  • Genetic profiling of Vietnamese population from large-scale genomic analysis of non-invasive prenatal testing data
  • 2020
  • Ingår i: Scientific Reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 10:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The under-representation of several ethnic groups in existing genetic databases and studies have undermined our understanding of the genetic variations and associated traits or diseases in many populations. Cost and technology limitations remain the challenges in performing large-scale genome sequencing projects in many developing countries, including Vietnam. As one of the most rapidly adopted genetic tests, non-invasive prenatal testing (NIPT) data offers an alternative untapped resource for genetic studies. Here we performed a large-scale genomic analysis of 2683 pregnant Vietnamese women using their NIPT data and identified a comprehensive set of 8,054,515 single-nucleotide polymorphisms, among which 8.2% were new to the Vietnamese population. Our study also revealed 24,487 disease-associated genetic variants and their allele frequency distribution, especially 5 pathogenic variants for prevalent genetic disorders in Vietnam. We also observed major discrepancies in the allele frequency distribution of disease-associated genetic variants between the Vietnamese and other populations, thus highlighting a need for genome-wide association studies dedicated to the Vietnamese population. The resulted database of Vietnamese genetic variants, their allele frequency distribution, and their associated diseases presents a valuable resource for future genetic studies.

  • Impact of terrestrial runoff on organic matter, trophic state, and phytoplankton in a tropical, upland reservoir
  • 2016
  • Ingår i: Aquatic Sciences. - : Springer Science and Business Media LLC. - 1015-1621 .- 1420-9055. ; 78:2, s. 367-379
  • Tidskriftsartikel (refereegranskat)abstract
    • The impact of organic matter inputs from agricultural, forest and domestic sources on aquatic processes has been considerably less studied in tropical reservoirs relative to temperate systems despite the high number of these small aquatic systems in the tropics. Here we present the results of an in situ mesocosm study that examined the impact of allochthonous organic matter on a headwater reservoir in Northern Vietnam. We examined the impact of wastewater and soils from floodplain paddies, Acacia mangium plantations and from upland slopes on the metabolic status of the reservoir. The addition of floodplain paddy soils to the reservoir water led to a rapid switch in metabolic status from net autotrophic to net heterotrophic. In contrast, the addition of wastewater in low concentrations had less impact on the metabolic status of the reservoir, reflecting the low population density in the area. The addition of floodplain paddy soils also increased phytoplankton diversity and evenness relative to the control. In summary, soils from floodplain paddies and from A. mangium plantations had the highest impact on the reservoir, with upland soils and wastewater having less of an impact. We also found that primary production in this reservoir was nitrogen limited. In order to avoid accelerating the impact of runoff on the reservoir, future management options should perhaps focus on minimizing water and sediment runoff from upstream paddy fields and from A. mangium plantations. These results also underline the importance of studying these upland tropical water bodies that can contribute an important but, on the whole, ignored part of the global carbon balance.

  • Burden of Hospital Acquired Infections and Antimicrobial Use in Vietnamese Adult Intensive Care Units
  • 2016
  • Ingår i: PLOS ONE. - : PUBLIC LIBRARY SCIENCE. - 1932-6203. ; 11:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Background Vietnam is a lower middle-income country with no national surveillance system for hospital-acquired infections (HAIs). We assessed the prevalence of hospital-acquired infections and antimicrobial use in adult intensive care units (ICUs) across Vietnam. Methods Monthly repeated point prevalence surveys were systematically conducted to assess HAI prevalence and antimicrobial use in 15 adult ICUs across Vietnam. Adults admitted to participating ICUs before 08: 00 a.m. on the survey day were included. Results Among 3287 patients enrolled, the HAI prevalence was 29.5% (965/3266 patients, 21 missing). Pneumonia accounted for 79.4% (804/1012) of HAIs Most HAIs (84.5% [855/1012]) were acquired in the survey hospital with 42.5% (363/855) acquired prior to ICU admission and 57.5% (492/855) developed during ICU admission. In multivariate analysis, the strongest risk factors for HAI acquired in ICU were: intubation (OR 2.76), urinary catheter (OR 2.12), no involvement of a family member in patient care (OR 1.94), and surgery after admission (OR 1.66). 726 bacterial isolates were cultured from 622/1012 HAIs, most frequently Acinetobacter baumannii (177/726 [24.4%]), Pseudomonas aeruginosa (100/726 [13.8%]), and Klebsiella pneumoniae (84/726 [11.6%]), with carbapenem resistance rates of 89.2%, 55.7%, and 14.9% respectively. Antimicrobials were prescribed for 84.8% (2787/ 3287) patients, with 73.7% of patients receiving two or more. The most common antimicrobial groups were third generation cephalosporins, fluoroquinolones, and carbapenems (20.1%, 19.4%, and 14.1% of total antimicrobials, respectively). Conclusion A high prevalence of HAIs was observed, mainly caused by Gram-negative bacteria with high carbapenem resistance rates. This in combination with a high rate of antimicrobial use illustrates the urgent need to improve rational antimicrobial use and infection control efforts.

