  • Overview of the JET preparation for deuterium-tritium operation with the ITER like-wall
  • 2019
  • Ingår i: Nuclear Fusion. - : IOP Publishing. - 1741-4326 .- 0029-5515. ; 59:11
  • Forskningsöversikt (refereegranskat)abstract
    • For the past several years, the JET scientific programme (Pamela et al 2007 Fusion Eng. Des. 82 590) has been engaged in a multi-campaign effort, including experiments in D, H and T, leading up to 2020 and the first experiments with 50%/50% D-T mixtures since 1997 and the first ever D-T plasmas with the ITER mix of plasma-facing component materials. For this purpose, a concerted physics and technology programme was launched with a view to prepare the D-T campaign (DTE2). This paper addresses the key elements developed by the JET programme directly contributing to the D-T preparation. This intense preparation includes the review of the physics basis for the D-T operational scenarios, including the fusion power predictions through first principle and integrated modelling, and the impact of isotopes in the operation and physics of D-T plasmas (thermal and particle transport, high confinement mode (H-mode) access, Be and W erosion, fuel recovery, etc). This effort also requires improving several aspects of plasma operation for DTE2, such as real time control schemes, heat load control, disruption avoidance and a mitigation system (including the installation of a new shattered pellet injector), novel ion cyclotron resonance heating schemes (such as the three-ions scheme), new diagnostics (neutron camera and spectrometer, active Alfven eigenmode antennas, neutral gauges, radiation hard imaging systems...) and the calibration of the JET neutron diagnostics at 14 MeV for accurate fusion power measurement. The active preparation of JET for the 2020 D-T campaign provides an incomparable source of information and a basis for the future D-T operation of ITER, and it is also foreseen that a large number of key physics issues will be addressed in support of burning plasmas.

  • Net-zero deep decarbonization pathways in Latin America : Challenges and opportunities
  • 2020
  • Ingår i: Energy Strategy Reviews. - : Elsevier BV. - 2211-467X .- 2211-4688. ; 30
  • Tidskriftsartikel (refereegranskat)abstract
    • This synthesis paper presents the objectives, approach and cross-cutting results of the Latin American Deep Decarbonization Pathways project (DDP-LAC). It synthesizes and compares detailed national and sectoral deep decarbonization pathways (DDPs) to 2050 compatible with the Paris Agreement objectives and domestic development priorities in Argentina, Colombia, Costa Rica, Ecuador, Mexico and Peru. The first five countries analysed in detail the energy system and agriculture, forestry and land use (AFOLU) at a high level, while Peru focussed on a detailed analysis of AFOLU given its predominance in its GHG emissions. While economy-wide results were produced, this paper focuses on the electricity, passenger transport, and AFOLU results because of their current emissions, potential to grow, and identification of successful strategies for decarbonization (e.g. switching to clean electricity and other net-zero emissions fuels across the economy; urban planning, mode shifting, and electrification in passenger transport; and intensive sustainable agriculture, assignment of land use rights and their enforcement and afforestation in AFOLU). It also highlights where significant emissions remain in 2050, notably in industry, AFOLU, freight, and oil and gas production, all areas for future research. It derives insights for the design of domestic policy packages and identifies priorities for international cooperation. This analysis provides critical information for Long-Term Strategies, Nationally Determined Contributions and Global Stocktaking in the context of the Paris Agreement.

