  • Micro- and Nanoscale Surface Analysis of Late Iron Age Glass from Broborg, a Vitrified Swedish Hillfort

    • Archaeological glasses with prolonged exposure to biogeochemical processes in the environment can be used to understand glass alteration, which is important for the safe disposal of vitrified nuclear waste. Samples of mafic and felsic glasses with different chemistries, formed from melting amphibolitic and granitoid rocks, were obtained from Broborg, a Swedish Iron Age hillfort. Glasses were excavated from the top of the hillfort wall and from the wall interior. A detailed microscopic, spectroscopic, and diffraction study of surficial textures and chemistries were conducted on these glasses. Felsic glass chemistry was uniform, with a smooth surface showing limited chemical alteration (<150 nm), irrespective of the position in the wall. Mafic glass was heterogeneous, with pyroxene, spinel, feldspar, and quartz crystals in the glassy matrix. Mafic glass surfaces in contact with topsoil were rougher than those within the wall and had carbon-rich material consistent with microbial colonization. Limited evidence for chemical or physical alteration of mafic glass was found; the thin melt film that coated all exposed surfaces remained intact, despite exposure to hydraulically unsaturated conditions, topsoil, and associated microbiome for over 1,500 years. This supports the assumption that aluminosilicate nuclear waste glasses will have a high chemical durability in near-surface disposal facilities.

  • Flow Alignment of Extracellular Vesicles: Structure and Orientation of Membrane-Associated Bio-macromolecules Studied with Polarized Light

    • Extracellular vesicles (EVs) are currently in scientific focus, as they have great potential to revolutionize the diagnosis and therapy of various diseases. However, numerous aspects of these species are still poorly understood, and thus, additional insight into their molecular-level properties, membrane–protein interactions, and membrane rigidity is still needed. We here demonstrate the use of red-blood-cell-derived EVs (REVs) that polarized light spectroscopy techniques, linear and circular dichroism, can provide molecular-level structural information on these systems. Flow-linear dichroism (flow-LD) measurements show that EVs can be oriented by shear force and indicate that hemoglobin molecules are associated to the lipid bilayer in freshly released REVs. During storage, this interaction ceases; this is coupled to major protein conformational changes relative to the initial state. Further on, the degree of orientation gives insight into vesicle rigidity, which decreases in time parallel to changes in protein conformation. Overall, we propose that both linear dichroism and circular dichroism spectroscopy can provide simple, rapid, yet efficient ways to track changes in the membrane–protein interactions of EV components at the molecular level, which may also give insight into processes occurring during vesiculation.

    • Self-assembling peptides offer a versatile set of tools for bottom-up construction of supramolecular biomaterials. Among these compounds, non-natural peptidic foldamers experience increased focus due to their structural variability and lower sensitivity to enzymatic degradation. However, very little is known about their membrane properties and complex oligomeric assemblies-key areas for biomedical and technological applications. Here we designed short, acyclic β3-peptide sequences with alternating amino acid stereoisomers to obtain non-helical molecules having hydrophilic charged residues on one side, and hydrophobic residues on the other side, with the N-terminus preventing formation of infinite fibrils. Our results indicate that these β-peptides form small oligomers both in water and in lipid bilayers and are stabilized by intermolecular hydrogen bonds. In the presence of model membranes, they either prefer the headgroup regions or they insert between the lipid chains. Molecular dynamics (MD) simulations suggest the formation of two-layered bundles with their side chains facing opposite directions when compared in water and in model membranes. Analysis of the MD calculations showed hydrogen bonds inside each layer, however, not between the layers, indicating a dynamic assembly. Moreover, the aqueous form of these oligomers can host fluorescent probes as well as a hydrophobic molecule similarly to e.g. lipid transfer proteins. For the tested, peptides the mixed chirality pattern resulted in similar assemblies despite sequential differences. Based on this, it is hoped that the presented molecular framework will inspire similar oligomers with diverse functionality.

    • Self-assembled peptide nanostructures with stimuli-responsive features are promising as functional materials. Despite extensive research efforts, water-soluble supramolecular constructs that can interact with lipid membranes in a controllable way are still challenging to achieve. Here, we have employed a short membrane anchor protein motif (GLFD) and coupled it to a spiropyran photoswitch. Under physiological conditions, these conjugates assemble into ∼3.5 nm thick, foil-like peptide bilayer morphologies. Photoisomerization from the closed spiro (SP) form to the open merocyanine (MC) form of the photoswitch triggers rearrangements within the foils. This results in substantial changes in their membrane-binding properties, which also varies sensitively to lipid composition, ranging from reversible nanofoil reformation to stepwise membrane adsorption. The formed peptide layers in the assembly are also able to attach to various liposomes with different surface charges, enabling the fusion of their lipid bilayers. Here, SP-to-MC conversion can be used both to trigger and to modulate the liposome fusion efficiency.

