  • Control of Wave Energy Converters in arrays

    • One way to lower the levelized cost of energy for wave power plants and paving so the way for commercial success, is to increase the power absorption by use of advanced control algorithms. This thesis investigates the influence of the generator inertia, the generator damping and the layout on power absorption and presents a new model free strategy of controlling wave energy converters.The evaluation of all control strategies was done in a numerical simulation and in experimental 1:10 model scale wave tank tests conducted in the COAST laboratory at the University of Plymouth. The WECs used are inspired by the wave energy concept developed at Uppsala University.The influence of the generator inertia on the power absorption was tested with an uncontrolled WEC. Compared to a conventional WEC the power output could be significantly increased for small waves and high wave periods.   As a simple and easy to implement control strategy, a WEC with sea state optimized generator damping was used to create a power matrix. The optimal damping factor depends on both, wave period and wave height. The power absorption increases with the wave height and when the wave period converges towards the oscillation period of the WEC.A genetic algorithm was used to obtain the optimized layouts for wave energy farms, which suggest that the converter should be placed in rows parallel to the wave front, and the position in the array has nearly no influence on the optimal control parameter.Then a collaborative learning approach using machine learning is presented, with several identical wave energy converters in a row to parallelise the search of the optimal control parameter. It was implemented to control the generator damping factor and the latching time. With the latter the power could be increased significantly.

  • Snake Gourds, Parasites and Mother Roasting : Medicinal plants, plant repellents, and Trichosanthes (Cucurbitaceae) in Lao PDR

    • Background. Traditional plant use was studied in Lao PDR. Research focused on medicinal plant use by the Brou, Saek and Kry ethnic groups, traditional plant repellents against parasitic arthropods and leeches, and the phylogeny and biogeography of the medicinally-important snake gourd genus (Trichosanthes, Cucurbitaceae).  Methods. The ethnobiology research used a combination of structured interviews, village surveys, botanical collecting, hydro-distillation, GC-MS analysis, literature studies, and laboratory experiments. The plant systematics research used a combination of morphological studies, molecular biology laboratory work, and phylogenetic, dating and biogeographical analysis.  Results. Informants reported the use of close to 100 species to repel arthropods and leeches, many of which have constituents with documented efficacy.  Brou, Saek and Kry informants use over 75 plant species for women’s healthcare, mainly during the postpartum period for steam sauna, steam bath, hotbed, mother roasting, medicinal decoctions and infusions, and postpartum diet.  A molecular phylogeny of Trichosanthes and Gymnopetalum using a broad sampling of ~60% of their species and 4756 nucleotides of nuclear and plastid DNA shows that Gymnopetalum is nested within Trichosanthes. Fossil-calibrated Bayesian molecular dating of the Trichosanthes phylogeny reveals an early Oligocene origin of the genus, and many of the extant sections originating and diversifying during the Miocene. Biogeographical analysis shows a likely East or South Asian origin of Trichosanthes, with lineages diversifying and spreading throughout Australasia from the early Pliocene to the Pleistocene.  Discussion. Traditional plant use in Lao PDR is common and widespread. The presence among the repellent species of economical alternatives to costly synthetic repellents is tenable, and the subject of ongoing studies.  Postpartum traditions and medicinal plant use are essential parts of childbirth and postpartum recovery in these ethnic groups, and many other groups in Lao PDR. Efforts to improve maternal healthcare and reduce maternal and infant mortality need to integrate these traditions with modern notions of healthcare to achieve wider adoption. Documenting all possible uses of commonly used medicinal plant species shows that similarity in use between these ethnic groups is relatively low considering that they share, and have shared for many generations, the same environment and resources. A lack of effective cures leads to a process of continuous innovation, where effective cures are shared between cultures, but remedies of only cultural importance, or those under evaluation are culture-specific.  The Trichosanthes phylogeny implies the merging of Gymnopetalum into Trichosanthes, and this is done using available names or new combinations. A synopsis of Trichosanthes, the new combinations, and a revision of the species in Australia, are made and presented.  Conclusions. Traditional plant use is widespread in Lao PDR, and of significance to many people as a source of primary healthcare and inexpensive repellents. The important medicinal plant genus Trichosanthes includes Gymnopetalum, and has a complex biogeographic history with multiple colonization events of Australasia.

