    • Förvaltningsplanen är framtagen för att ge vägledning och verktyg till Länsstyrelsen,Viltförvaltningsdelegationen samt alla samhällsintressen och allmänhet som berörs av förekomst avde stora rovdjuren i Gävleborgs län. Planen ska ge förutsättningar för en förutsägbar förvaltningmed tydliga mål och riktlinjer samt vilka verktyg som finns för att nå målen: information ochkommunikation, skadeförebyggande åtgärder, akuta åtgärder vid rovdjursangrepp, hantering avnärgångna rovdjur, användning av hundar, skyddsjakt, licensjakt samt en adaptiv viltförvaltning.Bland verktygen återfinns också förebyggande av illegal jakt och artskyddsbrott, forskning ochkunskapsuppbyggnad samt åtgärder för förbättring av genetisk status hos varg.Förhoppningen är att de konflikter och motsättningar som finns i synen på de stora rovdjuren skaförebyggas och minska genom öppenhet och dialog med allmänhet och företrädare för berördanäringar och intressen.Här beskrivs Länsstyrelsens mål för rovdjursförvaltningen i Gävleborg. Arbetet styrs avövergripande mål, populationsmål samt mål kopplade till rovdjurens förekomst. Länsstyrelsen harockså fastställt riktlinjer som stöd i arbetet med att uppnå målen.I förvaltningsplanen har följande övergripande mål fastställts: Länets rovdjurspopulationer ska bidra till att uppnå och bibehålla gynnsambevarandestatus för arterna ur ett nationellt perspektiv. För de arter där miniminivåer harbeslutats innebär detta att dessa med säkerhet inte får underskridas och att beslutadförvaltningsnivå är målet. Länets tamdjurshållning som näring ska inte påtagligt försvåras eller omöjliggöras till följdav rovdjursförekomst. Förvaltningen av de stora rovdjuren i länet ska ta hänsyn till ekonomiska, sociala ochkulturella omständigheter, både på regional nivå och lokal nivå. Det ska finnas ett regionalt förtroende för förvaltningen av stora rovdjur.Specifika mål har även upprättats för följande delområden: Populationsmål för rovdjurspopulationerna inom länet Förebyggande av skador orsakade av stora rovdjur på tamdjur, gröda och egendom samthundar Förebyggande av rovdjursangrepp på människor Ersättning för viltskador orsakade av rovdjur Illegal jakt och artskyddsbrott Rovdjur som resurs Dynamiken med andra arter Människors attityder till rovdjur Rovdjurens påverkan på människors livskvalitet Förtroendet för rovdjursförvaltningenPlanen har utarbetats av Länsstyrelsen tillsammans med en arbetsgrupp med ledamöter frånGävleborgs viltförvaltningsdelegation, och godkändes slutgiltigt av Viltförvaltningsdelegationen2017-05-19.

  • Inventering av stora rovdjur i Gävleborgs län 2014-2015

    •  Länsstyrelsen ansvarar för att årliga rovdjursinventeringar genomförs i Gävleborgs län. Syftet är attövervaka rovdjursstammarnas storlek, utbredning, genetiska status och hur stammarna utvecklasöver tiden. I den här rapporten redovisas resultaten från inventeringar av lodjur, varg, järv och kungsörn, samtlänsstyrelsens bedömning av förekomsten av björn i Gävleborg under inventeringssäsongen2014/2015. Vid lodjursinventeringen 2014/2015 kvalitetssäkrades 15 familjegrupper, det vill säga 15 honor somfött ungar 2014. Lodjursstammen i Gävleborg kan därmed uppskattas till cirka 83 individer vintern2014/2015. Fyra av familjegrupperna delades med angränsande län. Gävleborg berördes av tio vargrevir vintern 2014/2015: Prästskogen/Haverö, Naggen, Kukumäki,Sjösveden, Tandsjön, Lingbo, Åmot/Ockelbo, Bondskog, Björnås och Glamsen. I sju av revirenbekräftades att föryngring skett våren 2014. Sex av reviren delades med andra län. Det innebär attGävleborg anses ha haft 4,7 familjegrupper vintern 2014/2015, enligt Viltskadecenters fördelning.Vargarna i Korsåreviret, som tidigare delades mellan Gävleborg och Dalarna, har denna säsong vistatsväster om länsgränsen och har inte spårats i Gävleborg. Tandsjö-reviret försvann underinventeringssäsongen genom skyddsjakt i februari 2015. Ingen inventering av björn har genomförts denna säsong. Länsstyrelsen bedömer att björnstammensstorlek och spridning i länet sannolikt överensstämmer med resultatet från 2012 års spillningsinventering. Björnstammen uppskattades då till cirka 380 individer i Gävleborgs län. En riktad järvinventering har för första gången på många år genomförts i Gävleborgs län vintern2014/2015. Med hjälp av fotostationer har bilder på järv erhållits på flera ställen i hela norra delen avGävleborg och en föryngring 2014 har kvalitetssäkrats. Vid inventeringen av kungsörn i Gävleborg 2014 konstaterades totalt 25 revir, varav 8 hade lyckadehäckningar och 3 av dessa var dubbelkullar, det vill säga kullar med två ungar. Storleken på länetsfasta kungsörnspopulation 2014 uppskattas till 50-65 individer. Inventeringarna av lo, järv och varg utfördes av länsstyrelsens personal, medan kungsörninventerades av Kungsörnsgruppen Gävleborg på uppdrag av länsstyrelsen. Allmänhetensrapportering av rovdjursobservationer har varit till stor hjälp i arbetet med att göra så brainventeringar som möjligt utifrån de resurser som finns.

