    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Klebsiella (K.) pneumoniae is a common cause of pneumonia-derived sepsis. Myeloid related protein 8 (MRP8, S100A8) and MRP14 (S100A9) are the most abundant cytoplasmic proteins in neutrophils. They can form MRP8/14 heterodimers that are released upon cell stress stimuli. MRP8/14 reportedly exerts antimicrobial activity, but in acute fulminant sepsis models MRP8/14 has been found to contribute to organ damage and death. We here determined the role of MRP8/14 in K. pneumoniae sepsis originating from the lungs, using an established model characterized by gradual growth of bacteria with subsequent dissemination. Infection resulted in gradually increasing MRP8/14 levels in lungs and plasma. Mrp14 deficient (mrp14(-/-)) mice, unable to form MRP8/14 heterodimers, showed enhanced bacterial dissemination accompanied by increased organ damage and a reduced survival. Mrp14(-/-) macrophages were reduced in their capacity to phagocytose Klebsiella. In addition, recombinant MRP8/14 heterodimers, but not MRP8 or MRP14 alone, prevented growth of Klebsiella in vitro through chelation of divalent cations. Neutrophil extracellular traps (NETs) prepared from wildtype but not from mrp14(-/-) neutrophils inhibited Klebsiella growth; in accordance, the capacity of human NETs to kill Klebsiella was strongly impaired by an anti-MRP14 antibody or the addition of zinc. These results identify MRP8/14 as key player in protective innate immunity during Klebsiella pneumonia.

  • Cocreating injury prevention training for youth team handball : bridging theory and practice

    • Although it is advocated that end-users are engaged in developing evidence-based injury prevention training to enhance the implementation, this rarely happens. The € Implementing injury Prevention training ROutines in TEams and Clubs in youth Team handball (I-PROTECT)' uses an ecological participatory design incorporating the perspectives of multiple stakeholders throughout the project. Within the I-PROTECT project, the current study aimed to describe the development of holistic injury prevention training specifically for youth handball players through using knowledge from both end-users (coaches and players) and researchers/handball experts. Employing action evaluation within participatory action research, the cyclical development process included three phases: research team preparation, handball expert-based preparation and end-user evaluation to develop injury prevention training incorporating both physical and psychological perspectives. To grow the knowledge of the interdisciplinary research team, rethinking was conducted within and between phases based on participants' contributions. Researchers and end-users cocreated examples of handball-specific exercises, including injury prevention physical principles (movement technique for upper and lower extremities, respectively, and muscle strength) combined with psychological aspects (increase end-user motivation, task focus and body awareness) to integrate into warm-up and skills training within handball practice. A cyclical development process that engaged researchers/handball experts and end-users to cocreate evidence-based, theory-informed and context-specific injury prevention training specifically for youth handball players generated a first pilot version of exercises including physical principles combined with psychological aspects to be integrated within handball practice. © 2022 BMJ Publishing Group.   • Modelling potential yield capacity in conifers using Swedish long-term experiments

    • Information on forest site productivity is a key component to assess the carbon sequestration potential of boreal forests. While site index (SI) is commonly used to indicate forest site productivity, expressions of SI in the form of yield capacity (potential maximum mean annual volume increment) is desirable since volume yield is central to the economic and ecological analyses of a given species and site. This paper assessed the functional relationship between SI and yield capacity on the basis of yield plot data from long-term experiments measured over several decades for Norway spruce (Picea abies), Scots pine (Pinus sylvestris), Lodgepole pine (Pinus contorta) and Larch (Larix decidua and Larix sibirica) in Sweden. Component models of total basal area and volume yield were also developed. SI was determined by existing height development functions using top height and age, whereas functions for stand-level (m2 ha- 1) basal area development were constructed based on age, SI and initial stand density using difference equations and nonlinear mixed-effects models. The relation between volume yield (m3 ha- 1) and top height was adjusted with total basal area production through nonlinear mixed-effects models. Species-specific parametric regression models were used to construct functional relationships between SI and yield capacity. The root mean square errors of the species-specific models ranged from 2 to 6% and 10-18% of the average values for the basal area and volume equations, respectively. For the yield capacity functions, the explained variations (R2) were within 80-96%. We compared our yield capacity functions to earlier functions of the species and significant differences were observed in both lower and higher SI classes, especially, for Scots pine and Norway spruce. The new functions give better prediction of yield capacity in current growing conditions; hence, they could later be used for comparing tree species' production under similar site and management regimes in Sweden.

    • Nutrient supply to ecosystems has major effects on ecological diversity, but it is unclear to what degree the shape of this relationship is general versus dependent on the specific environment or community. Although the diet composition in terms of the source or proportions of different nutrient types is known to affect gut microbiota composition, the relationship between the quantity of nutrients supplied and the abundance and diversity of the intestinal microbial community remains to be elucidated. Here, we address this relationship using replicate populations of Drosophila melanogaster maintained over multiple generations on three diets differing in the concentration of yeast (the only source of most nutrients). While a 6.5-fold increase in yeast concentration led to a 100-fold increase in the total abundance of gut microbes, it caused a major decrease in their alpha diversity (by 45-60% depending on the diversity measure). This was accompanied by only minor shifts in the taxonomic affiliation of the most common operational taxonomic units (OTUs). Thus, nutrient concentration in host diet mediates a strong negative relationship between the nutrient abundance and microbial diversity in the Drosophila gut ecosystem.

