  • Imputation and subset-based association analysis across different cancer types identifies multiple independent risk loci in the TERT-CLPTM1L region on chromosome 5p15.33

    • Genome-wide association studies (GWAS) have mapped risk alleles for at least 10 distinct cancers to a small region of 63 000 bp on chromosome 5p15.33. This region harbors the TERT and CLPTM1L genes; the former encodes the catalytic subunit of telomerase reverse transcriptase and the latter may play a role in apoptosis. To investigate further the genetic architecture of common susceptibility alleles in this region, we conducted an agnostic subset-based meta-analysis (association analysis based on subsets) across six distinct cancers in 34 248 cases and 45 036 controls. Based on sequential conditional analysis, we identified as many as six independent risk loci marked by common single-nucleotide polymorphisms: five in the TERT gene (Region 1: rs7726159, P = 2.10 × 10(-39); Region 3: rs2853677, P = 3.30 × 10(-36) and PConditional = 2.36 × 10(-8); Region 4: rs2736098, P = 3.87 × 10(-12) and PConditional = 5.19 × 10(-6), Region 5: rs13172201, P = 0.041 and PConditional = 2.04 × 10(-6); and Region 6: rs10069690, P = 7.49 × 10(-15) and PConditional = 5.35 × 10(-7)) and one in the neighboring CLPTM1L gene (Region 2: rs451360; P = 1.90 × 10(-18) and PConditional = 7.06 × 10(-16)). Between three and five cancers mapped to each independent locus with both risk-enhancing and protective effects. Allele-specific effects on DNA methylation were seen for a subset of risk loci, indicating that methylation and subsequent effects on gene expression may contribute to the biology of risk variants on 5p15.33. Our results provide strong support for extensive pleiotropy across this region of 5p15.33, to an extent not previously observed in other cancer susceptibility loci.

    • The Cancer Genome Atlas (TCGA) Research Network has profiled and analyzed large numbers of human tumors to discover molecular aberrations at the DNA, RNA, protein and epigenetic levels. The resulting rich data provide a major opportunity to develop an integrated picture of commonalities, differences and emergent themes across tumor lineages. The Pan-Cancer initiative compares the first 12 tumor types profiled by TCGA. Analysis of the molecular aberrations and their functional roles across tumor types will teach us how to extend therapies effective in one cancer type to others with a similar genomic profile. © 2013 Nature America, Inc.   • Fabrication of Highly Porous CuAl Intermetallic by Thermal Explosion Using NaCl Space Holder

    • A high-porosity CuAl-based intermetallic compound with composition Cu-50 at.% Al has been successfully prepared by thermal explosion (TE) using NaCl as space holder. The results showed that the NaCl particles were completely removed from the green compact by water leaching. The temperature of the specimen during the TE and the evolution of the porous microstructure were investigated. The TE was ignited at 560°C, and the specimen temperature increased to 775°C in 3 s, resulting in formation of intermetallic CuAl and CuAl2 phases in the final product. A porous CuAl-based intermetallic compound with up to 62 vol.% open porosity was produced when adding 60 vol.% NaCl. The compound exhibited a bimodal pore size structure, including large pores (200 μm to 300 μm) that replicated the NaCl particles and small pores (5 μm to 10 μm) interspersed in the pore walls. Moreover, the large pores were interconnected by channels and formed an open CuAl-based intermetallic cellular structure, having great potential for use in heat exchange and filtration applications.

  • Oxidation Resistance of Highly Porous Fe-Al Foams Prepared by Thermal Explosion

    • Open-cell Fe-Al intermetallic foams were successfully prepared by a simple and energy-saving thermal explosion (TE) process. The effects of the Fe/Al molar ratio (Fe-(40–50) at. pct Al) and thermal treatment temperature on the TE temperature profile, phase composition, pore characteristics, and oxidation resistance of the prepared foams were investigated. The results showed that the Al content significantly influenced the ignition (Tig) and combustion (Tc) temperatures of the TE process; in particular, as the Al content decreased, Tig increased gradually from 623 °C to 636 °C and Tc decreased from 1059 °C to 981 °C. FeAl was identified as the dominant phase in the thermally treated foams. The Fe-Al intermetallic foams displayed an open porosity of 60 vol pct, with pores connected with each other to form an open pore structure. The formation of the pores was attributed to the expansion of interparticle pores in the pressed body during the TE reaction. X-ray photoelectron spectroscopy analysis of the Fe-50Al foam showed that the Al 2p and O 1s binding energies were 74.5 eV and at 531.4 eV, respectively. The formation of a surface alumina layer in the early stages of the oxidation process resulted in the parabolic oxidation rate law, and the Fe-50Al foams exhibited an excellent resistance to oxidation at 650 °C in air. These results suggest that the synthesized Fe-Al foams represent promising materials for applications involving an oxidizing environment and high temperatures.