    • In this essay, an interdisciplinary group of researchers sets out to address the period 1780–1840 in Sweden in a new way, by placing nature at its centre. With the help of ecocritical and transcultural theory, combined with renewed attention to the Swedish fine arts, learned discourses, and practices, we suggest a new approach to these revolutionary decades. The perceived dissonance, the interplay between climatic conditions and cultural template in early modern and modern Sweden, has not been fully addressed in current research, despite the fact that the relationship between humankind and the environment is a central issue in contemporary society and scholarship. Representations of nature situate the nation, they negotiate the relationship between a sensed reality and an ideal, between human and more-than-human beings. We suggest a focus on the unpredictable space created by negotiations of nature in Swedish representations during this crucial period, and, furthermore, on the ways in which this creative space is charged with utopian possibilities in the early Anthropocene. This is the background and the driving force of the planned research project ‘Cool Nature: Utopian Landscapes in Sweden 1780–1840’.

    • In order to elucidate the impact of bentonite on the process of radiation induced oxidative dissolution of UO2 in an aqueous system, the dissolution of U(VI) and consumption of H2O2 over time has been studied. In addition, γ-irradiation experiments were performed to study a more relevant and complex system, serving as a comparison with the previously stated system. In both cases, the experiments revealed that the presence of bentonite in water could either delay or prevent in part the release of uranium to the environment. The cause is mainly attributed to the scavenging of radiolytic oxidants rather than to the adsorption of uranium onto bentonite.

  • Temozolomide as monotherapy is effective in treatment of advanced malignant neuroendocrine tumors

    • Purpose: A retrospective analysis of the toxicity and efficacy of temozolomide in advanced neuroendocrine tumors. Experimental Design: Thirty-six patients with advanced stages of neuroendocrine tumor (1 gastric, 7 thymic and 13 bronchial carcinoids, 12 pancreatic endocrine tumors, 1 paraganglioma, 1 neuroendocrine foregut, and 1 neuroendocrine cecal cancer) were treated with temozolomide (200 mg/m2) for 5 days every 4 weeks. Patients had previously received a mean of 2.4 antitumoral medical regimens. Tumor response was evaluated radiologically according to the Response Evaluation Criteria in Solid Tumors every 3 months on an intent-to-treat basis. The circulating tumor marker plasma chromogranin A was also assessed. The expression of 06-methylguanine DNA methyltransferase, an enzyme implicated in chemotherapy resistance, was studied by immunohistochemistry (n = 23) and compared with response to temozolomide. Results: Median overall time to progression was 7 months (95% confidence interval, 3-10). Radiologic response was seen in 14% of patients and stable disease in 53%. Side effects were mainly hematologic; 14% experienced grade 3 or 4 thrombocytopenia (National Cancer Institute toxicity criteria). Ten patients had tumors with 06-methylguanine DNA methyltransferase immunoreactivity in <10% of nuclei, whereas four patients showed radiologic responses. Conclusions: Temozolomide as monotherapy had acceptable toxicity and antitumoral effects in a small series of patients with advanced malignant neuroendocrine tumors and four of these showed radiologic responses.

  • The Relationship between Health-Related Quality of Life, Subjective Scar Estimation, and Activity Performance in Adult Burn Patients 6 and 12 Months after Injury

    • A burn injury affects a person’s health-related quality of life (HRQoL) in different ways and might influence their daily life for months and years afterward. The aim of this study was to examine how activity performance and subjective scar estimation relate to self-rated health and whether this changes in the first year post-burn. Fifty consecutive patients who were scheduled for follow-up at the Burn Center’s outpatient clinic in Uppsala were included. Assessments of HRQoL (EQ-5D), activity performance (DASH), and subjective scar evaluation (POSAS) were conducted at 6 and 12 months post-burn. The results show a statistically significant correlation between self-rated HRQoL and activity performance (p = 0.001) and between self-rated HRQoL and subjective scar estimation (p = 0.000) at 6 but not at 12 months post-burn. A possible explanation of the lack of correlation at one year post-burn might be the patient´s expectations of his or her recovery. In future research, it would be interesting to investigate the long-term correlations between quality of life and activity performance.

