    • Sulphate records from Greenland ice-cores indicate that Marine Isotope Stages 4 and 5 were charactensed by a higher incidence of large volcanic eruptions than other periods during the last glacial period, however, few investigations have focused on tephra deposits associated with these volcanic eruptions and the nature and origin of the events. Here we present a detailed tephrochronological framework of the products of 15 volcanic events spanning this interval: the majority of which have been preserved as cryptotephra horizons within the Greenland records. The major element compositions of individual glass shards within these horizons indicate that 13 of the eruptions originated from Iceland and 6 of these events can be correlated to the specific volcanic systems of Katla, Grimsvotn, Grimsvotn-Kverkfjoll and either Reykjanes or Veidivotn-Bardarbunga. For the remaining Icelandic horizons a source from either the rift zone or a flank zone can be suggested based on rock suite affinities. Two horizons have been correlated to a source from the Jan Mayen volcanic system which represents the first discovery of material from this system within any Greenland ice-cores. The robust geochemical characterisations, independent ages for these horizons (derived from the GICCO5 ice-core chronology) and stratigraphic positions relative to the Dansgaard-Oeschger climate events recorded in the Greenland ice-cores represent a critical framework that provides new information on the frequency and nature of volcanic events occurring in the North Atlantic region during MIS 4 and 5. This framework can now be utilised in the assessment of the differential timing and rate of response to the millennial-scale climatic events that characterised this period, through the use of the tephra horizons as time-synchronous tie-lines to other palaeoclimatic sequences.

  • Controlling the carbon vacancy in 4H-SiC by thermal processing

    • The carbon vacancy (Vc) is perhaps the most prominent point defect in silicon carbide (SiC) and it is an efficient charge carrier lifetime killer in high-purity epitaxial layers of 4H-SÌC. The Vc concentration needs to be controlled and minimized for optimum materials and device performance, and an approach based on post-growth thermal processing under C-rich ambient conditions is presented. It utilizes thermodynamic equilibration and after heat treatment at 1500 °C for 1 h, the Vc concentration is shown to be reduced by a factor-25 relative to that in as-grown state-of-the-art epi-layers. Concurrently, a considerable enhancement of the carrier lifetime occurs throughout the whole of >40 urn thick epi-layers.

    • A visible tephra horizon in the NGRIP ice core has been identified by geochemical analysis as the Fugloyarbanki Tephra, a widespread marker horizon in marine cores from the Faroe Islands area and the northern North Atlantic. An age of 26 740 ± 390 yr b2k (1σ uncertainty) is derived for this tephra according to the new Greenland Ice Core Chronology (GICC05) based on multi-parameter counting of annual layers. Detection of this tephra for the first time within the NGRIP ice core provides a key tie-point between marine and ice-core records during the transition between MIS 3 and 2. Identification of this volcanic event within the Greenland records demonstrates the future potential of using tephrochronology to precisely correlate palaeoarchives in widely separated localities that span the last glacial period, as well as providing a potential method for examining the extent of the radiocarbon marine reservoir effect at this time.

    • The aim of the present study was to investigate seasonal and spatial patterns of soil oxygen consumption, nitrification, denitrification and fluxes of dissolved inorganic nitrogen (DIN) in a tidal salt marsh of the Lagoon of Venice, Italy. In the salt marsh, intact soil cores including overlying water were collected monthly at high tide from April to October in salt marsh creeks and in areas covered by the dominant vegetation, Limonium serotinum. In May, cores were also collected in areas with vegetation dominated by Juncus maritimus and Halimione portulacoides. In laboratory incubations at in situ temperature in the dark, flux rates of oxygen and DIN were monitored in the overlying water of the intact cores. 15N-nitrate was added to the overlying water and nitrification and denitrification were measured using isotope-dilution and -pairing techniques. The results show that highest soil oxygen consumption coincided with the highest water temperature in June and July. The highest denitrification rates were recorded in spring and autumn coinciding with the highest nitrate concentrations. Soil oxygen consumption and nitrification rates differed between sampling sites, but denitrification rates were similar among the different vegetation types. The highest rates were recorded in areas covered with L. serotinum. Burrowing soil macrofauna enhanced oxygen consumption, nitrification and denitrification in April and May. The data presented in this study indicate high temporal as well as spatial variations in the flux of oxygen and DIN, and nitrogen transformations in the tidal salt marshes of the Venice lagoon during the growth season. The results identify the salt marshes of the Venice lagoon as being metabolically very active ecosystems with a high capacity to process nitrogen.

