    • For the International Large Detector concept at the planned International Linear Collider, the use of time projection chambers (TPC) with micro-pattern gas detector readout as the main tracking detector is investigated. In this paper, results from a prototype TPC, placed in a 1. T solenoidal field and read out with three independent Gas Electron Multiplier (GEM) based readout modules, are reported. The TPC was exposed to a 6. GeV electron beam at the DESY II synchrotron. The efficiency for reconstructing hits, the measurement of the drift velocity, the space point resolution and the control of field inhomogeneities are presented.

  • Multi-ancestry genome-wide association analyses identify novel genetic mechanisms in rheumatoid arthritis

    • Rheumatoid arthritis (RA) is a highly heritable complex disease with unknown etiology. Multi-ancestry genetic research of RA promises to improve power to detect genetic signals, fine-mapping resolution and performances of polygenic risk scores (PRS). Here, we present a large-scale genome-wide association study (GWAS) of RA, which includes 276,020 samples from five ancestral groups. We conducted a multi-ancestry meta-analysis and identified 124 loci (P < 5 × 10−8), of which 34 are novel. Candidate genes at the novel loci suggest essential roles of the immune system (for example, TNIP2 and TNFRSF11A) and joint tissues (for example, WISP1) in RA etiology. Multi-ancestry fine-mapping identified putatively causal variants with biological insights (for example, LEF1). Moreover, PRS based on multi-ancestry GWAS outperformed PRS based on single-ancestry GWAS and had comparable performance between populations of European and East Asian ancestries. Our study provides several insights into the etiology of RA and improves the genetic predictability of RA.

  • Patient satisfaction and loyalty in Japanese primary care : a cross-sectional study

    • Background: This study aimed to explore associations between various elements of primary care, patient satisfaction, and loyalty. Methods: This cross-sectional study used a modified version of the Primary Care Assessment Tool (PCAT), which was adapted for Japan. We distributed the PCAT questionnaire to patients aged 20 years or older at five rural primary care centres in Japan. We confirmed the validity and reliability of the measure for our study. Next, we examined which elements of primary care were related to patient satisfaction and loyalty using Spearman’s correlation and structural equation modelling. Results: Of 220 eligible patients, 206 participated in this study. We developed nine component scales: first contact (regular access), first contact (urgent access), longitudinality, coordination, comprehensiveness (variety of care), comprehensiveness (risk prevention), comprehensiveness (health promotion), family-centeredness, and community orientation. Longitudinality and first contact (urgent access) were related with patient satisfaction. Longitudinality, first contact (regular access), and family-centeredness were related to patient loyalty. In the structural equation modelling analysis, two variables were significantly related to loyalty, namely a combined variable including longitudinality and first contact (regular access), along with family-centeredness. Conclusions: While a patient satisfaction model could not be distilled from the data, longitudinality, first contact (urgent access), and family-centeredness were identified as important elements for the cultivation of patient loyalty. This implies that primary care providers need to develop a deep understanding of patients’ contexts and concerns and pay attention to their level of access to cultivate greater patient loyalty.

  • Steeper Macular Curvature in Eyes With Non-Highly Myopic Retinitis Pigmentosa

    • Purpose: A posterior staphyloma has been reported to be present in some eyes with retinitis pigmentosa (RP), and the purpose of this study was to determine the macular curvature of non-highly myopic RP eyes. Methods: This was a retrospective, observational study. The medical charts of the right eyes of 143 patients with RP and 60 controls whose axial length ranged from 21.5 mm to 26.0 mm were reviewed. The mean curvature of Bruch's membrane within 6 mm of the central macula obtained from the horizontal optical coherence tomographic images were evaluated as the mean macular curvature index (MMCI). The relationships between the MMCI and other clinical factors were assessed. Results: The mean MMCI of RP patients (-13.73 ± 9.63 × 10-5 μm-1) was significantly lower than that of the controls (-6.63 ± 5.63 × 10-5 μm-1). This indicated a deeper concave shape of the macula in RP eyes (P < 0.001). The MMCI was significantly correlated with the age (r = 0.20; P = 0.016) and the axial length (r = -0.24; P = 0.004). Further analysis suggested a nonlinear effect of the ellipsoid zone width on the macular curvature in the RP eyes. Conclusions: There is a high incidence of steeper macular curvatures even in non-highly myopic RP eyes, and the steepness was also affected by the degree of photoreceptor degeneration.