    • INTRODUCTION: Atrial fibrillation (AF) is the most common arrhythmia and confers an increased risk of mortality, stroke, heart failure and cognitive decline. There is growing interest in AF screening; however, the most suitable population and device for AF detection remains to be elucidated. Here, we present the design of the CONSIDERING-AF (deteCtiON and Stroke preventIon by moDEl scRreenING for Atrial Fibrillation) study. METHODS AND ANALYSIS: CONSIDERING-AF is a randomised, controlled, siteless, non-blinded diagnostic superiority trial with four parallel groups and a primary endpoint of identifying AF during a 6-month study period set in Region Halland, Sweden. In each group, 740 individuals aged≥65 years will be included. The primary objective is to compare the intervention of AF screening enrichment using a risk prediction model (RPM), followed by 14 days of a continuous ECG patch, with no intervention (standard care). Primary outcome is defined as the incident AF recorded in the Region Halland Information Database after 6 months as compared with standard care. Secondary endpoints include the difference in incident AF between groups enriched or not by the RPM, with and without an invitation to 14 days of continuous ECG recording, and the proportions of oral anticoagulation treatment in the four groups. ETHICS AND DISSEMINATION: This study has ethical approval from the Swedish Ethical Review Authority. Results will be published in peer-reviewed international journals.     • Background: AdCD40L is an immunostimulatory gene therapy under evaluation for advanced melanoma, including ocular melanoma. Herein, we present the final data of a Phase I/IIa trial using AdCD40L alone or in combination with low dose cyclophosphamide +/- radiation therapy.Methods: AdCD40L is a replication-deficient adenovirus carrying the gene for CD40 ligand (CD40L). Twenty-four patients with advanced melanoma were enrolled and treated with AdCD40L monotherapy, or combined with cyclophosphamide +/- single fraction radiotherapy. The patients were monitored for 10 weeks using immunological and radiological evaluations and thereafter for survival.Results: AdCD40L treatment was safe and well tolerated both alone and in combination with cyclophosphamide as well as local radiotherapy. Four out of twenty-four patients had >1 year survival. Addition of cyclophosphamide was beneficial but adding radiotherapy did not further extend survival. High initial plasma levels of IL12 and MIP3b correlated to overall survival, whereas IL8 responses post-treatment correlated negatively with survival. Interestingly, antibody reactions to the virus correlated negatively with post IL6 and pre IL1b levels in blood.Conclusions: AdCD40L was safely administered to patients and effect was improved by cyclophosphamide but not by radiotherapy. Immune activation profile at baseline may predict responders better than shortly after treatment.

    • Aquatic chemical ecology is an important and growing field of research that involves understanding how organisms perceive and respond to chemical cues in their environment. Research assessing the preference or avoidance of a water source containing specific chemical cues has increased in popularity in recent years, and a variety of methods have been described in the scientific literature. Two-current choice flumes have seen the greatest increase in popularity, perhaps because of their potential to address the broadest range of research questions. Here, we review the literature on two-current choice flumes and show that there is a clear absence of standardized methodologies that make comparisons across studies difficult. Some of the main issues include turbulent flows that cause mixing of cues, inappropriate size of choice arenas for the animals, short experiments with stressed animals, failure to report how experiment and researcher biases were eliminated, general underreporting of methodological details, underutilization of collected data and inappropriate data analyses. In this review, we present best practice guidelines on how to build, test and use two-current choice flumes to measure the behavioural responses of aquatic animals to chemical cues, and provide blueprints for flume construction. The guidelines include steps that can be taken to avoid problems commonly encountered when using two-current choice flumes and analysing the resulting data. This review provides a set of standards that should be followed to ensure data quality, transparency and replicability in future studies in this field.

    • The study is a scoping review of 80 research articles in LIS and related fields (2004-2014) on the use of social network sites by researchers. The results show that social network sites are used as part of scholarly life, yet with disciplinary differences. It is also shown that the area lacks methodological, theoretical and empirical coherence and theoretical stringency. The most salient strands of research (General uptake, Outreach, Special tools/cases, Assessing impact, Practices/new modes of communication) are mapped and ways to improve research in the field are identified. This provides a first step towards a more comprehensive understanding of the roles of social network sites in scholarship.

