  • Effects of rosiglitazone on gene expression in subcutaneous adipose tissue in highly active antiretroviral therapy-associated lipodystrophy

    • Highly active antiretroviral therapy (HAART) has improved the prognosis of human immunodeficiency virus (HIV)-infected patients but is associated with severe adverse events, such as lipodystrophy and insulin resistance. Rosiglitazone did not increase subcutaneous fat in patients with HAART-associated lipodystrophy (HAL) in a randomized, double-blind, placebo-controlled trial, although it attenuated insulin resistance and decreased liver fat content. The aim of this study was to examine effects of rosiglitazone on gene expression in subcutaneous adipose tissue in 30 patients with HAL. The mRNA concentrations in subcutaneous adipose tissue were measured using real-time PCR. Twenty-four-week treatment with rosiglitazone (8 mg/day) compared with placebo significantly increased the expression of adiponectin, peroxisome proliferator-activated receptor-γ (PPARγ), and PPARγ coactivator 1 and decreased IL-6 expression. Expression of other genes involved in lipogenesis, fatty acid metabolism, or glucose transport, such as acyl-CoA synthase, adipocyte lipid-binding protein, CD45, fatty acid transport protein-1 and -4, GLUT1, GLUT4, keratinocyte lipid-binding protein, lipoprotein lipase, PPARδ, and sterol regulatory element-binding protein-1c, remained unchanged. Rosiglitazone also significantly increased serum adiponectin concentration. The change in serum adiponectin concentration was inversely correlated with the change in fasting serum insulin concentration and liver fat content. In conclusion, rosiglitazone induced significant changes in gene expression in subcutaneous adipose tissue and ameliorated insulin resistance in patients with HAL. Increased expression of adiponectin might have mediated most of the favorable insulin-sensitizing effects of rosiglitazone in these patients.

  • Disentangling the Evidence of Milankovitch Forcing From Tree-Ring and Sedimentary Records

    • Tree-ring records constitute excellent high-resolution data and provide valuable information for climate science and paleoclimatology. Tree-ring reconstructions of past temperature variations agree to show evidence for annual-to-centennial anomalies in past climate and place the industrial-era warming in the context of the late Holocene climate patterns and regimes. Despite their wide use in paleoclimate research, however, tree rings have also been deemed unsuitable as low-frequency indicators of past climate. The arising debate concerns whether the millennia-long tree-ring records show signals of orbital forcing due to the Milankovitch cycles. Here, we produce a summer-temperature reconstruction from tree-ring chronology running through mid- and late-Holocene times (since 5486 BCE) comprising minimum blue channel light intensity (BI). The BI reconstruction correlates with existing and new tree-ring chronologies built from maximum latewood density (MXD) and, unlike the MXD data, shows temperature trends on Milankovitch scales comparable to various types of sedimentary proxy across the circumpolar Arctic. Our results demonstrate an unrevealed potential of novel, unconventional tree-ring variables to contribute to geoscience and climate research by their capability to provide paleoclimate estimates from inter-annual scales up to those relevant to orbital forcing.

  • Future changes in the supply of goods and services from natural ecosystems : prospects for the European north

    • Humans depend on services provided by ecosystems, and how services are affected by climate change is increasingly studied. Few studies, however, address changes likely to affect services from seminatural ecosystems. We analyzed ecosystem goods and services in natural and seminatural systems, specifically how they are expected to change as a result of projected climate change during the 21st century. We selected terrestrial and freshwater systems in northernmost Europe, where climate is anticipated to change more than the global average, and identified likely changes in ecosystem services and their societal consequences. We did this by assembling experts from ecology, social science, and cultural geography in workshops, and we also performed a literature review. Results show that most ecosystem services are affected by multiple factors, often acting in opposite directions. Out of 14 services considered, 8 are expected to increase or remain relatively unchanged in supply, and 6 are expected to decrease. Although we do not predict collapse or disappearance of any of the investigated services, the effects of climate change in conjunction with potential economical and societal changes may exceed the adaptive capacity of societies. This may result in societal reorganization and changes in ways that ecosystems are used. Significant uncertainties and knowledge gaps in the forecast make specific conclusions about societal responses to safeguard human well-being questionable. Adapting to changes in ecosystem services will therefore require consideration of uncertainties and complexities in both social and ecological responses. The scenarios presented here provide a framework for future studies exploring such issues.

    • offers undergraduate courses in Cognitive Science. The studies are mainly organized as web-based courses or collaborative student projects. In the basic studies component, a student completes four one credit methodology courses and chooses a related assignment worth three credits for one of them. One of the methodology courses is entitled Networks in Learning Organization. It will be delivered entirely on the web, and it also will serve as a pilot course to help design other methodology courses. This paper will first briefly discuss the theoretical foundations of learning organizations and organizational learning. Then, we will review the role of networks as the technological foundation of a learning organization and continue by discussing the use of educational technology to support organizational learning and learn about it. Finally, we will describe the structure and the methods of the course and present topics that form the starting point for the discourse within the course.

  • Prognostic Role of Tumor Immune Microenvironment in Pleural Epithelioid Mesothelioma

    • Pleural mesothelioma (MPM) is an aggressive malignancy with an average patient survival of only 10 months. Interestingly, about 5%–10% of the patients survive remarkably longer. Prior studies have suggested that the tumor immune microenvironment (TIME) has potential prognostic value in MPM. We hypothesized that high-resolution single-cell spatial profiling of the TIME would make it possible to identify subpopulations of patients with long survival and identify immunophenotypes for the development of novel treatment strategies.MethodsWe used multiplexed fluorescence immunohistochemistry (mfIHC) and cell-based image analysis to define spatial TIME immunophenotypes in 69 patients with epithelioid MPM (20 patients surviving ≥ 36 months). Five mfIHC panels (altogether 21 antibodies) were used to classify tumor-associated stromal cells and different immune cell populations. Prognostic associations were evaluated using univariate and multivariable Cox regression, as well as combination risk models with area under receiver operating characteristic curve (AUROC) analyses.ResultsWe observed that type M2 pro-tumorigenic macrophages (CD163+pSTAT1−HLA-DRA1−) were independently associated with shorter survival, whereas granzyme B+ cells and CD11c+ cells were independently associated with longer survival. CD11c+ cells were the only immunophenotype increasing the AUROC (from 0.67 to 0.84) when added to clinical factors (age, gender, clinical stage, and grade).ConclusionHigh-resolution, deep profiling of TIME in MPM defined subgroups associated with both poor (M2 macrophages) and favorable (granzyme B/CD11c positivity) patient survival. CD11c positivity stood out as the most potential prognostic cell subtype adding prediction power to the clinical factors. These findings help to understand the critical determinants of TIME for risk and therapeutic stratification purposes in MPM.