  • Assortative mating and the evolution of desirability covariation
  • 2019
  • Ingår i: Evolution and human behavior. - : Elsevier. - 1090-5138 .- 1879-0607. ; 40:5, s. 479-491
  • Tidskriftsartikel (refereegranskat)abstract
    • Mate choice lies dose to differential reproduction, the engine of evolution. Patterns of mate choice consequently have power to direct the course of evolution. Here we provide evidence suggesting one pattern of human mate choice-the tendency for mates to be similar in overall desirability-caused the evolution of a structure of correlations that we call the d factor. We use agent-based models to demonstrate that assortative mating causes the evolution of a positive manifold of desirability, d, such that an individual who is desirable as a mate along any one dimension tends to be desirable across all other dimensions. Further, we use a large cross-cultural sample with n = 14,478 from 45 countries around the world to show that this d-factor emerges in human samples, is a cross-cultural universal, and is patterned in a way consistent with an evolutionary history of assortative mating. Our results suggest that assortative mating can explain the evolution of a broad structure of human trait covariation.

  • Contrasting Computational Models of Mate Preference Integration Across 45 Countries
  • 2019
  • Ingår i: Scientific Reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 9
  • Tidskriftsartikel (refereegranskat)abstract
    • Humans express a wide array of ideal mate preferences. Around the world, people desire romantic partners who are intelligent, healthy, kind, physically attractive, wealthy, and more. In order for these ideal preferences to guide the choice of actual romantic partners, human mating psychology must possess a means to integrate information across these many preference dimensions into summaries of the overall mate value of their potential mates. Here we explore the computational design of this mate preference integration process using a large sample of n = 14,487 people from 45 countries around the world. We combine this large cross-cultural sample with agent-based models to compare eight hypothesized models of human mating markets. Across cultures, people higher in mate value appear to experience greater power of choice on the mating market in that they set higher ideal standards, better fulfill their preferences in choice, and pair with higher mate value partners. Furthermore, we find that this cross-culturally universal pattern of mate choice is most consistent with a Euclidean model of mate preference integration.

  • 2020
  • Ingår i: Frontiers in Psychology. - : Frontiers Media SA. - 1664-1078. ; 11
  • Tidskriftsartikel (refereegranskat)abstract
    • Introduction: Seventy-nine percent of internet users use Facebook, and on average they access Facebook eight times a day (Greenwood et al., 2016). To put these numbers into perspective, according to Clement (2019), around 30% of the world's population uses this Online Social Network (OSN) site.Despite the constantly growing body of academic research on Facebook (Chou et al., 2009; Back et al., 2010; Kaplan and Haenlein, 2010; McAndrew and Jeong, 2012; Wilson et al., 2012; Krasnova et al., 2017), there remains limited research regarding the motivation behind Facebook use across different cultures. Our main goal was to collect data from a large cross-cultural sample of Facebook users to examine the roles of sex, age, and, most importantly, cultural differences underlying Facebook use.

  • 2017
  • Ingår i: Journal of Intensive Care. - : BioMed Central (BMC). - 2052-0492. ; 5
  • Tidskriftsartikel (refereegranskat)abstract
    • Ventilator-associated respiratory infection (VARI) is a significant problem in resource-restricted intensive care units (ICUs), but differences in casemix and etiology means VARI in resource-restricted ICUs may be different from that found in resource-rich units. Data from these settings are vital to plan preventative interventions and assess their cost-effectiveness, but few are available.

