  • Holocene forcing of aeolian dust activity over the Tibetan Plateau and its surroundings

    • Aeolian deposits on the Tibetan Plateau (TP) and its surroundings provide crucial source materials for the Asian dust cycle, which significantly affects Asian and global ecosystems and climate. However, it is unclear how the dust dynamics of the TP and its surroundings are linked to Earth's climate system. To address this issue, we examined the grain size and accumulation rate of six Holocene aeolian sections on the southern TP (a new, well-dated high-resolution section, two relatively low-resolution sections, and three published sections) and combined them with equivalent aeolian sedimentary records from eastern arid central Asia. The results suggest that dust activity in both regions decreased during the early to middle Holocene and then increased in the late Holocene. We hypothesize that the primary drivers of Holocene dust activity in both regions are similar. Cold-season insolation, as the primary driving factor, combined with ice volume and atmospheric CO2 concentration, collectively controlled the regional temperature, which determined the near-surface wind intensity via its influence on the TP High and Siberian High, respectively, thus ultimately controlling the regional dust activity. In this context, we project that dust activity on the TP and its neighboring areas will decrease under warm scenarios in the 21st century. Overall, our findings provide an extensive overview of the past, present, and future scenarios of Asian dust activity, especially of the TP dust.

  • Endothelial cell heterogeneity and microglia regulons revealed by a pig cell landscape at single-cell level

    • Pigs are valuable large animal models for biomedical and genetic research, but insights into the tissue- and cell-type-specific transcriptome and heterogeneity remain limited. By leveraging single-cell RNA sequencing, we generate a multiple-organ single-cell transcriptomic map containing over 200,000 pig cells from 20 tissues/organs. We comprehensively characterize the heterogeneity of cells in tissues and identify 234 cell clusters, representing 58 major cell types. In-depth integrative analysis of endothelial cells reveals a high degree of heterogeneity. We identify several functionally distinct endothelial cell phenotypes, including an endothelial to mesenchymal transition subtype in adipose tissues. Intercellular communication analysis predicts tissue- and cell type-specific crosstalk between endothelial cells and other cell types through the VEGF, PDGF, TGF-beta, and BMP pathways. Regulon analysis of single-cell transcriptome of microglia in pig and 12 other species further identifies MEF2C as an evolutionally conserved regulon in the microglia. Our work describes the landscape of single-cell transcriptomes within diverse pig organs and identifies the heterogeneity of endothelial cells and evolutionally conserved regulon in microglia.

  • Common variants in breast cancer risk loci predispose to distinct tumor subtypes

    • BackgroundGenome-wide association studies (GWAS) have identified multiple common breast cancer susceptibility variants. Many of these variants have differential associations by estrogen receptor (ER) status, but how these variants relate with other tumor features and intrinsic molecular subtypes is unclear.MethodsAmong 106,571 invasive breast cancer cases and 95,762 controls of European ancestry with data on 173 breast cancer variants identified in previous GWAS, we used novel two-stage polytomous logistic regression models to evaluate variants in relation to multiple tumor features (ER, progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2) and grade) adjusting for each other, and to intrinsic-like subtypes.ResultsEighty-five of 173 variants were associated with at least one tumor feature (false discovery rate < 5%), most commonly ER and grade, followed by PR and HER2. Models for intrinsic-like subtypes found nearly all of these variants (83 of 85) associated at p < 0.05 with risk for at least one luminal-like subtype, and approximately half (41 of 85) of the variants were associated with risk of at least one non-luminal subtype, including 32 variants associated with triple-negative (TN) disease. Ten variants were associated with risk of all subtypes in different magnitude. Five variants were associated with risk of luminal A-like and TN subtypes in opposite directions.ConclusionThis report demonstrates a high level of complexity in the etiology heterogeneity of breast cancer susceptibility variants and can inform investigations of subtype-specific risk prediction.

  • Gestational dietary betaine supplementation suppresses hepatic expression of lipogenic genes in neonatal piglets through epigenetic and glucocorticoid receptor-dependent mechanisms

    • Methyl donors play critical roles in nutritional programming through epigenetic regulation of gene expression. Here we fed gestational sows with control or betaine-supplemented diets (3 g/kg) throughout the pregnancy to explore the effects of maternal methyl-donor nutrient on neonatal expression of hepatic lipogenic genes. Betaine-exposed piglets demonstrated significantly lower liver triglyceride content associated with down-regulated hepatic expression of lipogenic genes acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), stearoyl-CoA desaturase (SCD) and sterol regulatory element-binding protein-1c. Moreover, s-adenosyl methionine to s-adenosyl homocysteine ratio was elevated in the liver of betaine-exposed piglets, which was accompanied by DNA hypermethylation on FAS and SCD gene promoters and more enriched repression histone mark H31K27me3 on SCD gene promoter. Furthermore, glucocorticoid receptor (GR) binding to SCD gene promoter was diminished along with reduced serum cortisol and liver GR protein content in betaine-exposed piglets. GR-mediated SCD gene regulation was confirmed in HepG2 cells in vitro. Dexamethasone (Dex) drastically increased the luciferase activity of porcine SCD promoter, while the deletion of GR response element on SCD promoter significantly attenuated Dex-mediated SCD transactivation. In addition, miR-let-7e, miR-1285 and miR-124a, which respectively target porcine SCD, ACC and GR, were significantly up-regulated in the liver of betaine-exposed piglets, being in accordance with decreased protein content of these three genes. Taken together, our results suggest that maternal dietary betaine supplementation during gestation attenuates hepatic lipogenesis in neonatal piglets via epigenetic and GR-mediated mechanisms.

