    • Summary There is a need for more integrated landscape planning due to rapid changes in landscape use through increased rationalization of land use (e.g. forestry), climate change and lack of planning in the landscape outside the cities. In this project, we have anticipated the reindeer herding Sami people and their interest to follow the prerequisites of land use and habitat changes over time. The project can partially be mentioned as a process consisting of a dialogue between reindeer herders, the Swedish Forest Agency, Vilhelmina Model Forest and the National Monitoring Programme NILS. The European project Baltic Landscape frames the project. The aim of Baltic Landscape is to demonstrate concrete examples of how conflicts related to land use claims can be solved. In this project, we worked specifically with the Model Forest concept that is based on a bottom-up perspective of working with issues related to landscapes and covering e.g. sector driven industry such as forestry, tourism, environmental values and governmental organizations. During the project all parts met and had theoretical sessions, field exercises and subsequent discussions that overall was about how best to monitor different habitats that are of large importance for the forage of reindeers (grazing types). More technical approaches such as Unmanned airplanes (so called UAV:s) were tested. Reindeer movements across the landscape range from the Alpine regions in the west to the coniferous forests on the east coast. The study includes the alpine landscape, mountain birch forest and marshlands in Vilhelmina Model Forest. The purpose: for reindeer herders was to find a method to conduct inventory on grazing types (reindeer habitats) that was rational to work with, i.e., not too time consuming (so that the inventory could be combined with the everyday work) and not too simple (so that it does not work as it is supposed to, as a support for discussion with other land users). for NILS was to initiate a dialogue with users and their needs of environmental monitoring data. More concretely, it was about to find out if there is data that can be of importance for reindeer herders which is not presently collected in the current field monitoring. Moreover to see if reindeer herders have the opportunity to collect data in a way so that it is possible to supplement existing NILS data. Finally to see if was possible to use unmanned aircraft, known as UAVs, to do inventory grazing types and especially hanging lichens (important as food supplement for reindeer). Moreover, interviews with stakeholders (N= 12) and reindeer herders (N= 1) , were made in order to deepen the knowledge of potential needs of NILS monitoring data for landuse and landscape planning by stakeholders in Vilhelmina Model Forest. for Vilhelmina Model Forest and the Swedish Forest Agency was to initiate dialogue between the parties and act as a coordinator and resource for increased dialogue. The results of the project (and process) revealed: that the importance of the possibility to conduct a dialogue. The dialogue led to the emergence of synergies that all parts could use. In this case the common denominator was to establish a specific area in the alpine region for field visits. The purpose was to use the area to promote and initiate dialogues about the landscape and in the longer perspective enable monitoring and increase knowledge about suitable usage of the landscape. NILS called such a surface Flagship areas, Vilhelmina Model forest call those areas Demo areas or geological parks (geopark). that it was possible to develop a rational functioning method of reindeer grazing types (linked to the Reindeer Husbandry Plans) based on field exercises and discussions conducted in this project 2012 -2014. During 2014 NILS personnel educate reindeer herders using the method developed within this project. that the methodology of inventory used by the NILS programme, both in field and by aerial photographs, is partly compatible with the existing reindeer grazing types. And also that NILS can benefit from dialogue with stakeholders and gain knowledge of which classification schemes can be converted into or what can be added to their inventory. that there is a wish to continue further dialogue between the partners. that interviews with stakeholder revealed the need of usefulness and costs for NILS data, that no no single variable was excluded as not interesting at all for any of the stakeholders (87 out of 356 NILS variables were asked for in the interview) and that stakeholders made suggestions of new variables that could complement the current NILS variable list.

  • Impact of Temperature Dependent Sampling Procedures in Proteomics and Peptidomics : A Characterization of the Liver and Pancreas Post Mortem Degradome

    • Little is known about the nature of post mortem degradation of proteins and peptides on a global level, the so-called degradome. This is especially true for nonneural tissues. Degradome properties in relation to sampling procedures on different tissues are of great importance for the studies of, for instance, post translational modifications and/or the establishment of clinical biobanks. Here, snap freezing of fresh(< 2 min post mortem time) mouse liver and pancreas tissue is compared with rapid heat stabilization with regard to effects on the proteome (using two-dimensional differential in-gel electrophoresis) and peptidome (using label free liquid chromatography). We report several proteins and peptides that exhibit heightened degradation sensitivity, for instance superoxide dismutase in liver, and peptidyl-prolyl cis-trans isomerase and insulin C-peptides in pancreas. Tissue sampling based on snap freezing produces a greater amount of degradation products and lower levels of endogenous peptides than rapid heat stabilization. We also demonstrate that solely snap freezing related degradation can be attenuated by subsequent heat stabilization. We conclude that tissue sampling involving a rapid heat stabilization step is preferable to freezing with regard to proteomic and peptidomic sample quality.