  • Sex Differences in Mate Preferences Across 45 Countries : A Large-Scale Replication
  • 2020
  • Ingår i: Psychological Science. - : SAGE Publications. - 0956-7976 .- 1467-9280. ; 31:4, s. 408-423
  • Tidskriftsartikel (refereegranskat)abstract
    • Considerable research has examined human mate preferences across cultures, finding universal sex differences in preferences for attractiveness and resources as well as sources of systematic cultural variation. Two competing perspectives-an evolutionary psychological perspective and a biosocial role perspective-offer alternative explanations for these findings. However, the original data on which each perspective relies are decades old, and the literature is fraught with conflicting methods, analyses, results, and conclusions. Using a new 45-country sample (N = 14,399), we attempted to replicate classic studies and test both the evolutionary and biosocial role perspectives. Support for universal sex differences in preferences remains robust: Men, more than women, prefer attractive, young mates, and women, more than men, prefer older mates with financial prospects. Cross-culturally, both sexes have mates closer to their own ages as gender equality increases. Beyond age of partner, neither pathogen prevalence nor gender equality robustly predicted sex differences or preferences across countries.

  • Determination of platinum in human subcellular microsamples by inductively coupled plasma mass spectrometry
  • 2007
  • Ingår i: Analytical Biochemistry. - : Elsevier BV. - 0003-2697. ; 363, s. 135-42
  • Tidskriftsartikel (refereegranskat)abstract
    • A fast and robust method for the determination of platinum in human subcellular microsamples by inductively coupled plasma mass spectrometry was developed, characterized, and validated. Samples of isolated DNA and exosome fractions from human ovarian (2008) and melanoma (T289) cancer cell lines were used. To keep the sample consumption to approximately 10 μl and obtain a high robustness of the system, a flow injection sample introduction system with a 4.6-μl sample loop was used in combination with a conventional pneumatic nebulizer and a spray chamber. The system was optimized with respect to signal/noise ratio using a multivariate experimental design. The system proved to be well suited for routine analysis of large sample series, and several hundreds of samples could be analyzed without maintenance or downtime. The detection limit of the method was 0.12 pg (26 pg/g) platinum. To avoid systematic errors from nonspectral interferences, it was necessary to use reagent matched calibration standards or isotope dilution analysis. An uncertainty budget was constructed to estimate the total expanded uncertainty of the method, giving a quantification limit of 2.3 pg (0.5 ng/g) platinum in DNA samples. The uncertainty was sufficiently low to study quantitative differences in the formation of Pt–DNA adducts after treatment with cisplatin using different exposure times and concentrations.

  • Recycled pangasius pond sediments as organic fertilizer for vegetables cultivation : strategies for sustainable food production
  • 2023
  • Ingår i: Clean Technologies and Environmental Policy. - : Springer Science and Business Media LLC. - 1618-954X .- 1618-9558. ; 25:2, s. 369-380
  • Tidskriftsartikel (refereegranskat)abstract
    • Intensive Pangasius aquaculture farming contributes to increased income and profits in Vietnam, but is also associated with many environmental problems, including pollution from nutrient-rich water and sediments that are released into the environment. This article investigates the feasibility of using Pangasius pond sediments (PPS) in combination with amendments of rice straw (RS) to fertilize cucumber (Cucumis sativus L.) and water spinach (Ipomoea aquatica Forssk) plant. The results showed that mixing a ratio of 30% PPS with 70% RS produced the highest nutrient concentrations. The study demonstrates that organic fertilizer produced from PPS in combination with chemical fertilizer can replace 2575% of the inorganic fertilizers used as a nutrient source for cucumber and water spinach, and also increase the vegetable production. The highest yields were found for treatment PPS-3 (a combination of 50% chemical fertilizer and 50% organic fertilizer used on cucumber), and for treatment PPSRS-4 (a combination of 25% chemical fertilizer and 75% organic fertilizer used on water spinach) in both the wet and dry season (P˂0.05). The lowest yields were found when growing cucumbers and water spinach either with 100% organic fertilizer (the treatment PPS-5) or with 100% chemical fertilizer (the control treatment PPS-1), respectively (P˂0.05). Overall, the results indicated that the recycling of PPS as organic fertilizer and chemical fertilizer in combination for vegetable cultivation is more environmentally friendly as compared to using only chemical fertilizers.