    • Snowy cold regions account for 60% of Japan's land area. These regions see many traffic accidents that stem from weather conditions peculiar to winter (hereinafter: "wintry accidents"). Typical examples include skidding accidents on snow-covered or icy roads and run-off-road accidents involving snowstorm-induced poor visibility. As the majority of those wintry accidents are skidding accidents, elucidating the impact of winter weather and winter road surface conditions on traffic accident occurrence may be useful in selecting priority sections for measures against wintry accidents and in appropriately identifying the sections and the times of day that necessitate road management, including snow removal and anti-icing-agent application.

    • Gold-mineralized quartz veins at the Trenggalek district of the Southern Mountains Range in East Java, Indonesia, are hosted by Oligo-Miocene volcaniclastic and volcanic rocks, and are distributed close to andesitic plugs in the northern prospects (Dalangturu, Suruh, Jati, Gregah, Jombok, Salak, and Kojan) and the southern prospects (Sentul and Buluroto). The plugs are subalkaline tholeiitic basaltic-andesite to calc-alkaline andesite in composition. Ar-40-Ar-39 dating of a quartz-adularia vein at the Dalangturu prospect yielded an age of 16.29 +/- 0.56 Ma (2 sigma), and a crystal tuff of a limestone-pyroclastic rock sequence at the southwest of the Dalangturu prospect was determined as 15.6 +/- 0.5 Ma (2 sigma). Statistic overlap of ages suggests that the gold mineralization in the northern prospects took place in a shallow marine to subaerial transitional environment. Hydrothermal alteration of the host rocks is characterized by the replacement of quartz, illite and adularia. Quartz veins in surface outcrops are up to 50 cm wide in the northern prospects and up to 3 m wide in the southern prospects, showing a banded or brecciated texture, and are composed of quartz, adularia, carbonates with pyrite, electrum, sphalerite, galena, and polybasite. Gold contents of quartz veins are positively correlated with Ag, Zn, Pb, and Cu contents in both the northern and southern prospects. The quartz veins at the Jati, Gregah, and Sentul prospects have relatively lower gold-silver ratios (Ag/Au = 23.2) compared to those at the Kojan, Dalangturu, Salak, and Suruh prospects (Ag/Au = 66.8). The quartz veins at the Dalangturu prospect are relatively rich in base metal sulfides. Ag/(Au+Ag) ratios of electrum in the Dalangturu prospect range from 45.2 to 65.0 at%, and FeS contents of sphalerite range from 1.2 to 6.4 mol%. Fluid inclusion microthermometry indicates ore-forming temperatures of 190-200 degrees C and 220-230 degrees C at the Sentul and Kojan prospects, respectively. Widely variable vapor/liquid ratio of fluid inclusions indicates that fluid boiling took place within the hydrothermal system at the Sentul prospect. Salinities of ore-fluids range from 0 to 0.7 wt% (av. 0.4 wt% NaCl equiv.) and from 0.5 to 1.4 wt% (av. 0. 9 wt%) for the Sentul and Kojan prospects, respectively. The boiling of hydrothermal fluid was one of the gold deposition mechanisms in the Sentul prospect.

  • Interethnic analyses of blood pressure loci in populations of East Asian and European descent

    • Blood pressure (BP) is a major risk factor for cardiovascular disease and more than 200 genetic loci associated with BP are known. Here, we perform a multi-stage genome-wide association study for BP (max N = 289,038) principally in East Asians and meta-analysis in East Asians and Europeans. We report 19 new genetic loci and ancestry-specific BP variants, conforming to a common ancestry-specific variant association model. At 10 unique loci, distinct non-rare ancestry-specific variants colocalize within the same linkage disequilibrium block despite the significantly discordant effects for the proxy shared variants between the ethnic groups. The genome-wide transethnic correlation of causal-variant effect-sizes is 0.898 and 0.851 for systolic and diastolic BP, respectively. Some of the ancestry-specific association signals are also influenced by a selective sweep. Our results provide new evidence for the role of common ancestry-specific variants and natural selection in ethnic differences in complex traits such as BP.