  • iPTF Archival Search for Fast Optical Transients

    • There has been speculation about a class of relativistic explosions with an initial Lorentz factor Gamma(init) smaller than that of classical gamma-ray bursts (GRBs). These dirty fireballs would lack prompt GRB emission but could be pursued via their optical afterglow, appearing as transients that fade overnight. Here we report a search for such transients (that fade by 5-sigma in magnitude overnight) in four years of archival photometric data from the intermediate Palomar Transient Factory (iPTF). Our search criteria yielded 50 candidates. Of these, two were afterglows to GRBs that had been found in dedicated follow-up observations to triggers from the Fermi GRB Monitor. Another (iPTF14yb) was a GRB afterglow discovered serendipitously. Eight were spurious artifacts of reference image subtraction, and one was an asteroid. The remaining 38 candidates have red stellar counterparts in external catalogs. The photometric and spectroscopic properties of the counterparts identify these transients as strong flares from M dwarfs of spectral type M3-M7 at distances of d approximate to 0.15-2.1 kpc; three counterparts were already spectroscopically classified as late-type M stars. With iPTF14yb as the only confirmed relativistic outflow discovered independently of a high-energy trigger, we constrain the all-sky rate of transients that peak at m = 18 and fade by Delta m = 2 mag in Delta t = 3 hr to be 680 yr(-1), with a 68% confidence interval of 119-2236 yr(-1). This implies that the rate of visible dirty fireballs is at most comparable to that of the known population of long-duration GRBs.

  • The millennium shift: Investigating the relationship between environment and growth trends of Norway spruce and Scots pine in northern Europe

    • For boreal forests in colder climates, changes in environmental conditions are hypothesised to substantially affect ecosystem processes. In this study, trends of top height growth of Scots pine (Pinus sylvestris L.) and Norway spruce (Picea abies (L.) Karst) were analysed using permanent sample plot data from more than 300 long-term experimental sites distributed from temperate zones to the boreal forest conditions in Sweden. By regression analyses, the effects of temperature-sum and precipitation-sum on top height growth were assessed in the period 1986-2018. A significant upward temporal trend in height growth was observed for both species, with the trend more pronounced after the millennium shift. The magnitude of the annual relative height growth after the millennium was about 16.92% and 9.54% higher than expected, respectively for Scots pine and Norway spruce. A potential climate response on height growth was found for both species with temperature-sum positively correlated with top height growth. No significant effect of precipitation-sum on height growth was observed for either species. Our results suggest improved growing conditions and forest sites became more productive in response to increasing temperature in the northern temperate and boreal regions. The increasing growth trends may offer shorter rotation periods and increased forest value for Norway spruce and Scots pine, coupled with contributions of boreal forests to the emerging bio-economy and the regulation of global atmospheric carbon.

  • Heavy work and disability pension : a long term follow-up of Swedish construction workers

    • OBJECTIVES: The objective of this study was to investigate the occurrence of disability pensions over time among workers with physically demanding jobs.METHODS: The occurrence of disability pension was prospectively studied between 1980-2008 among 325 549 Swedish construction workers. The risks for disability pension and years lost of working life were compared among 22 occupational groups, adjusting for age, body mass index, height, and smoking habits.RESULTS: The risk varied considerably among blue-collar workers. For example, rock workers had double the risk of disability pension [relative risk (RR) 2.16, 95% confidence interval (95% CI) 1.96-2.39] compared to electricians. Most working years lost due to disability pensions (about 75%) were found among men >50 years, mainly due to musculoskeletal and cardiovascular diseases. The years of working life lost due to disability pension varied from 0.7 (salaried employees) to 3.2 years (rock workers) among occupational groups.CONCLUSION: Work environment is an important predictor for disability pension among construction workers with those in physically heavy jobs having the highest burden of disability. If the purpose is to increase labor force participation for workers with heavy jobs, strategies to reduce physical demands at work among elderly workers are important.

    • Ambient energy harvesting has great potential to contribute to sustainable development and address growing environmental challenges. Converting waste energy from energy-intensive processes and systems (e.g. combustion engines and furnaces) is crucial to reducing their environmental impact and achieving net-zero emissions. Compact energy harvesters will also be key to powering the exponentially growing smart devices ecosystem that is part of the Internet of Things, thus enabling futuristic applications that can improve our quality of life (e.g. smart homes, smart cities, smart manufacturing, and smart healthcare). To achieve these goals, innovative materials are needed to efficiently convert ambient energy into electricity through various physical mechanisms, such as the photovoltaic effect, thermoelectricity, piezoelectricity, triboelectricity, and radiofrequency wireless power transfer. By bringing together the perspectives of experts in various types of energy harvesting materials, this Roadmap provides extensive insights into recent advances and present challenges in the field. Additionally, the Roadmap analyses the key performance metrics of these technologies in relation to their ultimate energy conversion limits. Building on these insights, the Roadmap outlines promising directions for future research to fully harness the potential of energy harvesting materials for green energy anytime, anywhere.
