    • Copy number variants (CNVs) account for a major proportion of human genetic polymorphism and have been predicted to have an important role in genetic susceptibility to common disease. To address this we undertook a large, direct genome-wide study of association between CNVs and eight common human diseases. Using a purpose-designed array we typed,19,000 individuals into distinct copy-number classes at 3,432 polymorphic CNVs, including an estimated similar to 50% of all common CNVs larger than 500 base pairs. We identified several biological artefacts that lead to false-positive associations, including systematic CNV differences between DNAs derived from blood and cell lines. Association testing and follow-up replication analyses confirmed three loci where CNVs were associated with disease-IRGM for Crohn's disease, HLA for Crohn's disease, rheumatoid arthritis and type 1 diabetes, and TSPAN8 for type 2 diabetes-although in each case the locus had previously been identified in single nucleotide polymorphism (SNP)-based studies, reflecting our observation that most common CNVs that are well-typed on our array are well tagged by SNPs and so have been indirectly explored through SNP studies. We conclude that common CNVs that can be typed on existing platforms are unlikely to contribute greatly to the genetic basis of common human diseases.

  • Sources, Composition, and Export of Particulate Organic Matter Across British Estuaries

    • Estuaries receive and process a large amount of particulate organic carbon (POC) prior to its export into coastal waters. Studying the origin of this POC is key to understanding the fate of POC and the role of estuaries in the global carbon cycle. Here, we evaluated the concentrations of POC, as well as particulate organic nitrogen (PON), and used stable carbon and nitrogen isotopes to assess their sources across 13 contrasting British estuaries during five different sampling campaigns over 1 year. We found a high variability in POC and PON concentrations across the salinity gradient, reflecting inputs, and losses of organic material within the estuaries. Catchment land cover appeared to influence the contribution of POC to the total organic carbon flux from the estuary to coastal waters, with POC contributions >36% in estuaries draining catchments with a high percentage of urban/suburban land, and <11% in estuaries draining catchments with a high peatland cover. There was no seasonal pattern in the isotopic composition of POC and PON, suggesting similar sources for each estuary over time. Carbon isotopic ratios were depleted (-26.7 +/- 0.42 parts per thousand, average +/- sd) at the lowest salinity waters, indicating mainly terrigenous POC (TPOC). Applying a two-source mixing model, we observed high variability in the contribution of TPOC at the highest salinity waters between estuaries, with a median value of 57%. Our results indicate a large transport of terrigenous organic carbon into coastal waters, where it may be buried, remineralized, or transported offshore. Plain Language Summary Estuaries transport and process a large amount terrigenous particulate organic matter (i.e., carbon and nitrogen) prior to its export to coastal waters. In order to understand the fate of organic carbon and the role of estuaries in the global carbon cycle it is essential to improve our knowledge on its composition, origin, and amount of carbon transported. We quantified the elemental concentrations and stable isotopes composition of carbon and nitrogen to quantify the amount of terrigenous particulate organic matter transported by 13 British estuaries, which drain catchments of diverse land cover under different hydrological conditions. We found a great variability in particulate organic carbon (POC) and particulate organic nitrogen concentrations across the salinity gradient, implying inputs, and losses of material within the estuaries. Each estuary had similar sources of particulate material throughout the year. In most of the estuaries, the POC had a terrigenous origin at the lowest salinity waters. The terrigenous organic carbon contribution decreased toward coastal waters with an average contribution of 57% at the highest salinity waters, indicating a large transport of terrigenous organic carbon into coastal waters.

