  • Toward an alternative approach for the preparation of low-temperature titanium dioxide blocking underlayers for perovskite solar cells

    • The anodic electrodeposition method is investigated as an alternative technique for the preparation of a titanium oxide (TiO 2 ) blocking underlayer (UL) for perovskite solar cells (PSCs). Extremely thin Ti IV -based films are grown from aqueous acidic titanium(iii) chloride in an electrochemical cell at room temperature. This precursor layer is converted to the UL (ED-UL), in a suitable state for PSC applications, by undertaking a sintering step at 450 °C for half an hour. PSCs with the composition of the light-absorbing material FA 0.85 MA 0.10 Cs 0.05 Pb(I 0.87 Br 0.13 ) 3 (FA and MA denote the formamidinium and methylammonium cations, respectively) based on the ED-UL are compared with PSCs with the UL of a standard type prepared by the spray-pyrolysis method at 450 °C from titanium diisopropoxide bis(acetylacetonate) (SP-UL). We obtain power conversion efficiencies (PCEs) of over 20% for mesoscopic perovskite devices employing both ED-ULs and SP-ULs. Slightly higher fill factor values are observed for ED-UL-based devices. In addition, ED-ULs prepared by the same method have also been applied in planar PSCs, resulting in a PCE exceeding 17%, which is comparable to that for similar PSCs with an SP-UL. The preparation of ED-ULs with a lower sintering temperature, 150 °C, has also been examined. The efficiency of a planar PSC incorporating this underlayer was 14%. These results point out to the possibility of applying ED-ULs in flexible planar PSCs in the future.

  • Life cycle assessment of pig slurry treatment technologies for nutrient redistribution in Denmark

    • Animal slurry management is associated with a range of impacts on fossil resource use and the environment. The impacts are greatest when large amounts of nutrient-rich slurry from livestock production cannot be adequately utilised on adjacent land. To facilitate nutrient redistribution, a range of different technologies are available. This study comprised a life cycle assessment of the environmental impacts from handling 1000 kg of pig slurry ex-animal. Application of untreated pig slurry onto adjacent land was compared with using four different treatment technologies to enable nutrient redistribution before land application: (a) separation by mechanical screw press, (b) screw press separation with composting of the solid fraction, (c) separation by decanter centrifuge, and (d) decanter centrifuge separation with ammonia stripping of the liquid fraction. Emissions were determined based on a combination of values derived from the literature and simulations with the Farm-N model for Danish agricultural and climatic conditions. The environmental impact categories assessed were climate change, freshwater eutrophication, marine eutrophication, terrestrial acidification, natural resource use, and soil carbon, nitrogen and phosphorus storage. In all separation scenarios, the liquid fraction was applied to land on the pig-producing (donor) farm and the solid fraction transported to a recipient farm and utilised for crop production. Separation, especially by centrifuge, was found to result in a lower environmental impact potential than application of untreated slurry to adjacent land. Composting and ammonia stripping either slightly increased or slightly decreased the environmental impact potential, depending on the impact category considered. The relative ranking of scenarios did not change after a sensitivity analysis in which coefficients for field emissions of nitrous oxide, ammonia and phosphorus were varied within the range cited in the literature. Therefore, the best technology to implement in a given situation depends on the environmental problem in question, local policy, cost and practicality.

    • Despite general agreement on the importance of adjusting each life cycle assessment (LCA) to its goal, the methodological choices in previously published LCAs on wastewater and sludge management systems are surprisingly similar, even when the information sought in the studies most likely differ. We argue that the potential of LCA may not currently be fully utilised, partly due to particular methodological challenges arising in both attributional and consequential LCAs for this type of systems. By developing the theory for handling of allocation problems in attributional LCAs, and by elaborating on the different possible foreseeable consequences in consequential LCA, we aim to facilitate both attributional and consequential LCAs, and to show the importance of this choice for a specific wastewater and sludge management system.