  • A catalog of genetic loci associated with kidney function from analyses of a million individuals

    • Chronic kidney disease (CKD) is responsible for a public health burden with multi-systemic complications. Through transancestry meta-analysis of genome-wide association studies of estimated glomerular filtration rate (eGFR) and independent replication (n = 1,046,070), we identified 264 associated loci (166 new). Of these,147 were likely to be relevant for kidney function on the basis of associations with the alternative kidney function marker blood urea nitrogen (n = 416,178). Pathway and enrichment analyses, including mouse models with renal phenotypes, support the kidney as the main target organ. A genetic risk score for lower eGFR was associated with clinically diagnosed CKD in 452,264 independent individuals. Colocalization analyses of associations with eGFR among 783,978 European-ancestry individuals and gene expression across 46 human tissues, including tubulo-interstitial and glomerular kidney compartments, identified 17 genes differentially expressed in kidney. Fine-mapping highlighted missense driver variants in 11 genes and kidney-specific regulatory variants. These results provide a comprehensive priority list of molecular targets for translational research.

  • Risk factors for nephropathy in persons with type 1 diabetes: a population-based study

    • Aims Albuminuria is strongly associated with risk of renal dysfunction, cardiovascular disease and mortality. However, clinical guidelines diverge, and evidence is sparse on what risk factor levels regarding blood pressure, blood lipids and BMI are needed to prevent albuminuria in adolescents and young adults with type 1 diabetes. Methods A total of 9347 children and adults with type 1 diabetes [mean age 15.3 years and mean diabetes duration 1.4 years at start of follow-up] from The Swedish National Diabetes Registry were followed from first registration until end of 2017. Levels for risk factors for a risk increase in nephropathy were evaluated, and the gradient of risk per 1 SD (standard deviation) was estimated to compare the impact of each risk factor. Results During the follow-up period, 8610 (92.1%) remained normoalbuminuric, 737 (7.9%) individuals developed micro- or macroalbuminuria at any time period of whom 132 (17.9% of 737) individuals developed macroalbuminuria. Blood pressure >= 140/80 mmHg was associated with increased risk of albuminuria (p <= 0.0001), as were triglycerides >= 1.0 mmol/L (p = 0.039), total cholesterol >= 5.0 mmol/L (p = 0.0003), HDL < 1.0 mmol/L (p = 0.013), LDL 3.5- < 4.0 mmol/L (p = 0.020), and BMI >= 30 kg/m(2) (p = 0.033). HbA1c was the strongest risk factor for any albuminuria estimated by the measure gradient of risk per 1 SD, followed by diastolic blood pressure, triglycerides, systolic blood pressure, cholesterol and LDL. In patients with HbA1c > 65 mmol/mol (> 8.1%), blood pressure > 140/70 mmHg was associated with increased risk of albuminuria. Conclusions Preventing renal complications in adolescents and young adults with type 1 diabetes need avoidance at relatively high levels of blood pressure, blood lipids and BMI, whereas very tight control is not associated with further risk reduction. For patients with long-term poor glycaemic control, stricter blood pressure control is advocated.

  • Risk of atrial fibrillation in persons with type 2 diabetes and the excess risk in relation to glycaemic control and renal function: a Swedish cohort study

    • Background To examine the incidence of atrial fibrillation in individuals with type 2 diabetes compared with age- and sex-matched controls from the general population and its variation in relation to glycaemic control and renal function. Methods A total of 421,855 patients with type 2 diabetes from the Swedish National Diabetes Registry and 2,131,223 controls from the Swedish Population Registry, matched for age, sex and county, were included and followed from January 1, 2001 to December 31, 2013. Results Overall, 8.9% of individuals with type 2 diabetes and 7.0% of controls were diagnosed with atrial fibrillation during follow-up, unadjusted incidence risk ratio (IRR) 1.35 (95% 1.33-1.36). Women < 55 years old with type 2 diabetes had an IRR of 2.36 (95% CI 2.10-2.66), in relation to controls, whereas the corresponding value for men < 55 years old with type 2 diabetes was IRR 1.78 (95% CI 1.67-1.90). In the fully adjusted Cox regression, the risk of type 2 diabetes on incident atrial fibrillation was 28% greater vs controls, hazard ratio (HR) 1.28 (95% CI 1.26-1.30), p < 0.0001. The excess risk of atrial fibrillation in individuals with type 2 diabetes increased with worsening glycaemic control and renal complications. For individuals with HbA1c <= 6.9% (<= 52 mmol/mol) and normoalbuminuria the excess risk vs controls was still increased, adjusted HR 1.16 (95% CI 1.14-1.19); p < 0.0001. Conclusions Individuals with type 2 diabetes had an overall 35% higher risk of atrial fibrillation compared to age- and sex-matched controls from the general population. The excess risk for atrial fibrillation increased with renal complications or with poor glycaemic control. Individuals with type 2 diabetes with good glycaemic control and normoalbuminuria had slightly increased risk.

    • To implement policies about sustainable landscapes and rural development necessitates social learning about states and trends of sustainability indicators, norms that define sustainability, and adaptive multi-level governance. We evaluate the extent to which social learning at multiple governance levels for sustainable landscapes occur in 18 local development initiatives in the network of Sustainable Bergslagen in Sweden. We mapped activities over time, and interviewed key actors in the network about social learning. While activities resulted in exchange of experiences and some local solutions, a major challenge was to secure systematic social learning and make new knowledge explicit at multiple levels. None of the development initiatives used a systematic approach to secure social learning, and sustainability assessments were not made systematically. We discuss how social learning can be improved, and how a learning network of development initiatives could be realized.