    • BACKGROUND: Current approaches for treating metastatic malignant melanoma (MM) are not effective enough and are associated with serious adverse events. Due to its immunogenicity, melanoma is an attractive target for immunostimulating therapy. In this phase I/IIa study, local AdCD40L immunostimulatory gene therapy was evaluated in patients with MM.METHODS: AdCD40L is an adenovirus carrying the gene for CD40 ligand. Patients that failed standard treatments were enrolled. Six patients received four weekly intratumoral AdCD40L injections. Next, nine patients received low-dose cyclophosphamide conditioning before the first and fourth AdCD40L injection. The blood samples were collected at multiple time points for chemistry, haematology and immunology evaluations. Radiology was performed at enrolment and repeated twice after the treatment.RESULTS: AdCD40L was safe with mild transient reactions. No objective responses were recorded by MRI, however, local and distant responses were seen on FDG-PET. The overall survival at 6 months was significantly better when cyclophosphamide was added to AdCD40L. The patients with the best survival developed the highest levels of activated T cells and experienced a pronounced decrease of intratumoral IL8.CONCLUSIONS: AdCD40L therapy for MM was well tolerated. Local and distant responses along with better survival in the low-dose cyclophosphamide group are encouraging.

    • Carbon-fiber-reinforced plastic (CFRP) is one of the most commonly used materials in the aerospace industry today. CFRP in pre-impregnated form is an anisotropic material whose properties can be controlled to a high level by the designer. Sometimes, these properties make the material hard to predict with regards to how the geometry affects manufacturing aspects. This chapter describes 11 design rules that describe geometrical design choices and deals with manufacturability problems that are connected to them, why they are connected, and how they can be minimized or avoided. Examples of design choices dealt with in the rules include double curvature shapes, assembly of uncured CFRP components, and access for nondestructive testing.

    • Carbon ber-reinforced plastic (CFRP) is one of the most commonly used materials in the aerospace industry today. CFRP in pre- impregnated form is an anisotropic material whose properties can be controlled to a high level by the designer. Sometimes, these properties make the material hard to predict with regards to how the geometry affects manufacturing aspects. This paper describes eleven design rules originating from different guidelines that describe geometrical design choices and deals with manufacturability problems that are connected to them, why they are connected and how they can be minimized or avoided. Examples of design choices dealt with in the rules include double curvature shapes, assembly of uncured CFRP components and access for non-destructive testing (NDT). To verify the technical content and ensure practicability, the rules were developed by, inter alia, studying literature and performing case studies at SAAB Aerostructures. The research was done through a collaboration between Linköping University and SAAB Aerostructures in a state-funded project. This ensured a balanced approach between academic advancement and usefulness in commercial projects.

    • What a company offers its customers has to fulfil several different needs, desires, constraints, which can originate from multiple different sources that affect the offering throughout its life cycle. All these criteria have to come together and be translated into statements that can support the designer’s understanding of the offering’s purpose. This translation is done through a requirements development process to provide a controlled process to define statements that describe what the offering is supposed to fulfil.This research provides insights on key challenges and success factors in requirements development to support the design of effective and resource-efficient offerings. Namely, it identifies crucial sources and aspects to be considered, and a requirements development process demonstrating how to overcome identified challenges. By getting the requirements right from the beginning, sub-optimisation and unnecessary time and risks can be avoided. The consideration of accurate sources and aspects is considered to be one of the most important factors for the successful design of offerings. It is also in the earliest phases of design, that is to say requirements development, where one has the greatest possibility to affect the environmental impact of the offering. What is missing, however, is sufficient and appropriate support in industry on how to do so.The gap between the three areas of effectiveness and resource efficiency, design of integrated offerings, and requirements development has been investigated. Results are based on findings in the literature and in industry, identified primarily by qualitative studies. In the research, 15 different companies have been included through a number of interviews and discussions.Key sources and aspects to consider in the requirements development process are identified along with challenges, and success factors that can be utilised to overcome the identified challenges. This research’s final results include an adapted requirements development process that considers the earlier-mentioned sources and aspect, challenges, and success factors. Such a requirements development process should support the design of effective and resource-efficient offerings.