  • Improvement and Assessment of Two-Dimensional Resistivity Models Derived from Radiomagnetotelluric and Direct-Current Resistivity Data

    • Two-dimensional (2-D) models of electrical resistivity are improved by jointly inverting radiomagnetotelluric (RMT) and direct-current resistivity (DCR) data or by allowing for displacement currents in the inversion of RMT data collected on highly resistive bedrock. Uniqueness and stability of the 2-D models are assessed with a model variance and resolution analysis that allows for the non-linearity of the inverse problem.Model variance and resolution are estimated with a truncated singular value decomposition (TSVD) of the sensitivity matrix. In the computation of model errors, inverse singular values are replaced by non-linear semi-axes and the number of included eigenvectors is increased until a given error threshold is reached. Non-linear error estimates are verified with most-squares inversions. For the obtained truncation levels, model resolution matrices are computed. For RMT data, non-linear error appraisals are smaller than linearized ones. Hence, the consideration of the non-linearity in RMT data leads to reduced model errors or enhanced model resolution.The dielectric effect on RMT data is investigated with a new 2-D forward and inverse code that allows for displacement currents. As compared to the quasi-static approximation, apparent resistivities and phases of the impedance tensor elements are found to be significantly smaller and the vertical magnetic transfer function exhibits more distinct sign reversals. More reliable models of electrical resistivity are obtained from areas with highly resistive bedrock, if displacement currents are allowed for. In contrast, inversions with a quasi-static scheme introduce artefactual structures with extremely low or high resistivities.A smoothness-constrained 2-D joint inversion of RMT and DCR data is presented. The non-linear model variance and resolution analysis is applied to single and joint inverse models. For DCR data, the errors estimated by most-squares inversions are consistently larger than those estimated by the non-linear semi-axes, indicating that DCR models are poorly resolved. Certain areas of the joint inverse models are better resolved than in the single inverse models.

    • This dissertation examines detective novels for children and young adults written and published in the German Democratic Republic (GDR). The aim of the thesis is to study how the genre developed under the conditions of a socialist society. The analysis of the 66 texts included in the corpus is based on a socio-historical approach assuming that dialogical interdependencies between texts and society exist and can be verified. Central to the analytical work with the texts is the thesis that detective novels written for young readers reflect the socio-political development in East-German society. It shows, however, that—because of their strong didactic impetus—the texts did generally not, like detective novels for adults, develop into a forum for socio-critical discussions. In the diachronic development, which extends from the beginning in the Soviet occupation zone to the post-reunification period, it is shown that changing socio-political conditions interact with the texts, which becomes particularly obvious in the changing presentation of the detectives and criminals.Studying the texts, the dissertation presents basic research and an overview of the genre. Ten texts from the corpus are subject to a detailed analysis in order to deepen the general insights with examples. This way, different aspects of detective novels for children and young adults in the GDR are emphasized, e.g. the interaction between text and illustration. The embedding of figures in a socialist community produces further motives frequently occurring in the texts such as: the Heimat motive or the anti-fascist society.Although the majority of the texts do not go beyond stereotypical representations of characters, criminal cases, and locations—and hence demonstrate the close link between (normative) ideas of society and their literary implementation—the body of texts contains some innovative exceptions in which the social development is questioned and even cautiously criticized.

