    • The Horizon 2020 Project EuPRAXIA ("European Plasma Research Accelerator with eXcellence In Applications") is preparing a conceptual design report of a highly compact and cost-effective European facility with multi-GeV electron beams using plasma as the acceleration medium. The accelerator facility will be based on a laser and/or a beam driven plasma acceleration approach and will be used for photon science, high-energy physics (HEP) detector tests, and other applications such as compact X-ray sources for medical imaging or material processing. EuPRAXIA started in November 2015 and will deliver the design report in October 2019. EuPRAXIA aims to be included on the ESFRI roadmap in 2020.

  • Analysis of laser wakefield dynamics in capillary tubes

    • A general approach to the modifications of the spectrum of a laser pulse interacting with matter is elaborated and used for spectral diagnostics of laser wakefield generation in guiding structures. Analytical predictions of the laser frequency red shift due to the wakefield excited in a capillary waveguide are confirmed by self-consistent modeling results. The role of ionization blue shift, and nonlinear laser pulse and wakefield dynamics on the spectrum modification, is analyzed for recent experiments on plasma wave excitation by an intense laser pulse guided in hydrogen-filled glass capillary tubes up to 8 cm long. The dependence of the spectral frequency shift, measured as a function of filling pressure, capillary tube length and incident laser energy, is in excellent agreement with the simulation results, and the associated longitudinal accelerating field is in the range 1-10 GV m(-1).

  • Inhibition of intestinal bile acid absorption improves cholestatic liver and bile duct injury in a mouse model of sclerosing cholangitis

    • Background and Aims: Approximately 95% of bile acids (BAs) excreted into bile are reabsorbed in the gut and circulate back to the liver for further biliary secretion. Therefore, pharmacological inhibition of the ileal apical sodium-dependent BA transporter (ASBT/SLC10A2) may protect against BA-mediated cholestatic liver and bile duct injury. Methods: Eight week old Mdr2(-/-) (Abcb4(-/-)) mice (model of cholestatic liver injury and sclerosing cholangitis) received either a diet supplemented with A4250 (0.01% w/w) - a highly potent and selective ASBT inhibitor - or a chow diet. Liver injury was assessed biochemically and histologically after 4 weeks of A4250 treatment. Expression profiles of genes involved in BA homeostasis, inflammation and fibrosis were assessed via RT-PCR from liver and ileum homogenates. Intestinal inflammation was assessed by RNA expression profiling and immunohistochemistry. Bile flow and composition, as well as biliary and fecal BA profiles were analyzed after 1 week of ASBT inhibitor feeding. Results: A4250 improved sclerosing cholangitis in Mdr2(-/-) mice and significantly reduced serum alanine aminotransferase, alkaline phosphatase and BAs levels, hepatic expression of proinflammatory (Tnf-alpha, Vcam1, Mcp-1) and pro-fibrogenic (Col1a1, Col1a2) genes and bile duct proliferation (mRNA and immunohistochemistry for cytokeratin 19 (CK19)). Furthermore, A4250 significantly reduced bile flow and biliary BA output, which correlated with reduced Bsep transcription, while Ntcp and Cyp7a1 were induced. Importantly A4250 significantly reduced biliary BA secretion but preserved HCO3- and biliary phospholipid secretion resulting in an increased HCO3-/BA and PL/BA ratio. In addition, A4250 profoundly increased fecal BA excretion without causing diarrhea and altered BA pool composition, resulting in diminished concentrations of primary BAs tauro-beta-muricholic acid and taurocholic acid. Conclusions: Pharmacological ASBT inhibition attenuates cholestatic liver and bile duct injury by reducing biliary BA concentrations in mice. (C) 2015 European Association for the Study of the Liver.     • In situ Na intercalation of the layered compound VSe2 has been studied with photoemission and scanning tunneling microscopy. Core-level spectroscopy proves that Na deposited in UHV onto the VSe2 surface rapidly intercalates, leaving only small amounts at the surface. The scanning tunneling microscopy measurements show that the intercalated Na is not uniformly distributed between the VSe2 layers, but preferentially in two-dimensional islands. Thus the surface region is divided into intercalated and nonintercalated areas. Hole-like features in the intercalated areas are interpreted as locally missing Na.

