  • Atmospheric new particle formation from sulfuric acid and amines in a Chinese megacity
  • 2018
  • Ingår i: Science. - : American Association for the Advancement of Science (AAAS). - 0036-8075 .- 1095-9203. ; 361:6399, s. 278-281
  • Tidskriftsartikel (refereegranskat)abstract
    • Atmospheric new particle formation (NPF) is an important global phenomenon that is nevertheless sensitive to ambient conditions. According to both observation and theoretical arguments, NPF usually requires a relatively high sulfuric acid (H2SO4) concentration to promote the formation of new particles and a low preexisting aerosol loading to minimize the sink of new particles. We investigated NPF in Shanghai and were able to observe both precursor vapors (H2SO4) and initial clusters at a molecular level in a megacity. High NPF rates were observed to coincide with several familiar markers suggestive of H2SO4-dimethylamine (DMA)water (H2O) nucleation, including sulfuric acid dimers and H2SO4-DMA clusters. In a cluster kinetics simulation, the observed concentration of sulfuric acid was high enough to explain the particle growth to similar to 3 nanometers under the very high condensation sink, whereas the subsequent higher growth rate beyond this size is believed to result fromthe added contribution of condensing organic species. These findings will help in understanding urban NPF and its air quality and climate effects, as well as in formulating policies to mitigate secondary particle formation in China.

  • Emerging contaminants: A One Health perspective
  • 2024
  • Ingår i: Innovation. - 2666-6758. ; 5
  • Forskningsöversikt (refereegranskat)abstract
    • Environmental pollution is escalating due to rapid global development that often prioritizes human needs over planetary health. Despite global efforts to mitigate legacy pollutants, the continuous introduction of new substances remains a major threat to both people and the planet. In response, global initiatives are focusing on risk assessment and regulation of emerging contaminants, as demonstrated by the ongoing efforts to establish the UN's Intergovernmental Science-Policy Panel on Chemicals, Waste, and Pollution Prevention. This review identifies the sources and impacts of emerging contaminants on planetary health, emphasizing the importance of adopting a One Health approach. Strategies for monitoring and addressing these pollutants are discussed, underscoring the need for robust and socially equitable environmental policies at both regional and international levels. Urgent actions are needed to transition toward sustainable pollution management practices to safeguard our planet for future generations.

  • The state of science on severe air pollution episodes : Quantitative and qualitative analysis
  • 2021
  • Ingår i: Environment International. - : Elsevier BV. - 1873-6750 .- 0160-4120. ; 156, s. 106732-106732
  • Tidskriftsartikel (refereegranskat)abstract
    • Severe episodic air pollution blankets entire cities and regions and have a profound impact on humans and their activities. We compiled daily fine particle (PM2.5) data from 100 cities in five continents, investigated the trends of number, frequency, and duration of pollution episodes, and compared these with the baseline trend in air pollution. We showed that the factors contributing to these events are complex; however, long-term measures to abate emissions from all anthropogenic sources at all times is also the most efficient way to reduce the occurrence of severe air pollution events. In the short term, accurate forecasting systems of such events based on the meteorological conditions favouring their occurrence, together with effective emergency mitigation of anthropogenic sources, may lessen their magnitude and/or duration. However, there is no clear way of preventing events caused by natural sources affected by climate change, such as wildfires and desert dust outbreaks.

  • 2021
  • Ingår i: Symmetry. - : MDPI AG. - 2073-8994. ; 13:9
  • Tidskriftsartikel (refereegranskat)abstract
    • With the continuous development of intelligent product interaction technology, the facial expression design of virtual images on the interactive interface of intelligent products has become an important research topic. Based on the current research on facial expression design of existing intelligent products, we symmetrically mapped the PAD (pleasure-arousal-dominance) emotion value to the image design, explored the characteristics of abstract expressions and the principles of expression design, and evaluated them experimentally. In this study, the experiment of PAD scores was conducted on the emotion expression design of abstract expressions, and the data results were analyzed to iterate the expression design. The experimental results show that PAD values can effectively guide designers in expression design. Meanwhile, the efficiency and recognition accuracy of human communication with abstract expression design can be improved by facial auxiliary elements and eyebrows.