  • Glycemic Control and Risk of Sepsis and Subsequent Mortality in Type 2 Diabetes

    • OBJECTIVE To investigate the nature of the relationship between HbA1c and sepsis among individuals with type 2 diabetes, and to assess the association between sepsis and all-cause mortality in such patients. RESEARCH DESIGN AND METHODS We included 502,871 individuals with type 2 diabetes recorded in the Swedish National Diabetes Register and used multivariable Cox regression and restricted cubic spline analyses to assess the association between time-updated HbA1c values and sepsis occurrence between 1 January 2005 and 31 December 2015. The association between sepsis and death was examined using multivariable Cox regression analysis. RESULTS Overall, 14,534 (2.9%) patients developed sepsis during the study period. On multivariable Cox regression analysis, compared with an HbA1c of 48–52 mmol/mol (6.5–6.9%), the adjusted hazard ratio for sepsis was 1.15 (95% CI 1.07–1.24) for HbA1c <43 mmol/mol (6.1%), 0.93 (0.87–0.99) for HbA1c 53–62 mmol/mol (7.0–7.8%), 1.05 (0.97–1.13) for HbA1c 63–72 mmol/mol (7.9–8.7%), 1.14 (1.04–1.25) for HbA1c 73–82 mmol/mol (8.8–9.7%), and 1.52 (1.37–1.68) for HbA1c >82 mmol/mol (9.7%). In the cubic spline model, a reduction of the adjusted risk was observed within the lower HbA1c range until 53 mmol/mol (7.0%), with a hazard ratio of 0.78 (0.73–0.82) per SD; it increased thereafter (P for nonlinearity <0.001). As compared with patients without sepsis, the adjusted hazard ratio for death among patients with sepsis was 4.16 (4.03–4.30). CONCLUSIONS In a nationwide cohort of individuals with type 2 diabetes, we found a U-shaped association between HbA1c and sepsis and a fourfold increased risk of death among those developing sepsis. © 2021 by the American Diabetes Association.

    • BACKGROUND: Substantial clinical data support the addition of low doses of atypical antipsychotic drugs to selective serotonin reuptake inhibitors (SSRIs) to rapidly enhance the antidepressant effect in treatment-resistant depression. Preclinical studies suggest that this effect is at least partly explained by an increased catecholamine outflow in the medial prefrontal cortex (mPFC).METHODS: In the present study we used in vivo microdialysis in freely moving rats and in vitro intracellular recordings of pyramidal cells of the rat mPFC to investigate the effects of adding the novel atypical antipsychotic drug asenapine to the SSRI escitalopram with regards to monoamine outflow in the mPFC and dopamine outflow in nucleus accumbens as well as glutamatergic transmission in the mPFC.RESULTS: The present study shows that addition of low doses (0.05 and 0.1 mg/kg) of asenapine to escitalopram (5 mg/kg) markedly enhances dopamine, noradrenaline, and serotonin release in the rat mPFC as well as dopamine release in the nucleus accumbens. Moreover, this drug combination facilitated both N-methyl-d-Aspartate (NMDA)- and α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA)-induced currents as well as electrically evoked excitatory postsynaptic potentials in pyramidal cells of the rat mPFC.CONCLUSIONS: Our results support the notion that the augmentation of SSRIs by atypical antipsychotic drugs in treatment-resistant depression may, at least in part, be related to enhanced catecholamine output in the prefrontal cortex and that asenapine may be clinically used to achieve this end. In particular, the subsequent activation of the D1 receptor may be of importance for the augmented antidepressant effect, as this mechanism facilitated both NMDA and AMPA receptor-mediated transmission in the mPFC. Our novel observation that the drug combination, like ketamine, facilitates glutamatergic transmission in the mPFC may contribute to explain the rapid and potent antidepressant effect obtained when atypical antipsychotic drugs are added to SSRIs.

    • Brexpiprazole (Rexulti(®)), a novel D2/3 receptor (R) partial agonist, was recently approved as monotherapy for schizophrenia, demonstrating effectiveness against both positive and negative symptoms, and also approved as add-on treatment to antidepressant drugs, inducing a potent antidepressant effect with a faster onset compared to an antidepressant given alone. Moreover, brexpiprazole has demonstrated pro-cognitive effects in preclinical studies. To explore whether the observed effects may be mediated via modulation of prefrontal glutamatergic transmission, we investigated the effect of brexpiprazole, alone and in combination with the SSRI escitalopram, on prefrontal glutamatergic transmission using in vitro electrophysiological intracellular recordings of deep layer pyramidal cells of the rat medial prefrontal cortex (mPFC). Nanomolar concentrations of brexpiprazole potentiated NMDAR-induced currents and electrically evoked EPSPs via activation of dopamine D1Rs, in similarity with the effect of the atypical antipsychotic drug clozapine. The effect of an ineffective concentration of brexpiprazole was significantly potentiated by the addition of escitalopram. When combined with escitalopram, brexpiprazole also potentiated AMPAR-mediated transmission, in similarity with the clinically rapid acting antidepressant drug ketamine. The effect on the AMPAR-mediated currents was also D1R dependent. In conclusion, our data propose that brexpiprazole exerts a clozapine-like potentiation of NMDAR-mediated currents in the mPFC, which can explain its efficacy on negative symptoms of schizophrenia and the pro-cognitive effects observed preclinically. Moreover, add-on brexpiprazole to escitalopram also potentiated AMPAR-mediated transmission, which may provide a neurobiological explanation to the faster antidepressant effect of add-on brexpiprazole in major depression.