    • Changes in the membrane composition of sub-populations of cells can influence different properties with importance to tumour growth, metastasis and treatment efficacy. In this study, we use correlated fluorescence microscopy and ToF-SIMS with C-60(+) and (CO2)(6k)(+) ion beams to identify and characterise sub-populations of cells based on successful transfection leading to over-expression of CCT delta, a component of the multi-subunit molecular chaperone named chaperonin-containing tailless complex polypeptide 1 (CCT). CCT has been linked to increased cell growth and proliferation and is known to affect cell morphology but corresponding changes in lipid composition of the membrane have not been measured until now. Multivariate analysis of the surface mass spectra from single cells, focused on the intact lipid ions, indicates an enrichment of phosphatidylethanolamine species in the transfected cells. While the lipid changes in this case are driven by the structural changes in the protein cytoskeleton, the consequence of phosphatidylethanolamine enrichment may have additional implications in cancer such as increased membrane fluidity, increased motility and an ability to adapt to a depletion of unsaturated lipids during cancer cell proliferation. This study demonstrates a successful fluorescence microscopy-guided cell by cell membrane lipid analysis with broad application to biological investigation. [GRAPHICS] .

    • Commercial use of lead halide perovskites requires improved thermal stability and therefore a better understanding of their degradation mechanisms. The thermal degradation of three clean perovskite single crystal surfaces (MAPbI3, MAPbBr3, FAPbBr3) was investigated using synchrotron-based photoelectron spectroscopy. Central findings are that the halide has a large impact on thermal stability and that the degradation of formamidnium results in the formation of a new organic species at the FAPbBr3 crystal surface.

    • Purpose Odour is an important aspect of systems for humanand agricultural waste management and many technologiesare developed with the sole purpose of reducing odour.Compared with greenhouse gas assessment and the assessmentof toxicity, odour assessment has received little attentionin the life cycle assessment (LCA) community. This articleaims to redress this.Methods Firstly, a framework for the assessment of odourimpacts in LCA was developed considering the classicalLCA framework of emissions, midpoint and endpoint indicators.This suggested that an odour footprint midpoint indicatorwas worth striving for. An approach to calculating an arealindicator we call “odour footprint”, which considers the odourdetection threshold, the diffusion rate and the kinetics ofdegradation of odourants, was implemented in MATLAB.We demonstrated the use of the characterisation factors wecalculated in a case study based on odour removal technologyapplied to a pig barn.Results and discussion We produced a list of 33 linear characterisation factors based on hydrogen sulphide equivalents, analogous to the linear carbon dioxide equivalency factors in use in carbon footprinting, or the dichlorobenzene equivalency factors developed for assessment of toxic impacts in LCA. Like the latter, this odour footprint method does not take local populations and exposure pathway analysis into account—its intent is not to assess regulatory compliance or detailed design. The case study showed that despite the need for materials and energy, large factor reductions in odour footprint andeutrophication potential were achieved at the cost of a smaller factor increase in greenhouse emissions.Conclusions The odour footprint method is proposed as animprovement on the established midpoint method for odourassessment in LCA. Unlike it, the method presented hereconsiders the persistence of odourants. Over time, we hopeto increase the number of characterised odourants, enablinganalysts to perform simple site-generic LCA on systems withodourant emissions.

    • The 1984/85 Stockholm Health of the Population Study (HPS) was a cross sectional study including a questionnaire and a health examination. It was conducted through the ordinary health personnel of four Primary Health Care (PHC) districts in the Stockholm County. The main objective was to broaden the scope for planning health care. One of the goals of the study was to involve those working in PHC in planning their activities after local needs and to use more time for prevention work. The main effort to reach this objective was through involvement of the PHC personal from the early planning phase. This strategy was very time consuming but, on the other hand experiences from the HPS have led to both new screening activities of e.g high consumers of alcohol and the elderly, and preventive work aimed at those with smoking and overweight problems in the local PHC area. In the implementation process and through education the scientific approach level and interest in primary health care increased. A keen interest of survey data for local planning is obvious.