  • 1α,25-dihydroxyvitamin D3 promotes osteogenic activity and downregulates proinflammatory cytokine expression in human periodontal ligament cells
  • 2015
  • Ingår i: Journal of Periodontal Research. - : John Wiley & Sons. - 0022-3484 .- 1600-0765. ; 50:5, s. 666-673
  • Tidskriftsartikel (refereegranskat)abstract
    • Background and ObjectiveThe aim of this study was to assess the impact of 1,25-dihydroxyvitamin D3 (vitamin D3) on osteogenic and inflammatory properties of human periodontal ligament (PDL) cells and investigate underlying mechanisms. Material and MethodsHuman PDL cells, obtained from four subjects, were stimulated with vitamin D3 for 4-48h. The bone markers osteopontin and osteocalcin and proinflammatory cytokine/chemokine expression was determined by quantitative real-time polymerase chain reaction and enzyme-linked immunosorbent assay. Cytokine and chemokine expression was determined after stimulation with the inflammation promoter lipopolysaccharide (LPS) in the presence or absence of vitamin D3. Alkaline phosphatase activity was assessed using p-nitrophenylphosphate substrate. ResultsTreatment with 30ng/mL of vitamin D3, corresponding to an optimal plasma concentration of vitamin D, for 24h had no effect on PDL cell number and morphology but increased PDL cell osteopontin and osteocalcin mRNA expression by about 70 and 40%, respectively, and, moreover, treatment with vitamin D3 for 48h enhanced PDL cell alkaline phosphatase activity by about two times showing that vitamin D3 exerts pro-osteogenic effects in human PDL cells. Stimulation with LPS (1g/mL) for 4h increased PDL cell interleukin (IL)-6 cytokine and chemokine ligand 1 (CXCL1) chemokine mRNA expression several fold. The LPS-induced increase in IL-6 and CXCL1 transcripts was attenuated by vitamin D3 (30ng/mL). Treatment with vitamin D3 (3-300ng/mL) for 24h reduced the LPS-evoked increase in PDL cell IL-6 protein by about 50%. Vitamin D3 (30ng/mL) had no effect on LPS-induced IL-1 and MCP-1 mRNA expression. ConclusionsVitamin D3 promotes osteogenic differentiation but also downregulates inflammation promoter-induced IL-6 cytokine and CXCL1 chemokine expression in human PDL cells, suggesting that vitamin D3 both stimulates bone regeneration and antagonizes inflammation in human periodontal tissue.

  • Secretory leukocyte protease inhibitor regulates human periodontal ligament cell production of pro-inflammatory cytokines
  • 2017
  • Ingår i: Inflammation Research. - : Springer Science and Business Media LLC. - 1023-3830 .- 1420-908X. ; 66:9, s. 823-831
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective: Regulation of immune-like cell properties of periodontal ligament (PDL) cells is not understood. We investigate the importance of secretory leukocyte protease inhibitor (SLPI) for production of pro-inflammatory cytokines in human PDL cells. Materials and methods: PDL cells were isolated from teeth extracted for orthodontic reasons. Cellular location of SLPI was investigated by immunocytochemistry. Cytokine transcript and protein expression were assessed by quantitative real-time RT-PCR and Western blotting. SLPI gene activity was knocked-down by siRNA. NF-κB signaling was assessed by measuring IκBα, and phosphorylated p65 and p105 protein expression. Results: PDL cells showed cytoplasmic expression of SLPI. Cellular expression level of SLPI negatively correlated to LPS-induced stimulation of IL-6 and MCP-1. Both SLPI gene activity and protein were reduced by about 70% in PDL cells treated with SLPI siRNA compared to cells treated with non-coding construct. Treatment with SLPI siRNA was associated with up-regulation of both basal and LPS-stimulated IL-6, MCP-1 and TLRs mRNA expression. The up-regulation of MCP-1 transcript in SLPI siRNA-treated cells was confirmed on protein level. SLPI siRNA-treatment enhanced the phosphorylated NF-κB p105 protein expression. Conclusions: SLPI regulates PDL cell pro-inflammatory cytokine expression and modulates NF-κB signaling, suggesting that SLPI governs the immune cell-like properties of PDL cells.

    • In the late 19th century, football entered Sweden's coastal cities, such as Malmö, Halmstad and Gothenburg. The sport grew quickly, and the Swedish Football Association (SvFF) was founded in 1904. In the following decades, the popularity of football increased and in the 1950s it was perceived as the national sport of Sweden. However, at that time the sport was non-professional and in practice only for men. In order to keep up with hardening international competition, SvFF overturned the amateur regulations in 1967. Professionalisation was slow due to the lack of revenue but accelerated for male players after the Bosman ruling in 1995. Women's football developed gradually from the 1960s and in 1972 a national league organised by SvFF was formed. Youth football also grew substantially. Despite the differences in resources football became well-established amongst both men and women. However, the tensions between idealism, voluntarism and inclusion on the one hand, and commercialism, professionalism and selection, on the other hand, remain. This is best exemplified by the 51% rule, which states that clubs must be majority-owned by the members. This is hailed by some as a guarantee for democratic football, while others argue that it restricts clubs’ financial development.