  • Precision electroweak measurements on the Z resonance

    • We report on the final electroweak measurements performed with data taken at the Z resonance by the experiments operating at the electron-positron colliders SLC and LEP. The data consist of 17 million Z decays accumulated by the ALEPH, DELPHI, L3 and OPAL experiments at LEP, and 600 thousand Z decays by the SLID experiment using a polarised beam at SLC. The measurements include cross-sections, forward-backward asymmetries and polarised asymmetries. The mass and width of the Z boson, m(Z) and Gamma(Z), and its couplings to fermions, for example the p parameter and the effective electroweak mixing angle for leptons, are precisely measured: m(Z) = 91.1875 +/- 0.0021 GeV, Gamma(Z) = 2.4952 +/- 0.0023 GeV, rho(l) = 1.0050 +/- 0.0010, sin(2)theta(eff)(lept) = 0.23153 +/- 0.00016. The number of light neutrino species is determined to be 2.9840 +/- 0.0082, in agreement with the three observed generations of fundamental fermions. The results are compared to the predictions of the Standard Model (SM). At the Z-pole, electroweak radiative corrections beyond the running of the QED and QCD coupling constants are observed with a significance of five standard deviations, and in agreement with the Standard Model. Of the many Z-pole measurements, the forward-backward asymmetry in b-quark production shows the largest difference with respect to its SM expectation, at the level of 2.8 standard deviations. Through radiative corrections evaluated in the framework of the Standard Model, the Z-pole data are also used to predict the mass of the top quark, m(t) = 173(+10)(+13) GeV, and the mass of the W boson, m(W) = 80.363 +/- 0.032 GeV. These indirect constraints are compared to the direct measurements, providing a stringent test of the SM. Using in addition the direct measurements of m(t) and m(W), the mass of the as yet unobserved SM Higgs boson is predicted with a relative uncertainty of about 50% and found to be less than 285 GeV at 95% confidence level.   • Aqueous graphene dispersions for paper packaging

    • Graphene is widely touted as the thinnest and the most versatile material available. As an atomically thin layer of carbon atoms arranged in a hexagonal configuration, graphene has a combination of technologically important properties, such as thermal and electrical conductivity, mechanical strength, and impermeability to gases. From an industrial perspective on applications, water as a dispersing media for graphene offers safer handling and environmental benefits compared with conventional organic solvents. However, the high surface tension of water and the attractive forces between graphene surfaces drive the sheets to aggregation. Although surfactants have been an important stepping stone in the advancement of aqueous graphene dispersions, these surface-active molecules are often needed in excess and have adverse effects on coatings during film formation. These challenges limit the industrial relevance of graphene as an effective barrier in composites. In general, gas barriers against both oxygen and water vapour, made from a single coating formulation, is seemingly a holy grail for the packaging industry. In this thesis work, the aim was to gain a fundamental understanding of aqueous graphene dispersions for gas barriers used in paper packaging. Biobased materials were systematically investigated as dispersing agents for graphene based on dispersing conditions and functional barrier performance. Flavin mononucleotide (FMN), a food additive, dispersed graphene using a relatively low amount of FMN and showed intriguing spectroscopic signatures of π-π interactions with graphene. Starch nanoparticles (SNPs) realised concentrated and stable aqueous graphene dispersions for composite films. The SNP-stabilized graphene sheets in starch films lowered the gas permeability of both oxygen and water vapour simultaneously by over 70% under all the conditions tested. In general, a combined gas barrier performance is unusual for both bioplastics and common petrochemical-based plastics used in the packaging industry. Motivated by the graphene network leading to the extraordinary barrier performance, the aqueous SNP-graphene dispersion was modified for inkjet printing. The printed patterns were flexible and electrically conductive in the order of 104 S m-1 that is on par with the highest reported values in the literature. These surfactant-free aqueous SNP-graphene dispersions have the potential and versatility for paper-based gas barriers with integrated electronics. Multifunctional composite films made from these dispersions, when optimized, could become competitive with commercial plastics, and meet the current and future demands of the packaging industry.

