared to that of 1 month. Hydraulic conductivity is reduced as a result of GLD and FA addition, since the porosity of the tailings decreased. If reduced porosity, improved water retention capacity and no cracks are ensured in the tailings, oxygen diffusion can be limited.

  • Sol-gel Synthesis and Photocatalytic Characterization of Immobilized TiO2 Films

    •    Contamination of surface and ground water from industrial wastes and anthropogenic activities represents one of the greatest challenges to the sustainable development of human society. Heterogeneous photocatalysis, a kind of advanced oxidation process characterized by the production of highly oxidative hydroxyl radicals, is a relatively novel subject with tremendous potential in water treatment applications.     The purpose of this research was first to develop feasible hydroxyl radical detection methods, which can be used to evaluate efficiency of photocatalytic process, and second to prepare immobilized TiO2 films with high photocatalytic activities by the sol gel method.     The feasibility of Indigo carmine and phthalic hydrazide as OH-radical probes was investigated. The organic dye Indigo carmine absorbs visual light strongly at 610 nm and its destruction can be monitored conveniently in a spectrophotometer. Results showed that both ·OH and HO2· can bleach Indigo carmine, and the bleaching yield of ·OH was pH independent. The photocatalytic dye bleaching in black light UV illuminated Degussa P25 TiO2 aerated suspensions was then investigated. A strong pH dependency of the bleaching yield was found.  This implies that the quantum yield of OH radical at pH 3 is one fourth compared to that at pH 10. The reaction of the OH radical with phthalic hydrazide will form strongly chemiluminescent 3-hydroxyphthalic hydrazide. Using the more specific phthalic hydrazide as OH radical probe, an even stronger pH dependent quantum yield of OH radical was found. At pH 10 the quantum yield reached the same magnitude as that obtained by using Indigo carmine, whereas the quantum yields at acidic pH were close to zero. However it was found that the addition of phosphate and fluoride anions can substantially enhance the OH radical yield at acidic pH by blocking the adsorption of phthalic hydrazide onto the TiO2 surfaces. Hence the adsorption of phthalic hydrazide to TiO2 is an important factor to consider when this method is used.     Photocatalytic TiO2 films coated on metal plates were prepared by a sol gel method using titanium isopropoxide as TiO2 precursor and isopropanol as solvent. The photocatalytic activity of the obtained films was evaluated by bleaching of indigo carmine at pH 9 under black light UV irradiation. The effect of the molar ratio of isopropanol, water and hydrochloric acid to titanium isopropoxide was studied. It was also shown that the activities of TiO2 films are considerably influenced by calcination temperature, coating cycles and the supporting materials.

    • The feasibility of using Indigo carmine (IC) as a probe of free radical production in photocatalysis was elucidated by comparative studies of its bleaching by gamma radiolysis and TiO2 photocatalysis. The gamma radiation result from pH 3 to 10 shows that both (OH)-O-center dot and HO2 center dot can bleach IC and that the bleaching yields of (OH)-O-center dot is pH independent at similar to 0.7 molecules per (OH)-O-center dot. The quantum yields (phi) of (OH)-O-center dot from Degussa p25 TiO2 photocatalysis from pH 3 to 10 was determined. It was found that phi was about 4% at pH 10 and dropped to below 1% at pH 3. This agrees well with the result of photocatalysis of Fricke dosimetry at pH 1. The effects of some common inorganic anions that may affect photocatalytic systems in technical applications (Cl-, SO42-, Br-, HPO42-, B4O72-, HCO3-/CO32-) was also investigated, showing that these ions does not inhibit the photocatalytic bleaching efficiency of IC.

    • Green liquor dregs (GLD), a residual product from sulfate paper mills, was blended with tailings, fly ash, and bark sludge with the aim of improving the material’s physical properties so that it could function as a sealing layer in dry covers on sulfidic mine waste. Geotechnical and geochemical investigations, including weathering cell tests, were carried out on GLD with admixtures to assess their effectiveness. Due to its alkaline character, GLD was shown to have the potential to improve leachate quality by decreasing metal mobility when blended with tailings. The admixtures showed favorable sealing layer properties such as high water retention capacity and low hydraulic conductivity. However, caution must be exercised when the dregs are blended with tailings containing large amounts of As and Mo, since increased leaching of these elements may be expected.

  • The role of SPINK1 in ETS rearrangement-negative prostate cancers

    • ETS gene fusions have been characterized in a majority of prostate cancers; however, the key molecular alterations in ETS-negative cancers are unclear. Here we used an outlier meta-analysis (meta-COPA) to identify SPINK1 outlier expression exclusively in a subset of ETS rearrangement-negative cancers ( approximately 10% of total cases). We validated the mutual exclusivity of SPINK1 expression and ETS fusion status, demonstrated that SPINK1 outlier expression can be detected noninvasively in urine, and observed that SPINK1 outlier expression is an independent predictor of biochemical recurrence after resection. We identified the aggressive 22RV1 cell line as a SPINK1 outlier expression model and demonstrate that SPINK1 knockdown in 22RV1 attenuates invasion, suggesting a functional role in ETS rearrangement-negative prostate cancers.