  • FXR activation protects against NAFLD via bile-acid-dependent reductions in lipid absorption

    • FXR agonists are used to treat non-alcoholic fatty liver disease (NAFLD), in part because they reduce hepatic lipids. Here, we show that FXR activation with the FXR agonist GSK2324 controls hepatic lipids via reduced absorption and selective decreases in fatty acid synthesis. Using comprehensive lipidomic analyses, we show that FXR activation in mice or humans specifically reduces hepatic levels of mono-and polyunsaturated fatty acids (MUFA and PUFA). Decreases in MUFA are due to FXR-dependent repression of Scd1, Dgat2, and Lpin1 expression, which is independent of SHP and SREBP1c. FXR-dependent decreases in PUFAs are mediated by decreases in lipid absorption. Replenishing bile acids in the diet prevented decreased lipid absorption in GSK2324-treated mice, suggesting that FXR reduces absorption via decreased bile acids. We used tissue-specific FXR KO mice to show that hepatic FXR controls lipogenic genes, whereas intestinal FXR controls lipid absorption. Together, our studies establish two distinct pathways by which FXR regulates hepatic lipids.

    • The photoion excitation spectra of (NO)-N-14 and (NO)-N-15 have been studied in the 105-112 Mn excitation region using high-order harmonic laser spectroscopy. Utilizing the small bandwidth offered by this technique, the natural widths of prominent lines in this region have been measured. These lines originate from interactions between high-lying Rydberg levels converging to the NO+ electronic ground state and a 'new' NO valence state situated close to the latter state. The present measurements show broad resonances corresponding to a decay time of 20-50 fs. This supports the interpretation that the decay takes place via rapid electronic autoionization.

    • We report preliminary experimental and computational studies of the temporal evolution of the harmonics, generated in xenon, by 50-ps pulses from a Nd: YLF laser, amplified in Nd: glass amplifiers, and focused to produce intensities in the range 1013--5 {\texttimes} 1013 W cm{\textminus}2. Measurements were made of the temporal profiles of the seventh- and ninth-order harmonics using a specially modified VUV streak camera with better than 7-ps temporal resolution. These results were compared with predictions of a numerical model that included the effects of neutral atom depletion and photoelectron dispersion in the calculation of the harmonic output as a function of time.

  • Analysis of x-ray emission and electron dynamics in a capillary-guided laser wakefield accelerator

    • The dynamics of electron acceleration driven by laser wakefield inside a 30.5 mm long dielectric capillary tube is analyzed using radiation emitted in the x-ray range. 3D particle-in-cell simulations, performed with parameters close to the experimental ones, show that in long plasmas, the accelerated electrons catch up and finally overrun the driving laser owing to a higher velocity of the electrons in the plasma. The electrons are then transversely scattered by the laser pulse, and penetrate the capillary wall where they generate bremsstrahlung radiation, modeled using geant4 simulations. The signature of bremsstrahlung radiation is detected using an x-ray camera, together with the betatron radiation emitted during electron acceleration in the plasma bubble. The reflection of betatron radiation from the inner capillary surface also accounts for a fraction of the observed signal on the x-ray camera. The simulation results are in agreement with the experimental ones and provide a detailed description of the electron and radiation properties, useful for the design of laser wakefield accelerators or radiation sources using long plasma media.

