    • EUSO-Balloon is a pathfinder mission for the Extreme Universe Space Observatory onboard the Japanese Experiment Module (JEM-EUSO). It was launched on the moonless night of the 25(th) of August 2014 from Timmins, Canada. The flight ended successfully after maintaining the target altitude of 38 km for five hours. One part of the mission was a 2.5 hour underflight using a helicopter equipped with three UV light sources (LED, xenon flasher and laser) to perform an inflight calibration and examine the detectors capability to measure tracks moving at the speed of light. We describe the helicopter laser system and details of the underflight as well as how the laser tracks were recorded and found in the data. These are the first recorded laser tracks measured from a fluorescence detector looking down on the atmosphere. Finally, we present a first reconstruction of the direction of the laser tracks relative to the detector.

  • Benchmark DEBORA: Assessment of MCFD compared to high-pressure boiling pipe flow measurements

    • A benchmark activity on two-fluid simulations of high-pressure boiling upward flows in a pipe is performed by 12 participants using different MCFD (Multiphase Computational Fluid Dynamics) codes and closure relationships. More than 30 conditions from DEBORA experiment conducted by CEA are considered. Each case is characterised by the flow rate, inlet temperature, wall heat flux and outlet pressure. High-pressure Freon (R12) at 14 bar and 26 bar is boiled in a 19.2mm pipe heated over 3.5m. Flow rates range from 2000 kg m−2 s−1 to 5000 kg m−2 s−1 and exit quality x ranges from single-phase conditions to x=0.1 which leads to a peak void fraction of α=70%. In these high pressure conditions, bubbles remain small and there is no departure from the bubbly flow regime (François et al., 2011; Hösler, 1968). However, different kind of bubbly flows are observed: wall-peak, intermediate peak or core-peak, depending on the case considered. Measurements along the pipe radius near the end of the heated section are compared to code predictions. They include void fraction, bubble mean diameter, vapour velocity and liquid temperature. The benchmark covered two phases. In the first phase of the benchmark activities, experimental data were given to the participants, allowing to compare the simulation results and to develop, to select or to adjust the models in the CMFD codes. The second phase included blind cases where the participants could not compare to the measurements. In between the two phases, possible additional model adjustments or calibrations were performed. Overall, the benchmark involved very different closures and a wide range of models’ complexity was covered. Yet, it is extremely difficult to have a robust closure for all conditions considered, even knowing experimental measurements. The wall-to-core peak transition is not captured consistently by the models. The degree of subcooling and the void fraction level are also difficult to assess. We were not capable of showing superiority of some physical closures, even for part of the model. The interaction between mechanisms and their hierarchy are extremely difficult to understand. Although departure from nucleate boiling (DNB) was not considered in this benchmarking exercise, it is expected that DNB predictions at high-pressure conditions depend strongly on the near-wall flow, temperature, and void fraction distributions. Therefore, the suitability of the closures also limits the accuracy of DNB predictions. The benchmark also demonstrated that in order to progress further in models development and validation, it is compulsory to have new measurements that include simultaneously as many variables as possible (including liquid temperature, velocity, cross-correlations and wall temperature); also, a better knowledge of the local bubble sizes distributions is the key to discriminate performances of interfacial area modelling (IATE, MUSIG or iMUSIG models, considering for instance the possibility of two classes of bubbles having totally different behaviour regarding the lift force). Following this benchmark impulse, we hope that future activities will be engaged on high-pressure boiling water experiments with a continuation of models’ comparisons and development.

  • Patients intention to consume prescribed and non-prescribed medicines: A study based on the theory of planned behaviour in selected European countries

    • What is known and objectivePolypharmacy has a significant impact on patients health with overall expenditure on over-the-counter (OTC) medicines representing a substantial burden in terms of cost of treatment. The aim of this study, which was conducted within the framework of a European Project funded by the European Union under the Seventh Framework Programme and was entitled OTC-SOCIOMED, was to report on possible determinants of patient behaviour regarding the consumption of medicines, and particularly OTCs, in the context of primary care. MethodsA multicentre, cross-sectional study was designed and implemented in well-defined primary healthcare settings in Cyprus, the Czech Republic, France, Greece, Malta and Turkey. Patients completed a questionnaire constructed on the basis of the theory of planned behaviour (TPB), which was administered via face-to-face interviews. Results and discussionThe percentage of patients who had consumed prescribed medicines over a 6-month period was consistently high, ranging from 79% in the Czech Republic and 82% in Turkey to 97% in Malta and 100% in Cyprus. Reported non-prescribed medicine consumption ranged from 33% in Turkey to 92% in the Czech Republic and 97% in Cyprus. TPB behavioural antecedents explained 43% of the variability of patients intention to consume medicines in Malta and 24% in Greece, but only 3% in Turkey. Subjective norm was a significant predictor of the intention to consume medicines in all three countries (Greece, Malta and Turkey), whereas attitude towards consumption was a significant predictor of the expectation to consume medicines, if needed. What is new and conclusionThis study shows that parameters such as patients beliefs and influence from family and friends could be determining factors in explaining the high rates of medicine consumption. Factors that affect patients behavioural intention towards medicine consumption may assist in the formulation of evidence-based policy proposals and inform initiatives and interventions aimed at increasing the appropriate use of medicines.