    • Despite rapid advances in development of the ecosystem services (ES) concept, challenges remain for its use in decision making. Cultural ES (CES) have proven particularly difficult to pin down and resultant “shades of grey” impede their consideration by decision-makers. This study undertakes a literature review of CES to highlight the shades of grey, briefly illustrates findings by reference to the Swedish mountain landscape, then addresses potential implications for practical decision making. The concept of CES is complex and difficult to operationalize. The root of confusion appears to be a lack of rigour in identifying CES, hindering identification of proper methods for determining: the ecosystem elements that underpin CES; the beneficiaries of CES and how they value benefits delivered; and how CES may vary in space and time. We conclude by proposing a framework of questions, which we relate to the ES cascade model, that is intended to help researchers and decision-makers to reflect when considering CES. Answers to the questions should enable decision-makers to prioritise policy development or implementation in relation to the differing needs of potentially competing beneficiaries and what needs to be done or not done to the ecosystem, where, when and by whom.

    • OBJECTIVE: Atherosclerotic lesions have regions that are hypoxic. Because the lesion contains macrophages that are loaded with lipid, we investigated whether hypoxia can influence the accumulation of lipids in these cells. METHODS AND RESULTS: Exposure of human macrophages to hypoxia for 24 hours resulted in an increased formation of cytosolic lipid droplets and an increased accumulation of triglycerides. Exposure of the macrophages to oxidized low-density lipoprotein (oxLDL) increased the accumulation of cytosolic lipid droplets because of an increase in cellular cholesterol esters. The accumulation of lipid droplets in oxLDL-treated cells was further increased after hypoxia, caused by an increased level of triglycerides. Expression analyses combined with immunoblot or RT-PCR demonstrated that hypoxia increased the expression of several genes that could promote the accumulation of lipid droplets. Hypoxia increased the mRNA and protein levels of adipocyte differentiation-related protein (ADRP). It is well known that an increased expression of ADRP increases the formation of lipid droplets. Hypoxia decreased the expression of enzymes involved in beta-oxidation (acyl-coenzyme A synthetase and acyl-coenzyme A dehydrogenase) and increased the expression of stearoyl-coenzyme A desaturase, an important enzyme in the fatty acid biosynthesis. Moreover, exposure to hypoxia decreased the rate of beta-oxidation, whereas the accumulation of triglycerides increased. CONCLUSIONS: The results demonstrate that exposure of human macrophages to hypoxia causes an accumulation of triglyceride-containing cytosolic lipid droplets. This indicates that the hypoxia present in atherosclerotic lesions can contribute to the formation of the lipid-loaded macrophages that characterize the lesion and to the accumulation of triglycerides in such lesions.

    • Several experimental evidences suggested that beta1 integrin-mediated adhesion of hematopoietic stem cells (HSC) is important for their function in the bone marrow (BM). Using induced deletion of the beta1 integrin gene restricted to the hematopoietic system, we show that beta1 integrin is not essential for HSC retention in the BM, hematopoiesis, and trafficking of lymphocytes. However, immunization with a T cell-dependent antigen resulted in virtually no IgM production and an increased secretion of IgG in mutant mice, while the response to a T cell-independent type 2 antigen showed decreases in both IgM and IgG. These data suggest that beta1 integrins are necessary for the primary IgM antibody response.

    • Från förordet: Årets rapport kartlägger det svenska entreprenörskapets utveckling under en turbulent tid präglad av pandemi, restriktioner och nedstängningar av hela samhällen vilket du kan läsa om i kapitel två. Trots pandemin ökar svenskt entreprenörskap under 2021 till nio procent (TEA) vilket är den högsta siffran som redovisats i GEM-projektet för Sveriges del sedan 2010. En minst sagt glädjande siffra.Det är emellertid inte en anledning att luta sig tillbaka. Världen står inför stora utmaningar i form av komponentbrist, störningar i leveranskedjor samt en oroande geopolitisk situation, inte minst i och med Rysslands invasion av Ukraina. Därtill kan tilläggas att Sverige dras med olösta och kända problem som till exempel en dysfunktionell arbetsmarknad präglad av kompetensbrist kombinerat med hög arbetslöshet och låg självförsörjningsgrad bland stora delar av befolkningen.I årets rapport görs även nedslag i entreprenörernas relation till miljömässig- och social hållbarhet. Svenska entreprenörer uppvisar på intet sätt det största engagemanget för bland annat miljömässig hållbarhet globalt sett. Men det finns anledning att tro att det åtminstone delvis kan bero på att företagare i Sverige ställer högre krav inom sin verksamhet, speciellt jämfört med företagare i utvecklingsländer.