    • Ion implantation is an important technique for a successful implementation of commercial SiC devices. Much effort has also been devoted to optimising implantation and annealing parameters to improve the electrical device characteristics. However, there is a severe lack of understanding of the fundamental implantation process and the generation and annealing kinetics of point defects and defect complexes. Only very few of the most elementary intrinsic point defects have been unambiguously identified so far. To reach a deeper understanding of the basic mechanisms SiC samples have been implanted with a broad range of ions, energies, doses, etc., and the resulting defects and damage produced in the lattice have been studied with a multitude of characterisation techniques. In this contribution we will review some of the results generated recently and also try to indicate where more research is needed. In particular, deep level transient spectroscopy (DLTS) has been used to investigate point defects at very low doses and transmission electron microscopy (TEM) and Rutherford backscattering spectrometry (RBS) are used for studying the damage build-up at high doses.

    • IRL790 ([2-(3-fluoro-5-methanesulfonylphenoxy)ethyl](propyl)amine, mesdopetam) is a novel compound in development for the clinical management of motor and psychiatric disabilities in Parkinson disease. The discovery of IRL790 was made applying a systems pharmacology approach based on in vivo response profiling. The chemical design idea was to develop a new type of DA D3/D2 receptor type antagonist built on agonist rather than antagonist structural motifs. We hypothesized that such a dopamine antagonist with physicochemical properties similar to agonists would exert antidyskinetic and antipsychotic effects in states of dysregulated dopaminergic signaling while having little negative impact on physiologic dopamine transmission and, hence, minimal liability for side effects related to dopamine-dependent functions. At the level of in vivo pharmacology, IRL790 displays balancing effects on aberrant motor phenotypes, reducing L-DOPA-induced dyskinesias in the rodent 6-hydroxydopamine lesion model and reducing psychostimulant-induced locomotor hyperactivity elicited by pretreatment with either d-amphetamine or dizocilpine, without negatively impacting normal motor performance. Thus, IRL790 has the ability to normalize the behavioral phenotype in hyperdopaminergic as well as hypoglutamatergic states. Neurochemical and immediate early gene (IEG) response profiles suggest modulation of DA neurotransmission, with some features, such as increased DA metabolites and extracellular DA, shared by atypical antipsychotics and others, such as increased frontal cortex IEGs, unique to IRL790. IRL790 also increases extracellular levels of acetylcholine in the prefrontal cortex and ventral hippocampus. At the receptor level, IRL790 appears to act as a preferential DA D3 receptor antagonist. Computational docking studies support preferential affinity at D3 receptors with an agonist-like binding mode. SIGNIFICANCE STATEMENT This paper reports preclinical pharmacology along with molecular modeling results on IRL790, a novel compound in clinical development for the treatment of motor and psychiatric complications in advanced Parkinson disease. IRL790 is active in models of perturbed dopaminergic and glutamatergic signaling, including rodent 6-hydroxydopamine L-DOPA-induced dyskinesias and psychostimulant-induced hyperactivity, in a dose range that does not impair normal behavior. This effect profile is attributed to interactions at dopamine D2/D3 receptors, with a 6- to 8-fold preference for the D3 subtype.