  • Whole-genome sequencing identifies EN1 as a determinant of bone density and fracture
  • 2015

  • Tidskriftsartikel (refereegranskat)abstract
    • The extent to which low-frequency (minor allele frequency (MAF) between 1-5%) and rare (MAF <= 1%) variants contribute to complex traits and disease in the general population is mainly unknown. Bone mineral density (BMD) is highly heritable, a major predictor of osteoporotic fractures, and has been previously associated with common genetic variants(1-8), as well as rare, population specific, coding variants(9). Here we identify novel non-coding genetic variants with large effects on BMD (n(total) = 53,236) and fracture (n(total) = 508,253) in individuals of European ancestry from the general population. Associations for BMD were derived from whole-genome sequencing (n = 2,882 from UK10K (ref. 10); a population-based genome sequencing consortium), whole-exome sequencing (n = 3,549), deep imputation of genotyped samples using a combined UK10K/1000 Genomes reference panel (n = 26,534), and de novo replication genotyping (n = 20,271). We identified a low-frequency non-coding variant near a novel locus, EN1, with an effect size fourfold larger than the mean of previously reported common variants for lumbar spine BMD8 (rs11692564(T), MAF51.6%, replication effect size510.20 s.d., P-meta = 2 x 10(-14)), which was also associated with a decreased risk of fracture (odds ratio = 0.85; P = 2 x 10(-11); ncases = 98,742 and ncontrols = 409,511). Using an En1cre/flox mouse model, we observed that conditional loss of En1 results in low bone mass, probably as a consequence of high bone turnover. We also identified a novel low frequency non-coding variant with large effects on BMD near WNT16 (rs148771817(T), MAF = 1.2%, replication effect size +10.41 s.d., P-meta = 1 x 10(-11)). In general, there was an excess of association signals arising from deleterious coding and conserved non-coding variants. These findings provide evidence that low-frequency non-coding variants have large effects on BMD and fracture, thereby providing rationale for whole-genome sequencing and improved imputation reference panels to study the genetic architecture of complex traits and disease in the general population.

  • Antimicrobial peptide LL-37 and its pro-form, hCAP18, in desquamated epithelial cells of human whole saliva
  • 2020
  • Ingår i: European Journal of Oral Sciences. - : Wiley. - 0909-8836 .- 1600-0722. ; 128:1, s. 1-6
  • Tidskriftsartikel (refereegranskat)abstract
    • The antimicrobial peptide LL-37 is active against oral bacteria and has been demonstrated to be present in human saliva, but its distribution in different fractions of saliva is not known. LL-37 is formed from its intracellular pro-form, hCAP18, in an extracellular enzymatic reaction catalyzed by proteinase 3 and kallikrein 5. Here, we prepared cell-containing and cell-free fractions of unstimulated human whole saliva by centrifugation after depolymerization of mucins with dithiothreitol, and measured the levels of hCAP18/LL-37 in these fractions using ELISA. Cellular expression of hCAP18/LL-37 was determined by western blotting and immunocytochemistry. The ELISA analyses demonstrated that both cells and cell-free saliva contained hCAP18/LL-37. Western blot analysis of cell-pellet homogenates showed a strong band corresponding to hCAP18 at the correct molecular weight and a weak band corresponding to LL-37. Phase-contrast and light microscopy revealed that the cells consisted of desquamated epithelial cells. These cells expressed cytoplasmic immunoreactivity for hCAP18/LL-37. The peripheral part of the cytoplasm, corresponding to the plasma membrane, was particularly rich in hCAP18/LL-37 immunoreactivity. No immunoreactivity was observed after omission of the primary antibody. We conclude that desquamated epithelial cells of human whole saliva contain antimicrobial hCAP18/LL-37, suggesting that these cells may take part in the innate immune system by harboring and releasing these peptides.

  • Strontium chloride promotes cell proliferation in a human osteoblast cell line
  • 2014
  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • Strontium ranelate (SrRan) is the active component of drugs currently used for reducing the risk of fractures in patients suffering from osteoporosis. Despite extensive use, the underlying mechanisms of action of Sr2+ are not fully understood. In the present study, we assess the impact of SrCl2 on human osteoblast activity and proliferation. Cultures of the human osteoblast-like cell line MG63 were treated for 72 h in presence of 0.1 mM, 1 mM, 5 mM and 10 mM SrCl2 or vehicle, used in control groups. Cells were counted manually using a Bürker chamber. Total protein content was determined by colorimetric analysis performed by a microplate reader using Bio-Rad protein assay. Alkaline phosphatase (ALP) activity was determined enzymatically and normalized to total protein content in each sample. Cell viability was assessed using the MTT assay. Treatment with 5 mM SrCl2 for 72 h enhanced total MG63 cell protein content by 37% compared to controls (p<0.01). A lower concentration (0.1 mM) of SrCl2 had no effect on total protein. Incubation with 5 mM SrCl2 for 72 h increased MG63 cell number by 38% compared to controls (p<0.001). The SrCl2-induced increase in cell number was associated with enhanced (+14% compared to controls, p<0.05) cell viability. Treatment with a higher concentration (10 mM) of SrCl2 enhanced cell number similar to 5 mM SrCl2 (+54% compared to controls, p<0.05). Treatment with 0.1 or 5 mM SrCl2 for 72 h had no effect (p>0.05) on MG63 cell ALP activity, while 1 mM SrCl2 reduced ALP activity as well as total protein content by about 25% compared to controls (p<0.05). The current results demonstrate that treatment with SrCl2 for 72 h, at concentrations higher than 1 mM promotes cell proliferation in human osteoblast-like cells, suggesting that Sr2+ may enhance bone formation through this mechanism.