A recent phase III clinical trial of an sPLA(2) inhibitor (varespladib) was stopped prematurely for lack of efficacy. less thanbrgreater than less thanbrgreater thanMethods We conducted a Mendelian randomization meta-analysis of 19 general population studies (8,021 incident, 7,513 prevalent major vascular events [MVE] in 74,683 individuals) and 10 acute coronary syndrome (ACS) cohorts (2,520 recurrent MVE in 18,355 individuals) using rs11573156, a variant in PLA2G2A encoding the sPLA(2)-IIA isoenzyme, as an instrumental variable. less thanbrgreater than less thanbrgreater thanResults PLA2G2A rs11573156 C allele associated with lower circulating sPLA(2)-IIA mass (38% to 44%) and sPLA(2) enzyme activity (3% to 23%) per C allele. The odds ratio (OR) for MVE per rs11573156 C allele was 1.02 (95% confidence interval [CI]: 0.98 to 1.06) in general populations and 0.96 (95% CI: 0.90 to 1.03) in ACS cohorts. In the general population studies, the OR derived from the genetic instrumental variable analysis for MVE for a 1-log unit lower sPLA(2)-IIA mass was 1.04 (95% CI: 0.96 to 1.13), and differed from the non-genetic observational estimate (OR: 0.69; 95% CI: 0.61 to 0.79). In the ACS cohorts, both the genetic instrumental variable and observational ORs showed a null association with MVE. Instrumental variable analysis failed to show associations between sPLA2 enzyme activity and MVE. less thanbrgreater than less thanbrgreater thanConclusions Reducing sPLA(2)-IIA mass is unlikely to be a useful therapeutic goal for preventing cardiovascular events.

  • Large-scale migration into Britain during the Middle to Late Bronze Age

    • Present-day people from England and Wales harbour more ancestry derived from Early European Farmers (EEF) than people of the Early Bronze Age1. To understand this, we generated genome-wide data from 793 individuals, increasing data from the Middle to Late Bronze and Iron Age in Britain by 12-fold, and Western and Central Europe by 3.5-fold. Between 1000 and 875 BC, EEF ancestry increased in southern Britain (England and Wales) but not northern Britain (Scotland) due to incorporation of migrants who arrived at this time and over previous centuries, and who were genetically most similar to ancient individuals from France. These migrants contributed about half the ancestry of Iron Age people of England and Wales, thereby creating a plausible vector for the spread of early Celtic languages into Britain. These patterns are part of a broader trend of EEF ancestry becoming more similar across central and western Europe in the Middle to Late Bronze Age, coincident with archaeological evidence of intensified cultural exchange2-6. There was comparatively less gene flow from continental Europe during the Iron Age, and Britain's independent genetic trajectory is also reflected in the rise of the allele conferring lactase persistence to ~50% by this time compared to ~7% in central Europe where it rose rapidly in frequency only a millennium later. This suggests that dairy products were used in qualitatively different ways in Britain and in central Europe over this period.

    • This paper presents the status of the work, by April 2008, in C4.110, a joint working group by CIGRE, CIRED and UIE. The scope of the working group is to gather technical knowledge on the immunity of equipment, installations and processes against voltage dips, and to use this knowledge in the further development of methods and standards. The activities of the working group are divided in seven "chapters". Chapters 1 and 7 are introduction and conclusions, respectively. Chapter 2 gives a general description of voltage dips as they appear at the terminals of sensitive equipment. Chapter 3 describes the performance of equipment and processes during voltage dips. This chapter also includes recommendations on the design of processes. In Chapter 4 the results from Chapter 2 and Chapter 3 are combined to set requirements for the dip characteristics that should be included in immunity testing. Chapter 5 is the data gathering chapter, covering data on voltage-dip statistics at different locations, but also data on the economics of equipment immunity and testing. Finally, in Chapter 6, recommendations for immunity objectives will be given. Important contributions of the working group are: a check-list of voltage dip characteristics to be used early in the design of equipment; a methodology to assess the performance of a complete installation and to include voltage-dip performance in the design of the installation; recommendations for characterization testing of equipment against voltage dips; recommendations for voltagedip immunity of equipment.