    • Differentiation of mesenchymal stem cells (MSCs) into osteoblasts is a critical process for proper skeletal development and acquisition/maintenance of bone mass. However, since this regulatory mechanism has not yet been fully elucidated, the treatment of severe osteoporosis and fractures is a challenge. Here, through a comprehensive analysis of gene expression during the differentiation of MSCs into osteoblasts, we show that the forkhead transcription factor Foxf2 is a crucial regulator of this process. Foxf2 expression transiently increased during MSC osteoblastic differentiation. Overexpression of Foxf2 in MSCs inhibited osteoblastic differentiation, and conversely, knockdown of Foxf2 expression promoted this process. Osteoprogenitor-specific Foxf2 knockout mice developed a high bone mass phenotype due to increased bone formation. RNA-seq analysis and molecular experiments revealed that Foxf2 regulation of bone formation is mediated by Wnt2b. Knockdown of Foxf2 in mouse femurs enhanced bone regeneration in vivo. FOXF2 expression was correlated with hip bone mineral density in postmenopausal women with low bone mass. Finally, inhibition of FOXF2 promoted osteoblastic differentiation of human MSCs. This study uncovers a critical role of Foxf2 in the differentiation of MSCs into osteoblasts and provides insight into the pathogenesis associated with bone-related diseases such as osteoporosis and nonunion after fracture.

  • The Kago low-sulfidation gold and silver deposit : A peripheral mineralisation to the Nansatsu high-sulfidation system, southern Kyushu, Japan

    • The Kago deposit is a small deposit located at the southern tip of the Satsuma Peninsula of Southern Kyushu, Japan. It lies proximal to the well-known Nansatsu-type mineralisation province dominated by high-sulfidation type epithermal deposits. The deposit was heavily mined in the 18 th Century, largely for its relatively higher gold compared to that of surrounding and regional deposits. The Kago deposit is a typical low-sulfidation deposit, characterised by adularia-quartz veins, composed of electrum, Ag-tetrahedrite, polybasite, chalcopyrite and pyrite. Based on mine records, the grade ranged from 4.1 to 13.3 g/t Au and 2.6-6.6 g/t Ag. Alteration grades from low to high temperature argillic into a propylitic zone at the extremes of the vein exposures. Carbonate is absent. Fluid inclusion microthermometry reveals a typical temperature range of 220-240 °C with salinity of 0.7-2.6 NaCl eq. wt%. Electrum from high-grade ore ranges from 66 to 69 wt% Au. 40Ar/39Ar age dating of adularia bearing colloform/crustiform and brecciated veins, suggests a mineralisation event from 4.23 to 4.0 Ma. δ18O of veined and silicified quartz ranges from +4.0 to +18.4‰. δ18OH2O of fluids in equilibrium with quartz, in the dominant range of measured fluid inclusion temperatures, ranges from -6.5‰ to -0.2‰. δ34S of pyrite has a narrow range from -1.8 to 2.7‰. The deposit lies at the northern extent of the classic Nansatsu high-sulfidation epithermal area, in which a number of silicified bodies punctuate the region in a roughly semi-circular shape. The Kago deposit lies within the principle mineralisation age range of the high-sulfidation deposits, which range from 5.5 to 3.7 Ma. The structural displacement of the Kago deposit from the Nansatsu mineralisation and the differing host rocks has greatly influenced alteration, ore and rock-water interaction of the ore depositing fluids. Here we seek to establish the relationship that this extended mineral province has between the differing styles of mineralisation.

    • Samples of the carbonaceous asteroid (162173) Ryugu were collected and brought to Earth by the Hayabusa2 spacecraft. We investigated the macromolecular organic matter in Ryugu samples and found that it contains aromatic and aliphatic carbon, ketone, and carboxyl functional groups. The spectroscopic features of the organic matter are consistent with those in chemically primitive carbonaceous chondrite meteorites that experienced parent-body aqueous alteration (reactions with liquid water). The morphology of the organic carbon includes nanoglobules and diffuse carbon associated with phyllosilicate and carbonate minerals. Deuterium and/or nitrogen-15 enrichments indicate that the organic matter formed in a cold molecular cloud or the presolar nebula. The diversity of the organic matter indicates variable levels of aqueous alteration on Ryugus parent body.
