    • CD1d-restricted natural killer T (NKT) cells can have multiple effects on an immune response, including the activation, regulation and attraction of innate immune cells, and modulation of adaptive immunity. Recent studies reveal that there are distinct subsets of NKT cells which selectively perform some of the functions attributed to CD1d-restricted cells, but the mechanisms underlying these functional differences have not been resolved. Our aim in this study was to identify novel NKT cell associated traits that would provide important insight into NKT cell activation and function. To this end, we have performed gene expression profiling of two separate subsets of NKT cells, analyzing genes differentially expressed in these cells compared to conventional CD4(+)NK1.1(-) T cells. We identify different sets of genes over expressed in each of the two NKT cell types, as well as genes that are common to the two CD1d-restricted NKT cell populations analyzed. A large number of these genes are highly relevant for NKT cell development, activation and function. Each NKT subtype displayed a unique set of chemokine receptors, integrins and molecules related to effector function, supporting the notion that distinct NKT cells can be selectively engaged and have diverse functions in different types of immune reactions.

  • Cultivating Compassion and Reducing Stress and Mental Ill-Health in Employees : A Randomized Controlled Study

    • Stress and mental ill-health carry considerable costs for both individuals and organizations. Although interventions targeting compassion and self-compassion have been shown to reduce stress and benefit mental health, related research in organizational settings is limited. We investigated the effects of a 6-week psychological intervention utilizing compassion training on stress, mental health, and self-compassion. Forty-nine employees of two organizations were randomly assigned to either the intervention (n = 25) or a physical exercise control condition (n = 24). Multilevel growth models showed that stress (p = 0.04) and mental ill-health (p = 0.02) decreased over 3 months in both groups (pre-intervention to follow-up: Cohen’s d = −0.46 and d = 0.33, respectively), while self-compassion only increased in the intervention group (p = 0.03, between group d = 0.53). There were no significant effects on life satisfaction in any of the groups (p > 0.53). The findings show promising results regarding the ability of compassion training within organizations to decrease stress and mental ill-health and increase self-compassion.

    • In Activity Based Working Environment (ABWE) offices, employees are allowed increased autonomy and are expected to choose where, when, with whom, and to some degree with what, to work; in other words, employees are expected to self-lead to a higher degree and to coordinate and align with colleagues. Effects of these expectations on employees’ cognitive stress and performance are understudied. In the present study, Swedish ABWE workers (N = 416) are compared with workers in cell offices (N = 30) and landscape offices (N = 64), and relationships of self-leadership, information richness, and autonomy with cognitive stress and performance were examined using regression analysis. Results show no relationship between office type and outcomes. For cognitive stress, information richness had the largest negative relationship, followed by self-leadership: goal-setting and autonomy. For performance, self-leadership: goal-setting had the largest positive relationship, followed by information richness. This suggests that when organizational situations cannot be strongly structured – for example because the best work process is not known, or innovation or different collaboration constellations are needed – they need instead to be enriched so that employee orientation and coordination do not become too much of a burden on the individual employee, disrupting cognitive functioning and performance.

    • In this paper, we advance the conversation about management education by outlining a future scenario of management teaching in which art is employed as a method, learning goal, and methodology. The scenario is informed by our personal experiences of teaching management and art expressed in terms of three design principles for management education: the principles of body, mind, and soul; and it is presented as an ethnography from the future. This future is envisioned to be characterized by four assumptions: that students are co-creators of knowledge; that the role of teachers is to facilitate the students’ learning processes; that Artificial Intelligence is an integral part of learning processes; and that the primary learning objective for the students is to develop their relations to the world and contribute with feasible future solutions to wicked problems and global challenges.

  • Diagnostic Efficacy of Rectal Suction Biopsy with Regard to Weight in Children Investigated for Hirschsprung's Disease

    • BACKGROUND/AIM: Diagnostic efficacy, defined as the percentage of rectal suction biopsy (RSB) specimens sufficient enough to determine the absence of ganglia cells in Hirschsprung's disease (HD) diagnosis, has been reported to be low, requiring repeated biopsies. The aim was to explore whether RSB diagnostic efficacy was influenced by the child's weight and to ascertain whether RSB efficacy differed between aganglionic and ganglionic tissue.MATERIALS AND METHODS: Efficacy analyses were conducted in a national HD-center's register on children 0-15 kg, examined between 2011-2019. First-time RSB diagnostic efficacy was correlated to the children's weight and final HD diagnosis.RESULTS: Among the 84 children who had first-time RSB, the overall diagnostic efficacy was 85% (71/84). The efficacy was higher among children weighing less than the identified cut-off of 9.0 kg (89% in 0-9.0 kg versus 62% in 9.01-15.0 kg, p = 0.026). Among children diagnosed with HD, 96% (26/27) weighed 0-9.0 kg. In this weight group, the diagnostic efficacy was lower in aganglionosis compared to ganglionosis (77%; 20/26 versus 96%; 43/45), p = 0.045).CONCLUSIONS: The RSB diagnostic efficacy was significantly higher in children weighing less than 9.0 kg and was less in aganglionic compared to ganglionic tissue. Therefore, weight can be useful to predict RSB diagnostic efficacy.

