  • Pathogenic Huntingtin Repeat Expansions in Patients with Frontotemporal Dementia and Amyotrophic Lateral Sclerosis.

    • We examined the role of repeat expansions in the pathogenesis of frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS) by analyzing whole-genome sequence data from 2,442 FTD/ALS patients, 2,599 Lewy body dementia (LBD) patients, and 3,158 neurologically healthy subjects. Pathogenic expansions (range, 40-64 CAG repeats) in the huntingtin (HTT) gene were found in three (0.12%) patients diagnosed with pure FTD/ALS syndromes but were not present in the LBD or healthy cohorts. We replicated our findings in an independent collection of 3,674 FTD/ALS patients. Postmortem evaluations of two patients revealed the classical TDP-43 pathology of FTD/ALS, as well as huntingtin-positive, ubiquitin-positive aggregates in the frontal cortex. The neostriatal atrophy that pathologically defines Huntington's disease was absent in both cases. Our findings reveal an etiological relationship between HTT repeat expansions and FTD/ALS syndromes and indicate that genetic screening of FTD/ALS patients for HTT repeat expansions should be considered.

    • Scholars across disciplines increasingly hear calls for more open and collaborative approaches to scientific research. The concept of Open Innovation in Science (OIS) provides a framework that integrates dispersed research efforts aiming to understand the antecedents, contingencies, and consequences of applying open and collaborative research practices. While the OIS framework has already been taken up by science of science scholars, its conceptual underpinnings require further specification. In this essay, we critically examine the OIS concept and bring to light two key aspects: 1) how OIS builds upon Open Innovation (OI) research by adopting its attention to boundary-crossing knowledge flows and by adapting other concepts developed and researched in OI to the science context, as exemplified by two OIS cases in the area of research funding; 2) how OIS conceptualises knowledge flows across boundaries. While OI typically focuses on well-defined organisational boundaries, we argue that blurry and even invisible boundaries between communities of practice may more strongly constrain flows of knowledge related to openness and collaboration in science. Given the uptake of this concept, this essay brings needed clarity to the meaning of OIS, which has no particular normative orientation towards a close coupling between science and industry. We end by outlining the essay's contributions to OI and the science of science, as well as to science practitioners.

  • Creating and Capturing Value through Crowdsourcing

    • Examples of the value that can be created and captured through crowdsourcing go back to at least 1714 when the UK used crowdsourcing to solve the Longitude Problem, obtaining a solution that would enable the UK to become the dominant maritime force of its time. Today, Wikipedia uses crowds to provide entries for the world's largest and free encyclopedia. Partly fueled by the value that can be created and captured through crowdsourcing, interest in researching the phenomenon has been remarkable.Despite this - or perhaps because of it - research into crowdsourcing has been conducted in different research silos, within the fields of management (from strategy to finance to operations to information systems), biology, communications, computer science, economics, political science, among others. In these silos, crowdsourcing takes names such as broadcast search, innovation tournaments, crowdfunding, community innovation, distributed innovation, collective intelligence, open source, crowdpower, and even open innovation. This book aims to assemble chapters from many of these silos, since the ultimate potential of crowdsourcing research is likely to be attained only by bridging them. Chapters provide a systematic overview of the research on crowdsourcing from different fields based on a more encompassing definition of the concept, its difference for innovation, and its value for both private and public sector.

    • Knowledge sharing is particularly important for co-creating, discussing, or acquiring innovative ideas. Crowdsourcing, as an enabler of open innovation, has raised the question about the kind of organising forms and/or managerial interventions it may require or underpin. However, there is little consensus in management studies on how to best design a crowdsourcing initiative (contest) with regard to the mechanisms to engage an online community. In this paper, starting from an exploratory case study on the project “Stati Generali della Formazione e del Lavoro” (General Assembly on Training and Work)—a crowdsourcing experience designed for a large community of professional trainers, planned and managed by University of Milano-Bicocca and AIF Academy (Associazione Italiana Formatori), a broad representative association of Italian trainers—we study the factors influencing the decision of the participants (a.k.a., solvers) to become involved (and to what extent) in a contest. The study could contribute to the debate on crowdsourcing by both underlining important governance factors involved and providing empirical evidence of the link between management strategies and crowdsourcing success.

    • According to conventional wisdom on crowdsourcing, the number of people defines the crowd and maximization of this number is often assumed to be the goal of any crowdsourcing exercise. However, some structural characteristics of the crowd might be more important than the sheer number of participants. These characteristics include (1) the growth rate and its attractiveness to members, (2) equality among members, (3) density within provisional boundaries, (4) goal orientation of the crowd, and (5) “seriality” of the interactions between members. Therefore, a typology is proposed that may allow managers to position their companies’ initiatives among four strategic types for driving innovation: crowd crystals, online communities, closed crowds, and open crowds. Incumbent companies may prefer closed and controlled access to the crowd, limiting the potential for gaining results and insights from fully open crowd-driven innovation initiatives. Thus, the effects on industries and organizations by open crowds are still to be explored.