    • The central theme of this thesis is the use of imaging Surface Plasmon Resonance (iSPR) as a tool in the characterization of surfaces with laterally varying properties. Within the scope of this work, an instrument for iSPR analysis was designed and built. SPR is a very sensitive technique for monitoring changes in optical properties in the immediate vicinity of a sensor surface, which is very useful in biosensing and surface science research. We have employed SPR in the Kretschmann configuration, wherein surface plasmons are excited by means of an evanescent field arising from total internal reflection from the backside of the sensor surface. In iSPR, the signal is the reflectivity of TM-polarized light which is measured using an imaging detector, typically a CCD camera. Advantages of this technique include extreme surface sensitivity and, because detection is done from the backside, compatibility with complex samples. In addition, SPR is a non-labeling technique, and in imaging mode, a lateral resolution in the µm range can be attained.The imaging SPR instrument could be operated in either wavelength interrogation mode or in intensity mode. In the former case, the objective is to find the SPR wave-length, λSPR, which is the wavelength at which the reflected intensity is at a minimum. In intensity mode, a snapshot of the intensity reflectance is taken at a fixed wavelength hand incidence angle.In biosensor science, the use of an imaging technique offers a major advantage by enabling parallelization and thereby increasing throughput. We have, for example, used iSPR in biochemical interaction analysis to monitor immobilization and specific binding to protein and synthetic polypeptide micro arrays. The primary interest has been the study of soft matter surfaces that possess properties interesting in the field of biomimetics or for applications in biosensing. Specifically, the surfaces studied in this thesis include patterned self-assembled monolayers of thiolates on gold, a graft polymerized poly(ethylene glycol) (PEG) based hydrogel, a dextran hydrogel, and a polyelectrolyte charge gradient. Our results show that the PEG-based hydrogel is very well suited for use as a platform in protein immobilization in an array format, owing to the very low unspecific binding. In addition, well defined microarray templates were designed by patterning of hydrophobic barriers on dextran and monolayer surfaces. A polypeptide affinity microarray was further designed and immobilized on such a patterned monolayer substrate, in order to demonstrate the potential of analyte quantification with high sensitivity over a large dynamic range.Furthermore, iSPR was combined with electrochemistry to enable laterally resolved studies of electrochemical surface reactions. Using this combination, the electrochemical properties of surfaces patterned with self assembled monolayers can be studied in parallel, with a spatial resolution in the µm regime. We have also employed electrochemistry and iSPR for the investigation of potential and current density gradients on bipolar electrodes.The imaging SPR instrument could be operated in either wavelength interrogation mode or in intensity mode.   • Automatic Instance-based Tailoring of Parameter Settings for Metaheuristics

    • Many industrial problems in various fields, such as logistics, process management, orproduct design, can be formalized and expressed as optimization problems in order tomake them solvable by optimization algorithms. However, solvers that guarantee thefinding of optimal solutions (complete) can in practice be unacceptably slow. Thisis one of the reasons why approximative (incomplete) algorithms, producing near-optimal solutions under restrictions (most dominant time), are of vital importance.Those approximative algorithms go under the umbrella term metaheuristics, each of which is more or less suitable for particular optimization problems. These algorithmsare flexible solvers that only require a representation for solutions and an evaluation function when searching the solution space for optimality.What all metaheuristics have in common is that their search is guided by certain control parameters. These parameters have to be manually set by the user andare generally problem and interdependent: A setting producing near-optimal resultsfor one problem is likely to perform worse for another. Automating the parameter setting process in a sophisticated, computationally cheap, and statistically reliable way is challenging and a significant amount of attention in the artificial intelligence and operational research communities. This activity has not yet produced any major breakthroughs concerning the utilization of problem instance knowledge or the employment of dynamic algorithm configuration.The thesis promotes automated parameter optimization with reference to the inverse impact of problem instance diversity on the quality of parameter settings with respect to instance-algorithm pairs. It further emphasizes the similarities between static and dynamic algorithm configuration and related problems in order to show how they relate to each other. It further proposes two frameworks for instance-based algorithm configuration and evaluates the experimental results. The first is a recommender system for static configurations, combining experimental design and machine learning. The second framework can be used for static or dynamic configuration,taking advantage of the iterative nature of population-based algorithms, which is a very important sub-class of metaheuristics.A straightforward implementation of framework one did not result in the expected improvements, supposedly because of pre-stabilization issues. The second approach shows competitive results in the scenario when compared to a state-of-the-art model-free configurator, reducing the training time by in excess of two orders of magnitude.

  • The Multiple Faces of Interfaces : Electron microscopy analysis of CuInSe2 thin-film solar cells