    • Prediction of 3D structures of membrane proteins, and of G-protein coupled receptors (GPCRs) in particular, is motivated by their importance in biological systems and the difficulties associated with experimental structure determination. In the present study, a novel method for the prediction of 3D structures of the membrane-embedded region of helical membrane proteins is presented. A large pool of candidate models are produced by repacking of the helices of a homology model using Monte Carlo sampling in torsion space, followed by ranking based on their geometric and ligand-binding properties. The trajectory is directed by weak initial restraints to orient helices towards the original model to improve computation efficiency, and by a ligand to guide the receptor towards a chosen conformational state. The method was validated by construction of the β1 adrenergic receptor model in complex with (S)-cyanopindolol using bovine rhodopsin as template. In addition, models of the dopamine D2 receptor were produced with the selective and rigid agonist (R)-N-propylapomorphine ((R)-NPA) present. A second quality assessment was implemented by evaluating the results from docking of a library of 29 ligands with known activity, which further discriminated between receptor models. Agonist binding and recognition by the dopamine D2 receptor is interpreted using the 3D structure model resulting from the approach. This method has a potential for modeling of all types of helical transmembrane proteins for which a structural template with sequence homology sufficient for homology modeling is not available or is in an incorrect conformational state, but for which sufficient empirical information is accessible.

  • Investigation of D1 receptor-agonist interactions and D1/D2 agonist selectivity using a combination of pharmacophore and receptor homology modelling

    • The aim of this study was to use a combined structure and pharmacophore modeling approach to extract information regarding dopamine D1 receptor agonism and D1/D2 agonist selectivity. A 3D structure model of the D1 receptor in its agonist-bound state was constructed with a full D1 agonist present in the binding site. Two different binding modes were identified using (+)-doxanthrine or SKF89626 in the modeling procedure. The 3D model was further compared with a selective D1 agonist pharmacophore model. The pharmacophore feature arrangement was found to be in good agreement with the binding site composition of the receptor model, but the excluded volumes did not fully reflect the shape of the agonist binding pocket. A new receptor-based pharmacophore model was developed with forbidden volumes centered on atom positions of amino acids in the binding site. The new pharmacophore model showed a similar ability to discriminate as the previous model. A comparison of the 3D structures and pharmacophore models of D1 and D2 receptors revealed differences in shape and ligand-interacting features that determine selectivity of D1 and D2 receptor agonists. A hydrogen bond pharmacophoric feature (Ser-TM5) was shown to contribute most to the selectivity. Non-conserved residues in the binding pocket that strongly contribute to D1/D2 receptor agonist selectivity were also identified; those were Ser/Cys3.36, Tyr/Phe5.38, Ser/Tyr5.41, and Asn/His6.55 in the transmembrane (TM) helix region, together with Ser/Ile and Leu/Asn in the second extracellular loop (EC2). This work provides useful information for the design of new selective D1 and D2 agonists. The combined receptor structure and pharmacophore modeling approach is considered to be general, and could therefore be applied to other ligand–protein interactions for which experimental information is limited.

    • This study is focused on the identification of structural features that determine the selectivity of dopamine receptor agonists toward D(1) and D(2) receptors. Selective pharmacophore models were developed for both receptors. The models were built by using projected pharmacophoric features that represent the main agonist interaction sites in the receptor (the Ser residues in TM5 and the Asp in TM3), a directional aromatic feature in the ligand, a feature with large positional tolerance representing the positively charged nitrogen in the ligand, and sets of excluded volumes reflecting the shapes of the receptors. The sets of D(1) and D(2) ligands used for modeling were carefully selected from published sources and consist of structurally diverse, conformationally rigid full agonists as active ligands together with structurally related inactives. The robustness of the models in discriminating actives from inactives was tested against four ensembles of conformations generated by using different established methods and different force fields. The reasons for the selectivity can be attributed to both geometrical differences in the arrangement of the features, e.g., different tilt angels of the pi system, as well as shape differences covered by the different sets of excluded volumes. This work provides useful information for the design of new D(1) and D(2) agonists and also for comparative homology modeling of D(1) and D(2) receptors. The approach is general and could therefore be applied to other ligand-protein interactions for which no experimental protein structure is available.