  • 2020
  • Ingår i: HCI International 2020 – Late Breaking Papers: Digital Human Modeling and Ergonomics, Mobility and Intelligent Environments. - Cham : Springer. - 9783030599867 - 9783030599874 ; , s. 236-248
  • Konferensbidrag (refereegranskat)abstract
    • Head-Up display (HUD) is increasingly applied to automobiles. However, HUD might have also shortcomings causing new driving problems. This paper investigates the effects of different Human Machine Interfaces (HMI) of HUD under ACC function on situation awareness (SA) and system usability for cut-in driving scenarios. The laboratory-based controlled experiment conducted used a driving simulator with a total of 8 participants. Each participant performed three different tasks, using three different HMIs including two HUD HMI and one baseline HMI (dashboard HMI). The results indicate that HUD display can influence the participants’ SA and system usability, and that different HUD design can have different effects. The HUD design with dynamic directivity of augmented reality brings about better SA and system usability. The research suggest that it is possible to improve the SA and system usability through improved HUD-HMI design.

  • Ingår i: Human Interaction, Emerging Technologies and Future Applications II. - Cham : Springer. - 9783030442668 - 9783030442675 ; , s. 285-291
  • Konferensbidrag (refereegranskat)abstract
    • Advances in vehicle technology open up both opportunities and challenges for human-machine interface (HMI) design in intelligent vehicles. Design guidelines for icons in central consoles of vehicles have been discussed in human-computer interaction community, but more study and innovation are needed in icon design. In this paper, we investigated the displayed line thickness and size of icons in the intelligent vehicle’s central control screen, based on international design guidelines and standards. The experiment involved 17 participants performing simulated in-vehicle secondary task. The result from the experiments shows that the usability of icons increases and the driver’s workload decreases as the icons get larger. We also found a set of recommended values for size and line thickness of icons in this context. Future work will focus on providing design guidelines for other aspects of HMI design in intelligent vehicles.

  • The landscape of somatic mutations in infant MLL-rearranged acute lymphoblastic leukemias.
  • 2015
  • Ingår i: Nature Genetics. - : Springer Science and Business Media LLC. - 1546-1718 .- 1061-4036. ; 47:4, s. 192-330
  • Tidskriftsartikel (refereegranskat)abstract
    • Infant acute lymphoblastic leukemia (ALL) with MLL rearrangements (MLL-R) represents a distinct leukemia with a poor prognosis. To define its mutational landscape, we performed whole-genome, exome, RNA and targeted DNA sequencing on 65 infants (47 MLL-R and 18 non-MLL-R cases) and 20 older children (MLL-R cases) with leukemia. Our data show that infant MLL-R ALL has one of the lowest frequencies of somatic mutations of any sequenced cancer, with the predominant leukemic clone carrying a mean of 1.3 non-silent mutations. Despite this paucity of mutations, we detected activating mutations in kinase-PI3K-RAS signaling pathway components in 47% of cases. Surprisingly, these mutations were often subclonal and were frequently lost at relapse. In contrast to infant cases, MLL-R leukemia in older children had more somatic mutations (mean of 6.5 mutations/case versus 1.3 mutations/case, P = 7.15 × 10(-5)) and had frequent mutations (45%) in epigenetic regulators, a category of genes that, with the exception of MLL, was rarely mutated in infant MLL-R ALL.