  • Adverse events in apheresis : an update of the WAA registry data

    • Apheresis with different procedures and devices are used for a variety of indications that may have different adverse events (AEs). The aim of this study was to clarify the extent and possible reasons of various side effects based on data from a multinational registry. The WAA-apheresis registry data focus on adverse events in a total of 50846 procedures in 7142 patients (42% women). AEs were graded as mild, moderate (need for medication), severe (interruption due to the AE) or death (due to AE). More AEs occurred during the first procedures versus subsequent (8.4 and 5.5%, respectively). AEs were mild in 2.4% (due to access 54%, device 7%, hypotension 15%, tingling 8%), moderate in 3% (tingling 58%, urticaria 15%, hypotension 10%, nausea 3%), and severe in 0.4% of procedures (syncope/hypotension 32%, urticaria 17%, chills/fever 8%, arrhythmia/asystole 4.5%, nausea/vomiting 4%). Hypotension was most common if albumin was used as the replacement fluid, and urticaria when plasma was used. Arrhythmia occurred to similar extents when using plasma or albumin as replacement. In 64% of procedures with bronchospasm, plasma was part of the replacement fluid used. Severe AEs are rare. Although most reactions are mild and moderate, several side effects may be critical for the patient. We present side effects in relation to the procedures and suggest that safety is increased by regular vital sign measurements, cardiac monitoring and by having emergency equipment nearby.

    • The excitation of plasma waves over a length of up to 8 cm is demonstrated using laser guiding of intense laser pulses through hydrogen-filled glass capillary tubes. The plasma waves are diagnosed by spectral analysis of the transmitted laser radiation. The dependence of the spectral redshift-measured as a function of filling pressure, capillary tube length, and incident laser energy-is in excellent agreement with simulation results. The longitudinal accelerating field inferred from the simulations is in the range of 1-10 GV/m.

  • Influence of atomic density in high-order harmonic generation

    • We have investigated how high-order harmonics generated in rare gases depend on the atomic density. The peak and the profile of the atomic density in the interaction region were measured as a function of the backing pressure and the distance from the nozzle by a differential interferometry technique, The conversion efficiency for the harmonics in the plateau was found to increase approximately quadratically over the entire range of peak pressures investigated (3-80 mbar). The intensity of the harmonics in the cutoff region, in contrast, increased only until an optimum peak pressure was reached, beyond which it decreased. This optimum peak pressure was found to be dependent on both the laser intensity and the process order. To understand this effect, we have performed extensive propagation calculations of both the fundamental and the harmonic fields, using ionization rates and dipole moments from a tunnel ionization model. We obtained good agreement with the experimental results. The observed effect is attributed to ionization-induced defocusing of the fundamental laser beam, which reduces the peak intensity obtained in the medium and shortens the extent of the plateau.   • The measurement of venous haematocrit in patients with polycythaemia vera.

    • OBJECTIVE: In clinical practice, patients with polycythaemia vera (PV) are monitored by measurement of venous packed cell volume (PCV). However, whereas treatment recommendations are still based upon studies in which the results were obtained with the centrifuged microhaematocrit, currently in most instances automated blood cell counters are used to calculate PCV. In a group of patients with polycythaemia we therefore compared the results obtained by the microhaematocrit method with PCV calculated by haematology analysers. DESIGN: The study was carried out on a prospective basis. Duplicate venous blood samples were collected. The centrifuged microhaemotocrit was obtained by using an IEC Micro-MB Centrifuge. Depending on different routine methods used in the participating hospitals, the blood cell counter PCV was calculated using Coulter STKS, Bayer Technicon H2 or H3. SETTING: Patients were included from four Swedish university hospitals: Akademiska (Uppsala), Huddinge and Karolinska (Stockholm) and Sahlgrenska (Göteborg). SUBJECTS: Seventy-four patients with PV and 10 patients with secondary polycythaemia were included and a total of 150 duplicate blood samples were analysed from these subjects. RESULTS: In the 150 measurements the mean blood cell counter calculated PCV was 0.448 +/- 0.037; the mean for centrifuged microhaematocrit was 0.467 +/- 0. 037 and the difference between means was highly significant (P = 6.8 x 10-25). The means for centrifuged haematocrit and calculated PCV differed significantly in the groups of PV patients treated with phlebotomy only, hydroxyurea or radiophosphorous (P < 0.0001, respectively). In PV patients treated with alpha-interferon and in patients with secondary polycythaemia the difference in means did not reach statistical significance (P = 0.07 and P = 0.13, respectively). The groups of patients with MCV <80 fL and >/=80 fL both presented significant differences between means for calculated PCV and centrifuged haematocrit. CONCLUSIONS: If PV patients are monitored with blood cell counter calculated PCV it appears that the therapeutic goal should be to maintain the calculated PCV below 0.43, provided the local differences in calculated PCV and centrifuged haematocrit are of the same magnitude as in this study.