    • Universal stress proteins (Usps) are found in all kingdoms of life and can be divided into four classes by phylogenic analysis. According to available structures, Usps exist as homodimers, and genetic studies show that their cellular assignments are extensive, including functions relating to stress resistance, carbon metabolism, cellular adhesion, motility, and bacterial virulence. We approached the question of how Usps can achieve such a variety of functions in a cell by using a new procedure for statistical analysis of multiple sequence alignments, based on physicochemically related values for each amino acid residue of Usp dimer interfaces. The results predicted that Usp proteins within a class may, in addition to forming homodimers, be able to form heterodimers. Using Escherichia coli Usps as model proteins, we confirmed the existence of such interactions. We especially focused on class I UspA and UspC and demonstrated that they are able to form homo- and heterodimers in vitro and in vivo. We suggest that this ability to form both homo- and heterodimers may allow for an expansion of the functional repertoire of Usps and explains why organisms usually contain multiple usp paralogues.

    • This study reports the chemical composition of particles present along Greenland's North Greenland Eemian Ice Drilling (NEEM) ice core, back to 110,000years before present. Insoluble and soluble particles larger than 0.45 mu m were extracted from the ice core by ice sublimation, and their chemical composition was analyzed using scanning electron microscope and energy dispersive X-ray spectroscopy and micro-Raman spectroscopy. We show that the dominant insoluble components are silicates, whereas NaCl, Na2SO4, CaSO4, and CaCO3 represent major soluble salts. For the first time, particles of CaMg(CO3)(2) and Ca(NO3)(2)center dot 4H(2)O are identified in a Greenland ice core. The chemical speciation of salts varies with past climatic conditions. Whereas the fraction of Na salts (NaCl+Na2SO4) exceeds that of Ca salts (CaSO4+CaCO3) during the Holocene (0.6-11.7kyr B.P.), the two fractions are similar during the BOlling-AllerOd period (12.9-14.6kyr B.P.). During cold climate such as over the Younger Dryas (12.0-12.6kyr B.P.) and the Last Glacial Maximum (15.0-26.9kyr B.P.), the fraction of Ca salts exceeds that of Na salts, showing that the most abundant ion generally controls the salt budget in each period. High-resolution analyses reveal changing particle compositions: those in Holocene ice show seasonal changes, and those in LGM ice show a difference between cloudy bands and clear layers, which again can be largely explained by the availability of ionic components in the atmospheric aerosol body of air masses reaching Greenland.

    • We here describe, document, and make available a wide range of data sets used for annual-layer identification in ice cores from DYE-3, GRIP, NGRIP, NEEM, and EGRIP. The data stem from detailed measurements performed both on the main deep cores and shallow cores over more than 40 years using many different setups developed by research groups in several countries and comprise both discrete measurements from cut ice samples and continuous-flow analysis data.The data series were used for counting annual layers 60 000 years back in time during the construction of the Greenland Ice-Core Chronology 2005 (GICC05) and/or the revised GICC21, which currently only reaches 3800 years back. Now that the underlying data are made available (listed in Table 1) we also release the individual annual-layer positions of the GICC05 timescale which are based on these data sets.We hope that the release of the data sets will stimulate further studies of the past climate taking advantage of these highly resolved data series covering a large part of the interior of the Greenland ice sheet.

    • Psychosis in Parkinson's disease is a common phenomenon associated with poor outcomes. To clarify the pathophysiology of this condition and the mechanisms of antipsychotic treatments, we have here characterized the neurophysiological brain states induced by clozapine, pimavanserin, and the novel prospective antipsychotic mesdopetam in a rodent model of Parkinson's disease psychosis, based on chronic dopaminergic denervation by 6-OHDA lesions, levodopa priming, and the acute administration of an NMDA antagonist. Parallel recordings of local field potentials from eleven cortical and sub-cortical regions revealed shared neurophysiological treatment effects for the three compounds, despite their different pharmacological profiles, involving reversal of features associated with the psychotomimetic state, such as a reduction of aberrant high-frequency oscillations in prefrontal structures together with a decrease of abnormal synchronization between different brain regions. Other drug-induced neurophysiological features were more specific to each treatment, affecting network oscillation frequencies and entropy, pointing to discrete differences in mechanisms of action. These findings indicate that neurophysiological characterization of brain states is particularly informative when evaluating therapeutic mechanisms in conditions involving symptoms that are difficult to assess in rodents such as psychosis, and that mesdopetam should be further explored as a potential novel antipsychotic treatment option for Parkinson psychosis.