  • The NF-κB-modulated miR-19a-3p enhances malignancy of human ovarian cancer cells through inhibition of IGFBP-3 expression
  • 2019
  • Ingår i: Molecular Carcinogenesis. - : Wiley. - 0899-1987 .- 1098-2744. ; 58:12, s. 2254-2265
  • Tidskriftsartikel (refereegranskat)abstract
    • Ovarian cancer is the most lethal gynecologic malignancy due to the lack of symptoms until advanced stages, and new diagnosis and treatment strategy is in urgent need. In this study, we found higher expression of miR-19a-3p in ovarian cancer tissues compared with that in the adjacent normal tissues. By chromatin immunoprecipitation (ChIP) and electrophoretic mobility shift assay (EMSA) analysis, we showed that nuclear factor-kappaB (NF-κB) binds to the promoter of miR-19a-3p, leading to reduced expression in ovarian cancer cells. Further study indicated that miR-19a-3p inhibits the expression of insulin-like growth factor binding protein-3 (IGFBP-3), resulting in enhanced growth and migration of ovarian cancer cells in vitro and tumor growth in vivo. These results showed that miR-19a-3p enhances the oncogenesis of ovarian cancer through inhibition of IGFBP-3 expression, and which can be inhibited by NF-κB, suggesting an NF-κB/miR-19a-3p/IGFBP-3 pathway in the oncogenesis of ovarian cancer, which expands our understanding of ovarian cancer and they may contribute to the development of new diagnosis and treatment of ovarian cancer.

  • Reconciling modeling with observations of radiative absorption of black carbon aerosols
  • 2017
  • Ingår i: Journal of Geophysical Research - Atmospheres. - 2169-897X .- 2169-8996. ; 122:11, s. 5932-5942
  • Tidskriftsartikel (refereegranskat)abstract
    • The physical treatment of internal mixing and aging of black carbon (BC) aerosols that allow for enhanced solar absorption of the BC is an important parameterization in climate models. Many climate models predict a factor of 2-3 lower aerosol absorption optical depth (AAOD) than the atmospheric columnar absorption observed from ground-based networks such as AERONET, likely because these models do not parameterize properly the BC absorption enhancement (E-MAC). Models that are configured with an internal mixing have predicted large variations of E-MAC, which are poorly constrained from ambient measurements. We determined the BC E-MAC from aerosol coatings with a two-step solvent experiment to remove both organic and inorganic coatings in ambient fine particulate matter (PM2.5). Observations in a rural North China site showed that the E-MAC varied from 1.4 to 3. The E-MAC increases simultaneously with SO42-/EC ratios, suggesting the photochemical production of sulfate coatings enhanced BC absorption. A global climate model, parameterized to account for these observational constraints, verifies that sulfates are primary drivers of the BC absorption enhancement in severely polluted area in China. This magnification of the radiative forcing of coated BC is stronger by a factor of similar to 2 than predicted by the standard parameterization (external mixing) in the climate model and is in better agreement with AERONET observations of AAOD. This result would be useful for testing the representation of solar absorption by BC-containing particles in the newer generation of climate models. Plain Language Summary Atmospheric black carbon (BC) or soot in fine particulate matter (PM2.5) is emitted from incomplete combustion of fossil fuel or biomass/biofuel. The BC is an important pollutant for both air quality and Earth's energy balance, and the BC radiative forcing maybe second only to that of CO2. The photochemical production of nonabsorbing secondary aerosols may create a coating on BC and may thereby act as a lens which may enhance the light absorption. However, this absorption enhancement is poorly constrained by ambient measurements, and thus the estimates of BC climate forcing remain highly uncertain. To this end, an aerosol filter dissolution-filtration (AFD) with two-step solvent dissolution protocol was employed to remove both organic and inorganic coatings and then investigate their effects on BC light absorption. The observations and model simulation showed that the BC warming effect likely doubled due to lens effect from secondary aerosols.