    • The CIS solar cell family features both a high stability and world-class performances. They can be deposited on a wide variety of substrates and absorb the entire solar spectrum only using a thickness of a few micrometers. These particularities allow them to feature the most positive Energy returned on energy invested (EROI) values and the shortest Energy payback times (EPBT) of all the main photovoltaic solar cells. Using mainly electron microscopy characterization techniques, this thesis has explored the questions related to the interface control in thin-film photovoltaic solar cells based on CuInSe2 (CIS) absorber materials. Indeed, a better understanding of the interfaces is essential to further improve the solar cell conversion efficiency (currently around 23%), but also to introduce alternative substrates, to implement various alloying (Ga-CIS (CIGS), Ag-CIGS (ACIGS)…) or even to assess alternative buffer layers.The thread of this work is the understanding and the improvement of the interface control. To do so, the passivation potential of Al2O3 interlayers has been studied in one part of the thesis. While positive changes were generally measured, a subsequent analysis has revealed that a detrimental interaction could occur between the NaF precursor layer and the rear Al2O3 passivation layer. Still within the passivation research field, incorporation of various alkali-metals to the CIS absorber layer has been developed and analyzed. Large beneficial effects were ordinarily reported. However, similar KF-post deposition treatments were shown to be potentially detrimental for the silver-alloyed CIGS absorber layer. Finally, part of this work dealt with the limitations of the thin-barrier layers usually employed when using steel substrates instead of soda-lime glass ones. The defects and their origin could have been related to the steel manufacturing process, which offered solutions to erase them.Electron microscopy, especially Transmission electron microscopy (TEM), was essential to scrutinize the local changes occurring at the different interfaces within a few nanometers. The composition variation was measured with both Electron energy loss spectroscopy (EELS) and Energy dispersive X-ray spectroscopy (EDS) techniques. Finally, efforts have been invested in controlling and improving the FIB sample preparation, which was required for the TEM observations in our case.

  • The interplay between quark and hadronic degrees of freedom and the structure of the proton

    • We study the low-energy sector of the strong interaction which is the least understood part of the Standard Model, the theory that describes the interactions of all known particles. The ideal particles for this study are the proton and the neutron, collectively called the nucleon. They make up the nucleus of all the atoms of our world and understanding them has been of high priority ever since their discovery. We show that one cannot neglect the effects of other hadrons, such as neutrons and pions when studying the proton. A large part of the proton's hadronic wavefunction is shown to consist of the wavefunctions of other hadrons. In other words, when probing the proton there is a sizeable probability that one is probing some other hadron surrounding the proton as a quantum fluctuation.The nucleon itself consists of elementary particles known as quarks and gluons, collectively called partons. Exactly how the properties of these partons make up the properties of the nucleon has been the subject of active research ever since their discovery. Two main issues are the flavor asymmetry of the proton sea and the spin structure of the nucleon. To address these questions we study the interplay between the partonic and hadronic degrees of freedom. We introduce a model based on a convolution between hadronic quantum fluctuations as described by chiral perturbation theory, and partonic degrees of freedom motivated by a physical model of the nucleon having only few physically constrained parameters.We present the hadronic distribution functions and the parton distribution functions. The results are in agreement with a large set of experimental data. These include the structure functions of the proton and the neutron. Agreement with the sum rules of the spin structure functions offers new insight into the spin structure of the nucleon.

    • This paper describes the conceptual design of the Mechanical Tritium Pumping System (MTPS) that shall be installed and tested at JET during the next Deuterium-Tritium-Experiment (DTE2). This pump train uses a two-stage liquid ring pump in combination with a booster pump to cover a pressure regime from 10(-1) Pa to 10(5) Pa. As working fluid for all pumps, mercury will be used for tritium compatibility reasons. Starting from the requirements to MTPS, the pumps and their arrangement will be described in this paper as well as the mercury containment strategy and safety- and control issues.

  • Genetic variability and nitrogenase activity of cyanobacterial communities associated with tropical seagrass meadows (western Indian Ocean)

    • Tropical seagrass ecosystems are highly productive and important for sustaining marine life and associated coastal societies. In this study, the diversity and role of nitrogen-fixing cyanobacteria associated with five common seagrass genera in coastal regions of the western Indian Ocean (WIO; Tanzania) were examined, as well as the impact of anthropogenic activities. Cyanobacteria were characterized morphologically and genetically (16S rRNA and nifH gene phylogeny), as were diel variations in nifH gene expression, NifH protein levels and nitrogenase activity. The results revealed that WIO seagrass beds supported rich cyanobacterial diversity and that these represented approx. 83% of total clones obtained (DNA and RNA nifH clone libraries). Non-heterocystous genera, such as Oscillatoria, Lyngbya, Leptolyngbya, Phormidium and Microcoleus dominated, while heterocystous morphotypes such as Calothrix were less frequent and unicellular morphotypes (e.g. Gloeocapsa, Chroococcus and Chroococcidiopsis) were few. Additionally, the phylogenetic analysis revealed several novel uncharacterized cyanobacterial clades. Cyanobacterial composition and nitrogenase activity varied over seasons and between the seagrass species. Day time nitrogenase activity originated primarily from heterocystous phylotypes, while non-heterocystous filamentous phylotypes fixed nitrogen at night. The highest activity in the diel cycle was 358 ± 232 nmol C2H4 g-1 h-1at 09.00 associated with epiphytes of the seagrass Cymodocea. Nitrogenase activity was consistently lower in anthropogenically disturbed (eutrophication) seagrass sites. Such data suggest that diazotrophic cyanobacteria may be a significant source of ‘new’ nitrogen in the often oligotrophic coastal regions of tropical oceans. It is also proposed that the rapid shifts in the cyanobacterial population and function found may also be used as early disturbance indicator in coastal management practices.