    • We introduce the AusTraits database - a compilation of values of plant traits for taxa in the Australian flora (hereafter AusTraits). AusTraits synthesises data on 448 traits across 28,640 taxa from field campaigns, published literature, taxonomic monographs, and individual taxon descriptions. Traits vary in scope from physiological measures of performance (e.g. photosynthetic gas exchange, water-use efficiency) to morphological attributes (e.g. leaf area, seed mass, plant height) which link to aspects of ecological variation. AusTraits contains curated and harmonised individual- and species-level measurements coupled to, where available, contextual information on site properties and experimental conditions. This article provides information on version 3.0.2 of AusTraits which contains data for 997,808 trait-by-taxon combinations. We envision AusTraits as an ongoing collaborative initiative for easily archiving and sharing trait data, which also provides a template for other national or regional initiatives globally to fill persistent gaps in trait knowledge.

  • A case-matched gender comparison transcriptomic screen identifies eIF4E and eIF5 as potential prognostic markers in male breast cancer

    • Purpose: Breast cancer affects both genders, but is understudied in men. Although still rare, male breast cancer (MBC) is being diagnosed more frequently. Treatments are wholly informed by clinical studies conducted in women, based on assumptions that underlying biology is similar. Experimental Design: A transcriptomic investigation of male and female breast cancer was performed, confirming transcriptomic data in silico. Biomarkers were immunohistochemically assessed in 697 MBCs (n = 477, training; n = 220, validation set) and quantified in pre- and posttreatment samples from an MBC patient receiving everolimus and PI3K/mTOR inhibitor. Results: Gender-specific gene expression patterns were identified. eIF transcripts were upregulated in MBC. eIF4E and eIF5 were negatively prognostic for overall survival alone (log-rank P = 0.013; HR = 1.77, 1.12-2.8 and P = 0.035; HR = 1.68, 1.03-2.74, respectively), or when coexpressed (P = 0.01; HR = 2.66, 1.26-5.63), confirmed in the validation set. This remained upon multivariate Cox regression analysis [eIF4E P = 0.016; HR = 2.38 (1.18-4.8), eIF5 P = 0.022; HR = 2.55 (1.14-5.7); coexpression P = 0.001; HR = 7.04 (2.22-22.26)]. Marked reduction in eIF4E and eIF5 expression was seen post BEZ235/everolimus, with extended survival. Conclusions: Translational initiation pathway inhibition could be of clinical utility in MBC patients overexpressing eIF4E and eIF5. With mTOR inhibitors that target this pathway now in the clinic, these biomarkers may represent new targets for therapeutic intervention, although further independent validation is required.

  • Blood Pressure Loci Identified with a Gene-Centric Array.

    • Raised blood pressure (BP) is a major risk factor for cardiovascular disease. Previous studies have identified 47 distinct genetic variants robustly associated with BP, but collectively these explain only a few percent of the heritability for BP phenotypes. To find additional BP loci, we used a bespoke gene-centric array to genotype an independent discovery sample of 25,118 individuals that combined hypertensive case-control and general population samples. We followed up four SNPs associated with BP at our p < 8.56× 10(-7) study-specific significance threshold and six suggestively associated SNPs in a further 59,349 individuals. We identified and replicated a SNP at LSP1/TNNT3, a SNP at MTHFR-NPPB independent (r(2) = 0.33) of previous reports, and replicated SNPs at AGT and ATP2B1 reported previously. An analysis of combined discovery and follow-up data identified SNPs significantly associated with BP at p < 8.56× 10(-7) at four further loci (NPR3, HFE, NOS3, and SOX6). The high number of discoveries made with modest genotyping effort can be attributed to using a large-scale yet targeted genotyping array and to the development of a weighting scheme that maximized power when meta-analyzing results from samples ascertained with extreme phenotypes, in combination with results from nonascertained or population samples. Chromatin immunoprecipitation and transcript expression data highlight potential gene regulatory mechanisms at the MTHFR and NOS3 loci. These results provide candidates for further study to help dissect mechanisms affecting BP and highlight the utility of studying SNPs and samples that are independent of those studied previously even when the sample size is smaller than that in previous studies.