    • KTH, Norrköpings kommun och Trafikverket deltar i det europeiska FoI-projektet Triple Access Planning for Uncertain Futures med fokus på hur scenarieplanering kan utgöra ett verktyg för att utforska hur tillgängligheten kan utvecklas, givet stora osäkerheter och komplexitet i samhällsutvecklingen. Projektet bygger kunskap om hur ett brett fokus på tillgänglighet, som en kombination av mobilitet, rumslig närhet och digitala lösningar, kan påverka strategisk trafik- och samhällsplanering och bidra till en mer robust och hållbar samhällsutveckling. Med andra ord: Hur skapar vi institutionell kapacitet för tillgänglighet scenarioplanering för hållbar urban transportplanering? I en workshopserie under hösten 2022 och våren 2023 prövade KTH en scenarioansats för att tillsammans med planerare i Norrköpings kommun utforska framtida urban tillgänglighet n och vilka konsekvenser det skulle kunna få för stads- och trafikutvecklingen. Norrköping är en av sju städer i Europa som deltar i TAP-projektet med liknande undersökningar. Projektet tillämpar kvalitativ scenariometodik, med successiv och gemensam reflektion och utvärdering, tillsammans med trafik- och samhällsplanerare i Norrköpings kommun. Arbetet har gett insikter om förutsättningar och behov för att med scenarioplanering utforska framtida tillgänglighet i en urban geografi, som rör bl.a. följande organisatoriska och institutionella teman: planeringssammanhangets avgränsning och inramning; arbetets integrering med etablerade processer och målbilder; tillgång till data om tillgänglighet; svårigheter att avgränsa osäkerheter som interna eller externa, samt grad av integrering av berörda aktörer och professioner, horisontellt och vertikalt. Resultaten visar på institutionella osäkerheter och behov av stärkt institutionell kapacitet som påverkar förutsättningarna att med scenarier stärka analyser och planering för framtida hållbar tillgänglighet.

The focus of this paper is to identify and study the approximation curve of a class of threshold predicates that allow for non-trivial approximation. Arguably the simplest such predicate is the majority predicate sgn(x 1+⋯+xn ), for which we obtain an almost complete understanding of the asymptotic approximation curve, assuming the Unique Games Conjecture. Our techniques extend to a more general class of "majority-like" predicates and we obtain parallel results for them. In order to classify these predicates, we introduce the notion of Chow-robustness that might be of independent interest.

    • The marketplace for probiotic foods is burgeoning, measured in billions of euro per annum. It is imperative, however, that all bacterial strains are fully assessed for human safety. The ability to bind fibrinogen is considered a potential pathogenicity trait that can lead to platelet aggregation, serious medical complications, and in some instances, death. Here we examined strains from species frequently used as probiotics for their ability to bind human fibrinogen. Only one strain (CCUG 47825), a Lactobacillus salivarius isolate from a case of septicaemia, was found to strongly adhere to fibrinogen. Furthermore, this strain was found to aggregate human platelets at a level comparable to the human pathogen Staphylococcus aureus. By sequencing the genome of CCUG 47825, we were able to identify candidate genes responsible for fibrinogen binding. Complementing the genetic analysis with traditional molecular microbiological techniques enabled the identification of the novel fibrinogen receptor, CCUG_2371. Although only strain CCUG 47825 bound fibrinogen under laboratory conditions, homologues of the novel fibrinogen binding gene CCUG_2371 are widespread among L. salivarius strains, maintaining their potential to bind fibrinogen if expressed. We highlight the fact that without a full genetic analysis of strains for human consumption, potential pathogenicity traits may go undetected. © 2012 Blackwell Publishing Ltd.

  • Potential for the valorization of carbon dioxide from biogas production in Sweden

    • Biogas solutions offer many advantages to improve sustainable development, but there is still untapped potential in its environmental performance. During biogas upgrading, CO2 is separated from the gas to deliver a flow with high methane concentration and thus high energy content. In this practice, CO2 is commonly emitted to the atmosphere without contributing to a net addition of climate gases because of its biological origin, being a missed opportunity for carbon capture. In this paper, CO2 valorization is an option that has been evaluated using a qualitative and quantitative approach, taking Sweden as an example. Results showed that around 140 kt of CO2 can potentially be captured and utilized from biogas upgrading, which can significantly increase in future scenarios. If CO2 were turned into methane using power-to-gas technology, an additional 35% of biogas could be produced in the short term, meaning up to additional 0.7 TWh in 2020. By 2050, around 600 to 1600 kt of CO2 could be available, depending on how well the biogas production develops and how much of the biogas is upgraded, resulting in up to 6.2 TWh of biomethane. The qualitative assessment suggested that only minor modifications in the upgrading process are required for this practice. Biogas actors are interested in getting involved in valorization projects that enhance their circular business and avoid carbon lock-in mainly to improve the environmental performance of biomethane. Moreover, the application of CO2 valorization requires collaboration with different actors to integrate current CO2 demand or innovative transformation technologies.