  • The host defense peptide LL-37 is internalized by human periodontal ligament cells and prevents LPS-induced MCP-1 production
  • 2019
  • Ingår i: Journal of Periodontal Research. - : Wiley. - 0022-3484 .- 1600-0765. ; 54:6, s. 662-670
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective: The human host defense peptide LL-37 both shows antimicrobial effects and modulates host cell properties. Here, we assess the effects of synthesized LL-37 on lipopolysaccharide (LPS)-induced inflammation in human periodontal ligament (PDL) cells and investigates underlying mechanisms. Background: LL-37 has been detected in the periodontal tissues, but its functional importance for PDL cell innate immune responses is not known. Methods: Human PDL cells were obtained from premolars extracted on orthodontic indications. Cellular pro-inflammatory monocyte chemoattractant protein-1 (MCP-1) mRNA expression was determined using quantitative real-time RT-PCR. MCP-1 protein production was assessed by western blot and ELISA. Internalization of LL-37 by PDL cells was visualized by immunocytochemistry. Nuclear factor kappa-light-chain-enhancer of activated B-cell (NF-κB) activity was assessed by western blot of phosphorylated p65, phosphorylated p105, and IκBα proteins. Binding of LL-37 to PDL cell DNA was determined by isolation and purification of DNA and dot blot for LL-37 immunoreactivity. Results: Treatment with LL-37 (1 µmol/L) for 24 hours prevented LPS-induced stimulation of MCP-1 expression analyzed both on transcript and on protein levels. Stimulation with LL-37 (1 µmol/L) for 24 hours had no effect on toll-like receptor (TLR)2 and TLR4 transcript expression, suggesting that LL-37 acts downstream of the TLRs. Preincubation with LL-37 for 60 minutes followed by stimulation with LPS for 24 hours in the absence of LL-37 completely prevented LPS-evoked MCP-1 transcript expression, implying that LL-37 acts intracellularly and not via binding and neutralization of LPS. In PDL cells stimulated with LL-37 for 60 minutes, the peptide was internalized as demonstrated by immunocytochemistry, suggesting an intracellular mechanism of action. LL-37 immunoreactivity was observed both in the cytosol and in the nucleus. Downregulation of LPS-induced MCP-1 by LL-37 was not mediated by reduction in NF-κB activity as shown by unaltered expression of phosphorylated p65, phosphorylated p105, and IκBα NF-κB proteins in the presence of LL-37. Immunoreactivity for LL-37 was observed in PDL cell DNA treated with but not without 0.1 and 1 µmol/L LL-37 for 60 minutes in vitro. Conclusion: LL-37 abolishes LPS-induced MCP-1 production in human PDL cells through an intracellular, NF-κB-independent mechanism which probably involves direct interaction between LL-37 and DNA.

