    • The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental studies. Over the past decades, rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and habitats. Yet, in spite of the progress of molecular methods, knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains challenging. In order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis levels. Combining the information from previous efforts such as FUNGuild and Fun(Fun) together with involvement of expert knowledge, we reannotated 10,210 and 151 fungal and Stramenopila genera, respectively. This resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera, designed for rapid functional assignments of environmental studies. In order to assign the trait states to fungal species hypotheses, the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS sequences. On the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1% dissimilarity threshold.

    • Early-life inorganic arsenic exposure influences not only child health and development but also health in later life. The adverse effects of arsenic may be mediated by epigenetic mechanisms, as there are indications that arsenic causes altered DNA methylation of cancer-related genes. The objective was to assess effects of arsenic on genome-wide DNA methylation in newborns. We studied 127 mothers and cord blood of their infants. Arsenic exposure in early and late pregnancy was assessed by concentrations of arsenic metabolites in maternal urine, measured by high performance liquid chromatography-inductively coupled plasma mass spectrometry. Genome-wide 5-methylcytosine methylation in mononuclear cells from cord blood was analyzed by Infinium HumanMethylation450K BeadChip. Urinary arsenic in early gestation was associated with cord blood DNA methylation (Kolmogorov-Smirnov test, P-value<10-15), with more pronounced effects in boys than in girls. In boys, 372 (74%) of the 500 top CpG sites showed lower methylation with increasing arsenic exposure (r S -values>-0.62), but in girls only 207 (41%) showed inverse correlation (r S -values>-0.54). Three CpG sites in boys (cg15255455, cg13659051 and cg17646418), but none in girls, were significantly correlated with arsenic after adjustment for multiple comparisons. The associations between arsenic and DNA methylation were robust in multivariable-adjusted linear regression models. Much weaker associations were observed with arsenic exposure in late compared with early gestation. Pathway analysis showed overrepresentation of affected cancer-related genes in boys, but not in girls. In conclusion, early prenatal arsenic exposure appears to decrease DNA methylation in boys. Associations between early exposure and DNA methylation might reflect interference with de novo DNA methylation.

  • Screening of arsenic in tubewell water with field test kits : Evaluation of the method from public health perspective

    • There is an urgent need for Bangladesh to identify the arsenic (As) contaminated tubewells (TWs) in order to assess the health risks and initiate appropriate mitigation measures. This will involve testing water in millions of TWs and raising community awareness about the health problems related to chronic As exposure from drinking water. Field test kits offer the only practical tool within the time frame and financial resources available for screening and assessment of the As contaminated TWs as well as their monitoring than that of the laboratory measurement. A comparison of field test kit and laboratory measurements by AAS as "gold standard" for As in water of 12,532 TWs in Matlab Upazila in Bangladesh, indicates that the field kit correctly determined the status of 91% of the As levels compared to the Bangladesh Drinking Water Standard (BDWS) of 50 mu g/L, and 87% of the WHO guideline value of 10 mu g/L. Nevertheless, due to analytical and human errors during the deten-nination of As by the field test kits, some misclassification of wells is inevitable. Cross-checking of the field test kit results, both by Field Supervisor and by the laboratory analyses reveal considerable discrepancies in the correct screening mainly at As concentration ranges of 10-24.9 mu g/L and 50-99.9 mu g/L, critical from a public health point of view. The uncertainties of misclassification of these two groups of TWs have severe public health implications due to As exposure from drinking water sources. This can be reduced through proper training of the field personnel, cross verification of the field test kit results with laboratory analyses and further development of the field test kits to determine As at low concentrations.

  • Cadmium exposure in pregnancy and lactation in relation to iron status

    • Objectives. The purpose of this study was to determine the impact of iron status on cadmium dose among pregnant women. Methods. Iron status and cadmium concentration in blood, urine, and placenta were determined among women followed for 2 years from early pregnancy. Results. Blood cadmium and urinary cadmium were correlated with iron status throughout the study period. Urinary cadmium increased longitudinally among women with exhausted iron stores during their pregnancy. The increase in urinary cadmium with age was more pronounced in multiparous than in nulliparous women. Conclusions. Iron deficiency during pregnancy leads to increased cadmium absorption and body burden. Multiparous women exhibit additional increases with increasing age.