  • Hydrogen gas sensor based on self-heating effect of SnO2/Pt thin film with ultralow power consumption
  • 2024
  • Ingår i: International journal of hydrogen energy. - : Elsevier. - 0360-3199 .- 1879-3487. ; 61, s. 774-782
  • Tidskriftsartikel (refereegranskat)abstract
    • Self-heating of sensing elements on gas sensors is an effective solution to avoid using external heaters. In this paper, a self-heated hydrogen gas sensor is presented. The sensor was created using the DC sputtering method, which involved fabricating it on a thermal-insulating Kapton flexible substrate. This process utilized a thin film of SnO2 with thick 50 nm that was modified with nanoclusters of Pt, serving as the sensing material. The SnO2/Pt material film was analyzed for microstructure and composition by SEM, XRD, and XPS analysis. Infrared images show that the self-heating effect is mainly concentrated in the strip of gas-sensitive material. It showed many good performances, such as high sensitivity (able to detect down to 50 ppm of H2), good selectivity (poor response to CO, NH3, H2S, and NO2), the sensor's performance is little changed by environmental humidity, and low power consumption (89 μW at 5V). The sensor is also stable and low-cost, suitable for portable H2 detection devices due to its low generated heat and small size.

  • 2022
  • Ingår i: Aquaculture Economics & Management. - : Informa UK Limited. - 1365-7305 .- 1551-8663. ; 26:2, s. 152-170
  • Tidskriftsartikel (refereegranskat)abstract
    • This study assesses capital assets and factors influencing on farmers’ decision to participate in social networks, and how these contribute to and affect household’s profits of small-scale shrimp farmers in four coastal provinces in the Mekong Delta, Vietnam. Data was collected through observations, and informal discussions and interviews with 350 small-scale shrimp farmer households. The results show that farmer’s age, education of household head, dependency ratio, village residence time, village population density are important factors influencing positively on shrimp farmers´ participation in social networks. The study also shows that contacts with fisheries association, traders, wholesale agencies, trust, household savings, labors’ education, household’s agricultural land size, post larvae quality and equipment significantly influenced on the profit variable with 1–10%. The results also indicate that a 10% increase in social capital would bring an increase in household income of 3.9%.

  • Design and fabrication of effective gradient temperature sensor array based on bilayer SnO2/Pt for gas classification
  • 2022
  • Ingår i: Sensors and actuators. B, Chemical. - : Elsevier. - 0925-4005 .- 1873-3077. ; 351
  • Tidskriftsartikel (refereegranskat)abstract
    • Classification of different gases is important, and it is possible to use different gas sensors for this purpose. Electronic noses, for example, combine separated gas sensors into an array for detecting different gases. However, the use of separated sensors in an array suffers from being bulky, high-energy consumption and complex fabrication processes. Generally, gas sensing properties, including gas selectivity, of semiconductor gas sensors are strongly dependent on their working temperature. It is therefore feasible to use a single device composed of identical sensors arranged in a temperature gradient for classification of multiple gases. Herein, we introduce a design for simple fabrication of gas sensor array based on bilayer Pt/SnO2 for real-time monitoring and classification of multiple gases. The study includes design simulation of the sensor array to find an effective gradient temperature, fabrication of the sensors and test of their performance. The array, composed of five sensors, was fabricated on a glass substrate without the need of backside etching to reduce heat loss. A SnO2 thin film sensitized with Pt on top deposited by sputtering was used as sensing material. The sensor array was tested against different gases including ethanol, methanol, isopropanol, acetone, ammonia, and hydrogen. Radar plots and principal component analysis were used to visualize the distinction of the tested gases and to enable effective classification.

  • Artemisinin pharmacokinetics and efficacy in uncomplicated-malaria patients treated with two different dosage regimens.
  • 2002
  • Ingår i: Antimicrobial agents and chemotherapy. - 0066-4804. ; 46:4, s. 1026-31
  • Tidskriftsartikel (refereegranskat)abstract
    • The immediate efficacies of two oral dosage regimens of artemisinin were investigated in 77 male and female adult Vietnamese falciparum malaria patients randomly assigned to treatment with either 500 mg of artemisinin daily for 5 days (group A; n = 40) or artemisinin at a dose of 100 mg per day for 2 days, with the dose increased to 250 mg per day for 2 consecutive days and with a final dose of 500 mg on the fifth day (group B; n = 37). Parasitemia was monitored every 4 h. The average parasite clearance time was longer in group B than in group A (means +/- standard deviations, 50 +/- 23 and 34 +/- 14 h, respectively; P < 0.01). Artemisinin concentrations in saliva samples obtained on days 1 and 5 were quantified by high-performance liquid chromatography. The average oral clearance, based on saliva drug concentrations in group B patients, was twofold higher than that in group A patients on day 1 (P < 0.01), with no differences in drug half-lives (P = 0.40), indicating a saturable first-pass metabolism. Female patients had higher oral clearance values on day 1. Artemisinin's pharmacokinetic parameters were similar on day 5 in both groups, although a significant increase in oral clearance from day 1 to day 5 was evident. Thus, artemisinin exhibited both dose- and time-dependent pharmacokinetics. The escalating dose studied did not result in higher artemisinin concentrations toward the end of the treatment period.