  • Ingår i: Intelligent Human Systems Integration 2019. - Cham : Springer. - 9783030110505 - 9783030110512 ; , s. 570-575
  • Konferensbidrag (refereegranskat)abstract
    • With the development of society, technical products become more complex and intelligent. Interaction design of intelligent products is meeting new challenges. It requires practitioners to wade into the whole product development process and grasp basic interdisciplinary knowledge. This paper focuses on design methodologies for Human-Artificial Systems design with interdisciplinary knowledge. In the Interaction Design Method Framework (IDMF) we proposed, design runs through the whole product development process to help practitioners propose emerging technology-based designs. The IDMF framework is composed of 102 design methods and six phases. We deconstructed each phase of the IDMF according to three dimensions, design, information, and business. This paper offers a practical guide to using the IDMF framework by providing a high-level summary of the automotive AR-HUD design. It presented how the IDMF framework help practitioners effectively guide the design team and propose emerging technology-based designs.

  • Radiative absorption enhancement from coatings on black carbon aerosols
  • 2016
  • Ingår i: Science of the Total Environment. - : Elsevier BV. - 0048-9697 .- 1879-1026. ; 551, s. 51-56
  • Tidskriftsartikel (refereegranskat)abstract
    • The radiative absorption enhancement of ambient black carbon (BC), by light-refractive coatings of atmospheric aerosols, constitutes a large uncertainty in estimates of climate forcing. The direct measurements of radiative absorption enhancement require the experimentally-removing the coating materials in ambient BC-containing aerosols, which remains a challenge. Here, the absorption enhancement of the BC core by non-absorbing aerosol coatings was quantified using a two-step removal of both inorganic and organic matter coatings of ambient aerosols. The mass absorption cross-section (MAC) of decoated/pure atmospheric BC aerosols of 4.4 +/- 0.8 m(2)g(-1) was enhanced to 9.6 +/- 1.8 m(2)g(-1) at 678-nm wavelength for ambiently-coated BC aerosols at a rural Northern China site. The enhancement of MAC (E-MAC) rises from 1.4 +/- 0.3 in fresh combustion emissions to similar to 3 for aged ambient China aerosols. The three-week high-intensity campaign observed an average E-MAC of 2.25 +/- 0.55, and sulfates were primary drivers of the enhanced BC absorption.

  • A New Machine-Learning Prediction Model for Slope Deformation of an Open-Pit Mine : An Evaluation of Field Data
  • 2019
  • Ingår i: Energies. - : MDPI. - 1996-1073. ; 12:7
  • Tidskriftsartikel (refereegranskat)abstract
    • Effective monitoring of the slope deformation of an open-pit mine is essential for preventing catastrophic collapses. It is a challenging task to accurately predict slope deformation. To this end, this article proposed a new machine-learning method for slope deformation prediction. Ground-based interferometric radar (GB-SAR) was employed to collect the slope deformation data from an open-pit mine. Then, an ensemble learner, which aggregated a set of weaker learners, was proposed to mine the GB-SAR field data, delivering a slope deformation prediction model. The evaluation of the field data acquired from the Anjialing open-pit mine demonstrates that the proposed slope deformation model was able to precisely predict the slope deformation of the monitored mine. The prediction accuracy of the super learner was superior to those of all the independent weaker learners.

  • The genomic landscape of core-binding factor acute myeloid leukemias
  • 2016
  • Ingår i: Nature Genetics. - : Springer Science and Business Media LLC. - 1546-1718 .- 1061-4036. ; 48, s. 1551-1556
  • Tidskriftsartikel (refereegranskat)abstract
    • Acute myeloid leukemia (AML) comprises a heterogeneous group of leukemias frequently defined by recurrent cytogenetic abnormalities, including rearrangements involving the core-binding factor (CBF) transcriptional complex. To better understand the genomic landscape of CBF-AMLs, we analyzed both pediatric (n = 87) and adult (n = 78) samples, including cases with RUNX1-RUNX1T1 (n = 85) or CBFB-MYH11 (n = 80) rearrangements, by whole-genome or whole-exome sequencing. In addition to known mutations in the Ras pathway, we identified recurrent stabilizing mutations in CCND2, suggesting a previously unappreciated cooperating pathway in CBF-AML. Outside of signaling alterations, RUNX1-RUNX1T1 and CBFB-MYH11 AMLs demonstrated remarkably different spectra of cooperating mutations, as RUNX1-RUNX1T1 cases harbored recurrent mutations in DHX15 and ZBTB7A, as well as an enrichment of mutations in epigenetic regulators, including ASXL2 and the cohesin complex. This detailed analysis provides insights into the pathogenesis and development of CBF-AML, while highlighting dramatic differences in the landscapes of cooperating mutations for these related AML subtypes.