    • Biogas solutions offer many benefits for the environment and society, including organic waste treatment as well as being an enabler for energy and nutrient recovery. The products of anaerobic digestion are a biogas, which contains a share of 30 to 50% carbon dioxide (CO2) and 50 to 70% methane, and a liquid remanent, rich in nutrients. The biogas can be upgraded by removing the CO2 to increase the energy content, producing biomethane. At present, CO2 is considered a waste in biomethane production systems, and hence it is emitted into the atmosphere. Nevertheless, biogas upgrading technologies separate a pure-grade CO2 and, likewise, carbon capture processes, providing a pure CO2 flow that can be stored or utilized. Compared to storage, carbon capture and utilization (CCU) technologies deliver valuable carbon-based products required to sustain human activities. The valorization of green CO2 could aid the transition towards defossilization of the economy. Indeed, several CO2 utilization technologies could be incorporated into biomethane production systems, but there is still a limited understanding of the available alternatives and their potential impacts on biomethane systems.This thesis aims to investigate the integration of CO2 utilization technologies in biomethane production systems by revealing its potential, identifying alternatives, and assessing the impacts of the integration. Using Sweden as an example, scenarios of future biomethane production were employed to estimate the potential CO2 available for utilization. To complement the analysis, a qualitative approach made possible the identification of aspects that could affect CO2 utilization in biomethane production. Moreover, a multi-criteria analysis (MCA) framework was developed to identify relevant indicators for assessment and available alternatives for CO2 utilization. The research also includes a life cycle assessment (LCA) to evaluate the climate performance of relevant CCU alternatives in the biomethane production system.Results show that 160 kt of CO2 could be obtained from biomethane production in Sweden, which could potentially increase threefold from 2020 to 2030. The evaluation of alternatives for CO2 utilization includes environmental, technical, economic, and social criteria with sound indicators within an MCA framework. Indicators to evaluate each criterion provide valuable information to identify feasible and sustainable alternatives that can be integrated into biomethane plants. The identified alternatives with a high readiness level are additional methane through methanation, horticulture, mineral carbonates, fuels, pH control, bulk chemicals, and liquefied CO2 for direct use. The results provide information to decision-makers in relation to considerations to take before implementation, like energy requirements, the existence of regulations and standards, and uncertainty. In terms of the climate performance of biomethane with the inclusion of CCU alternatives, the results show a possible reduction of CO2 emissions that depends on the possibility of substituting fossil-based products. The investigated alternatives all result in lower emissions, but concrete curing and methanation using renewable hydrogen produce the best results.To conclude, the potential future increase of green CO2 from biomethane in Sweden creates opportunities to substitute fossil carbon in current applications and mature conversion pathways. Moreover, the inclusion of CCU in biomethane production contributes to reducing biomethane system emissions and diversifying its products. Possible alternatives of CCU that can be integrated into biomethane production systems in the short term include methanation and concrete curing. Other alternatives could be possible but present lower performance and higher uncertainties at the moment.

    • Carbon capture and utilization has been proposed as an essential climate change mitigation strategy, but only a few implemented cases exist. During biomethane production from anaerobic digestion, CO₂ is commonly separated and emitted into the atmosphere, which can be utilized as raw material for various products. This research aims to identify and assess CO₂ utilization alternatives for possible integration with biogas upgrading from anaerobic digestion by developing a soft multi-criteria analysis (MCA). A literature review complemented with stakeholder participation enabled the identification of relevant alternatives and criteria for assessment. Potential alternatives for CO₂ utilization include methane, mineral carbonates, biomass production, fuels, chemicals, pH control, and liquefied CO₂. Results show that although no alternative performs well in all indicators, there is an opportunity for short-term implementation for methane, biomass production, mineral carbonates, liquefied CO₂, and pH control. Moreover, the uncertainty analysis reveals that even though the technologies have a high technological development, more information on critical aspects is still required. The soft MCA provides information to decision-makers, practitioners, and the academic community on learning opportunities of the alternatives and indicators to step from development into implementation. For instance, the method can be used to assess more specific systems with different locations and scales or to direct efforts to ease the implementation of CCU.