  • Learning from dialogue groups--physicians' perceptions of role

    • The purpose of this paper is to examine the aspects of being a physician that such medical professionals mention in dialogue groups when given the opportunity to choose their own topics of discussion.Design/methodology/approachOver a period of two years, 60 physicians participated in eight dialogue groups at one of the main hospitals in Stockholm, Sweden. Five focus group interviews were performed after the final dialogue group session.FindingsQualitative content analysis showed that three themes dominated in the physicians' perceptions of their role: hierarchy and subgroups; understanding of learning and knowledge; clinical work. Very little time in the dialogue groups was spent discussing the third theme, i.e. problems or issues related to patients or their families. The hierarchy among doctors seemed to influence many aspects of the role of these individuals, their healthcare organisation and their work environment. The methodology in the dialogue groups challenged the prevailing hierarchical structures and seemed to improve the relations between different groups of doctors in the hierarchy. For some of the physicians, this also resulted in a new way of perceiving and acting in their professional role.Research limitations/implicationsThe results of this study represent only one hospital.Practical implicationsThe findings may help healthcare managers understand physicians' conceptions of their role.Originality/valueFew intervention studies have considered management programmes directed towards physicians. The present investigation is the first qualitative analysis of the use of dialogue groups within a healthcare setting.

  • Structural characterization of precious-mean quasiperiodic Mo/V single-crystal superlattices grown by dual-target magnetron sputtering

    • A class of quasiperiodic superlattice structures, which can be generated by the concurrent inflation rule A→AmB and B→A (where m=positive integer), has been studied both theoretically and experimentally. Given that the ratios between the thicknesses of the two superlattice building blocks, A and B, are chosen to be γ(m)=[m+(m2+4)1/2]/2 (known as the ‘‘precious means’’), then the x-ray- and electron-diffraction peak positions are analytically found to be located at the wave vectors q=2πΛ−1r[γ(m)]k, where r and k are integers and Λ is an average superlattice wavelength. The analytically obtained results have been compared to experimental results from single-crystalline Mo/V superlattice structures, generated with m=1, 2, and 3. The superlattices were grown by dual-target dc-magnetron sputtering on MgO(001) substrates kept at 700 °C. X-ray diffraction (XRD) and selected-area electron diffraction (SAED) showed that the analytical model mentioned above predicts the peak positions of the experimental XRD and SAED spectra with a very high accuracy. Furthermore, numerical calculations of the diffraction intensities based on a kinematical model of diffraction showed good agreement with the experimental data for all three cases. In addition to a direct verification of the quasiperiodic modulation, both conventional and high-resolution cross-sectional transmission electron microscopy (XTEM) showed that the superlattices are of high crystalline quality with sharp interfaces. Based on lattice resolution images, the width of the interfaces was determined to be less than two (002) lattice-plane spacings (≊0.31 nm).

    • Natural radiative lifetimes have been measured along the 6sns S-3(1) (n=7-10) and 6snd D-3(1) (n=6-11) Rydberg series of neutral mercury using time-resolved laser-induced fluorescence spectroscopy in a laser-produced mercury plasma. The states of Hg I investigated in the present work were populated from the ground state by a stepwise excitation process. Relativistic Hartree-Fock calculations, including intravalence interactions and core-polarization effects, have been performed and the theoretical lifetimes have been compared with the measurements.

    • The time-dependent soft X-ray emission of helium and nitrogen plasmas generated by optical-field ionization is reported. The experiments were carried out by focusing pulses of the high-power Ti:sapphire laser of the Lund Institute of Technology (lambda = 796 nm, pulse duration 150 fs, pulse energy 150 mJ) to a 50-mu m diameter spot close to a nozzle, using He and N-2 as target gases. The emission on He+, N4+, and N3+ resonance lines was recorded by means of a flat-field grating spectrometer coupled to an X-ray streak camera. A pronounced difference in the temporal shape of the emission of the Lyman-alpha line of hydrogen-like helium and of the 2p-3d resonance lines of lithium-like and beryllium-like nitrogen was observed. The helium line exhibited an initial spike followed by a slow revival of the emission, whereas the nitrogen lines showed a slow decay after a fast initial rise. These observations are explained with the help of simulations.

