The aim of this investigation was to evaluate factors that may influence neurodevelopmental outcome in congenital hypothyroidism patients. The developmental quotient (DQ) of 155 children with congenital hypothyroidism was evaluated at 24 months of age, using Gesell Developmental Schedules (GDS), and compared with that of 310 healthy controls.   • A global survey on the use of the international classification of diseases codes for metabolic dysfunction-associated fatty liver disease.

    • With the implementation of the 11th edition of the International Classification of Diseases (ICD-11) and the publication of the metabolic dysfunction-associated fatty liver disease (MAFLD) nomenclature in 2020, it is important to establish consensus for the coding of MAFLD in ICD-11. This will inform subsequent revisions of ICD-11.Using the Qualtrics XM and WJX platforms, questionnaires were sent online to MAFLD-ICD-11 coding collaborators, authors of papers, and relevant association members.A total of 890 international experts in various fields from 61 countries responded to the survey. We also achieved full coverage of provincial-level administrative regions in China. 77.1% of respondents agreed that MAFLD should be represented in ICD-11 by updating NAFLD, with no significant regional differences (77.3% in Asia and 76.6% in non-Asia, p=0.819). Over 80% of respondents agreed or somewhat agreed with the need to assign specific codes for progressive stages of MAFLD (i.e. steatohepatitis) (92.2%), MAFLD combined with comorbidities (84.1%), or MAFLD subtypes (i.e., lean, overweight/obese, and diabetic) (86.1%).This global survey by a collaborative panel of clinical, coding, health management and policy experts, indicates agreement that MAFLD should be coded in ICD-11. The data serves as a foundation for corresponding adjustments in the ICD-11 revision.

    • The intensity of 9Be + p nuclear fusion reactions was experimentally studied during second harmonic (2ω CH) ion-cyclotron resonance heating (ICRH) and further analyzed during fundamental hydrogen minority ICRH of JET-ILW hydrogen and deuterium plasmas. In relatively low-density plasmas with a high ICRH power, a population of fast H+ ions was created and measured by neutral particle analyzers. Primary and secondary nuclear reaction products, due to 9Be + p interaction, were observed with fast ion loss detectors, γ-ray spectrometers and neutron flux monitors and spectrometers. The possibility of using 9Be(p, d)2α and 9Be(p, α)6Li nuclear reactions to create a population of fast alpha particles and study their behaviour in non-active stage of ITER operation is discussed in the paper.

    • Population stratification is a useful approach for a better understanding of complex biological problems in human health and wellbeing. The proposal that such stratification applies to the human gut microbiome, in the form of distinct community composition types termed enterotypes, has been met with both excitement and controversy. In view of accumulated data and re-analyses since the original work, we revisit the concept of enterotypes, discuss different methods of dividing up the landscape of possible microbiome configurations, and put these concepts into functional, ecological and medical contexts. As enterotypes are of use in describing the gut microbial community landscape and may become relevant in clinical practice, we aim to reconcile differing views and encourage a balanced application of the concept.

    • The Populus genus is one of the major plant model systems, but genomic resources have thus far primarily been available for poplar species, and primarily Populus trichocarpa (Torr. & Gray), which was the first tree with a whole-genome assembly. To further advance evolutionary and functional genomic analyses in Populus, we produced genome assemblies and population genetics resources of two aspen species, Populus tremula L. and Populus tremuloides Michx. The two aspen species have distributions spanning the Northern Hemisphere, where they are keystone species supporting a wide variety of dependent communities and produce a diverse array of secondary metabolites. Our analyses show that the two aspens share a similar genome structure and a highly conserved gene content with P. trichocarpa but display substantially higher levels of heterozygosity. Based on population resequencing data, we observed widespread positive and negative selection acting on both coding and noncoding regions. Furthermore, patterns of genetic diversity and molecular evolution in aspen are influenced by a number of features, such as expression level, coexpression network connectivity, and regulatory variation. To maximize the community utility of these resources, we have integrated all presented data within the PopGenIE web resource (PopGenIE.org).

  • Alternative positions of internal heat exchanger for CO2 booster refrigeration system : Thermodynamic analysis and annual thermal performance evaluation

    • Refrigeration systems running on transcritical CO2 cycle are considered an alternative to phase-down the use of hydrofluorocarbons (HFCs) in response to the Kigali Amendment. Responding to the need to improve the efficiency of CO2 booster systems and to identify optimum system designs, thermodynamic models of three booster systems are proposed with the use of an internal heat exchanger (IHX). Findings from this study suggest that placing the IHX with a low-temperature fluid side at the suction line of high pressure stage compressor and high-temperature fluid side at the outlet of gas cooler represents the most optimal approach in improving the coefficient of performance (COP) of the booster system. When operating in the transcritical conditions, the COP values can be improved by 6.35% at the IHX thermal effectiveness of 0.8 and by 6.48% at the ratio of medium temperature load to low temperature load of 6. Using IHX can significantly reduce the compressor discharge pressure, which can be reduced by 0.55 MPa at the ambient temperature of 40 degrees C. Furthermore, by adding IHX, the annual performance factor of CO2 system can be improved significantly by 1.68% and the annual total power consumption can be decreased by 6.48% in the tropical climate. It can be concluded that IHX can improve the COP values of a booster system when operating in the subtropical and tropical regions.