    • Due to the high exothermic characteristic of NiAl during the reaction synthesis process from Ni–Al elemental powders, the NiAl intermetallic melts frequently, and the specimens are difficult to maintain their original shape, which leads to the inhomogeneity of the pore size and morphology. To tackle this problem, porous NiAl intermetallic monolith with controllable shape and pore structure was prepared through thermal explosion (TE) using NaCl as space holder. The TE behavior was recorded, and the effect of the volume fraction of NaCl on the phase composition, macroscopic feature, pore morphology and open porosity were investigated. The results showed that NiAl was the main phase in the products, and the specimen was free from cracking or deforming when NaCl content reached 30 vol%. The interconnected channel and pore windows were formed, and the open porosity was improved greatly to 63% by adding 50 vol% NaCl. The leachable space holder route provides a simple way to control the shape, pore structure and open porosity of the synthesized porous NiAl intermetallic.

    • Absorption by pairs of H2 molecules is an important opacity source in the atmospheres of the outer planets, and thus of special astronomical interest. The emission spectra of cool white dwarf stars differ significantly from the expected blackbody spectra, amongst other reasons due to absorption by H2-H2, H2-He, and H2-H collisional complexes in the stellar atmospheres. To model the radiative processes in these atmospheres, which have temperatures of several thousand kelvin, one needs accurate knowledge of the induced dipole (ID) and potential energy surfaces (PES) of such collisional complexes. These come from quantum-chemical calculations with the H2 bonds stretched or compressed far from equilibrium. Laboratory measurements of collision-induced (CI) absorption exist only at much lower temperature. For H2 pairs at room temperature, the calculated spectra of the rototranslational band, the fundamental band, and the first overtone match the experimental data very well. In addition, with the newly obtained IDS it became possible to reproduce the measurements in the far blue wing of the rototranslational spectrum of H2 at 77.5 K, as well as at 300 K. Similarly good agreement between theory and measurement is seen in the fundamental band of molecular deuterium at room temperature. Furthermore, we also show the calculated absorption spectra of H2-He at 600 K and of H2-H2 at 2,000 K, for which there are no experimental data for comparison

    • A well-functioning placenta is essential for fetal and maternal health throughout pregnancy. Using placental weight as a proxy for placental growth, we report genome-wide association analyses in the fetal (n=65,405), maternal (n=61,228) and paternal (n=52,392) genomes, yielding 40 independent association signals. Twenty-six signals are classified as fetal, four maternal and three fetal and maternal. A maternal parent-of-origin effect is seen near KCNQ1. Genetic correlation and colocalization analyses reveal overlap with birth weight genetics, but 12 loci are classified as predominantly or only affecting placental weight, with connections to placental development and morphology, and transport of antibodies and amino acids. Mendelian randomization analyses indicate that fetal genetically mediated higher placental weight is causally associated with preeclampsia risk and shorter gestational duration. Moreover, these analyses support the role of fetal insulin in regulating placental weight, providing a key link between fetal and placental growth.

    • Head impact usually causes fatalities of riders in motorcycle accidents. The use of helmet can protect head effectively and the study of head injury mechanism is useful in safety helmet design and head injury risk decrease. A real motorcycle-passenger car accident was reconstructed in order to simulate and analyze the motion, impact velocity, impact angle in the collision. In order to study the dynamic response of helmeted head during the accident impact processing, an effective helmet model was constructed and validated. Coupling with the finite element model of human head, the injury mechanism of head and relevant parameters of biomechanics were studied and the protection ability of helmet was analyzed. It was found that the presence of helmet reduces the probility of head injury.