  • Tidskriftsartikel (refereegranskat)abstract
    • Context: Requirements-elicitation is one of the essential steps towards software design and construction. Business analysts and stakeholders often face challenges in gathering or conveying key software requirements. There are many methods, and tools designed by researchers and practitioners, but with the invention of new technologies, there appears to be a need to make requirements gathering and design-rationale process more efficient. Storytelling is an emerging concept and researchers are witnessing its effectiveness in education, community-building, information system, and requirement elicitation. Objective: Objectives of this study are to i) devise a method for requirements elicitation and improving design-rationales using story-based technique; ii) evaluate the effectiveness of the aforementioned proposed activity. Methodology: To answer the research objectives, we have i) conducted open-ended interviews to get feedback on our proposed method; ii) case requirement from a running project to map how this method can be useful; and iii) performed empirical evaluation of the proposed card-based activity. Result: i) Our regression model has shown that participants' perception regarding the ease of use and the fun in the game has an ultimate effect on requirements elicitation through enhancing user's desire to play the game, hence, increasing the collaborative learning outcomes of the game; ii) Our results have shown that using team-based activities helps the less-experienced designers to argue through design rationales and better elicit software requirements. Our results have reinforced the finding that using game-based solutions not only enhances communication and develops trust between stakeholders but also helps in motivating participants of requirements activity; iii) Initial results (from interview and empirical evaluation) for the proposed technique and method show positive results. Improvement in the process and activity as suggested by the participants will be accommodated in future studies.

    • Context: Researchers around the globe invest a lot of time searching the literature for performing reviews (Systematic Literature Review (SLR), Multivocal Literature Review (MLR)). The steps to performing the review includes inclusion of the grey literature, preprints, and quality assessed non-peer reviewed literature (the purpose is to minimize the publication bias). The initial screening of the papers takes time and bibliographic information is only available online for the researcher(s). Objective: Objective of our study is to propose, design, and develop a method that will help the research community to download the basic information of the papers (title, abstract, author) for the searched query from arxiv, SSRN, and SocArxiv (Social Science ArXiv). Method: We used Web scraping to extract data from the servers and save it in excel file. To retrieve the desired query from the databases, a Python code is used. Two methods have been discussed in the study to download the metadata of the searched query. Results: We have used different queries (such as "grey literature", "testing software", and "python" etc.) to see the results of our proposed method. Furthermore, we cross-verified the results with the online search results of the databases. Conclusion: Initial results from the preliminary pilot evaluations show that it is a viable method to search, download, and shortlist the research articles information (title, abstract etc.) from arXiv,1 SSRN,2 and SocArXiv.3 For external validity more evaluations are needed.