    • The magnetic and transport properties of Ni81Fe 19/Al2O3 granular multilayer films were studied in relation to their structural properties as the nominal thickness t of the permalloy (Ni81Fe19) layer was varied near the percolation limit: in the range of 8≤t≤ 16 Å while keeping the nominal thickness of the Al2O3 layers constant at 16 Å. A good structural quality of the multilayers was demonstrated by low angle x-ray reflectivity measurements, and transmission electron microscopy showed the transition from continuous permalloy layers separated by aluminium oxide layers for t= 16 Å to metal grains dispersed in the insulator at t=8 Å. Magnetization measurements showed the gradual transition from ferromagnetic layers to superparamagnetic clusters and grains that successively become blocked as the temperature decreases. A strong correlation between transport and structural properties was observed in the temperature (T) dependence of the electrical resistance measured with the current in the plane in the range of 2 ≤T≤300 K: a gradual change of behavior from continuous permalloy layers with conducting interlayer connections for t=16 Å. to isolated permalloy grains in a dielectric for the film with t= 10 Å. The percolation occurs between 12 and 10 Å, as deduced both from the magnetic and resistive properties. The discontinuous metal films were analyzed within models for thermally assisted tunneling, yielding estimates of the tunneling barrier for intralayer conduction of about 20 meV for t= 10 Å. A significant magnetic field dependence of the resistance increasing with decreasing temperature was observed in all samples.

    • Bile salt export pump (Bsep) (Abcb11)(-/-) mice are protected from acquired cholestatic injury due to metabolic preconditioning with a hydrophilic bile acid (BA) pool with formation of tetrahydroxylated bile acids (THBAs). We aimed to explore whether loss of Bsep and subsequent elevation of THBA levels may have immunomodulatory effects, thus improving liver injury in the multidrug resistance protein 2 (Mdr2) (Abcb4)(-/-) mouse. Cholestatic liver injury in Mdr2(-/-)Bsep(-/-) double knockout (DKO), Mdr2(-/-), Bsep(-/-), and wild-type mice was studied for comparison. Mdr2(-/-) mice were treated with a THBA (3 alpha,6 alpha,7 alpha,12 alpha-Tetrahydroxycholanoic acid). RNA/protein expression of inflammatory/fibrotic markers were investigated. Serum BA-profiling was assessed by ultra-performance liquid chromatography tandem mass spectrometry. Hepatic immune cell profile was quantified by flow cytometric analysis (FACS). In vitro, the THBA effect on chenodeoxycholic acid (CDCA)-induced inflammatory signaling in hepatocyte and cholangiocytes as well as lipopolysaccharide (LPS)/interferon-gamma (IFN-gamma)-induced macrophage activation was analyzed. In contrast to Mdr2(-/-), DKO mice showed no features of sclerosing cholangitis. Sixty-seven percent of serum BAs in DKO mice were polyhydroxylated (mostly THBAs), whereas Mdr2(-/-) mice did not have these BAs. Compared with Mdr2(-/-), DKO animals were protected from hepatic inflammation/fibrosis. THBA feeding in Mdr2(-/-) mice improved liver injury. FACS analysis in DKO and Mdr2(-/-) THBA-fed mice showed changes of the hepatic immune cell profile towards an anti-inflammatory pattern. Early growth response 1 (EGR1) protein expression was reduced in DKO and in Mdr2(-/-) THBA-fed mice compared with Mdr2(-/-) control mice. In vitro, THBA-reduced CDCA induced EGR1 protein and mRNA expression of inflammatory markers in hepatocytes and cholangiocytes. LPS/IFN-gamma-induced macrophage activation was ameliorated by THBA. THBAs repress EGR1-related key pro-inflammatory pathways. Conclusion: THBA and their downstream targets may represent a potential treatment strategy for cholestatic liver diseases.