    • This paper aims to determine the correlation of the throw distance of cyclist and electric bicycle in traffic accidents with throw vehicle impact speed. A total of 22 vehicle-electric bicycle accidents were sampled from IVAC database, China. The selected accidents were reconstructed by using PC-Crash and Madymo programs. The relationship between throw distance and vehicle impact velocity was analyzed by using logistic regression model The relationship curves were obtained based on the calculated parameters. With increasing of the vehicle impact velocity, an increase of throw distance was observed. When the vehicle impact velocity was above 30km/h, the throw distance of electric bicycle was larger than that of cyclist. By comparing of results from the PC-Crash and MADYMO simulations with observations from in-depth accident investigation, the feasibility of the reconstruction method and the validity of the numerical model were evaluated. This research could provide a basis for further study on the dynamic response and injury prevention of cyclist.

  • Toward a Coordinated Global Observing System for Seagrasses and Marine Macroalgae

    • In coastal waters around the world, the dominant primary producers are benthic macrophytes, including seagrasses and macroalgae, that provide habitat structure and food for diverse and abundant biological communities and drive ecosystem processes. Seagrass meadows and macroalgal forests play key roles for coastal societies, contributing to fishery yields, storm protection, biogeochemical cycling and storage, and important cultural values. These socio-economically valuable services are threatened worldwide by human activities, with substantial areas of seagrass and macroalgal forests lost over the last half-century. Tracking the status and trends in marine macrophyte cover and quality is an emerging priority for ocean and coastal management, but doing so has been challenged by limited coordination across the numerous efforts to monitor macrophytes, which vary widely in goals, methodologies, scales, capacity, governance approaches, and data availability. Here, we present a consensus assessment and recommendations on the current state of and opportunities for advancing global marine macrophyte observations, integrating contributions from a community of researchers with broad geographic and disciplinary expertise. With the increasing scale of human impacts, the time is ripe to harmonize marine macrophyte observations by building on existing networks and identifying a core set of common metrics and approaches in sampling design, field measurements, governance, capacity building, and data management. We recommend a tiered observation system, with improvement of remote sensing and remote underwater imaging to expand capacity to capture broad-scale extent at intervals of several years, coordinated with strati fied in situ sampling annually to characterize the key variables of cover and taxonomic or functional group composition, and to provide ground-truth. A robust networked system of macrophyte observations will be facilitated by establishing best practices, including standard protocols, documentation, and sharing of resources at all stages of work flow, and secure archiving of open-access data. Because such a network is necessarily distributed, sustaining it depends on close engagement of local stakeholders and focusing on building and long-term maintenance of local capacity, particularly in the developing world. Realizing these recommendations will producemore effective, efficient, and responsive observing, a more accurate global picture of change in vegetated coastal systems, and stronger international capacity for sustaining observations.

    • Quantum chemical methods are used to obtain the interaction-induced dipole surfaces (IDS) of complexes of two interacting (i.e. colliding) molecules, for example H2–H2, H2–He, etc., collisional complexes, along with their potential energy surfaces (PES). Eight H2 bond distances, from 0.942 to 2.801 bohr, are chosen for each H2 molecule to account for rotovibrational excitations. Rotovibrational matrix elements of these ID and PE surfaces are computed as necessary for the study of supermolecular (‘collision-induced’) absorption spectra of dense hydrogen gas, and of gaseous mixtures of hydrogen and helium, at temperatures up to several thousand kelvin and for frequencies from 0 to those of several H2 overtone bands. Rotovibrational state to state scattering calculations couple the collisional complex perturbatively to single photons. The absorption process causes rotovibrational transitions in one molecule, or simultaneous transitions in both molecules (when H2–H2 collisional complexes are considered). The spectral profiles of tens of thousands of such transitions are computed from first principles. Individual ‘lines’ are very broad so that they overlap substantially, forming a supermolecular quasi-continuum. The comparison of the computed collision-induced absorption (CIA) spectra with existing laboratory measurements at low temperatures (≤ 300 K) shows close agreement so that our results for higher temperatures, where laboratory experiments do not exist, may be used with confidence. Similar calculations of CIA spectra at high temperatures and frequencies are underway for other collisional systems (e.g. H2–H) of interest in astrophysical applications (e.g. ‘cool’ stellar atmospheres). Collision-induced Raman spectra (CIRS) have been similarly obtained; computed Raman spectra also compare favourably with existing laboratory measurements.