  • 2024
  • Ingår i: Journal of manufacturing systems. - : Elsevier BV. - 0278-6125 .- 1878-6642. ; 72, s. 406-423
  • Tidskriftsartikel (refereegranskat)abstract
    • Intelligent models for tool wear condition monitoring (TWCM) have been extensively researched. However, in industrial scenarios, limited acquired monitoring signals and variations of machining parameters lead to insufficient training samples and data distribution shifts for the models. To address the issues, this research presents a novel residual attention hybrid adaptation network (RAHAN) model based on a residual attention network (ResAttNet) and a hybrid adaptation strategy. In the RAHAN model, by integrating a channel attention mechanism and deep residual modules, ResAttNet is designed as a feature extractor to acquire features from monitoring signals for tool wear conditions. Embedding subdomain adaptation into a condition recognizer while establishing separate adversarial learning in a domain obfuscator, the hybrid adaptation strategy is developed to eliminate global distribution shifts and align local distributions of each tool wear phase simultaneously. Six migration tasks under a laboratory and two factory machining platforms were conducted to evaluate the effectiveness of the RAHAN model. Compared with a baseline model, four ablation models, and six state-of-the-art transfer learning models, the RAHAN model achieved the highest average accuracy of 92.70% on six migration tasks. Furthermore, the RAHAN model shows clearer feature representations of each tool wear condition than other compared models. The comparative results demonstrate that the RAHAN model has superior transferability and therefore can be considered as a good potential solution to support cross-domain TWCM under different machining processes.

    • Tyrosine phosphorylation is an important post-translation modification of proteins that is controlled by tyrosine kinases and phosphatases. Disruption of the balance between the activity of tyrosine kinases and phosphatases may result in diseases. Receptor type protein tyrosine phosphatase epsilon (PTPRE) is closely related with receptor type protein tyrosine phosphatase alpha (PTPRA). PTPRE has been studied in osteoclast cells, nerve cells, hematopoietic cells, cancer cells and others, and it has different functions among various tissues. In this review, we summarized the current knowledge about the regulation of PTPRE on cellular signal transduction and its function under normal and pathological conditions.

  • DDR1/2 enhance KIT activation and imatinib resistance of primary and secondary KIT mutants in gastrointestinal stromal tumors
  • 2024
  • Ingår i: Molecular Carcinogenesis. - 0899-1987. ; 63:1, s. 75-93
  • Tidskriftsartikel (refereegranskat)abstract
    • Gastrointestinal stromal tumors (GISTs) are predominantly initiated by KIT mutations. In this study, we observed that discoidin domain receptors 1 and 2 (DDR1 and DDR2) exhibited high expression in GISTs, were associated with KIT, and enhanced the activation of both wild-type KIT and primary KIT mutants. Inhibition of DDR1/2 led to a reduction in the activation of KIT and its downstream signaling molecules, ultimately impairing GIST cell survival and proliferation in vitro. Consequently, treatment of mice carrying germline KIT/V558A mutation with DDR1/2 inhibitor significantly impeded tumor growth, and the combined use of DDR1/2 inhibitor and imatinib, the first-line targeted therapeutic agent for GISTs, markedly enhanced tumor growth suppression. In addition, DDR1/2 inhibition resulted in decreased KIT expression, while KIT inhibition led to upregulation of DDR1/2 expression in GISTs. The presence of DDR1/2 also decreased the sensitivity of wild-type KIT or primary KIT mutants to imatinib, indicating a possible role for DDR1/2 in promoting GIST survival during KIT-targeted therapy. The development of drug-resistant secondary KIT mutations is a primary factor contributing to GIST recurrence following targeted therapy. Similar to primary KIT mutants, DDR1/2 can associate with and enhance the activation of secondary KIT mutants, further diminishing their sensitivity to imatinib. In summary, our data demonstrate that DDR1/2 contribute to KIT activation in GISTs and strengthen resistance to imatinib for both primary and secondary KIT mutants, providing a rationale for further exploration of DDR1/2 targeting in GIST treatment.

  • Konferensbidrag (refereegranskat)abstract
    • With the rapid development of the intelligent vehicle industry, in order to improve the efficiency and usability of systems, the interface design of the Human-Computer Interaction of intelligent vehicles has received great attention. In this article, firstly, combined with the domestic and foreign taxonomy of automated driving systems and collaborative design concepts, a framework of human-computer collaborative interaction (Human-Engaged Automated Driving, HEAD) is proposed. Secondly, we discussed the engagement of people and intelligent vehicle at different levels. We analyzed the interaction flow chart under the Human-Computer Collaboration, established the information architecture of Human-Computer Engagement, and conducted the design practice on the Human-Machine Interface. Finally, a case study of the HMI design of Adaptive Cruise Control was performed to validate the HEAD framework. SUS usability test was performed together with an experiment evaluating interface elements in two interface designs: Original UI design and Iterative UI design. This also including a Likert scale questionnaire. The results show that HEAD can guide and improve the Human-Machine Interface design to enhance the efficiency of Human-Computer Interaction and user experience.