  • Linear dust polarization during the embedded phase of protostar formation: Synthetic observations of bridge structures

    • Measuring polarization from thermal dust emission can provide important constraints on the magnetic field structure around embedded protostars. However, interpreting the observations is challenging without models that consistently account for both the complexity of the turbulent protostellar birth environment and polarization mechanisms. Aims. We aim to provide a better understanding of dust polarization maps of embedded protostars with a focus on bridge-like structures such as the structure observed toward the protostellar multiple system IRAS 16293-2422 by comparing synthetic polarization maps of thermal reemission with recent observations. Methods. We analyzed the magnetic field morphology and properties associated with the formation of a protostellar multiple based on ideal magnetohydrodynamic 3D zoom-in simulations carried out with the » RAMSES code. To compare the models with observations, we postprocessed a snapshot of a bridge-like structure that is associated with a forming triple star system with the radiative transfer code » POLARIS and produced multiwavelength dust polarization maps. Results. The typical density in the most prominent bridge of our sample is about 10-16 g cm-3, and the magnetic field strength in the bridge is about 1 to 2 mG. Inside the bridge, the magnetic field structure has an elongated toroidal morphology, and the dust polarization maps trace the complex morphology. In contrast, the magnetic field strength associated with the launching of asymmetric bipolar outflows is significantly more magnetized (∼100 mG). At λ = 1.3 mm, and the orientation of the grains in the bridge is very similar for the case accounting for radiative alignment torques (RATs) compared to perfect alignment with magnetic field lines. However, the polarization fraction in the bridge is three times smaller for the RAT scenario than when perfect alignment is assumed. At shorter wavelength (λ 200 μm), however, dust polarization does not trace the magnetic field because other effects such as self-scattering and dichroic extinction dominate the orientation of the polarization. Conclusions. Compared to the launching region of protostellar outflows, the magnetic field in bridge-like structures is weak. Synthetic dust polarization maps of ALMA Bands 6 and 7 (1.3 mm and 870 μm, respectively) can be used as a tracer of the complex morphology of elongated toroidal magnetic fields associated with bridges.

  • Mutational Processes Molding the Genomes of 21 Breast Cancers

    • All cancers carry somatic mutations. The patterns of mutation in cancer genomes reflect the DNA damage and repair processes to which cancer cells and their precursors have been exposed. To explore these mechanisms further, we generated catalogs of somatic mutation from 21 breast cancers and applied mathematical methods to extract mutational signatures of the underlying processes. Multiple distinct single- and double-nucleotide substitution signatures were discernible. Cancers with BRCA1 or BRCA2 mutations exhibited a characteristic combination of substitution mutation signatures and a distinctive profile of deletions. Complex relationships between somatic mutation prevalence and transcription were detected. A remarkable phenomenon of localized hypermutation, termed "kataegis,'' was observed. Regions of kataegis differed between cancers but usually colocalized with somatic rearrangements. Base substitutions in these regions were almost exclusively of cytosine at TpC dinucleotides. The mechanisms underlying most of these mutational signatures are unknown. However, a role for the APOBEC family of cytidine deaminases is proposed.

  • Multi-proxy identification of the Laschamp geomagnetic field excursion in Lake Pupuke, New Zealand