    • A molecule or a molecular system always consists of excited states of different spin multiplicities. With conventional optical excitations, only the (bright) states with the same spin multiplicity of the ground state could be directly reached. How to reveal the dynamics of excited (dark) states remains the grand challenge in the topical fields of photochemistry, photophysics, and photobiology. For a singlet-triplet coupled molecular system, the (bright) singlet dynamics can be routinely examined by conventional femtosecond pump-probe spectroscopy. However, owing to the involvement of intrinsically fast decay channels such as intramolecular vibrational redistribution and internal conversion, it is very difficult, if not impossible, to single out the (dark) triplet dynamics. Herein, we develop a novel strategy that uses an ultrafast broadband white-light continuum as a excitation light source to enhance the probability of intersystem crossing, thus facilitating the population flow from the singlet space to the triplet space. With a set of femtosecond time-reversed pump-probe experiments, we report on a proof-of-concept molecular system (i.e., the malachite green molecule) that the pure triplet dynamics can be mapped out in real time through monitoring the modulated emission that occurs solely in the triplet space. Significant differences in excited-state dynamics between the singlet and triplet spaces have been observed. This newly developed approach may provide a useful tool for examining the elusive dark-state dynamics of molecular systems and also for exploring the mechanisms underlying molecular luminescence/photonics and solar light harvesting.

    • The pancreatic islet is a highly structured micro-organ that produces insulin in response to rising blood glucose. Here we develop a label-free and automatic imaging approach to visualize the islets in situ in diabetic rodents by the synchrotron radiation X-ray phase-contrast microtomography (SRμCT) at the ID17 station of the European Synchrotron Radiation Facility. The large-size images (3.2 mm × 15.97 mm) were acquired in the pancreas in STZ-treated mice and diabetic GK rats. Each pancreas was dissected by 3000 reconstructed images. The image datasets were further analysed by a self-developed deep learning method, AA-Net. All islets in the pancreas were segmented and visualized by the three-dimension (3D) reconstruction. After quantifying the volumes of the islets, we found that the number of larger islets (=>1500 μm3) was reduced by 2-fold (wt 1004 ± 94 vs GK 419 ± 122, P < 0.001) in chronically developed diabetic GK rat, while in STZ-treated diabetic mouse the large islets were decreased by half (189 ± 33 vs 90 ± 29, P < 0.001) compared to the untreated mice. Our study provides a label-free tool for detecting and quantifying pancreatic islets in situ. It implies the possibility of monitoring the state of pancreatic islets in vivo diabetes without labelling.

  • A population-based survey of the epidemiology of symptom-defined gastroesophageal reflux disease : the Systematic Investigation of Gastrointestinal Diseases in China

    • Background: The epidemiology of gastroesophageal reflux disease (GERD) has yet to be investigated using the symptomatic threshold criteria recommended by the Montreal Definition. This study aimed to determine the prevalence of symptom-defined GERD across five regions of China, and to investigate variables associated with GERD. Methods: A representative sample of 18 000 adults (aged 18-80 years) were selected equally from rural and urban areas in each region (n = 1800). According to the Montreal Definition, GERD is present when mild symptoms of heartburn and/ or regurgitation occur on >= 2 days a week, or moderate-to-severe symptoms of heartburn and/or regurgitation occur on >= 1 day a week. Results: In total, 16 091 participants completed the survey (response rate: 89.4%) and 16 078 responses were suitable for analysis. Applying the Montreal criteria, the prevalence of symptom-defined GERD was 3.1% and varied significantly (p < 0.001) among the five regions (from 1.7% in Guangzhou to 5.1% in Wuhan) and between rural and urban populations (3.8% vs 2.4%). Factors significantly associated with GERD included living in a rural area and a family history of gastrointestinal diseases. Conclusions: This population-based survey found that the prevalence of symptom-defined GERD in China was 3.1%, which is lower than that found in Western countries.