  • Globular C1q receptor (p33) binds and stabilizes pro-inflammatory MCP-1 : a novel mechanism for regulation of MCP-1 production and function
  • 2018
  • Ingår i: Biochemical Journal. - 0264-6021. ; 475:4, s. 775-786
  • Tidskriftsartikel (refereegranskat)abstract
    • The protein gC1qR (globular C1q receptor), also named p33, was originally identified as a binding partner of the globular heads of C1q in the complement system. gC1qR/p33 is abundantly expressed in many cell types, but the functional importance of this protein is not completely understood. Here, we investigate the impact of gC1qR/p33 on the production and function of the pathophysiologically important chemokine monocyte chemoattractant protein-1 (MCP-1) and the underlying molecular mechanisms. Knockdown of gC1qR/p33 negatively regulated the production of MCP-1, but had no effect on the expression of transcript for MCP-1 in human periodontal ligament cells, suggesting a translational/post-translational mechanism of action. Laser scanning confocal microscopy showed considerable cytosolic co-localization of gC1qR/p33 and MCP-1, and co-immunoprecipitation disclosed direct physical interaction between gC1qR/p33 and MCP-1. Surface plasmon resonance analysis revealed a high-affinity binding (KD = 10.9 nM) between gC1qR/p33 and MCP-1. Using a transwell migration assay, we found that recombinant gC1qR/p33 enhances MCP-1-induced migration of human THP-1 monocytes, pointing to a functional importance of the interaction between gC1qR/p33 and MCP-1. An in vitro assay revealed a rapid turnover of the MCP-1 protein and that gC1qR/ p33 stabilizes MCP-1, hence preventing its degradation. We propose that endogenous gC1qR/p33 physically interacts with MCP-1 causing stabilization of the MCP-1 protein and stimulation of its activity in human periodontal ligament cells, suggesting a novel gC1qR/p33-mediated pro-inflammatory mechanism of action.

  • Don't buy a pig in a poke: Considering challenges of and problems with performance analysis technologies in Swedish men's elite football
  • 2021
  • Ingår i: Performance Enhancement and Health. - : Elsevier BV. - 2211-2669. ; 9:1
  • Tidskriftsartikel (refereegranskat)abstract
    • During the last decades, technologies to monitor, test and analyze athletes’ performance and health have rapidly developed. At present, global positioning systems (GPS), stadium camcorders, heart rate monitors and mobile applications are prominent performance analysis technologies (PATs) used in most elite sport environments. While PATs is understood as an aid, there is a growing body of literature that points to negative consequences. These negative consequences are concerning and call for research and measures to develop strategies for effective and productive implementation. To achieve this, this article first outlines key challenges and problems of PATs, using sport sociological research on coaching and athletes, historical knowledge of the scientization of training and the changing role of the coach, as well as scientific and experiential knowledge of performance analysis. Our findings show that key challenges and problems occur in a chain of six steps that concern the implementing of PATs: 1. Investment in PATs; 2. Production of performance data; 3. Interpretation of performance data; 4. Communication of performance data; 5. Decision-making based on performance data; and 6. Influence of PATs on coaches and athletes. The article then answers these challenges and problems by outlining recommendations for how sport managers and administrators can prevent buying “a pig in a poke” by acquiring competence about performance analysis and PATs, investing time, and developing effective communication between those working with PATs. © 2021 The Author(s)

    • Abstract—We introduce the Set MHT, a tracking algorithmthat maintains multiple hypotheses and produces “smooth”estimates without the track coalescence often associated withMinimum Mean Squared Error (MMSE) estimation or thejitter associated with Maximum Likelihood (ML) estimation. It does this by utilizing Minimum Mean Optimal SubpatternAssignment (MMOSPA) estimation techniques coupled with atheoretically-grounded approach for probabilistically determiningthe identities of the state estimates. Unlike traditional MHTalgorithms, the Set MHT does not “forget” uncertainty in targetidentities, i.e. display an unjustifiably high confidence level inthe target identities, as a result of pruning out competinghypotheses. Rather, it uses merging techniques while avoiding theshortcomings of traditional Gaussian mixture reduction trackers.

  • Ingår i: Nature genetics. - : Springer Science and Business Media LLC. - 1546-1718 .- 1061-4036. ; 44:5, s. 491-501
  • Tidskriftsartikel (refereegranskat)abstract
    • Bone mineral density (BMD) is the most widely used predictor of fracture risk. We performed the largest meta-analysis to date on lumbar spine and femoral neck BMD, including 17 genome-wide association studies and 32,961 individuals of European and east Asian ancestry. We tested the top BMD-associated markers for replication in 50,933 independent subjects and for association with risk of low-trauma fracture in 31,016 individuals with a history of fracture (cases) and 102,444 controls. We identified 56 loci (32 new) associated with BMD at genome-wide significance (P < 5 × 10(-8)). Several of these factors cluster within the RANK-RANKL-OPG, mesenchymal stem cell differentiation, endochondral ossification and Wnt signaling pathways. However, we also discovered loci that were localized to genes not known to have a role in bone biology. Fourteen BMD-associated loci were also associated with fracture risk (P < 5 × 10(-4), Bonferroni corrected), of which six reached P < 5 × 10(-8), including at 18p11.21 (FAM210A), 7q21.3 (SLC25A13), 11q13.2 (LRP5), 4q22.1 (MEPE), 2p16.2 (SPTBN1) and 10q21.1 (DKK1). These findings shed light on the genetic architecture and pathophysiological mechanisms underlying BMD variation and fracture susceptibility.