  • High-Resolution Greenland Ice Core Data Show Abrupt Climate Change Happens in Few Years

    • The last two abrupt warmings at the onset of our present warm interglacial period, interrupted bythe Younger Dryas cooling event, were investigated at high temporal resolution from the NorthGreenland Ice Core Project ice core. The deuterium excess, a proxy of Greenland precipitationmoisture source, switched mode within 1 to 3 years over these transitions and initiated a moregradual change (over 50 years) of the Greenland air temperature, as recorded by stable waterisotopes. The onsets of both abrupt Greenland warmings were slightly preceded by decreasingGreenland dust deposition, reflecting the wetting of Asian deserts. A northern shift of theIntertropical Convergence Zone could be the trigger of these abrupt shifts of Northern Hemisphereatmospheric circulation, resulting in changes of 2 to 4 kelvin in Greenland moisture sourcetemperature from one year to the next.

  • A Utopian Quest for Universal Knowledge : Diachronic Histories of Botanical Collections between the Sixteenth Century and the Present

    • This thesis explores the history of botany as a global collection-based science by tracing parallels between utopian traditions and botanical collecting, from their sixteenth-century beginnings to the present. A range of botanical collections, such as gardens, herbaria and classification systems, have played a central role in the struggle to discover a global or universal scientific order for the chaotic, diverse and locally shaped kingdom of plants. These collections and utopia intersect historically, and are characterised by the same epistemology of collecting: the creation of order through confined collecting spaces or “no-place.” They are manipulations of space and time. Between chaos and order, both seek to make a whole from – often unruly – parts. The long history of botanical collecting is characterised by a degree of continuity of practice that is unusual in the sciences.  For instance, the basic technology of the herbarium – preserving plants by mounting and labelling dried specimens on paper – has been in use for almost five centuries, from sixteenth-century Italy to ongoing digitisation projects. The format of the compilation thesis is well-suited to handling the historiographical challenge of tracing continuity and discontinuity with such a long chronological scope. The thesis is structured as a walled quadripartite garden, with the Kappa enclosing four research papers and an epilogue. The papers take a diachronic approach to explore different perspectives on botanical collections: botanical collecting in seventeenth-century Oxford, pressed plants in books that are not formally collections; and the digitisation of botanical collections. These accounts are all shaped by the world of books, text and publication, historically a male-dominated sphere. In order to acknowledge marginalisation of other groups and other ways of knowing plants, the epilogue is an explanation of an embroidered patchwork of plant-dyed fabric, which forms the cover of the thesis.

    • The last glacial period is characterized by a number of millennial climate events that have been identified in both Greenland and Antarctic ice cores and that are abrupt in Greenland climate records. The mechanisms governing this climate variability remain a puzzle that requires a precise synchronization of ice cores from the two hemispheres to be resolved. Previously, Greenland and Antarctic ice cores have been synchronized primarily via their common records of gas concentrations or isotopes from the trapped air and via cosmogenic isotopes measured on the ice. In this work, we apply ice core volcanic proxies and annual layer counting to identify large volcanic eruptions that have left a signature in both Greenland and Antarctica. Generally, no tephra is associated with those eruptions in the ice cores, so the source of the eruptions cannot be identified. Instead, we identify and match sequences of volcanic eruptions with bipolar distribution of sulfate, i.e. unique patterns of volcanic events separated by the same number of years at the two poles. Using this approach, we pinpoint 82 large bipolar volcanic eruptions throughout the second half of the last glacial period (12-60ka). This improved ice core synchronization is applied to determine the bipolar phasing of abrupt climate change events at decadal-scale precision. In response to Greenland abrupt climatic transitions, we find a response in the Antarctic water isotope signals (δ18O and deuterium excess) that is both more immediate and more abrupt than that found with previous gas-based interpolar synchronizations, providing additional support for our volcanic framework. On average, the Antarctic bipolar seesaw climate response lags the midpoint of Greenland abrupt δ18O transitions by 122±24 years. The time difference between Antarctic signals in deuterium excess and δ18O, which likewise informs the time needed to propagate the signal as described by the theory of the bipolar seesaw but is less sensitive to synchronization errors, suggests an Antarctic δ18O lag behind Greenland of 152±37 years. These estimates are shorter than the 200 years suggested by earlier gas-based synchronizations. As before, we find variations in the timing and duration between the response at different sites and for different events suggesting an interaction of oceanic and atmospheric teleconnection patterns as well as internal climate variability.