  • Application of CDIO in Non-Engineering Programmes - Motives, Implementation and Experiences
  • 2016
  • Ingår i: Proceedings of 2016 International CDIO Conference, Turku, Finland.
  • Konferensbidrag (refereegranskat)abstract
    • The aims of the paper are to present a set of general recommendations for how to implement CDIO in non-engineering programmes, to show how they can be applied in practice, and to discuss associated benefits and challenges. The application of the recommendations is demonstrated in case studies of non-engineering programmes that have implemented CDIO. The subject areas of the programmes include art, science, food processing, business, and library science. The case descriptions are purposively relatively detailed aiming to enable a transfer of approaches and experiences from the cases to other non-engineering programmes. Common benefits of applying CDIO to non-engineering programmes include a stronger connection to the professional context, and strengthened programme development and quality assurance. Common CDIO implementation challenges for non-engineering programmes are found to be similar as for engineering programmes, for example, the training of faculty to teach skills beyond their subject specialty, such as design and communication.

  • Landslide Susceptibility Mapping Using Single Machine Learning Models : A Case Study from Pithoragarh District, India
  • 2021
  • Ingår i: Advances in Civil Engineering / Hindawi. - : Hindawi Publishing Corporation. - 1687-8086 .- 1687-8094. ; 2021
  • Tidskriftsartikel (refereegranskat)abstract
    • Landslides are one of the most devastating natural hazards causing huge loss of life and damage to properties and infrastructures and adversely affecting the socioeconomy of the country. Landslides occur in hilly and mountainous areas all over the world. Single, ensemble, and hybrid machine learning (ML) models have been used in landslide studies for better landslide susceptibility mapping and risk management. In the present study, we have used three single ML models, namely, linear discriminant analysis (LDA), logistic regression (LR), and radial basis function network (RBFN), for landslide susceptibility mapping at Pithoragarh district, as these models are easy to apply and so far they have not been used for landslide study in this area. The main objective of this study is to evaluate the performance of these single models for correctly identifying landslide susceptible zones for their further application in other areas. For this, ten important landslide affecting factors, namely, slope, aspect, curvature, elevation, land cover, lithology, geomorphology, distance to rivers, distance to roads, and overburden depth based on the local geoenvironmental conditions, were considered for the modeling. Landslide inventory of past 398 landslide events was used in the development of models. The data of past landslide events (locations) was randomly divided into a 70/30 ratio for training (70%) and validation (30%) of the models. Standard statistical measures, namely, accuracy (ACC), specificity (SPF), sensitivity (SST), positive predictive value (PPV), negative predictive value (NPV), Kappa, root mean square error (RMSE), and area under the receiver operating characteristic curve (AUC), were used to evaluate the performance of the models. Results indicated that the performance of all the models is very good (AUC > 0.90) and that of the LR model is the best (AUC = 0.926). Therefore, these single ML models can be used for the development of accurate landslide susceptibility maps. Our study demonstrated that the single models which are easy to use and can compete with the complex ensemble/hybrid models can be applied for landslide susceptibility mapping in landslide-prone areas.

    • Massive MIMO (Multiple Input Multiple Output) has demonstrated as a potential candidate for 5G-and-beyond wireless networks. Instead of using Gaussian signals as most of the previous works, this paper makes a novel contribution by investigating the transmission quality of image data by utilizing the Massive MIMO technology. We first construct a framework to decode the image signal from the noisy received data in the uplink Massive MIMO transmission by utilizing the alternating direction method of multipliers (ADMM) approach. Then, a low-pass filter is exploited to enhance the efficiency of the remaining noise and artifacts reduction in the recovered image. Numerical results demonstrate the necessity of a post-filtering process in enhancing the quality of image recovery.