    • I samhället uppmärksammas tekniken för sin ökande komplexitet och systemiska karaktär. l vilken mån avspeglar sig detta i skolans teknikämne? Denna avhandling berör undervisning om tekniska system i ett skolperspektiv, med det huvudsakliga syftet att undersöka hur undervisning om tekniska system tas upp till behandling i skolan.Den övergripande frågeställningen är: Hur har skolan hanterat den  didaktiska uppgiften att utveckla ett undervimingsområde om tekniska system?Avhandlingen är en monografi som bygger på delstudier av tre arenor  med relevans för skolan. De tre arenorna omfattar huvudsakligen nationella styrdokument, läromedel respektive lärares arbete.Resultaten pekar bland annat på att tekniska system successivt  förstärkts som undervisningsinnehåll i styrdokument och läromedel sedan slutet på 1970-talet. Denna förändring har skett parallellt med att teknikämnets beskrivning förändrats i grundskolan. I öäverenstämmelse med modern teknikfilosofi har teknik i skolsammanhang i allt högre grad kommit att framställas som en egen kunskapskultur och inte som en del av naturvetenskaperna. Detta har varit förutsättningar för att tekniska system har kunnat etableras som innehåll. Diskursen talet om tekniska rys/em har dock skiftat i karaktär över tiden.Gemensamt för de tre arenorna är att två hegemoniska diskurser har hämmat introduktionen av tekniska system. För det första teknikundervisningens egen fascination för enkla artefakter och, för det andra, en dominant naturvetenskaplig diskurs. Härmed fokuseras undervisningen på systemens komponenter, snarare än mot systemnivån. Så tenderar t.ex. undervisning om energisystem att handla om energiomvandlingar eller kemiska reaktioner, istället för systemens uppbyggnad, funktioner, eller dess relationer till människor och samhälle.Avhandlingen visar även att tekniska system framställs som faktiskt existerande och inte som mentala konstruktioner vars gränser måste bestämmas. Produktions- och transportsystem ges framträdande positioner i teknikinnehållet. Tekniska system belyses snarare som något som påverkar omgivningen, än att de är möjliga att påverka. Överlag finns en brist på systembegrepp. Dock har styr- och reglerteknik nått en viss position. 1 avhandlingen visas även hur människans roll relativt systemen tas upp och, i samband med detta, hur ett etiskt och politiskt tema vuxit fram inom arenornas teknikdiskurs. Temat har medborgerliga förtecken och tar sina motiv från en strävan mot en hållbar utveckling. Utifrån en teknikhistorisk emfas används argument för att eleverna bör studera tekniska systems framväxt och förändring.