  • Systematic orientation of fresh rectal suction biopsies improves histopathological diagnostics in hirschsprung’s disease – a method description and preliminary report

    • Background: Optimizing rectal suction biopsy (RSB) diagnostics in Hirschsprung’s disease (HD) may shorten diagnostic time and prevent need for repeated biopsies. Aim: To explore whether systematic orientation of fresh RSB specimens increased biopsy quality, diagnostic times, diagnostic efficacy, and histopathologic workload, and to explore these outcome measures for aganglionic specimens. Materials/Methods: This was an observational case-control study conducted at a national referral center for HD on data collected from the local HD-diagnostic register. From 2019 each fresh RSB was oriented by the collector in a notch in a foam cushion, placed in a separate cassette, and sent in formalin for pathological analysis. Outcome measures of oriented RSB samples collected 2019–2021 were compared to those of non-oriented RSB samples collected 2015–2018. Staining/immunohistochemistry consisted of hematoxylin eosin, S-100 and calretinin. Results: 78 children with 81 RSBs and 242 biopsy analyzes were included. The frequency of high-quality RSB specimens was higher in oriented: 40% (42/106) versus non-oriented 25% (34/136) (p = 0.018), the diagnostic turnaround time was shorter: 2 days (1–5) versus 3 days (2–8) (p = 0.015), and the number of additional sectioning/leveling/re-orientation per biopsy was lower: 7 (3–26) versus 16 (7–72) (p = 0.011). Specifically for aganglionic specimens, the frequency of high-quality biopsies was generally higher in oriented than in non-oriented RSB specimens: 47% (28/59) versus 14% (7/50) (p < 0.001); the diagnostic efficacy was higher 95% (19/20) versus 60% (9/15) (p = 0.027) and the diagnostic turnaround time shorter: 2 days (2–3) versus 3 days (2–8) (p = 0.036). Conclusions: Systematic orientation of fresh RSB specimens improves HD diagnostics. Improvement was consistent in aganglionic specimens.

    • Spatial and temporal variations among individual human cell proteomes are comprehensively mapped across the cell cycle using proteomic imaging and transcriptomics. The cell cycle, over which cells grow and divide, is a fundamental process of life. Its dysregulation has devastating consequences, including cancer(1-3). The cell cycle is driven by precise regulation of proteins in time and space, which creates variability between individual proliferating cells. To our knowledge, no systematic investigations of such cell-to-cell proteomic variability exist. Here we present a comprehensive, spatiotemporal map of human proteomic heterogeneity by integrating proteomics at subcellular resolution with single-cell transcriptomics and precise temporal measurements of individual cells in the cell cycle. We show that around one-fifth of the human proteome displays cell-to-cell variability, identify hundreds of proteins with previously unknown associations with mitosis and the cell cycle, and provide evidence that several of these proteins have oncogenic functions. Our results show that cell cycle progression explains less than half of all cell-to-cell variability, and that most cycling proteins are regulated post-translationally, rather than by transcriptomic cycling. These proteins are disproportionately phosphorylated by kinases that regulate cell fate, whereas non-cycling proteins that vary between cells are more likely to be modified by kinases that regulate metabolism. This spatially resolved proteomic map of the cell cycle is integrated into the Human Protein Atlas and will serve as a resource for accelerating molecular studies of the human cell cycle and cell proliferation.

Ett vidgat rum, där till exempel stiftelser också ryms, kan bidra till att förlänga tidsperspektivet - och det som kortsiktigt inte går att räkna hem, kan på lång sikt visa sig vara det nyttiga.

Evidence of the effect of this collaboration will be drawn from interviews, observations, and reflections gathered from the business students.FindingsThis study and its related counterparts show that liberal arts studies incorporated into business programs enhance creativity, professional judgment, social contribution, and personal fulfillment in students. The addition of multiple framing was found to be particularly healthy to the students’ educational development.Originality/valueThe involvement of creative processes in business education leads to a more fulfilling and beneficial program for students.

Under de första fyra terminerna, får de, parallellt med andra kurser, läsa ett spår kallat ”Global Challenges”, varvid en termin ägnas åt mänsklig hållbarhet. Förutom att hålla själva, behöver de lära sig att skapa hållbara organisationer, men också att tänka i termer av meningsfullhet och att släppa sitt ego, till förmån för ekosystemet.