    • Background Type 1 diabetes is associated with an increased risk of developing several cardiovascular complications. To our knowledge, the independent association between type 1 diabetes and atrial fibrillation has not been studied. Methods We did a prospective case-control study of individuals with type 1 diabetes in the Swedish National Diabetes Registry who were each matched with five controls for age, sex, and county of residence who were randomly selected from the Swedish Population Register. Cases of atrial fibrillation were obtained from the Swedish National Patient Registry. Findings We followed up 36 258 patients with type 1 diabetes and 179 980 controls between Jan 1, 2001, and Dec 31, 2013. Median follow-up was 9.7 years (IQR 5.2-13.0) for patients and 10.2 years (5.7-13.0) for controls. 749 (2%) individuals with type 1 diabetes and 2882 (2%) controls were diagnosed with atrial fibrillation, with an adjusted hazard ratio (HR) of 1.13 (95% CI 1.01-1.25; p = 0.029) in men and 1.50 (1.30-1.72; p < 0.0001) in women (p = 0.0019 for interaction). The excess risk of atrial fibrillation in individuals with type 1 diabetes increased with worsening glycaemic control and renal complications. Among individuals with normoalbuminuria, no excess risk of atrial fibrillation was noted in men with type 1 diabetes who had HbA(1c) lower than 9.7% (< 83 mmol/mol) or in women with type 1 diabetes who had HbA(1c) lower than 8.8% (< 73 mmol/mol). Interpretation Compared with the general population, the risk of atrial fibrillation in men with type 1 diabetes was slightly raised, whereas for female patients it was 50% higher. The risk of atrial fibrillation in people with type 1 diabetes increased with renal complications and poor glycaemic control.

Methods: In a 3-year longitudinal study, obese children (n = 684) were divided into three groups based on age at the start of treatment, age 6-9 years, 10-13 years, and 14-16 years. Results: The mean BMI standard deviation score (BMI-SDS) decline was age-dependent (p = 0.001), independently of adjustment for missing data: -1.8 BMI-SDS units in the youngest, -1.3 in the middle age group, and -0.5 in the oldest age group. SES and parental BMI status did not affect the results. 30% of the adolescents remained in treatment at year 3. There was only a weak correlation between BMI-SDS change after 1 and 3 years: r = 0.51 (p < 0.001). Among children with no BMI-SDS reduction during year 1 (n = 46), 40% had a clinically significantly reduced BMI-SDS after year 3. Conclusion: Behavioral treatment should be initiated at an early age to increase the chance for good results.     • The aim of the present study was to synthesise and screen a set of novel nicotine hapten immunogens used for the treatment of nicotine dependence. In the screening process we studied the amount of antibodies generated and their selectivity, using ELISA techniques, and their effects on nicotine-induced dopamine release in the NAC(shell) of the rat, assessed by in vivo voltammetry. We conclude that even small changes such as the linker attachment on the nicotine molecule as well as the structure of the linker may greatly influence the selectivity of the antibodies and the central neurobiological effects of nicotine that are considered critical for its dependence producing properties.     • Introduction Renal complications are both a marker of previous suboptimal glycaemic control and a major risk factor for cardiovascular disease in persons with type 1 diabetes (T1D). The aim of the study was to evaluate the prevalence of renal complications in persons with T1D in four geographical regions. Methods Nationwide registry data from Austria/Germany, Sweden and the US were used to estimate the prevalence of renal complications from January 2016 until September 2018. Chronic kidney disease (CKD) and albuminuria in the study population and each registry were analysed by diabetes duration. Risk factors for renal complications were described by registry. Results In the total cohort of 78.926 adults with T1D, mean age was 44.4 +/- 18.43 years and mean diabetes duration was 21.6 +/- 22 years. Mean estimated glomerular filtration rate (eGFR) was 94.0 +/- 31.45 ml/min, 13.0% had microalbuminuria and 3.9% had macroalbuminuria. Mean age, diabetes duration, use of insulin pumps and continuous glucose monitoring, as well as presence of albuminuria, varied between registries. Albuminuria was present in approximately 10% of persons with diabetes duration < 20 years and impaired renal function (eGFR < 60 ml/min) was present in 17%. In persons with diabetes duration > 40 years, approximately one-third had albuminuria and 25% had impaired renal function. Conclusions This analysis used three nationwide registries of persons with T1D. Despite recent use of more effective diabetes therapies, a substantial proportion of persons with T1D have renal complications at < 20 years after diagnosis. Efficient glucose-lowering and renal-protective strategies are needed in persons with T1D.

    • På senare år har allt fler främmande, invasiva arter börjat breda ut sig i våra sötvatten. En främmande art är en organism som har spridits utanför sitt naturliga utbredningsområde med människans hjälp. Detta kan ha skett avsiktligt eller av misstag. En del av dessa nya arter ställer till problem, andra gör det inte. Här beskrivs övervakningen av några invasiva arter samt hur man kan arbeta för att begränsa dem. Ullhandskrabba, vandrarmussla och sjögull kommer av allt att döma att öka sin utbredning i Sverige de närmaste åren. Det finns idag ett stort antal främmande arter rapporterade från svenska sötvatten och i vår närhet finns ett femtiotal nya arter som kan tänkas etablera sig här. Alla dessa arter kommer inte nödvändigtvis att skapa problem men kunskap om deras biologi och observationer från allmänheten kommer att betyda mycket även i det framtida övervakningsarbetet.