    • AIM: The use of cardiopulmonary bypass (CPB) can cause changes in serum creatinine and cystatin C independent of glomerular filtration rate. We aimed to quantify the temporal changes of these biomarkers and C-reactive protein (CRP) after CPB.METHODS: This was a prospective study at an academic medical center between April and October 2013. We compared postoperative changes in serum creatinine and cystatin C in 38 patients with normal preoperative kidney function who underwent cardiac surgery using CPB and did not develop perioperative acute kidney injury (AKI). The effect of inflammation on intra-individual changes was examined in mixed effects regressions, using measurements of pre- and postoperative CRP.RESULTS: Both serum creatinine (79.9 ± 22.7 vs. 92.6 ± 21.4 µmol/L, p = 0.001) and cystatin C (1.16 ± 0.39 vs. 1.33 ± 0.37 mg/L, p = 0.012) decreased significantly in the first 8 hours postoperatively compared to preoperatively, as a result of hemodilution. Thereafter serum creatinine returned to preoperative levels, whereas serum cystatin C continued to rise and was significantly elevated at 72 hours post-CPB compared to preoperative levels (1.53 ± 0.48 vs. 1.33 ± 0.37 mg/L, p = 0.003). CRP levels increased significantly post-CPB and were significantly associated with increases in both serum creatinine and cystatin C.CONCLUSIONS: Serum creatinine and cystatin C appear not to be interchangeable biomarkers during and immediately after CPB. Processes unrelated to kidney function such as acute inflammation have a significant effect on post-CPB changes in these biomarkers, and may result in significant increases in serum cystatin C that could erroneously be interpreted as AKI.     • PurposeSerum creatinine is used ubiquitously to estimate glomerular filtration rate and to diagnose acute kidney injury after cardiac surgery. Serum cystatin C is a novel biomarker that has emerged as a possible diagnostic alternative to serum creatinine. It is unclear if the dynamic changes in serum cystatin C immediately following cardiopulmonary bypass (CPB) differ from those of serum creatinine in patients with normal preoperative kidney function.MethodsWe compared changes in serum levels of creatinine and cystatin C by measuring them serially in 19 patients undergoing CPB. Within-patient differences for serum creatinine and serum cystatin C were compared by repeated measures ANOVA.ResultsSerum creatinine and cystatin C levels showed significant correlation with each other. Both biomarkers showed a significant decrease after CPB, but their serum concentrations reverted to pre-CPB levels within 12 h. Serum levels of serum creatinine remained unchanged from baseline levels throughout 72-h post-CPB. In contrast, serum cystatin C levels rose further and became significantly higher compared to baseline within 48 h. Serum cystatin C remained significantly elevated at 48- and 72-h post-CPB.ConclusionsProcesses that determine the serum concentrations of serum creatinine and cystatin C in the post-CPB period affect the two biomarkers differently, suggesting that the two are not interchangeable as diagnostic markers of glomerular filtration rate. Future studies are needed to examine if these discrepancies are related to differences in their production rates, in their ability to detect small changes in glomerular filtration rate, or to a combination of these, and to determine the effect of such differences on the diagnostic and prognostic accuracy of the two biomarkers.