  • Shooting two birds with two bullets: how to find Minimum Mean OSPA estimates
  • 2010

  • Konferensbidrag (refereegranskat)abstract
    • Most area-defense formulations followfrom the assumption that threats must first be identifiedand then neutralized. This is reasonable, but inherentto it is a process of labeling: threat A must be identifiedand then threat B, and then action must be taken. This manuscript begins from the assumption that suchlabeling (A & B) is irrelevant. The problem naturallydevolves to one of Random Finite Set (RFS) estimation:we show that by eschewing any concern of targetlabel we relax the estimation procedure, and it is perhapsnot surprising that by such a removal of constraint (oflabeling) performance (in terms of localization) is enhanced.A suitable measure for the estimation of unlabeledobjects is the Mean OSPA (MOSPA). We derive ageneral algorithm which provided the optimal estimatorwhich minimize the MOSPA. We call such an estimatora Minimum MOSPA (MMOSPA) estimator.

  • Evidence of 9 Be + p nuclear reactions during 2ω CH and hydrogen minority ICRH in JET-ILW hydrogen and deuterium plasmas
  • 2018
  • Ingår i: Nuclear Fusion. - : IOP Publishing. - 1741-4326 .- 0029-5515. ; 58:2
  • Tidskriftsartikel (refereegranskat)abstract
    • The intensity of 9Be + p nuclear fusion reactions was experimentally studied during second harmonic (2ω CH) ion-cyclotron resonance heating (ICRH) and further analyzed during fundamental hydrogen minority ICRH of JET-ILW hydrogen and deuterium plasmas. In relatively low-density plasmas with a high ICRH power, a population of fast H+ ions was created and measured by neutral particle analyzers. Primary and secondary nuclear reaction products, due to 9Be + p interaction, were observed with fast ion loss detectors, γ-ray spectrometers and neutron flux monitors and spectrometers. The possibility of using 9Be(p, d)2α and 9Be(p, α)6Li nuclear reactions to create a population of fast alpha particles and study their behaviour in non-active stage of ITER operation is discussed in the paper.

  • A control oriented strategy of disruption prediction to avoid the configuration collapse of tokamak reactors
  • 2024
  • Ingår i: Nature Communications. - 2041-1723 .- 2041-1723. ; 15:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The objective of thermonuclear fusion consists of producing electricity from the coalescence of light nuclei in high temperature plasmas. The most promising route to fusion envisages the confinement of such plasmas with magnetic fields, whose most studied configuration is the tokamak. Disruptions are catastrophic collapses affecting all tokamak devices and one of the main potential showstoppers on the route to a commercial reactor. In this work we report how, deploying innovative analysis methods on thousands of JET experiments covering the isotopic compositions from hydrogen to full tritium and including the major D-T campaign, the nature of the various forms of collapse is investigated in all phases of the discharges. An original approach to proximity detection has been developed, which allows determining both the probability of and the time interval remaining before an incoming disruption, with adaptive, from scratch, real time compatible techniques. The results indicate that physics based prediction and control tools can be developed, to deploy realistic strategies of disruption avoidance and prevention, meeting the requirements of the next generation of devices.

    • Contemporary images of desirable work (for example at gaming companies or at one of the tech giants) foregrounds creativity and incorporates and idealises elements of play. Simultaneously, becoming one of the best in some particular leisure activity can require many long hours of hard, demanding work. Between on the one hand work and on the other hand leisure and play, we enter the domain of games and sports. Most classical sports originally developed from physical practices of moving the human body and these practices were, through standardization, organization and rationalization, turned into sports. Many sport researchers, (sport) historians and (sport) sociologists have pointed out that sports have gone through a process of “sportification”. Cross-country skiing is an example of an activity that has gone through a historical process of sportification, over time becoming progressively more managed and regulated. Computer games are today following a similar trajectory and have gone from being a leisure activity to becoming a competitive activity, “e-sports”, with professional players, international competitions, and live streams that are watched by tens of millions of viewers. In this paper we look at similarities between the sportification of cross-country skiing and e-sports. While there are many similarities, one important difference is that where classical sports (e.g. cross-country skiing) often originated in work-related practices (in this case forestry), the formation of new and emerging sports (for example e-sports) instead often originates in playful leisure activities. We will in this text show how sports and sportification processes can function as a lens with which to better understand the intersection of work and play, and well as their overlapping and hybrid albeit non-mutually exclusive combinations; laborious play and playful work.