    • The objective of this paper is to explore what challenges exist when setting requirements for an Integrated Product Service Offering (IPSO). An IPSO, sometimes called Product Service System, is a concept with increased interest from manufacturing companies. It consists of a combination of products and services that, based on a life cycle perspective, have been integrated to fit targeted customer needs. In order to achieve a successful IPSO, it is important to collect aspects from many actors, something which sometimes is challenging for companies moving towards providing IPSOs.The four challenges found when setting requirements in IPSO development are; identification and inclusion of relevant aspects from relevant actors throughout the IPSO’s life cycle, understanding of the underlying aspects for all requirements for all elements of the offering, prioritization of requirements, and the difficulty to track how requirements affect each other between different elements in the IPSO.The methodology used to find these challenges was a combination of a structured literature review and an interview study at three manufacturing companies moving towards providing IPSOs.

  • Inventering av stora rovdjur i Gävleborgs län 2015-2016

    • Länsstyrelsen ansvarar för att årliga rovdjursinventeringar genomförs i Gävleborgs län. Syftet är attövervaka rovdjursstammarnas storlek, utbredning, genetiska status och hur stammarna utvecklasöver tiden. I den här rapporten redovisas resultaten från inventeringar av lodjur, varg och kungsörn, samtlänsstyrelsens bedömning av förekomsten av björn och järv i Gävleborg inventeringssäsongen2015/2016.Vid lodjursinventeringen 2015/2016 kvalitetssäkrades 19,5 länsegna familjegrupper, det vill sägahonor som fött ungar 2015. Fem av familjegrupperna delades med angränsande län.Lodjursstammen som berör Gävleborg kan därmed uppskattas till cirka 110 individer vintern2015/2016. Gävleborg berördes av 14 vargrevir vintern 2015/2016. 11 av reviren bestod av familjegrupper ellerrevirmarkerande par: Prästskogen, Kukumäki, Sjösveden, Lingbo, Åmot/Ockelbo, Björnås, Blyberget,Hålnan, Lortglo, Krokvattnet och Glamsen. I åtta av reviren bekräftades att föryngring skett våren2015. Sju av reviren delades med andra län. Utöver dessa elva revir har tre ensamma stationäravargar spårats. Dessa är inte DNA-profilerade men kvalitetsäkrade som stationära vargar enligtinventeringsföreskrifterna. Vid inventeringen av kungsörn i Gävleborg 2015 konstaterades totalt 23 intakta revir med 14häckande par, varav 8 hade lyckade häckningar. Storleken på länets fasta kungsörnspopulation 2015uppskattas till drygt 50 individer. Ingen inventering av björn har genomförts denna säsong. Länsstyrelsen bedömer att björnstammensstorlek i länet minskat något sedan den senaste inventeringen som genomfördes 2012.Björnstammen upp-skattades då till cirka 380 individer i Gävleborgs län. Järvinventeringen blev bristfällig vintern 2015/2016. Huvudsyftet med inventeringen är att fastställaantalet föryngringar som skett inom länet. Järvarna föder sina ungar i februari och honan lämnar sinlya efter någon vecka med ungarna. Snön försvann redan i början på mars och förutsättningarna attspåra en hona tillbaka till lyan försvann med snön. Länsstyrelsen bedömer att det sker mellan 4-8föryngringar inom Gävleborgs län. Spår av järv blir allt vanligare söderut i länet. Länets järvpopulationär som tätast i de norra och nordvästra delarna. Inventeringarna av lo, järv och varg utfördes av länsstyrelsens personal, medan kungsörninventerades av Kungsörnsgruppen Gävleborg på uppdrag av länsstyrelsen. Allmänhetensrapportering av rovdjursobservationer har varit till stor hjälp i arbetet med att göra så brainventeringar som möjligt utifrån de resurser som finns. Rapportering av rovdjursobservationer görs till skandobs.se eller till mobilapplikationen Skandobstouch, som är gratis att ladda ner.

  • Individualization of Hematopoietic Stem Cell Transplantation Using Alpha/Beta T-Cell Depletion

    • Allogeneic hematopoietic stem cell transplantation (HSCT) is associated with several potentially lethal complications. Higher levels of CD3+ T-cells in the graft have been associated with increased risk of graft-versus-host disease (GVHD), but also beneficial graft-versus-leukemia effect and reduced infections. To tackle post-transplant complications, donor lymphocyte infusions have been used but with an increased risk of GVHD. To reduce this risk, we performed depletion of alpha beta T-cells and treated 12 patients post-HSCT suffering from infections and/or poor immune reconstitution. The alpha beta T-cell depleted cell products were characterized by flow cytometry. The median log depletion of alpha beta T-cells was -4.3 and the median yield of gamma delta T-cells was 73.5%. The median CD34+ cell dose was 4.4 x 10(6)/kg. All 12 patients were alive 3 months after infusion and after 1 year, two patients had died. No infusion-related side effects were reported and no severe acute GVHD (grade III-IV) developed in any patient post-infusion. Overall, 3 months after infusion 11 out of 12 patients had increased levels of platelets and/or granulocytes. In conclusion, we describe the use of alpha beta T-cell depleted products as stem cell boosters with encouraging results.

