  • PRL2 Phosphatase Promotes Oncogenic KIT Signaling in Leukemia Cells through Modulating CBL Phosphorylation

    • Receptor tyrosine kinase KIT is frequently activated in acute myeloid leukemia (AML). While high PRL2 (PTP4A2) expression is correlated with activation of SCF/KIT signaling in AML, the underlying mechanisms are not fully understood. We discovered that inhibition of PRL2 significantly reduces the burden of oncogenic KIT-driven leukemia and extends leukemic mice survival. PRL2 enhances oncogenic KIT signaling in leukemia cells, promoting their proliferation and survival. We found that PRL2 dephosphorylates CBL at tyrosine 371 and inhibits its activity toward KIT, leading to decreased KIT ubiquitination and enhanced AKT and ERK signaling in leukemia cells.

    • The objective of this study was to examine the psychometric properties of the Chinese version of the Dermatology Life Quality Index (DLQI) and to assess the invariance of its items with respect to several patient parameters via Rasch analysis. Data were aggregated from 9,845 patients with various skin diseases across 9 hospitals in different regions of China. The response structure, local independence, and reliability of the DLQI scale were analysed in a partial credit model, and differential item functioning (DIF) across region, disease, sex, and age were assessed with a Mantel-Haenszel procedure. Although acceptable scale reliability (Person Separation Index=2.3) was obtained, several problems were revealed, including disordered response thresholds, misfitting items, DIF by geographical region and disease, and mis-targeting patients with mild impairment regarding health-related quality of life (HRQL). In conclusion, the DLQI provides inadequate information on patients' impairments in HRQL, and the application of the DLQI in Chinese patients with skin disease is limited.

    • As a major challenge and opportunity for traditional manufacturing, intelligent manufacturing is facing the needs of sustainable development in future. Sustainability assessment undoubtedly plays a pivotal role for future development of intelligent manufacturing. Aiming at this, the paper presents the digital twin driven information architecture of sustainability assessment oriented for dynamic evolution under the whole life cycle based on the classic digital twin mapping system. The sustainability assessment method segment of the architecture includes indicator system building, indicator value determination, indicator importance degree determination and intelligent manufacturing project assessing. A novel approach for treating the ambiguity of expert judgment in indicator value determination by introducing trapezoidal fuzzy number into analytic hierarchy process is proposed, while the complexity of the influence relationship among the indicators is processed by the integration of complex networks modeling and PROMETHEE II for the indicator importance degree determination. A two-stage evidence combination model based on evidence theory is built for intelligent manufacturing project assessing lastly. The presented digital-twin-driven information architecture and the sustainability assessment method is tested and validated on a study of sustainability assessment of 8 intelligent manufacturing projects of an air conditioning enterprise. The results of the presented method were validated by comparing them with the results of the fuzzy and rough extension of the PROMETHEE II, TOPSIS and VIKOR methods, indicator importance degree determining method by entropy and indicator value determining method by accurate expert scoring.

  • Influence of O-O formation pathways and charge transfer mediator on lipid bilayer membrane-like photoanodes for water oxidation

    • Inspired by the function of crucial components in photosystem II (PSII), electrochemical and dyesensitized photoelectrochemical (DSPEC) water oxidation devices were constructed by the selfassembly of well-designed amphipathic Ru(bda)-based catalysts (bda = 2,2'-bipyrdine-6,6'-dicarbonoxyl acid) and aliphatic chain decorated electrode surfaces, forming lipid bilayer membrane (LBM)-like structures. The Ru(bda) catalysts on electrode-supported LBM films demonstrated remarkable water oxidation performance with different O-O formation mechanisms. However, compared to the slow charge transfer process, the O-O formation pathways did not determine the PEC water oxidation efficiency of the dyesensitized photoanodes, and the different reaction rates for similar catalysts with different catalytic paths did not determine the PEC performance of the DSPECs. Instead, charge transfer plays a decisive role in the PEC water oxidation rate. When an indolo[3,2-b] carbazole derivative was introduced between the Ru (bda) catalysts and aliphatic chain-modified photosensitizer in LBM films, serving as a charge transfer mediator for the tyrosine-histidine pair in PSII, the PEC water oxidation performance of the corresponding photoanodes was dramatically enhanced.