    • Electron beams and betatron X-ray radiation generated by laser wakefield acceleration in long plasma targets are studied. The targets consist of hydrogen filled dielectric capillary tubes of diameter 150 to 200 microns and length 6 to 20 mm. Electron beams are observed for peak laser intensities as low as 5x10(17) W/cm(2). It is found that the capillary collects energy outside the main peak of the focal spot and contributes to keep the beam self-focused over a distance longer than in a gas jet of similar density. This enables the pulse to evolve enough to reach the threshold for wavebreaking, and thus trap and accelerate electrons. No electrons were observed for capillaries of large diameter (250 mu m), confirming that the capillary influences the interaction and does not have the same behaviour as a gas cell. Finally, X-rays are used as a diagnostic of the interaction and, in particular, to estimate the position of the electrons trapping point inside the capillary.

    • Two Swedish families with Huntington's disease (HD) have been investigated for linkage with G8 (D4S10). In one family from northern Sweden (Family 1) 48 family members were examined, and in another family from the southwestern part of Sweden (Family 2) 14 family members were examined. The lod scores were 1.531 for Family 1 and 2.057 for Family 2, and the combined lod score was 3.59. The HD gene was segregating with the haplotype C in Family 1 and with haplotype A in Family 2. The predictive value of the test was obvious. Before the testing with the G8 probe, 84.2% of the family members in Family 1 had a theoretical risk of 25% or 50% of having the HD gene. After the testing with the G8 probe, only 23.7% of the family members remained at the same risk, and it could also be certified that 63.2% had no or little risk of having the HD gene. Only one asymptomatic person was predicted to have HD.

    • Prompted by an increased interest of both research participants and the patient advocacy community in obtaining information about research outcomes and on the use of their biological samples; the international community has begun to debate the emergence of an ethical 'duty' to return research results to participants. Furthermore, the use of new technologies (e.g., whole-genome and -exome sequencing) has revealed both genetic data and incidental findings with possible clinical significance. These technologies together with the proliferation of biorepositories, provide a compelling rationale for governments and scientific institutions to adopt prospective policies. Given the scarcity of policies in the context of stem cell research, a discussion on the scientific, ethical and legal implications of disclosing research results for research participants is needed. We present the International Stem Forum Ethics Working Party's Policy Statement and trust that it will stimulate debate and meet the concerns of researchers and research participants alike.

    • A pump-probe technique has been applied for measuring the lifetimes and absolute photoionization cross-sections of excited He states. The 1s2p P-1 and 1s3p P-1 states of He are excited by using the 13th and the 14th harmonic, respectively, of a tunable 70 ps dye laser generated in a Kr gas jet. The states are ionized after a varying time delay, by absorption of probe photons with energies between 1.6 and 4.5 eV. Lifetimes of tau(1s2p) = 0.57 ns and tau(1s3p) = 1.76 ns are determined with a precision of about 15%. A significant enhancement of the number of ions present in the lifetime curves at zero time delay for pressures above 6 x 10(-5) mbar is attributed to direct two-photon ionization of He in combination with AC Stark broadening of the excited state and absorption of the XUV light in the medium. Absolute photoionization cross-sections from the He 1s2p P-1 and He 1s3p P-1 states in the threshold region are determined by measuring the saturation of the ionization process with a precision of similar to 25%. In addition, the variation of the relative orientation between the polarization vectors of the pump and probe beams enables the determination of partial photoionization cross-sections.

X-rays with a synchrotron-like spectrum and associated critical energy ∼ 5 keV, with a peak brightness of ∼ 1×1021 ph/s/mm2/mrad2/0.1%BW, were achieved by employing 16 TW laser pulses.

    • We demonstrate a new scheme for extreme ultraviolet (xuv) Fourier-transform spectroscopy based on the generation of two phase-locked high-harmonic beams. It allows us to measure for the first time interferograms at wavelengths as short as 90 nm, and open the perspective of performing high-resolution Fourier-transform absorption spectroscopy in the xuv. Our measurements also demonstrate that a precise control of the relative phase of harmonic pulses can be obtained with an accuracy on an attosecond time scale, of importance for future xuv pump-xuv probe attosecond spectroscopy.