    • Purpose[11C]Metomidate positron emission tomography (PET) is currently used for staging of adrenocortical carcinoma and for lateralization in primary aldosteronism (PA). Due to the short half-life of carbon-11 and a high non-specific liver uptake of [11C]metomidate there is a need for improved adrenal imaging methods. In a previous pre-clinical study para-chloro-2-[18F]fluoroethyletomidate has been proven to be a specific adrenal tracer. The objective is to perform a first evaluation of para-chloro-2-[18F]fluoroethyletomidate positron emission computed tomography ([18F]CETO-PET/CT) in patients with adrenal tumours and healthy volunteers.MethodsFifteen patients underwent [18F]CETO-PET/CT. Five healthy volunteers were recruited for test-retest analysis and three out of the five underwent additional [15O]water PET/CT to measure adrenal blood flow. Arterial blood sampling and tracer metabolite analysis was performed. The kinetics of [18F]CETO were assessed and simplified quantitative methods were validated by comparison to outcome measures of tracer kinetic analysis.ResultsUptake of [18F]CETO was low in the liver and high in adrenals. Initial metabolization was rapid, followed by a plateau. The kinetics of [18F]CETO in healthy adrenals and all adrenal pathologies, except for adrenocortical carcinoma, were best described by an irreversible single-tissue compartment model. Standardized uptake values (SUV) correlated well with the uptake rate constant K1. Both K1 and SUV were highly correlated to adrenal blood flow in healthy controls. Repeatability coefficients of K1, SUV65–70, and SUV120 were 25, 22, and 17%.ConclusionsHigh adrenal uptake combined with a low unspecific liver uptake suggests that 18F]CETO is a suitable tracer for adrenal imaging.   • Radiation dosimetry of para-chloro-2-[18F]fluoroethyl-etomidate:a PET tracer for adrenocortical imaging

    • Introduction[11C]metomidate, a methyl ester analogue of etomidate, is used for positron emission tomography of adrenocortical cancer, and has been tested in recent clinical trials for lateralization in primary aldosteronism (PA). However, in PA, visualization as well as uptake quantification are hampered by the tracer’s rather high non-specific liver uptake, and its overall clinical usefulness is also limited by the short 20-minute half-life of carbon-11. Therefore, we evaluated para-chloro-2-[18F]fluoroethyl-etomidate, [18F]CETO, a fluorine-18 (T1/2=109.8 min) analogue, as a potential new adrenocortical PET tracer.ObjectivesThe aim of this study was to assess in vivo and in-human radiation dosimetry of [18F]CETO.Methods: Residence times were calculated based on uptake data from rats (n=30, biodistribution study with ex vivo measurements) as well as in vivo PET/CT in cynomolgus (n=1) and humans (n=9). OLINDA 1.1 was used to obtain absorbed doses in human organs (mGy/MBq) and effective dose (mSv/MBq).Results[18F]CETO showed a high uptake in human adrenal glands, still increasing at 90 minutes post injection. Regardless of species used for input data (rat, cynomolgus or human), adrenal glands (absorbed dose 0.093 ± 0.038 mGy/MBq based on human data) were confirmed as the dose-limiting organs. The effective dose based on human data was 18.2 μSv/MBq and varied little when using rat (18.4 μSv/MBq) or cynomolgus data (16.1 μSv/MBq).  Conclusions[18F]CETO has a favourable biodistribution in humans for adrenal imaging. The effective dose for a typical clinical PET/CT examination with 200 MBq [18F]CETO  is 3.6 mSv.

    • The Haemophilus surface fibril (Hsf) is an extraordinary large (2413 amino acids) trimeric autotransporter, present in all encapsulated Haemophilus influenzae. It contributes to virulence by directly functioning as an adhesin. Furthermore, Hsf recruits the host factor vitronectin thereby inhibiting the host innate immune response resulting in enhanced survival in serum. Here we observed by electron microscopy that Hsf appears as an 100. nm long fibril at the bacterial surface albeit the length is approximately 200. nm according to a bioinformatics based model. To unveil this discrepancy, we denaturated Hsf at the surface of Hib by using guanidine hydrochloride (GuHCl). Partial denaturation induced in the presence of GuHCl unfolded the Hsf molecules, and resulted in an increased length of fibres in comparison to the native trimeric form. Importantly, our findings were also verified by E. coli expressing Hsf at its surface. In addition, a set of Hsf-specific peptide antibodies also indicated that the N-terminal of Hsf is located near the C-terminal at the base of the fibril. Taken together, our results demonstrated that Hsf is not a straight molecule but is folded and doubled over. This is the first report that provides the unique structural features of the trimeric autotransporter Hsf.