  • The proto-oncogene transcription factor Ets1 regulates neural crest development through Histone Deacetylase 1 to mediate output of bone morphogenetic protein signaling.
  • 2015
  • Ingår i: Journal of Biological Chemistry. - 1083-351X. ; 290:36, s. 21925-21938
  • Tidskriftsartikel (refereegranskat)abstract
    • The neural crest (NC) is a transient, migratory cell population that differentiates into a large variety of tissues including craniofacial cartilage, melanocytes, and peripheral nervous system. NC is initially induced at the border of neural plate and non-neuralectoderm by balanced regulation of multiple signaling pathways, among which an intermediate bone morphogenetic protein (BMP) signaling is essential for NC formation. Ets1, a proto-oncogene playing important roles in tumor invasion, has also been implicated in delamination of NC cells. In this study, we investigated Ets1 function in NC formation using Xenopus. Overexpression of ets1 repressed NC formation through down-regulation of BMP signaling. Moreover, ets1 repressed the BMP-responsive gene id3 that is essential for NC formation. Conversely, overexpression of id3 can partially rescue the phenotype of NC inhibition induced by ectopic ets1. Mechanistically, we found that Ets1 binds to id3 promoter as well as Histone Deacetylase 1 (HDAC1), suggesting that Ets1 recruits HDAC1 to the promoter of id3, thereby inducing Histone deacetylation of the id3 promoter. Thus, our studies indicate that Ets1 regulates NC formation through attenuating BMP signaling epigenetically.

    • In our work, we focused on the traffic conflict between car and motorcycle, which is common in China. In our design requirement process, we presented two traffic conflict situations and analyzed their key factors. Initial positioning and trajectory of encountered motorcycles were found to be the most important but difficult point in car driver’s awareness. Therefore, we propose two different prototypes of a LED visualization components for the car driver. This include changing color schemes and alteration of the amount of light in lamps. The goal was to design support to help the driver to detect motorcycles in the two corresponding traffic situations. The results show that changes in the amount of light in the lamps was performing better. In addition, a large set of qualitative feedback was collected and analyzed in the experiment.

  • Hypermethylation of endoplasmic reticulum disulfide oxidase 1α leads to trophoblast cell apoptosis through endoplasmic reticulum stress in preeclampsia

    • Abnormal trophoblast cell apoptosis is implicated in the pathogenesis of pregnancy-related disorders including preeclampsia (PE), and endoplasmic reticulum (ER) stress has been considered as a novel pathway in the regulation of cell apoptosis. In this study, we observed that both apoptosis and ER stress are triggered in trophoblast cells under hypoxia as well as in the placenta of PE rats. Quantitative polymerase chain reaction and Western blot analysis showed that the expression of endoplasmic reticulum disulfide oxidase 1α (ERO1α) is suppressed in trophoblast cells under hypoxia due to the hypermethylation of the ERO1α promoter region, and the inhibition of ERO1α expression plays an important role in ER stress and trophoblast cell apoptosis. Furthermore, we found that DNA methyltransferase 1 (DNMT1) is a key methyltransferase for DNA methylation in the regulation of ERO1α expression, and the binding level of DNMT1 to the ERO1α promoter is markedly elevated under hypoxia although DNMT1 expression is inhibited by hypoxia, suggesting that the binding level of DNMT1 to the ERO1α promoter region rather than the DNMT1 expression level contributes to the hypermethylation of ERO1α. Taken together, these results demonstrate that the hypermethylation of ERO1α mediated by increased binding of DNMT1 to the ERO1α promoter leads to trophoblast cell apoptosis through ER stress in the placenta of PE rats, which shed insight into the etiology of PE and might present a validated therapeutic target for the treatment of PE.