  • Evaluation of starvation status in the early developmental stages of black rockfish (Sebastes schlegelii) based on morphological and histological characteristics

    • Assessing the nutritional status and identifying major causes of mortality in larvae experiencing varying degrees of starvation are crucial for establishing appropriate feeding protocols and enhancing the welfare of hatchery-reared fish. The black rockfish Sebastes schlegelii is an important species in aquaculture and stock enhancement efforts in China, Japan, and Korea. This study aimed to identify optimal diagnostic morphometric indicators of starvation in newly hatched (0–6 days post-hatch, DPH) and postlarval stages (27–37 DPH) of this valuable fish species through histological analyses. Our findings revealed that certain morphometric parameters, including body length, the ratios of eye diameter to head height, body height to body length, and abdomen height to body height, exhibit sensitivity to starvation during both larval and postlarval stages. Particularly, the ratios of body height to body length and abdomen height to body height emerged as the most sensitive morphometric indicators of starvation. Histological examinations of the digestive system revealed rapid alterations in the morphology of hepatic parenchymal cells, accompanied by a significant decrease in the number of lipid cells in the liver during episodes of food deprivation. Starvation induced cellular degeneration in the digestive organs, manifested by reduced heights of epithelial cells and mucosal layers in the intestine, oesophagus, and stomach, along with degeneration and separation of muscle fibers. Among these variables, the height of the intestinal submucosa and muscle layer emerged as the most sensitive indicators reflecting nutritional conditions in newly hatched larvae. In contrast, the height of intestinal striated borders and mucosal folds proved to be the most sensitive indicators in the postlarval stage. Furthermore, the height of intestinal epithelial cells and the number of lipid vacuoles in enterocytes exhibited high sensitivity to food deprivation in both newly hatched larvae and postlarvae. These findings underscore the varying resilience of fish to starvation during different developmental phases and highlight the utility of morphological sensitivity characteristics as reliable diagnostic indices for assessing nutritional status in relation to starvation or suboptimal feeding during the early developmental stages of black rockfish in hatchery-reared processes.

    • Millimeter wave (mmWave) communication can realize high transmission rates in vehicular networks. Nevertheless, severe blocking effects and high mobility of vehicles would seriously affect downlink services for vehicles. To ensure communication quality and stability, this paper jointly explores beam allocation and relay selection in mmWave vehicular networks from the perspective of artificial intelligence-driven model. We utilize queuing theory to simulate dynamic distributions of vehicles and firstly propose a deep reinforcement learning (DRL) based joint beam allocation and relay selection scheme to mitigate the blocking effects and optimize the total communication capacity. When the expected downlink is blocked, mmWave base station (mmBS) can select appropriate idle vehicles as the relay nodes for service. Besides, we set the capacity threshold when designing the scheme to guarantee each target vehicle can obtain the ideal service. Through proper training, mmBS can intelligently find an optimal solution for the constantly updated vehicular networks based on the location of vehicles. Simulation results demonstrate the effectiveness of our scheme, which can restrain the transmission outage caused by random blockage and improve the total communication capacity of vehicular networks.

  • Assessment of the degradation efficiency of full-scale biogas plants : A comparative study of degradation indicators

    • Increasing popularity and applications of the anaerobic digestion (AD) process has necessitated the development and identification of tools for obtaining reliable indicators of organic matter degradation rate and hence evaluate the process efficiency especially in full-scale, commercial biogas plants. In this study, four biogas plants (A1, A2, B and C) based on different feedstock, process configuration, scale and operational performance were selected and investigated. Results showed that the biochemical methane potential (BMP) based degradation rate could be use in incisively gauging process efficiency in lieu of the traditional degradation rate indicators. The BMP degradation rates ranged from 70 to 90% wherein plants A2 and C showed the highest throughput. This study, therefore, corroborates the feasibility of using the BMP degradation rate as a practical tool for evaluating process performance in full-scale biogas processes and spots light on the microbial diversity in full-scale biogas processes.

  • Improving the real-time experience for software-measurement system end-users

    • Background: Software measurement systems are used in large companies to provide developers with up-to-date feedback and metrics. Aim /Problem: However, the front-ends of these systems are often not ready to provide a real-time experience for the end-users, who sometimes have to wait minutes before visualizations are provided. Method: In this paper we compare four alternative technological setups for these front-ends that were created within a large international telecommunication provider. We use a publicly available dataset for a performance evaluation and to analyze the results. Results: Our results indicate that the choice of the visualization component has a larger impact on the performance than the choice of the data storage. However, performance is also impacted by the combination of storage and visualization tools used.