  • Identification of small molecules blocking the Pseudomonas aeruginosa type III secretion system protein PcrV

    • Pseudomonas aeruginosa is an opportunistic bacterial pathogen that employs its type III secretion system (T3SS) during the acute phase of infection to translocate cytotoxins into the host cell cytoplasm to evade the immune system. The PcrV protein is located at the tip of the T3SS, facilitates the integration of pore-forming proteins into the eukaryotic cell membrane, and is required for translocation of cytotoxins into the host cell. In this study, we used surface plasmon resonance screening to identify small molecule binders of PcrV. A follow-up structure-activity relationship analysis resulted in PcrV binders that protect macrophages in a P. aeruginosa cell-based infection assay. Treatment of P. aeruginosa infections is challenging due to acquired, intrinsic, and adaptive resistance in addition to a broad arsenal of virulence systems such as the T3SS. Virulence blocking molecules targeting PcrV constitute valuable starting points for development of next generation antibacterials to treat infections caused by P. aeruginosa.

    • PurposeThe purpose of this paper is to understand how meaning is assigned to online searching by viewing it as a mundane, yet often invisible, activity of everyday life and an integrated part of various social practices.Design/methodology/approachSearching is investigated with a sociomaterial approach with a starting point in information searching as entangled across practices and material arrangements and as a mundane part of everyday life. In total, 21 focus groups with 127 participants have been carried out. The study focusses particularly on peoples’ experiences and meaning-making and on how these experiences and the making of meaning could be understood in the light of algorithmic shaping.FindingsAn often-invisible activity such as searching is made visible with the help of focus group discussions. An understanding of the relationship between searching and everyday life through two interrelated narratives is proposed: a search-ification of everyday life and a mundane-ification of search.Originality/valueThe study broadens the often narrow focus on searching in order to open up for a research-based discussion in information science on the role of online searching in society and everyday life.

    • Understanding UO2 matrix dissolution is of major importance for the safety assessment of a future deep geological repository. Oxidative dissolution of UO2 and SIMFUEL pellets have been extensively studied in HCO3- solutions, while less is known about systems with no or very low HCO3- concentrations. The aim of this work is to elucidate the oxidative dissolution of UO2 and SIMFUEL pellets in HCO3- free solutions by studying the dissolution of U (VI) and consumption of H2O2 over time. The results are compared with previous experiments performed in HCO3- solutions. The oxidative dissolution rate is higher for the UO2 pellet in HCO3- compared to the other systems. It is evident that the kinetics of the reaction with H2O2 is qualitatively different for SIMFUEL in comparison with pure UO2. For the UO2 pellet in pure water, the presence of a secondary phase (meta) studtite, on the surface of the pellet is confirmed by Raman spectroscopy. The kinetic impact of the secondary phase is evaluated in separate UO2 powder experiments. The (meta) studtite (surface) precipitation leads to a slower release of uranium into the solution. Numerical simulations using experimentally determined rate constants are used to evaluate a simple mechanism of surface precipitation. The numerical results are in fair agreement with the experimental observations given certain criteria. In addition, the γ-radiation induced dissolution of UO 2 and SIMFUEL pellets were investigated in pure water, and compared with HCO3- systems. Also here the dissolution rate of uranium is higher for UO2 in HCO3- compared to pure water, while for SIMFUEL longer irradiation times are needed to observe any difference between pure and HCO3- containing water.