    • Det talas om företag som konst: om estetik, kreativitet, immateriella värden, upplevelser och fantasi. Likaså talas det om konst som företag: om ekonomisk styrning, marknadsföring, varumärken, tydliga mål och genomtänka strategier. Frågan är vad som har hänt. Det var inte länge sedan konst och företag sågs som skilda världar. Konst associerades med sådant som frihet, originalitet, irrationalitet, känslor, intuition och geniförklarade individer. Företag associerades med sådant som nytta, massproduktion, kontroll, kalkyler, rationalitet, förnuft och strukturer. Idag ser det annorlunda ut. Idag verkar föreställningarna om konst och företag nästan ha bytt plats med varandra. Konstnären har blivit företagsledare: hon eller han ska kunna räkna, samarbeta med näringslivet, skapa evenemang, vara entreprenör och göra sig själv till ett varumärke. Parallellt har företagsledaren blivit konstnär: hon eller han ska kunna skapa visioner, vara kreativ, inspirerande och fantasifull, lita till sin intuition och kunna iscensätta sig själv och sitt företag. Utifrån litteraturstudier, intervjuer med konstnärliga ledare och företagsledare, direktobservationer samt en poetisk närläsning, studeras i boken föreställningar om konst och företag. Tre parallella tendenser i samtiden står i fokus: företagens estetisering, ledarskapets romantisering samt konstens ekonomisering. Dessutom diskuteras om och hur konsten - som praktik och uttryck - kan ge insikter om företag och ledarskap.

    • The nucleolus is essential for ribosome biogenesis and is involved in many other cellular functions. We performed a systematic spatiotemporal dissection of the human nucleolar proteome using confocal microscopy. In total, 1,318 nucleolar proteins were identified; 287 were localized to fibrillar components, and 157 were enriched along the nucleoplasmic border, indicating a potential fourth nucleolar subcompartment: the nucleoli rim. We found 65 nucleolar proteins (36 uncharacterized) to relocate to the chromosomal periphery during mitosis. Interestingly, we observed temporal partitioning into two recruitment phenotypes: early (prometaphase) and late (after metaphase), suggesting phase-specific functions. We further show that the expression ofMKI67 is critical for this temporal partitioning. We provide the first proteome-wide analysis of intrinsic protein disorder for the human nucleolus and show that nucleolar proteins in general, and mitotic chromosome proteins in particular, have significantly higher intrinsic disorder level compared to cytosolic proteins. In summary, this study provides a comprehensive and essential resource of spatiotemporal expression data for the nucleolar proteome as part of the Human Protein Atlas.

    • This thesis focuses on the spatiotemporal mapping of proteins at a subcellular level. In other words, determining the cellular location of proteins over time. From a biological point of view, knowledge about protein location is fundamental to understand protein function. In the longer run, this also means better understanding of cells in the context of health and disease, since protein malfunction and mislocalization are two important factors during disease development. Using an antibody-based imaging approach, Paper I contains a subcellular map of 12 003 protein in 30 different cellular structures, freely accessible as part of the Human Protein Atlas (www.proteinatlas.org). Apart from enabling exploration of the organellar proteomes, we conclude that half of the human proteins localize to multiple compartments, and that almost one fifth display cell-to-cell variations in terms of protein expression. Paper II aimed to decrease the cumbersome work of manual protein location annotations by leveraging the power of the crowd through citizen science. By integrating the image-classification task into a video game with a massive player base, EVE online, protein location labels could be efficiently and rapidly assessed compared to manual curation from a few experts. To compare the performance of the players, a deep learning classifier was developed. The algorithm was capable of classifying protein location in images containing several challenging localization problems, such as multilocalizing proteins, cell line variations and rare classes. Using the protein location data from Paper I and Paper II, Paper III presents an image-based characterization of the nucleolar proteome. In total, 1 318 nucleolar proteins are included, of which 157 localizes to a fourth nucleolar compartment, the nucleolar rim. Additionally, 65 proteins were detected on the chromosomal periphery during mitosis, and these could be further divided into two recruitment phenotypes with different temporal profiles. Also, the mitotic chromosome proteins are enriched for intrinsically disordered domains, suggesting liquid-like properties of the perichromosomal layer. Paper IV presents a systematic dissection of the variable proteome drafted in Paper I. We show evidence for 539 proteins being correlated to cell cycle variations, of which a minority are also cycling at a transcriptional level, suggesting protein regulation at a translational or post-translational level. Additionally, we detected hundreds of proteins with previously unknown relations to mitosis and the cell cycle, many being linked to proliferation and oncogenic functions.In conclusion, Paper I and Paper II provide a basis for further in-depth studies of proteins at a subcellular level, while Paper III and Paper IV show how this resource can be used to study proteins in space and time. The results enable system-level investigation of protein dynamics, as well as provide exciting insights into organellar organization, such as the nucleolus.

    • The research cluster Vulnerability as Virtuosity is to collectively examine human and more-than-human vulnerabilities withinthe context of crisis, with a specific focus on exploring the following questions:- How are human responses and vulnerabilities understood and manifested across differentfields and disciplines?- How can vulnerabilities be addressed fairly and sustainably?- What insights can be gained from human and more-than-human vulnerabilities?- How can the diverse knowledge on vulnerability from various fields be shared andleveraged?- What practices can be initiated to align vulnerability with a more sustainable and justworld?"