  • Novel Time Series Bagging Based Hybrid Models for Predicting Historical Water Levels in the Mekong Delta Region, Vietnam
  • 2022
  • Ingår i: CMES - Computer Modeling in Engineering & Sciences. - : Tech Science Press. - 1526-1492 .- 1526-1506. ; 131:3, s. 1431-1449
  • Tidskriftsartikel (refereegranskat)abstract
    • Water level predictions in the river, lake and delta play an important role in flood management. Every year Mekong River delta of Vietnam is experiencing flood due to heavy monsoon rains and high tides. Land subsidence may also aggravate flooding problems in this area. Therefore, accurate predictions of water levels in this region are very important to forewarn the people and authorities for taking timely adequate remedial measures to prevent losses of life and property. There are so many methods available to predict the water levels based on historical data but nowadays Machine Learning (ML) methods are considered the best tool for accurate prediction. In this study, we have used surface water level data of 18 water level measurement stations of the Mekong River delta from 2000 to 2018 to build novel time-series Bagging based hybrid ML models namely: Bagging (RF), Bagging (SOM) and Bagging (M5P) to predict historical water levels in the study area. Performances of the Bagging-based hybrid models were compared with Reduced Error Pruning Trees (REPT), which is a benchmark ML model. The data of 19 years period was divided into 70:30 ratio for the modeling. The data of the period 1/2000 to 5/2013 (which is about 70% of total data) was used for the training and for the period 5/2013 to 12/2018 (which is about 30% of total data) was used for testing (validating) the models. Performance of the models was evaluated using standard statistical measures: Coefficient of Determination (R2), Root Mean Square Error (RMSE) and Mean Absolute Error (MAE). Results show that the performance of all the developed models is good (R2 > 0.9) for the prediction of water levels in the study area. However, the Bagging-based hybrid models are slightly better than another model such as REPT. Thus, these Bagging-based hybrid time series models can be used for predicting water levels at Mekong data.

    • Objective: To establish a hospital-based surveillance network with national coverage for antimicrobial resistance (AMR) and antibiotic consumption in Viet Nam. Methods: A 16-hospital network (Viet Nam Resistance: VINARES) was established and consisted of national and provincial-level hospitals across the country. Antimicrobial susceptibility testing results from routine clinical diagnostic specimens and antibiotic consumption data in Defined Daily Dose per 1000 bed days (DDD/1000 patient-days) were prospectively collected and analysed between October 2012 and September 2013. Results: Data from a total of 24 732 de-duplicated clinical isolates were reported. The most common bacteria were: Escherichia coli (4437 isolates, 18%), Klebsiella spp. (3290 isolates, 13%) and Acinetobacter spp. (2895 isolates, 12%). The hospital average antibiotic consumption was 918 DDD/1000 patient-days. Third-generation cephalosporins were the most frequently used antibiotic class (223 DDD/1000 patient-days, 24%), followed by fluoroquinolones (151 DDD/1000 patient-days, 16%) and second-generation cephalosporins (112 DDD/1000 patient-days, 12%). Proportions of antibiotic resistance were high: 1098/1580 (69%) Staphylococcus aureus isolates were methicillin-resistant (MRSA); 115/344 isolates (33%) and 90/358 (25%) Streptococcus pneumoniae had reduced susceptibility to penicillin and ceftriaxone, respectively. A total of 180/2977 (6%) E. coli and 242/1526 (16%) Klebsiella pneumoniae were resistant to imipenem, respectively; 602/1826 (33%) Pseudomonas aeruginosa were resistant to ceftazidime and 578/1765 (33%) to imipenem. Of Acinetobacter spp. 1495/2138 (70%) were resistant to carbapenems and 2/333 (1%) to colistin. Conclusions: These data are valuable in providing a baseline for AMR among common bacterial pathogens in Vietnamese hospitals and to assess the impact of interventions. (C) 2019 The Authors. • Cryogels with interconnected channels allow high flow-through properties and mass transfer when dealing with complex mixtures such as non-clarified crude extracts. However, their mechanical strength can be challenged due to a large void volume inside the polymeric network. We have addressed this problem by forming a double-layer cryogel applied as a dye-affinity chromatography gel. In this study, poly(acrylamide-co-allyl glycidyl ether) cryogel was prepared at sub-zero temperature. The second layer was then prepared inside the primary cryogel under the same conditions to form a double-layer network. Cibacron Blue F3GA, a dye molecule, was immobilized on the surface of the cryogels. Bovine serum albumin was used as a model molecule to study the adsorption/elution procedure in batch and continuous modes. The maximum batch binding capacity and the dynamic binding capacity for the single-layer cryogel were 18 and 0.11, and for the double-layer cryogel were 7.5 and 0.9 mg/g of gel, respectively. However, the mechanical stability of the double-layer cryogel increased 7-fold (144 kPa). It was found that the kinetic and adsorption isotherms follow pseudo-second-order and Freundlich models, respectively. The regeneration of the columns after adsorption/elution cycles was evaluated, and no significant loss of capacity was observed after 10 cycles.