    • Background & Aims: Severe cholestasis may cause cholemic nephropathy that can be modeled in common bile duct ligated (CBDL) mice. We aimed to explore the therapeutic efficacy and mechanisms of norursodeoxycholic acid (norUDCA) in cholemic nephropathy. Methods: In 8-week CBDL mice fed with norUDCA (prior or post CBDL) or chow we evaluated serum urea levels, urine cytology and urinary neutrophil gelatinase associated lipocalin (uNGAL), kidney and liver tissue quantification of fibrosis by hydroxyproline content and gene chip expression looking at key genes of inflammation and fibrosis. Moreover, we comprehensively analysed bile acid profiles in liver, kidney, serum and urine samples. Results: NorUDCA-fed CBDL mice had significantly lower serum urea and uNGAL levels and less severe cholemic nephropathy as demonstrated by normal urine cytology, significantly reduced tubulointerstitial nephritis, and renal fibrosis as compared to controls. NorUDCA underwent extensive metabolism to produce even more hydrophilic compounds that were significantly enriched in kidneys. Conclusion: NorUDCA ameliorates cholemic nephropathy due to the formation of highly hydrophilic metabolites enriched in kidney. Consequently, norUDCA may represent a medical treatment for cholemic nephropathy. Lay summary: The term cholemic nephropathy describes renal dysfunction together with characteristic morphological alterations of the kidney in obstructive cholestasis that can be mimicked by ligation of the common bile duct in mice. Feeding the hydrophilic bile acid norUDCA to bile duct ligated mice leads to a significant amelioration of the renal phenotype due to the formation of highly hydrophilic metabolites enriched in the kidney and may therefore represent a medical treatment for cholemic nephropathy. The state is excited by the 13th harmonic of a tunable 80 ps laser and ionized by a synchronous ultraviolet laser, with a variable time delay. This experiment demonstrates that the high harmonics generated in a jet of rare gas exposed to an intense laser field provide a tunable short-pulse xuv source ideally suited for pump/probe type of studies and, in particular, short lifetime measurements.

    • Using time-resolved UV laser-induced fluorescence from a laser-produced plasma, natural radiative lifetimes have been measured for 21 levels of Ge I belonging to the odd-parity 4p4d, 4p5d, and 4p6s configurations. Stimulated Brillouin scattering in water has allowed us to compress Nd:YAG laser pulses pumping a dye laser (where YAG denotes yttrium aluminum garnet), thus yielding 1-ns tunable laser pulses to enable accurate measurements of short lifetimes. Branching ratios of Ge I have been measured by inductively coupled plasma emission spectrometry, the intensity calibration being performed by means of Ar lines emitted by a hollow-cathode lamp. The experimental lifetimes and branching ratios have been combined in order to provide a set of accurate transition probabilities for the 4p-5s and 4p-4d transitions. A relativistic Hartree-Fock; calculation, taking configuration-interaction and polarization effects into account, has been combined with a least-squares optimization procedure of the Slater and spin-orbit integrals in order to test the ability of this approach to correctly predict radiative properties of the group-IV elements. Good agreement between experimental and theoretical transition probability values has been achieved for most of the transitions considered.

    • Laser wakefield accelerators offer the possibility of compact electron acceleration. However one of the key outstanding issues with the results reported to date is the electron beam stability. Experiments on two laser systems reveal that the contrast ratio between the ASE pedestal and main pulse is an important factor in determining the quality of the electron beam. With a high contrast ratio (10^8) the electron beam profile is a well collimated single beam having a low pointing instability (<10 mrad rms). With a lower contrast (10^6) the beam profile contains multiple beamlets which exhibit a large pointing instability (~50 mrad rms). Ahigh contrast ratio not only improves the beam pointing stability (~6 mrad) but also stabilizes the electron beam energy reproducibility (5%).