    • Media criticism is studied as a concept, as critical expression and as a force for social change. The concept of media criticism is developed in relation to different forms of critical practice, theory about criticism and critique and as a part of theories about media accountability. Media criticism as a force of social change is approached by exploring concepts for the analysis of social and cultural forms of media criticism.Four kinds of media texts published in Sweden between 1998 and 2013 were analysed to investigate critical expression: television columns, reflective books on journalism written by journalists, debate articles and letters to the editor. They were studied in terms of how they address actors, content and forms of critique and responsiveness. The methodology used was reflexive interpretation mainly driven by insight, but also with an emancipative intention. Qualitative text analysis was the major method used, combined with quantitative content analysis.The concept of media criticism is developed on three levels as normative and institutional, focused on established norms and values, norm shaping and formative, focused on establishing new norms and values and openness and practice, making it possible for many people to contribute to critique. Media criticism can further be specified in three dimensions comprising its intentions, the object of critique and the process of critique. Taken together they establish different forms of criticality in which a communicative intention is seen as essential. The dissertation shows that media criticism should be given a more independent role in relation to the media accountability frame. Critical cultures and practices should be analysed in their relations to accountability cultures and practices. The concepts of institution, formative and formation were used for analysing the social and cultural forms of media criticism, where institutions are understood as the stable forms of media criticism, formatives as the changeable forms and formations as the combination of the two. Criticism and critical practice potentially have an important role to play for change in the media, journalism and society by addressing issues in an open and reflexive way. The approach to media criticism developed in the dissertation is termed critical institutionalism and aims to bridge the gap between critical social science, the sociology of critique and institutional analysis as applied in media studies.

    • This paper summarizes the state-of-the-art with respect to wetlands for nutrient removal and biodiversity enhancement, as expressed at a research workshop in December 2002. In the end of 2002, 260 and 884 wetlands had been constructed in Sweden with subsidies from LIP and the European Union, respectively. Most frequently, the aims were to remove nutrients from water and enhance biodiversity. The question raised is if we know how to design multifunctional wetlands. Should a wetland be deep or shallow, with or without macrophytes to be an efficient nutrient sink? Diverging opinions are presented, but generally it appears that fairly shallow wetlands at least partly covered by emergent macrophytes are favourable. The importance of extreme high flows and hydraulic short-circuiting is highlighted, and Danish and Norwegian approaches to wetlands construction are presented. Also, there is a risk that nutrient retaining wetlands develop a fairly trivial flora and fauna unless special care is taken. Intentional establishment of desirable and less common species, as well as creation of a variation of depth gradients to favour such species are such measures. Others are allowing for water level variations, as well as vegetation management such as grazing and harvesting.

    • Myocardial infarction (MI) with obstructive coronary artery disease (MI-CAD) and MI in the absence of obstructive coronary artery disease (MINOCA) affect different populations and may have separate pathophysiological mechanisms, with greater inflammatory activity in MINOCA compared to MI-CAD. Helicobacter pylori (Hp) can cause systemic inflammation and has been associated with cardiovascular disease (CVD). We aimed to investigate whether Hp infection is associated with concentrations of protein biomarkers of inflammation and CVD. In a case-control study, patients with MINOCA (n = 99) in Sweden were included, complemented by matched subjects with MI-CAD (n = 99) and controls (n = 100). Protein biomarkers were measured with a proximity extension assay in plasma samples collected 3 months after MI. The seroprevalence of Hp and cytotoxin-associated gene A (CagA) was determined using ELISA. The associations between protein levels and Hp status were studied with linear regression. The prevalence of Hp was 20.2%, 19.2%, and 16.0% for MINOCA, MI-CAD, and controls, respectively (p = 0.73). Seven proteins were associated with Hp in an adjusted model: tissue plasminogen activator (tPA), interleukin-6 (IL-6), myeloperoxidase (MPO), TNF-related activation-induced cytokine (TRANCE), pappalysin-1 (PAPPA), soluble urokinase plasminogen activator receptor (suPAR), and P-selectin glycoprotein ligand 1 (PSGL-1). Hp infection was present in one in five patients with MI, irrespective of the presence of obstructive CAD. Inflammatory proteins were elevated in Hp-positive subjects, thus not ruling out that Hp may promote an inflammatory response and potentially contribute to the development of CVD.