  • Galactic archaeology with metal-poor stars

    • The chemical fingerprints of old, metal-poor stars can be used to unravel the events of the newborn Universe and help us understand the properties of the first stars and star clusters. The study of nearby stars to infer properties in the distant past is often referred to as Galactic archaeology. However, the chemical composition of stars cannot be observed directly, but must be inferred by means of spectroscopic modelling. Traditionally, this modelling utilises one-dimensional (1D) stellar atmospheres in hydrostatic and local thermodynamic equilibrium (LTE). Today, we know that departures from LTE (known as NLTE), and differences between 1D model atmospheres and their hydrodynamical three-dimensional (3D) counterparts, become increasingly severe at lower metallicity. The development of NLTE modelling of spectral line formation in 3D atmospheres is still in its infancy, but constitutes a remarkable step forward that has been made possible by parallelised codes and supercomputers. The central theme of this thesis is the application of NLTE analyses to metal-poor stars, to help usher the field of Galactic archaeology forward with important consequences for the nature of the first stellar generations.I present a theoretical NLTE study of aluminium, where I validate the analysis using a set of bright standard stars and provide calculated NLTE effects for a large parameter space. I perform 3D NLTE calculations for the solar spectrum to better constrain the zero-point of the cosmic abundance scale, and find excellent agreement with the meteoritic aluminium abundance.I also present NLTE analyses of metal-poor stars in the globular clusters NGC 6397 and M4. While globular cluster stars were long expected to form from a chemically homogeneous medium, star-to-star abundance variations of light elements indicate multiple epochs of star formation. Massive first-generation stars polluted the interstellar medium from which later generations formed, and I use the observed abundance variations to deduce the properties of the polluting stars. Among the heavier elements, I uncover evolutionary abundance variations that match predictions of stellar evolution models with atomic diffusion. The results indicate that the chemical abundance ratios of unevolved metal-poor stars are affected by gravitational settling, with a bias of the order 25-50 %, increasing towards lower metallicity. This atmospheric depletion mechanism is a probable explanation to why the stellar abundances of lithium fall short of the predictions from standard Big Bang nucleosynthesis.Finally, I apply a 3D NLTE abundance analysis to the red giant SMSS 0313-6708, which is the most iron-deficient star known. The chemical abundance pattern of this star indicates that it formed from gas affected only by Big Bang nucleosynthesis and a single faint supernova. Comparison of the inferred abundance pattern to theoretical predictions leads to constraints on the explosion mechanism and the mass of the metal-free progenitor star.

    • Carriers of large recurrent copy number variants (CNVs) have a higher risk of developing neurodevelopmental disorders. The 16p11.2 distal CNV predisposes carriers to e.g., autism spectrum disorder and schizophrenia. We compared subcortical brain volumes of 12 16p11.2 distal deletion and 12 duplication carriers to 6882 non-carriers from the large-scale brain Magnetic Resonance Imaging collaboration, ENIGMA-CNV. After stringent CNV calling procedures, and standardized FreeSurfer image analysis, we found negative dose-response associations with copy number on intracranial volume and on regional caudate, pallidum and putamen volumes (β = −0.71 to −1.37; P < 0.0005). In an independent sample, consistent results were obtained, with significant effects in the pallidum (β = −0.95, P = 0.0042). The two data sets combined showed significant negative dose-response for the accumbens, caudate, pallidum, putamen and ICV (P = 0.0032, 8.9 × 10−6, 1.7 × 10−9, 3.5 × 10−12 and 1.0 × 10−4, respectively). Full scale IQ was lower in both deletion and duplication carriers compared to non-carriers. This is the first brain MRI study of the impact of the 16p11.2 distal CNV, and we demonstrate a specific effect on subcortical brain structures, suggesting a neuropathological pattern underlying the neurodevelopmental syndromes.

  • Diagnostic and prognostic potential of joint imaging in patients with anti-citrullinated protein antibodies

    • The introduction of novel therapeutic strategies set new goals for the patients’ outcome, which aims to achieve remission. This goal requires early diagnosis of RA and prompt efficient pharmacotherapy. The introduction of anti-citrullinated protein antibodies (ACPA) two decades ago allowed an earlier RA diagnosis. However, there are indications that ACPA positivity is still associated with higher rates of radiographic damage. As the small joints in hands and feet commonly are the first involved sites of inflammation, the role of different imaging modalities were studied regarding their diagnostic and prognostic impact for assessment of arthritis in RA. Further, ultrasound (US) and radiography were used to study the association between RA-specific antibodies and the occurrence of arthritis and joint damage in systemic lupus erythematosus (SLE).The use of US allows assessment of soft tissue like joint capsules, tendons and bursae. Used for a live scanning, it is easy to detect effusions and edema. Doppler indicates vasoproliferation were inflammation is present. Also, US seems to be more sensitive than radiography to detect minimal structural changes located at bone surfaces. We wanted to investigate whether US findings in a pre-RA stage can predict development of arthritis.Digital X-ray radiogrammetry (DXR) is a technique based on computerized analyses of standard hand radiographs to calculate peripheral bone mineral density (BMD) of the three middle metacarpal bones (DXR-BMD). In order for early treatment decisions, we aimed to study whether changes in DXR-BMD loss after 3 months can predict radiographic damage in early RA.In conclusion, the studies showed that ACPA-positivity is still associated with a higher risk of radiographic damage regardless of early treatment decisions. Therefore, close radiographic monitoring and readiness to intensive treatment is warranted in ACPA-positive patients. This thesis also shows that erosions detected by US in ACPA-positive patients with arthralgia predict development of clinical arthritis. Also, the magnitude of DXR-BMD loss helps identify patients at higher risk for future radiographic damage, and may therefore help to improve early treatment decisions. Finally, US and radiography confirm a higher rate of arthritis and erosions also in SLE patients who are positive for RA-specific antibodies.