  • 3D printed nanocellulose scaffolds as a cancer cell culture model system

    • Current conventional cancer drug screening models based on two-dimensional (2D) cell culture have several flaws and there is a large need of more in vivo mimicking preclinical drug screening platforms. The microenvironment is crucial for the cells to adapt relevant in vivo characteristics and here we introduce a new cell culture system based on three-dimensional (3D) printed scaffolds using cellulose nanofibrils (CNF) pre-treated with 2,2,6,6-tetramethylpyperidine-1-oxyl (TEMPO) as the structural material component. Breast cancer cell lines, MCF7 and MDA-MB-231, were cultured in 3D TEMPO-CNF scaffolds and were shown by scanning electron microscopy (SEM) and histochemistry to grow in multiple layers as a heterogenous cell population with different morphologies, contrasting 2D cultured mono-layered cells with a morphologically homogenous cell population. Gene expression analysis demonstrated that 3D TEMPO-CNF scaffolds induced elevation of the stemness marker CD44 and the migration markers VIM and SNAI1 in MCF7 cells relative to 2D control. T47D cells confirmed the increased level of the stemness marker CD44 and migration marker VIM which was further supported by increased capacity of holoclone formation for 3D cultured cells. Therefore, TEMPO-CNF was shown to represent a promising material for 3D cell culture model systems for cancer cell applications such as drug screening. © 2021 by the authors.     • The cancer microenvironment influences tumor progression and metastasis and is pivotal to consider when designing in vivo-like cancer models. Current preclinical testing platforms for cancer drug development are mainly limited to 2D cell culture systems that poorly mimic physiological environments and traditional, low throughput animal models. The aim of this work was to produce a tunable testing platform based on 3D printed scaffolds (3DPS) with a simple geometry that, by extracellular components and response of breast cancer reporter cells, mimics patient-derived scaffolds (PDS) of breast cancer. Here, the biocompatible polysaccharide alginate was used as base material to generate scaffolds consisting of a 3D grid containing periostin and hydroxyapatite. Breast cancer cell lines (MCF7 and MDA-MB-231) produced similar phenotypes and gene expression levels of cancer stem cell, epithelial-mesenchymal transition, differentiation and proliferation markers when cultured on 3DPS and PDS, contrasting conventional 2D cultures. Importantly, cells cultured on 3DPS and PDS showed scaffold-specific responses to cytotoxic drugs (doxorubicin and 5-fluorouracil) that were different from 2D cultured cells. In conclusion, the data presented support the use of a tunable alginate-based 3DPS as a tumor model in breast cancer drug discovery. © 2021 The Author(s). Published by IOP Publishing Ltd.

  • Sodium glucose cotransporter 2 inhibitors and risk of serious adverse events: nationwide register based cohort study

    • OBJECTIVE: To assess the association between the use of sodium glucose cotransporter 2 (SGLT2) inhibitors and seven serious adverse events of current concern. DESIGN: Register based cohort study. SETTING: Sweden and Denmark from July 2013 to December 2016. PARTICIPANTS: A propensity score matched cohort of 17213 new users of SGLT2 inhibitors (dapagliflozin, 61%; empagliflozin, 38%; canagliflozin, 1%) and 17213 new users of the active comparator, glucagon-like peptide 1 (GLP1) receptor agonists. MAIN OUTCOME MEASURES: The primary outcomes were lower limb amputation, bone fracture, diabetic ketoacidosis, acute kidney injury, serious urinary tract infection, venous thromboembolism, and acute pancreatitis, as identified from hospital records. Hazard ratios and 95% confidence intervals were estimated by using Cox proportional hazards models. RESULTS: Use of SGLT2 inhibitors, as compared with GLP1 receptor agonists, was associated with an increased risk of lower limb amputation (incidence rate 2.7 v 1.1 events per 1000 person years, hazard ratio 2.32, 95% confidence interval 1.37 to 3.91) and diabetic ketoacidosis (1.3 v 0.6, 2.14, 1.01 to 4.52) but not with bone fracture (15.4 v 13.9, 1.11, 0.93 to 1.33), acute kidney injury (2.3 v 3.2, 0.69, 0.45 to 1.05), serious urinary tract infection (5.4 v 6.0, 0.89, 0.67 to 1.19), venous thromboembolism (4.2 v 4.1, 0.99, 0.71 to 1.38) or acute pancreatitis (1.3 v 1.2, 1.16, 0.64 to 2.12). CONCLUSIONS: In this analysis of nationwide registers from two countries, use of SGLT2 inhibitors, as compared with GLP1 receptor agonists, was associated with an increased risk of lower limb amputation and diabetic ketoacidosis, but not with other serious adverse events of current concern.