    • We present palaeomagnetic and cosmogenic radionuclide records of the Laschamp geomagnetic excursion in Lake Pupuke, a maar lake in Auckland, New Zealand. Laschamp was identified by a combination of relative palaeointensity, (10)Be and (14)C data from the lake sediments and represents the first such record from the Southern Hemisphere. Despite the high organic carbon content, which causes relatively weak natural remanent magnetisations, the geomagnetic intensity minimum associated with the Laschamp excursion is identifiable as a relative palaeointensity minimum that is synchronous with (i) a peak in (10)Be concentration and (ii) an anomaly in Delta(14)C. The Lake Pupuke time scale, provided by (14)C data calibrated with INTCAL09, places the (10)Be maximum at the same time as a (10)Be maximum in Greenland ice cores when secured to the GICC05 time scale. The central age of the Laschamp geomagnetic excursion in Lake Pupuke as defined by the (10)Be prediction peak is c. 41 kyr, which confirms its global application as a palaeomagnetic isochron. Anomalous palaeomagnetic directional data at c. 32 kyr in the Lake Pupuke sediments may represent the Mono Lake geomagnetic excursion, but tephra layers caused by frequent eruptions in the Auckland volcanic field during this excursion probably disrupted the palaeointensity signal. The study highlights the value of combining traditional palaeomagnetic methods with measurements of cosmogenic radionuclides in the quest for accurate and precise geochronologies during MIS3, a time of rapid global climate change.     • Obesity is globally prevalent and highly heritable, but its underlying genetic factors remain largely elusive. To identify genetic loci for obesity susceptibility, we examined associations between body mass index and ~2.8 million SNPs in up to 123,865 individuals with targeted follow up of 42 SNPs in up to 125,931 additional individuals. We confirmed 14 known obesity susceptibility loci and identified 18 new loci associated with body mass index (P < 5 × 10−8), one of which includes a copy number variant near GPRC5B. Some loci (at MC4R, POMC, SH2B1 and BDNF) map near key hypothalamic regulators of energy balance, and one of these loci is near GIPR, an incretin receptor. Furthermore, genes in other newly associated loci may provide new insights into human body weight regulation.

    • Humans began to leave lasting impacts on Earth’s surface starting 10,000 to 8000 years ago. Through a synthetic collaboration with archaeologists around the globe, Stephens et al. compiled a comprehensive picture of the trajectory of human land use worldwide during the Holocene (see the Perspective by Roberts). Hunter-gatherers, farmers, and pastoralists transformed the face of Earth earlier and to a greater extent than has been widely appreciated, a transformation that was essentially global by 3000 years before the present.Science, this issue p. 897; see also p. 865Environmentally transformative human use of land accelerated with the emergence of agriculture, but the extent, trajectory, and implications of these early changes are not well understood. An empirical global assessment of land use from 10,000 years before the present (yr B.P.) to 1850 CE reveals a planet largely transformed by hunter-gatherers, farmers, and pastoralists by 3000 years ago, considerably earlier than the dates in the land-use reconstructions commonly used by Earth scientists. Synthesis of knowledge contributed by more than 250 archaeologists highlighted gaps in archaeological expertise and data quality, which peaked for 2000 yr B.P. and in traditionally studied and wealthier regions. Archaeological reconstruction of global land-use history illuminates the deep roots of Earth’s transformation and challenges the emerging Anthropocene paradigm that large-scale anthropogenic global environmental change is mostly a recent phenomenon.

  • Capital and income breeding: the role of food supply.

    • An aspect of life history that has seen increasing attention in recent years is that of strategies for financing the costs of offspring production. These strategies are often described by a continuum ranging from capital breeding, in which costs are met purely from endogenous reserves, to income breeding, in which costs are met purely from concurrent intake. A variety of factors that might drive strategies towards a given point on the capital-income continuum has been reviewed, and assessed using analytical models. However, aspects of food supply, including seasonality and unpredictability, have often been cited as important drivers of capital and income breeding but are difficult to assess using analytical models. Consequently, we used dynamic programming to assess the role of the food supply in shaping offspring provisioning strategies. Our model is parameterized for a pinniped (one taxon remarkable for the range of offspring strategies that it illustrates). We show that increased food availability, increased seasonality and, to a lesser extent, increased unpredictability can all favor the emergence of capital breeding. In terms of the conversion of energy into offspring growth, the shorter periods of care associated with capital breeding are considerably more energetically efficient than income breeding, because shorter periods of care are associated with a higher ratio of energy put into offspring growth, to energy spent on parent and offspring maintenance metabolism. Moreover, no clear costs are currently associated with capital accumulation in pinnipeds. This contrasts with general assumptions about endotherms, which suggest that income breeding will usually be preferred. Our model emphasizes the role of seasonally high abundances of food in enabling mothers to pursue an energetically efficient capital breeding strategy. We discuss the importance of offspring development for dictating strategies for financing offspring production.