  • The impact of helminth infection in patients with active tuberculosis

    • The geographic distribution of helminth infection and tuberculosis (TB) overlap substantially. Experimental animal models and limited data from humans have shown that intestinal helminths could subvert the host immune response towards a T-helper 2 (Th2)-type immune response and an increased regulatory T-cell activity (Tregs). This in turn affects the host's ability to mount an effective Th1 immune-mediated protection against Mycobacterium tuberculosis. However, evidence for this hypothesis in the human setting from helminth infected TB patients is limited. This thesis primarily focuses on the immunological and clinical impact of helminth infection on pulmonary TB. The kinetics of the Quantiferon-Gold (QFN) assay, which measures IFN-³ response to TB-specific antigens in whole blood was assessed and showed a modest decline during TB treatment to the level observed for healthy blood donors. We further assessed another clinical monitoring tool, the-TB-score, composed of clinical signs and symptoms of TB, and found an early decline two weeks after initiation of TB- treatment where a failure of decline correlated with increased mortality. Overall, the helminth co-infection rate was significantly higher in TB patients compared to healthy controls. Helminth co-infection was associated to a significantly higher rate of eosinophilia and IgE-levels in healthy controls and patients with tuberculosis. During the first weeks of anti-TB treatment, a marked decrease in the rate of helminth infection was observed in HIV co-infected compared to HIV-negative TB patients. However, helminth co-infection was more common in HIV negative than HIV positive TB patients. There was no detectable impact of helminth infection on the clinical presentation of pulmonary tuberculosis. At baseline, helminth co-infected TB patients showed an increased frequency of Tregs compared to helminth negative TB patients and healthy controls. This was accompanied by an increased rate of PPD stimulated IL-5 and spontaneous production of IL-10 by peripheral blood mononuclear cells among helminth co-infected TB patients. A placebo controlled randomized trial was conducted in order to test the hypothesis that albendazole treatment of helminth positive TB patients may improve the clinical response of TB by reducing the immunmodulatory effect of helminthes on TB immunity. A total of 140 helminth co-infected TB patients were randomized to albendazole (400 mg per os for three consecutive days) or placebo. No significant difference was observed between the albendazole and placebo group in terms of the primary outcome (TB score change between baseline and week 8). Among the secondary outcomes, a significant decline of peripheral eosinophil cells was observed in the albendazole treated group, but no effect on other outcome variables (changes in chest x-ray findings, IgE level and sputum smear conversion). Regarding the immunological assessment no significant difference was observed for changes in Tregs, and PPD-induced production of IFN- ³ or IL-5 although a non-significant trend of a decrease in IL-10 expressing PBMCs were observed in the albendazole group. Taken together, the burden of helminth infection was higher in TB patients than in a healthy control group. Helminth co-infection during pulmonary TB in the human setting induces an immune response characterized by increased IgE production, eosinophilia as well as increased levels of Tregs and spontaneous IL-10 production. Thus, the immunological impact of helminth infection on the outcome and risk for developing TB merits further investigation.

    • The divertor solutions obtained with the plasma edge modelling tool SOLPS5.0 are discussed. The code results are benchmarked against carefully analysed L-mode discharges at various density levels with and without impurity seeding in the full-metal tokamaks ASDEX Upgrade and JET. The role of the cross-field drifts and currents in the solutions is analysed in detail, and the improvements achieved by fully activating the drift and current terms in view of matching the experimental signals are addressed. The persisting discrepancies are also discussed.