  • Tryptophan-accelerated electron flow through proteins

    • Energy flow in biological structures often requires submillisecond charge transport over long molecular distances. Kinetics modeling suggests that charge-transfer rates can be greatly enhanced by multistep electron tunneling in which redox-active amino acid side chains act as intermediate donors or acceptors. We report transient optical and infrared spectroscopic experiments that quantify the extent to which an intervening tryptophan residue can facilitate electron transfer between distant metal redox centers in a mutant Pseudomonas aeruginosa azurin. CuI oxidation by a photoexcited ReI-diimine at position 124 on a histidine(124)-glycine(123)-tryptophan(122)-methionine(121) β strand occurs in a few nanoseconds, fully two orders of magnitude faster than documented for single-step electron tunneling at a 19 angstrom donor-acceptor distance.

    • Discoveries of diverse microbial eukaryotes and their inclusion in comprehensive phylogenomic analyses have crucially re-shaped the eukaryotic tree of life in the 21st century.(1) At the deepest level, eukaryotic diversity comprises 9-10 "supergroups." One of these supergroups, the Metamonada, is particularly important to our understanding of the evolutionary dynamics of eukaryotic cells, including the remodeling of mitochondrial function. All metamonads thrive in low-oxygen environments and lack classical aerobic mitochondria, instead possessing mitochondrion-related organelles (MROs) with metabolisms that are adapted to low-oxygen conditions. These MROs lack an organellar genome, do not participate in the Krebs cycle and oxidative phosphorylation,(2) and often synthesize ATP by substrate-level phosphorylation coupled to hydrogen production.(3,4) The events that occurred during the transition from an oxygen-respiring mitochondrion to a functionally streamlined MRO early in metamonad evolution remain largely unknown. Here, we report transcriptomes of two recently described, enigmatic, anaerobic protists from the genus Anaeramoeba.(5) Using phylogenomic analysis, we show that these species represent a divergent, phylum-level lineage in the tree of metamonads, emerging as a sister group of the Parabasalia and reordering the deep branching order of the metamonad tree. Metabolic reconstructions of the Anaeramoeba MROs reveal many "classical" mitochondrial features previously not seen in metamonads, including a disulfide relay import system, propionate production, and amino acid metabolism. Our findings suggest that the cenancestor of Metamonada likely had MROs with more classical mitochondrial features than previously anticipated and demonstrate how discoveries of novel lineages of high taxonomic rank continue to transform our understanding of early eukaryote evolution.

  • Midbrain Gene Screening Identifies a New Mesoaccumbal Glutamatergic Pathway and a Marker for Dopamine Cells Neuroprotected in Parkinson's Disease

    • The ventral tegmental area (VTA) and substantia nigra pars compacta (SNc) of the midbrain are associated with Parkinson's disease (PD), schizophrenia, mood disorders and addiction. Based on the recently unraveled heterogeneity within the VTA and SNc, where glutamate, GABA and co-releasing neurons have been found to co-exist with the classical dopamine neurons, there is a compelling need for identification of gene expression patterns that represent this heterogeneity and that are of value for development of human therapies. Here, several unique gene expression patterns were identified in the mouse midbrain of which NeuroD6 and Grp were expressed within different dopaminergic subpopulations of the VTA, and TrpV1 within a small heterogeneous population. Optogenetics-coupled in vivo amperometry revealed a previously unknown glutamatergic mesoaccumbal pathway characterized by TrpV1-Cre-expression. Human GRP was strongly detected in non-melanized dopaminergic neurons within the SNc of both control and PD brains, suggesting GRP as a marker for neuroprotected neurons in PD. This study thus unravels markers for distinct subpopulations of neurons within the mouse and human midbrain, defines unique anatomical subregions within the VTA and exposes an entirely new glutamatergic pathway. Finally, both TRPV1 and GRP are implied in midbrain physiology of importance to neurological and neuropsychiatric disorders.