    • We present the first continuous paleolimnological reconstruction from the North Island of New Zealand (37A degrees S) that spans the last 48.2 cal kyr. A tephra- and radiocarbon-based chronology was developed to infer the timing of marked paleolimnological changes in Lake Pupuke, Auckland, New Zealand, identified using sedimentology, magnetic susceptibility, grain size and geochemistry (carbon, nitrogen and sulphur concentrations and fluxes, carbon and nitrogen stable isotopes). Variable erosional influx, biomass and benthic REDOX conditions are linked to changing effective precipitation and seasonality within three inferred broad intervals of climatic change: (1) the Last Glacial Coldest Phase (LGCP) of reduced effective precipitation and cooler temperatures, from 28.8 to 18.0 cal kyr BP, (2) the Last Glacial Interglacial Transition (LGIT) of increasing effective precipitation and warmer conditions, from 18.0 to 10.2 cal kyr BP, and (3) a Holocene interval of high effective precipitation, beginning with a warm period of limited seasonality from 10.2 cal kyr BP and followed by increasing seasonality from 7.6 cal kyr BP. The LGCP and LGIT also contain millennial-scale climate events, including the coldest inferred glacial conditions during the LGCP from 27.8 to 26.0 and 22.0-19.0 cal kyr BP, and a climate reversal in the LGIT associated with lower lake level, from 14.5 to 13.8 cal kyr BP, coeval with the Antarctic Cold Reversal. The onset of seasonal thermal stratification occurred at 5.7 cal kyr BP and was linked to natural eutrophication of Lake Pupuke, which produced enhanced organic sedimentation.

  • Discovery of Molecular-line Polarization in the Disk of TW Hya

    • We report observations of polarized line and continuum emission from the disk of TW Hya using the Atacama Large Millimeter/submillimeter Array. We target three emission lines, (CO)-C-12 (3-2), (CO)-C-13 (3-2), and CS (7-6), to search for linear polarization due to the Goldreich-Kylafis effect, while simultaneously tracing the continuum polarization morphology at 332 GHz (900 mu m), achieving a spatial resolution of 0.'' 5 (30 au). We detect linear polarization in the dust continuum emission; the polarization position angles show an azimuthal morphology, and the median polarization fraction is similar to 0.2%, comparable to previous, lower frequency observations. Adopting a "shift-and-stack" technique to boost the sensitivity of the data, combined with a linear combination of the Q and U components to account for their azimuthal dependence, we detect weak linear polarization of (CO)-C-12 and (CO)-C-13 line emission at a similar to 10 sigma and similar to 5 sigma significance, respectively. The polarization was detected in the line wings, reaching a peak polarization fraction of similar to 5% and similar to 3% for the two molecules between disk radii of 0.'' 5 and 1 ''. The sign of the polarization was found to flip from the blueshifted side of the emission to the redshifted side, suggesting a complex, asymmetric polarization morphology. Polarization is not robustly detected for the CS emission; however, a tentative signal, comparable in morphology to that found for the (CO)-C-12 and (CO)-C-13 emission, is found at a less than or similar to 3 sigma significance. We are able to reconstruct a polarization morphology, consistent with the azimuthally averaged profiles, under the assumption that this is also azimuthally symmetric, which can be compared with future higher-sensitivity observations.