  • First Report on the Occurrence and Bioaccumulation of Hexafluoropropylene Oxide Trimer Acid : An Emerging Concern

    • Here, we report on the occurrence of a novel perfluoroalkyl ether carboxylic acid, ammonium perfluoro-2-[(propoxy)propoxy]-1-propanoate (HFPO-TA), in surface water and common carp (Cyprinus carpio) collected from the Xiaoqing River and in 'residents residing near a fluoropolymer production plant in Huantai County, China. Compared with the levels upstream of the Xiaoqing River, HFPO-TA concentrations (5200-68500 ng/L) were approximately 120-1600-times higher downstream after receiving fluoropolymer plant effluent from a tributary. The riverine discharge' of HFPO-TA was estimated to be 4.6 t/yr, accounting for 22% of total PFAS discharge. In the wild common carp collected downstream from the point source, HFPO-TA was detected in the blood (median: 1510 ng/mL), liver (587 ng/g ww), and muscle (118 ng/g ww). The log BCFbiood of HFPO-TA (2.18) was significantly higher than that of PFOA (1.93). Detectable levels of HFPO-TA were also found in the sera of residents (median: 2.93 ng/mL). This is the first report on the environmental occurrence and bioaccumulation of this novel chemical. Our results indicate an emerging usage of HFPO-TA in the fluoropolymer manufacturing industry and raise concerns about the toxicity and potential health risks of HFPO-TA to aquatic organisms and humans.

  • Worldwide Distribution of Novel Perfluoroether Carboxylic and Sulfonic Acids in Surface Water

    • Driven by increasingly stringent restrictions on long-chain per- and polyfluoroalkyl substances (PFASs), novel fluorinated compounds have emerged on the market. Here we report on the occurrences of several perfluoroalkyl ether carboxylic and sulfonic acids (PFECAs and PFESAs), including hexafluoropropylene oxide dimer and trimer acids (HFPO-DA and HFPO-TA), ammonium 4,8-dioxa-3 H-perfluorononanoate (ADONA), chlorinated polyfluorinated ether sulfonic acid (6:2 Cl-PFESA), and its hydrogen-substituted analogue (6:2 H-PFESA) in surface waters from China ( n = 106), the United States ( n = 12), the United Kingdom ( n = 6), Sweden ( n = 10), Germany ( n = 14), The Netherlands ( n = 6), and Korea ( n = 6). Results showed that HFPO-DA, HFPO-TA, and 6:2 Cl-PFESA (median = 0.95, 0.21, and 0.31 ng/L, respectively) were frequently detected in all countries, indicating ubiquitous dispersal and distribution in global surface waters. The presence of 6:2 H-PFESA was widely detected in China (detection rate > 95%) but not in any other country. Only trace levels of ADONA (0.013-1.5 ng/L) were detected in the Rhine River flowing through Germany. The estimated total riverine mass discharges of HFPO-DA, HFPO-TA, and ΣPFESAs reached 2.6, 6.0, and 4.3 ton/year in five of the major river systems in China. Our results indicated that novel PFECAs and PFESAs might become global contaminants, and future investigations are warranted.

  • Highly dispersed Cu atoms in MOF-derived N-doped porous carbon inducing Pt loads for superior oxygen reduction and hydrogen evolution

    • The preparation of oxygen reduction reaction (ORR) and hydrogen evolution reaction (HER) catalysts with high activity, stability and low platinum loading has always been the focus of research. The single-atom platinum supported catalyst greatly improves the utilization of platinum, but the catalytic activity and selectivity are greatly affected by the platinum coordination environment, and the preparation of the material is difficult. Doping base metals to adjust the electronic structure of platinum is an effective strategy to improve catalyst performance. In this work, copper-platinum alloy nanoparticles were loaded on N-doped porous carbon via a targeted route guided by highly dispersed Cu atoms derived from MOF. The product C-ZIF-CuPt has high activity and high stability ORR, HER bifunctional catalytic performance, which is better than commercial Pt/C (20 wt%). The Pt activity in C-ZIF-CuPt is 4.4 times (ORR) and 6.7 times (HER) than Pt/C. Spectromicroscopic determinations unveiled that strong interactions between carbon carrier and the CuPt alloys contribute to the overall stabilities. DFT calculations show that Cu doping can increase the d-band center of Pt, reduce the ORR overpotential, and the activation energy barrier to water molecules, which is beneficial to ORR and HER catalysis. Under the same carrier conditions, the performance of sub-nano (single atoms, clusters) platinum-supported catalysts (C-ZIF-Cu-Pt) is inferior to C-ZIF-CuPt. This further shows that platinum alloying can effectively improve the performance of the catalyst.