  • Merging-based forward-backward smoothing on Gaussian mixtures
  • 2014

  • Konferensbidrag (refereegranskat)abstract
    • Conventional forward-backward smoothing (FBS) for Gaussian mixture (GM) problems are based on pruning methods which yield a degenerate hypothesis tree and often lead to underestimated uncertainties. To overcome these shortcomings, we propose an algorithm that is based on merging components in the GM during filtering and smoothing. Compared to FBS based on the N-scan pruning, the proposed algorithm offers better performance in terms of track loss, root mean squared error (RMSE) and normalized estimation error squared (NEES) without increasing the computational complexity.

  • Ingår i: FUSION 2013, 9-12 July 2013, Istanbul, Turkey. - 9786058631113 ; , s. 1296 - 1303
  • Konferensbidrag (refereegranskat)abstract
    • This paper presents the Smoothed Probabilistic Data Association Filter (SmPDAF) that attempts to improve the Gaussian approximations used in the Probabilistic Data Association Filter (PDAF). This is achieved by using information from future measurements. Newer approximations of the densities are obtained by using a combination of expectation propagation, which provides the backward likelihood information from the future measurements, and pruning, which uses these backward likelihoods to reduce the number of components in the Gaussian mixture. Performance comparison between SmPDAF and PDAF shows us that the root mean squared error performance of SmPDAF is significantly better than PDAF under comparable track loss performance.

  • Two-filter Gaussian mixture smoothing with posterior pruning

  • Konferensbidrag (refereegranskat)abstract
    • In this paper, we address the problem of smoothing on Gaussian mixture (GM) posterior densities using the two-filter smoothing (TFS) strategy. The structure of the likelihoods in the backward filter of the TFS is analysed in detail. These likelihoods look similar to GMs, but are not proper density functions in the state-space since they may have constant value in a subspace of the state space. We present how the traditional GM reduction techniques can be extended to this kind of GMs. We also propose a posterior-based pruning strategy, where the filtering density can be used to make further approximations of the likelihood in thebackward filter. Compared to the forward–backward smoothing(FBS) method based on N-scan pruning approximations, the proposed algorithm is shown to perform better in terms of track loss, normalized estimation error squared (NEES), computational complexity and root mean squared error (RMSE).

  • A New Multiple Model Filter with Switch Time Conditions
  • 2010
  • Ingår i: IEEE Transactions on Signal Processing. - 1941-0476 .- 1053-587X. ; 58:1, s. 11-25
  • Tidskriftsartikel (refereegranskat)abstract
    • The interacting multiple model filter has long been the method of choice for performing target tracking using multiple motion models. The filter finds a suboptimal solution to a problem which has the implicit assumption that immediate model shifts have the highest probability. When the sampling rate of the underlying continuous process is high compared to the target dynamics, this is not a reasonable assumption. Instead, changes in dynamics persist for some time. In this paper we propose an alternative switching model, which forces the dynamic models to persist for at least a model-specific time. The model is semi-Markov in nature, with a sojourn time probability mass function which is zero for a model-specific number of time steps, and then follows a geometrical distribution. Through this assumption a less complex problem in terms of model hypotheses arises, and to that problem we derive a state estimation algorithm that is close to optimal when the model assumptions are valid. Three other semi-Markov-based multiple-model filters are discussed and compared to in a qualitative sense. We also derive a new aircraft motion model for start and termination of turns. Finally, the proposed filter is evaluated on a benchmark scenario for tracking, and the results show a performance increase compared to the Interacting Multiple Model (IMM) filter for the trajectories considered.

  • An Alternative Derivation of the Cardinalized Probability Hypothesis Density Filter
  • 2008
  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • In this report, an alternative approach to derive the Gaussian mixture cardinalized probability hypothesis density (GM-CPHD) filter is presented. The derivations differ in that the presented ones are based on "ordinary" statistics, while the original GM-CPHD derivation started from the finite set statistics (FISST) description of the CPHD filter. The results of the derivations are compared with filter update equations presented in another paper. The sets of equations are not completely equivalent. However, initial performance evaluations of the approaches indicate similar performance. Future work is needed to understand the differences between different GM-CPHD filter equations.