    • OBJECTIVE: Type 2 diabetes is strongly associated with obesity, but the mortality risk related to elevated body weight in people with type 2 diabetes compared with people without diabetes has not been established. RESEARCH DESIGN AND METHODS: We prospectively assessed short- and long-term mortality in people with type 2 diabetes with a recorded diabetes duration /=40 kg/m(2) compared with control subjects after multiple adjustments. Long-term, all weight categories showed increased mortality, with a nadir at BMI 25 to <30 kg/m(2) and a stepwise increase up to HR 2.00 (95% CI 1.58-2.54) among patients with BMI >/=40 kg/m(2), that was more pronounced in patients <65 years old. CONCLUSIONS: Our findings suggest that the apparent paradoxical findings in other studies in this area may have been affected by reverse causality. Long-term, overweight (BMI 25 to <30 kg/m(2)) patients with type 2 diabetes had low excess mortality risk compared with control subjects, whereas risk in those with BMI >/=40 kg/m(2) was substantially increased.

    • OBJECTIVE Low weight has been associated with increased mortality risks in type 1 diabetes. We aimed to investigate the importance of weight and weight gain/loss in the Swedish population diagnosed with type 1 diabetes. RESEARCH DESIGN AND METHODS Patients with type 1 diabetes (n = 26,125; mean age 33.3 years; 45% women) registered in the Swedish National Diabetes Registry from 1998 to 2012 were followed from the first day of study entry. Cox regression was used to calculate risk of death from cardiovascular disease (CVD), major CVD events, hospitalizations for heart failure (HF), and total deaths. RESULTS Population mean BMI in patients with type 1 diabetes increased from 24.7 to 25.7 kg/m(2) from 1998 to 2012. Over a median follow-up of 10.9 years, there were 1,031 deaths (33.2% from CVD), 1,460 major CVD events, and 580 hospitalizations for HF. After exclusion of smokers, patients with poor metabolic control, and patients with a short follow-up time, there was no increased risk for mortality in those with BMI <25 kg/m(2), while BMI >25 kg/m(2) was associated with a minor increase in risk of mortality, major CVD, and HF. In women, associations with BMI were largely absent. Weight gain implied an increased risk of mortality and HF, while weight loss was not associated with higher risk. CONCLUSIONS Risk of major CVD, HF, CVD death, and mortality increased with increasing BMI, with associations more apparent in men than in women. After exclusion of factors associated with reverse causality, there was no evidence of an obesity paradox.

    • Background Body mass index (BMI) may be a stronger risk factor for heart failure than for coronary heart disease in type 2 diabetes mellitus, but prior studies have not been powered to investigate the relative and absolute risks for acute myocardial infarction and heart failure in type 2 diabetes mellitus by BMI and glycemic level combined as compared with age- and sex-matched general population comparators. Methods and Results We identified 181045 patients from The Swedish National Diabetes Registry, registered during 1998 to 2012 and 1538434 general population comparators without diabetes mellitus, matched for age, sex, and county, all without prior major cardiovascular disease. Cases and comparators were followed with respect to the outcomes through linkage to the Swedish Inpatient Registry. Over a median follow-up time of 5.7years, there were 28855 acute myocardial infarction and 33060 heart failure cases among patients and comparators. Excess risk (above that of comparators in whom no data on hemoglobin A1c and BMI was available), incidence rates and hazard ratios for heart failure were substantially higher among the obese patients compared with those with low BMI, where very obese patients (BMI ≥40kg/m2) who also had poor glycemic control, suffered a 7-fold risk of heart failure versus comparators (reference level). By contrast, for acute myocardial infarction, the highest absolute and relative risks were found among patients with poor glycemic control, with no additional risk conferred by increasing BMI. Conclusions BMI is a strong independent risk factor for heart failure but not for acute myocardial infarction among patients with type 2 diabetes mellitus.

    • In type 1 diabetes, potential loss of life-years is greatest in those who are youngest at the time of onset. Using data from a nationwide cohort of patients with type 1 diabetes, we aimed to study risk factor trajectories by age at diagnosis. We stratified 30005 patients with type 1 diabetes aged 18–75 years into categories based on age at onset: 0–10, 11–15, 16–20, 21–25, and 26–30 years. HbA1c, albuminuria, estimated glomerular filtration rate (eGFR), body mass index (BMI), low-denisty lipoprotein (LDL)-cholesterol, systolic blood pressure (SBP), and diastolic blood pressure trends were analyzed using mixed models. Variable importance for baseline HbA1c was analyzed using conditional random forest and gradient boosting machine approaches. Individuals aged ≥16 years at onset displayed a relatively low mean HbA1c level (~55–57mmol/mol) that gradually increased. In contrast, individuals diagnosed at ≤15 years old entered adulthood with a mean HbA1c of approximately 70mmol/mol. For all groups, HbA1c levels stabilized at a mean of approximately 65mmol/mol by about 40 years old. In patients who were young at the time of onset, albuminuria appeared at an earlier age, suggesting a more rapid decrease in eGFR, while there were no distinct differences in BMI, SBP, and LDL-cholesterol trajectories between groups. Low education, higher age, and poor risk factor control were associated with higher HbA1c levels. Young age at the diabetes onset plays a substantial role in subsequent glycemic control and the presence of albuminuria, where patients with early onset may accrue a substantial glycemic load during this period. © Author(s) (or their employer(s)) 2021.