  • Homocysteine accelerates hepatocyte autophagy by upregulating TFEB via DNMT3b-mediated DNA hypomethylation

  • Ingår i: Acta Biochimica et Biophysica Sinica. - : China Science Publishing & Media Ltd.. - 1672-9145. ; 55:8, s. 1184-1192

    • Autophagy plays a critical role in the physiology and pathophysiology of hepatocytes. High level of homocysteine (Hcy) promotes autophagy in hepatocytes, but the underlying mechanism is still unknown. Here, we investigate the relationship between Hcy-induced autophagy level and the expression of nuclear transcription factor EB (TFEB). The results show that Hcy-induced autophagy level is mediated by upregulation of TFEB. Silencing of TFEB decreases the level of autophagy-related protein LC3BII/I and increases p62 expression level in hepatocytes after exposure to Hcy. Moreover, the effect of Hcy on the expression of TFEB is regulated by hypomethylation of the TFEB promoter catalyzed by DNA methyltransferase 3b (DNMT3b). In summary, this study shows that Hcy can activate autophagy by inhibiting DNMT3b-mediated DNA methylation and upregulating TFEB expression. These findings provide another new mechanism for Hcy-induced autophagy in hepatocytes.

    • The development of intelligent automobiles puts forward higher requirements for HMI design. The information obtained by drivers through vision accounts for 80%. As a key part of visual perception, color affects driving performance. This article investigates vehicle interface design about icon-background contrast. Three colors commonly used in car-machine interface are chosen: red, green and blue, and contrast is divided into five levels under daytime and nighttime. The participants tested three colors and the five contrasts in an experiment using classic paradigm of secondary tasks. We collected data from their driving behavior and subjective experience, trying to find a most appropriate value. The results show that recommended contrast values under different lighting conditions are 3:1 to 4:1 for red, blue, and green during the day; 3:1 for red at night, and 5:1 for blue and green, so that the vehicle central console color design references are formed.

  • Dose-effect relationship of apoptosis induced by fission-neutron in murine thymocytes
  • 2000
  • Ingår i: Chinese Journal of Radiological Medical Protection. ; 20, s. 87-87
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective To investigate the effectiveness of high LET fission-neutron to induce apoptosis in murine thymocytes and to compare it with that of low LET 60Co γ-ray. Methods Apoptosis induction was studied qualitatively by light and transmission electron microscopy and DNA gel electrophoresis, also quantitatively by flow cytometry(FCM) and diphenylamine (DPA)methods. Results DNA ladders of murine thymocytes were detectable, the typical apoptosis of thymocytes could be observed morphologically by means of light and electron microscopy at 6 h after fission-neutron irradiation with doses ranging from 0.5 to 5.0 Gy,meanwhile the percentages of apoptosis increased with increasing doses. After exposure to γ-rays with doses ranging from 1.0 to 30 Gy,the expermiental results were similar to those from neutron radiation. The incidence of apoptosis peaked at about 20 Gy,the percentages did not increase further when doses increased. Conclusion Apoptosis of murine thymocytes can be induced when mice are exposed to either fission-neutron (0.5-5.0 Gy) or to γ-ray (1-30 Gy).Although the relationship between apoptosis and radiation doses is similar, the percentage of apoptosis induced by neutron irradiation is higher than that induced by γ-irradiation.The RBE values of fission-neutron for inducing apoptosis murine thymocytes are 2.09 (by FCM method) and 2.37 (by DPA method),respectively. These results also suggest that fission-neutron-induced murine immune tissue is more severe than that induced by γ-rays at several hours post-irradiation and this might be the basis for heavy damage to immune tissues induced by fission-neutron-irradiation in later period.