  • Data-driven distance metrics for kriging - Short-term urban traffic state prediction

    • Estimating traffic flow states at unmeasured urban locations provides a cost-efficient solution for many ITS applications. In this work, a geostatistical framework, kriging is extended in such a way that it can both estimate and predict traffic volume and speed at various unobserved locations, in real-time. In the paper, different distance metrics for kriging are evaluated. Then, a new, data-driven one is formulated, capturing the similarity of measurement sites. Then, with multidimensional scaling the distances are transformed into a hyperspace, where the kriging algorithm can be used. As a next step, temporal dependency is injected into the estimator via extending the hyperspace with an extra dimension, enabling for short horizon traffic flow prediction. Additionally, a temporal correction is proposed to compensate for minor changes in traffic flow patterns. Numerical results suggest that the spatio-temporal prediction can make more accurate predictions compared to other distance metric-based kriging algorithms. Additionally, compared to deep learning, the results are on par while the algorithm is more resilient against traffic pattern changes.

  • Energy-aware predictive control for electrified bus networks

    • For an urban bus network to operate efficiently, conflicting objectives have to be considered: providing sufficient service quality while keeping energy consumption low. The paper focuses on energy efficient operation of bus lines, where bus stops are densely placed, and buses operate frequently with possibility of bunching. The proposed decentralized, bus  fleet control solution aims to combine four conflicting goals incorporated into a multi-objective, nonlinear cost function. The multi-objective optimization is solved under a receding horizon model predictive framework. The four conflicting objectives are as follows. One is ensuring periodicity of headways by watching leading and following vehicles i.e. eliminating bus bunching. Equal headways are only a necessary condition for keeping a static, predefifined, periodic timetable. The second objective is timetable tracking, and passenger waiting time minimization. In case of high passenger demand, it is desirable to haste the bus in order to prevent bunching. The final objective is energy efficiency. To this end, an energy consumption model is formulated considering battery electric vehicles with recuperation during braking. Alternative weighting strategies are compared and evaluated through realistic scenarios, in a calibrated microscopic traffic simulation environment. Simulation results confirm of 3-8% network level energy saving compared to bus holding control while maintaining punctuality and periodicity of buses.

  • Network-Level Optimal Control for Public Bus Operation

    • The paper presents modeling, control and analysis of an urban public transport network. First, a centralized system description is given, built up from the dynamics of individual buses and bus stops. Aiming to minimize three conflicting goals (equidistant headways, timetable adherence, and minimizing passenger waiting times), a reference tracking model predictive controller formulated based on the piecewise-affine system model. The closed-loop system is analyzed with three methods. Numerical simulations on a simple experimental network showed that the temporal evolution of headways and passenger numbers could maintain their periodicity with the help of velocity control. With the help of randomized simulation scenarios, sensitivity of the system is analyzed. Finally, infeasible regions for the bus network control was sought using by formulating an explicit model predictive controller.

    • This paper presents a model-based multiobjective control strategy to reduce bus bunching and hence improve public transport reliability. Our goal is twofold. First, we define a proper model, consisting of multiple static and dynamic components. Bus-following model captures the longitudinal dynamics taking into account the interaction with the surrounding traffic. Furthermore, bus stop operations are modeled to estimate dwell time. Second, a shrinking horizon model predictive controller (MPC) is proposed for solving bus bunching problems.The model is able to predict short time-space behavior of public transport buses enabling constrained, finite horizon, optimal control solution to ensure homogeneity of service both in time and space. In this line, the goal with the selected rolling horizon control scheme is to choose a proper velocity profile for the public transport bus such that it keeps both timetable schedule and a desired headway from the bus in front of it (leading bus). The control strategy predicts the arrival time at a bus stop using a passenger arrival and dwell time model. In this vein, the receding horizon model predictive controller calculates an optimal velocity profile based on its current position and desired arrival time. Three different weighting strategies are proposed to test (i) timetable only, (ii) headway only or (iii) balanced timetable - headway tracking. The controller is tested in a high fidelity traffic simulator with realistic scenarios. The behavior of the system is analyzed by considering extreme disturbances. Finally, the existence of a Pareto front between these two objectives is also demonstrated.

  • Optimal headway merging for balanced public transport service in urban networks

    • This paper presents a velocity control/advise algorithm relying on vehicle-to-vehicle communication, to ensure the headway homogeneity of buses on a joint corridor, i.e. when multiple lines merge and travel on the same route. The proposed control method first schedules merging buses prior to entering a common line. Second, based on the position and velocity of the bus ahead of the controlled one, a shrinking horizon model predictive controller (MPC) calculates a proper velocity profile for the merging bus. The model is able to predict short time- space behavior of public transport buses enabling constrained, finite horizon, optimal control solution to reach the merging point with equidistant headways, taking all buses from different lines into account. The controller is tested in a high fidelity traffic simulator with realistic scenarios.

    • Models for long‐term glass alteration are required to satisfy performance predictions of vitrified nuclear waste in various disposal scenarios. Durability parameters are usually extracted from short‐term laboratory tests, and sometimes checked with long‐term natural experiments on glasses, termed analogues. In this paper, a unique potential ancient glass analogue from Sweden is discussed. The hillfort glass found at Broborg represents a unique case study as a vitrified waste glass analogue to compare to Low Activity Waste glass to be emplaced in near surface conditions at Hanford (Washington State). Glasses at Broborg have similar and dissimilar compositions to LAW glass, allowing the testing of long‐term alteration of different glass chemistries. Additionally, the environmental history of the site is reasonably well documented. Initial investigations on previously collected samples established methodologies for handling and characterizing these artifacts by laboratory methods while preserving their alteration layers and cultural context. Evidence of possible biologically influenced glass alteration, and differential alteration in the two types of glass found at the Broborg site is presented.