  • Esports and Sportification : A View From Sweden
  • 2024
  • Ingår i: Critical Perspectives on Esports. - London : Routledge. - 9781003383178
  • Bokkapitel (refereegranskat)abstract
    • The rapid development of esports over the last two decades has been in some ways a challenge to the established sporting world. However, esports has largely developed according to the sportification model (Guttman, 1978), over time becoming progressively more organised, specialised, standardised, regimented, and rationalised. Competitive computer gaming is today following a similar trajectory as other sports have done in the past and has gone from being a leisure activity to becoming a competitive activity with organisations, professional players, and international competitions. This chapter gives a closer look at the ongoing sportification of esports in general and, more specifically, how this process has played out in Sweden. Similarities and differences in the sportification of esports and a more traditional sport - cross-country skiing - will be analysed. How have esports worked with organisation, regimentation, rationalisation, and standardisation? Building upon archival sources, earlier research, and interviews with representatives from Swedish esports organisations will conclude the chapter.

  • Human endogenous peptide p33 inhibits detrimental effects of LL-37 on osteoblast viability
  • 2015
  • Ingår i: Journal of Periodontal Research. - : John Wiley & Sons. - 0022-3484 .- 1600-0765. ; 50:1, s. 80-88
  • Tidskriftsartikel (refereegranskat)abstract
    • Background and Objective: High levels of the antimicrobial peptide, LL-37, are detected in gingival crevicular fluid from patients with chronic periodontitis. LL-37 not only shows antimicrobial activity but also affects host-cell viability. The objective of the present study was to identify endogenous mechanisms that antagonize the detrimental effects of LL-37 on osteoblast viability, focusing on the human peptide p33 expressed on the surface of various cell types. Material and Methods: Human osteoblast-like MG63 cells and human hFOB1.19 osteoblasts were treated with or without LL-37 in the presence or absence of p33. Recombinant human p33 was expressed in an Escherichia coli expression system. Lactate dehydrogenase (LDH) was assessed using an enzymatic spectrophotometric assay. DNA synthesis was determined by measuring [3H]-thymidine incorporation. Cell number was assessed by counting cells in a Bu€rker chamber. Intracellular Ca2+ was monitored by recording Fluo 4-AM fluorescence using a laser scanning confo- cal microscope. Cellular expression of p33 was determined by western blotting. Results: LL-37 caused a concentration-dependent release of LDH from human osteoblasts, showing a half-maximal response value (EC50) of 4 lM and a rapid and sustained rise in the intracellular Ca2+ concentration of osteoblasts, sug- gesting that LL-37 forms pores in the cell membrane. p33 (10 lM) inhibited the LL-37-induced LDH release and LL-37-evoked rise in intracellular Ca2+ con- centration, suggesting that p33 prevents LL-37-induced permeabilization of the cell membrane. Moreover, p33 blocked LL-37-induced attenuation of osteoblast numbers. Also, mucin antagonized, at concentrations representative for nonsti- mulated whole saliva, LL-37-evoked LDH release, whilst cationic endogenous polyamines had no impact on LL-37-induced LDH release from osteoblasts. Conclusions: The endogenous peptide p33 prevents LL-37-induced reduction of human osteoblast viability. Importantly, this mechanism may protect the osteo- blasts from LL-37-induced cell damage in patients suffering from chronic peri- odontitis associated with high levels of LL-37 locally.

  • Performance Evaluation of MHT and GM-CPHD in a Ground Target Tracking Scenario
  • 2009
  • Ingår i: Proceedings of the 12th International Conference on Information Fusion. - 9780982443804 ; , s. 300-307
  • Konferensbidrag (refereegranskat)abstract
    • Performance evaluations of multi-target tracking algorithms are often limited to consider comparisons within the same algorithm family. In this paper, two conceptually different multi-target tracking algorithms are evaluated, namely a multiple-hypothesis tracking (MHT) algorithm and the Gaussian mixture cardinalized probability hypothesis density (GM-CPHD) filter. As a reference, a conventional single-hypothesis tracking algorithm is included in the evaluation. The performance is assessed using the root-mean square error of the estimated number of targets, and the recently published optimal subpattern assignment (OSPA) measure. The scenario under consideration is tracking of nine closely spaced ground targets, using simulated measurements from an airborne radar. By observing the estimation of the number of targets, as well as of the target states, conclusions are drawn regarding the behavior of MHT and GM-CPHD. For example, GM-CPHD is more responsive to changes in the number of targets, whereas MHT is less responsive, but produces a more stable output.