    • We present Atacama Large Millimeter/submillimeter Array (0.87 mm) and Very Large Array (9 mm) observations toward OMC-2 FIR4 and OMC-2 FIR3 within the Orion integral-shaped filament, thought to be two of the nearest regions of intermediate-mass star formation. We characterize the continuum sources within these regions on ?40 au (01) scales and associated molecular line emission at a factor of ?30 better resolution than previous observations at similar wavelengths. We identify six compact continuum sources within OMC-2 FIR4, four in OMC-2 FIR3, and one additional source just outside OMC-2 FIR4. This continuum emission is tracing the inner envelope and/or disk emission on less than 100 au scales. HOPS-108 is the only protostar in OMC-2 FIR4 that exhibits emission from high-excitation transitions of complex organic molecules (e.g., methanol and other lines) coincident with the continuum emission. HOPS-370 in OMC-2 FIR3, with L;?;360 L, also exhibits emission from high-excitation methanol and other lines. The methanol emission toward these two protostars is indicative of temperatures high enough to thermally evaporate it from icy dust grains; overall, these protostars have characteristics similar to hot corinos. We do not identify a clear outflow from HOPS-108 in (CO)-C-12, but we find evidence of interaction between the outflow/jet from HOPS-370 and the OMC-2 FIR4 region. A multitude of observational constraints indicate that HOPS-108 is likely a low- to intermediate-mass protostar in its main mass accretion phase and is the most luminous protostar in OMC-2 FIR4. The high-resolution data presented here are essential for disentangling the embedded protostars from their surrounding dusty environments and characterizing them.

    • We have conducted a survey of 328 protostars in the Orion molecular clouds with the Atacama Large Millimeter/submillimeter Array at 0.87 mm at a resolution of ∼0.″1 (40 au), including observations with the Very Large Array at 9 mm toward 148 protostars at a resolution of ∼0.″08 (32 au). This is the largest multiwavelength survey of protostars at this resolution by an order of magnitude. We use the dust continuum emission at 0.87 and 9 mm to measure the dust disk radii and masses toward the Class 0, Class I, and flat-spectrum protostars, characterizing the evolution of these disk properties in the protostellar phase. The mean dust disk radii for the Class 0, Class I, and flat-spectrum protostars are 44.9-3.4+5.8, 37.0-3.0+4.9, and 28.5-2.3+3.7 au, respectively, and the mean protostellar dust disk masses are 25.9-4.0+7.7, 14.9-2.2+3.8, 11.6-1.9+3.5 M⊙, respectively. The decrease in dust disk masses is expected from disk evolution and accretion, but the decrease in disk radii may point to the initial conditions of star formation not leading to the systematic growth of disk radii or that radial drift is keeping the dust disk sizes small. At least 146 protostellar disks (35% of 379 detected 0.87 mm continuum sources plus 42 nondetections) have disk radii greater than 50 au in our sample. These properties are not found to vary significantly between different regions within Orion. The protostellar dust disk mass distributions are systematically larger than those of Class II disks by a factor of >4, providing evidence that the cores of giant planets may need to at least begin their formation during the protostellar phase.

  • Seventy-five genetic loci influencing the human red blood cell

    • Anaemia is a chief determinant of global ill health, contributing to cognitive impairment, growth retardation and impaired physical capacity. To understand further the genetic factors influencing red blood cells, we carried out a genome-wide association study of haemoglobin concentration and related parameters in up to 135,367 individuals. Here we identify 75 independent genetic loci associated with one or more red blood cell phenotypes at P < 10(-8), which together explain 4-9% of the phenotypic variance per trait. Using expression quantitative trait loci and bioinformatic strategies, we identify 121 candidate genes enriched in functions relevant to red blood cell biology. The candidate genes are expressed preferentially in red blood cell precursors, and 43 have haematopoietic phenotypes in Mus musculus or Drosophila melanogaster. Through open-chromatin and coding-variant analyses we identify potential causal genetic variants at 41 loci. Our findings provide extensive new insights into genetic mechanisms and biological pathways controlling red blood cell formation and function.