    • Rhenium oxides belonging to the family AReO4 where A is a metal cation, exhibit interesting electronic and magnetic properties. In this study we have utilized the muon spin rotation/relaxation (mu+SR) technique to study the magnetic properties of the MgReO4 compound. To the best of our knowledge, this is the first investigation reported on this interesting material, that is stabilized in a wolframite crystal structure using a special highpressure synthesis technique. Bulk magnetic studies show the onset of an antiferromagnetic (AF) long range order, or a possible singlet spin state at T-C1 approximate to 90 K, with a subtle second hightemperature transition at T-C2 approximate to 280 K. Both transitions are also confirmed by heat capacity (Cp) measurements. From our mu+SR measurements, it is clear that the sample enters an AF order below T-C1 = T-N approximate to 85 K. We find no evidence of magnetic signal above TN, which indicates that T-C2 is likely linked to a structural transition. Further, via sensitive zero field (ZF) mu(+) SR measurements we find evidence of a spin reorientation at T-Cant approximate to 65 K. This points towards a transition from a collinear AF into a canted AF order at low temperature, which is proposed to be driven by competing magnetic interactions.

    • Dry film resist SU-8 was used to make a thick mold for soft lithography of a poly(dimethylsiloxane) (PDMS) microfluidic chip with deep channels. The stacking of the SU-8 film enabled an ultra-thick (up to 500 μm) resist process on Si wafer. This process was fast and highly reproducible compared with the conventional liquid SU-8 process. The deep channel in the PDMS chip was utilized as a micro-flow cell for sensitive absorbance measurement. Sunset Yellow FCF dye was used to demonstrate absorption spectroscopy in the deep channel. Since the channel depth was proportional to the optical path length, which was proportional to the absorbance value, the PDMS chip achieved a detection limit (15.9 μM) comparable to U- or Z-shaped microfabricated absorbance detection cells in glass. Calibration curves for different solution concentrations were obtained with good R2 values (∼1). © 2012 The Royal Society of Chemistry.

    • An organic lateral resolution test device has been developed to measure the performance of imaging mass spectrometry (IMS) systems. The device contains periodic gratings of polyethylene glycol (PEG) and lipid bars covering a wide range of spatial frequencies. Microfabrication technologies were employed to produce well-defined chemical interfaces, which allow lateral resolution to be assessed using the edge-spread function (ESF). In addition, the design of the device allows for the direct measurement of the modulation transfer function (MTF) to assess image quality. Scanning electron microscopy (SEM) and time-of-flight secondary ion mass spectrometry (TOF-SIMS) were used to characterize the device. TOF-SIMS imaging was used to measure the chemical displacement of biomolecules in matrix-assisted laser desorption/ionization (MALDI) matrix crystals. In a proof-of-concept experiment, the platform was also used to evaluate MALDI matrix application methods, specifically aerosol spray and sublimation methods.

    • Membrane fusion involves the merging of two phospholipid bilayers in an aqueous environment and is involved in many cellular processes, such as cell exocytosis. Synaptic vesicles are nanometer-sized organelles, which are packaged with chemical messengers (e.g., neurotransmitters, neurohormones, and neuropeptides). Each presynaptic nerve terminal contains hundreds of synaptic vesicles. When an action potential depolarizes the presynaptic plasma membrane, Ca2+ channels open, and Ca2+ flows into the nerve terminal to trigger the exocytosis of synaptic vesicles, thereby vesicles migrate to the plasma membrane of a cell, fuse, and release their contents into the extracellular space. These messengers can then bind to receptors on a target cell, thus inducing a cascade of signaling events in a complex network (Figure 14.1). Until now, several types of neurotransmitters, such as amino acids, monoamines, and peptides, have been found, but their function in the brain is not always clear. Exocytotic events occur on a millisecond timescale with transmitter release proportions varying from zepto- to femtomole amounts per vesicle, making them experimentally challenging to monitor. Several different kinds of bioanalytical techniques have been developed to measure chemical messengers in the extracellular fluid following exocytosis from tissue in vivo and to measure individual exocytotic events at single cells under in vitro experimental conditions with biological or artificial models.