    • We demonstrate that the spatial mode of high-order harmonics can be continuously controlled. The control is achieved by spatially modulating the degree of elliptical polarization of the fundamental field using birefringent optics. A highly sensitive relationship between the efficiency of harmonic generation and the degree of laser elliptical polarization leads to atoms emitting harmonics mainly in regions of linear polarization. The harmonics are emitted as annular beams whose angles of divergence can be continuously varied.

    • The far-field spatial distributions of high-order harmonics in an ionized medium have been investigated using a computer model that includes both the single atom effects and the collective effects of the medium. The results show that depletion of the non-linear medium due to ionization and radial phase variations across the gas jet cause broadening and complicated structures in the far-field spatial distributions. These phase variations are due to three factors: focusing, ionization, i.e. depletion of the non-linear medium and electron dispersion, and intensity-dependent phase variations of the atomic dipoles.

    • Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease of unknown aetiology for which there are no approved therapeutic options. Patients with PSC display changes in gut microbiota and in bile acid (BA) composition; however, the contribution of these alterations to disease pathogenesis remains controversial. Here we identify a role for microbiota-dependent changes in BA synthesis that modulates PSC pathophysiology. In a genetic mouse model of PSC, we show that loss of microbiota-mediated negative feedback control of BA synthesis results in increased hepatic BA concentrations, disruption of bile duct barrier function and, consequently, fatal liver injury. We further show that these changes are dependent on decreased BA signalling to the farnesoid X receptor, which modulates the activity of the rate-limiting enzyme in BA synthesis, CYP7A1. Moreover, patients with advanced stages of PSC show suppressed BA synthesis as measured by serum C4 levels, which is associated with poor disease prognosis. Our preclinical data highlight the microbiota-dependent dynamics of BA metabolism in cholestatic liver disease, which could be important for future therapies targeting BA and gut microbiome interactions, and identify C4 as a potential biomarker to functionally stratify patients with PSC and predict disease outcomes. Patients with primary sclerosing cholangitis (PSC), a chronic cholestatic liver disease, display changes in the gut microbiota and in bile acid composition. Schneider, Candels and colleagues identify a role for microbiota-dependent regulation of bile acid synthesis through farnesoid X receptor signalling, which is relevant for PSC disease progression.

    • We report on a time-resolved study of the fifth harmonic generated in xenon by 140 ps pulses from a Nd:YAG laser in the 10(13) W cm-2 intensity regime. Absolute timing between the driving laser pulse and the harmonic pulse could be determined by means of reference harmonics generated in non-linear crystals and a single common streak camera. Above a certain laser intensity the centre of the observed harmonic pulse was shifted earlier in time relative to the laser pulse. We found the intensity dependence of this shift to be approximately linear within the intensity range used which is consistent with the results of numerical simulations taking the ionization and dispersion of the xenon gas into account.

    • Seaweed biomass is a renewable resource with multiple applications. Sea-based cultivation of seaweeds can provide high biomass yields, low construction, operation, and maintenance costs and could offer an environmentally and economically sustainable alternative to land-based cultivations. The biochemical profile of sea-grown biomass depends on seasonal variation in environmental factors, and the optimization of harvest time is important for the quality of the produced biomass. To identify optimal harvest times of Swedish sea-based cultivated sea lettuce (Ulva fenestrata), this study monitored biomass yield, morphology, chemical composition, fertility, and biofouling at five different harvesting times in April - June 2020. The highest biomass yields (approx. 1.2 kg fw [m rope]-1) were observed in late spring (May). The number and size of holes in the thalli and the amount of fertile and fouled tissue increased with prolonged growth season, which together led to a significant decline in both biomass yield and quality during summer (June). Early spring (April) conditions were optimal for obtaining high fatty acid, protein, biochar, phenolic, and pigment contents in the biomass, whereas carbohydrate and ash content, as well as essential and non-essential elements, increased later in the growth season. Our study results show that the optimal harvest time of sea-based cultivated U. fenestrata depends on the downstream application of the biomass and must be carefully selected to balance yield, quality, and desired biochemical contents to maximize the output of future sea-based algal cultivations in the European Northern Hemisphere.