    • Signal transduction from the T cell receptor (TCR) is crucial for T cell-mediated immune responses and, when deregulated, also contributes to the development of autoimmunity. How TCR signaling is regulated is incompletely understood. In this study, we demonstrate a ubiquitin-dependent mechanism in which the deubiquitinase Otud7b has a crucial role in facilitating TCR signaling. Upon TCR ligation, Otud7b is rapidly recruited to the tyrosine kinase Zap70, a central mediator of TCR-proximal signaling. Otud7b deficiency attenuates the activation of Zap70 and its downstream pathways and impairs T cell activation and differentiation, rendering mice refractory to T cell-mediated autoimmune and inflammatory responses. Otud7b facilitated Zap70 activation by deubiquitinating Zap70, thus preventing the association of Zap70 with the negative-regulatory phosphatases Sts1 and Sts2. These findings establish Otud7b as a positive regulator of TCR-proximal signaling and T cell activation, highlighting the importance of deubiquitination in regulating Zap70 function.

    • Climate change brings about significant changes in the physical environment in the Arctic. Increasing temperatures, sea ice retreat, slumping permafrost, changing sea ice regimes, glacial loss and changes in precipitation patterns can all affect how contaminants distribute within the Arctic environment and subsequently impact the Arctic ecosystems. In this review, we summarized observed evidence of the influence of climate change on contaminant circulation and transport among various Arctic environment media, including air, ice, snow, permafrost, fresh water and the marine environment. We have also drawn on parallel examples observed in Antarctica and the Tibetan Plateau, to broaden the discussion on how climate change may influence contaminant fate in similar cold-climate ecosystems. Significant knowledge gaps on indirect effects of climate change on contaminants in the Arctic environment, including those of extreme weather events, increase in forests fires, and enhanced human activities leading to new local contaminant emissions, have been identified. Enhanced mobilization of contaminants to marine and freshwater ecosystems has been observed as a result of climate change, but better linkages need to be made between these observed effects with subsequent exposure and accumulation of contaminants in biota. Emerging issues include those of Arctic contamination by microplastics and higher molecular weight halogenated natural products (hHNPs) and the implications of such contamination in a changing Arctic environment is explored.

    • The structural phase transition accompanied by a Jahn-Teller switch has been studied over a range of H/D ratios in (NH4)2[Cu(H2O)6](SO4)2 (ACTS). In particular, single-crystal neutron diffraction investigations of crystals with deuteration in the range 50 to 82% are shown to be consistent with previous electron paramagnetic resonance (EPR) experiments exhibiting a phase boundary at 50% deuteration under ambient pressure. Polycrystalline samples show that the two phases can co-exist. In addition, single-crystal neutron and polycrystalline X-ray diffraction pressure experiments show a shift to lower pressure at 60% deuteration versus previous measurements at 100% deuteration.

    • To recycle elemental molybdenum from waste molybdenum disilicide (MoSi2) heating elements, the MoSi2 was first disintegrated to MoO3 and SiO2 powders in air at a pest oxidation temperature of 500 °C. X-ray diffraction (XRD) patterns confirmed the completion of the pest oxidation reaction. The mixture of MoO3 and SiO2 powders were heated to 950 °C in a tube furnace to evaporate MoO3, and the XRD patterns of the residue showed that only SiO2 was left in the crucible, confirming that the MoO3 was removed through thermal evaporation. The collected MoO3 crystals had a striped morphology. Photocatalytic performance of MoO3 showed superior activity in comparison with commercial MoO3 and P25 for the degradation of methylene blue under visible light irradiation. The photocatalytic degradation activity of MoO3 synthesized by thermal evaporation at 950 °C was 99.25% in 60 min.