    • Naturvårdsbränning är en skötselåtgärd som syftar till att återskapa den ursprungliga dynamiken i skogen eftersom brand haft en omfattande ekologisk roll innan brandbekämpningenseffektiviserande. De senaste 100 åren har bränning av skog varit mer eller mindre tillämpat i skogsbruket, då främst för att främja återväxt på hyggen. Sedan början på 1990-talet har branden på allvar fungerat som en åtgärd för att gagna tall- och lövstrukturer samt brandgynnade arter.År 2006 genomförde länsstyrelsen i Gävleborg sina första naturvårdsbränningar, det hann bli tre stycken under sommaren. Två av bränningarna finansierades och genomfördesav Åtgärdsprogram för hotade arter och den tredje av Skötsel, båda funktioner på dåvarande Naturenheten och samtliga tre skedde på reservatsmark i Gävleborgs län. Denna rapport är en sammanställning av separata rapporter för de tre bränningarna skrivna av Andreas Wedman (Hagåsen och Bursjöberget) och Carola Orrmalm (Stora Korpimäki)Praktiskt enkla objekt valdes, med små arealer och säkra avgränsningar. Detta kriterium var viktigt eftersom bränning är en riskverksamhet som aldrig tidigare skett i länsstyrelsensegen regi. Ett annat kriterium som avgjorde var att objekten bedömdes strukturmässigtlämpliga för naturvårdsbränning ur ekologiperspektiv, dvs. objekt med föreligganderisk för igenväxning av gran samt objekt med önskad tallföryngring och tidigare brandpåverkan. Länsstyrelsen har sedan ett par år tillbaka arbetat med en brandstrategi som pekar ut lämpliga trakter, som även togs i beaktande. En exkursion med Anders Granström från SLU låg till slutgiltig grund för valet av två av objekten, Hagåsen och Bursjöberget. Det tredje objektet Stora Korpimäki var sedan tidigare utpekat som passande.Samtliga tre reservats skötselplaner anger bränning som skötselåtgärd och detta utgjorde ett ytterligare skäl till att just dessa objekt valdes.Rapporten som följer behandlar bränningarnas genomförande samt de initiala resultaten. En mer systematisk och grundlig uppföljning av mortalitet, återväxt, skapande av substratsamt en insektsinventering kommer att följa upp denna rapport i ett senare skede.

  • Spontana eller anlagda bränder : Vilka ger mest naturvårdsnytta? En studie av Gävleborgs län

    • Skogsbrandens omfattande påverkan på den boreala skogens ekologi har sedan länge varitkänd. Branden är den naturliga störning som haft störst betydelse för dynamiken i skogen. Enbrand medför att naturligt konkurrenssvaga arter får chans att etablera sig och bidrar därmedtill en större diversitet. Brandens omfattning och intensitet avgör hur pass stor störningen blir.Denna studie jämförde anlagda naturvårdsbränder med spontana bränder med avseende påvärdefulla naturvårdsmässiga egenskaper. Parametrar som jämfördes var bland annat antalbranddödade och sotade träd, sothöjd, branddjup, vegetationstyp och ungefärlig areal.Dessutom dokumenterades brandfälten med kamera och kartlades i ArcGIS för att det ska gålätt att återfinna dem i framtiden.Resultatet visade att anlagda bränder i de flesta avseenden har större naturvärden än spontanabränder. De förekom i regel på områden där diversiteten var större och träden äldre och de varstörre till ytan. De spontant uppkomna brandytorna visade sig vara små, ha färre brända trädoch vara svagare brända med ett antal undantag. Sothöjden var ungefär dubbelt så hög vidanlagda som vid spontana bränder. Dock fanns ingen signifikant skillnad mellan anlagda ochspontana bränder med avseende på antalet branddödade träd. Tidsmässigt uppträder anlagdabränder mestadels i början och slutet av brandsäsongen, medan spontana bränder förekommermer utspritt över året och har sin topp under högsommaren.En slutsats är att de anlagda naturvårdsbränderna är avgörande för att skapa värdefullabrandsubstrat. De spontana bränderna är för små och lågintensiva för att kunna bli värdefullaför brandgynnade arter och för att kunna skapa nya successioner. Om de brandgynnadearterna och hela skogens mångfald ska bevaras, måste naturvårdsbränningarna fortskrida ochfungera som en avgörande faktor i skogens framväxt. Människan har nästan elimineratskogselden och måste därför aktivt verka för att få den tillbaka som en ekologisk störning.

    • In this work we have studied the influence of H2S on radiation induced dissolution of spent nuclear fuel using simple model systems. The reaction between H2O2 and H2S/HS- has been studied experimentally as well as the effect of H2S/HS - on γ-radiation induced dissolution of a UO2 pellet. The experiments clearly show that the reaction of H2O 2 and H2S/HS- is fairly rapid and that H 2O2 and H2S/HS- stoichiometry is favorable for inhibition. Radiolysis experiments show that H2S/ HS- can effectively protect UO2 from oxidative dissolution. The effect depends on sulfide concentration in combination with dose rate. Autoclave experiments were also conducted to study the role of H 2S/HS- in the reduction of U(VI) in the presence and absence of H2 and Pd particles in anoxic aqueous solution. The aqueous solutions were pressurized with H2 or N2 and two different concentrations of H2S/HS- were used in the presence and absence of Pd. No catalytic effect of Pd on the U(VI) reduction by H2S/HS- could be found in N2 atmosphere. U(VI) reduction was found to be proportional to H2S/HS- concentration in H2 and N2 atmosphere. It is clearly shown the Pd catalyzed H2 effect is more powerful than the effect of H2S/HS-. H2S/HS- poisoning of the Pd catalyst is not observed under the present conditions.