  • Hypospadias : gene mapping and candidate gene studied
  • 2009
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Hypospadias is a common congenital malformation in boys, characterized by incomplete fusion of the urethral folds, abnormal opening of urethra and different degrees of curvature of the penis. In Sweden, the incidence of hypospadias is 1.14 per 300 male live-births according to the annual Swedish Malformation Registry. Hypospadias is considered to be a complex genetic disorder caused by the interplay between environmental factors and the additive effects of multiple genes. Several observations suggest that hypospadias are under genetic influence. To identify disease genes in complex disorders, two main strategies have been used. We have performed a whole genome screening for linkage in families with additional cases of hypospadias and also performed DNA sequencing of candidate genes for hypospadias. To identify the chromosomal loci involved in the pathogenesis of hypospadias, a genome-wide linkage analysis in a three-generational family showing autosomal dominant inheritance of hypospadias was performed. Fifteen individuals, whereof seven affected, were genotyped within a total of 426 microsatellite markers and the genotyping results were analyzed using parametric and non-parametric linkage analyses. The genome-wide linkage analysis and subsequent fine mapping gave a maximum linkage in both parametric (LOD score 2.71) and non-parametric (NPL score 5.01) single-point analyses for marker D7S640. A susceptibility haplotype shared by all affected boys was identified by markers D7S2519 and D7S2442, respectively. This finding suggests a novel hypospadias locus (25 cM) at chromosome 7q32.2- q36.1 (Study I). In a previous genome-wide scan for familial hypospadias, we have identified suggestive linkage in nine chromosomal regions. To extend this analysis new families and additional markers were included. The fine mapping analysis displayed an increased LOD score on chromosome 8q24.1 and 10p15 in altogether 82 families. From the chromosome region 10p15, we sequenced the AKR1C3 and KLF6 genes that have possible roles during male urethra development. Sequencing analysis showed one mutation (c.697A>G, p.A215T) in the AKR1C3 gene and one mutation (c.496T>C, p.P166S) in the KLF6 gene. In addition, three polymorphisms (rs3763676, rs12529 and rs7741) in the 5´end of the AKR1C3gene showed a significant association with hypospadias. These findings indicate that the AKR1C3 and possibly also the KLF6 genes function as geneticrisk factors for hypospadias (Study II). Several different pathways are implicated in the male genital development; of those the androgen pathway iscrucially important. Therefore the androgen receptor (AR) and the 5 alpha reductase (SRD5A2) genes were sequenced in 38 isolated hypospadias cases. One mutation in the AR gene (p.Q798E) and another in the SRD5A2 gene (p.K199S) was detected. Interestingly, also a high frequency of the rare leucine allele in theV89L polymorphism of the SRD5A2 gene was found. The leucine allele has been shown to confer a decreased activity of enzyme, thus this allele may be a risk factor for hypospadias. This finding was confirmed in total 158 hypospadias cases compared to 96 controls (Study III). The MAMLD1 (CXorf6) gene is the first gene causing isolated hypospadias. The MAMLD1 gene was sequenced in DNA from 97 sporadic hypospadias cases to elucidate the role of this gene in hypospadias. One new mutation, p.Q529K, that is predicted to affect the splicing process, was found in a boy with severe hypospadias. The variants, p.V432A and p.531ins3Q, have been reported previously and are indicated in this study as polymorphisms. Additionally, a significant association between the p.N589S polymorphism (rs2073043) that may alter the predicted protein structure and hypospadias was detected (p-value <0.05). The combination of two rare alleles of p.N589S and p.P286S (rs41313406) is over represented among cases. It may be that these two polymorphisms are inherited together as a haplotype (T-G), thus increasing the risk for hypospadias. These findings suggest that in a few cases hypospadias is caused by mutations in the MAMLD1 gene, and that the polymorphism (p.N589S) is a genetic risk factor (Study IV).