    • New ab initio results are reported for the interaction-induced changes in the dipole moments and polarizabilities of pairs of hydrogen molecules, computed using finite-field coupled-cluster methods in MOLPRO 2000 and GAMESS, with an aug-cc-pV5Z (spdf) basis set. Earlier work by X. Li, C. Ahuja, J. F. Harrison, and K. L. C. Hunt, J. Chem. Phys. 126, 214302 (2007), on collision-induced polarizabilities Δα has been extended with 170 additional geometrical configurations of the H2 pairs. In calculations of Δα, we have used a "random field" technique, with up to 120 different field strengths, having components that range from 0.001 to 0.01 a.u. Numerical tests show that the pair dipoles Δμ can be obtained accurately from calculations limited to 6 values of the field in each direction, so this approach has been used to compute Δμ by X. Li, K. L. C. Hunt, F. Wang, M. Abel, and L. Frommhold, Int. J. Spectroscopy 2010, 371201 (2010). We have evaluated the collision-induced dipoles of H2 pairs for 28 combinations of bond lengths (ranging from 0.942 a.u. to 2.801 a.u.), 7 intermolecular separations R, and 17 different relative orientations. In our work on Δα, the bond lengths are fixed at 1.449 a.u. Our results agree well with the previous ab initio work of W. Meyer, A. Borysow, and L. Frommhold, Phys. Rev. A 40, 6931 (1989), and of Y. Fu, C. G. Zheng and A. Borysow, J. Quant. Spectroscopy and Rad. Transfer, 67, 303 (2000)-where those data exist-for Δμ of H2 pairs. For Δα, our results agree well with the CCSD(T) results obtained by G. Maroulis, J. Phys. Chem. A 104, 4772 (2000) for two pair orientations and fixed R. The pair polarizability anisotropies also agree well with the small-basis self-consistent field results of D. G. Bounds, Mol. Phys. 38, 2099 (1979), although the trace of the polarizability differs by factors of 2 or more from Bounds' results. We have determined the expansion coefficients for Δμ and Δα, expressed as series in the spherical harmonics of the orientation angles of the intermolecular vector and of unit vectors along the molecular axes. The leading coefficients converge at long range to the predictions from perturbation theory, derived by J. E. Bohr and K. L. C. Hunt, J. Chem. Phys. 87, 3821 (1987); T. Bancewicz, W. G.az, and S. Kielich, Chem. Phys. 128, 321 (1988); and X. Li and K. L. C. Hunt, J. Chem. Phys. 100, 7875 (1994); ibid, 9276 (1994). Based on our results for Δμ, we find excellent agreement for the binary rototranslational absorption spectrum of H2 at 297.5 K as calculated by X. Li, K. L. C. Hunt, F. Wang, M. Abel, and L. Frommhold, Int. J. Spectroscopy 2010, 371201 (2010) and as determined experimentally by G. Bachet, E. R. Cohen, P. Dore, and G. Birnbaum, Can. J. Phys. 61, 591 (1983), out to ∼1500 cm-1. We have also calculated the vibrational spectra out to 20,000 cm-1, at T = 600 K, 1000 K, and 2000 K, for which there are no experimental data. We are currently working to extend the temperature range in the calculations to 7000 K, for application in modeling the spectra of cool white dwarf stars. We have used the results for Δα to calculate collision-induced rototranslational Raman spectra for H2 pairs [M. Gustafsson, L. Frommhold, X. Li, and K. L. C. Hunt, J. Chem. Phys. 130, 164314 (2009)]. Experimental results for the Raman spectra have been reported by U. Bafile, M. Zoppi, F. Barocchi, M. S. Brown, and L. Frommhold, Phys. Rev. A 40, 1654 (1989); U. Bafile, L. Ulivi, M. Zoppi, F. Barocchi, M. Moraldi, and A. Borysow, Phys. Rev. A 42, 6916 (1990); and M. S. Brown, S.-K. Wang, and L. Frommhold, Phys. Rev. A 40, 2276 (1989). Agreement between our calculations and experiment is good for both the polarized and depolarized spectra, with the remaining discrepancies probably attributable to the difference between the static (calculated) and frequency-dependent (experimental) values of Δα.

    • We propose a new approach to fabricate the graphene nanomesh through the local catalytic hydrogenation of carbon by Cu nanoparticles. It allows to tune the size and density of the holes in the nanomesh as well as the total edge length of the holes through the control of the thickness of the Cu film. The upshift of both G and 2D peaks in Raman spectra of the graphene nanomeshes indicates that the nanomesh is spontaneously p-type doped. Moreover, the split of G peak reveals that the doping is localized near the edge region of the hole in the nanomesh. Importantly, the nanomesh shows improved chemical enhancement for Raman spectra of absorbed RhB molecules as compared to the graphene. The edges in the nanomesh can enhance Raman spectroscopy via increasing both the local charge transfer and the ability to absorb RhB molecules. The results show that the graphene nanomesh has a great potential for the rapid and sensitive detection for the environmental monitoring and food security.