    • We reported an optical DNA/protein microfluidic sensor which consists of single stranded (ss) DNA-Cy3 probes on gold surface and simple line-shape microfluidic channel. These ssDNA-Cy3 probes with random sequence in bulk solution or on gold surface exhibits fluorescence enhancement after binding with complementary ssDNA (cssDNA) targets. Particularly it did not require complicated design or hairpin-like stem-loop conformation, which made it easier to be made and applied in analytes detection by fluorescence switching techniques. Using ssDNA-cy3 probes attached on gold surface in a microfluidic channel, strong fluorescence enhancement was measured by ssDNA with cssDNA binding or ssDNA with cssDNA-biotin binding. The following introduction of streptavidin resulted in fluorescence quenching (fluorescence decrease) because of the binding of hybridized DNA-biotin with streptavidin. This sensor showed strong affinity and high sensitivity toward the streptavidin, the minimum detectable concentration for streptavidin was 1 pM, equating to an absolute detection limit of 60 amol in this microfluidic channel. Microfluidic channel height and flow rate is optimized to increase surface reaction efficiency and fluorescence switching efficiency. In contrast to previously reported optical molecular beacon approach, this sensor can be used not only for the detection of cssDNA target, but also for the detection of streptavidin. This microfluidic sensor offers the promise of analyzing kinds of molecular targets or immunoreactions.

    • We report the application of individually addressable microelectrode arrays (MEAs) to study the heterogeneity of cell exocytosis at the subcellular level. Multiple subcellular-size electrodes are covered by a single PC12 cell for the investigation of subcellular exocytosis. PC12 cells have been seeded and cultured on top of three kinds of MEAs containing 16, 25, or 36 square microelectrodes (4 mm width in a 4 by 4 MEA, 3 mu m width in a 5 by 5 MEA, 2 mm width in a 6 by 6 MEA). After collagen coating, single cells were found to cover several electrodes and these were selected for the study of subcellular exocytosis. Amperometric results show that single cell and subcellular heterogeneity in single cell exocytosis can be electrochemically detected with these MEAs. The results also show that these MEAs are suitable for detecting fast chemical events at single cells, as well as for developing multifunctional electrochemical sensors.

    • We report the fabrication and characterization of micro well-based individually addressable ultra-microelectrode arrays (MEAs) and their application to spatially and temporally resolved detection of neurotransmitter release across a single pheochromocytoma (PC 12) cell. The MEAs consist of sixteen 4-μm-width square microelectrodes, or twenty-five 3-μm-width square microelectrodes, or thirty-six 2-jim-width square microelectrodes. Each MEA is tightly defined in a 30×30 μm square area, which is further encased inside a 40×40 μm SU-8 microwell. We demonstrate the excellent stability and reproducibility of these microelectrodes by using cyclic voltammetry and we have performed recording of spatially resolution of single cell exocytosis with multiple ultra-microelectrodes in 2-μm resolution.

    • We report the fabrication and characterization of microwell-based individually addressable microelectrode arrays (MEAs) and their application to spatially and temporally resolved detection of neurotransmitter release across a single pheochromocytoma (PC12) cell. The microwell-based MEAs consist of 16 4-μm-width square ultramicroelectrodes, 25 3-μm-width square ultramicroelectrodes, or 36 2-μm-width square ultramicroelectrodes, all inside a 40 × 40 μm square SU-8 microwell. MEAs were fabricated on glass substrates by photolithography, thin film deposition, and reactive ion etching. The ultramicroelectrodes in each MEA are tightly defined in a 30 × 30 μm square area, which is further encased inside the SU-8 microwell. With this method, we demonstrate that these microelectrodes are stable, reproducible, and demonstrate good electrochemical properties using cyclic voltammetry. Effective targeting and culture of a single cell is achieved by combining cell-sized microwell trapping and cell-picking micropipet techniques. The surface of the microelectrodes in the MEA was coated with collagen IV to promote cell adhesion and further single-cell culture, as good adhesion between the cell membrane and the electrode surface is critical for the quality of the measurements. Imaging the spatial distribution of exocytosis at the surface of a single PC12 cell has also been demonstrated with this system. Exocytotic signals have been successfully recorded from eight independent 2-μm-wide ultramicroelectrodes from a single PC12 cell showing that the subcellular heterogeneity in single-cell exocytosis can be precisely analyzed with these microwell-based MEAs.

    • We report the lithographic microfabrication of a movable thin film microelectrode array (MEA) probe consisting of 16 platinum band electrodes placed on top of a supporting borosilicate glass substrate. These 1.2 mu m wide electrodes were tightly packed and positioned parallel in two opposite rows within a 20 mu m x 25 mu m square area and with a distance less than 10 mu m from the edge of the glass substrate. We demonstrate the ability to control and place the probe in close proximity to the surface of adherent bovine chromaffin cells and to amperometrically record single exocytosis release events with high spatiotemporal resolution. The two-dimensional position of single exocytotic events occurring in the center gap area separating the two rows of MEA band electrodes and that were codetected by electrodes in both rows was determined by analysis of the fractional detection of catecholamine released between electrodes and exploiting random walk simulations. Hence, two-dimensional electrochemical imaging recording of exocytosis release between the electrodes within this area was achieved. Similarly, by modeling the current spikes codetected by parallel adjacent band electrodes positioned in the same electrode row, a one-dimensional imaging of exocytosis with submicrometer resolution was accomplished within the area. The one- and twodimensional electrochemical imaging using the MEA probe allowed for high spatial resolution of exocytosis activity and revealed heterogeneous release of catecholamine at the chromaffin cell surface.