  • A novel fabrication strategy for highly porous FeAl/Al2O3 composite by thermal explosion in vacuum

    • The high porosity of FeAl/Al2O3 composites was successfully prepared via a low-energy consumption method of thermal explosion (TE) in vacuum from reactant mixtures of Fe, Al and Fe2O3. The temperature profiles, phase compositions, microstructure, porosity and pore size of the products were investigated. The TE reactions were ignited between 639 and 648 °C and maximum combustion temperatures reached to 1196–1867 °C. XRD patterns showed that FeAl, Fe2Al5 and Al2O3 were formed via TE reaction, and FeAl and Al2O3 were evolved as dominant phase after the final sintering at 1100 °C. The FeAl/Al2O3 composites exhibited an interconnected pore structure with porosities and pore size of 52–61% and 27–32 μm, respectively.

    • The ability of light to cause pain is paradoxical. The retina detects light but is devoid of nociceptors while the trigeminal sensory ganglia (TG) contain nociceptors but not photoreceptors. Melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs) are thought to mediate light-induced pain but recent evidence raises the possibility of an alternative light responsive pathway independent of the retina and optic nerve. Here, we show that melanopsin is expressed in both human and mouse TG neurons. In mice, they represent 3% of small TG neurons that are preferentially localized in the ophthalmic branch of the trigeminal nerve and are likely nociceptive C fibers and high-threshold mechanoreceptor Aδ fibers based on a strong size-function association. These isolated neurons respond to blue light stimuli with a delayed onset and sustained firing, similar to the melanopsin-dependent intrinsic photosensitivity observed in ipRGCs. Mice with severe bilateral optic nerve crush exhibit no light-induced responses including behavioral light aversion until treated with nitroglycerin, an inducer of migraine in people and migraine-like symptoms in mice. With nitroglycerin, these same mice with optic nerve crush exhibit significant light aversion. Furthermore, this retained light aversion remains dependent on melanopsin-expressing neurons. Our results demonstrate a novel light-responsive neural function independent of the optic nerve that may originate in the peripheral nervous system to provide the first direct mechanism for an alternative light detection pathway that influences motivated behavior.

    • The ecological effects of mass-flowering crops on pollinator abundance and species richness of neighbouring habitats are well established, yet the potential evolutionary consequences remain unclear. We studied effects of proximity to a mass-flowering crop on the pollination of local co-flowering plants and on patterns of natural selection on a pollination-generalised plant on the Tibetan Plateau. We recorded pollinator visitation rates and community composition at different distances (near vs. far) to oilseed rape (Brassica napus) fields in two habitat types and quantified pollinator-mediated selection on attractive traits of Trollius ranunculoides. The proximity to oilseed rape increased pollinator visitation in neighbouring alpine meadows and changed pollinator composition in neighbouring shrub meadows. Trollius ranunculoides in the alpine meadow near oilseed rape received three times more pollinator visits (mainly bees) and consequently had a 16.5% increase in seed set but also received slightly more heterospecific pollen per stigma. In contrast, pollinator visitation to T. ranunculoides in the shrub meadow near oilseed rape was three times lower (mainly flies), leading to a 10.7% lower seed despite no effect on pollen deposition. The proximity to the oilseed rape field intensified pollinator-mediated selection on flower size and weakened selection on flower height of T. ranunculoides in the alpine meadow but did not affect phenotypic selection on either trait in the shrub meadow. Our study highlights context-dependent variation in plant–pollinator interactions close to mass-flowering oilseed rape, suggesting potential effects on the evolution of flower traits of native plants through altered pollinator-mediated selection. However, context dependence may make these effects difficult to predict.

    • The impact scenarios, impact velocity were investigated based on 224 car-electric bicycle collision cases in IVAC (In-depth Investigation of Car Accident in Changsha) database. Cars were classified into three types: sedan, SUV and one box. According to analysis of traffic accident data, car-electric bicycle collision models were built up by using MADYMO. In order to analyze the kinematics of electric-bicycle-riders, this paper conducted a series of numerical experiments by changing the types of cars. Results show that the most prominent impact modes were frontal impact (54%) and side impact (33%), rider's head tend to strike sedan at upper part of the windscreen', 'SUV at the front end of bonnet, and one box at lower part of the windscreen, The peak values of head's and pelvis' acceleration were higher and occurred earlier when a SUV or one box impacted with electric bicycle.

  • A critical role of the mechanosensor PIEZO1 in glucose-induced insulin secretion in pancreatic beta-cells

    • Glucose-induced insulin secretion depends on beta-cell electrical activity. Inhibition of ATP-regulated potassium (K-ATP) channels is a key event in this process. However, K-ATP channel closure alone is not sufficient to induce beta-cell electrical activity; activation of a depolarizing membrane current is also required. Here we examine the role of the mechanosensor ion channel PIEZO1 in this process. Yoda1, a specific PIEZO1 agonist, activates a small membrane current and thereby triggers beta-cell electrical activity with resultant stimulation of Ca2+-influx and insulin secretion. Conversely, the PIEZO1 antagonist GsMTx4 reduces glucose-induced Ca2+-signaling, electrical activity and insulin secretion. Yet, PIEZO1 expression is elevated in islets from human donors with type-2 diabetes (T2D) and a rodent T2D model (db/db mouse), in which insulin secretion is reduced. This paradox is resolved by our finding that PIEZO1 translocates from the plasmalemma into the nucleus (where it cannot influence the membrane potential of the beta-cell) under experimental conditions emulating T2D (high glucose culture). beta-cell-specific Piezo1-knockout mice show impaired glucose tolerance in vivo and reduced glucose-induced insulin secretion, beta-cell electrical activity and Ca2+ elevation in vitro. These results implicate mechanotransduction and activation of PIEZO1, via intracellular accumulation of glucose metabolites, as an important physiological regulator of insulin secretion. Insulin secretion depends on action potential firing in pancreatic islet beta-cells, but the underlying mechanism is unclear. Here, the authors show that activation of the mechanosensor ion channel PIEZO1 plays a central role in beta-cell electrical activity and insulin release.

    • This paper proposes a spatial three degrees of freedom (DOF) parallel kinematic machine enhanced by a passive leg and a web-based remote control system. First, the geometric model of the parallel kinematic machine is addressed. In the mechanism, a fourth kinematic link-a passive link connecting the base center to the moving platform center-is introduced. Each of the three parallel limbs is actuated by one prismatic joint, respectively. The additional link has three passive DOF, namely two rotations around x and y axes and one translation along z axis. With the existence of this link, the unwanted motion of the tool (located in the moving platform) is constrained. The fourth link also enhances the global stiffness of the structure and distributes the torque from machining. With the kinematic model, a web-based remote control approach is applied. The concept of the web-based remote manipulation approach is introduced and the principles behind the method are explored in detail. Finally, a remote manipulation is demonstrated to the proposed structure using web-based remote control concept.

    • Graphene oxide (GO) is an attractive alternative for large-scale production of graphene, but its general structure is still under debate due to its complicated nonstoichiometric nature. Here we perform a set of femto-second pump-probe experiments on as-synthesized GO to extrapolate structural information in situ. Remarkably, it is observed that, in these highly oxidized GO samples, the ultrafast graphene-like dynamics intrinsic to pristine graphene is completely dominant over a wide energy region and can be modified by the localized impurity states and the electron-phonon coupling under certain conditions. These observations, combined with the X-ray photoelectron spectroscopy analysis and control experiments, lead to an important conclusion that GO consists of two types of domain, namely the carbon-rich graphene-like domain and the oxygen-rich domain. This study creates a new understanding of the realistic domain structure and properties of as-synthesized GO, offering useful guidance for future applications based on chemically modified/functionalized graphenes.

    • Here we present a finished sequence of human chromosome 15, together with a high-quality gene catalogue. As chromosome 15 is one of seven human chromosomes with a high rate of segmental duplication, we have carried out a detailed analysis of the duplication structure of the chromosome. Segmental duplication in chromosome 15 are largely clustered in two regions, on proximal and distal 15q; the proximal region is notable because recombination among the segmental duplications can result in deletions causing Prader-Willi and Angelman syndromes. Sequence analysis shows that the proximal and distal regions of 15q share extensive ancient similarity. Using a simple approach, we have been able to reconstruct many of the events by which the current duplication structure arose. We find that most of the intrachromosomal duplications seem to share a common ancestry. Finally, we demonstrate that some remaining gaps in the genome sequence are probably due to structural polymorphisms between haplotypes; this may explain a significant fraction of the gaps remaining in the human genome.
